Abstract

Description
本発明は、一般に、新脈管形成インヒビターを含む融合タンパク質を作製および使用するための方法および組成物に関する。より詳細には、本発明は、免疫グロブリンFc領域および新脈管形成インヒビターを含む、イムノフュージン(immunofusin)と呼ばれる融合タンパク質を作製および使用するための方法および組成物に関する。

本発明は、商業的量の臨床グレードの新脈管形成インヒビターの生成に有用であった、本明細書中でイムノフュージンと呼ばれる、融合タンパク質を提供する。この新脈管形成インヒビターは、イムノフュージンタンパク質構築物から使用前に切断され得る。しかし、イムノフュージンまたはこのイムノフュージンをコードする核酸が、新脈管形成インヒビターで処置する必要がある哺乳動物に直接投与され得ることが意図される。
(実施例1.huFc−huエンドスタチンの発現)
ヒトエンドスタチンを、Loら(1998)(Protein Engineering 11:495)の教示に従って、ヒトFc−ヒトエンドスタチン(huFc−huEndo)融合タンパク質として発現させた。Fcは、ヒト免疫グロブリンγのFcフラグメントをいう(配列番号1に示されるDNA配列;配列番号2に示されるアミノ酸配列)。(ポリメラーゼ連鎖反応PCR)を使用して、Fc−Endo融合タンパク質における発現に、エンドスタチンcDNA(配列番号3;このアミノ酸配列は、配列番号4に開示される)を合わせた。正方向プライマーは、5’−CC CCG GGT AAA CAC AGC CAC CGC GAC TTC C(配列番号5;配列番号6に開示されるアミノ酸をコードした)、または5’−C AAG CTT CAC AGC CAC CGC GAC TTC C(配列番号7;コードされるアミノ酸を配列番号8に開示する)のいずれかであって、ここでXmaI部位またはHindIII部位にはエンドスタチンのN末端をコードする配列が続いた。XmaI部位を有するプライマーは、IgG Fc領域のCH3ドメインの末端でのXmaI部位への連結に、エンドスタチンcDNAを合わせた。HindIII部位を有するプライマーは、融合タンパク質の連結部にエンテロキナーゼ認識部位であるAsp4−Lys(La Vallieら(1993)J.Biol.Chem.268:23311−23317)を含むpdCs−Fc(D4K)ベクターのHindIII部位への連結に、エンドスタチンcDNAを合わせた。逆方向プライマーは、5’−C CTC GAG CTA CTT GGA GGC AGT CAT G(配列番号9)(これは、エンドスタチンのC末端の直後に翻訳終止コドン(アンチコドン、CTA)を置くように設計された)であり、これにはXhoI部位が続く。PCR産物をクローン化および配列決定し、そして得られたXmaIおよびXhoI消化pdCs−FcベクターにXmaI−XhoIフラグメントを連結させた。同様に、エンドスタチンをコードするHindIII−XhoIフラグメントを、適切に消化したpdCs−huFc(D4K)ベクター内に連結した。Fc−endoまたはFc(D4K)−エンドスタチンを発現する安定なクローンを、NS/0細胞のエレクトロポレーション、次いで、100nMメトトレキサートを含有する増殖培地における選択によって得た。タンパク質発現レベルを、抗ヒトFcELISA(実施例3)によってアッセイし、SDS−PAGEによって確認した。SDS−PAGEによって、約52kDのタンパク質産物が示された。最良の産生クローンを、限界希釈によってサブクローニングした。
一過性トランスフェクションについて、リン酸カルシウムとプラスミドDNAとの沈降によって(Sambrookら(1989)Molecular Cloning−A Laboratory Manual、Cold Spring Harbor、NY)か、またはLipofectAMINE Plus(Life Technologies、Gaithersburg、MD)を製造業者の指示に従って用いたリポフェクションによって、プラスミドをヒト腎臓293細胞内に導入した。
