Abstract

Proceso para la producción de ácido hialurónico en Bacillus subtilis, que comprende los siguientes etapas: (a) cultivar células bacterianas huésped de Bacillus subtilis transformadas con el sistema grac-lac, en condiciones adecuadas para la producción de ácido hialurónico, y en presencia de isopropil-ß-tiogalactopiranósido (IPTG) como inductor, en donde dichas células bacterianas huésped se caracterizan por estar transformadas con: (i) al menos un vector de plásmido episómico que comprende una secuencia que codifica para la enzima hialuronano sintasa, una secuencia que codifica para la enzima UDP-glucosa deshidrogenasa en tándem, bajo el control del promotor fuerte inducible Pgrac que usa el represor lac, o (ii) al menos un vector de plásmido episómico que comprende una secuencia que codifica para la enzima hialuronano sintasa, una secuencia que codifica para la enzima UDP-glucosa deshidrogenasa, una secuencia que codifica para la enzima UDP-glucosa pirofosforilasa, y una secuencia que codifica para la enzima glucosa 6 fosfato isomerasa, bajo el control del promotor fuerte inducible Pgrac que usa el represor lac; (b) recuperar el ácido hialurónico del medio de cultivo; en donde dichas células huésped bacterianas de Bacillus subtilis transformadas con el vector de plásmido (i) o (ii) capaces de producir ácido hialurónico de la etapa a) se preseleccionan en la placa en gradiente de IPTG.
