- 【特許請求の範囲】 1.複数の分析物を系列的にプロセシングするための装置であって、該装置は: 微粒子の集団を平面アレイに配置するためのフローチャンバーであって、該複 数の微粒子の集団の各微粒子がそこに係留された分析物を有する、フローチャン バー; 該複数の微粒子に係留された分析物が系列的にプロセシング試薬に曝露される ように、1つ以上のリザーバーからプロセシング試薬をフローチャンバーに系列 的に送達するための流体手段;および 該複数の微粒子の各々からの光学シグナルのシーケンスを検出するための検出 手段であって、該シーケンスの各光学シグナルがプロセシング試薬とそこに係留 された該分析物との間の相互作用の指標である、検出手段、 を備える、装置。 2.前記フローチャンバーが軸を有し、そして該軸に沿って連続して、入口、平 面キャビティ、ダム、および出口を、前記プロセシング試薬が該入口を通って該 フローチャンバーに進入し、そして該軸の方向に該平面キャビティを通って該出 口へと流動するように備える、請求項1に記載の装置。 3.前記フローチャンバーの前記平面キャビティがさらに光学的に透過性の天井 および床を有し、該光学的に透過性の天井および床が互いに平行であり、かつ該 床が複数の平行な隆起を有し、該平行な隆起が該フローチャンバーの前記軸に対 して平行に配置され、そして該平行な隆起が、前記微粒子が該平面キャビティ中 で該平行な隆起の間に列を形成するように間隔を空けている、請求項1または2 に記載の装置。 4.前記検出手段がさらに、光学列であって、前記微粒子の前記平面アレイから の前記光学シグナルの画像を、該光学シグナルのデジタル画像を発生させるため の電気光学検出器上に焦点を合わせる光学列と、該微粒子の該平面アレイの複数 のデジタル画像を記録するための記録手段とを備える、請求項1〜3のいずれか に記載の装置。 5.前記検出手段がさらに、シグナル追跡手段であって、前記微粒子の各々から の前記光学シグナルを前記デジタル画像の各々において相関させて、前記複数の 該微粒子の各々について、該光学シグナルのシーケンスを形成する、シグナル追 跡手段を備える、請求項4に記載の装置。 6.前記電気光学検出器がCCDカメラである、請求項5に記載の装置。 7.前記分析物がDNAであり、そしてここで前記光学シグナルが蛍光シグナルで ある、請求項1〜6のいずれかに記載の装置。 8.フローチャンバーであって: 該フローチャンバーの一端の入口と; 該フローチャンバーの他端の出口と; 該入口および出口の各々と流体連通した平面キャビティとを備え、ここで該平 面キャビティが緊密に充填された微粒子の平面アレイを保持し、プロセシング試 薬が各微粒子にプロセスの各工程の間に接近し得るように、一連のプロセシング 工程の間、各々がそれに係留した分析物を有する、フローチャンバー。 9.前記平面アレイに関して緊密に充填された微粒子が、該平面アレイにおける 単位面積当たりの微粒子の数が、等面積の六方晶系アレイにおける微粒子の数の 少なくとも80%であるか、または隣接する微粒子の中心間の平均距離が微粒子直 径の2倍未満であることのいずれかを必要とする、請求項8に記載のフローチャ ンバー。 10.前記平面が光学的に透過性の天井および床を備え、該光学的に透過性の天 井および床は互いに実質的に平行であり、そして該床が、前記フローチャンバー の長軸に平行して配置される平行な複数の隆起を有し、かつ前記平面キャビティ 中の微粒子が該平行な隆起の間に列を形成するように間隔を空けて配置される、 請求項8に記載のフローチャンバー。 11.複数の微粒子の各々からの光学シグナルのシーケンスを検出するための検 出装置であって: 該微粒子からの光学シグナルを集めて、デジタルシグナルに変換するための光 学列; 該微粒子の複数のデジタル画像を記録するための画像化装置;および シグナル追跡手段であって、該光学シグナルのシーケンスを該微粒子の各々に ついて形成するために該光学シグナルの該デジタル画像の各々において、該微粒 子の各々からの該光学シグナルを相関させるためのシグナル追跡手段、 を備える、検出装置。 12.