Abstract
Description

。従って、GPVIを組換え形態で提供し、かつ直接的な治療標的としてのその
効力、または小さな化合物、特に、自然のコラーゲン−血小板相互作用を阻害ま
たは阻止する能力を有する化学合成化合物または化学合成可能な化合物、をスク
リーニングするためのツールとしてのその効力を示すことは本発明の目的であっ
た。

管腔拡張術(PTCA)などの手術的処置のときに適用することができると考え
られる。組換えGPVIフラグメントの利点は、現在の処置法と比較した場合、
フラグメントが、GPIIb−IIIaアンタゴニストによる凝集などの血小板
活性化後の事象を抑制することによるのではなく、むしろ血小板の活性化を妨げ
るか、または血小板の活性化を低下させることによって、より早い段階で作用す
るということである。従って、より少ない量の血小板顆粒内容物が放出される。
これには、外傷修復だけでなく、平滑筋遊走による血管壁の再構築、およびアテ
ローム性動脈硬化症の一因であることが知られている単球などの食細胞の引き寄
せにも関与している増殖因子およびケモカインが含まれる。GPVIに対するヒ
ト化マウスモノクローナル抗体のFabフラグメントは、上記のような類似する
適用で血小板表面のGPVIを阻止するために類似した効果で使用することがで
きる。
つコラーゲン−血小板相互作用の阻害剤である治療化合物を開発するために使用
され得る(例えば、コンビナトリアルライブラリーの)小さな分子をスクリーニ
ングするために、コラーゲンに対する結合アッセイにおいて使用することができ

血小板の老化および機能性のマーカーとしての適用である。若い血小板は、一般
に、老化した血小板よりも活性であり、かつ機能的であると考えられる。若い血
小板は、GPVIに特異的なヘビ毒液C型レクチンのコンブルキシンに結合し、
かつコンブルキシンにより活性化され、そして血小板は老化するに従って、その
結合および活性化度はともに低下する。これは、循環における血小板の活性化ま
たは損傷によるGPVIにおけるタンパク質分解的または立体配座的な変化ある
いはFcγとのその会合のいずれかのためであり得る。これは、医学的に管理さ
れている患者ならびに健常者における血小板の老化プロファイルおよび機能プロ
ファイルを測定するための有用なパラメータであり得る。血小板の老化プロファ
イルは、骨髄または免疫系を冒す多くの疾患において変化し、そのより良好な決
定方法が得られる場合には重要な診断基準になり得る。例えば、血小板の増大し
た代謝回転が関与する疾患患者はより多くの若い血小板を示し、これに対して、
化学療法または放射線処置を受けている患者は一様に老化した集団を示す。従っ
て、そのような老化プロファイルは、処置の精密なモニターリングに使用するこ
とができる。正常である健康な集団では、老化プロファイル分布について、そし

