Description
において使用するためのDNA構築物及びベクター、並びに該ベクターで形質転
換した酵母細胞に関する。
、例えば、欧州特許出願第0088632A号、第0116201A号、第0123294A号、第012354
4A号、第0163529A号、第0123289A号、第0100561A号、第0189998A号及び第019598
6A号、PCT特許出願第WO95/01421号、第95/02059号及び第90/10075号、並びに米
国特許第4,546,082号を参照のこと。
般的な特徴である。そのようなマーカー遺伝子は、大腸菌(E.coli)における最初
のクローニング工程に由来し、ここで該マーカー遺伝子は、形質転換した細胞を
選別するために用いられるか、又はベクターとして用いられるプラスミドを維持
するために用いられる。抗生物質マーカー遺伝子は、形質転換した酵母細胞の培
養に不都合な影響を決して与えないと考えられており、従ってそのようなDNA
を欠失させる何れの工程も行わないのが通例であった。さらに、プラスミド構築
物の特性は、形質転換した酵母細胞からのプラスミドの単離、及び大腸菌への単
離したプラスミドの形質転換の後、抗生物質による選択によって調査される。従
って、抗生物質耐性マーカー遺伝子を保有することは、実際には好都合であった
。
ているが、誤って幾らか細胞が環境に放出されるという小さな危険性が常に存在
する。それらの非常に複雑な性質のために、そのような遺伝子工学的に操作され
た微生物は非常に短期間生存するのみであり、環境を害する危険は極めて低い。
勿論、このことが、そのような形質転換した微生物が研究及び大規模な実施の両
方において使用が許可されてきた理由である。
むプラスミドが誤って環境に廃棄される可能性があり、プラスミドが自発的に獲
得される場合に、細菌中において抗生物質に対する耐性が導入されるという理論
上の危険が存在する。
対して耐性にさせる遺伝子による潜在的な環境汚染の如何なる危険性も、可能な
限り最小にすべきである。 従って、今日まで用いられてきた方法よりも、いっそう安全な方法を開発する
必要があり、そのような改良方法を提供することが本発明の目的である。
あって、ここで、産生のために用いる酵母形質転換株は発現ベクターを含有し、
該ベクターにおいて、最初のクローニング工程で用いられる抗生物質マーカー遺
伝子を、酵母宿主の形質転換の前に、イン・ビトロでの修飾により非機能的にす
る方法に関する。また本発明は、そのような方法において使用するためのDNA
配列及び発現ベクター、並びに形質転換した酵母細胞に関する。

ドpKV228における種々の欠失が、Arg34GLP−1(7-37)発酵収率に影
響を及ぼすかどうかを評価した(表3)。

在的な危険性がなくなる。

スリン類縁体、インスリン様増殖因子I又はII、ヒト成長ホルモン又はウシ成長
ホルモン、インターロイキン、組織プラスミノゲン賦活剤、トランスホーミング
増殖因子a又はb、グルカゴン、グルカゴン様ペプチド1(GLP−1)、グル
カゴン様ペプチド2(GLP−2)、GRPP、第VII因子、第VIII因子、第XII
I因子、血小板由来増殖因子、及びリパーゼのような酵素が挙げられる。
リンに見られるような3つのジスルフィド架橋が正しく形成された2つの鎖のイ
ンスリン又はインスリン類縁体分子へ変換され得るプロインスリンを含む単一鎖
のポリペプチドは、「インスリン前駆体」又は「インスリン類縁体の前駆体」と
して理解され得る。インスリン前駆体は、インスリンのA鎖及びB鎖を架橋する
修飾C−ペプチドを、典型的に含有するであろう。さらに、好ましいインスリン
前駆体は、B(30)アミノ酸残基を欠如するであろう。最も好ましいインスリン前
駆体は、例えばEP163529、及びPCT出願第95/00550及び95/07931に記載されるよ
うなものである。インスリンの例としては、ヒトインスリン、好ましくはdes(B
30)ヒトインスリン及びブタインスリンが挙げられる。好ましいインスリン類縁
体は、1以上の天然アミノ酸残基、好ましくは1、2または3の天然のアミノ酸
残基が、別のコード可能なアミノ酸残基によって置換されたものである。従って
、源のインスリンは、位置A21において、Asnの代わりに、Ala、Gln
、Glu、Gly、His、Ile、Met、Ser、Thr、Trp、Try
又はValを含む群から選択されるアミノ酸残基、特にGly、Ala、Ser
及びThrを含む群から選択されるアミノ酸残基を有していてもよい。同様に、
源のインスリンは、位置B28において、Proの代わりに、Asp、Lys等
を含む群から選択されるアミノ酸残基を有していてもよく、位置B29において
、Lysの代わりに、アミノ酸Proを有していてもよい。

