Abstract
Verfahren zur Herstellung von enantiomerenangereicherte Aminen durch Umsetzung eines Ketons mit Ammoniak oder einem Ammoniumsalz und einem Reduktionsmittel in Gegenwart eines katalytischen Systems mit den Komponenten: a) eine Aminosäuretransaminase, b) eine alpha-Aminosäure, die ein Substrat der Aminosäuretransaminase ist, c) eine zur Herstellung der alpha-Aminosäure geeignete Aminosäuredehydrogenase, d) ein NAD(P)+ und e) reduzierendes Enzym, das NAD(P)+ mit dem Reduktionsmittel zu NAD(P)H umsetzt. Das Verfahren kann mit katalytischen Mengen an alpha-Aminosäure und NAD(P)+ durchgeführt werden und ermöglicht eine enantioselektive reduktive Aminierung von Ketonen.
Description
Verfahren zur Herstellung von enantiomerenangereicherten
Aminen
Enantiomerenreine Amine finden Anwendung als chirale Bausteine, sogenannte „Chiral Building Blocks", bei der Herstellung von pharmazeutischen und Agrowirkstoffen .
Prominente Beispiele dafür sind Rivastigmin, Chephalosporin und chirale 1-Amino-l-arylalkane . Einzelne Vertreter dieser Verbindungen werden bereits in Mengen von mehr als 1000 Tonnen produziert. Für die Herstellung von pharmazeutischen und Agrowirkstoffen werden optisch reine Amine benötigt, da nur entweder das (R)- oder das (S) -Enantiomer die erwünschte Wirkung erzielt. In einigen Fällen kann von dem nicht erwünschten Enantiomer sogar eine schädliche Wirkung ausgehen. Deshalb besteht ein Bedarf an Verfahren zur Herstellung von enantiomerenangereicherten Aminen.
Der bisher am meisten genutzte Weg zu enantiomerenangereicherten oder enantiomerenreinen Aminen ist die Racematspaltung ausgehend vom Amin in racemischer Form. Klassisch erfolgt die Racematspaltung über diastereomere Salze. Dazu werden stöchiometrische Mengen einer chiralen Carbonsäure zum Amin in racemischer Form gegeben und die resultierenden diastereomeren Salze werden dann durch fraktionierende Kristallisation getrennt. Danach muss die chirale Carbonsäure wieder abtrennt werden und in der Regel aus Kostengründen recycliert werden. Das unerwünschte Enantiomer muss entweder entsorgt oder in das Racemat überführt und in den Produktionsprozeß zurückgeführt werden. Diese Schritte sind mit erheblichem Aufwand und Kosten verbunden.
Eine Alternative zur Racematspaltung über diastereomere Salze ist die biokatalytische Racematspaltung, bei der mittels eines Enzyms aus einem racemischen Amin enantioselektiv ein Aminderivat erzeugt wird oder aus einem racemischen Aminderivat enantioselektiv Amin freigesetzt
wird. Hierzu können Lipasen, Acylasen, Proteasen und viele andere Hydrolasen eingesetzt werden, von denen bekannt ist, dass sie Racemate stereospezifisch spalten können. Solche Verfahren sind bekannt aus Bornscheuer und Kazlauskas, Hydrolases in Organic Synthesis (2005), Wiley-VCH Weinheim, sowie Enzyme Catalysis in Organic Synthesis, 2nd Edition (2002 ), ed. Drauz und Waldmann, Wiley-VCH Weinheim.
Bei diesen Verfahren entfallt der Einsatz stochiometrischer Mengen an chiralen Hilfsreagenzien . Es bleibt aber der Nachteil einer Limitierung der theoretischen Ausbeute auf 50% bezogen auf den als Racemat vorliegenden Ausgangsstoff und gegebenenfalls der zusatzliche Arbeitsschritt für das Recycling des ungewunschten Enantiomers mit den damit verbundenen Kosten.
