Abstract
Oppfinnelsen vedrører glykoprotein VI (GPVI), dets isolering, rensing og fremgangsmåter for rekombinant fremstilling. Spesielt vedrører oppfinnelsen anvendelsen av GPVI, fortrinnsvis rekombinant GPVI, ved behandling av forstyrrelser og patologiske følger korrelert direkte eller indirekte til blodkoagulasjonsforstyrrelser, slik som trombotiske og kardiovaskulære sykdommer. Det ekstracellulære rekombinante protein kan også anvendes til etablering av screeningsanalyser for å finne potensielle inhibitorer for det membranbundne GPVI, for å inhibere binding av henholdsvis trombocytter og blodplater til kollagen. Endringer i GPVI kan anvendes til å overvåke blodplatealder og eksponering for trombotiske og kardiovaskulære sykdommer.
Description
Oppfinnelsen vedrører glykoprotein VI (GPVI), dets isolering, rensing og fremgangsmåter for rekombinant fremstilling. Oppfinnelsen vedrører spesielt anvendelsen av GPVI, fortrinnsvis rekombinant GPVI, ved behandling av forstyrrelser og patologiske følger som er korrelert direkte eller indirekte til blodkoagulasjonsforstyrrelser, slik som trombotiske og kardiovaskulære sykdommer. Det ekstracellulære rekombinante protein kan også anvendes til å etablere screeningsanalyser for å finne potensielle inhibitorer for det membranbundne GPVI for å inhibere interaksjon mellom blodplater og kollagen. GPVI på blodplateoverflaten modifiseres i løpet av blodplatens levetid in vivo, og kan derfor brukes som en markør for blodplatealdersprofilen.
Glykoprotein VI er et 62/65 kDa (henholdsvis ikke-redusert/redusert) stort blod-platemembranglykoprotein som danner et kompleks sammen med den felles Fcy-underenhet. GPVI-underenheten inneholder det kollagenbindende sete og Fcy-underenheten er ansvarlig for signalgivning. Komplekset danner én av hovedkollagenreseptorene på blodplatens overflate, av avgjørende betydning for blodplateaktivering som respons på kollagen. Gjenkjennelsessekvensen på kollagen består av (GlyProHyp)n-sekvenser. Det er kjent pasienter fra Japan som har en genetisk mangel i GPVI. De har svake blødnings-problemer og deres blodplater gir bare svak respons på kollagen, sannsynligvis via andre reseptorer. Det er blitt funnet ut ganske mye vedrørende signalkaskadene som skriver seg fra GPVI, og som sterkt ligner på de fra immunreseptorer, inkludert T-cellereseptorer, B-cellereseptorer og naturlige drepercellereseptorer. Disse kaskadene omfatter src-familie-tyrosinkinaser, slik som Fyn og Lyn, samt p72<SYK> og mange andre tyrosinkinaser og fosfataser, og adaptorproteiner, slik som LAT. Et hovedmål for disse kaskadene er aktivering av fosfolipase Cy2, som splitter fosfolipider slik at man får de andre budbringerne diacylglyserol og IP3. GPVI antas å være involvert i aktivering av blod-integrinet a2|31 som har en hovedrolle i blodplateadhesjon til skadd karvegg. Mus med Fcy-underenheten "utslått" har blodplater som fortsatt oppviser responser på kollagen, noe som tyder på at den hvilende tilstand til a2|31 kan reguleres ved hjelp av GPVI/Fcy-komplekset.
Blodplatekollagenreseptoren GPVI er nært beslektet med de naturlige dreperakti-vatorreseptorer av p58KAR-familien samt FcaR.
Adhesjonen og aktiveringen av hvilende blodplater i omløp på et sted for vasku-lær skade er det første trinn i en prosess som fører til dannelsen av en trombe som om-dannes til en hemostatisk plugg. Kollagen er én av hovedkomponentene i karveggen som er ansvarlig for blodplateaktivering. Mange typer av kollagen foreligger og syv av disse finnes i subendotellagene. Flere forskjellige reseptorer for kollagen er blitt identifisert på blodplater, men de viktigste er nå ansett for å være integrinet a2|3i og ikke-integrinet GPVI. Selv om o^Pi er godt karakterisert og begge underenhetene ble klonet og sekvensert for flere år siden, har strukturen til GPVI forblitt uklar selv om flere trekk er blitt identifisert. Det ble bestemt for ca. tyve år siden at GPVI er et viktig blodplateglyko-protein med en molekylmasse i området 60-65 kDa og en sur pl. Dets rolle som en putativ kollagenreseptor ble fastslått etter identifikasjonen av en pasient i Japan med en svak blødningsforstyrrelse, hvis blodplater oppviste en spesifikk defekt i respons på kollagen og manglet denne reseptoren. Denne pasienten hadde også utviklet autoanti-stoffer mot den manglende reseptor og disse ble brukt til å karakterisere molekylet ytterligere. Nylig ble det fastslått at GPVI er forbundet ikke-kovalent med den felles Fcy-underenhet som virker som den signalgivende del av komplekset. Det ble også demonstrert at gjenkjennelsessekvensen på kollagen på GPVI er en gjentatt Gly-Pro-Hyp-triplett i kollagenets trippelheliksstruktur, og at de syntetiske peptidene er basert på denne strukturen kan brukes som spesifikke GPVI-rettede agonister. GPVI/Fcy-komplekset ble påvist å signalisere til blodplatens indre ved hjelp av en immunreseptor-lignene mekanisme som involverer aktivering av p72<SYK>, og som fører til en kaskade av kinase/fosfatase/adaptor-proteininteraksjoner for aktivering av PLCy2, og følgelig for frigjørelse av granuler og blodplateaggregasjon. Et ytterligere trinn i karakterisering av dette molekylet var påvisningen av at slange-C-type-lektinet, convulxin, fra den tropiske klapperslange, Crotalus durissus terrificus, var i stand til å aktivere blodplater ved å klumpe sammen GPVI gjennom en multimer interaksjon. Convulxin ble påvist å binde spesifikt til GPVI, noe som gir en fremgansmåte for rensing av denne reseptoren sammen med etablerte fremgansmåter.
