Abstract

CD8 T細胞免疫応答を発生するワクチン接種のための方法および試薬
本発明はCD8+ T細胞エピトープ源として異なった初回刺激および追加刺激組成物を使用した標的抗原に対する保護的CD8+ T細胞免疫応答の発生に関している。
ワクチン学における一般的問題は、免疫化により高レベルのCD8 T細胞を発生できないことである。このことはマラリアを含むいくつかの疾患に対するワクチンの開発を妨げてきた。
(i) 標的抗原の一つまたはそれ以上のCD8+ T細胞エピトープ源、ならびに医薬品として受容可能な担体から成る初回刺激組成物;および
(ii)初回刺激組成物のCD8+ T細胞エピトープと同一である少なくとも一つのCD8+ T細胞エピトープ源を含む標的抗原の一つまたはそれ以上のCD8+ T細胞エピトープ源、ここでCD8+ T細胞エピトープ源は非複製または複製欠陥組換えポックスウイルスベクターである、ならびに医薬品として受容可能な担体から成る追加刺激組成物;
ただし、(i)のエピトープ源がウイルスベクターであるならば、(ii)におけるウイルスベクターは異なったウイルスから誘導される。
本発明の一つの好適な態様において、初回刺激組成物中のCD8+ T細胞エピトープ源は核酸(DNAでもRNAでもよい)、特に組換えDNAプラスミドである。DNAまたはRNAは包まれていてもよいし(例えば、リソソーム中に)または遊離の形でもよい。
非複製または複製欠陥であるウイルスは自然にそうなるかまたはインビトロでの繁殖によりまたは例えば、複製に決定的である遺伝子の欠損のような遺伝子操作により人工的に作製される。MVAに対するCEF細胞のように、ウイルスが増殖できる一つまたは少数の細胞型が一般的に存在するであろう。
1)MRC−5細胞(ヒト細胞株)においてワクシニアウイルスのコペンハーゲン株と比較してDNA合成に1log(10倍)の減少を示す;
2)ヒーラー細胞(ヒト細胞株)においてワクシニアウイルスのコペンハーゲン株と比較してウイルス力価に2logの減少を示す。
本発明に従った初回刺激に使用するための別の好適なウイルスベクターには、遺伝学上非複製または複製欠陥であるようになされた種々の異なったウイルスが含まれる。非複製または複製欠陥ベクターを産生するようなウイルスの遺伝子操作は文献に多く記載されている(例えば、McLeanら1994)。
病原体または腫瘍に対する保護的CD8+ T細胞免疫応答を発生させるための方法であって、但し本方法は少なくとも一つの病原体または癌のエピトープまたは抗原を含む組換えタンパク質または粒子の少なくとも一回量、続いて同一のエピトープまたは抗原をコードしている組換えMVAベクターの少なくとも一回量を投与することから成る;
CD8+ T細胞免疫応答を追加刺激するための医薬品の製造における組換え非複製または複製欠陥ポックスウイルスベクターの使用;
CD8+ T細胞免疫応答を追加刺激するための医薬品の製造におけるMVAベクターの使用;
標的抗原の一つまたはそれ以上のCD8+ T細胞エピトープ源を含む、少なくとも一つの標的抗原またはエピトープに対する初回刺激CD8+ T細胞応答を追加刺激するための医薬品ならびに医薬として受容可能な担体、ここでCD8+ T細胞エピトープ源は非複製または複製欠陥組換えポックスウイルスベクターである;および 遺伝子銃による送達に適した特別な形の本明細書に記載されている初回刺激および/または追加刺激組成物;および遺伝子銃法による組成物の送達から成る免疫化法。

プロトコール:初回刺激組成物の1mgをi.m.で0および3週に2回、続いて追加刺激物を6および9週に皮内に投与する。

追加刺激組成物: MVA、108ffu PBS溶液
プロトコール:初回刺激組成物をi.m.で0および3週に2回、続いて追加刺激物を6および9週に投与する。腫瘍処置のためには、MVAは最も有効な経路としてi.v.で投与される。

初回刺激組成物: タンパク質500μg+アジュバント(QS−21)
追加刺激組成物: 組換えMVA、108ffu PBS溶液