3つの異なるELISAを使用して、MTX耐性クローンおよび他の試験サンプルの上清におけるタンパク質産物の濃度を決定した。抗ヒトFc(huFc)ELISAを使用して、ヒトFc含有タンパク質の量を測定した。抗マウスFc(muFc)抗体および抗イヌ(caFc)抗体をELISAで使用して、それぞれ、マウスFc含有タンパク質およびイヌFc含有タンパク質の量を測定した。抗huFc ELISAについての手順は、本明細書中で以下に詳細に記載される。
ELISAプレートを、AffiniPure ヤギ抗ヒトIgG(H+L)(Jackson ImmunoResearch Laboratories、West Grove、PA)(PBS中で5μg/ml)で、96ウェルプレート(Nunc−Immuno Plate MaxiSorpTM、Nalge Nunc International、Rochester、NY)において100μl/ウェルにてコーティングした。コーティングされたプレートを被覆し、そして4℃で一晩インキュベートした。次いで、プレートをPBS中の0.05%Tween20で4回洗浄し、そしてPBS中の1%BSA/1%ヤギ血清(200μl/ウェル)でブロックした。37℃で2時間のブロッキング緩衝液とのインキュベーションの後、プレートをPBS中の0.05%Tweenで4回洗浄し、そしてペーパータオルで軽く叩いて乾かした。
試験サンプルを、PBS中の1%BSA/1%ヤギ血清/0.05%Tweenを含有するサンプル緩衝液において適切な濃度に希釈した。濃度が既知であったキメラ抗体(ヒトFcを有する)を用いて、検量線を作成した。検量線を作成するために、サンプル緩衝液中に連続希釈を作製して、125ng/ml〜3.9ng/mlの範囲にわたる検量線を得た。希釈サンプルおよび基準物をプレートに添加し(100μl/ウェル)、次いで、このプレートを37℃で2時間インキュベートした。インキュベーション後、プレートをPBS中の0.05% Tweenで8回洗浄した。次いで、各ウェルに、サンプル緩衝液中で約1:120,000で希釈された100μlの2次抗体である西洋ワサビペルオキシダーゼ(HRP)結合体化抗ヒトIgG(Jackson ImmunoResearch Laboratories、Inc.West Grove、PA)を添加した。2次抗体の正確な希釈は、HRP結合体化抗ヒトIgGの各ロットについて決定されなければならない。37℃での2時間のインキュベーションの後、このプレートをPBS中の0.05% Tweenで8回洗浄した。
基質溶液を、0.03%の新たに添加されたH2O2を含有する15mlの0.025Mクエン酸/0.05M Na2HPO4緩衝液(pH5)中に、30mg(1錠)のo−フェニレンジアミンジヒドロクロリド(OPD)を溶解することによって調製した。この基質溶液を、100μl/ウェルでプレートに添加した。色は、暗黒下にて室温で、30分間で呈色が可能であった。呈色時間は、コーティングされたプレート、2次抗体などのロット毎の変動性に依存して、変更され得る。この反応を、100μl/ウェルで4NのH2SO4を添加することによって停止させた。このプレートを、プレートリーダー(これを、490nmおよび650nmの両方に設定し、そして490nmでのODから650nmでのバックグラウンドODを差し引くようにプログラムした)で読み取った。
ヒトアンギオスタチン(配列番号10に示されるDNA配列;配列番号11に示されるアミノ酸配列)を、本質的に実施例1に記載されるように、ヒトFc−ヒトアンギオスタチン(huFc−huAngio)融合タンパク質として発現させた。pdCs−huFcベクターまたはpdCs−huFc(D4K)ベクターにおける発現にアンギオスタチンcDNA(配列番号3)を合わせるために、PCRを使用した。それぞれの正方向プライマーは、5’−CC CCG GGT AAG AAA GTG TAT CTC TCA GAG(配列番号12;配列番号13に開示されるアミノ酸をコードした)、および5’−C CCC AAG CTT AAA GTG TAT CTC TCA GAG(配列番号14;配列番号15に開示されるアミノ酸をコードした)であり、ここでXmaI部位またはHindIII部位の後には、アンギオスタチンのN末端をコードする配列が続いた。逆方向プライマーは、アンギオスタチンのC末端の直後に翻訳終止コドン(アンチコドン、CTA)を配置するように設計された、5’−CCC CTC GAG CTA CGC TTC TGT TCC TGA GCA(配列番号16)であり、そしてこれには、XhoI部位が続いた。このPCR産物をクローニングし、そして配列決定し、そしてアンギオスタチンをコードする得られたXmaI−XhoIフラグメントおよびHindIII−XhoIフラグメントを、それぞれ、pdCs−huFcベクターおよびpdCs−huFc(D4K)ベクターに連結した。huFc−huAngioおよびhuFc(D4K)−huAngioを発現している安定なNS/0クローンを、実施例2および3に記載されるように選択し、そしてアッセイした。
