Las caracterizaciones de TLC analíticas se llevaron a cabo con placas de 0,2 mm: Merck, gel de sílice 60 F254. La elución se realiza con EA, hept, DCM, MeOH o mezclas de los mismos. La detección se realiza con UV o con una solución de KMnO4 (3 g), K2CO3 (20 g), 5 % de NaOH (3 ml) y H2O (300 ml) con calentamiento posterior.
Las CC se realizan utilizando gel de sílice Brunschwig 60A (0,032-0,63 mm); la elución se realiza con EA, Hept, DCM, MeOH o mezclas de los mismos. En los casos de los compuestos que contienen una función ácida, 1 % de AcOH se añade al o a los eluyentes.
Los compuestos se caracterizan por RMN 1H (300 MHz) (Varian Oxford), o mediante RMN 1H (400 MHz) (Bruker Advance 400). Los desplazamientos químicos δ se proporcionan en ppm con relación al disolvente utilizado; multiplicidades: s = singlete, d = doblete, t = triplete, q = cuadruplete, p = pentuplete, hex = sextuplete, hep = heptuplete, m = multiplete, a = amplio, las constantes de acoplamiento se proporcionan en Hz. Alternativamente, los compuestos se caracterizan por CL-EM (Sciex API 2000 con Bomba Binaria Agilent 1100 con DAD y ELSD o un Agilent cuadrupolo EM 6140 con Bomba Binaria Agilent 1200, DAD y ELSD); por TLC (placas de TLC de Merck, gel de sílice 60 F254 ), o por punto de fusión. Los compuestos se purifican mediante cromatografía sobre gel de sílice 60A. NH4OH como se usa para CC es 25 % ac.

El compuesto del título se preparó en analogía con el Ejemplo 2, partiendo del anhídrido del ácido 1-ciclopropil-7fluoro-1,4-dihidro-8-metoxi-4-oxo-3-quinolinacarboxílico con ácido bórico (1:1) y con ácido acético (1:2) (101 mg; preparado de acuerdo con el documento WO 2010/056633) y 6-[(5R)-5-[(3-azetidinilamino)-metil]-2-oxo-3oxazolidinil]-2H-1,4-benzotiazin-3(4H)-ona (preparada de acuerdo con el documento WO 2008/126034; 167 mg). Al final de la reacción, la mezcla de reacción se concentró a presión reducida y el residuo se concentró en MeOH y se trató con HCl 1N en MeOH (3 ml) durante 15 min. La mezcla de reacción se diluyó con agua (2 ml) y el sólido se filtró. La fase orgánica se concentró a presión reducida y se purificó mediante CC (DCM/MeOH 9:1 seguido por DCM/MeOH 9:1+1 % de AcOH), para proporcionar después de la evaporación y agitación del residuo con MeOH/TBME un sólido de color beis. RMN 1H (DMSO-d6) δ: 11,40 (s, 1H); 11,22 (s, 1H); 8,93 (s, 1H), 8,31 (d, J = 8,9 Hz, 1H); 7,62-7,56 (m, 1H); 7,467,39 (m, 1H); 6,75 (d, J = 8,9 Hz, 1H); 4,60 (s, 2H); 4,53-4,36 (m, 4H); 4,32-4,15 (m, 4H); 3,83 (dd, J = 6,9 Hz, J = 10,5 Hz, 1H); 3,48-3,24 (m, 4H); 1,38 (t, J = 7,0 Hz, 3H), EM (IEN, m/z): 536,19 [M+H+] para C25H25N7O7; tR = 0,92 min (EM1).
Ejemplo 10: Ácido 1-ciclopropil-4-oxo-7-(3-{[(R)-2-oxo-3-(3-oxo-3,4-dihidro-2H-benzo[1,4]tiazin-6-il)-oxazolidin -5-ilmetil]-amino}-azetidin-1-il)-1,4-dihidro-[1,8]naftiridin-3-carboxílico:
10.1. Ácido 7-cloro-1-ciclopropil-4-oxo-1,4-dihidro-[1,8]naftiridin-3-carboxílico:
Una suspensión de éster etílico del ácido 7-cloro-1-ciclopropil-1,4-dihidro-4-oxo-1,8-naftiridin-3-carboxílico (preparada de acuerdo con el documento EP 607825; 500 mg) en HCl 6N (6 ml) se agitó a 100 ºC durante 30 min. La mezcla de reacción se dejó que alcanzara ta y los cristales resultantes se recolectaron mediante filtración y se lavaron secuencialmente con agua y MeOH, para proporcionar un sólido de color beis. RMN 1H (DMSO-d6) δ: 11,40 (s, 1H), 8,81 (s, 1H); 8,70 (d, J = 8,4 Hz, 1H); 7,78 (d, J = 8,4 Hz, 1H); 3,85-3,74 (m, 1H); 1,27-1,07 (m, 4H), EM (IEN, m/z): 265,1 [M+H+] para C12H9N2O3Cl; tR = 0,7 min (EM3).

21.3. éster terc-butílico del ácido (RS)-3-{[(S)-3-(5-etoxicarbonilmetoxi-6-nitro-piridin-2-il)-2-oxo-oxazolidin-5-ilmetil]amino}-pirrolidin-1-carboxílico:
El compuesto del título se preparó en analogía con el Ejemplo 17, etapa 17.1, partiendo de (6-bromo-2-nitropiridin-3iloxi)acetato de etilo (6,84 g; preparado tal como se describió en WO 2007/118130) y el intermedio 21.2 (6,4 g), para proporcionar una espuma de color amarillento (11,34 g; 99 % de rendimiento). EM (IEN, m/z): 454,08 [M+H+] para C22H31N5O9; tR = 0,70 min (EM2).