000pfuをスクリーニングすることにより、4個の陽性pfuが得られた。
3つは、制限酵素SalIまたはEcoRIで切断しても1350bpの挿入断
片を有し、IgGスーパーファミリーに属した。4個目は、SalI消化による
4.6kbの挿入断片を有し、EcoRIで処理したときにそれぞれ2300b
pおよび1300bpの2つのフラグメントをもたらした。そのDNAは、GP
VIのアミノ酸配列決定に由来する10個のペプチドに対する配列をコードして
いたが、アミノ末端に達しないところで停止していた。開始メチオニンまたはリ
ーダー配列を見出すことができなかったが、Alu配列で終了する、以前に配列
決定された非リーディングフレームDNAの2000bp以上が存在していた。
5’末端RACE実験を、それらのペプチドの配列によって裏付けられたGPV
I配列の一部分内に位置するプライマーを用いて血小板のポリA RNAに対し
て行った。4番目のクローンの配列における278bpを含む348bpのフラ
グメント、および最初のメチオニンを含む14アミノ酸に対応する1987bp
に由来する新しい70bpが見出され、その後、確立されたGPVI配列に達し
た。従って、合計で1249bp、すなわち、開始コドンの上流の25bpの5
’配列、リーダー配列を含み、339アミノ酸を有するタンパク質をコードする
1017bpのオープンリーディングフレーム、および停止コドンを含む207
bpの3’領域を含有するcDNAを配列決定することができた。
血小板mRNAを用いたRACEを使用して、ヒト骨髄のcDNAライブラリー
からクローン化され、そして配列決定された。1017bpのオープンリーディ
ングフレームは339個のアミノ酸をコードし、非翻訳3’領域を有する。アミ
ノ酸配列の疎水性分析により、2つの推定的な膜貫通ドメイン、推定的な20ア
ミノ酸シグナル配列、および成熟タンパク質の残基247と残基265との間の
19アミノ酸ドメインが存在することが明らかにされた。配列およびそのアミノ
酸翻訳を図2および図1に示す。GenBankの検索において見出された最も
類似する分子のアミノ酸配列と比較することにより、GPVIは免疫グロブリン
スーパーファミリーに属すること、およびその細胞外ドメインは、2つのジスル
フィド架橋により形成された2つのIgC2ドメインループを含有することが明
瞭にされる。GPVIは、N末端を外側に有する膜横断タンパク質クラスの分子
であり、膜を1回横断する。最も近縁の分子は、阻害タイプおよび活性化タイプ
の両方を含有するナチュラルキラー受容体クラスに属する。GPVIは、その機
能によるだけでなく、阻害クラスとは異なり、ITIM配列をその細胞質ドメイ
ンに含有しないために活性化サブクラスに属することは明らかである。GPVI
は、リン酸化に関与し得るチロシン残基を1つも含有しない。このドメインには
数個のトレオニン残基およびセリン残基が存在するが、それらは、キナーゼのコ
ンセンサス配列に対する基準を何ら満たさない。NK受容体の活性化クラスと同
様に、GPVIは、Fcγサブユニットとの複合体形成に関与する膜横断ドメイ
ンの3番目のアミノ酸としてアルギニン残基を含有する。細胞質ドメインは、こ
のファミリーの他のメンバーの細胞質ドメインに対する(膜のすぐ下流の領域に
おける)わずかな類似性のみを示す51個のアミノ酸を含有する。このことは、
GPVIにおけるこのドメインは、それ以外のファミリーメンバーとは異なるタ
イプの細胞質分子と会合し得ることを示唆している。GPVIは、1個の推定的
なN−グリコシル化部位をAsn69に含有するだけである。しかし、Igドメ
インのβシートが終了した後の膜のすぐ上流のドメインは、GPIbαおよびG
PVにおいて見出されているようなO−グリコシル化部位を提供し得るトレオニ
ン残基およびセリン残基が多い。このO−グリコシル化の主要な機能は、その嵩
張ったリガンドとの相互作用を容易にするために血小板表面から十分に広がった
受容体構造体を提供することであると考えられる。GPVIはシアロ糖タンパク
質としてより以前には明らかにされていたので、理論的なアミノ酸質量(37k
Da)とゲル電気泳動で決定された質量(65kDa、還元)との分子量の差は
、このグリコシル化のためであるに違いない。

るペプチド保有HLA抗原の受容体部位とともに鋭角を形成していることが示さ
れた。HLAペプチド結合部位の構造をコラーゲンの構造と直接比較することに
より、それが含有するHLA結合部位およびペプチドの多数のα−らせん構造が
コラーゲンの三重らせん構造に非常に類似しているので、直ちに、なぜこれらの
受容体が共通の起源を有するかが示唆される。ナチュラルキラー受容体は、ナチ
ュラルキラー細胞が調べている細胞における2つの離れたHLA部位を認識する
2つの受容体による二量体化機構によって作用することが推定される。おそらく
は、この二量体化は、受容体のクラスに依存した活性化機構または失活化機構の
一部である。GPVIの場合、2つのGPVI分子が1つのFcγと会合する可
能性があり得る。これは、Fcγダイマーの各モノマーが認識部位を有するから
である。しかしながら、化学量論は未だ知られておらず、そしてコラーゲンの構
造、GPVIおよびコンブルキシンを介して作用するコラーゲン様ペプチドとの
構造に基づいて、シグナルの強さが、一緒にクラスター化しているGPVI/F
cγ複合体の数に関連することは確からしい。このIgファミリーに属する他の
血小板受容体には、ICAM−2(CD102)およびPECAM(CD31)
が含まれる。

