Claims (1)
- 【特許請求の範囲】 1. 一方では、ラクトフェリン、特に哺乳類のラクトフェリン、好まし くはウシ、ブタ、ヤギ、ヒトラクトフェリン、又は由来タンパク質をコードする cDNAを、また他方では、当該cDNAによりコードされるラクトフェリン又は由来タ ンパク質を植物細胞に生成されることに必要な要素、特に植物細胞の転写機械が 認識する転写プロモーター及びターミネーターを含む組換え核酸配列を、細胞か らもしくはこれから取得した植物から、ラクトフェリン又は由来タンパク質を得 る目的として、植物細胞を形質転換する為の利用。 2. 請求項1記載の組換え核酸配列の利用において、利用された核酸配 列は組換えポリペプチドの分泌に必要なシグナルペプチドをコードする配列を含 むことを特徴とする利用。 3. 請求項2記載の組換え核酸配列の利用において、利用されたシグナ ルペプチドはサツマイモのスポラミンAのシグナルペプチドであることを特徴と する利用。 4. 一方では、ラクトフェリン、特に哺乳類のラクトフェリン、好まし くはウシ、ブタ、ヤギ、ヒトラクトフェリン、又は由来タンパク質をコードする 配列を、また他方では、当該配列によりコードされるラクトフェリン又は由来タ ンパク質を植物細胞に生成されることに必要な要素を、特に植物細胞の転写機械 が認識する転写プロモーター及びター ミネーターを含むことを特徴とする組換え核酸配列。 5. 請求項4記載の組換え核酸配列において、組換えポリペプチドの分 泌に必要なシグナルペプチドをコードする配列を含むことを特徴とする核酸配列 。 6. 請求項5記載の組換え核酸配列において、利用されたシグナルペプ チドはサツマイモのスポラミンAのシグナルペプチドであることを特徴とする核 酸配列。 7. 場合によっては、その複製に影響を与えない部位に挿入した、請求 項4から6記載の核酸配列を含むベクター、特にプラスミド。 8. 請求項7記載のベクターによって形質転換した宿主細胞、特にアグ ロバクテリウム・ツメファシェンス等のあらゆるバクテリア。 9. ラクトフェリン、特にヒトラクトフェリン又は由来タンパク質の取 得方法において、 − 特にそれ自身は、請求項7記載のベクターによって形質転換された、 請求項8記載の宿主細胞によって、請求項4から6記載の組換え配列を細胞のゲ ノムに組込むように植物細胞を形質転換すること、 − 場合によっては、上記の形質転換細胞からの形質転換植物を取得する こと、 − 特に抽出と、場合により、その後の精製によって、当該上記形質転換 した植物又は細胞内のラクトフェリン、特にヒトラクトフェリン又は由来タンパ ク質を回収すること、 から成ることを特徴とする取得方法。 10. 請求項4から6記載の一つ(又は複数)の組換え核酸配列を含み 、ゲノム内に安定した形式で組込んだことを特徴とする、特にアブラナ、タバコ 、トウモロコシ、エンドウマメ、トマト、ニンジン、コムギ、オオムギ、ジャガ イモ、ダイズ、ヒマワリ、レタス、コメ、ウマゴヤシ及びテンサイの中から選択 する遺伝形質転換の植物、植物抽出物又は植物の部分、特に葉、及び/又は果実 及び/又は種子、及び/又は植物細胞。 11. 請求項9記載の方法によって取得されたことを特徴とするラクト フェリン、特にヒトラクトフェリン又は由来タンパク質。 12. 本発明によるラクトフェリン、特にヒトラクトフェリン又は由来 タンパク質を含むことを特徴とする特にアブラナ、タバコ、トウモロコシ、エン ドウマメ、トマト、ニンジン、コムギ、オオムキ、ジャガイモ、ダイズ、ヒマワ リ、レタス、コメ、ウマゴヤシ及びテンサイの中から選択した遺伝形質転換の植 物、植物抽出物又は植物の部分、特に葉、及び/又は果実及び/又は種子、及び /又は植物細胞。 13. 医薬、医用、歯科、化粧又はバイオテクノロジー組成物を得る為 に、請求項10又は12記載の植物、植物抽出物又は植物部分、 及び/又はタンパク質(ラクトフェリン、特にヒトラクトフェリン又は由来タン パク質)の利用。 14. 請求項10又は12記載の植物、植物抽出物、植物の部分及び/ 又は請求項11記載のタンパク質(ラクトフェリン、特にヒトラクトフェリン又 は由来タンパク質)から成ることを特徴とするバイオ物質又は医薬、医用、歯科 、化粧又はバイオテクノロジー組成物。
