Abstract

Description
本発明は、悪性B細胞およびB細胞由来の腫瘍によって特徴付けられる疾患に対する、アンタゴニスト抗CD40抗体またはその抗原結合性フラグメントを用いた治療の方法に関する。
B細胞はインビボでの通常の免疫応答の間に、重要な役割を果たす。外来の抗原は特定B細胞の免疫グロブリン表面に結合し、エンドサイトーシス、プロセシング、クラスII−MHC分子上にプロセシングされたペプチドの提示、およびB細胞表面上へのB7抗原の上方調節を含む一連の事象を誘発する。その後、特定のT細胞は、クラスII−MHC分子上への提示されたプロセシングされた抗原のT細胞受容体(TCR)認識を介してB細胞に結合する。TCRを介した刺激によって、T細胞が活性化され、T細胞がサイトカインの産生を開始する。T細胞をさらに活性化する二番目のシグナルは、T細胞上のCD28抗原とB細胞上のB7抗原との間の相互作用である。上記シグナルが受容されると、CD40リガンド(これは休止ヒトT細胞上には提示されていない)が、T細胞表面上で上方調節される。CD40リガンドが、B細胞表面上のCD40抗原に結合することにより、B細胞が刺激され、B細胞を可溶性免疫グロブリンを高レベルで分泌する形質細胞へ成熟させる。
非ホジキンリンパ腫(高悪性度リンパ腫、中悪性度リンパ腫、および低悪性度リンパ腫)、ホジキン病、急性リンパ芽球性白血病、骨髄腫、慢性リンパ球性白血病、および骨髄芽球性白血病のようなリンパ腫を含む、悪性B細胞を含む疾患の患者を処置する方法を、提供する。本方法は、抗CD40抗体またはその抗原結合フラグメントを用いて患者を処置する工程を含む。この抗CD40抗体またはその抗原結合フラグメントは、正常ヒトB細胞上のCD40抗原に結合した場合、有意なアゴニスト活性がなく、悪性のヒトB細胞上のCD40抗原に結合した場合、アンタゴニスト活性を示す。本発明の方法での使用に適したモノクローナル抗体およびその抗原結合フラグメントは、以下の性質を示す:1)ヒト細胞の表面に発現されているヒトCD40抗原へ特異的に結合し得る、2)正常ヒトB細胞上のCD40抗原へ結合した場合、有意なアゴニスト活性がない、および3)悪性ヒトB細胞上のCD40抗原へ結合した場合、アンタゴニスト活性を示す。いくつかの実施形態において、本抗CD40抗体またはそのフラグメントはまた、正常ヒトB細胞上のCD40抗原へ結合した場合、アンタゴニスト活性を示す。モノクローナル抗体は、CD40へ強い親和性を示し、少なくとも10−5M、好ましくは少なくとも約10−8Mから約10−20M、より好ましくは少なくとも約5×10−9Mから約10−16Mの解離定数(Kd)によって特徴付けられる。適切なモノクローナル抗体は、ヒト定常領域を有する。その定常領域は、好ましくは、全体的または部分的にヒト化したフレームワークを有し、および、最も好ましくは、完全にヒト抗体またはその抗原結合フラグメントを有する。そのようなモノクローナル抗体の例は、本明細書中で15B8と記載されている抗体、15B8と記載されているハイブリドーマ細胞株から産生されるモノクローナル抗体、配列番号2および配列番号4からなる群から選択されたアミノ酸配列を含むモノクローナル抗体、配列番号1および配列番号3からなる群から選択されたヌクレオチド配列を含む核酸分子によりコードされるアミノ酸配列を含むモノクローナル抗体;およびヒトCD40への特異的な結合能を有している、これらモノクローナル抗体の抗原結合フラグメントである。
上記目的を達成するために、本発明は、例えば、以下の手段を提供する。
(項目1) 悪性B細胞を含む疾患を有する患者を処置する方法であって、該方法は、治療有効量のヒト抗CD40モノクローナル抗体またはその抗原結合フラグメントを、該患者に投与する工程を包含し、ここで、該抗体またはそのフラグメントが正常ヒトB細胞上のCD40抗原に結合する場合、該抗CD40モノクローナル抗体またはそのフラグメントには有意なアゴニスト活性がなく、かつ該抗体またはそのフラグメントが、悪性ヒトB細胞上のCD40抗原に結合する場合、該抗CD40モノクローナル抗体またはそのフラグメントがアンタゴニスト活性を示し、ここで、該ヒト抗CD40モノクローナル抗体は、以下:
a)ハイブリドーマ細胞株15B8により生成されるモノクローナル抗体;
b)配列番号2および配列番号4からなる群より選択されるアミノ酸配列を含むモノクローナル抗体;
c)配列番号1および配列番号3からなる群より選択されるヌクレオチド配列を含む核酸分子によりコードされるアミノ酸配列を有するモノクローナル抗体;ならびに
d)モノクローナル抗体であって、該モノクローナル抗体が、a)、b)、またはc)に記載のモノクローナル抗体のフラグメントであり、かつ該フラグメントが、ヒトCD40に特異的に結合する能力を保持する、モノクローナル抗体からなる群より選択され、ここで、該患者を処置する方法が、該患者における望ましい治療応答を促進する、方法。
(項目2) 項目1に記載の方法であって、前記抗体またはそのフラグメントが、前記正常ヒトB細胞上の前記CD40抗原に結合する場合、該ヒト抗CD40モノクローナル抗体またはそのフラグメントがアンタゴニスト活性を示す、方法。
(項目3) 前記ヒト抗CD40モノクローナル抗体が、少なくとも約10 −5 Mの解離定数(K d )を有する、項目1に記載の方法。
(項目4) 前記ヒト抗CD40モノクローナル抗体が、少なくとも約10 −8 M〜約10 −20 MのK d を有する、項目3に記載の方法。

(項目7) 項目1に記載の方法であって、前記悪性B細胞が、B細胞リンパ腫細胞、非ホジキンリンパ腫細胞、高悪性度B細胞リンパ腫細胞、中悪性度B細胞リンパ腫細胞、低悪性度B細胞リンパ腫細胞、B細胞急性リンパ芽球性白血病細胞、多発性骨髄腫細胞、慢性リンパ性白血病細胞、骨髄芽球性白血病細胞およびホジキン病細胞からなる群より選択される、方法。
(項目8) 項目1に記載の方法であって、前記処置が、前記ヒト抗CD40モノクローナル抗体またはそのフラグメントを含む薬学的組成物の、少なくとも1治療有効用量を、前記患者に投与する工程を包含する、方法。
(項目9) 前記ヒト抗CD40モノクローナル抗体またはそのフラグメントの治療有効用量が、約0.01mg/kg〜約40mg/kgの範囲内である、項目8に記載の方法。
(項目10) 前記処置が、複数の治療有効用量の前記ヒト抗CD40モノクローナル抗体またはそのフラグメントを投与する工程を包含する、項目1に記載の方法。
(項目11) 前記抗原結合フラグメントが、Fabフラグメント、F(ab’) 2 フラグメント、Fvフラグメントおよび単鎖Fvフラグメントからなる群より選択される、項目1に記載の方法。

(項目13) 項目12に記載の方法であって、前記悪性細胞が、B細胞リンパ腫細胞、非ホジキンリンパ腫細胞、高悪性度B細胞リンパ腫細胞、中悪性度B細胞リンパ腫細胞、低悪性度B細胞リンパ腫細胞、B細胞急性リンパ芽球性白血病細胞、多発性骨髄腫細胞、慢性リンパ性白血病細胞、骨髄芽球性白血病細胞およびホジキン病細胞からなる群より選択される、方法。
(項目14) 前記ヒト抗CD40モノクローナル抗体が、少なくとも約10 −5 Mの解離定数(K d )を有する、項目12に記載の方法。

本発明は、悪性B細胞由来の疾患を有するヒト患者を治療するための方法に関する。本方法は、抗CD40抗体またはその抗原結合フラグメントを用いた処置を含み、その処置法において、抗体またはその抗原結合フラグメントの投与が、この治療法を受けた患者内に正の治療上の応答を促進する。本発明の方法での使用に適切な抗CD40抗体は、以下の特徴を有する:1)これらはヒト細胞表面上に発現しているヒトCD40抗原に特異的に結合する;2)これらは、正常ヒトB細胞上のCD40抗原へ結合した場合、有意なアゴニスト活性がない;および3)これらは、悪性ヒトB細胞上のCD40抗原へ結合した場合、アンタゴニスト活性を示す。これら抗CD40抗体およびその抗原結合フラグメントは、本明細書では、アンタゴニスト抗CD40抗体として、参照されている。このような抗体は、以下に記載されている完全なヒトモノクローナル抗体15B8およびモノクローナル抗体15B8の結合特性を有するモノクローナル抗体を含むが、これらに限定されない。以下により詳細に議論されているように、これら抗体は、CD40受容体に特異的である。