Diese Nachteile, die prinzipiell für alle
Racematspaltungstrategien gelten, lassen sich durch eine asymmetrische Synthese unter Verwendung von prochiralen Ausgangsverbindungen vermeiden. Die bekannten asymmetrischen Synthesen unter Verwendung von ubergangsmetallhaltigen Katalysatoren erreichen allerdings oft nicht die erforderliche Enantioselektivitat . Darüber hinaus können sich aus der Verwendung der ubergangsmetallhaltigen Katalysatoren auch für pharmazeutische Anwendungen unerwünschte Gehalte an Ubergangsmetallen im resultierenden Produkt ergeben.
Ein weiterer bekannter enzymatischer Weg zu enantiomerenangereicherten Aminen ist der durch Transaminasen katalysierte Austausch von Ketogruppe und Aminogruppe zwischen zwei Substraten.
Die bekannten Transaminasen sind enantioselektiv sowohl hinsichtlich des eingesetzten Amingruppendonors als auch
hinsichtlich des gebildeten Amins. Ausgehend von prochiralen Ketonen werden bei der Reaktion deshalb enantiomerenangereicherte Amine gebildet. Nachteilig ist allerdings, dass es sich bei der Umsetzung um eine Gleichgewichtsreaktion handelt und deshalb in der Regel nur ein Teil des eingesetzten prochiralen Ketons in das gewünschte enantiomerenangereicherte Amin umgesetzt wird.
Aus Matcham et al . , Chimica Oggi 14 (1996) 20-24; Matcham et al., Chimia 53 (1999) 584-589; sowie WO99/46398 ist bekannt, dass sich das Gleichgewicht der Reaktion auf die gewünschte Produktseite verschieben lässt, wenn Isopropylamin als Aminsubstrat verwendet wird (R3, R4 = CH3) und das bei der Reaktion gebildete Aceton destillativ aus der Reaktionsmischung entfernt wird. Das Verfahren kann allerdings nur mit Transaminasen durchgeführt werden, die Isopropylamin als Amingruppendonor akzeptieren. Unter den Prozessbedingungen, die für die Entfernung von Aceton erforderlich sind, sind allerdings nur wenige Transaminasen ausreichend stabil.
In Shin et al . , Biotechnology and Bioengineering 65 (1999) 206-211 und EP 1 818 411 wird vorgeschlagen, das Gleichgewicht durch die Entfernung des gebildeten Ketons zu verschieben, indem Alanin als Aminsubstrat eingesetzt wird und das daraus gebildete Pyruvat enzymatisch mit einer gleichzeitig eingesetzten Pyruvatdecarboxylase,
Lactatdehydrogenase oder Acetolactatsynthase weiter umgesetzt wird. Die Beschränkung auf Alanin als Aminsubstrat schränkt die Wahl der Transaminasen allerdings deutlich ein. Um enantiomerenangereicherte Amine mit (R) -Konfiguration herzustellen, sind (R) -selektive
Transaminasen erforderlich und das Aminsubstrat muss ebenfalls in der (R) -Form vorliegen. Das setzt bei diesem Verfahren die Verwendung von D-Alanin voraus, welches nur mit zusätzlichem Aufwand zugänglich ist. Die Alternative ist die Verwendung von DL-Alanin, dabei verbleibt aber
L-Alanin im Reaktionsgemisch und muss abgetrennt und entsorgt werden.
US 3,183,170 beschreibt ein Verfahren zur Herstellung von L-Aminosäuren durch Umsetzung einer alpha-Ketocarbonsäure mit L-Glutaminsäure in Gegenwart einer
L-Glutaminsäuredehydrogenase, einer Hydrogenase, einer Transaminase, eines Wasserstoffakzeptorfarbstoffs, eines Elektronentransportsystems, einer Stickstoffquelle und von gasförmigem Wasserstoff. Das Verfahren hat den Nachteil, dass die verwendeten Wasserstoffakzeptorfarbstoffe starke Zellgifte sind. Außerdem ist das Verfahren beschränkt auf die aminosäurespezifische L-Glutaminsäuredehydrogenase und kann nicht mit Aminosäuredehydrogenasen angewendet werden, die unterschiedliche Aminosäuren als Substrat tolerieren.