わす。

るホスホアミダイト法、又はMatthesら, EMBO Journal 3, 1984, pp801-805に記
載される方法)により合成的に調製され得る。ホスホアミダイト法に従って、オ
リゴヌクレオチドを、例えば、自動DNA合成機にて合成し、精製し、二重構造
にし、連結させて合成DNA構築物を形成する。一般に好ましいDNA構築物の
調製方法は、例えば、Sambrookら, Molecular Cloning: A Laboratory Manual,
Cold Spring Harbor, NY, 1989に記載されるようなポリメラーゼ連鎖反応(PC
R)に基づく。

参照)に従って、ゲノムライブラリーまたはcDNAライブラリーを調製し、合
成オリゴヌクレオチドプローブを用いてハイブリダイゼーションすることにより
本発明のポリペプチドの全て又は一部をコードするDNA配列をスクリーニング
することによって得られる、ゲノム起源又はcDNA起源であってもよい。
成起源、ゲノム起源又はcDNA起源(適切なもの)のアニーリングフラグメン
ト(このフラグメントは、全DNA構築物の種々の部分に相当する)によって調
製される、合成起源とゲノム起源の混合物、合成起源とcDNA起源の混合物、
又はゲノム起源とcDNA起源の混合物であってもよい。
体の複製に無関係な、染色体外の独立体として存在するベクター(例えば、プラ
スミド、染色体外因子、ミニクロモソーム、又は人工染色体)であってもよい。
ベクターは、確実に自己複製する何れの手段を含有してもよい。また代わりとし
て、ベクターは、宿主細胞に導入されると、ゲノムに組み込まれ、組み込まれた
染色体とともに複製されるものであってもよい。ベクター系は、宿主細胞のゲノ
ムに導入されて全DNAをともに含有する、単一のベクター若しくはプラスミド
、又は2以上のベクター若しくはプラスミド、又はトランスポゾンであってもよ
い。
せた所望のタンパク質又はポリペプチドをコードするDNA配列を含有してもよ
いであろう。プロモーターは、酵母において転写活性を示す何れのDNA配列で
あってもよく、酵母に対して同種又は異種のタンパク質をコードする遺伝子由来
であってもよい。適切なプロモーターの例は、サッカロミセス・セレビシエMα
1、TPI、ADH又はPGKプロモーターである。
TPIターミネーターのような適切なターミネーター(T. Alber 及びG. Kawasak
i, J. Mol. Appl. Genet. 1, 1982, pp.419-434参照)に使用可能なように連結さ
せてもよい。
ようなDNA配列を含有するであろう。そのような配列の例は、酵母プラスミド
2μ複製遺伝子REP1-3及び複製起点である。また該ベクターは、選択可能
なマーカー(例えば、P. R. Russell, Gene 40, 1985, pp.125-130に記載される
ようなシゾサッカロミセスポンペTPI遺伝子)を含有していてもよい。