正常なヒトB細胞表面上に提示されたCD40に、これら抗体が結合した場合、その抗体は有意なアゴニストの活性がない。いくつかの実施形態において、正常なヒトB細胞表面上に提示されたCD40へのそれら抗体の結合は、結果として、これら正常なヒトB細胞の増殖および分化を阻害する。従って、本発明の方法における使用に適切なアンタゴニスト抗CD40抗体は、細胞表面にCD40抗原を発現している正常なヒトB細胞へのアンタゴニスト活性を示し得るこれらのモノクローナル抗体を含む。アンタゴニスト抗CD40抗体が悪性ヒトB細胞表面上に提示されているCD40に結合した場合、抗体は本明細書中の他の箇所で定義されたアンタゴニスト活性を示す。

ック動物、好ましくはヒト免疫グロブリン遺伝子座を有するマウス内で調製され得る。好ましい実施形態において、CD40を発現しているSf9細胞は、免疫原として使用される。免疫は、生理食塩水中に、好ましくはフロイントの完全アジュバンドのようなアジュバンド中に、抗原含有溶液を混合または乳化し、非経口的(一般的には皮下または筋肉内に)この混合物または乳濁物を注射することによって実行され得る。一投与あたり50−200μgの投与量は、代表的には十分量である。免疫は、生理食塩水中、好ましくはフロイント不完全アジュバンドを用いて、タンパク質の一度以上の注射によって、一般的に2−6週間後に増強される。あるいは、当該技術分野中で公知の方法を使用したインビトロでの免疫によって、当業者は抗体を産生し得る。本発明の目的に対して、この方法は、インビトロ免疫と等価であると考えられている。ポリクローナル抗血清は、ガラスまたはプラスチック容器へ免疫された動物から採血し、25度で1時間血液をインキュベーションし、続いて4度で2−18時間インキュベーションすることによって得られる。その血清は遠心分離(例えば、1000×gで10分間)によって回収される。1回の採血あたり約20−50mlがウサギから得られ得る。

下記の実施例において使用されるアンタゴニスト抗CD40抗体は、15B8である。15B8は、ヒトIgG2重鎖の遺伝子座およびヒトK軽鎖の遺伝子座を有するトランスジェニックマウス(Xenomouse、Abgenix)の免疫により生産される、ヒトIgG2サブタイプの抗ヒトCD40モノクローナル抗体である。FACS分析により示されるように、15B8は、ヒトCD40に特異的に結合し、かつサル(カニクイザル、アカゲザルおよびヒヒ)およびチンパンジー由来の末梢血B細胞上のCD40と交差反応する。15B8は、霊長類でない動物種由来のCD40と交差反応せず、ELISAおよびFACS分析により実証されるように、TNFレセプターファミリーの他のメンバーと結合もしない。ヒトCD40に対する15B8の結合親和力は、BIACoreアッセイにより測定した場合、3.1×10−9Mである。
CD40結合との直接競合が15B8のアンタゴニスト活性の機構であるか否かを決定するために、競合結合アッセイを実施した。CD40Lをコードする遺伝子を含みかつ細胞表面上にCD40Lを発現する、チャイニーズハムスター卵母細胞(CHO)株を作製した。このCD40L発現CHO細胞を、15B8とCD40とのインキュベーションの前および後に、精製されたCD40と共にインキュベートした。フルオレセインイソチオシアネート(FITC)標識した抗huIgGを、上記細胞に添加した。FACS分析を実施し、CD40を介してこのCHO細胞に結合した15B8を検出した。CD40への15B8の結合は、その後のCD40へのCD40Lの結合を阻害した。しかし、CD40LおよびCD40を、15B8の添加前に一緒にインキュベートした場合、15B8は、その後CD40への結合が可能であった。いかなる作用機構にも結び付けられないが、このことは15B8がCD40上の結合部位に対してCD40Lと直接は競合しないこと、そしてCD40への15B8の結合は、おそらくCD40分子内にCD40へのCD40Lの結合を妨げる高次構造変化を引き起こしたことを、示唆する。15B8の結合により誘導されるCD40分子の推定構造変化もまた、上記細胞にネガティブシグナルを伝達し得、アンタゴニスト効果を起こし得る。
