본 발명은 단백질을 생산할 수 있으며 무글루타민 배지에서 성장할 수 있는 신규한 글루타민-요구성 인간세포에 관한 것이다. 나아가, 이는 단백질을 생산하는 신규한 방법 및 글루타민-요구성 인간세포에서 선택성 마커로서의 글루타민 합성효소(GS)의 용도에 관한 것이다.
포유류세포 배양에 의한 단백질 생산은 치료 및 임상 적용용 단백질을 공급하는데 사용된다. 오늘날 포유류세포 배양은 인간 및 동물 약품, 특히 비교적 크고, 복잡하며 글리코실화된 약품들에서의 사용을 위한 다수의 중요한 단백질의 선호되는 소스(source)이다 (N.B. Finter 등, Large-Scale Mammalian Cell Culture Technology, 1990, ed. A.S. Lubiniecki, Marcel Dekker, Inc., New York).
예를 들면, 세포 배양에 의한 인간 단백질 에리쓰로포이에틴(erythropoietin)(EPO)의 생산이 WO 93/09222에 기술되어 있다. 인간 EPO는 이를 코딩하는 외인성 유전자로 전달이입(transfection)된 인간섬유모세포를 사용하여 상당한 특이적 생산 속도로 수득되었다. 인간 EPO의 생산을 위한 추가의 방 법(WO 94/12650)에서, 내인성 코딩된 EPO를 활성화할 수 있는 DNA 서열로 전달이입된 인간 섬유육종 세포주 HT1080이 사용되었다. 유사한 접근법이 WO 99/09268에 기술되어 있다. Namalwa, Hela S3 및 HT 1080세포 같은 불멸화된 인간세포가 내인성 코딩된 EPO를 활성화할 수 있는 DNA 서열로 전달이입되었다.
인간 EPO 생산을 위해 사용된 WO 93/09222, WO 94/12650 및 WO 99/09268에 기술된 세포들은 모두 글루타민을 포함하는 배지에서 배양되어 왔다. 이는 글루타민이 배양되는 세포에 의해 에너지 기질로 사용되는 경우에, 세포독성이 있으며, 세포성장을 방해하는 암모니아가 생산되는 이화대사산물이기 때문에 불리하다. 더구나, 이는 세포의 골지체내의 pH에의 영향으로 인해 단백질의 글리코실화를 방해할 수 있다.
글리코실화 단백질인, 높은 수준의 조직 플라스미노겐 활성제 생산이 WO 87/04462에 기술되어 있다. GS는 여기에서, GS를 코딩하는 유전자를 세포에 전달이입함으로써 글루타민-기본영양(prototrophic) Chinese Hamster Ovary (CHO) 세포에서 조직 플라스미노겐 활성제 (tPA)를 코딩하는 유전자를 공증폭하기 위한 증폭 시스템으로서 사용된다. 그러나 EP-A 148 605에서 발견되는 경우와 같이, 글리코실화된 인간 단백질의 생산을 위해 CHO 세포를 사용하는 것은 불리하다. CHO 세포에 의해 합성된 단백질은 자연적으로 나타나는 글리코실화된 인간 단백질과 평균 탄수화물 조성에 있어서 다를 수 있다. 이는 인간 세포가 α2.3 시알릴트렌스퍼라제 및 α2.6 시알릴트렌스퍼라제 효소를 가지고 있다는 사실에 기인한다. WO 89/10404에서는, 마우스 하이브리도마, 마우스 플라즈마사이토마 세포 및 래트 하이브리도마 세포와 같은 골수종 세포를 이들을 GS로 형질전환하여 글루타민-비의존적으로 만드는 방법이 보고되었다. 여기에서는 GS가 면역글로블린 분자의 경쇄 및 중쇄를 코딩하는 유전자의 공증폭 및 골수종세포주의 섬유소용해효소를 코딩하는 유전자의 공증폭에 사용될 수 있다는 것이 추가로 증명되었다. 그러나 설치류 세포주는 불리한 면, 즉 N-아세틸뉴라민산 대신에 N-글리콜릴뉴라민산 잔기의 부착, 설페이션 수행 불능 및 α1.3 갈락토실트렌스퍼라제 효소의 존재를 나타낸다. 설치류 세포에서 합성된 글리코 단백질의 올리고사카리드 구조는 그러므로 인간에서 면역원성일 것으로 기대될 수도 있다.
본 발명의 목적은 단백질의 생산, 특히 높은 단백질 역가(titre)를 나타내는 글리코단백질의 생산에 있어서 상기 언급된 불리한 점을 갖지 않는 개선된 방법을 제공하는 것이다.
상기 목적은 청구항 1에 따른 신규한 글루타민-요구성 인간세포 및 청구항 7에 따른 신규한 방법으로 달성되었다.
본 발명에 따른 글루타민-요구성 인간세포는 (첫 번째) 단백질을 코딩하는 외인성 DNA 서열 또는 단백질을 코딩하는 내인성 유전자의 발현을 변경할 수 있는 외인성 DNA 서열로 전달이입되었으며, 추가로 (두 번째)글루타민 합성효소(GS), 바람직하게는 포유류 GS를 코딩하는 외인성 DNA 서열로 전달이입되어 제공되며, 여기에서 상기의 외인성 DNA 서열은 하나 이상의 DNA 구조체(construct)상에 위치하며, 상기 전달이입된 세포는 상기 단백질을 생산할 수 있으며 무글루타민 배지에서 성장할 수 있다.
