Claims (1)
- 【特許請求の範囲】 1.式(I)で表される化合物であって、 式中: R1は、水素、ヒドロキシ、アルキル、アルコキシ、ヒドロキシアルキル、ト リフルオロメチル、アミノ、アルキルカルボニルアミノ、アルキルカルボニルオ キシ、ハロゲンであり、 R2およびR3は、互いに独立に、水素、ヒドロキシ、アルキル、アルコキシ、 ヒドロキシアルキル、ハロゲン、アルコキシカルボニルアルキル、カルバモイル 、スルファモイルであり、 R4は、ω−ヒドロキシアルコキシ、ω−アミノアルコキシ、ω−アミノアル キルアミノ、アルコキシアルキル、ヒドロキシアルコキシアルキルアミノアルキ ル、アルコキシカルボニルアルキル、ジヒドロキシアルキル、ホルミル、アルキ ルカルボニル、アルコキシカルボニル、アルキルカルボニルアミノアルキ ル、アミノアルキル、アルキルアミノアルキル、ジアルキルアミノアルキル、カ ルボキシアルキル、カルバモイルアルキル、カルボキシアミドアルキル、カルボ キシル、アミノ、ニトロ、シアノ、ニトリロ、シアノアルキル、アジド、炭素原 子を少なくとも2個有するアルキル、炭素原子を少なくとも2個有するアルコキ シ、炭素原子を少なくとも2個有するヒドロキシアルキルであり、 R5は、水素、ハロゲン、アルキルであり、 Arは、アルキル、アルコキシ、ヒドロキシ、ヒドロキシアルキル、ハロゲン 、アルコキシカルボニルアルキル、カルバモイル、スルファモイルで一置換され ていても互いに独立に二置換されていてもよい、アリールまたはヘテロアリール であり、 R6およびR7は、同じであっても異なってもよく、共に炭素原子を少なくとも 3個含み、1個または複数の水酸基を担持してもよく、酸素原子によって炭素原 子が相互に連結していてもよい、ヒドロカルビル基であって、R6およびR7は、 アミン窒素とともに環を形成してもよく、 但し、(a) (i)R2、R3およびR5のうち少なくとも2つが水素以外で あるか、 (ii)R1がヒドロキシまたはメトキシ以外であり、且つArがヒドロキシま たはメトキシでオルト置換されているフェニル以外であるか、 (iii)Arがヘテロアリールであるか、あるいは (iv)R6およびR7の少なくとも1つが、芳香族ヒドロカルビルかシクロアル キルであるとき、 R4は、水素、メチル、メトキシ、ヒドロキシメチル、ヒドロキシ、ハロゲン 、カルバモイル、スルファモイルでもよく、 (b)Arが未置換のフェニルであるとき、R1、R2、R3、R4およびR5は、 全てが水素であってはならない化合物、それらの生理学的に許容できる酸との塩 類、および化合物が光学異性体の形であってもよいとき、ラセミ混合物および個 々の鏡像異性体。 2.R4が、ω−ヒドロキシアルコキシ、ω−アミノアルコキシ、ω−アミノア ルキルアミノ、アルコキシアルキル、ヒドロキシアルコキシアルキルアミノアル キル、ジヒドロキシアルキル、ホルミル、アルキルカルボニル、アルコキシカル ボニル、アルコキシカルボニルアルキル、アルキルカルボニルアミノアルキ ル、アミノアルキル、アルキルアミノアルキル、ジアルキルアミノアルキル、カ ルボキシアルキル、カルバモイルアルキル、カルボキシアミドアルキル、カルボ キシル、アミノ、ニトロ、シアノ、ニトリロ、シアノアルキル、またはアジドで ある、請求の範囲第1項に記載の化合物。 3.R1が水素またはメチルであり、R2、R3およびR5が全て水素であるか、ま たはR2、R3およびR5の1つがメチル、メトキシ、ヒドロキシ、カルバモイル 、スルファモイルまたはハロゲンであって、その他は水素であるかのいずれかで あり、Arが、フェニルであるか、またはメチル、メトキシ、ヒドロキシ、ヒド ロキシメチル、カルバモイル、スルファモイルまたはハロゲンで一置換されてい るか互いに独立に二置換されているフェニルである、請求の範囲第2項に記載の 化合物。 