非ホジキンリンパ腫(NHL)の前臨床インビトロモデルにおいて、15B8の可能な効力を実証するために、Rituximab処置されたNHL患者または未処置のNHL患者のいずれかより採取された悪性B細胞(NHL細胞)を使用して、15B8を試験した。Rituximab(IDEC−C2B8;Rituxan(登録商標);IDEC Pharmaceuticals Corp.,San Diego,California)は、再発性または治療抵抗性の、低悪性度または濾胞性の、NHL処置用抗CD20モノクローナル抗体である。
chidaら(1999)Science 286:300−303;Farrelら(1997)Biomed.Pharmacother.51:258−267)。NHL細胞および正常B細胞におけるCD40シグナル伝達とは対照的に、CD40シグナル伝達は、EBV形質転換バーキットリンパ腫細胞株において増殖阻止をもたらす(Fukudaら(2000)Viral Immunol.13:215−229;Bakerら(1998)Blood 92:2830−2843)。従って、この異種移植モデルにおけるアンタゴニスト抗CD40 MAb(15B8)の試験の結果から、NHL患者によるこの抗体(15B8)に対する応答を推定することは、不可能である。
15B8がインビトロにおいてCD40L様の増殖シグナルを提供するかどうかを試験するため、腫瘍に浸潤されたリンパ節由来のB細胞(NHL細胞)を、抗体未処置のNHL患者1人、Rituximab感受性のNHL患者1人およびRituximab抵抗性のNHL患者1人より、採取した。これらのNHL細胞を、以下の4つの異なる培養条件下で研究した:抗体の添加なし(培地);ヒトアイソタイプ抗体IgGの2添加(コントロール;huIgG2と呼ぶ);抗CD40抗体MS81(アゴニスト抗体)の添加;および15B8の添加。IL−4の存在下または非存在下にて、全ての抗体を1μg/ml、2μg/mlおよび5μg/mlで試験した。2人の患者由来のNHL細胞を、上記のように同じ4つの条件下でIL−4(2ng/ml)の存在下で、培養した。B細胞増殖を、上記の3H−チミジン取り込みにより測定した。
インビトロにおいてCD40Lによって提供される増殖シグナルをブロックする15B8の能力を評価するため、患者由来のNHL細胞を、4つの異なる条件下で上記のようにCD40L発現フィーダー細胞上の懸濁物中にて培養した:抗体の添加なし(medium);ヒトアイソタイプ抗体IgG2の添加(コントロール);抗CD40抗体MS81(アゴニスト抗体)の添加;および15B8の添加。全ての抗体を、IL−4存在下または非存在下で1μg/ml、2μg/mlおよび5μg/mlの濃度で添加した。抗体未処置の患者1人、Rituximab感受性の患者2人、およびRituximab抵抗性の患者5人(合計8人の患者)由来のNHL細胞を、上記と同じ4つの条件下で、IL−4の存在下(2ng/ml)にて培養した。Rituximab感受性の患者3人およびRituximab抵抗性の患者4人(合計7人の患者)由来のNHL細胞を、同様の条件下でIL−4の非存在下にて培養した。NHL細胞の増殖を、3H−チミジン取り込みにより測定した。

15B8がアゴニスト抗CD40であるか、またはアンタゴニスト抗CD40であるかを決定するために、15B8を、ヒトおよび5つの異なる霊長類種(チンパンジー(chimp)、マーモセット、カニクイザル、アカゲザル、およびヒヒを含む)由来の細胞を用いて、以下に記載するいくつかのインビトロアッセイにて試験した。
2.細胞増殖の結果を、huIgG2コントロールに対する15B8の3H−チミジン取り込みの比として記録する。いくつかのサンプルからのデータは、CD40L(陽性コントロール)により誘導されるCPMが<2000のため、表中に含まれない