プロトコール:初回刺激組成物を0および3週に2回、続いて追加刺激物を6および9週にi.d.で投与する。

実施例1
材料および方法
エピトープ列の発生
マラリアエピトープ列は各々が表1に示されたような三つのエピトープをコードしている一連のカセットから作製された(カセットは各々の末端に制限酵素部位を含んでいる)。各々のカセットは四つの合成オリゴヌクレオチドから構築され、それらはお互いにアニールされ、クローニングベクター内へ結合され、続いてエラーが導入されていないことを検査するために配列決定された。個々のカセットは次に必要に応じてお互いに連結された。カセットCの3’末端のBamHI部位はカセットAの5’末端のBglII部位と融合され、両方の制限酵素部位を破壊して二つのカセット間に二つのアミノ酸スペーサー(GS)がコードされる。カセットB,DおよびHは次に同一の方法で一列に連結された。CABDHFEを含むより長い列も同じ方法で構築された。
カセットCABDHを含んでいるエピトープ列はC末端をTyAタンパク質読み枠内融合するように酵母発現ベクター内へ導入された。TyAまたはTyA融合タンパク質が酵母内でこのベクターから発現された場合、タンパク質は自発的にウイルス様粒子を形成し、それはショ糖濃度勾配遠心分離により酵母の細胞質から分離できる。組換えTy−VLPはこの方法で調製され、注射前にショ糖を除去するためにPBSに対して透析された(Laytonら1996参照)。
E1遺伝子の欠損した複製欠陥組換えアデノウイルスが本研究で使用された(McGroryら1988)。アデノウイルスはCMV IEプロモーター調節下で大膳菌β−ガラクトシダーゼを発現した。免疫化のためには、107pfuのウイルスが耳垂内に皮内で投与された。
ペブチドはResearch Genetics(USA)から購入され、10mg/mlでDMSO(Sigma)に溶解され、さらにPBSで1mg/mlに希釈された。実験に使用されたCTLエピトープを含んでいるペプチドは表3に記載されている。
多数の異なったベクターがDNAワクチンを構築するために使用された。プラスミドpTHは抗原コード配列およびウシ成長ホルモン転写終止配列の導入を可能にするため、ポリリンカーが続いたイントロンAを持つCMV IEプロモーターを含んでいる。本プラスミドはアンピシリン耐性遺伝子を運んでおり、大腸菌内では複製できるが哺乳動物細胞では複製できない。これは以下の抗原の各々を発現するDNAワクチンを製作するために使用された:P.ブルゲイTRAP、P.ブルゲイCS、熱帯熱マラリア原虫TRAP、熱帯熱マラリア原虫LSA−1(C末端の278アミノ酸のみ)、カセットCABDHを含んでいるエピトープ列、およびHMエピトープ列(HIVエピトープにカセットCABが続いたもの)。プラスミドpSG2は抗生物質耐性遺伝子を除いてpTHと同じである。
組換えMVAは最初に抗原配列をプラスミドpSC11のようなウイルスプロモーターを持つシャトルベクター内へクローニングすることにより作製された(Chakrabartiら1985;Morrisonら1989)。P.ベルゲイCSおよび熱帯熱マラリア原虫TRAP、インフルエンザ核タンパク質およびHMおよびマウス腫瘍エピトープポリエピトープ列はP7.5プロモーター(Mackettら1984)を用いて発現され、およびP.ベルゲイTRAPは強力合成プロモーター(SSP;Carrollら1995)を用いて発現された。次に、プロモーター、抗原コード配列およびマーカー遺伝子に隣接するウイルス配列をMVAと組換えして組換え体が生成するように、シャトルベクターpSC11またはpMCO3が野生型MVAで感染させた細胞の形質転換に使用された。組換えウイルスはマーカー遺伝子(βグルクロニダーゼまたはβガラクトシダーゼ)を発現するので組換えウイルスを含んでいるプラークの同定が可能である。組換え体は免疫化に使用する前に繰り返してプラーク精製された。組換えNYVAC−PbCSPワクシニアは以前に報告されている(Lanarら1996)。PbCSPをコードしている組換えワクシニアの野生型またはウェスタンリザーブ(WR)株は以前に報告されている(Satchidanandamら1991)。