4.Arがヘテロアリールである、請求の範囲第1項に記載の化合物。 5.R1が水素またはメチルであり、R2、R3、R4およびR5が全て水素である か、またはR2、R3、R4およびR5の1つがメチル、メトキシ、ヒドロキシ、ヒ ドロキシメチル、カルバモイル、スルファモイルまたはハロゲンであって、その 他は水 素であるかのいずれかである、請求の範囲第4項に記載の化合物。 6.R1が、水素、アルキル、ヒドロキシアルキル、トリフルオロメチル、アミ ノ、アルキルカルボニルアミノ、アルキルカルボニルオキシ、またはハロゲンで あり、Arが、ヒドロキシまたはアルコキシでオルト置換されているフェニル以 外である、請求の範囲第1項に記載の化合物。 7.R1が水素またはメチルであり、R2、R3、R4およびR5が全て水素である か、またはR2、R3、R4およびR5の1つがメチル、メトキシ、ヒドロキシ、カ ルバモイル、スルファモイルまたはハロゲンであって、その他は水素であるかの いずれかであり、Arが、フェニルであるか、またはメチル、メトキシ、ヒドロ キシ、ヒドロキシメチル、カルバモイル、スルファモイルまたはハロゲンで一置 換されているか互いに独立に二置換されているフェニルである、請求の範囲第6 項に記載の化合物。 8.R6およびR7の少なくとも1つが、芳香族ヒドロカルビル、シクロアルキル 、または少なくとも1つの位置で炭素原子が酸素原子によって相互に連結してい てもよいヒドロカルビル鎖で ある、請求の範囲第1項に記載の化合物。 9.R1が、水素またはメチルであり、R2、R3、R4およびR5が全て水素であ るか、またはR2、R3、R4およびR5の1つがメチル、メトキシ、ヒドロキシ、 カルバモイル、スルファモイルまたはハロゲンであって、その他は水素であるか のいずれかであり、Arが、フェニルであるか、またはメチル、メトキシ、ヒド ロキシ、ヒドロキシメチル、カルバモイル、スルファモイルまたはハロゲンで一 置換されているか互いに独立に二置換されているフェニルである、請求の範囲第 8項に記載の化合物。 10.R1が、ヒドロキシ、ハロゲン、トリフルオロメチル、アミノ、メトキシ またはヒドロキシメチルである、請求の範囲第1項から第9項のいずれか一項に 記載の化合物。 11.R2およびR3が、互いに独立に水素、ヒドロキシまたはヒドロキシメチル である、請求の範囲第1項から第10項のいずれか一項に記載の化合物。 12.R4が、水素、ホルミル、アルコキシカルボニル、アルキルカルボニル、 ヒドロキシアルキル、アルコキシアルキル、カルボキシアミドアルキル、カルバ モイルアルキル、アミノアル キル、アミノ、アジド、シアノアルキル、カルボキシまたはカルボキシアルキル である、請求の範囲第1項から第10項のいずれか一項に記載の化合物。 13.R4が、水素、ホルミル、ヒドロキシメチル、ヒドロキシエチル、ヒドロ キシプロピル、ヒドロキシブチル、ヒドロキシペンチル、ヒドロキシヘキシル、 エトキシメチル、メトキシカルボニル、アミノ、アミノプロピル、アセチル、1 ,2−ヒドロキシエチル、エチルアミノメチル、またはヒドロキシエトキシエチ ル−アミノエチルである、請求の範囲第12項に記載の化合物 14.R5が水素である、請求の範囲第1項から第13項のいずれか一項に記載 の化合物。 