Gibbins et al., i FEBS letters, vol. 413, nr. 2,1997, s. 255-259 identifiserer GPVI som kollagenreseptoren som utgjør bindeleddet mellom stimulering av blodplater og fosforylering av Fc reseptoren og videre cellesignalering.
Jandrot-Perrus et al., i Journal of Biological Chemistry, vol. 272, nr. 43, 1997, s. 27035-27041 beskriver blant annet interaksjonen mellom convulxin og blodplater og hvordan denne interaksjonen ble inhibert av antistoffer mot GPVI.
Det er således klart fra teknikkens stand at GPVI synes å være en svært interes-sant forbindelse på mange terapeutiske områder, fremfor alt vedrørende applikasjoner som er beslektet, direkte eller indirekte, til blodkoagulasjonshendelser som avhenger av interaksjon mellom kollagen og blodplater. Det var derfor et mål ved foreliggende oppfinnelse å tilveiebringe GPVI i en rekombinant form, og å påvise dets virkningsfullhet som direkte terapeutisk mål eller som redskap for screening av korte forbindelser, spesielt kjemisk syntetiserte eller syntetiserbare forbindelser som evnen til å inhibere eller blokkere den naturlige interaksjon mellom blodplate og kollagen.
Oppfinnelsen vedrører også deler eller fragmenter av GPVI-proteinet som har beholdt sin biologiske aktivitet som er bindingen til kollagen.
Oppfinnelsen var vellykket når det gjelder å rense adekvate mengder av GPVI for preliminær karakterisering, og for peptidsekvensering. Sekvensene ble brukt til å utforme primere for PCR for å identifisere en positiv sekvens i et DNA-bibliotek. Denne DNA-sekvens blir så brukt som en probe for å isolere en nesten komplett cDNA-sekvens fra biblioteket, og manglende 5'-sekvens ble erholdt ved å anvende en RACE-metode fra et blodplate-cDNA-bibliotek.
Oppfinnelsen var også vellykket ved å påvise bruken av rekombinant GPVI som terapeutisk anvendbar forbindelse som, når den administreres til en pasient med f.eks. skadde blodkar, er i stand til å binde kollagen og således forhindre blodplater som bærer membranbundet GPVI, fra å binde til kollagenet. Det rekombinante oppløselige ekstracellulære domene i GPVI inneholder kollagenbindingssetet og kan forhindre blodplateaktivering ved hjelp av kollagen. Det kan derfor være anvendbart til behandling av sykdomstilstander som involverer forøkt blodplateaktivering med kollagen, slik som aterosklerotisk plakkoppbrytning, ved slike sykdommer som ustabil angina, eller under kirurgisk behandling, slik som perkutan transluminal koronar-angioplasti (PTCA), hvor arterier åpnes på nytt ved oppblåsning av et ballongkateter som forårsaker betydelig skade på karveggen og mye blodplateaktivering, og som ofte resulterer i ny lukning av karet senere. Fordelen ved rekombinante GPVI-fragmenter sammenlignet med foreliggende behandlingsmetoder er at de virker på et tidlig stadium ved å forhindre eller redu-sere blodplateaktivering snarere enn ved å undertrykke hendelser etter blodplateaktivering, slik som aggregasjon ved hjelp av GPIIb-IIIa-antagonister. Mindre mengder av blodplategranulinnhold frigjøres således, inkludert vekstfaktorer og kjemokiner, som er involvert ikke bare i sårreparasjon, men i remodellering av karveggen ved hjelp av glatt-muskelmigrasjon, og tiltrekning av fagocyttiske celler, slik som monocytter som er kjent for å bidra til aterosklerose. Fab-fragment av humaniserte monoklonale museantistoffer mot GPVI kan brukes med lignende effekt for å blokkere GPVI på blodplateoverflaten, med lignende applikasjoner som ovenfor.
Rekombinant GPVI ifølge denne oppfinnelsen kan også anvendes i en bindings-analyse for kollagen for å screene med hensyn på små molekyler (f.eks. i kombinasjons-biblioteker) som er i stand til å inhibere denne interaksjonen, og som kan brukes til å utvikle terapeutiske forbindelser som er inhibitorer for interaksjoner mellom kollagen og blodplate. Ved hjelp av egnet derivatisering gjøres disse forbindelsene oralt tilgjengelige. Igjen er hovedmålet å fremstille forbindelser som reduserer GPVI-kollagen-interaksjoner og følgelig blodplateaktivering i situasjoner hvor blodplater kommer i kontakt med kollagen. Screeningsteknologien som sådan, som anvendes ved denne oppfinnelsen, er godt etablert innen teknikkens stand. Ved hjelp av slike screeningsanalyser gjør oppfinnelsen det mulig å finne og utvikle nye mål som kan inhibere det naturlige membranbundne GPVI på blodplateoverflaten som en kollagenantagonist. Slike mål som kan være små kjemiske molekyler, kan så være grunnlaget for ytterligere oppfinnelser.