21.4. éster terc-butílico del ácido (RS)-3-{[(S)-2-oxo-3-(3-oxo-3,4-dihidro-2H-pirido[3,2-b][1,4]oxazin-6-il)-oxazolidin-5ilmetil]-amino}-pirrolidin-1-carboxílico:
En analogía con el Ejemplo 17, etapa 17.4, el compuesto del título se preparó a partir del intermedio 21.3 (11,34 g), para proporcionar una espuma de color amarillento (7,13 g; 74 % de rendimiento). EM (IEN, m/z): 434,16 [M+H+] para C20H27N5O6; tR = 0,57 min (EM2).
21.5. Diclorhidrato de 6-((S)-2-oxo-5-(((RS)-pirrolidin-3-ilamino)metil)oxazolidin-3-il)-2H-pirido[3,2-b][1,4]oxazin3(4H)-ona:
En analogía con el Ejemplo 17, etapa 17.5, el compuesto del título se preparó a partir del intermedio 21.4 (7,0 g), para proporcionar un sólido de color amarillento (6,7 g, 100 % de rendimiento). EM (IEN, m/z): 334,22 [M+H+] para C15H21N5O4Cl2; tR = 0,40 min (EM2).
21.6. Ácido 1-ciclopropil-6-fluoro-4-oxo-7-((RS)-3-{[(S)-2-oxo-3-(3-oxo-3,4-dihidro-2H-pirido[3,2-b][1,4]oxazin-6-il)oxazolidin-5-ilmetil]-amino}-pirrolidin-1-il)-1,4-dihidro-[1,8]naftiridin-3-carboxílico:
En analogía con el Ejemplo 2, a partir del intermedio 21,5 (406 mg) y el ácido 7-cloro-1-ciclopropil-6-fluoro-4-oxo-1,4dihidro-[1,8]naftiridin-3-carboxílico (comercial; 283 mg), el compuesto del título se obtuvo en forma de un sólido de color beis (mezcla de diastereómeros; 398 mg; 68 % de rendimiento). EM (IEN, m/z): 580,07 [M+H+] para C27H26N7O7F; tR = 0,62 min (EM2).
Ejemplo 22: Ácido 1-ciclopropil-6-fluoro-4-oxo-7-[(S)-3-({[(S)-2-oxo-3-(3-oxo-3,4-dihidro-2H-pirido[3,2b][1,4]oxazin-6-il)-oxazolidin-5-ilmetil]-amino}-metil)-pirrolidin-1-il]-1,4-dihidro-[1,8]naftiridin-3-carboxílico:
22.1. éster terc-butílico del ácido (S)-3-[((S)-3-benciloxicarbonilamino-2-hidroxi-propilamino)-metil]-pirrolidin-1carboxílico:
El compuesto del título se preparó en analogía con el Ejemplo 17, etapa 17.1, partiendo de éster bencílico del ácido [(2R)-oxiranilmetil]-carbámico (5,17 g; preparado de acuerdo con el documento WO 2004/002973) y éster tercbutílico del ácido (S)-3-aminometilpirrolidin-1-carboxílico (5,00 g; comercial) en MeCN (120 ml), para proporcionar un aceite incoloro (6,4 g; 63 % de rendimiento). EM (IEN, m/z): 408,21 [M+H+] para C21H33N3O5; tR = 0,66 min (EM2).
22.2. éster terc-butílico del ácido (S)-3-{[((S)-2-oxo-oxazolidin-5-ilmetil)-amino]-metil}-pirrolidin-1-carboxílico:
Una solución del intermedio 22.1 (6,4 g) en THF (100 ml) se trató a 0 ºC con KOtBu (1,76 g) y se agitó adicionalmente a ta durante 1 h. La mezcla de reacción se concentró al vacío y el residuo se recogió en EA/agua. La fase ac. se extrajo dos veces con EA. Las fases org combinadas se lavaron con salmuera, se secaron sobre MgSO4, se filtraron, se evaporaron y se purificaron mediante CC (EA/MeOH 19:1 a 9:1 +1 % de NH4OH), para proporcionar un aceite incoloro (4,34 g; 92 % de rendimiento). EM (IEN, m/z): 300,17 [M+H+] para C14H25N3O4; tR = 0,50 min (EM2).
22.3. éster terc-butílico del ácido (S)-3-({[(S)-3-(5-etoxicarbonilmetoxi-6-nitro-piridin-2-il)-2-oxo-oxazolidin-5-ilmetil]amino}-metil)-pirrolidin-1-carboxílico:
El compuesto del título se preparó en analogía con el Ejemplo 17, etapa 17.3, partiendo de (6-bromo-2-nitropiridin-3iloxi)acetato de etilo (4,01 g; preparado de acuerdo con el documento WO 2007/118130) y el intermedio 22.2 (3,94 g), para proporcionar una espuma de color amarillento (7,01 g; 100 % de rendimiento). EM (IEN, m/z): 524,22 [M+H+] para C23H33N5O9; tR = 0,71 min (EM2).