Es besteht deshalb Bedarf an einem Verfahren, mit dem sich enantiomerenangereicherte Amine aus Ketonen herstellen lassen und das nicht die Nachteile der aus dem Stand der Technik bekannten Verfahren aufweist.
Diese Aufgabe wird gelöst durch das erfindungsgemäße Verfahren zur Herstellung eines enantiomerenangereicherten Amins, bei dem ein Keton mit Ammoniak oder einem Ammoniumsalz und einem Reduktionsmittel umgesetzt wird in Gegenwart eines katalytischen Systems, das folgende Komponenten umfasst: a) eine Aminosäuretransaminase, b) eine alpha-Aminosäure, die ein Substrat der Aminosäuretransaminase ist, c) eine zur Herstellung der alpha-Aminosäure geeignete Aminosäuredehydrogenase, d) NAD(P)+ und e) ein NAD(P)+ reduzierendes Enzym, das NAD(P)+ mit dem Reduktionsmittel zu NAD(P)H umsetzt.
Aminosäuretransaminasen im Sinne der Erfindung sind amingruppentransferierende Enzyme der Enzymklasse
E. C. 2.6.1.X, die eine alpha-Aminosäure als Substrat akzeptieren .
Aminosäuredehydrogenasen im Sinne der Erfindung sind Enzyme, die alpha-Ketocarbonsäuren mit Ammoniak zu alpha- Aminosäuren umsetzen und gleichzeitig NAD(P)H zu NAD(P)+ oxidieren .
NAD(P)+ steht sowohl für Nicotinamidadenindinucleotid (NAD+) und dessen Salze als auch für
Nicotinamidadenindinucleotidphosphat (NADP+) und dessen Salze. In gleicher Weise steht NAD(P)H sowohl für
Dihydronicotinamidadenindinucleotid (NADH) und dessen Salze als auch für Dihydronicotinamidadenindinucleotidphosphat (NADPH) und dessen Salze.
Das erfindungsgemäße Verfahren koppelt drei enzymatisch katalysierte Reaktionen zu einem Gesamtverfahren. In einer ersten durch eine Aminosäuretransaminase katalysierten, Reaktion wird das Keton mit einer alpha-Aminosäure zum enantiomerenangereicherten Amin und der der alpha- Aminosäure entsprechenden alpha-Ketocarbonsäure umgesetzt. In einer zweiten durch eine Aminosäuredehydrogenase katalysierten Reaktion wird die alpha-Ketocarbonsäure mit Ammoniak oder einem Ammoniumsalz sowie NAD(P)H umgesetzt unter Rückbildung der alpha-Aminosäure und Bildung von NAD(P)+. In einer dritten durch ein NAD(P)+ reduzierendes Enzym katalysierten Reaktion wird NAD(P)+ mit einem
Reduktionsmittel umgesetzt und so NAD(P)H zurückgebildet. Als Gesamtreaktion resultiert eine enantioselektive reduktive Aminierung des Ketons, bei der die alpha- Aminosäure und NAD(P)+ nur in katalytischen Mengen benötigt werden und für die Herstellung des enantiomerenangereicherten Amins nicht verbraucht werden.
Das nachfolgende Formelschema zeigt die Kopplung der drei enzymatischen Reaktionen zum erfindungsgemäßen Verfahren am Beispiel von Ammoniumformiat als Reduktionsmittel und
Ammoniumsalz und Formiatdehydrogenase als NAD+ reduzierendem Enzym. Dabei bezeichnet ASTA eine Aminosauretransaminase, ASDH eine Aminosauredehydrogenase und FDH eine Formiatdehydrogenase.

Gegenstand der vorliegenden Erfindung ist somit auch die Verwendung eines katalytischen Systems, umfassend
a) eine Aminosauretransaminase,
b) eine alpha-Aminosaure, die ein Substrat der Aminosauretransaminase ist,
c) eine zur Herstellung der alpha-Aminosaure geeignete Aminosauredehydrogenase,
d) NAD(P)+ und
e) ein NAD(P)+ reduzierendes Enzym, das NAD(P)+ mit dem Reduktionsmittel zu NAD(P)H umsetzt,
zur Herstellung eines enantiomerenangereicherten Amins aus einem Keton.
