Abstract
Изобретение относится к слитому белку, обладающему in vivo повышенной активностью эритропоэтина. В слитом белке пептидный фрагмент карбоксильного конца тромбопоэтина аминокислоты 337-353 слит с карбоксильным концом эритропоэтина человека. Изобретение также включает нуклеиновую кислоту, кодирующую слитый белок, и способ его получения. Преимущество изобретения заключается в том, что слитый белок имеет значительно увеличенный период полужизни in vivo вследствие повышенного содержания углеводов без потери активности, присущей эритропоэтину, и не вызывает никакой антигенности в случае применения в организме человека. 3 с. и 2 з.п.ф-лы, 5 ил.

Claims (5)
1. Слитый белок, обладающий in vivo повышенной активностью эритропоэтина (ЕРО) человека, в котором карбоксиконцевой пептид (СТР), содержащий, по меньшей мере, аминокислоты 337-353 тромбопоэтина (ТРО), слит с ЕРО человека по карбоксильному концу ЕРО человека.
2. Слитый белок по п.1, в котором карбоксиконцевой пептид имеет аминокислотную последовательность SEQ ID NO:2.
3. Слитый белок по п.1, в котором карбоксиконцевой пептид включает в себя аминокислоты ТРО в положениях 176-353, последовательность которых приведена в SEQ ID NO:1.
4. Нуклеиновая кислота, кодирующая слитый белок по любому из пп.1-3.
5. Способ получения слитого белка по любому из пп.1-3, предусматривающий культивирование линии клеток-хозяев, трансформированных рекомбинантным вектором, содержащим нуклеиновую кислоту по п.4.