15B8がアンタゴニスト抗CD40であるか否かを決定するために、CD40−CD40L相互作用を阻害するその能力を、CD40L媒介性のヒトB細胞増殖アッセイ(Kwekkeboomら(1993)Immunology 79:439−444)にて試験した。ヒトCD40Lを発現するトランスフェクトされたCHO細胞株を使用して、精製されたヒト末梢血B細胞またはPBMCの増殖を刺激した。10人の健常提供者由来のヒトB細胞および3人の健常提供者由来のヒトPBMCを試験した。試験されたサンプル全てにおいて、15B8は、濃度範囲0.2〜5μg/mlで、CD40L発現CHO細胞媒介性の増殖を42%〜88%抑制した(表5)。図5は、3人の個体由来の細胞を使用した代表的な用量応答曲線を示す。15B8の無効用量は0.008μg/mlであり、そして0.2μg/mlで飽和用量に達する(図5)。この観察は、アンタゴニスト抗CD40抗体としての15B8がヒトB細胞およびPBMCにて細胞表面に発現されたCD40Lにより提供される増殖信号を阻害し得ることを示す。

2.「15B8 %阻害」=100−(15B8 cpm/CD40L cpm × 100)
いくつかのサンプルからのデータは、CD40L(陽性コントロール)によって誘導される増殖が<5倍なので、表中にはない。
FACS分析は、15B8がサル(カニクイザル、アカゲザル、およびヒヒ)末梢血由来のB細胞表面上に発現されたCD40に結合することを実証する。新たに単離されたカニクイザルPBMCに対する15B8の効果を、ヒトおよびチンパンジーについて上記した同一の増殖アッセイ(Coliganら(1998)Current Protocols in Immunology 13:12;Kwekkeboomら(1993)Immunology 79:439−444)にて、試験した。3H−メチルチミジン取り込みにより測定する場合、ヒトPBMCとは対照的に、15B8が、カニクイザルPBMCを刺激してインビトロにおいて増殖させることを見出した(下記の表6)。1μg/mlのレベルで、15B8は、試験された17匹のサル由来の22サンプル(5匹のサル由来のサンプルを2回試験した)にて、huIgG2コントロールと比較してPBMCの増殖を6倍〜129.7倍刺激した(下記の表6)。5μg/mlのレベルで、15B8により刺激された増殖は、2匹のサル由来の4つのサンプルにおいて14倍〜24倍、および2匹のサル由来の2つのサンプルにおいて約1.25倍または約1.85倍である(表6)。このことは、15B8が5μg/mlの濃度レベルで、カニクイザル由来のPBMCに対するその増殖刺激効果についての飽和用量を超える限界であり得ることを、示唆する。表面マーカーによる活性化状態についてのB細胞のさらなるFACS分析は、15B8がサルB細胞に対してCD69、CD86およびHLA−DRの上方制御を誘導することを示した(表7)。こららのデータは、15B8が、カニクイザル由来の末梢血B細胞上に発現されるCD40に対するインビトロでのアゴニスト抗体であることを、示唆する。

CD40LをトランスフェクトしたCHO細胞を、実験前にホルムアルデヒドを用いて固定化した。
2.「N/A」は、測定されなかったか、または成功しなかったことを意味する。
3.細胞数が制限されたので、1匹のカニクイザル由来の細胞のみをFACSによって3日目に分析した。
15B8は、カニクイザル由来のPBMCにて、インビトロで増殖を刺激し得、そして細胞表面の活性化マーカーを上方制御し得る。15B8がこれらのサルにおいてインビボでのアゴニスト抗CD40抗体であるか否かを決定するために、15B8の生体分布および影響を受けた末梢血B細胞の運命(すなわち、血管外分布、アポトーシス、状態の活性化、または補体の溶解)を試験するため、研究を実施した「Biodistribution of 15B8.72 Antibodies following Intravenous Administration to Non−Naive Male and Female Cynomologus Monkeys(SNBL.218.3、SNBL(USA))」。