Für das erfindungsgemäße Verfahren können prinzipiell alle zur Umsetzung von Ketonen geeigneten Transaminasen verwendet werden. Geeignete Aminosäuretransaminasen sind aus Yun et al . , Applied and Environmental Microbiology 70 (2004) 2529-2534; Shin et al . , Bioscience, Biotechnology, and Biochemistry 65 (2001) 1782-1788; Kaulmann et al . , Enzyme and Microbial Technology 41 (2007) 628-637; Shin et al . , Applied Microbiology and Biotechnology 61 (2003) 463- 471; Shin et al . , Biotechnology and Bioengineering 65 (1999) 206-211; Matcham et al . , Chimia 53 (1999) 584-589; sowie WO 99/46398 bekannt.
Vorzugsweise werden omega-Transaminasen verwendet, die aus EP 0 404 146 bekannt sind. Besonders geeignet sind omega- Transaminasen aus Vibrio fluvialis, insbesondere Vibrio fluvialis Stamm JS17; Alcaligenes denitrificans , insbesondere Alcaligenes denitrificans Stamm Y2K-2; Klebsiella pneumoniae, insbesondere Klebsiella pneumoniae Stamm YS2F; sowie Bacillus thuringiensis, insbesondere Bacillus thuringiensis Stamm JS64.
Als alpha-Aminosäure kann jede alpha-Aminosäure eingesetzt werden, die ein Substrat der Aminosäuretransaminase ist. Vorzugsweise wird als alpha-Aminosäure eine proteinogene Aminosäure verwendet, wobei sowohl die natürlichen L-Aminosäuren als auch deren D-Enantiomere oder beliebige Mischungen der Enantiomere, wie z. B. eine racemische
Mischung, verwendet werden können. Besonders bevorzugte Aminosäuren sind Leucin, Alanin, Phenylalanin und Glutaminsäure .
Die alpha-Aminosäure kann bei dem erfindungsgemäßen Verfahren in einer katalytischen Menge eingesetzt werden. Vorzugsweise liegt die Gesamtmenge an alpha-Aminosäure und der alpha-Aminosäure entsprechender alpha-Ketosäure, bezogen auf die Gesamtmenge an Keton, im Bereich von 1 bis 50 mol-%, vorzugsweise 2 bis 10 mol-%.
In einer bevorzugten Ausführungsform des erfindungsgemäßen Verfahrens wird zu Beginn der Umsetzung nicht die alpha- Aminosäure vorgelegt, sondern die der alpha-Aminosäure entsprechende alpha-Ketocarbonsäure, die an Stelle der Aminogruppe eine Ketogruppe aufweist. Diese Ausführungsform ist besonders vorteilhaft für die Herstellung von enantiomerenangereicherten Aminen, die unter Verwendung einer D-Aminosäure oder einer nicht proteinogenen L-Aminosäure hergestellt werden müssen, da an Stelle einer schwer zugänglichen Aminosäure die der Aminosäure entsprechende leichter zugängliche alpha-Ketocarbonsäure für das Verfahren eingesetzt werden kann.
Als Aminosäuredehydrogenase kann jede NAD (P) H-Cofaktorabhängige Dehydrogenase verwendet werden, mit der sich die alpha-Aminosäure aus der der alpha-Aminosäure entsprechenden alpha-Ketocarbonsäure herstellen lässt. Geeignete Aminosäuredehydrogenasen sind aus Oshima et al, International Industrial Biotechnology 9 (1989) 5-11; Ohsima et al . , European Journal of Biochemistry 191 (1990) 715-720; Khan et al . , Bioscience, Biotechnology and
Biochemistry 69 (2005) 1861-1870; Hummel et al . , Applied Microbiology and Biotechnology 26 (1987) 409-416 und Bommarius in Enzyme Catalysis in Organic Synthesis, 2nd Edition (2002) , ed. Drauz und Waldmann, Wiley-VCH Weinheim bekannt.