マウス細胞およびエプスタイン−ーバールウイルス形質転換チンパンジーおよびマカクB細胞(BCL)は10%熱不活性化ウシ胎児血清(FCS)を補給したRPMIで培養した。脾細胞は10%FCS、2mMグルタミン、50U/mlペニシリン、50μM 2−メルカプトエタノールおよび10mMヘペスpH7.2(Gibco,UK)を加えたMEM培地中、示されたペプチド(最終濃度1μg/ml)で再刺激された。
示された系統のマウス、6−8週齢はHarlan Olac(Shaws Farm,Blackthorn,UK)から購入された。チンパンジーH1およびH2はBiomedical Primate Research Centre,Rijswick,The Netherlandsで研究された。マカクはUniversity of Oxfordで研究された。
マウスのプラスミドDNA免疫化は麻酔下、頸骨筋内へのDNAの筋肉内免疫化により実施された。マウス筋肉は時によりDavisら(1993)により記載されているように免疫化の5−9日前に50μlの1mMカルジオトキシン(Latoxan,France)で前処理されたが、そのような前処置は免疫原性または保護効能には何の有意な影響も与えなかったことが観察された。マウスのMVA免疫化は筋肉内(i.m.)、静脈内(外側尾静脈内)(i.v.)、皮内(i.d.)腹腔内(i.p.)または皮下(s.c.)免疫化により実施された。チンパンジーH1およびH2のプラスミドDNAおよびMVA免疫化は麻酔下で脚筋肉の筋肉内免疫化により実施された。これらのチンパンジー免疫化には、アジュバントとして15マイクログラムのヒトGM−CSFがプラスミドDNAと同時に投与された。チンパンジーへの組換えMVA投与は獣医監督下での筋肉内免疫化によるものであった。組換えヒトGM−CSFはSandoz(Camberley,UK)から購入された。遺伝子銃を用いるプラスミドDNA免疫化のため、DNAは金粒子上に沈殿させた。皮内送達のためには、二つの異なった型の遺伝子銃が使用された、AcellおよびOxford Bioscience装置(PowderJect Pharmaceuticals,Oxford,UK)。
CD8+ T細胞は示されたペプチドエピトープおよびMiyaharaら(1993)により記載されているようなELISPOTアッセイを用いてインビトロ再刺激なしで免疫化マウスの脾臓で定量された。簡単に説明すると、96−ウェルニトロセルロースプレート(Miliscreen MAHA,Millipore,Bedford UK)を15μg/mlの抗マウスインターフェロン−γモノクローナル抗体R4(EACC)のリン酸緩衝化塩溶液(PBS)50μlで被覆した。4℃で一夜インキュベーションした後、ウェルを一度PBSで洗い、10%FCSを含む100μlのRPMIを用いて室温で1時間ブロックした。免疫化したマウスからの脾細胞は1x107細胞/mlで再懸濁し、二重に抗体被覆ウェルへ加え、連続的に希釈した。ペプチドは1μg/mlの最終濃度で各々のウェルへ加えられた。ペプチドを加えない追加のウェルはインターフェロン−γ分泌のペプチド依存性のための対照として使用された。5%CO2中、37℃で12−18時間インキュベートした後、プレートをPBSおよび水で6回洗浄した。ウェルは次に1μg/mlのビオチニル化抗マウスインターフェロン−γモノクローナル抗体XMG1.2(Pharmingen,CA,USA)PBS溶液と、室温で3時間インキュベートした。PBSで洗浄後、1μg/mlのストレプトアビジン−アルカリ性ホスファターゼポリマー(Sigma)溶液50μlを室温で2時間加えた。50μlのアルカリ性ホスファターゼ結合体基質溶液(Biorad,Hercules,CA,USA)を加えることによりスポットが現れた。スポット出現後、水で洗うことにより反応を停止させた。スポットの数が立体顕微鏡の助けを借りて決定された。