En annen hovedapplikasjon for GPVI og reagenser som gjenkjenner spesifikke
domener i GPVI, er som markører for blodplatealder- og funksjonalitet. Unge blodplater antas generelt å være mer aktive og funksjonelle enn eldre. Unge blodplater binder seg til og aktiveres ved hjelp av slangegift-C-type-lektinet convulxin, som er spesifikt for GPVI,
og ettersom de eldes avtar både bindingen og aktiveringsgraden. Dette kan skyldes enten proteolytisk eller konformasjonelle endringer i GPVI eller dets binding til Fcy på grunn av blodplateaktivering eller skade i blodomløpet. Dette kan være en anvendbar parameter for å måle alderen og funksjonsprofilen til blodplater hos pasienter, samt hos friske per-soner under medisinske kontroller. Blodplatealderprofilen endrer seg ved mange sykdommer som påvirker benmargen eller immunsystemet, og kan være et viktig diagnostisk
kriterium dersom bedre metoder for dens bestemmelse var tilgjengelige. For eksempel vil pasienter med sykdommer som omfatter forøkt blodplateomsetning, oppvise yngre blodplater, mens pasienter på kjemoterapi eller strålebehandling vil oppvise en jevnt aldrende populasjon. En slik aldersprofil kan således brukes til en nøyaktig overvåkning av behandling. Hos en normalt frisk populasjon er svært lite kjent om aldersprofilfordelingen og dens rolle som en forutsiger for endringer i helse. Det er ikke kjent hvorvidt endring-ene i GPVI skyldes den delvise involvering av blodplater i hemostatiske hendelser, og hvorvidt endringer vi kunne være mer uttalt hos pasienter med omfattende kardiovasku-lær sykdom. For tiden brukes tiazolorange for å påvise unge retikulerte blodplater som inneholder mRNA. Dette mRNA går snart i oppløsning, noe som begrenser metoden til bare de unge blodplater. Reagenser som kan brukes i en slik analyse, vil omfatte GPVI-spesifikke slangegiftproteiner, slik som convulxin, eller monoklonale eller polyklonale antistoffer som gjenkjenner den N-terminale region i GPVI, eller monoklonale antistoffer som gjenkjenner nye seter eller konformasjoner eksponert ved hjelp av proteolyse av det N-terminale domene, eller spesifikke konformasjoner som er til stede enten i det intakte molekyl og ikke i det gamle, eller vice versa, eller små kjemiske enheter utvalgt for å gjenkjenne spesifikk intakt GPVI eller dets modifiserte form. Disse reagensene vil være merket med en fluorescensmarkør, eller sammen med et fluorescensmerket andre anti-stoff eller affinitetsreagens, og brukes i strømningscytometri for å måle blodplatebind-ingsprofilen. Ved et alternativ på et senere stadium kan det tas i bruk mindre arbeids-intensive måleteknikker basert på automatisert måling av blodplateprofiler. Ved å anvende cellesorteringsmetoder med strømningscytometri eller magnetiske kuler burde det være mulig å isolere unge og gamle blodplater for å undersøke faktorene som er involvert i fjerning av gamle blodplater fra blodomløpet. Reagenser som gjenkjenner spesifikke former av GPVI, vil være en nøkkel for slike undersøkelser.
Det er derfor et formål ved foreliggende oppfinnelse å tilveiebringe et DNA som koder for glykoprotein VI som omfatter aminosyresekvensen SEQ ID NR: 3 eller biologisk aktive fragmenter derav som binder til kollagen.
Det er et ytterligere formål med denne oppfinnelsen å tilveiebringe et DNA som koder for glyokoprotein VI, hvor DNAet omfatter SEQ ID NR: 1 eller fragmenter derav.
Det er et annet formål ved denne oppfinnelsen å tilveiebringe et farmasøytisk preparat som omfatter rekombinant GPVI som omfatter aminosyresekvensen fra SEQ ID NR: 3 sammen med en farmasøytisk akseptabel fortynner, bærer eller eksipiens, og dets anvendelse til fremstilling av et medikament innenfor det terapeutiske område av trombotiske og kardiovaskulære hendelser og forstyrrelser relatert til blodplate-kollagen-interaksjoner.
Et annet formål ved oppfinnelsen er anvendelsen av rekombinant GPVI som omfatter aminosyresekvensen SEQ ID NR: 3 i et screeningsredskap for påvisning av spesifikke inhibitorer for interaksjoner mellom blodplate og kollagen.
Et annet formål ved oppfinnelsen er anvendelsen av GPVI som omfatter aminosyre-sekvensen SEQ ID NR: 3 som en markør for blodplatealder og -eksponering for kardiovaskulære sykdommer.
Mulige medisinske indikasjoner og applikasjoner er henholdsvis f.eks. ustabil angina pectoris, PTC A, anvendelse av stenter i dette området, operasjoner på koronarkar, generelle operasjoner på blodkar, operasjoner som kan skade store blodkar, slik som hofteleddsoperasjoner. Dessuten er alle indikasjoner som vedrører tromboembolske hendelser forårsaket av forstyrrelser i interaksjonen mellom blodkarveggen og koagula-sjonssystemet med en høy risiko for dannelse av tromber og blokkering av blodårer, inkludert.