"단백질을 코딩하는 외인성 DNA 서열" 또는 "단백질을 코딩하는 내인성 유전자의 발현을 변경할 수 있는 외인성 DNA 서열" 및 "GS를 코딩하는 외인성 DNA 서열"은 통상적으로 발현벡터 또는 감염성 벡터와 같은 DNA 구조체 상에 위치한다. 발현벡터로서 플라스미드가 사용될 수 있다. 감염성 벡터로는 예를 들면, 레트로바이러스, 헤르페스, 아데노바이러스, 아데노바이러스-연관, 볼거리 및 폴리오바이러스 벡터가 적용될 수 있다. 바람직하게는 발현벡터로, 구체적으로 플라스미드가 사용된다.

일반적으로, "단백질을 코딩하는 내인성 유전자의 발현을 변경할 수 있는 외인성 DNA서열"은 세포로의 전달이입 후에 세포의 염색체 DNA로 편입된다. 상동성 재조합 또는 표적은 본원에서 조절서열과 함께 통상적으로 내인성 유전자와 연관된 조절 부위를 대체 또는 불활성시키기 위해 사용된다. 조절서열로서 예를 들면 WO 93/09222에 기술된 상응하는 비전달이입된 세포에서 분명히 나타난 것 보다 유전자를 더 높은 수준으로 발현되도록 하는 예를 들면, 프로모터 및/또는 인헨서가 기능할 수 있다. 적절한 프로모터는 조절성 또는 상시 발현되는 프로모터일 수 있다. 적절한 프로모터는 사용되는 세포주에 따라서 강한 프로모터, 예를 들면 인간 사이토메갈로바이러스 주요 즉발초기 프로모터(hCMV-MIE), SV40 초기 및 후발 프로모터, 아데노바이러스의 다른 프로모터, 폴리오마 바이러스 또는 파포바-바이러스의 임의의 초기 및 후발 프로모터, 인터페론 α1 프로모터, 마우스 메탈로씨오네인(metallothionein)프로모터, 라우스 사코마 바이러스 긴 말단 반복 프로모터, β-글로빈 프로모터, 콘알부민 프로모터, 오브알부민 프로모터, 마우스 β-글로빈 프로모터 및 인간 β-글로빈 프로모터일 수 있다.
본 발명에 따른 "GS를 코딩하는 외인성 DNA 서열"은 약한 프로모터의 조절하 및 강한 프로모터의 조절하에 있을 수 있다. 만약 외인성 DNA 서열이 단순히 GS를 코딩하는 유전자를 발현하기 위해 필요하다면 강한 프로모터가 사용된다. 만약 외인성 DNA 서열이 선택성 마커로 사용되며, 만약 GS가 증폭을 위해 사용된다면 약한 프로모터가 사용된다. 적합한 프로모터는 조절성 또는 상시 발현되는 프로모터 일 수 있다. 상기 프로모터는 예컨대, 세포 배양에서 글루타민 이화대사산물 암모니아를, 통상적으로 4mM 이하, 바람직하게는 2mM 이하, 더욱 바람직하게는 2mM 미만인, 고수준으로 생산하지 않지만 전달이입된 세포의 성장에는 충분한 농도로 GS가 발현되게 선택될 수 있다.
"선택성 마커"는 수용체 세포를 규명하고 단리하는 것을 가능하게 하는 선택성 표현형을 부여한다. GS는 본 발명에서 GS를 코딩하는 외인성 DNA 서열이 편입되고 이를 발현하는 성공적으로 전달이입된 글루타민-요구성 인간세포를 선택하기 위해 선택성 마커로서 사용될 수 있다.
강한 프로모터는, 사용되는 세포주에 따라서, 예를 들면 hCMV-MIE, SV40 초기 및 후발 프로모터, 아데노바이러스의 다른 프로모터, 폴리오마 바이러스 또는 파포바-바이러스의 임의의 초기 및 후발 프로모터, 인터페론 α1프로모터, 마우스 메탈로씨오네인 프로모터, 라우스 사코마 바이러스 긴 말단 반복 프로모터, β-글로빈 프로모터, 콘알부민 프로모터, 오브알부민 프로모터, 마우스 β-글로빈 프로모터 및 인간 β-글로빈 프로모터 일 수 있다.
약한 프로모터는, 사용되는 세포주에 따라서, 예를 들면 뮤린 류케미아 바이러스 긴말단 반복, 헤르페스 심플렉스 바이러스 티미딘 키나제 및 마우스 멤머리 종양 바이러스-긴말단 반복일 수 있다. 바람직하게 GS를 코딩하는 유전자는 강한 프로모터의 조절하에 있으며, 더욱 바람직하게는 hCMV-MIE 프로모터의 조절하에 있다. 가능한 구현예, 햄스터 유래의 증폭될 수 있는 포유류 GS 서열 및 포유류 세포에서 선택성 마커로서의 이의 용도는 기술분야에 잘 공지되어 있으며 예를 들면 WO 87/004462, WO91/06657 및 WO 89/01036 에 기술된다; 본 발명의 실시예는 이러한 햄스터 GS 발현 단위 및 참조문헌에 개시된 각각의 선택 방법을 채택한다.

글루타민-요구성 인간세포는 GS를 발현하지 않거나 또는 GS를 매우 약하게 발현하는, 따라서 글루타민을 포함하는 배양배지에서는 성장할 수 있지만, 무글루타민 배지에서는 성장할 수 없거나 단지 겨우 성장할 수 있는 모든 인간 세포를 의미한다. 본 발명에 사용되는 글루타민-요구성 인간세포는 유한(mortal) 글루타민-요구성 인간세포 또는 불멸화된 글루타민-요구성 인간세포이다. 유한 글루타민-요구성 인간세포는 배양에서 제한된 수명을 나타내는 글루타민-요구성 인간세포이다. 불멸화된, 또한 "영구적인" 또는 "확립된"으로도 불리는 글루타민-요구성 인간세포는 기술분야의 당업자에게 잘 공지된 대로 올바르게 계대배양 및 서브컬쳐된 경우 배양에서 명백한 무제한의 수명을 나타내는 글루타민-요구성 세포이다.