起源、ベクターの制限部位あるいは他のフラグメントまたは部分は、すべて市販
されているか、そうでなければ、一般に手に入れることができる。他の示唆が与

マウス抗体およびペルオキシダーゼ結合ヤギ抗ウサギ抗体、ウシ血清アルブミン
、Crotalus durissus terrificus毒液、コムギ胚
芽凝集素(wheat germ aggulutinin;WGA)、N−ヒ
ドロキシスクシンイミジルクロロホルマート活性化架橋4%ビーズ状アガロース
ならびにトリトンX−114をSigma Chemicals Co.(St
Louis、MO)から入手した。オクタノイル−N−メチルグルカミド(O
NMG)およびノナノイル−N−メチルグルカミド(NNMG)をOxyl C
hemie(Bobingen、ドイツ)から入手した。
たようにして血小板から単離した。簡単に記載すると、血小板(40の軟膜に由
来)を洗浄し、プロテアーゼ阻害剤の存在下、2%トリトンX−114中で溶解
した。トリトンX−114および水相を分離し、界面活性剤相を、セファロース
4Bに結合させたコムギ胚芽凝集素のカラムに負荷した。血小板糖タンパク質を
、10mM Tris HCl(pH7.4)、30mM NaCl、0.2%
オクタノイル−N−メチルグルカミド(ONMG)および2%N−アセチルグル
コサミンにおいて溶出した。透析および濃縮を行った後、糖タンパク質の溶液を
、N−ヒドロキシスクシンアミジル−p−ニトロフェニルクロロホルマート活性
化架橋4%ビーズ状アガロースに結合させたコンブルキシンのカラム(1mg/
ml)に負荷した。カラムを、4容量の10mM TrisHCl(pH7.4
)、30mM NaCl、0.2%ノナノイル−N−メチルグルカミド(NNM
G)で洗浄し、次いで4容量の10mM Tris HCl(pH7.4)、3
0mM NaClおよび2%NNMGで洗浄した。GPVIを、0.08%SD
Sを含む10mMのTris/HCl(pH7.4)で溶出した。溶液を濃縮し
、491型Prep Cell(BioRad、CA)を使用する8.5%ポリ
アクリルアミドの調製用ゲルに負荷した。調製用電気泳動を製造者の説明書に従
って非還元条件下で行った。GPVIは65kDaの単一バンドとして溶出した
。この画分をまとめ、Centricon−30(Amicon、Beverl
y、MA)で濃縮し、10mM Tris/HCl(pH7.4)および0.1
%ONMGに再懸濁した。
CおよびエンドプロテイナーゼAspN(Boehringer Mannhe
im、ドイツ)で消化した。得られた10個のペプチドを逆相HPLCで分離し
、120A型オンラインフェニルチオヒダントインアミノ酸分析計を備えたAp
plied Biosystem社の477A型パルス液相タンパク質シーケン
サで配列決定した。
cDNAライブラリーからの増幅− ヒト骨髄ライブラリー(Clonetech、Palo Alto、CA)に
由来するサンプル(1010pfu)(プラーク形成単位)を、4つの縮重プライ
マーの2つの組合せを使用して増幅した。プライマーの最終濃度は2μMであり
、dNTP濃度は200μMであり、2U/100μl反応物のAmpliTa
q Gold(Perkin Elmer、Rotkreuz、スイス)を使用
した。PCR条件は、37℃での5サイクル、その後、44℃での30サイクル
であった。5’TYATHCCNGCNATGAARMG3’のセンス19me
rおよび5’TTRTANARNGCRAAYTGRTC3’のアンチセンス2
0merにより、221bpのフラグメントが増幅され、これをBluescr
iptKS+(Stratagene、La Jolla、CA)にサブクロー
ン化し、T7Sequenase kit(Amersham、スイス)を使用
して配列決定した。
ブラリーのスクリーニング−221bpのフラグメントをプラスミドから切断し
、清浄化し、High Prime Labelling kit(Boehr
inger Mannheim、スイス)を使用してα32P−ATP(20MB
q/50μl、Hartmann Analytik、Braunschwei
g、ドイツ)で標識した。ヒト骨髄ライブラリーを製造者の説明書に従ってスク
リーニングした。陽性ファージを増殖させ、そのDNAを単離し、EcoRI部
位またはSalI部位のいずれかを使用してBlueScriptにサブクロー
ン化し、配列決定した。配列決定を、RACEのABSシステムを用いて行った
。血小板のポリA RNAを、以前に記載されたようにして調製した(Powe
