Abstract
【解決手段】 細胞を保持するための細胞保持キャビティ12を複数有し、前記細胞保持キャビティの底面20は、細胞接着性を示す1つの接着性領域30と、当該接着性領域30を囲み、細胞非接着性を示す非接着性領域32と、を含む細胞組織体マイクロチップとする。

Description
次に、本マイクロチップ1を用いて細胞組織体を形成させた実施例について説明する。
本実施例においては、本マイクロチップ1の基板10として、ポリメチルメタクリレート製の平板(24mm×24mm、厚さ200μm)を用いた。すなわち、このポリメチルメタクリレート平板の表面の一部に、10mm角の矩形範囲にわたって、マシニングセンタ(卓上型NC微細加工機、株式会社ピーエムティー製)を用いた穿孔加工処理を施すことにより、直径300μmの円形の貫通孔を約1000個形成した。この円形貫通孔は、互いにその中心間距離が400μmとなるように規則的に配置した。
本実施例においては、細胞として、ラットの肝臓から公知のコラゲナーゼ灌流法を用いて調製した初代肝細胞を用いた。ここで、この肝細胞の調製法について簡単に説明する。すなわち、まず、7週齢のウィスター系ラット(体重250g)の門脈(肝臓につながる血管の一つ)にカニューレを挿入し、当該カニューレから肝臓に所定の組成の脱血液を導入した後、コラゲナーゼ(和光純薬社製)を0.5mg/mLの濃度で溶解した消化液を灌流し、肝臓内における肝細胞同士の結合等を切断する消化処理を行った。そして、この消化処理された肝臓を摘出し、メスやピペット等を用いた分散処理と培養液等を用いた洗浄処理により、個々の細胞が分散した単離状態の肝細胞を得た。
このようにして得た肝細胞を、5.0×105個/mLの密度となるように培養液に分散した。そして、ポリスチレン製の培養容器(直径35mm、ファルコン社製)の底部に上記滅菌処理後の本マイクロチップ1を載置し、当該本マイクロチップ1上に、この細胞分散液の一部を2.0mL加え、5%炭酸ガス、95%空気の雰囲気下、37℃にて静置状態で培養を行った。

- 細胞を保持するための細胞保持キャビティを複数有し、
前記細胞保持キャビティの底面は、細胞接着性を示す1つの接着性領域と、当該接着性領域を囲み、細胞非接着性を示す非接着性領域と、を含む、
ことを特徴とする細胞組織体マイクロチップ。 - 前記接着性領域は、前記細胞保持キャビティ底面の中央付近に形成される、
ことを特徴とする請求項1に記載の細胞組織体マイクロチップ。 - 前記細胞保持キャビティ底面の径は、前記細胞の径の2〜50倍の範囲である、
ことを特徴とする請求項1又は2に記載の細胞組織体マイクロチップ。 - 前記接着性領域には、生体から取得され若しくは合成された細胞接着性物質又はこれらの誘導体が固定されている、
ことを特徴とする請求項1乃至3のいずれか一項に記載の細胞組織体マイクロチップ。 - 前記非接着性領域には、生体から取得され若しくは合成された細胞非接着性物質又はこれらの誘導体が固定されている、
ことを特徴とする請求項1乃至4のいずれか一項に記載の細胞組織体マイクロチップ。 - 前記接着性領域に、前記細胞から形成された細胞組織体が接着している、
ことを特徴とする請求項1乃至5のいずれか一項に記載の細胞組織体マイクロチップ。 - 請求項1乃至5のいずれか一項に記載の細胞組織体マイクロチップの前記細胞保持キャビティ内において細胞を培養し、前記接着性領域において細胞組織体を形成させる、
ことを特徴とする細胞組織体形成方法。 - 前記細胞を、前記各細胞保持キャビティあたり2〜1.5×105個の範囲で培養する、
ことを特徴とする請求項7に記載の細胞組織体形成方法。