ナル配列は、より大きなポリペプチドの成分として、それが合成される細胞の分
泌経路を通ってより大きなポリペプチドを誘導するポリペプチド(分泌ペプチド
)をコードするDNA配列である。より大きなポリペプチドは、分泌経路を通過
する間に一般に切断されて、分泌ペプチドを除去する。
泌経路へ誘導する何れのシグナルペプチドをコードしてもよい。シグナルペプチ
ドは、天然に存在するシグナルペプチド若しくはそれらの機能部分であってもよ
く、又は合成ペプチドであってもよい。酵母宿主細胞に有用なシグナルペプチド
は、サッカロミセス・セレビシエa−因子及びサッカロミセス・セレビシエイン
ベルターゼ、マウス唾液のアミラーゼのシグナルペプチド(O. Hagenbuchleら, N
ature 289, 1981, pp.643-646参照)、修飾カルボキシぺプチダーゼシグナルペプ
チド(L. A. Vallsら, Cell 48, 1987, pp.887-897参照)、酵母BAR1シグナル
ペプチド(WO87/02670参照)、又は酵母アスパラギン酸プロテアーゼ3(YAP3
)シグナルペプチド(M. Egel-Mitaniら, Yeast 6, 19909, pp.127-137参照)の遺
伝子から得られる。
が、シグナル配列の下流かつポリペプチドをコードするDNA配列の上流に挿入
されてもよい。リーダーペプチドの機能は、培地への分泌のために、発現したポ
リペプチドを小胞体からゴルジ装置へ、さらには分泌小胞へ誘導させることであ
る(即ち、細胞壁を横切ったポリペプチドの輸送、又は少なくとも酵母細胞の細
胞周辺腔への細胞膜を通ったポリペプチドの輸送)。リーダーペプチドは、酵母
a−因子リーダーであってもよい(例えば、その使用はUS 4,546,082, EP 16201
, EP 123294, EP 123544及びEP 163529に記載される)。また代わりとして、リ
ーダーペプチドは、天然に存在しない合成リーダーペプチドであってもよい。合
成リーダーペプチドは、WO89/02463又はWO92/11378、及びKjeldsenらによる「タ
ンパク質の発現と精製9, 331〜336(1997年)」に記載されるように構築されて
もよい。
培地への分泌のために、小胞体からゴルジ装置へ、さらには分泌小胞へ誘導され
る(即ち、存在するのであれば、細胞膜及び細胞壁を横切った発現したタンパク
質若しくはポリペプチドの輸送、又は少なくとも細胞壁を有する細胞の細胞周辺
腔への細胞膜を通った発現したタンパク質若しくはポリペプチドの輸送)プロペ
プチド配列の形態にあるペプチドを意味すると理解される。
びターミネーターそれぞれを連結し、それらを酵母複製に必要な情報を含む適切
な酵母ベクターにそれらを挿入するために用いられる方法は、当業者にとって公
知である(例えば、上述のSambrook参照)。第一に本発明のポリペプチドをコー
ドする全DNA配列を含むDNA構築物を調製し、次にこのフラグメントを適切
な発現ベクターに挿入するか、又は個々の要素(シグナル、リーダー又は異種タ
ンパク質のような)の遺伝情報を含むDNAフラグメントを順次挿入し、続いて
連結するかによってベクターを構築してもよいことは理解されるであろう。

酵母有機体の例は、酵母種サッカロミセス・セレビシエ、サッカロミセス・クル

varum)、クルイベロミセス(Kluyveromyces)・ラクチス(lactis)、ハンセヌラ(Ha
nsenula)・ポリモルファ(polymorpha)、ピチア(Pichia)・パストリス(pastoris)
、ピチア・メタノリカ(methanolica)、ピチア・クルイベリ、ヤロウィア(Yarrow
ia)・リポリチカ(lipolytica)、カンジダ種、カンジダ・ウチリス(utilis)、カ
ンジダ・カカオイ(cacaoi)、ゲオトリクム種及びゲオトリクム・ファメンタンズ
(fermentans)から選択される株であってよく、好ましくは酵母種サッカロミセス
・セレビシエである。

地は、酵母有機体の成長に適切な従来の何れの培地であってもよい。そのかなり
の割合が正確に処理された形態で培地中に存在するであろう分泌された異種タン
パク質は、遠沈法又は濾過による培地からの酵母細胞の分離、上澄み液又は濾液
のタンパク様成分の塩による沈殿(例えば、硫酸アンモニウム)、続いて種々の
クロマトグラフィ法による精製(例えば、イオン交換クロマトグラフィ、アフィ
ニティクロマトグラフィ等)を含む従来の方法によって、培地から回収され得る
。タンパク質が細胞周辺腔に分泌される場合、酵素的又は機械的に細胞を崩壊さ
せる。