r他、Cytokine、7、479〜482、1995)。逆転写(30μl
)を、5μgのポリA RNAを、プライマー5’TGAATGAGACGGT
CAGTTCAGC3’(20μM)、dNTP(1mM)、RNAsin(4
0U)、1X AMV緩衝液、および20UのAMV逆転写酵素とともに使用し
て45℃で20分間行い、その後、52℃で20分間行った。反応混合物を2μ
lの6N NaOHで65℃において30分間処理し、2μlの6N酢酸で中和
して、Centricon30(Amicon)で濃縮した。アンカーをApt
esおよびSiebertのプロトコル(BioTechniques、15:
890〜893、1993)に従って第1鎖DNAに連結した。ネスティッドP
CRを、アンカーに相補的なプライマーおよびプライマー5’TTGTACAG
AGCAAATTGGTC3’を使用して行い(35サイクル、55℃)、その
後、プライマー5’GACCAGAGGCTTCCGTTCTG3’を使用して
行った(53℃で30サイクル)。最も大きなバンド(350bp)を、それよ
りも小さいバンドからアガロース電気泳動で分離し、BlueScriptにサ
ブクローン化して、配列決定した。
するポリクローナル抗血清をウサギにおいて生成させた。IgGを、ウサギ抗G
PVI抗血清から、記載されたようにして精製した。Fabフラグメントを生成
させるための固定化パパイン(Pierce)によるIgGの消化を供給者の標
準的なプロトコルに従って行った。Fabフラグメントを、固定化プロテインA
(Sigma)カラムを使用して未消化IgGおよびFcフラグメントから分離
した。素通り部分を透析チューブに移し、固体のポリエチレングリコール20,
000を使用して濃縮し、20mMのHepes、140mMのNaCl、4m
MのKCl、pH7.4に対して徹底的に透析し、そして使用するまで4℃で保
存した。F(ab’)2フラグメントを、0.5M酢酸塩緩衝液(pH4.0)
中における37℃で18時間にわたるIgGのペプシン消化(1:50の酵素対
基質比(w/w))によって調製した。pHを希NaOHで7.4に調節し、サ
ンプルを20mMリン酸塩(pH7.4)に対して透析し、その後、F(ab’
)2フラグメントを、プロテインAクロマトグラフィーを使用して未消化IgG
およびFcフラグメントから分離した。素通り部分を透析チューブに移し、固体
のポリエチレングリコール20,000を使用して濃縮し、20mMのHepe
s、140mMのNaCl、4mMのKCl、pH7.4に対して徹底的に透析
し、小分けして−20℃で保存した。洗浄した血小板をトリトンX−114中で
溶解し、相分離を可溶物に対して行い、その後、トリトンX−114相と会合し
た膜糖タンパク質を、以前に記載されたようにしてコムギ胚芽凝集素−セファロ
ース4Bでのアフィニティークロマトグラフィーで分離した。GPVIは血小板
膜糖タンパク質プールの非常に小さな画分を示すので、本発明者らは、この受容
体を分離するために、ヘビC型レクチンのコンブルキシンの特異性を使用した。
セファロース4Bに結合させたコンブルキシンでのアフィニティークロマトグラ
フィーにより、65kDaのタンパク質が主生成物として得られた。しかし、そ
れよりも大きなMrおよびそれよりも小さなMrの両方の特徴付けられない物質
がGPVIと同時に溶出し、これらはカラムの徹底的な洗浄で除去できなかった
。8.5%ポリアクリルアミドでの調製用ゲル電気泳動を精製の最終ステップと
して加えた。GPVIを含有する画分をまとめ、これは再分析で単一バンドを示
した。精製GPVIを、コラーゲンによる血小板凝集を阻止するその能力につい
て試験した。わずかな阻害作用が、GPVI溶液の一部を血小板懸濁液に加えた
ときに認められた。しかし、混合物を血小板懸濁物に加える前にGPVIをコラ
ーゲンとプレインキュベーションすることによって、凝集は用量依存的に阻害さ
れた。これらの血小板は、新たなコラーゲンが添加されたときにもやはり凝集し
た。非還元条件下において、この単離されたタンパク質は62kDaのMrを有
し、還元条件下ではわずかに大きなMr(65kDa)に変化した。GPVIの
アミノ末端はブロックされていることが見出されたので、タンパク質を、配列が
得られる4個のペプチドおよび6個のペプチドをそれぞれもたらす酵素LysC
および酵素LysC/AspNで消化した。これらのペプチドをC4カラムでの
逆相HPLCで分離し、エドマン法を使用して配列決定した。これらのペプチド
のアミノ酸配列には、翻訳されたcDNA配列(図1)において下線が引かれて
いる。