Som angitt ovenfor, er GPVI-proteinet og fragmentene derav ifølge foreliggende oppfinnelse egnet for farmasøytisk effektive forbindelser i farmasøytiske preparater og kombinasjoner.
De farmasøytiske preparatene ifølge oppfinnelsen kan eventuelt omfatte ytterligere aktive bestanddeler som anti-koagulanter, slik som hirudin eller heparin, eller trombolytiske midler, slik som plasminogenaktivator eller hementin, eller antagonister for andre blodplatereseptorer, slik som GPIIb-IIIa-antagonister som abciximab eller eptifibatid, eller ADP-reseptorantagonister slik som clopidogrel.
Det nye proteinet og dets biologisk aktive fragmenter ifølge oppfinnelsen kan danne farmasøytisk akseptable salter med hvilken som helst ikke-toksisk, organisk eller uorganisk syre. Uorganiske syrer er f.eks. saltsyre, hydrobromsyre, svovelsyre eller fosforsyre, og syremetallsalter, slik som natriummonohydrogenortofosfat og kalium-hydrogensulfat. Eksempler på organiske syrer er mono-, di- og trikarboksylsyrene, slik som eddiksyre, glykolsyre, melkesyre, pyrodruesyre, malonyre, ravsyre, glutarsyre, fumarsyre, eplesyre, vinsyre, sitronsyre, askorbinsyre, maleinsyre, hydroksymaleinsyre, benzosyre, hydroksybenzosyre, fenyleddiksyre, kanelsyre, salicylsyre og sulfonsyrer, slik som metansulfonsyre. Salter av karboksyendeaminosyreresten omfatter de ikke-toksiske karboksylsyresaltene som dannes med hvilke som helst egnede uorganiske eller organiske baser. Disse saltene omfatter f.eks. alkalimetaller, slik natrium og kalium, jordalkali-metaller, slik som kalsium og magnesium, lettmetaller i gruppe HIA, inkludert alumin-ium, og organiske primære, sekundære og tertiære aminer, slik som trialkylaminer, inkludert trietylamin, prokain, dibenzylamin, 1-etenamin, N,N'-dibenzyletylen-diamin, dihydroabietylamin og N-alkylpiperidin.
Slik det her er brukt, betyr uttrykket "farmasøytisk akseptabel bærer" et inert, ikke-toksisk, fast eller flytende fyllstoff, fortynningsmiddel eller innkapslingsmateriale, som ikke reagerer på skadelig måte med den aktive forbindelse eller med pasienten. Egnede, fortrinnsvis flytende bærere er velkjent innenfor teknikken, slik som sterilt vann, saltoppløsning, vandig dekstrose, sukkeroppløsninger, etanol, glykoler og oljer, inkludert de av petroleums-, animalsk, vegetabilsk eller syntetisk opprinnelse, for eksempel peanøttolje, soyabønneolje og mineralolje.
Preparatene ifølge oppfinnelsen kan adminstreres som enhetsdoser som inneholder vanlige ikke-toksiske, farmasøytisk akseptable bærere, fortynningsmidler, adjuvanser og vehikler, som er vanlig for parenteral administrering.
Uttrykket "parenteral" omfatter her subkutan, intravenøs, intraartikulær og intra-tracheal injeksjon og infusjonsteknikker. Også andre administreringsmåter, slik oral administrering og topisk applikasjon, er egnet. Parenterale preparater og kombinasjoner administreres mest foretrukket intravenøst, enten i en bolusform eller som en konstant innsprøyting ifølge kjente fremgangsmåter. Tabletter og kapsler for oral administrering inneholder vanlige eksipienser, slik som bindemidler, fyllstoffer, fortynningsmidler, tabletteringsmidler, smøremidler, desintegrasjonsmidler og fuktemidler. Tablettene kan belegges i henhold til metoder som er godt kjent innenfor teknikken.
Orale væskepreparater kan være i form av vann- eller olj esuspensj oner, -opp-løsninger, -emulsjoner, -syruper eller -eliksirer, eller kan presenteres som et tørt produkt for rekondisjonering med vann eller en annen egnet vehikkel før bruk. Slike væskepreparater kan inneholde vanlige additiver som oppslemningsmidler, emulgeringsmidler, ikke-vandige vehikler og konserveringsmidler. Topiske applikasjoner kan være i form av vann- eller olj esuspensj oner, -oppløsninger, -emulsjoner, -geleer eller fortrinnsvis emulsjonssalver.
Enhetsdoser ifølge oppfinnelsen kan inneholde daglig påkrevde mengder av proteinet ifølge oppfinnelsen, eller undermultipler derav som til sammen utgjør den ønskede dose. Den optimale terapeutisk akseptable dosering og dosehastighet for en bestemt pasient (pattedyr, inkludert mennesker) avhenger av mange forskjellige faktorer, slik som aktiviteten av det bestemte aktive materiale som anvendes, alderen, kropps-vekten, den generelle helse, kjønn, kost, administreringstid og -vei, utskillingshastighet, formålet ved behandlingen, dvs. terapi eller profylakse, og typen av den trombotiske sykdom som skal behandles, antiblodplate- eller antikoagulantaktivitet.