15.R6およびR7が、それぞれ互いに独立に飽和ヒドロカルビル基、特に、飽 和脂肪族ヒドロカルビル基、たとえば、C1-8アルキル、特にC1-6アルキル、ま たはアダマンチルを示し、R6とR7が共に炭素原子を少なくとも3個、好ましく は少なくとも4個含む、請求の範囲第1項から第14項のいずれか一項に記載の 化合物。 16.R6およびR7が共にアミン窒素と環を形成する、請求の 範囲第1項から第14項のいずれか一項に記載の化合物。 17.R6およびR7の少なくとも1つが分枝炭素鎖を含む、請求の範囲第1項か ら第16項のいずれか一項に記載の化合物。 18.Arが、チエニル、ピリル、チアゾリル、オキサゾリル、メチルチアゾリ ルまたはメチルピリルである、請求の範囲第1項から第17項のいずれか一項に 記載の化合物。 19.N,N−ジイソプロピル−3−(2−フルオロフェニル)−3−フェニル プロパンアミン塩酸塩、 N,N−ジイソプロピル−3−(5−ホルミル−2−ヒドロキシ−フェニル) −3−フェニルプロパンアミン、またはその(R)−異性体、 N,N−ジイソプロピル−3−(2−ヒドロキシ−5−メチルオキシカルボニ ル−フェニル)−3−フェニルプロパンアミン、またはその(R)−異性体、 N,N−ジイソプロピル−3−(5−アセチル−2−ヒドロキシフェニル)− 3−フェニルプロパンアミン、またはその(R)−異性体、 N,N−ジイソプロピル−3−[2−ヒドロキシ−5−(2−ドロキシエチル )−フェニル]−3−フェニルプロパンア ミン、またはその(R)−異性体、 N,N−ジイソプロピル−3−[2−ヒドロキシ−5−(1−ヒドロキシエチ ル)−フェニル]−3−フェニルプロパンアミン、またはその3(R)−異性体 、 N,N−ジイソプロピル−3(R)−[5−(1(R*)、2−ジヒドロキシ エチル)−2−ヒドロキシフェニル]−3−フェニルプロパンアミン、またはそ の1(S*)−異性体、 N,N−ジイソプロピル−3−[2−ヒドロキシ−5−(6−ヒドロキシヘキ シル)−フェニル]−3−フェニルプロパンアミン、またはその(R)−異性体 、 N,N−ジイソプロピル−3−(5−エトキシメチル−2−ヒドロキシフェニ ル)−3−フェニルプロパンアミン、またはその(R)−異性体、 N,N−ジイソプロピル−3−[5−(3−アミノプロピル)−2−ヒドロキ シフェニル]−3−フェニルプロパンアミン、またはその(R)−異性体、 N,N−ジイソプロピル−3−[5−(3−アセトアミドプロピル)−2−ヒ ドロキシフェニル]−3−フェニルプロパンアミン、またはその(R)−異性体 、 N,N−ジイソプロピル−3−[5−(2−シアノエチル)−2−ヒドロキシ フェニル]−3−フェニルプロパンアミン、またはその(R)−異性体、 N,N−ジイソプロピル−3−(5−アミノ−2−ヒドロキシフェニル)−3 −フェニルプロパンアミン、またはその(R)−異性体、 N,N−ジイソプロピル−3−(5−アジド−2−ヒドロキシフェニル)−3 −フェニルプロパンアミン、またはその(R)−異性体、 N,N−ジイソプロピル−3−[2−ヒドロキシ−5−(3−ヒドロキシプロ ピル)−フェニル]−3−フェニルプロパンアミン、またはその(R)−異性体 、 N−シクロブチル−N−メチル−3−(2−ヒドロキシフェニル)−3−フェ ニルプロパンアミン、 N,N−ジイソプロピル−3−(2−ヒドロキシフェニル)−3−(2−チエ ニル)プロパンアミン、または N,N−ジイソプロピル−3−(2−ヒドロキシ−5−メチルフェニル)−3 −(2−チエニル)プロパンアミン、またはその(R)−異性体 である、請求の範囲第1項に記載の化合物。 20.