유한 글루타민-요구성 인간세포의 예는 인간 섬유모세포 및 인간 태아 허파조직세포일 수 있다. 불멸화된 글루타민-요구성 인간세포의 예는 인간 섬유육종 세포, 예컨대 HT1080 세포주 (예를 들면 DSMZ No. ACC-315 또는 ATCC No.CCL 121) 및 B-림프블라스토이드 인간 세포 예컨대 HL60 (DSMZ No. ACC-3)일 수 있다. 불멸화된 글루타민-요구성 인간세포가 본 발명에 바람직하게 사용된다. 추가로 바람직한 이러한 불멸화된 글루타민-요구성 인간세포는 B-림프블라스토이드 세포 또는 섬유육종 세포이며, 더욱 바람직하게는 인간 섬유육종세포, 가장 바람직하게는 HT1080 세포주 (예를 들면 ATCC No.CCL 121)가 사용된다.
글루타민-요구성 인간세포는 공지된 유전공학적 기술에 의해 외인성 DNA 서열로 전달이입될 수 있다.
외인성 DNA 서열로의 전달이입은 서열이 하나 이상의 DNA 구조체 상에 위치하는 지 여부에 좌우된다. 만약 서열이 하나 이상의 DNA 구조체 상에 위치한다면, 전달이입은 각각의 서열로 별개로 하거나 또는 공전달이입으로 한다. 각각의 서열을 별개로 전달이입하는 경우에 서열의 전달이입의 순서는 일반적으로 임의적이다. 각각 서열의 별개의 전달이입은 바람직하게는 먼저, "상기 단백질을 코딩하는 외인성 DNA 서열" 또는 "상기 단백질을 코딩하는 내인성 유전자의 발현을 변경할 수 있는 외인성 DNA 서열"로 하며 두 번째로 "GS를 코딩하는 외인성 DNA 서열"로 한다. 전달이입된 글루타민-요구성 세포는 각각의 별도의 전달이입 후에 배양될 수 있으며 단백질 생산에 대해 평가될 수 있다.
성공적으로 전달이입된 세포를 선택하기 위해, 이들은 무글루타민 배지에서 성장한다. 세포는 직접 무글루타민 배지에서 성장할 수도 있으며 또는 처음에는,단계적으로 무글루타민 배지로 희석될 글루타민을 포함하는 배지에서 성장할 수 있는데, 예를 들면 2mM에서 0mM의 단계로 희석될 수 있는 10mM의 글루타민 농도로 시작할 수 있다. 적절한 선택 절차는 사용되는 세포주에 따라서 선택될 수 있다. 상기 언급된 것으로부터 명백하듯이, 단백질을 생산할 수 있으며 무글루타민 배지에서 성장할 수 있는 본 발명의 글루타민-요구성 인간세포는 상기 세포를 상기 단백질을 코딩하는 외인성 DNA 서열 또는 상기 단백질을 코딩하는 내인성 유전자의 발현을 변경할 수 있는 외인성 DNA 서열 및 글루타민 합성효소를 코딩하는 외인성 DNA 서열로 전달이입하여 수득될 수 있으며, 여기에서 이들 외인성 DNA 서열은 하나 이상의 DNA 구조체 상에 위치한다.
단백질을 코딩하는 외인성 DNA 서열 또는 단백질을 코딩하는 내인성 유전자의 발현을 변경할 수 있는 외인성 DNA 서열은 전달이입 후에 예를 들면 WO 94/12650 에 기술된 유전자 증폭의 공지된 방법에 따라 증폭될 수 있다. 예를 들면 DHFR(디히드로폴레이트 리덕타제), GS, 아데노신 데아미나제, 아스파라긴 합성효소, 아스파르테이트 트렌스카바밀라제, 메탈로씨오네인-1, 오르니틴 데카르복실라제, P-당단백질, 리보뉴클레오티드 리덕타제, 티미딘 키나제 또는 크산틴-구아닌 포스포리보실 트렌스페라제와 같은 효소를 코딩하는 증폭할 수 있는 유전자가 본 목적을 위해 사용될 수 있다. 증폭된 상기 유전자 카피를 포함하는 세포는 예를 들면, 상기 효소의 대사 산물이 결여된 배지 또는 상응하는 선택성 작용제를 포함하는 배지로의 처리를 견딜 수 있다. 상응하는 선택성 작용제는 예를 들면 DHFR의 경우에는 메토트렉세이트(MTX) 및 GS의 경우에는 메티오닌 술폭시민(sulphoximine)(MSX)이다.
본 발명의 전달이입된 글루타민-요구성 인간 세포에 의해 생산되는 단백질은 비글리코실화 및 글리코실화된 단백질이다. 글리코실화 단백질은 하나 이상의 올리고사카리드 사슬을 갖는 단백질을 언급하는 것이다.

기술분야에 공지된 임의의 적합한 배양절차 및 배양기구가 본 발명의 전달이입된 인간세포를 키우는데 사용될 수 있다. 배양배지로서, 무혈청, 무글루타민의 통상적 기본배지 및 약 0.1 % 내지 20%, 바람직하게는 0.5% 내지 15%의 혈청으로 보충된 통상적 무글루타민 기본배지가 사용될 수 있다. 또한, 동물성기원의 단백 질이 없는 통상적 무글루타민 기본배지가 사용될 수 있다. 바람직하게는 무혈청 무글루타민의 통상적 기본배지가 사용된다.
