Claims (26)

細胞で機能し、セレクターコドンを認識する直交tRNA(O−tRNA)と;
p−アセチル−L−フェニルアラニンと前記O−tRNAと配列番号18〜20から選択されたアミノ酸配列を含むポリペプチドとを含む翻訳系の効率の少なくとも50%の効率でO−tRNAをp−アセチル−L−フェニルアラニンで優先的にアミノアシル化する直交アミノアシルtRNAシンテターゼ(O−RS)を含み;
更に前記核酸中の前記少なくとも1個のセレクターコドンの所定位置が前記該当蛋白質中のp−アセチル−L−フェニルアラニンの前記特定位置に対応するよう該当蛋白質の翻訳中に該当蛋白質の特定位置にp−アセチル−L−フェニルアラニンを組込むことにより、特定位置にp−アセチル−L−フェニルアラニンを組込んだ該当蛋白質を生産する段階を含む前記方法。 - O−RSが配列番号18〜20のいずれか1種又はその保存変異体を含むアミノ酸配列を含む請求項22に記載の方法。
- O−tRNAが配列番号21に記載のポリヌクレオチド配列又はその相補的ポリヌクレオチド配列を含むか又はこれによりコードされる請求項22に記載の方法。
- 細胞が非真核細胞である請求項22に記載の方法。
- 非真核細胞が大腸菌細胞である請求項25に記載の方法。