マウスエンドスタチン(配列番号17に示されるDNA配列;配列番号18に示されるアミノ酸配列)およびマウスFc(配列番号19に示されるDNA配列;配列番号20に示されるアミノ酸をコードした)を、本質的に実施例1に記載されるように、マウスFc−マウスエンドスタチン(muFc−muEndo)融合タンパク質として発現させた。pdCs−muFc(D4K)ベクターにおける発現めにエンドスタチンcDNA(配列番号4)を合わせるために、PCRを使用した。正方向のプライマーは、5’−C CCC AAG CTT CAT ACT CAT CAG GAC TTT C(配列番号21;配列番号22に開示されるアミノ酸をコードした)であり、ここでHindIII部位には、エンドスタチンのN末端をコードする配列が続いた。逆方向プライマーは、エンドスタチンのC末端の直後に翻訳終止コドン(アンチコドン、CTA)を配置するように設計された5’−CCC CTC GAG CTA TTT GGA GAA AGA GGT C(配列番号23)であり、そしてこれには、XhoI部位が続いた。このPCR産物をクローニングし、そして配列決定し、そしてエンドスタチンをコードする得られたHindIII−XhoIフラグメントを、pdCs−muFc(D4K)ベクター中に連結した。muFc(D4K)−muEndoを発現している安定なNS/0クローンを、実施例2および3に記載されるように選択し、そしてアッセイ(抗muFc ELISA)した。
マウスアンギオスタチン(配列番号24に示されるDNA配列;配列番号25に示されるアミノ酸配列)を、本質的に実施例1に記載されるように、マウスFc−マウスアンギオスタチン(muFc−muAngio)融合タンパク質として発現させた。PCRを、pdCs−Fs(D4K)ベクターにおける発現にアンギオスタチンcDNA(配列番号6)を合わせるために使用した。正方向プライマーは、5’−C CCC AAG CTT GTG TAT CTG TCA GAA TGT AAG CCC TCC TGT CTC TGA GCA(配列番号26;配列番号27に開示されるアミノ酸をコードする)であり、ここでHindIII部位には、アンギオスタチンのN末端をコードする配列が続いた。逆方向プライマーは、アンギオスタチンのC末端の直後に翻訳終止コドン(アンチコドン、CTA)を配置するように設計された5’−CCC CTC GAG CTA CCC TCC TGT CTC TGA GCA(配列番号28)であり、そしてこれには、XhoI部位(CTCGAG)が続いた。このPCR産物をクローニングし、そして配列決定し、そしてアンギオスタチンをコードするHindIII−XhoIフラグメントを、pdCs−muFc(D4K)ベクターに連結した。muFc(D4K)−muAngioを発現している安定なNS/0クローンを、実施例2および3に記載されるように選択し、そしてアッセイ(抗muFc ELISA)した。
イヌの血液から単離されたイヌ末梢血単球細胞(PBMC)を使用して、mRNAを調製した。逆転写酵素およびオリゴ(dT)を用いる第1鎖cDNAの合成後に、正方向プライマー5’−CC TTA AGC GAA AAT GGA AGA GTT CCT CGC(配列番号29;配列番号30に開示されるアミノ酸をコードした)(ここでAfIII部位を、イヌFcのヒンジ領域をコードする配列のすぐ上流に導入した)および逆方向プライマー5’−C CTC GAG TCA TTT ACC CGG GGA ATG GGA GAG GGA TTT CTG(配列番号31)(ここでXhoI部位を、イヌFcの翻訳終止コドン(アンチコドン、TCA)の後に導入した)を使用して、イヌFc(Kazuhikoら、(1992)JP 1992040894−A1)を増幅するために、PCRを実行した。