사용될 수 있는 혈청은 예를 들면, 소태아혈청 또는 성체소혈청이다. 바람직하게는 소태아혈청이 사용된다. 사용될 수 있는 통상적 무글루타민 기본배지는 예를 들면 무글루타민 Eagle's 최소기본배지(minimal essential medium)(MEM), 무글루타민 Dulbecco's Modification of Eagle's Medium (DMEM), 무글루타민 Iscove's DMEM 배지 (N. Iscove 및 F. Melchers, Journal of Experimental Methods, 1978, 147, 923), 무글루타민 Ham's F12 배지 (R.G. Ham, Proceedings of National Academy of Science, 1965, 53, 288), 무글루타민 L-15 배지 (A. Leibovitz, American Journal of Hygiene, 1963, 78, 173), 무글루타민 RPMI 1640 배지 (G.E. Morre 등, The Journal of the American Medical Association, 1967, 199, 519), 무글루타민 시판(proprietary) 배지 및 이들의 적합한 비의 혼합물이다. 고밀도 세포배양을 위한 통상적 세포배양 성장 배지의 강화는 기술분야에서 잘 공지되어 있으며 예를 들면 GB 2251249에 기술된다. 이는 본 발명의 무글루타민배지에도 또한 잘 적용된다.
통상적 보충제가 통상적 무글루타민 기본배지에 첨가될 수 있다. 일반적으로 첨가될 수 있는 보충제는 일반적으로 혈청에 존재하는 단백질을 포함하며 그리고 세포 성장 및/또는 세포 생활도에 긍적적 영향을 미치는 추가의 성분을 임의적으로 포함한다. 혈청에 일반적으로 존재하는 단백질은 예를 들면 소혈청알부민(BSA), 트렌스퍼린 및/또는 인슐린이다. 세포 성장 및/또는 세포 생활도에 긍적 적 영향을 미치는 추가의 성분은 예를 들면 대두지질, 셀레늄 및 에타놀아민이다. 글루타민 및/또는 뉴클레오시드를 대체하는 아미노산은 세포주에 따라 배양배지에 추가될 수 있다. 아미노산의 예는 이소루신, 루신, 발린, 리신, 아스파라긴, 아스파르트산, 글루탐산, 세린, 알라닌이다. 임의적으로 글루타민은 통상적 무글루타민 기본배지에 일반적으로 1mg/ℓ 미만, 바람직하게는 0.5mg/ℓ 미만의 낮은 농도로 이의 생합성기능(예를 들면 트렌스아미네이션 반응)을 지지하기 위해 첨가될 수 있다.
단백질을 코딩하는 외인성 DNA 서열 또는 단백질을 코딩하는 내인성 유전자의 발현을 변경할 수 있는 외인성 서열이 증폭할 수 있는 유전자를 사용하여 전달이입된 후에 증폭된다면, 상응하는 선택성 작용제를 통상적 무글루타민 기본배지에 첨가할 수 있다. 적용된 선택성 작용제의 농도범위는 사용되는 세포주에 의존한다. 일반적으로, 10μM 이상의 농도가 사용된다.
전달이입된 세포가 단백질을 생산할 수 있으며, 나아가 본 발명에 따른 무글루타민 배지에서 성장할 수 있는 본 발명에 따른 전달이입된 글루타민-요구성 인간세포를 수득하기 위한 개시물질로서 사용될 수 있는 글루타민-요구성 인간세포는 부착의존성 또는 부착비의존성일 수 있다. 만약 부착의존성 인간세포, 예를 들면 HT1080 세포주(ATCC No. CCL 121)가 사용된다면, 아직까지 문헌에 기술된 적이 없는 무혈청배지 중에서 부유상태로 성장할 수 있는 부착비의존성 HT1080 세포주로 적응될 수 있다.
적응은 단백질을 코딩하는 외인성 DNA 서열 또는 단백질을 코딩하는 내인성 유전자의 발현을 변경할 수 있는 외인성 DNA 서열 및 GS를 코딩하는 외인성 DNA 서열로의 전달이입 전 또는 후에 일어날 수 있다. 바람직하게는, 상기 세포는 먼저 단백질을 코딩하는 외인성 DNA 서열 또는 단백질을 코딩하는 내인성 유전자의 발현을 변경할 수 있는 외인성 DNA 서열로 전달이입되고, 그리고 두 번째로 무혈청배지 중에서 부유상태로의 성장에 적응되며 이어서 GS를 코딩하는 외인성 DNA 서열로 추가로 전달이입된다. 만약 필요하다면 전달이입된 세포는 다시 무혈청, 무글루타민 배지 중에서 부유상태로의 성장에 다시 적응될 수 있다.
본 발명의 전달이입된 글루타민-요구성 인간세포는 부착의존성 또는 부착비의존성일 수 있으며 무혈청, 무글루타민배지 중에서 부유상태로 성장할 수 있다. 바람직한 전달이입된 글루타민-요구성 인간세포는 부착비의존성이며 무혈청, 무글루타민배지 중에서 부유상태로 성장할 수 있다.