、本技術分野で公知のように、イン・ビトロでの化学的又は酵素的切断によって
、回収したタンパク質から除去されてもよい。酵素を用いて切断するのがより好
ましい。そのような酵素の例としては、トリプシン又はアクロモバクター(Achro
mobacter)・リチクス(lyticus)プロテアーゼIが挙げられる。
法でも本発明の特許請求の範囲を限定するものではない。
-21)インスリン前駆体、WO97/22706を参照)の発現のために構築した酵母プラス
ミドpAK729は、2つのApaLI酵素制限部位ApaLI(4477)及びAp
aLI(5723)を含む(図1参照)。これらの制限部位は、AMPマーカー遺伝子
の各側に位置する。pAK729における2つのApaLI部位間の1246ヌクレ
オチドを除去することにより、AMPマーカー遺伝子及び更に幾らかの大腸菌由
来のプラスミドDNAが取り除かれるであろう。
泳動を行い、単離し、再連結させ、次にコンピテントS.セレビシエ細胞(MT
663、EPB0163529参照)に形質転換して、形質転換した酵母株NN729.1
−ΔAMPが得られる。酵母株NN729.1−ΔAMPから該修飾発現プラス
ミドを再分離し、PCR生成、続く欠失を特徴とするDNA領域のサブクローニ
ング後にDNA配列を確認した。同様に、酵母株NN729.1−ΔAMPから
再分離したプラスミドDNAにおいて、インスリン前駆体をコードするDNA配
列を確認した。 酵母株NN729.1−ΔAMPを、YPD培地中で、30℃にて、72時間
培養した。インスリン前駆体の発酵収率は、RP−HPLCによって決定した。

択されたDNA配列を確認した。pAK729.5の制限プラスミド地図を図2
に示してある。制限酵素部位XhoI(5676)及びXhoI(5720)間のDNAフラ
グメントを、プラスミドpAK729.5から欠失させて、AMP遺伝子内に位
置する44ヌクレオチドを欠失させることが可能である。
泳動を行い、単離し、再連結させ、次にコンピテントMT663S.セレビシエ
細胞に形質転換させ、酵母形質転換株NN729.5−ΔAMPを得た。酵母株
NN729.5−ΔAMPから該修飾発現プラスミドを再分離し、PCR生成、
続く欠失を特徴とするDNA領域のサブクローニング後に、DNA配列を確認し
た。同様に、酵母株NN729.5−ΔAMPから再分離したプラスミドDNA
において、インスリン前駆体をコードするDNA配列を確認した。pAK729
.5−ΔAMPにおける44のヌクレオチドの欠失が、βーラクタマーゼ活性の
喪失に関してAMP遺伝子の完全な欠失と同程度に効率的であることがわかった
。 酵母株NN729.5−ΔAMPを、YPD培地中で、30℃にて、72時間
培養した。インスリン前駆体の発酵収率は、RP−HPLCにより決定した。

K729.6において、制限酵素部位AtaII(4982)及びAtaII(5978)間のD

くことができる。これにより、AMP遺伝子全て及びプロモーターが除去される
であろう。
ス電気泳動を行い、単離し、再連結させ、次にコンピテントMT663S.セレ
ビシエ細胞に形質転換した。酵母株NN729.6−ΔAMPから該発現プラス
ミドを再分離し、PCR生成、続く欠失を特徴とするDNA領域のサブクローニ
ング後に、DNA配列を確認した。同様に、酵母株NN729.6−ΔAMPか
ら再分離したプラスミドDNAにおいて、インスリン前駆体をコードするDNA
配列を確認した。AMP遺伝子を欠如しているプラスミドpAK729.6−Δ
AMPを図4に示してある。 酵母株NN729.6−ΔAMPを、YPD培地中で、30℃にて、72時間
培養した。インスリン前駆体の発酵収率は、RP−HPLCにより決定した。

7の選択されたDNA配列を確認した。pAK729.7において、制限酵素部
位AtaII(3801)およびAatII(5978)間のDNAフラグメントを欠失させて、
該発現プラスミドから2177のヌクレオチドを除去することかできる。pAK72
9.7プラスミドは、AMP遺伝子及び大腸菌の複製起点の両方を欠失させるこ
とができるように設計された。pAK729.7の制限プラスミド地図は図5に
示してある。
ース電気泳動を行い、単離し、再連結させ、次にコンピテントMT663S.セ
レビシエ細胞に形質転換した。酵母株NN729.7−ΔAMPから該修飾発現
プラスミドを分離し、PCR生成、続く欠失を特徴とするDNA領域のサブクロ
ーニング後に、DNA配列を確認した。同様に、酵母株NN729.7−ΔAM
Pから再分離したプラスミドDNAにおいて、インスリン前駆体をコードするD
NA配列を確認した。酵母株NN729.7−ΔAMPを、YPD培地中で、3
0℃にて、72時間培養した。インスリン前駆体の発酵収率は、RP−HPLC
により決定した。