I preparater og kombinasjoner som kan anvendes som antikoagulanter hos en behandlet pasient (in vivo), er derfor en farmasøytisk effekt daglig dose av peptidene ifølge denne oppfinnelsen mellom ca. 0,01 og 100 mg/kg kroppsvekt, fortrinnsvis mellom 0,1 og 10 mg/kg kroppsvekt. Alt etter applikasjonsformen kan en enkeltdose inneholde mellom 0,5 og 10 mg av kollageninhibitoren. For å oppnå en antikoagulant-effekt i utenomkroppslig blod, er en farmasøytisk effektiv mengde av peptidene ifølge oppfinnelsen mellom 0,2 og 150 mg/l, fortrinnsvis mellom 1 mg og 20 mg/l utenomkroppslig blod.

Nærmere beskrivelse av oppfinnelsen
To sekvenser med 7 aminosyrer som viser den minste degenerasjon i den gene-tiske koden, ble valgt for syntesen av DNA-primere for å amplifisere en del av GPVI-cDNA-et ved hjelp av PCR. Ettersom lokaliseringen av begge peptidene i proteinet var totalt ukjent, ble det for hver av dem fremstilt to degenererte primere, én sense og en antisense. Disse primerne ble brukt til å amplifisere ett humant benmargsbibliotek. Kombinasjonen av sense-5' TYA THC CNG CNA TGA ARMG 3'-primeren som koder for sekvensen PAMKRSL, og antisense-5' TTR TAN ARN GCR AAY TGR TC 3'-primeren som tilsvarer DQFALYK, amplifiserte et DNA-fragment på 221 bp. I tillegg til de utvalgte peptider kodet det amplifiserte DNA for LysC/AspN-peptidet DQLELVATGVFAKPSLSAQPGPAVSS, noe som klart knytter sekvensen til cDNA-et for GPVI.
Screening av 600.000 pfu fra et benmargsbibliotek med dette 221 bp store DNA-fragment ga 4 positive pfu. Tre hadde innføyelser av 1350 bp enten de ble kuttet ved hjelp av restriksjonsenzymene Sal I eller EcoR I, og tilhørte IgG-superfamilien. Den fjerde hadde en 4,6 kb stor innføyelse ved Sal I-fordøyelse og ga to fragmenter på henholdsvis 2300 bp og 1300 bp når den ble behandlet med EcoRI. Dens DNA kodet for sekvensen for de 10 peptidene avledet fra aminosyresekvensering av GPVI, men stanset like før aminoenden. Ikke noe startmetionin eller noen ledersekvens kunne finnes, men mer enn 2000 bp av tidligere sekvensert ikke-leseramme-DNA som terminerer i en Alu-sekvens, var til stede. 5'-ende-RACE-forsøket ble fullført på blodplate-poly-A-RNA med primere lokalisert i en del av GPVI-sekvensen som var blitt bekreftet ved hjelp av sekvensen til peptidene. Et fragment på 348 bp inkludert 278 bp på sekvensen til den fjerde klon og 70 bp nye fra bp 1987 som tilsvarer 14 aminosyrer, inkludert det første metionin, ble funnet før man falt tilbake til den etablerte GPVI-sekvens. Et cDNA som inneholder i alt 1249 bp, en 25 bp stor 5'-sekvens oppstrøms for startkodonet, en åpen leseramme på 1017 bp som koder for et protein, inkludert ledersekvens, med 339 aminosyrer, og en 3'-region på 207 bp inkludert stoppkodonet, kunne således sekvenseres.
Et cDNA som koder for blodplate-GPVI ble klonet og sekvensert fra et humant benmargs-cDNA-bibliotek under anvendelse av RACE med blodplate-mRNA for å supplere manglende 5'-sekvens. Den åpne leseramme på 1017 bp koder for 339 aminosyrer og en utranslatert 3'-region. Hydrofobisitetsanalyse av aminosyresekvensen avslørte tilstedeværelsen av to putative transmembrandomener, en putativ 20-aminosyresignal-sekvens og et 19-aminosyrers domene mellom restene 247 og 265 i det fullt ferdige protein. Sekvensen og dens aminosyretranslasjon er vist i figur 2 og figur 1. En sammenligning med aminosyresekvensen til de mest like molekyler som finnes ved et søk i GenBank, avslører klart at den tilhører immunoglobulinsuperfamilien og at det ekstracellulære domene inneholder to Ig-C2-domenesløyfer dannet ved hjelp av to disulfid-bruer. Det er et molekyl i en membrankrysningsklasse med N-enden på utsiden og som krysser membranen én gang. De nærmest beslektede molekyler tilhører den naturlige dreperreseptorklasse som inneholder både inhibitor- og aktivatortyper. GPVI tilhører klart aktivatorunderklassen ikke bare gjennom dets funksjon, men også på grunn av at det, i motsetning til inhibitorklassen, ikke inneholder ITIM-sekvenser i sitt cytoplasmatiske domene. Det inneholder ikke noen tyrosinrester som vil kunne være involvert i fosforylering. Det er noen treonin- og serinrester i dette domene, men de passer ikke til noen kriterier for kinasekonsensussekvenser. På samme måte som aktivatorklassen av NK-reseptorer, inneholder GPVI en argininrest som tredje aminosyre i membrankrys-ningsdomenet som er involvert i kompleksdannelsen med Fcy-underenheten. Det cytoplasmatiske domene inneholder 51 aminosyrer, og oppviser bare en mindre likhet (i regionen like under membranen) med de cytoplasmatiske domenene til andre medlemmer av denne familien. Dette tyder på at dette domene i GPVI kan assosieres med andre typer av cytoplasmatiske molekyler enn de øvrige familiemedlemmene. GPVI inneholder bare et enkelt putativt N-glykosyleringssete i Asn69. Domenet like over membranen etter P-arkene i Ig-domenets slutt er imidlertid rike på treonin- og serinrester som kan gi O-glykosyleringsseter, slik det finnes i GPIba og GPV. Hovedfunksjonen til denne O-glykosylering synes å være å presentere reseptorstrukturene i god avstand fra blodplateoverflaten for å lette interaksjonene med deres voluminøse ligander. Ettersom GPVI tidligere ble fastlagt som et sialoglykoprotein, må forskjellen i molekyl vekt mellom den teoretiske aminosyrevekt (37 kDa) og vekten bestemt ved hjelp av gelelektroforese (65 kDa redusert) skyldes denne glykosyleringen.
