Claims (1)
- 【特許請求の範囲】 1.下記一般式(II): 〔式中、Rは水素原子、直鎖若しくは分岐鎖のC1−C4アルキル基又はベンジル 基であり、 R1は窒素、酸素及びイオウから選択される1個又は2個のヘテロ原子を有す る任意に置換されていてもよい5又は6員環の芳香族複素環基である。〕 で表されるヘテロアリール−フェニルアラニン類の製造方法であって、下記一般 式(III) R1−Zn−Y (III) 〔式中、R1は上記と同義であり、Yは塩素原子、臭素原子又はヨウ素原子であ る〕 で表される化合物と、下記一般式(IV) 〔式中、Rは上記と同義であり、R’は任意に保護されることかあるアミノ基で あり、Xはヨウ素原子若しくは臭素原子又はメタンスルホニルオキシ基、フルオ ロメタンスルホニルオキシ基、p−トルエンスルホニルオキシ基若しくはトリフ ルオロメタンスルホニルオキシ基である〕 で表される化合物とを遷移金属(0)触媒の存在下に反応させ、R’が保護され たアミノ基である場合には、脱保護反応させることを特徴とする製造方法。 2.化合物III及び化合物IVが、1:1〜3:1のモル比で使用される請求 項1記載の製造方法。 3.モル比が1:1〜2:1である請求項2記載の製造方法。 4.Xがヨウ素原子である式(IV)の化合物が使用される請求項1記載の製造 方法。 5.触媒の量が、式(III)の有機亜鉛化合物に対し0.05〜5mol%で ある請求項1記載の製造方法。 6.触媒が、配位子の存在下で任意に担持されたパラジウム又はニッケルベース の触媒である請求項1記載の製造方法。 7.触媒がパラジウムテトラキス(トリフェニルホスフィン)、ニッケルテトラ キス(トリフェニルホスフィン)又はトリフェニルホスフィン存在下の炭担持パ ラジウムより選択されるものである請求項6記載の製造方法。 8.R’がtert−ブトキシカルボニルアミノ基又はホルミルアミノ基である 式(IV)の化合物を使用する請求項1記載の製造方法。 9.式(III)の化合物が、対応するヘテロアリール−リチウム誘導体又はヘ テロアリール−マグネシウム誘導体を無水ハロゲン化亜鉛と反応させその場で調 製されたものである請求項1記載の製造方法。 10.R1がチアゾリル基又はチエニル基より選択されるヘテロ環基である式(I I)の化合物の製造方法である請求項1記載の製造方法。 11.下記一般式(I) 〔式中、R及びR1は請求項1に記載と同義であり、 R2はC2−C4の直鎖若しくは分岐鎖のアルキル基、又はアルキル部分に1個 ないし6個の炭素原子を有するアリール基若しくはアリールアルキル基であり、 前記アリール基はフェニル基又は窒素、酸素及びイオウから選択される1個若し くは2個のヘテロ原子を有する5又は6員環の芳香族複素環基であり、ハロゲン 原子、水酸基、アルキル部分の炭素数が1−6のアルコキシ、アルキル、アルキ ルチオ、アルキルスルホニル若しくはアルコキシカルボニル基、1個以上のフッ 素原子を有するC1−C3アルキル基、カルボキシ基、ニトロ基、アミノ若しくは アミノカルボニル基、アシルアミノ基、アミノスルホニル基又はアルキル部分の 炭素数が1−6のモノ若しくはジアルキルアミノ若しくはモノ若しくはジ−アル キルアミノカルボニル基から選択される同一若しくは異なる1個以上の置換基と 任意に置換されていてもよい。 R3はメルカプト基又は基内においてメルカプト基に変換可能なR4COS基で あり、R4は直鎖若しくは分岐鎖のC1−C4アルキル基又はフェニル基である。 〕 で表されるフェニルアラニンのN−メルカプトアシル誘導体の製造方法であって 、下記一般式(III) R1−Zn−Y (III) 〔式中、R1及びYは請求項1に記載されたものと同義である〕 で表される化合物と下記一般式(IV) 〔式中、R、R’及びXは請求項1に記載されたものと同義である〕 で表される化合物とを遷移金属(0)触媒の存在下に反応させ、R’が保護され たアミノ基である場合には、脱保護反応させて次の一般式(II) 〔式中、R及びR1は前記と同義である〕 で表される化合物を得る反応を含むことを特徴とする製造方法。