무혈청 배지 중에서 부유상태로 성장할 수 있는 부착비의존성 세포가 되기위한 적응은 세포를 첫 번째 단계로 혈청 포함 배지를 사용하여 부착비의존성으로 적응시킴으로서 달성될 수 있다. 이는 예를 들면, 세포의 트립신처리 및 후속의 아지테이션 또는 아지테이션에 의한 세포의 방출에 의해 달성될 수 있다. 상기 세포는 이어서 두 번째 단계에서 후속적 혈청함량의 감소에 의해 무혈청배지에 적응된다. 적응동안 만약 사용된다면 선택성 작용제의 양은 세포의 성장을 억제하는 경우에는 감축될 수 있다. 그러나, 세포는 첫 번째 단계에서 후속적 혈청함량의 감소에 의해 무혈청배지 중에 성장하도록 적응되고 두 번째 단계에서 예를 들면 세포의 트립신처리 및 후속의 아지테이션(agitation) 또는 아지테이션에 의한 세포의 방출에 의해 부유상태에서 성장할 수 있는 부착비의존성 세포로 적응될 수도 있다. 양단계는 동시에 적용되는 것이 좋을 것이다.
바람직하게 세포는 첫 번째 단계에서 혈청을 포함하는 배지를 사용하여 아지테이션에 의해 세포를 방출시키는 것에 의해 부착비의존성 세포로 적응되며 이어서 두 번째 단계에서 후속적인 혈청 함량의 감소에 의해 무혈청배지로 세포를 적응시킨다.
혈청을 포함하는 배지의 기본으로서, 상기 기술된 배양배지가 사용될 수 있다. 본원에서 정의된 선택성 작용제가 배양배지에 추가될 수 있다. 선택성 작용제의 적용된 농도 범위는 사용된 세포주에 의존한다. 통상적으로 10μM 이상의 농도가 사용된다. 혈청을 포함하는 배지는 통상적으로 약 0.1 내지 20% 바람직하게는 0.2 내지 10% 가장 바람직하게는 0.5 내지 5% 혈청으로 보충된다. 사용될 수 있는 혈청은 상기 언급된 것들이다. 혈청 함량의 후속적 감소는 혈청 함량을 단계적으로 예를 들면 10% 부터 1%로, 0%로 감소하는 것에 의해 달성될 수 있다.
본 발명의 전달이입된 글루타민-요구성 인간세포는 상기 단백질의 발현 및 상기 단백질의 회수에 적합한 조건하에서 배양 배지 중에서 상기 세포를 배양함으로서 단백질 생산을 위한 방법에 사용된다. 생산된 단백질은 상기 기술한 바와 같다. 배양배지로서 상기 기술된 통상적 무글루타민 기본 배지 및 통상적 보충제가 사용될 수 있다. 적합한 배양배지는 예를 들면 WO 96/39488에 기재된 포유류세포의 생체외 배양을 위해 통상적으로 사용된 것들이다.

인간 EPO 유전자 카피를 다수 포함하는 부착의존성 인간 HT1080-R223 세포주는 EPO의 공업적인 생산에 사용되는 세포주이며 최초로 Transkaryotic Therapies, Inc. Cambridge, MA 02139(US)에서 만들어졌다. 이는 부착의존성 인간 섬유육종 세포 HT1080 세포주로부터 유래된다. 모 HT1080 세포주(ATTC No. CCL 121)는 DNA 구조체 pREPO22로 전달이입에 의해 EPO를 생산하는 능력을 갖게되었는데, 상기 구조체는 WO 95/31 560에 기재된 DNA 구조체 pREPO18과 DHFR 유전자가 반대 방향으로 있으며 pREPO22는 pREPO18 보다 대략 600 염기쌍의 덜 상동적인 서열을 포함하는 점을 제외하고는 유사하다.

무혈청배지
중에서 부유 성장으로의 적응
정적인 플라스크에서 부착 배양으로 자란 세포를 트립신을 처리하여 방출하 고 10% 투석된 소태아혈청(dFBS) 및 500nM MTX로 보충되고 나아가 "HM9"으로 언급되는 시판되는 글루타민을 포함하는 무혈청배지로 재현탁하고 교반 플라스크 배양으로 인큐베이션하였다. 6일 후에 성장이 개시되었고 세포를 동일한 배지로 서브컬쳐하였다(도1). 일단 신뢰할 수 있는 성장 패턴이 확립되면 배지의 혈청 함량을 1%로 감소시켰다. 혈청보충을 완전히 제거하기 전에 다시 신뢰할 수 있는 성장이 재확립되도록 하였다.
혈청-보충 및 무혈청 배양은 생산성을 평가하기 위해 과성장되었다. 결과는 부착 배양물로부터의 데이터와 함께 표1에 나타낸다. 적합한 적응 후에 부유배양 중의 특이적 성장속도는, 혈청이 없는 경우에도, 혈청-보충된 부착 배양의 특이적 성장속도와 동등하였다.
혈청 보충된 배지 중에서 부착 배양에서 부유배양으로 적응하면, EPO 합성의 특이적 속도가 24에서 12 EU/106세포/h으로 50% 감소되었다. 그러나 상기 속도는 무혈청 성장으로 적응 후에 18 EU/106세포/h으로 상당히 회복되었다.

중에서 부유 성장으로의 적응
HT1080 세포주 ATCC CCL121은 American Type Culture Collection (Rockville, Maryland, USA)에서 입수하였다. 초기에 세포는 10% 소태아혈청(FBS)를 함유하는 DMEM 중에서 부착 배양으로 성장되었다.
부유 및 무혈청 적응의 단계는 도 2에 도식적으로 나타낸 것에 따라 수행될 수 있다. 부유배양을 개시하기 위해, 세포는 트립신처리하여 부착 배양으로부터 방출되었고 방출된 세포는 HM9 플러스 2% FBS로 재현탁되었다. 이들은 이어서 교반배양으로 인큐베이션되었다. HM9 플러스 2% FBS 중에서 세포의 부유성장이 일단 확립되었으면 배양을 동일한 배지로 희석하여 딸 배양을 만들었다. HT1080 세포주의 신뢰할 만한 부유성장이 배양에서 30일 후에 확립되었다 (도3). 이후 배지의 혈청 함량을 감소하여, 마지막에는 완전히 제거하였다. 상기 세포는 혈청이 없는 상태에서 일련의 서브컬쳐에서 계속 성장하였다.