Strukturen til naturlige dreperreseptorer av to-domenetypen er blitt fastlagt ved hjelp av røntgenkrystallografiske undersøkelser, og de to Ig-domene ble påvist å danne en spissvinkel med reseptorsetet for de peptidbærende HLA-antigener som ligger på utsiden av knekken. En direkte sammenligning av strukturen til HLA-peptidbindingssetet med strukturen til kollagen gir umiddelbart en antydning om hvorfor disse reseptorene har et felles opprinnelsessted, ettersom de multiple a-heliske strukturer til HLA-bindingssetet og peptidet som det inneholder, ligner svært på den trippelheliske struktur til kollagen. De naturlige dreperreseptorer postuleres til å arbeide ved hjelp av en dimeriserings-mekanisme med to reseptorer som gjenkjenner to separate HLA-seter på cellen som den naturlige drepercelle undersøker. Denne dimeriseringen er sannsynligvis del av aktiver-ings- eller deaktiveringsmekanismen, avhengig av reseptorklassen. I tilfellet med GPVI kan det også være mulig at to GPVI-molekyler inngår forbindelse med en Fcy, ettersom hver monomer i Fcy-dimeren har en gjenkjennelsessekvens. Støkiometrien er imidlertid ikke kjent ennå, og basert på strukturen til kollagener, kollagenlignende peptider som virker via GPVI og convulxin, synes det sannsynlig at styrken på signalet står i forhold til antallet GPVI/Fcy-komplekser som er klumpet sammen. Andre blodplatereseptorer som tilhører denne Ig-familien, omfatter ICAM-2 (CD102) og PECAM (CD31).
Alle mikroorganismer, cellelinjer, ekspresjonssystemer, ekspresjons verter, plas-mider, promotorer, resistensmarkører, replikasjonsopprinnelsessteder, restriksjonsseter eller andre fragmenter eller deler av vektorer som er nevnt i beskrivelsen ikke direkte i forbindelse med den krevede oppfinnelse, er kommersielt eller på annen måte generelt tilgjengelig. Forutsatt at det ikke er gitt noen annen antydning, anvendes de bare som eksempler og er ikke av avgjørende betydning i forhold til oppfinnelsen, og kan erstattes henholdsvis av andre egnede redskaper og biologiske materialer.
Teknikkene som er av avgjørende betydning ifølge oppfinnelsen, er beskrevet nærmere nedenunder og ovenfor. Andre teknikker som ikke er nærmere beskrevet, tilsvarer kjente standardmetoder som er godt kjent for fagfolk innen teknikken, eller er beskrevet nærmere i de anførte litteraturhenvisninger og patentsøknader, og i standard-litteraturen (f.eks. Sambrook et al., 1989, Molecular Cloning: A Laboratory Manual, 2. utg., Cold Spring Harbor; Harlow, Lane, 1988, Antibodies: A Laboratory Manual, Cold Spring Harbor).
EKSEMPLER
Eksempel 1: Materialer. Protein-A-Sepharose, peroksidase-konjugert geit-anti-mus- og -anti-kanin-antistoffer, bovint serumalbumin, Crotalus durissus tern/?cw5-slangegift, hvetekimagglutinin (WGA), N-hydroksysuksinimidyl-klorformiat-aktivert kryssbundet 4% agarosekuler og Triton X-l 14 var fra Sigma Chemical Co. (St. Louis, MO), oktanoyl-N-metyl-glukamid (ONMG) og nonanoyl-N-metylglukamid (NNMG) var fra Oxyl Chemie (Bobingen, Tyskland).