CTLアッセイはAllsoppら(1996)により記載されているように、クロム標識標的細胞およびエフェクター細胞として培養マウス脾臓細胞を使用して実施された。チンパンジーまたはマカク細胞を用いるCTLアッセイは、標的細胞としてEBV−形質転換自己由来チンパンジーまたはマカクB細胞株を使用し、Hillら(1992)によりヒトCTLの検出で記載されているように実施された。
マウスは記載されているように(Lanarら1996)200μlのRPMIに含まれる2000(BALB/c)または200(C57BL/6)のP.ベルゲイANKA株スポロゾイトの静脈内接種により攻撃誘発させた。これらのスポロゾイトは感染マウスへ給餌後、20−25日間18℃に維持されたアノフェレス ステフェンシ蚊の唾液腺から切開された。血液期マラリア感染(免疫化の失敗を示している)は、攻撃誘発の5−12日後に採られたギームザ染色血液塗抹標本中のP.ベルゲイの環形出現を観察することにより検出された。
チンパンジーは20,000のアノフェレス ガンビエ蚊の唾液腺から切開されたNF54株熱帯熱マラリア原虫スポロゾイトの麻酔下での静脈内接種により攻撃誘発させた。これらのチンパンジーからの血液試料は、末梢血中の低レベルの熱帯熱マラリア原虫寄生虫の出現を検出するために攻撃誘発の5日後から毎日、顕微鏡および寄生虫培養により試験された。
マウスは200μlのPBSに含まれる1x105P815細胞の静脈内接種により攻撃誘発させた。動物は生存率がモニターされた。
マウスはインフルエンザウイルスA/PR/8/34の100ヘマグルチニン化単位(HA)の静脈内接種により攻撃誘発させた。攻撃誘発後、動物は毎日秤量され、生存率がモニターされた。
Mamu−A*01−重鎖およびβ2−ミクログロブリンから成る四量体複合体はOggら(1998)により記載されているように作製された。リーダーを持たないMamu−A*01 MHCクラスI重鎖の細胞外部分は5’プライマーMamuNdel:5’−CCT GAC TCA GAC CAT ATG GGC TCT CAC TCC ATG[配列ID番号:74]および3’プライマー:5’−GTG ATA AGC TTA ACG ATG ATT CCA CAC CAT TTT CTG TGC ATC CAG AAT ATG ATG CAG GGA TCC CTC CCA TCT CAG GGT GAG GGG C[配列ID番号:75]を用いてcDNAからPCR−増幅された。前者のプライマーはNdeI制限部位を含み、後者はHindIII部位を含んでおり、ビオチニル化酵素BirA基質ペプチドをコードしている。PCR生成物はNdeIおよびHindIIIで切断され細菌発現ベクターpGMT7ポリリンカーの同じ部位へ結合された。リーダーを持たないβ2−ミクログロブリンをコードしているアカゲザル遺伝子はプライマーB2MBACK:5’−TCA GAC CAT ATG TCT CGC TCC GTG GCC [配列ID番号:76]およびB2MFOR:5’−TCA GAC AAG CTT TTA CAT GTC TCG ATC CCA C[配列ID番号:77]を用いてcDNAからPCR−増幅され、同様pGMT7のNdeIおよびHindIII部位にクローン化された。両方の鎖とも大月易菌株BL−21で発現され、封入体から精製され、ペプチドCTPYDINQM[配列ID番号:54]の存在下で再び折り畳まれ、BirA酵素(Avidity)を用いてビオチニル化され、およびFPLCおよびmonoQイオン交換カラムで精製された。ビオチニル化され、再び折り畳まれたMHC−ペプチド複合体の量は、単量体複合体がコンホメーション感受性モノクローナル抗体W6/32により最初に捕捉され、アルカリ性ホスファターゼ(AP)−結合ストレプトアビジン(Sigma)、続いてのAPの比色基質により検出されるELISAアッセイにより見積もられた。四量体複合体の形成はフィコエリスリン(PE)−結合ストレプトアビジン(ExtrAvidin;Sigma)を再折り畳まれ、ビオチニル化された単量体へ4:1のMHC−ペプチド:PE−ストレプトアビジンモル比で加えることにより誘導された。複合体は暗所で4℃にて保存された。これらの四量体は免疫したマカクの末梢血リンパ球(PBL)中のMamu−A*01/gag特異的CD8+ T細胞の頻度を分析するために使用された。