逆方向プライマーはまた、XmaI制限部位を作製するためにサイレント変異を導入し、これによりリンカー−アダプターを通じたpdCs−caFc(D4K)ベクターの構築およびイヌのエンドスタチンまたはアンギオスタチンをコードするDNA構築物への連結が容易になった。Loら(Loら、Protein Engineering(1998)11:495)に詳細に記載されたpdCs−huFcの構築と同様に、pdCs−caFcについての発現ベクターを、以下の通りに構築した。イヌFcをコードするAfIII−XhoIフラグメントを、軽鎖シグナルペプチドをコードするXbaI−AfIIIフラグメントおよびXbaI−XhoIで消化したpdCsベクターに連結した。次いで、得られたpdCs−caFc発現ベクターを使用して、293細胞をトランスフェクトした。トランスフェクションのおよそ3日後、この上清を、プロテインAクロマトグラフィーにより精製した。イヌFc(配列番号32に示されるDNA配列;配列番号33に示されるアミノ酸配列)の発現を、ペルオキシダーゼ結合体化ウサギ抗イヌIgG(Fcフラグメント特異的)(Jackson ImmunoResearch、West Grove、PA)を使用して、SDS−PAGE、続いてウエスタンブロット分析により確認した。
イヌエンドスタチンのコード配列(配列番号34に示されるDNA配列;配列番号35に示されるアミノ酸配列)を、本質的に実施例5に記載されるように、Fc融合タンパク質としての発現のために、HindIII−XhoIフラグメントに合わせた。3’末端にて、終止コドンを、例えば、PCRによって、C末端のリジン残基をコードするコドンのすぐ後に導入し、そしてこれには、NotI制限酵素部位が続いた。しかし、5’末端では、再構築に都合のよいDraIII制限部位が存在した。HindIII粘着末端およびDraIII粘着末端からなるオリゴヌクレオチド2重鎖を化学的に合成し、そしてイヌエンドスタチンcDNAの残りをコードするDraIII−XhoI制限フラグメントに連結するために使用した。使用された二重鎖を以下に示す:
全長のイヌアンギオスタチンをコードするcDNA(配列番号39に示されるDNA配列;配列番号40に示されるアミノ酸配列)を、本質的に前述の実施例に記載されるように、caFc融合タンパク質として、発現に合わせた。簡潔には、3’末端にて、終止コドンを、例えば、PCRによって、C末端のリジン残基をコードするコドンのすぐ後に導入し、そしてこれには、XhoI部位の代わりのNotI制限部位が続いた。なぜなら、イヌアンギオスタチンのcDNAにおいて、内側XhoI制限部位が存在したからであった。5’末端にて、HindIII部位を、アンギオスタチンのN末端のすぐ上流にインフレーム(in−frame)で導入した。次いで、全長のイヌアンギオスタチンをコードするHindIII−NotIフラグメントを、発現のために、HindIII−NotIで消化したpdCs−caFcベクターに連結した(ここでNotI部位を、リンカー連結を通してXhoI部位に導入した)。caFc−caAngioを発現している安定なNS/0クローンを、実施例2および3に記載されるように選択し、そして抗caFc ELISAによりアッセイした。このタンパク質産物を、SDS−PAGEで分析し、そしてウエスタンブロット分析により確認した。
アンギオスタチンは、プラスミノーゲンの5つのKringleドメインのうちの初めの4つを含む。いずれか1個または数個のKringleドメインがアンギオスタチンの観察された抗脈管形成活性を担うか否かを決定するために、試験のために、1個のKringleドメインを単独でまたはそれらの組合せで産生することが、可能である。融合タンパク質パートナーとしてのFcの有用性を実証するために、マウスアンギオスタチンの第1のKringleドメイン(K1)の発現を、以下の方法で達成した。第1のKringleドメインは、マウスアンギオスタチン(配列番号25)のGlu−87で終了する。cDNA中のこの位置には、都合のよいNsiI制限部位が存在したので、その結果、NsiIによる消化の後に、4塩基の3’突出をT4ポリメラーゼにより除去して、平滑末端を作製した。翻訳終止停止コドンを、パリンドロームリンカーTGA CTC GAG TCA(配列番号41)への連結を通して、Glu−87をコードするGAAのすぐ下流に導入した。ここで終止コドンTGAには、XhoI部位が続いた。次いで、この短縮型アンギオスタチン(すなわち、第1のKringleのみ)をコードするHindIII−XhoIフラグメントを、発現のためにpdCs−muFc(D4K)ベクターに連結した。抗muFc ELISAおよびSDS−PAGEにより分析したとき、高レベルの発現が、一過性発現および安定な発現の両方において得られた。