4. 암모니아의
R223
세포주의 성장 및 생산성에 대한 영향
암모니아는 글루타민이 에너지 기질로서 사용되는 경우 배양되는 세포에서 생산되는 이화대사산물이다. 이는 세포독성이 있으며 성장을 방해할 수 있다. 나아가, 이는 세포의 골지체내의 pH에 대한 영향으로 인해 단백질의 글리코실화를 억제할 수 있다. 플라스크 배양 중의 R223세포는 전형적으로 무영양공급 배양에서 5mM의 암모니아를 생산하여 영양공급을 받은 발효기 배양에서는 10mM을 생산한다.
암모니아 영향에 대한 초기 검사에서, R223세포는 첨가된 암모니아 없이, 또는 2, 5 또는 10mM 로 암모니아가 첨가된 교반 플라스크에서 성장되었다. 각각의 암모니아 농도에 대해, 오버레이 가스의 CO2 함량을 변화시킴으로서 세개의 상이한 pH 값으로 복제 배양이 만들어졌다. 이의 주목적은 암모니아에 의해 발휘되는 성장억제의 정도를 결정하는 것 및 감소된 pH 가 이러한 성장억제를 극복할 것인지를 시험하는 것이었다.
암모니아는 세포성장을 억제(표2)하는 것으로 밝혀진 반면, 비록 pH를 감소시키기 위해 요구되는 상승된 pCO2 자체가 어느 정도 성장억제를 야기했을 수는 있지만, 감소된 pH는 암모니아의 억제효과를 경감시키는 것에는 실패하였다.
배양은 단지 3일 후에 종결되었다. 더 이상의 계속하는 것은 비휘발성 산 이화대사산물의 축적이 모든 배양의 pH를 수 포인트의 pH 단위로 떨어뜨리도록 하기 때문에 타당하지 않았다.
후속적 실험(표3)에서 오버레이 가스의 CO2 함량은 비억제 농도로 유지하면서 배지의 NaHCO3 함량을 조정하는 것에 의해 pH를 변화하였다. 시험된 pH의 범위는 pH 7.0 내지 7.5 이었다 (플라스크 배양에서는 배지 pH는 조절될 수 없으며 초기 배양 이틀만 지나도 pH가 상당히 떨어진다는 것이 강조되어야만 한다. 언급된 pH값은 각 배양의 초기 pH이다). 특이적 성장속도는 pH7.5(3g/ℓ의 NaHCO3 )에서 최대이었으나, 이 pH에서는 10mM 암모니아의 존재 중에서 성장 속도 및 최대 세포농도는 모두 반이 되었지만, EPO의 특이적 합성속도는 4배 감소하였다(표3).
pH7.5에서의 배양과 비교하여, pH7.25(1.5g/ℓ의 NaHCO3 )에서 특이적 성장속도는 감소되었으며, EPO의 특이적 합성속도는 증가하였다. 그러나 10mM 암모니아의 존재에 의해서, 성장속도는 덜 영향을 받았으며, EPO의 특이적 합성속도는 영향을 받지 않았다.
연구한 가장 낮은 pH인 pH7.0(0.75g/ℓ의 NaHCO3 )에서, 특이적 성장속도는 더욱 감소되었으나, 역시 pH7.5에서 보다는 암모니아에 의해 덜 영향을 받았다. EPO의 특이적 합성속도는 추가된 암모니아의 존재 중에서 증가되었다(이는 성장속도의 감소에 기인한 것일 수 있다).

5. 5리터 스케일의 발효기 배양에서
R223
세포주의 생산성
R223 세포주에 대해, 6.95 내지 7.15 사이의 상이한 pH 값으로 조절되는 세개의 5리터 발효가 수행되었다 (표4 참조). 각각의 배양은 주로 소비되는 영양분을 충분한 농도로 유지하기 위해 고안된 아미노산 및 글루코스를 포함하는 농축된 영양분을 받았다. 배양물은 pH 7.15 및 pH 7.05 에서는 잘 자랐으나, pH6.95에서의 배양물은 성장을 하지 않았는데, 이는 아마도 이 pH를 조절하기 위해 요구되는 높은 CO2의 농도에 기인한 것으로 추측된다.
실시예 6. 부착배양에서 R223의 GS로의 전달이입 및 GS 전달이입체의 생산성.
전달이입에 사용된 세포는 실시예 2의 부유-적응된 무혈청 스톡의 R223 세포주에서 유래하였다. 이들 세포가 큰 다세포 무리(aggregates)로 자라면, 이들을 트립신 처리하여 실질적으로 단세포 부유물로 감소시켰다.
전달이입을 위해 실시예 2에서 수득된 단일-부유 적응된 R223 세포주 (칼슘 또는 마그네슘이 없는 인산 완충용액 중)의 대략 107개 세포의 분취물(aliquot)을 GS유전자(Bebbington 등, 1992, Biotechnology 10, 169-175에 기재된 DNA 서열 pCMGS BamH1) 를 포함하는 20㎍의 선형화된 DNA와 혼합하고 Biorad Gene Pulser (450 볼트, 250㎌)을 이용하여 전기천공(electroporation)하였다. 대조군으로서, 동등한 세포의 분취물을 DNA 없이 전기천공하였다. 세포를 0 또는 10% dFBS를 포함하는 MTX가 없는 HM9 배지로 희석하여 96-웰 배양 플레이트 또는 25cm2 플라스크로 분배하고 35.5 내지 37℃에서 배양하였다. HM9 배지는 초기에는 2mM 글루타민을 포함하였지만 이는 하루 후 0.5mM로 희석되었으며, 이어서 10일 후에 무글루타민 HM9배지로 대체되었다. 또한 1일째 또는 10일째에, MTX(500nM)를 배양에 재도입하였다.
