Claims (1)
- 【特許請求の範囲】 1. 試料からターゲット分子を分離するにあたり、 1)(i)油体と、(ii)ターゲット分子を含有する試料とを接触させ 、前記ターゲット分子を前記油体に会合させる工程:及び 2)前記試料から、前記ターゲット分子と会合している前記油体を分離す る工程を含むことを特徴とする、試料からターゲット分子を分離する方法。 2. リガンド分子が更に含まれており、前記リガンド分子が前記油体及び前記 ターゲット分子と会合していることを特徴とする、請求項1記載の方法。 3. 前記リガンド分子が、前記油体に共有結合的に付着していることを特徴と する、請求項2記載の方法。 4. 前記リガンド分子が、前記油体中の油体タンパク質に共有結合的に付着し ていることを特徴とする、請求項3記載の方法。 5. 前記リガンドが、2種の分子を含んでおり、第1の分子が前記油体と会合 し、及び第2の分子が前記ターゲットと会合し、前記第1の分子と前記第2の分 子とが互いに会合していることを特徴とする、請求項2記載の方法。 6.前記第1及び第2のリガンド分子が互いに共役していることを特徴とする、 請求項5記載の方法。 7. 前記リガンド分子がタンパク質であることを特徴とする、請求項4記載の 方法。 8. 前記タンパク質リガンドが、前記油体タンパク質を有する融合タンパク質 であることを特徴とする、請求項7記載の方法。 9. 前記ターゲット分子が、タンパク質、ペプチド、有機分子、脂質、糖質、 核酸、細胞、細胞小器官、細胞成分、ウイルス、金属、金属イオン及びイオンか らなる群より選ばれることを特徴とする、請求項1〜8のいずれか一項記載の方 法。 10.前記リガンド分子がヒルジンであり、及び前記ターゲット分子がトロンビ ンであることを特徴とする、請求項8記載の方法。 11.前記リガンド分子がプロテインAであり、及び前記ターゲット分子が免疫 グロブリンであることを特徴とする、請求項8記載の方法。 12.前記リガンド分子がメタロチオネインであり、及び前記ターゲット分子が カドミウムであることを特徴とする、請求項8記載の方法。 13.前記リガンド分子がセルロース結合タンパク質であり、及び前記ターゲッ ト分子がセルロースであることを特徴とする、請求項8記載の方法。 14.前記リガンド分子が核酸結合タンパク質であり、及び前記ターゲット分子 が核酸であることを特徴とする、請求項8記載の方法。 15.前記リガンドが一本鎖DNA結合タンパク質又はRNA結合タンパク質で あり、及び前記ターゲットが一本鎖核酸分子であることを特徴とする、請求項1 4記載の方法。 16.前記リガンドが抗体であり、前記抗体が前記油体又は油体タンパク質に結 合していることを特徴とする、請求項2記載の方法。 17.前記ターゲットが、前記リガンド抗体に結合し得る、細胞、細胞小器官又 は細胞成分であることを特徴とする、請求項16記載の方法。 18.前記細胞が、Staphylococcus aureusであることを 特徴とする、請求項16記載の方法。 19.前記リガンドが二価抗体であり、前記二価抗体が前記油体及び前記ターゲ ットの双方に結合することを特徴とする、請求項16記載の方法。 20.前記リガンドが、アビジンに共役している抗体であり、及び前記ターゲッ ト分子がビオチンであることを特徴とする、請求項6記載の方法。 21.前記油体タンパク質がオレオシンであることを特徴とする、請求項4、7 、8、9、10、11、12、13、14及び15のいずれか一項記載の方法。 22.前記オレオシンが、ナタネ(Brassica spp.)、ダイズ(Gly cine max )、ヒマワリ(Helianthus annuus)、ギネア アブラヤシ(Elaeis uineeis)、ココヤシ(Cocus nuci fera )、トウゴマ(Ricinus communis)、ベニバナ(Cart hamus tinctorius )、アブラナ(Brassica spp.及 びSinapis alba)、コエンドロ(Coriandrum sativ um )、アマニ/アマ(Linum usitatissimum)、シロイヌナ ズナ(Arabidopsis thaliana)及びトウモロコシ(Zea mays )からなる群より選ばれた植物から誘導されることを特徴とする、請 求項21記載の方法。 23.工程1)において、前記油体及び前記試料を混合し、及び次いで約1分か ら約24時間までの間でインキュベーションすることを特徴とする、請求項1か ら22のいずれか一項記載の方法。 24.前記混合した油体及び試料を約4℃から約室温までの温度範囲でインキュ ベーションすることを特徴とする、請求項23記載の方法。 25.工程2)において、前記ターゲット分子と会合している前記油体を、遠心 分離、浮上又はサイズ排除によって、前記試料から分離することを特徴とする、 請求項1から24のいずれか一項記載の方法。 26.さらに、3)前記油体から前記ターゲット分子を分離する工程を含むこと を特徴とする、請求項1から25のいずれか一項記載の方法。 27.前記ターゲット分子を、適切な条件下の溶出によって分離することを特徴 とする、請求項26記載の方法。 28.前記油体が、ナタネ(Brassica spp.)、ダイズ(Glycin e max )、ヒマワリ(Helianthus annuus)、ギネアアブラ ヤシ(Elaeis uineeis)、ココヤシ(Cocus nucifer a )、トウゴマ(Ricinus communis)、ベニバナ(Cartham us tinctorius )、アブラナ(Brassica spp.及びSi napis alba )、コエンドロ(Coriandrum sativum) 、アマニ/アマ(Linum usitatissimum)、シロイヌナズナ(Arabidopsis thalian a)及びトウモロコシ(Zeamay s )からなる植物の群から得られることを特徴とする、請求項1から27のいず れか一項記載の方法。 29.分子と会合している油体を含むことを特徴とする、組成物。 30.前記分子が、有機分子、脂質、糖質、核酸、細胞、細胞小器官、細胞成分 、ウイルス、金属、金属イオン及びイオンからなる群より選ばれていることを特 徴 とする、請求項29記載の組成物。 31.リガンド分子が更に含まれており、前記リガンド分子が前記油体及び前記 ターゲット分子と会合していることを特徴とする、請求項30記載の組成物。 32.前記分子がリガンド分子であることを特徴とする、請求項29記載の組成 物。 33.前記リガンド分子が、前記油体に共有結合的に付着していることを特徴と する、請求項31又は32記載の組成物。 34.前記リガンドがビオチンであることを特徴とする、請求項33記載の組成 物。 35.試料からターゲット分子を分離する際に用いるアフィニティマトリクスで あって、 前記アフィニティマトリクスが、前記ターゲット分子と会合し得る油体を含む ことを特徴とする、アフィニティマトリクス。 36.試料からターゲット分子を分離する際に用いるアフィニティマトリクスで あって、 前記アフィニティマトリクスが、(a)油体と、(b)前記油体と会合してい るリガンド分子とを含んでおり、前記リガンド分子が前記ターゲット分子と会合 し得ることを特徴とする、アフィニティマトリクス。