マウスにおける免疫原性研究
プラスモジウム ベルゲイおよびプラスモジウム ヨエリイのスポロゾイト周囲(CS)タンパク質中のエピトープに対するCTL誘導の以前の研究は異なった送達系による種々のレベルのCTL誘導を示している。プラスミドDNA(Sedegahら1994)、複製するワクシニアウイルスで追加刺激されたインフルエンザウイルス(Liら1991)、アデノウイルス(Rodriguesら1997)および粒子送達系(Schodelら1994)で部分保護が報告されている。50マイクログラムのCSタンパク質をコードしているプラスミドによる筋肉内でのマウス免疫化は、1回注射後にこれらのマウスの脾臓中に中程度のCD8+細胞およびCTL活性を生み出した(図3、4)。
免疫原性および攻撃誘発
組換えMVAで追加刺激できる、皮内にプラスミドをDNA送達するための遺伝子銃の使用およびそれによる免疫応答の初回刺激が研究された。BALB/cマウスは以下の投与法で免疫された:
I)2週間隔での3回のpTH.PbCSP(免疫化当たり4mg)による遺伝子銃免疫化
II)2回の遺伝子銃免疫化に続く2週間後のMVAi.v.
III)1回の筋肉内DNA免疫化に続く2週間後のMVAi.v.
三つの免疫化法の免疫原性はELISPOTアッセイを使用して分析された。
特異的T細胞の最も高い頻度は2回の遺伝子銃免疫化に続くMVAi.v.追加刺激および1回の筋肉内DNA免疫化に続くMVAi.v.追加刺激で観察された(図6)。
二つのCTLエピトープ(P.ベルゲイから誘導されたSYIPSAEKI[配列ID番号:67]およびHIVから誘導されたRGPGRAFVTI[配列ID番号:68])によるBALB/cマウスでの前記の追加刺激効果が普遍的な現象であるかどうかを追求するため、2組の実験が実施された。インフルエンザ核タンパク質に対するCTL応答は5つの近交マウス系で研究された。最初の実験において、インフルエンザ核タンパク質から誘導された公表されているマウスCTLエピトープで研究された(表3参照)。三つの異なったH−2ハプロタイプ、BALB/cおよびDBA/2(H−2d)、C57BL/6および129(H−2d);CBA/J(H−2d)のマウスが使用された。動物の一つの組はインフルエンザ核タンパク質をコードしているプラスミドV1J−NPを用いて2週間隔で2回免疫された。同一動物の別の組はV1J−NPで初回刺激され、2週間後にインフルエンザウイルスNPを発現している106ffuのMVA.NPを用いて静脈内で追加刺激された。個々のマウスのCTLレベルはペプチド再刺激された脾臓細胞の51Cr放出アッセイで決定された。図7に示されているように、DNA初回刺激/MVA追加刺激免疫化法が分析されたすべてのマウス系においてより高いレベルの溶解を誘導し、2回のDNA注入よりも優れている。
プラスミドDNA初回刺激免疫応答に対するMVA追加刺激効果はさらに異なった抗原および異なった近交マウス系を用いて研究された。異なった系統のマウスは2回のDNA免疫化を用いる異なった抗原で免疫され、DNA/MVA免疫化と比較された。使用された抗原は大腸菌β−ガラクトシダーゼ、マラリア/HIVエピトープ列、マウス腫瘍エピトープ列および熱帯熱マラリア原虫TRAPであった。二つのDNA免疫化を比較すると、DNA初回刺激/MVA追加刺激法がすべての異なったマウス系および試験された抗原の組み合わせでより高いレベルのCTLを誘導した(図8)。
マラリアが風土病である地域に住んでいるヒトは連続的にスポロゾイト接種に曝されている。マラリア特異的CTLはこれらの自然に曝されている個体においては低レベルであることが観察されている。低レベルのスポロゾイト誘導CTL応答がMVAにより追加刺激できるかどうかという問題を扱うため、BALB/cマウスを照射した(マラリア感染を防ぐため)P.ベルゲイスポロゾイトで免疫し、MVAで追加刺激した。最後の免疫化から2週間後、脾臓細胞を再刺激して溶解活性を試験した。50または300+500スポロゾイトによる2回の注射では非常に低レベルまたは検出できないレベルの溶解しか誘導されなかった。