3匹の雄チンパンジーの2つの集団は、静脈内投与により0.03mg/kgまたは3mg/kgのいずれかの15B8を受けた。血清15B8濃度および末梢B細胞数を、15B8の投与直後より投与後29日にかけてモニターした。この実験結果を図6に示す。3mg/kgでの15B8の投与後、血清15B8濃度は、短期拡散期、対数線形性の消失期および非線形性の消失期を含む3相の様式で減少した。およそ10μg/ml未満の濃度では、非線形性の消失期が優先的であった。対数線形期中の半減期はおよそ4日であった。末梢B細胞数は15B8投与後直ぐに減少し、そして3〜4週間以内に回復した。15B8は、血清中において検出され、循環しているB細胞上の表面CD40レセプターに結合した。結合の程度は、2日目から投与後8日まで相対的に変わらないままであり、そしてそれから投与後29日にかけて消失するようであった。
ヒトIgMおよびヒトIgGの濃度をELISAアッセイにより測定した。96−ウェルELISAプレートを、4μg/mlの抗ヒトIgG mAb、または1.2μg/mlの抗ヒトIgM mAbを用いて、0.05Mの炭酸緩衝液(pH=9.6)中で16時間4℃でコートした。プレートをPBS−0.05%Tween−20(PBS−Tween)を用いて3回洗浄し、BSAで1時間飽和させた。2回洗浄後、プレートを1時間37℃で希釈率の異なる試験サンプルと共にインキュベートした。3回洗浄後、1μg/mlのペルオキシダーゼ標識された、マウス抗ヒトIgG mAbまたはマウス抗ヒトIgM mAbと共に1時間37℃でインキュベートすることにより、結合したIgを検出した。プレートを4回洗浄し、そして結合したペルオキシダーゼ活性をo−フェニレンジアミンを基質として添加することにより示した。
(インビトロアッセイの要約)
本結果は、15B8が、カニクイザル、アカゲザルおよびヒヒにおいて、アゴニスト性の抗CD40抗体であること、および、ヒト、チンパンジーおよびマーモセットにおいてアンタゴニスト性の抗体であることを示唆する。実施された実験を以下の表に要約する。
Claims (18)
- 患者のB細胞悪性腫瘍を処置するための組成物であって、該組成物は、治療有効量の結合体化されたヒト抗CD40モノクローナル抗体または結合体化されたその抗原結合フラグメントを含み、ここで、該結合体化された抗体または結合体化されたそのフラグメントが正常ヒトB細胞上のCD40抗原に結合する場合、該結合体化された抗CD40モノクローナル抗体または結合体化されたそのフラグメントには有意なアゴニスト活性がなく、かつ該結合体化された抗体または結合体化されたそのフラグメントが、悪性ヒトB細胞上のCD40抗原に結合する場合、該結合体化された抗CD40モノクローナル抗体または結合体化されたそのフラグメントがアンタゴニスト活性を示し、ここで、該結合体化されたヒト抗CD40モノクローナル抗体は、以下:
a)配列番号2および配列番号4からなる群より選択されるアミノ酸配列を含むモノクローナル抗体;
b)配列番号1および配列番号3からなる群より選択されるヌクレオチド配列を含む核酸分子によりコードされるアミノ酸配列を有するモノクローナル抗体;ならびに
c)モノクローナル抗体であって、該モノクローナル抗体が、a)またはb)に記載のモノクローナル抗体のフラグメントであり、かつ該フラグメントが、ヒトCD40に特異的に結合する能力を保持する、モノクローナル抗体、
からなる群より選択される抗体を含み、ここで、該患者を処置するための組成物が、該患者における望ましい治療応答を促進する、組成物。 - 請求項1に記載の組成物であって、前記結合体化されたヒト抗CD40モノクローナル抗体または結合体化されたそのフラグメントが、前記正常ヒトB細胞上の前記CD40抗原に結合する場合、該結合体化された抗体または結合体化されたそのフラグメントがアンタゴニスト活性を示す、組成物。
- 前記結合体化されたヒト抗CD40モノクローナル抗体が、少なくとも10 −5 Mの解離定数(K d )を有する、請求項1に記載の組成物。
- 前記結合体化されたヒト抗CD40モノクローナル抗体が、10 −8 M〜10 −20 MのK d を有する、請求項3に記載の組成物。
- 前記結合体化されたヒト抗CD40モノクローナル抗体が、10 −9 M〜10 −16 MのK d を有する、請求項4に記載の組成物。