첨가된 DNA에 없이 전기천공된 대조군 세포로 셋업된 배양의 경우는, 성장이 없었다.
DNA로 전기천공된 세포의 경우는, 6개의 96웰 플레이트에서 15개의 GS 전달이입체가 규명되었다. GS전달이입체는 초기 배지 중에 MTX 존재 및 부재 모두의 경우에 수득되었다. 15개의 GS전달이입체 중에, 7개를 성공적으로 플라스크 배양으로 확장하였다 (표5). 나머지 8개의 GS전달이입체는 비정상 세포형태를 나타내며, 성장이 불량하여 포기하였다.
단리된 각각의 초기 7개의 GS 전달이입체에 대해, 10% 혈청 보충된 무글루타민 HM9 배지를 사용하여 복제 정적 플라스크 배양세트를 셋업하였다. 1 내지 4일 간격으로, 배양을 중단하여 세포수를 세고 EPO-ELISA에 의해 EPO의 농도를 측정하였다. 이들 데이터의 집합으로부터, 각각의 GS 전달이입체에 대한 EPO의 특이적 합성속도를 측정할 수 있었다. 데이터는 표5에 요약하였으며, 비교를 위해, 비전달이입된 R223 세포주에 대한 데이터를 포함하였다. 모든 GS전달이입체가 비전달이입된 세포주와 비교하여 상승된 EPO의 특이적 합성속도를 나타내었다. 가장 좋은 GS전달이입체인 3E10은 비전달이입된 R223세포주보다 5배 내지 6배 더높은 합성속도를 가졌다.
데이터는 10% dFBS의 존재 중에 성장한 부착 배양물로부터 유래된 것이다. 메토트렉세이트는 전달이입체를 만드는데 사용된 배지로부터 빠졌으나, 전달이입 후 1일 또는 10일째에 재도입되었다.

실시예 6에서 수득된 7개의 전달이입체 중에서, GS 전달이입체 #3E10 및 GS 전달이입체 #8G3을 부유배양으로 진행시켰다.
세포를 35.5 내지 37℃에서 10% dFBS 및 500nM MTX로 보충된 무글루타민 HM9 배지로 정적 플라스크에서 부착 배양으로 키웠다. 상기 세포는 5 또는 6일 후에 아지테이션으로 방출하여, 동일한 배지에 재현탁하고, 동일한 온도에서 교반 플라스크 배양으로 인큐베이션하였다. 성장은 단지 2 또는 3일 후에 개시되었다. 세포를 동일한 배지에서 한번 서브컬쳐하였고 이어서 생산성을 평가하기 위해 과성장 시켰다. 역가 및 생산물 합성속도는 10% dFBS 및 500nM MTX에서 자란 비전달이입된 R223 세포주의 역가 및 합성속도 보다 2배 이상 더 높았다(표6).

9. 생산물 품질 분석
플라스크 배양에서 GS 전달이입체 3E10에 의해 생산된 EPO는 면역정제되어 글리코형태의 분포에 대해 분석되었다. 도4는 피크 세포농도에서 수득된 EPO 및 실시예 8에 기술된 무글루타민 HM9 배지에서 자란 3E10 세포 배양 수확물의 등전점초점(IEF) 겔 분석을 보여준다. HM9 배지에서 자란 비전달이입된 R223 세포주 유래의 비교 시료도 포함된다. GS 전달이입체 3E10에서 생산된 EPO는 비전달이입된 R223세포주에 비교하여 더 산성인 이소형태의 강화를 나타내었다. IEF 겔을 스캐닝하여 이소형태 분포의 정량 및 이론적 이소형태 상대적 활성도(IRA)의 계산을 하였다.
상기 데이터는, 비전달이입된 R223 세포주의 대조군 배양에 대한 것 보다 거의 두배 더 높은 IRA를 갖는, GS 전달이입체 3E10에서 생산물 품질의 상당한 향상을 보여주고 있다(표9 및 표10).
또한 가정적 N-글리칸 전하 "Z"가 GS 전달이입체 3E10 유래의 EPO에 대해 결정되었다. "Z"는 표10에 있는 비전달이입된 R223 세포주에 대해서는 결정되지 않았다. 그럼에도 불구하고, GS 전달이입체 3E10에 대한 "Z" 값(피크 세포농도에서 273 및 수확시 265)은 비전달이입된 R223 세포주의 플라스크 배양(183-228)에서 수득된 값을 초과하였다. "Z"는 Hermentin 등, Glycobiology, 1996, 6, 217-230에 따라 결정되었다; "Z"는 특정 시알릴화된 이소형태 각각의 %-쉐어(share)를 아시알로(asialo)/중성(neutral), 모노시알로, 트리-, 테트라 또는 펜타시알로인지 여부에 의존하는 상기 이소형태의 상응하는 음전하를 곱하여 수득된다. 상기 생산물 항(term)의 수학적 합계가 Z이다. 상기 Z 값은 주어진 치료용 글리코단백질의 생체내 제거속도와 연관된다(Hermentin, 상기문헌).