に比較して、インスリン前駆体を10〜20%も多く発現するということが、上
記より明らかである。
ラスミドを用いた酵母におけるArg34GLP−1(7-37)の発現 図1〜5に例示されるLA19X5M13をコードするpAK729構築物の
EcoRI(940)−XbaI(1403)配列を、本例のMFアルファ*−Arg34GL
P−1(7-37)をコードする配列で置き換えた(図6)。MFα1プレ−プロリー
ダーペプチド(Kurjan 及び Herskowitz, Cell 30, 1982. pp.993)の修飾(ここ
で、位置82のLeu及び位置83のAspは、それぞれMet及びAlaで置
換され、DNA配列にNcoI切断部位を導入する)を、この構築物にて行った
。該リーダー配列を、MFα1*と称した(Kjeldsen T.ら, 1996年)。MFα1シ
グナルMFα1*リーダーペプチド配列は、二塩基のKex2p認識モチーフ(
Lys−Arg)を含有し、該モチーフはArg34GLP−1(7-37)のコード配
列とリーダーとを分離する。ペプチドArg34GLP−1(7-37)は、ヒトGLP
−1(7-37)変異体(S. Mojsovら, J Biol. Chem. 261, 1986, pp.11880-11889)
であり、ここで、位置34の天然のアミノ酸残基はArg残基で置換されている
。

、2つのアスパラチルプロテアーゼ(即ち、(1)一塩基又は二塩基アミノ酸残
基のC末端側を切断する酵母アスパラチルプロテアーゼ3(YAP3)(Egel-Mi
taniら, YEAST 6: 127-137, 1990年)及び(2)プロテアーゼB、カルボキシペ
プチダーゼY,アミノペプチダーゼI、RNase、アルカリホスファターゼ、
酸性トレハラーゼ及びエキソポリホスファターゼのような他のプロテアーゼの活
性化を招く液胞型プロテアーゼA)を欠如した表現型を有する。さらに、トリオ
ースホスフェートイソメラーゼ遺伝子(TPI)を崩壊させ、その表現型は、グ
ルコースを含有する培地における形質転換株の成長においてグルコースを利用す
ることを可能にする。ME1719の遺伝的背景は、MATa/a Dyap3::
ura3/Dyap3::URA3 pep4-3/pep4-3 tpi::LEU2/
Dtpi::LEU2 leu2/leu2 Dura3/Dura3である。

Claims (8)
- 【請求項1】 細菌細胞を抗生物質耐性にすることができない組換え型酵母
発現ベクターであって、前記ベクターが、異種遺伝子及びイン・ビトロでの修飾
により非機能的にした抗生物質耐性マーカー遺伝子をコードする遺伝子を含む組
換え型酵母発現ベクター。 - 【請求項2】 前記抗生物質マーカー遺伝子が、AMP遺伝子である請求項
1に記載の組換え型酵母発現ベクター。 - 【請求項3】 適切な条件下にて請求項1又は2に記載のベクターを含有す
る酵母株を培養し、その培地から所望の産物を単離することを含んでなる、所望
のポリペプチド又はタンパク質の製造方法。 - 【請求項4】 適切な条件下にて酵母発現プラスミドを含有する酵母株を培
養することを含んでなる、所望のポリペプチド又はタンパク質の製造方法であっ
て、前記プラスミドにおいて、細菌における最初のクローニング工程に用いられ
る機能的抗生物質マーカー遺伝子を、酵母宿主に挿入する前にイン・ビトロにて
前記マーカー遺伝子の一部又は全マーカー遺伝子を欠失させることにより、非機
能的にする方法。 - 【請求項5】 前記抗生物質耐性マーカー遺伝子の周囲に適切な制限部位を
挿入し、イン・ビトロにて適切な制限酵素で処理することにより前記マーカー遺
伝子を欠失させることを含んでなる、請求項4に記載の方法。 - 【請求項6】 前記酵母株が、サッカロミセス属の株、好ましくはサッカロ
ミセス・セレビシエ株である請求項3〜5の何れか1項に記載の方法。 - 【請求項7】 請求項1又は2に記載のベクターを含有する、形質転換した
酵母細胞。 - 【請求項8】 サッカロミセス属の株、好ましくはサッカロミセス・セレビ
シエ株である、請求項7に記載の形質転換した酵母細胞。