Vorzugsweise ist die Aminosäuredehydrogenase eine Leucindehydrogenase, eine Alanindehydrogenase, eine Phenylalanindehydrogenase oder eine Glutamatdehydrogenase. Besonders geeignet sind Alanindehydrogenasen aus Bacillus sphaericus, insbesondere Bacillus sphaericus Stamm DSM642; Glutamatdehydrogenasen aus Bacillus subtilis, insbesondere Bacillus subtilis Stamm ISW1214; Phenylalanindehydrogenasen aus Rhodococcus sp . , insbesondere Rhodococcus sp. Stamm M4, Bacillus sphaericus und Thermoactinomyces intermedius . Besonders bevorzugt wird eine Leucindehydrogenase aus
Bacillus cereus verwendet. Für die Herstellung von D-Aminosäuren wird vorzugsweise eine
D-Aminosäuredehydrogenase verwendet, die in Vedha-Peters et al . , Journal of the American Chemical Society 128 (2006) 10923-10929 beschrieben ist als Mutante der meso-
Diaminopimelinsäure-D-dehydrogenase von Corynebacterium glutamicum.
Vorzugsweise werden eine Aminosäuretransaminase und eine Aminosäuredehydrogenase in Kombination verwendet, die in ihrer Stereoselektivität bezüglich der alpha-Aminosäure aufeinander abgestimmt sind, d.h. dass entweder die Aminosäuredehydrogenase die der alpha-Aminosäure entsprechende alpha-Ketosäure selektiv zum S-Enantiomer der alpha-Aminosäure umsetzt und die Aminosäuretransaminase selektiv das S-Enantiomer der alpha-Aminosäure mit dem
Keton umsetzt oder dass die Aminosäuredehydrogenase die der alpha-Aminosäure entsprechende alpha-Ketosäure selektiv zum R-Enantiomer der alpha-Aminosäure umsetzt und die Aminosäuretransaminase selektiv das R-Enantiomer der alpha-Aminosäure mit dem Keton umsetzt.
Bei dem erfindungsgemäßen Verfahren wird außerdem ein Reduktionsmittel eingesetzt in Kombination mit einem NAD(P)+ reduzierenden Enzym, das NAD(P)+ mit dem Reduktionsmittel zu NAD(P)H umsetzt. Geeignete NAD(P)+ reduzierende Enzyme sind aus Enzyme Catalysis in Organic Synthesis, 2nd Edition (2002) , ed. Drauz und Waldmann, Wiley-VCH Weinheim bekannt.
In einer bevorzugten Ausführungsform wird als Reduktionsmittel ein Salz der Ameisensäure und als NAD(P)+ reduzierendes Enzym eine Formiatdehydrogenase verwendet. Besonders bevorzugt ist das Reduktionsmittel Ammoniumformiat, das auch in situ aus Ameisensäure und Ammoniak erzeugt werden kann. Vorzugsweise wird eine Formiatdehydrogenase aus Candida boidinii oder eine davon abgeleitete Mutante verwendet. Ebenfalls geeignet sind die
von Tishkov et al . in Biomolecular Engineering 23 (2006) 89-11 beschriebenen Formiatdehydrogenasen . Diese Ausführungsform hat den Vorteil, dass als Reaktionsprodukt des Reduktionsmittels nur Kohlendioxid entsteht, was die Aufarbeitung der Reaktionsmischung vereinfacht.
In einer weiteren bevorzugten Ausführungsform wird als Reduktionsmittel Glucose und als NAD(P)+ reduzierendes Enzym eine Glucosedehydrogenase verwendet. Besonders geeignet sind Glukosedehydrogenasen aus Bacillus subtilis, Bacillus megaterium und Thermoplasma acidophilum.
Alternativ kann als Reduktionsmittel ein Salz der phosphorigen Säure und als NAD(P)+ reduzierendes Enzym eine Phosphitdehydrogenase verwendet werden. Geeignete Phosphitdehydrogenasen sind aus Relyea et al . , Bioorganic Chemistry 33 (2005) 171-189 bekannt.
Ebenfalls geeignet ist Glucose-6-phosphat als Reduktionsmittel in Kombination mit einer Glucose- 6-phosphatdehydrogenase als NAD(P)+ reduzierendem Enzym.