デバイス読みとり可能な媒体であって、該デバイスによって実行される命 令のプログラムを包含し、該命令のプログラムによって、微粒子の平面アレイの 画像を生成する方法を実行して、該微粒子の位置が一連のプロセシング工程の間 に追跡され、該命令のプログラムが: 該微粒子で発生する光学シグナルに基づいて、該一連のプロセシング工程の間 の該微粒子の該平面アレイの複数のデジタル画像を与えるための命令;および 該プロセシングが、該各微粒子で発生する光学シグナルを該複数のデジタル画 像の各々の中の対応する画像と相関させて、該一連のプロセシング工程の間に、 該各微粒子を追跡する工程を包含する、該複数のデジタル画像をプロセシングす るための命令、 を包含する、媒体。 13.前記プロセシングがさらに、各微粒子の少なくとも1つの光学特性を記録 して、該各微粒子の近似中心を決定する工程を包含する、請求項12に記載のデ バイスで読みとり可能な媒体。 14.前記プロセシングがさらに、各微粒子で発生する光学シグナルの少なくと も1つの特性を決定するために複数の画素を該各微粒子に割り当てる工程を包含 する、請求項13に記載のデバイスで読みとり可能な媒体。 15.所与の微粒子に割り当てられる前記画素の数が: (i)該所与の微粒子の前記近似中心が幾何学的中心から逸脱している可能性 がある程度; (ii)該所与の微粒子のサイズ;および (iii)該所与の微粒子の直径および形状の均一性 のうちの少なくとも1つに基づいて決定される、請求項14に記載のデバイスで 読みとり可能な媒体。 16.前記複数の画素の最初の画素が、その微粒子の前記近似中心を囲む各微粒 子に割り当てられる、請求項14に記載のデバイスで読みとり可能な媒体。 17.前記最初の画素が割り当てられた後、さらなる画素が該最初の画素にすぐ に隣接して各微粒子に割り当てられる、請求項16に記載のデバイスで読みとり 可能な媒体。 18.一連のプロセシング工程の間、微粒子の位置を追跡するために該微粒子の 平面アレイの画像を生成する方法であって、該方法は: 該微粒子で発生する光学シグナルに基づいて、該一連のプロセシング工程の間 の該微粒子の該平面アレイの複数のデジタル画像を与える工程;および 該複数のデジタル画像をプロセシングする工程であって、各微粒子で発生する 光学シグナルを該複数のデジタル画像の各々における対応する画像と相関させて 、該一連のプロセシング工程の間、該各微粒子を追跡する工程、 包含する、方法。 19.前記プロセシング工程がさらに、各微粒子の少なくとも1つの光学特性を 記録して、該各微粒子の前記近似中心を決定する工程を包含する、請求項18に 記載の方法。 20.前記プロセシング工程がさらに、各微粒子で発生する前記光学シグナルの 少なくとも1つの特性を決定するために複数の画素を該各微粒子に割り当てる工 程を包含する、請求項19に記載の方法。 21.前記所与の微粒子に割り当てられる前記画素の数が: (i)該所与の微粒子の前記近似中心が幾何学的中心から逸脱している可能性 がある程度; (ii)該所与の微粒子のサイズ;および (iii)該所与の微粒子の直径および形状の均一性 のうちの少なくとも1つに基づいて決定される、請求項20に記載の方法。 22.前記複数の画素の最初の画素が、その微粒子の前記近似中心を囲む各微粒 子に割り当てられる、請求項20に記載の方法。 23.前記最初の画素が割り当てられた後、さらなる画素が該最初の画素にすぐ に隣接して各微粒子に割り当てられる、請求項22に記載の方法。 24.ポリヌクレオチドのアレイであって: 緊密に充填された微粒子の平面アレイ;および 複数の異なるポリヌクレオチドであって、各々異なるポリヌクレオチドが、異 なる微粒子に付着されるように、該微粒子の各々に付着するポリヌクレオチドを 含む、アレイ。 25.前記平面アレイに関して緊密に充填した微粒子が、前記平面アレイにおけ る単位面積当たりの微粒子の数が、等面積の六方晶系アレイにおける微粒子の数 の少なくとも80%であるか、または隣接する微粒子の中心間の平均距離が微粒子 直径の2倍未満であることのいずれかを必要とする、請求項24に記載のアレイ 。 26.前記微粒子の各々の直径が約0.1μmと100μmとの間である、請求項24 に記載のアレイ。 27.前記複数の異なるポリヌクレオチドがcDNAライブラリーを含む、請求項2 4に記載のアレイ。 28.前記微粒子の平面アレイがフローチャンバー中に配置される、請求項24 に記載のアレイ。