図9はスポロゾイト初回刺激CTL応答がMVAにより有意に追加刺激されることを示している。マウスはAにおいては照射されたスポロゾイトの2回の低用量(50+50)で、Bにおいてはスポロゾイトの2回の高用量(300+500)で免疫された;マウスはDでは低用量のスポロゾイト初回刺激、Eでは高用量のスポロゾイト初回刺激に続いてMVA.PbCSPで追加刺激された。MVA.PbCSPでの免疫化後のCTL応答はCに示されている。
初回刺激−追加刺激免疫化法が初回刺激薬物としてプラスミドDNAおよび組換えTy−VLPを使用して例示されてきた。ここでは初回刺激薬物として非複製アデノウイルスを用いる例が提供される。大腸菌β−ガラクトシダーゼを発現する複製欠陥組換えアデノウイルス(Adeno−GAL)が使用された。BALB/cマウス群はプラスミドDNAに続いてMVAでまたはアデノウイルスに続いてMVAで免疫された。使用されたすべての抗原送達系は大腸菌β−ガラクトシダーゼをコードしていた。プラスミドDNAまたはアデノウイルスによるCTL応答の初回刺激およびMVAによる追加刺激は類似のレベルのCTLを誘導した(図10)。
複製適性が違う異なった株はDNA初回刺激CTL応答を追加刺激する能力が異なっているかどうかを決定するために、組換えワクシニアウイルスの異なった株を使用して初回刺激−追加刺激法が試験された。MVAおよびNYVACのような複製欠陥組換えワクシニアウイルスによる追加刺激は、同量の複製適性WRワクシニアウイルスによる追加刺激後のCTL応答と比較してより強いCTL応答の誘導を起こした(図11)。
組換えカナリアポックスウイルス(rCPV)またはニワトリポックスウイルス(rFPV)は以前に記載されているシャトルベクターを使用して作製された(TaylorらVirology 1992,187:321−328およびTay1orらVaccine 1988,6:504−508)。これらのシャトルベクターのための戦略は、CPVまたはFPVゲノムから誘導された配列から成る二つの隣接領域間のワクシニア特異的プロモーターの後ろに問題とするタンパク質をコードしている遺伝子を挿入することである。これらの隣接領域は必須ウイルス遺伝子内への挿入を避けるように選択される。組換えCPVまたはFPVは許容トリ細胞株(即ち、初代ニワトリ胎児線維芽細胞)でのインビボ組換えにより発生させた。任意の抗原またはエピトープ列のタンパク質配列がニワトリポックスまたはカナリアポックスウイルスを用いて発現できる。組換えCPVまたはFPVは抗原特異的抗体を用いて、または組換え遺伝子内へ抗体エピトープを含ませ、問題とするタンパク質の発現で特徴付けられる。組換えウイルスは初代CEFで増殖させる。免疫応答は、材料および方法で説明したようなプラスミドDNAを用いて初回刺激される。このプラスミドDNA初回刺激免疫応答は、静脈内に、皮内に、または筋肉内に接種された107ffu/pfuのrCPVまたはrFPVにより追加刺激される。CD8+ T細胞応答がモニターされ、攻撃誘発は前記のように実施された。
マウスにおけるマラリア攻撃誘発の研究
CD8+ T細胞応答の誘導レベルの保護的効能を評価するため、免疫化BALB/cまたはC57BL/6マウスを2000または200のP.ベルゲイスポロゾイトの静脈注射により攻撃誘発した。このことはスポロゾイトによる肝臓細胞の感染を導く。しかしながら、肝臓内寄生虫に対する十分に強いTリンパ球応答の存在下では生きている寄生虫は肝臓を離れないであろうので、血液期寄生虫は観察されないであろう。攻撃誘発の5−12日後、攻撃誘発したマウスからの血液薄層は顕微鏡により寄生虫が評価された。