Bei dem erfindungsgemäßen Verfahren können NAD(P)+ bzw. NAD(P)H in katalytischen Mengen eingesetzt werden.
Vorzugsweise liegt die Gesamtmenge an NAD(P)+ und NAD(P)H, bezogen auf die Gesamtmenge an Keton, im Bereich von 0,001 bis 5 mol-%, vorzugsweise 0,01 bis 1 mol-%. Zu Beginn der Umsetzung kann wahlweise sowohl NAD(P)+ als auch NAD(P)H vorgelegt werden.
Die im erfindungsgemäßen Verfahren eingesetzten Enzyme, d.h. die Aminosäuretransaminase, Aminosäuredehydrogenase und das NAD(P)+ reduzierende Enzym können im Verfahren sowohl gelöst als auch auf einem Träger immobilisiert eingesetzt werden. In einer bevorzugten Ausführungsform werden die Enzyme in Form eines Ganzzellkatalysators eingesetzt, d.h. in Form einer Zelle, die alle drei Enzyme exprimiert. Vorzugsweise wird ein rekombinanter
Ganzzellkatalysators verwendet, d.h. eine Zelle, die gentechnisch so verändert wurde, dass sie mindestens eines der drei Enzyme aus einer nicht nativen Gensequenz exprimiert. Besonders bevorzugt wird als rekombinanter Ganzzellkatalysator ein Bakterium, insbesondere ein Escherichia coli Bakterium, verwendet, das Aminosäuretransaminase, Aminosäuredehydrogenase und das NAD(P)+ reduzierende Enzym überexprimiert .
Bei dem erfindungsgemäßen Verfahren erfolgt die Umsetzung vorzugsweise in einem wässrigen Reaktionsmedium. Die wässrige Phase kann zusätzlich ein mit Wasser mischbares
Lösungsmittel enthalten, vorzugsweise in einer Menge von 1 bis 20 Gew.-% bezogen auf die gesamte Reaktionsmischung.
Geeignete Lösungsmittel sind Alkohole, insbesondere Methanol, Ethanol und Isopropanol, Glykole, insbesondere
Ethylenglykol und Propylenglykol, sowie Tetrahydrofuran,
Dimethylsulfoxid und Dimethylformamid.
Das wässrige Reaktionsmedium weist vorzugsweise einen pH-Wert im Bereich von 6 bis 9, besonders bevorzugt 7 bis 8, auf. Vorzugsweise wird ein pH-Wert in diesem Bereich durch einen Puffer aus einer geeigneten anorganischen oder organischen Säure und deren Ammoniumsalz eingestellt. Beispielsweise kann die Einstellung des pH-Werts durch Puffer aus Phosphorsäure und einem Ammoniumphosphat oder Puffer aus einer Carbonsäure und deren Ammoniumcarboxylat erfolgen. Alternativ kann der pH-Wert auch durch pH-geregelte Zudosierung von Ammoniak oder einer anorganischen oder organischen Säure zum Reaktionsmedium eingestellt werden. In einer bevorzugten Ausführungsform des erfindungsgemäßen Verfahrens erfolgt die Umsetzung in einem Zweiphasensystem aus einer wässrigen und einer organischen Phase. Die organische Phase kann dabei aus dem eingesetzten Keton und/oder dem gebildeten Amin bestehen. Vorzugsweise wird aber zur Bildung einer organischen Phase ein mit Wasser
nicht mischbares Lösungsmittel zugesetzt. Geeignete Lösungsmittel sind aliphatische Kohlenwasserstoffe, insbesondere Hexan und Heptan, aromatische Kohlenwasserstoffe, insbesondere Toluol und Xylole, Dialkylether, insbesondere Methyl-tert-butylether und
Ethyl-tert-butylether, sowie Carbonsäureester, insbesondere Essigsäureethylester . Die Umsetzung in einem Zweiphasensystem ermöglicht auch bei Einsatz von schlecht wasserlöslichen Ketonen oder bei der Herstellung von schlecht wasserlöslichen Aminen eine hohe Volumenausbeute, die nicht durch die Löslichkeit von Keton oder Amin limitiert ist. Darüber hinaus lassen sich bei der Umsetzung in einem Zweiphasensystem auch Inhibierungen eines der verwendeten Enzyme durch das eingesetzte Keton oder das gebildete Amin vermeiden. Die Umsetzung in einem
Zweiphasensystem ermöglicht außerdem eine einfache Abtrennung des Aminprodukts von den eingesetzten Enzymen und deren Rückgewinnung durch eine Phasentrennung, gegebenenfalls nach vorheriger Einstellung eines pH-Werts im Bereich von 9 bis 11.