Eksempel 2: GPVI-isolering fra blodplater. Membranglykoproteiner ble isolert fra blodplater som tidligere beskrevet. I korte trekk ble blodplater (fra 40 "buffy coats") vasket og ly sert i 2 % Triton X-l 14 i nærvær av proteaseinhibitorer. Triton X-l 14 og vannfaser ble adskilt og detergentfasen ble fylt på en kolonne med hvetekimagglutinin koblet til Sepharose 4B. Blodplateglykoproteinene ble eluert med 10 mM tris HC1, pH 7,4, 30 mM NaCl, 0,2 % oktanoyl-N-metylglukamid (ONMG) og 2 % N-acetylglukosamin. Etter dialyse og konsentrering ble oppløsningen av glykoproteiner på en kolonne av convulxin bundet til N-hydroksysuksinamidyl-p-nitrofenylklorformiat-aktivert, kryssbundet 4 % agarosekuler (1 mg/ml). Kolonnen ble vasket med 4 volumdeler 10 mM tris HC1, pH 7,4,30 mM NaCl, 0,2 % nonanoyl-N-metylglukamid (NNMG), og så med 4 volumdeler lOmM tris HC1, pH 7,4, 30 mM NaCl og 2 % NNMG. GPVI ble eluert med 0,08 % SDS i 10 mM tris/HCl, pH 7,4. Oppløsningen ble konsentrert og fylt på en preparativ gel av 8,5 % polyakrylamid under anvendelse av en modell 491 Prep Cell (BioRad, CA). Den prepara-tive elektroforese ble utført under ikke-reduserende betingelser ved å følge produsentens instruksjoner. GPVI eluerte som et enkeltbånd ved 65 kDa. Fraksjonene ble slått sammen, konsentrert på Centricon-30 (Amicon, Beverly, MA) og på nytt oppslemmet i 10 mM tris/HCl, pH 7,4 og 0,1 % ONMG.
Eksempel 3: Aminosyreanalyse av GPVI. GPVI ble fordøyd med endoproteinasene LysC og AspN (Boehringer Mannheim, Tyskland). De 10 peptidene som ble generert, ble separert ved hjelp av reversfase-HPLC og sekvensert på et Applied Biosystem modell 477A pulset væskefaseproteinsekvenseringsapparat med et modell 120A on-line fenyl-tiohydantoin-aminosyre-analyseapparat.
Eksempel 4: Amplifikasjon av et 221 bp stort fragment som koder for en del av GPVI, fra et kgtl 1-cDNA-bibliotek.
En prøve (10<10> pfu) (plakkdannende enheter) fra et humant benmargsbibliotek (Clonetech, Palo Alto, CA) ble amplifisert ved å anvende 2 kombinasjoner av 4 degenererte primere. Sluttprimerkonsentrasjonene var 2 u,m, dNTP-konsentrasjonen var 200 u,m og 2 U/100 ul reaksjons-AmpliTaq Gold (Perkin Eimer, Rotkreuz, Sveits) ble brukt. PCR-betingelsene var 5 sykluser ved 37 °C etterfulgt av 30 sykluser ved 44 °C. Sens-19-meren 5' TYATHCCNGCNATGAARMG 3' og antisens-20-meren 5' TTRTANARNGCRAAYTGRTC 3' amplifiserte et 221 bp stort fragment som ble subklonet i Bluescript KS<+> (Stratagene, La Jolla, CA) og sekvensert ved å anvende T7-Sequenase kit (Amersham, Sveits).
Eksempel 5: Screening av Xgtl 1-cDNA-biblioteket med den 221 bp store GPVI-probe. Det 221 bp store fragment ble kuttet fra plasmidet, rengjort og merket med a<32>P-ATP (20 MBq/50 ul, Hartmann Analytik, Braunschweig, Tyskland) under anvendelse av High Prime Labelling kit (Boehringer Mannheim, Sveits). Det humane benmargsbibliotek ble screenet ved å følge produsentens instruksjoner. Positive fager ble dyrket, deres DNA isolert og subklonet i BlueScript under anvendelse enten av EcoRI- eller Sal I-seter, og sekvensert. Sekvensering ble utført ved å anvende ABS-systemet til RACE. Blodplate-polyA-RNA ble fremstilt som tidligere beskrevet (Power et al., Cytokine 7,479-482, 1995). Reverstranskripsjon (30 ul) ble utført ved å anvende 5 u,g polyA-RNA med primeren 5TGAATGAGACGGTCAGTTCAGC 3' (20 nm), dNTP (1 mM), RNAsin (40 U), IX AMV-buffer og 20 U AMV-revers-transkriptase i 20 min ved 45 °C etterfulgt av 20 minutter ved 52 °C. Reaksjonsblandingen ble behandlet med 2 ul 6N NaOH ved
65 °C i 30 min, nøytralisert med 2 ul 6N eddiksyre og konsentrert i et Centricon 30 (Amicon). Et anker ble ligert til førstetråds-DNAet ved å følge fremgangsmåten til Aptes og Sibert (Bio Techniques 15: 890-893, 1993). Nestet PCR ble utført ved å anvende en primer som er komplementær til ankeret og primeren 5'
TTGTACAGAGCAAATTGGTC 3' (35 sykluser, 55 °C) og etterfulgt av primeren 5' GACCAGAGGCTTCCGTTCTG 3' (30 sykluser ved 53 °C). Det høyeste bånd (350 bp) ble fraskilt ved hjelp av agaroseelektroforese fra de lavere, subklonet i BlueScript, og sekvensert.