실시예 10. 6리터 발효기에서 무혈청 부유 배양 중의 GS 전달이입체 3E10의 생산성, 대사 및 생산물 품질
GS 전달이입체 3E10을 에어리프트 발효기에서 회분 배양으로 키웠다. 배양 배지는 혈청이 없는 무글루타민 HM9이었다. 각각의 배양은 주로 소비되는 영양분을 충분한 농도로 유지하도록 고안된 아미노산 및 글루코스를 포함하는 농축된 영양공급을 받았다. 결과는 표11에 있으며 비전달이입된 모세포 R223의 발효에 대한 데이터가 비교를 위해 포함된다. GS 전달이입체는 증가된 생활도, 즉 배양 기간의 증가를 나타내었다. 생산물 합성의 특이적 속도는 변화되지 않은 비전달이입된 모세포주의 것과 동등하였으나, 더 긴 배양수명으로 인해 최대 산물의 농도가 증가되었다. 암모니아 축적은 GS 전달이입체의 경우 적어도 4배 더 낮았다. RIA에 의해 측정된 생산물 품질은 GS전달이입체의 경우 향상되었다.
실시예 11. Iscove's 기재 배지에서 무혈청 부유배양 중의 GS 전달이입체의 생산성 및 대사.
실시예 7의 무혈청 성장에 적응된 GS 전달이입체 3E10을 무혈청, 무글루타민 버젼의 Iscove's 배지로 교반 플라스크에서 배양하였다. 모세포주, R223을 글루타민으로 보충된 동등한 배지에 병행으로 키웠다.
사용된 배지는 Iscove's 보충제 (0.4g/L의 소 혈청 알부민, 30mg/L의 인간 홀로-트렌스페린), 10mg/L의 재조합 인간 인슐린, 1g/L의 루트롤 F68 및 60㎕/L의 에탄올아민이 들어 있는 Iscove's Modified Dulbecco 배지였다. 상기 배지는 세포주 R223의 배양을 위해 4mM 글루타민을 포함하지만, GS 전달이입된 세포주의 경우에는 글루타민은 빼고 4mM 소디움 글루타메이트 플러스 4mM 아스파라긴으로 대체되었다.
생산물 합성의 특이적 속도는 GS 전달이입된 세포주인 3E10이 비전달이입된 모세포주 R223 보다 대략 50% 더 높았다 (표12). 암모니아 생산의 특이적 속도는 전달이입된 세포주 3E10의 경우가 7배 더 낮았다(표13).
생산물은 이들 각각의 배양물로부터 순수 정제되고 등전점초점(IEF)겔로 분석되었다. 상기 분석의 결과는 도5에 있으며, 이는 염색 후의 IEF 겔을 보여준다. pH2.5 내지 6.5 IEF 겔은 변성조건에서 전기영동되었으며 쿠마시 블루로 염색되었다.

R223 유래의 생산물의 경우, 겔의 길이에 걸쳐 퍼져있는 13개 이상의 가시적 밴드가 있다. GS 전달이입된 세포주 3E10에 의해 생산된 생산물의 경우, 더 염기성인 밴드(겔의 윗부분에서 분리된)는 세포주 R223 유래의 생산물에 대한 것 보다 강도가 훨씬 덜 했으며, 반면 더 산성인 밴드(겔의 아랫부분)는 세포주 3E10에 대하여 강도가 증가한다. 모세포주 R223 유래의 생산물에서는 검출할 수 없는, GS-전달이입체 3E10을 사용하여 만든 생산물에서 검출할 수 있는 하나 이상의 추가의 산성밴드가 또한 있다. 이는 GS 전달이입된 세포주 3E10으로부터 만들어진 생산물에 대한 시알릴화의 증가도를 나타낸다.

Claims (12)
- 글루타민 합성효소를 코딩하는 외인성 DNA 서열로 글루타민 요구성 인간 세포를 전달이입시키는 단계를 포함하는, 글리코실화 단백질을 생산하는 글루타민 요구성 인간 세포를 개질시키는 방법으로서, 상기 개질이 상기 단백질의 시알릴화 증가, 상기 단백질의 생산율 증가, 상기 세포의 생존력의 연장 및 이들의 조합으로 이루어지는 군에서 선택되는 것인 방법.
- 제 1 항에 있어서, 상기 방법이 상기 글리코실화 단백질을 상기 전달이입 세포의 배양으로부터 회수하는 단계를 더 포함하는 방법.
- 삭제
- 제 1 항 또는 제 2 항에 있어서, 상기 개질이 상기 단백질의 시알릴화의 증가를 포함하는 것인 방법.
- 제 1 항 또는 제 2 항에 있어서, 상기 개질이 상기 단백질의 생산율 증가를 포함하는 것인 방법.
- 제 1 항 또는 제 2 항에 있어서, 상기 개질이 상기 세포의 생존력의 연장을 포함하는 것인 방법.
- 제 1 항 또는 제 2 항에 있어서, 상기 개질이 상기 단백질의 시알릴화 증가,상기 단백질의 생산율 증가 및 상기 세포의 생존력의 연장을 포함하는 것인 방법.
- 제 1 항 또는 제 2 항에 있어서, 상기 전달이입된 세포가 부착비의존성이고, 무혈청, 무글루타민 배지 중에서 부유상태로 성장할 수 있는 것인 방법.
- 제 1 항 또는 제 2 항에 있어서, 상기 글루타민 요구성 인간 세포가 인간 섬유육종 세포인 방법.
- 제 9 항에 있어서, 상기 인간 섬유육종 세포가 HT1080 세포주인 방법.
- 제 1 항 또는 제 2 항에 있어서, 상기 글리코실화 단백질이 에리쓰로포이에틴(erythropoietin) 인 방법.
- 제 11 항에 있어서, 상기 에리쓰로포이에틴이 인간 에리쓰로포이에틴인 방법.