薬学的に活性な物質、特に抗コリン作用剤として使用するための、請求の 範囲第1項から第19項のいずれか一項に記載の化合物。 21.請求の範囲第1項から第19項のいずれか一項に記載の化合物、および好 ましくは適合する製薬用担体を含む薬剤用組成物。 22.抗コリン作用薬を調製するための、請求の範囲第1項から第19項のいず れか一項に記載の化合物の使用。 23.尿失禁関連疾患を罹っている生体を治療する方法であって、請求の範囲第 1項から第19項のいずれか一項に記載の化合物の有効量を前記生体に投与する ステップを含む方法。 24.請求の範囲第1項から第19項のいずれか一項に記載の化合物を調製する 方法であって、 a)式IIの化合物 (式中、R1〜R5およびArは、請求の範囲第1項に規定した通りであり、Yは 脱離基である)を、アミンHNR6、R7(式中、R6およびR7は上記の規定通り である)と反応させること、または、 b)式IIIの化合物 (式中、R1〜R7およびArは、請求の範囲第1項に規定した通りであり、あら ゆる水酸基は保護されていてもよい)を還元すること、または c)式IVの第二級アミン(式中、R1〜R5およびArは、請求の範囲第1項に規定した通りであり、あら ゆる水酸基は保護されていてもよく、Zは、 R6およびR7と同じ意味を持つ)をN−アルキル化すること、または、 d)式Vaまたは式Vbの化合物 (式中、R1〜R7およびArは、請求の範囲第1項に規定した通りであり、あら ゆる水酸基は保護されていてもよく、Wは水酸基またはハロゲンを表す)を還元 すること、または e)式VIの化合物(R2〜R7およびArは、請求の範囲第1項に規定した通りであり、R1aはカ ルボキシルまたはアルコキシである)において、R1aをヒドロキシに変換する こと、または f)式VIIの化合物 (式中、R1、R6、R7およびArは、請求の範囲第1項に規定した通りであり 、R2b〜R5bのうち1つはアルキレンであり、その他はR2〜R5に関して請求 の範囲第1項に規定した通りである)において、アルキレンをアルキル、ヒドロ キシアルキルまたはジヒドロキシアルキルに還元すること、または g)請求の範囲第1項に規定した式Iの化合物において、基R1〜R5のうち1つ または複数を他の基R1〜R5に変換すること、または h)式VIIIの化合物 (式中、R1〜R7は、請求の範囲第1項に規定した通りであり、Xは酸素または イオウである)を、式IX CH3N=C: IX の化合物と反応させて、式Iaの化合物 (式中、R1〜R7およびXは上述の規定通りである)を生成すること、または i)上記式VIII(式中Xは酸素である)の化合物を、式Xの化合物 と反応させて、式Ibの化合物 (式中、R1〜R7は、請求の範囲第1項に規定した通りである)を生成すること 、または、 j)式XIの化合物(式中、R1〜R7は、請求の範囲第1項に規定した通りである)を、式XIIの 化合物 (式中、R1〜R7は、請求の範囲第1項に規定した通りである)に変換すること 、または k)式XIIIの化合物 (式中、R1〜R7は、請求の範囲第1項に規定した通りであり、Xは酸素または イオウである)を、式XIVの化合物(式中、R1〜R7およびXは、上記の通りであり、R8およびR9は、互いに独立 に水素またはアルキルである)に変換すること、および、 i)必要な時に、得られた化合物のヒドロキシ保護基を取るこ と、 ii)必要に応じて、得られた式Iの塩基を、生理学的に許容できる酸との塩に 変換すること、またはその逆、および/または、 iii)必要に応じて、得られた光学異性体の混合物を、個々の鏡像異性体に分 割すること を含む方法。