Eksempel 6: Fremstilling av anti-GPVI-Fab og F(ab')2. Polyklonale antisera mot humant GPVI ble generert i kaniner. IgG fra kanin-anti-GPVI-antiserum ble renset som beskrevet. Spalting av IgG med immobilisert papain (Pierce) for å generere Fab-fragmenter, ble utført i henhold til standardfremgangsmåten til leverandøren. Fab-fragmenter ble skilt fra uspaltet IgG- og Fc-fragmenter under anvendelse av en immobilisert protein A-kolonne (Sigma). Gjennomstrømningen ble overført til dialyserør, konsentrert under anvendelse av fast polyetylenglykol 20.000, grundig dialysert mot 20 mM Hepes, 140 mM NaCl, 4 mM KC1, pH 7,4 og lagret ved 4 °C inntil det ble brukt. F(ab')2-fragmenter ble fremstilt ved hjelp av pepsinspalting av IgG, forhold mellom enzym og substrat 1:50 (vekt/vekt), i 0,5 M acetatbuffer, pH 4,0, ved 37 °C i 18 timer. pH-verdien ble korrigert til 7,4 med fortynnet NaOH og prøven ble dialysert mot 20 mM fosfat, pH 7,4. F(ab')2-fragmenter ble separert fra uspaltet IgG- og Fc-fragmenter ved å anvende protein A-kromatografi. Gjennomstrømningen ble overført til dialyserør, konsentrert under anvendelse av fast polyetylenglykol 20.000, grundig dialysert mot 20 mM Hepes, 140 mM NaCl, 4 mM KC1, pH 7,4 og lagret i alikvoter ved -20 °C. Vaskede blodplater ble lysert i Triton X-l 14 og faseseparasjon ble utført på det oppløselige materiale før isolering av membran-glykoproteinene forbundet med Triton X-l 14-fasen ved hjelp av affinitetskromatografi på hvetekimagglutinin-Sepharose 4B som beskrevet tidligere. Ettersom GPVI utgjør en svært liten fraksjon av blodplatemembranglykoproteinblandingen, brukte vi spesifisitieten til orme-C-type-lektinet convulxin for isolering av denne reseptoren. Affinitetskromatografi på convulxin koblet til Sepharose 4B ga et 65 kDa protein som hovedprodukt. Ukarakterisert materiale med både høyere og lavere Mr sameluerte imidlertid med GPVI og kunne ikke fjernes ved hjelp av grundig vasking av kolonnen. Preparativ gelelektroforese på 8,5 % polyakrylamid ble tilføyet som et rensesluttrinn. Fraksjoner inneholdende GPVI ble slått sammen og ga et enkeltband etter ny analyse. Renset GPVI ble testet med hensyn på dets evne til å blokkere blodplateaggregasjon ved hjelp av kollagen. En svak inhibitoreffekt ble observert når alikvoter av GPVI-oppløsning ble tilsatt blodplatesuspensjonen. Ved å forinkubere GPVI med kollagen før tilsetning av blandingen til blodplatesuspensjonen, kunne imidlertid aggregasjon inhiberes på en dose-avhengig måte. Disse blodplatene aggregerte fortsatt når nytt kollagen ble tilsatt. Under ikke-reduserende betingelser har det isolerte protein en Mr på 62 kDa med et skifte mot en litt høyere Mr (65 kDa) under reduserende betingelser. Ettersom aminoenden til GPVI ble funnet å være blokkert, ble proteinet spaltet med enzymene LysC og LysC/AspN som ga henholdsvis 4 og 6 peptider, hvorfra sekvens ble erholdt. Peptidene ble separert ved hjelp av reversfase-HPLC på en C4-kolonne og sekvensert ved å anvende Edman-metoden. Aminosyresekvensene til disse peptidene er understreket i den translaterte cDNA-sekvens (figur 1).
Claims (10)
1. DNA,
karakterisert ved at det koder for glykoprotein VI som omfatter aminosyresekvensen ifølge SEQ ID NO: 3 eller biologisk aktive fragmenter derav som binder til kollagen.
2. DNA ifølge krav 1, hvor DNAet omfatter sekvensen ifølge SEQ ID NO:l eller fragmenter derav som koder for biologisk aktive fragmenter av glykoprotein VI som binder til kollagen.
3. DNA ifølge kravene 1 eller 2, hvor DNAet består av sekvensen ifølge SEQ ID NO:l.
4. DNA ifølge hvilke som helst av kravene 1-3, hvor DNAet koder for glykoprotein VI som omfatter aminosyresekvensen ifølge SEQ ID NO:2 og SEQ ID NO:3.
5. Rekombinant humant glykoprotein VI til anvendelse som medikament, karakterisert ved at det omfatter aminosyresekvensen ifølge SEQ ID NO:3.
6. Farmasøytisk preparat,
karakterisert ved at det omfatter glykoprotein VI omfattende aminosyre-sekvensen fra SEQ ID NO:3 og et farmasøytisk akseptabelt fortynningsmiddel, bærer eller eksipiens derfor.
7. Farmasøytisk preparat ifølge krav 6, hvor det i tillegg omfatter en farmakologisk aktiv forbindelse.
8. Anvendelse av GPVI omfattende aminosyresekvensen fra SEQ ID NO:3 i et screeningsredskap for påvisning av spesifikke inhibitorer for blodplate-kollagen-interaksjoner.
9. Anvendelse av GPVI omfattende aminosyresekvensen fra SEQ ID NO:3 til fremstilling av et medikament til bruk innenfor det terapeutiske område trombotiske og kardiovaskulære følger og forstyrrelser relatert til blodplate-kollagen-interaksjoner.
10. Anvendelse av endringer i GPVI omfattende aminosyresekvensen fra SEQ ID NO:3 som en markør for blodplatealder og blodplateeksponering for trombotiske og kardiovaskulære tilstander eller følger.