- 請求項1に記載の組成物であって、前記B細胞悪性腫瘍が、B細胞リンパ腫、非ホジキンリンパ腫、高悪性度B細胞リンパ腫、中悪性度B細胞リンパ腫、低悪性度B細胞リンパ腫、B細胞急性リンパ芽球性白血病、多発性骨髄腫、慢性リンパ性白血病、骨髄芽球性白血病およびホジキン病からなる群より選択される、組成物。
- 前記結合体化された抗原結合フラグメントは、Fabフラグメント、F(ab')フラグメント、Fvフラグメントおよび単鎖Fvフラグメントからなる群より選択される抗原結合フラグメントを含む、請求項1に記載の組成物。
- 患者のB細胞悪性腫瘍を処置するためのキットであって、該キットは、以下:
請求項1に記載の組成物、および
指示書であって、該処置が、前記結合体化されたヒト抗CD40モノクローナル抗体または結合体化されたそのフラグメントを含む該組成物の、少なくとも1治療有効用量を、該患者に投与する工程を包含することを示す、指示書、
を備える、キット。 - 前記結合体化されたヒト抗CD40モノクローナル抗体または結合体化されたそのフラグメントの治療有効用量が、0.01mg/kg〜40mg/kgの範囲内である、請求項7に記載のキット。
- 患者のB細胞悪性腫瘍を処置するためのキットであって、該キットは、以下:
請求項1に記載の組成物、および
指示書であって、該処置が、複数の治療有効用量の前記結合体化されたヒト抗CD40モノクローナル抗体または結合体化されたそのフラグメントの投与を含むことを示す、指示書、
を備える、キット。 - B細胞系統の悪性細胞の増殖を阻害するための組成物であって、該組成物が、有効量の結合体化されたヒト抗CD40モノクローナル抗体または結合体化されたその抗原結合フラグメントを含み、該結合体化された抗体または結合体化されたそのフラグメントには有意なアゴニスト活性がなく、ここで、該結合体化された抗体または結合体化されたそのフラグメントが該悪性細胞上のCD40抗原に結合する場合、該悪性細胞の増殖は阻害され、ここで、該結合体化されたヒト抗CD40モノクローナル抗体は以下:
a)配列番号2および配列番号4からなる群より選択されるアミノ酸配列を含むモノクローナル抗体;
b)配列番号1および配列番号3からなる群より選択されるヌクレオチド配列を含む核酸分子によりコードされるアミノ酸配列を有するモノクローナル抗体;ならびに
c)モノクローナル抗体であって、該モノクローナル抗体が、a)またはb)に記載のモノクローナル抗体のフラグメントであり、かつ該フラグメントが、ヒトCD40に特異的に結合する能力を保持する、モノクローナル抗体、
からなる群より選択される抗体を含む、組成物。 - 請求項11に記載の組成物であって、前記悪性細胞が、B細胞リンパ腫細胞、非ホジキンリンパ腫細胞、高悪性度B細胞リンパ腫細胞、中悪性度B細胞リンパ腫細胞、低悪性度B細胞リンパ腫細胞、B細胞急性リンパ芽球性白血病細胞、多発性骨髄腫細胞、慢性リンパ性白血病細胞、骨髄芽球性白血病細胞およびホジキン病細胞からなる群より選択される、組成物。
- 前記結合体化されたヒト抗CD40モノクローナル抗体が、少なくとも10 −5 Mの解離定数(K d )を有する、請求項11に記載の組成物。
- 前記結合体化されたヒト抗CD40モノクローナル抗体が、10 −8 M〜10 −20 MのK d を有する、請求項11に記載の組成物。
- 前記結合体化されたヒト抗CD40モノクローナル抗体が、10 −9 M〜10 −16 MのK d を有する、請求項14に記載の組成物。
- 前記結合体化されたヒト抗CD40モノクローナル抗体または結合体化されたそのフラグメントは、治療剤の部分と結合体化されているものである、請求項1〜7、11〜15までのいずれか1項に記載の組成物、または請求項8〜10のいずれか1項に記載のキット。
- 前記治療剤の部分は、サイトトキシン、細胞傷害剤、化学療法剤、治療タンパク質、または放射性金属イオンからなる群より選択されるものである、請求項16に記載の組成物またはキット。
- 前記結合体化されたヒト抗CD40モノクローナル抗体または結合体化されたそのフラグメントは、第2の抗体と結合体化されているものである、請求項1〜7、11〜15までのいずれか1項に記載の組成物、または請求項8〜10のいずれか1項に記載のキット。