組換えMVAの静脈内注射はヒト免疫化には好適な経路ではなく、大量免疫化には実現不可能である。それ故、MVA追加刺激の異なった経路での免疫原性および保護効能が試験された。マウスはプラスミドDNAによりi.m.で初回刺激された。2週間後、それらは以下の経路で投与されたMVAで追加刺激された:静脈内(i.v.)、皮下(s.c.)、腹腔内(i.p.)、筋肉内(i.m.)および皮内(i.d.)。この追加刺激2週間後、ペプチド特異的CD8+ T細胞はELISPOTアッセイにより決定された。最も高いレベルを誘導した最も有効な経路はMVAのi.v.およびi.d.接種であった。他の経路は中程度から弱い応答しか与えなかった(図12)。
動物は筋肉内プラスミドDNA注射により初回刺激され、2週間後、示された組換えMVA(106ffu/マウス)が示された経路により投与された。マウスは最後の免疫化から16日後に2000のP.ベルゲイスポロゾイトで攻撃誘発し、攻撃誘発の8および10日後に血液期寄生虫血症がスクリーニングされた。エピトープはポリペプチド列HMを示している。
遺伝子銃送達は例えばEisenbraunら DNA Cell Biol.1993,12:791−797およびDeganoら Vaccine 1998,16:394−398に詳細に記載されている。
C57BL/6マウスはP.ベルゲイスポロゾイト攻撃誘発に対して非常に感受性である。C57BL/6マウスは前赤血球抗原PbCSPおよびPbTRAPの両方によるDNA−MVA初回刺激−追加刺激法を用いて免疫化され、マウス当たり200または1000の感染性スポロゾイトで攻撃誘発した(200はこの系統での感染を誘導するために必要とされる量の2倍以上に相当する)。200のスポロゾイトで攻撃誘発された10匹のマウスすべてが滅菌免疫性を示した。1000のスポロゾイトで攻撃誘発された群でさえも、60%のマウスが保護された(表12)。天然のままのC57BL/6マウスはすべてスポロゾイト攻撃誘発で感染された。
マウスのさらに二つの疾患モデルにおけるDNA−初回刺激/MVA−追加刺激法の保護効能
免疫原性研究に続いて、DNA−初回刺激/MVA−追加刺激法の保護効能が二つの追加のマウス攻撃誘発モデルで試験された。二つの攻撃誘発モデルはP815腫瘍モデルおよびインフルエンザAウイルス攻撃誘発モデルであった。両方のモデル系においてCTLが保護を仲介していることが示された。
DBA/2マウス群(n=10)はDNA、続いてHMエピトープ列の腫瘍エピトープ列を発現するMVAの組み合わせで免疫された。最後の免疫化から2週間後、マウスは105のP815細胞の静脈内投与で攻撃された。この攻撃後、マウスは定期的に腫瘍関連徴候および生存率でモニターされた。
BALB/cマウス群はプラスミドDNAによる3回の遺伝子銃免疫化、2回の筋肉内プラスミドDNA注射、1回のi.m.DNA注射に続く1回のMVA.NP追加刺激i.v.または2回の遺伝子銃免疫化に続く1回のMVA.NP追加刺激i.v.で免疫された。プラスミドDNAおよび組換えMVAはインフルエンザウイルス核タンパク質を発現した。最後の免疫化から2週間後、マウスに鼻腔内で100HAのインフルエンザA/PR/8/34ウイルスを攻撃誘発した。動物は攻撃誘発後毎日生存でモニターした。

非ヒト霊長類での免疫原性研究非ヒト霊長類での初回刺激追加刺激法の免疫原性および保護効能
マウスで観察されたDNA初回刺激/MVA追加刺激法の強力な免疫原性が、霊長類でも強力な免疫原性を誘導することを示すために、本方法がマカクにおいて試験された。ワクチンはHIVおよびSIV配列から誘導されたCTLエピトープの列から成っており、プラスミドDNAまたはMVAにおいては各々DNA.HおよびMVA.Hと表示されている。ポリエピトープ列中での定義されたCTLエピトープの使用はマカクにおけるSIV特異的CTLを試験することを可能にする。抗原性ペプチドのMHCクラスI制限のため、マカクはそのMHCクラスIハロタイプでスクリーニングされ、Mamu−A*A01陽性動物が記載された実験に選択された。
三匹の動物(CYD,DIおよびDORIS)が下記の本免疫化法で免疫された
0週 DNA(8μg、i.d.、遺伝子銃)
8週 DNA(8μg,i.d.、遺伝子銃)
17週 MVA(5x108pfu、i.d.)