Kringleドメインは、複数のループ(外側のループおよび内側のループを含む)からなる。マウスアンギオスタチンの第1のKringleでは、この内側のループはCys55およびCys79によって規定され、これは一緒になって、このループの基部でジスルフィド結合を形成する。内側のループのCys−67は、外側のループのCys残基と別のジスルフィド結合を形成し、Kringle構造を生じる。この内側のループが任意の抗脈管形成活性を有するか否かを試験するために、これを、以下の通りにmuFc−内側K1(Kringle1)として発現させた。テンプレートとして第1のKringleをコードするDNAフラグメントとともに、配列5’GGG CCT TGG AGC TAC ACT ACA(配列番号42;配列番号43に開示されるアミノ酸をコードした)を有する変異原性プライマーを使用して、PCRによって、TGC(Cys−67)からAGC(Ser)に変異誘発した。このことは、Kringle1の内側のループが外側のループなしに発現される場合に、Cys−67がジスルフィド結合を形成しないことを保証する。配列5’GCGGATCCAAGCTT AGT ACA CAT CCC AAT GAG GG(配列番号44;配列番号45に開示されるアミノ酸をコードした)を有する上流プライマーを使用して、Ser−43(AGT)に対するコドンのすぐ5’にインフレームでHindIII部位を導入した。BamHI部位もまた、このHindIII部位のすぐ上流に導入した。このBamHI部位は、以下の実施例12に示される可撓性Gly−Serリンカーの末端に、BamHI部位を連結するために有用である。従って、マウスアンギオスタチンのSer−43からGlu−87をコードするHindIII−XhoI DNAフラグメントを、発現のためにpdCs−muFc(D4K)ベクターに連結した。muFc−内側K1の高レベルの発現は、抗muFc ELISAおよびSDS−PAGEによって分析したとき、一過性発現および安定な発現の両方において得られた。

ハイブリッド分子huAngio−huFc−huEndoは、huFc−huEndoにペプチド結合により連結されたヒトアンギオスタチンを含む。このDNA構築物を、以下の通りに構築した。終止コドンを有さないヒトアンギオスタチンをコードするHindIII−XhoIフラグメントを、PCRによって初めに作製し、その結果、アンギオスタチンの最後のアミノ酸残基についてのコドンには、XhoI部位のCTCGAGがすぐ続いた。5’末端のHindIIIを、AfIII−HindIII’アダプターを通して、軽鎖シグナルペプチド(Loら、Protein Engineering(1998)11:495)のXbaI−AfIIIフラグメントに連結した:
薬物動態学研究の1つのセットでは、100〜200mm3のLewis肺腫瘍を移植されたC57/BL6マウスに尾静脈において、1匹のマウス当たり720μgのhuFu−huAngioを注射した。本研究で使用された腫瘍のサイズおよびhuFc−huAngioの用量を選択して、O’Reilly(O’Reillyら、(1996)Nature Medicine 2:689)により記載される実際の処置プロトコルをシミュレートした。血液を眼窩後方の採血により、注射後1/2、1、2、4、8、24、および48hrに回収した。この血液サンプルを、抗huFc ELISA、続いてウエスタン分析によって分析した。huFc−huAngioは、マウスにおいて約32時間の循環半減期を有することが見出され、そしてウエスタン分析は、90%を超えるhu−Fc−huAngioが循環中にインタクトな分子として残っていたことを示した。