22週 MVA(5x108pfu、i.d.)。
3匹の異なったマカク(CYD、DIおよびDORIS)からのPBMCは18、19および23週に単離され、インビトロでペプチドCTPYDINQM[配列ID番号:54]で再刺激された。ペプチドCTPYDINQM[配列ID番号:54]による2回の再刺激後、培養物はペプチド−パルス自己由来標的細胞に対するそれらの溶解活性が試験された。強いCTL活性が観察された。
チンパンジーでの免疫原性および攻撃誘発の研究
プラスミドDNAによる最初の免疫化および続いての組換えMVAによる免疫化の同様な方法が、より高等な霊長類において熱帯熱マラリア原虫マラリアに対して有効であることを示すため、2匹のチンパンジーで免疫化および攻撃誘発の研究が実施された。チンパンジーH1はイントロンAなしでCMVプロモーターから熱帯熱マラリア原虫TRAPを発現するプラスミド(CMV−TRAP)の500μgによる最初の免疫化を受けた。チンパンジーH2は、熱帯熱マラリア原虫LSA−1遺伝子のC末端部分を発現するCMV−LSA−1を同量受けた。両方のチンパンジーは続いての2ヶ月かけてさらに3回の免疫化を受けたが、その各々の免疫化は三つのプラスミドで行われた。H1は前記のCMV−TRAPに、イントロンAを持つCMVプロモーターを使用してTRAPを発現するpHT−TRAPが加えられ、より高い発現レベルが導かれた。H1はまたRSVプロモーターからLSA−1のC末端部分を発現するRSV−LSA−1も受けた。H2は第二、第三および第四の免疫化時にCMV−LSA−1、pTH−LSA−1およびRSV−TRAPを受けた。用量はいつも各々のプラスミド500μgであった。
これらの実施例はヒトマラリア感染の齧歯動物モデルにおける高レベルの保護的CD8+ T細胞を誘導する、マラリアに対する免疫化のための新しい方法を示している。また、サブユニットワクチンを使用するスポロゾイト攻撃誘発に対する先例のない完全保護も示されている(CSエピトープ含有ワクチンによるDNA初回刺激およびMVA追加刺激を用いて表6では36匹のマウスの内36匹が保護された)。DNA初回刺激/MVA追加刺激法を用いた保護的免疫応答の誘導がウイルス感染インフルエンザAモデルおよび癌(P816腫瘍モデル)の二つの追加のマウスモデルで示された。ヒトでの使用のためのワクチンにより重要であるのは、この免疫化法はまた霊長類のCD8+ T細胞に対して高度に免疫原性であることである。強いSIV−gag特異的CTLが、エピトープ列を発現するプラスミドDNAおよびMVAにより3匹のマカクの内の3匹で誘導された。誘導されたレベルはSIV感染動物で観察されたレベルに匹敵する。チンパンジー研究からのデータは、同じ免疫化法がより高等な霊長類で熱帯熱マラリア原虫に対する強いCD8+ Tリンパ球応答を誘導でき、熱帯熱マラリア原虫スポロゾイト攻撃誘発に対する保護のいくつかの証拠があることを示している。

Claims (1)
- 少なくとも一つの標的抗原に対する保護的CD8+ T細胞免疫応答を発生するためのキットであって、以下:

(ii)初回刺激組成物のCD8+ T細胞エピトープと同一である少なくとも一つのCD8+ T細胞エピトープを含む標的抗原の一つまたはそれ以上のCD8+ T細胞エピトープ源、ここでCD8+ T細胞エピトープ源は非複製または複製欠陥組換えポックスウイルスベクターである、ならびに医薬品として受容可能な担体から成る追加刺激組成物;
からなり、ただし、(i)のエピトープ源がウイルスベクターであるならば、(ii)におけるウイルスベクターは異なったウイルスから誘導される、キット。