Als Keton werden vorzugsweise Dialkylketone, Alkylarylketone, Alkylheteroarylketone und Alkylaralkylketone eingesetzt, wobei die Alkylgruppen, Arylgruppen und Heteroarylgruppen mit nicht enzyminhibierenden Gruppen substituiert sein können. Das Keton kann im erfindungsgemäßen Verfahren zu Beginn der Umsetzung vorgelegt werden. Vorzugsweise wird aber zu Beginn der Umsetzung nur ein Teil des umzusetzenden Ketons vorgelegt und der restliche Teil des umzusetzenden Ketons entsprechend dem Umsatz an Keton zudosiert. Durch eine solche Zudosierung des Ketons lässt sich eine Inhibierung der verwendeten Enzyme durch das Keton vermeiden. Ketone mit einem Schmelzpunkt von mehr als 300C werden vorzugsweise als Lösungen in einem Lösungsmittel eingesetzt, wobei sowohl die zuvor beschriebenen mit Wasser mischbaren Lösungsmittel als auch die zuvor beschriebenen
mit Wasser nicht mischbaren Losungsmittel verwendet werden können. Vorzugsweise betragt die Gesamtmenge an Keton bezogen auf das Reaktionsvolumen mehr als 10 g/l, besonders bevorzugt mehr als 50 g/l und insbesondere mehr als 100 g/l. Die obere Grenze für die Gesamtmenge an Keton bezogen auf das Reaktionsvolumen ergibt sich aus den Loslichkeiten von eingesetztem Keton und gebildetem Amin, insbesondere aus der Loslichkeit des dem Amin entsprechenden Ammoniumsalzes, wenn die Umsetzung bei einem pH-Wert unterhalb des pKa-Werts dieses Ammoniumsalzes durchgeführt wird. Vorzugsweise betragt die Gesamtmenge an Keton bezogen auf das Reaktionsvolumen weniger als 500 g/l.
Der im erfindungsgemaßen Verfahren benotigte Ammoniak oder das Ammoniumsalz können sowohl zu Beginn vorgelegt als auch wahrend der Umsetzung entsprechend dem Umsatz an Keton zudosiert werden. Vorzugsweise werden Ammoniak oder Ammoniumsalz so vorgelegt und gegebenenfalls zudosiert, dass wahrend der Umsetzung die Gesamtmenge an Ammoniak und Ammoniumsalz stets in einem stochiometrischen Überschuß zum Keton vorliegt.
Das erfindungsgemaße Verfahren kann sowohl im Batch als auch kontinuierlich durchgeführt werden. Bei einer Reaktionsfuhrung im Batch erfolgt die Umsetzung vorzugsweise in einem Ruhrkesselreaktor oder in einem Membranreaktor. Bei einer kontinuierlichen Reaktionsfuhrung erfolgt die Umsetzung vorzugsweise in einem Membranreaktor oder mit einem auf einem festen Trager immobilisierten Enzym in einem Festbettreaktor. Die Umsetzung in einem Membranreaktor ermöglicht eine einfache Ruckhaltung der im erfindungsgemaßen Verfahren verwendeten Enzyme im Reaktor. Bei Verwendung eines rekombinanten Ganzzellkatalysators können die Enzyme auch durch Abtrennung der Zellen mittels Filtration oder Zentrifugation zurückgewonnen werden.
Wie das Vergleichsbeispiel zeigt, konnten mit den Reduktionsbedingungen gemäß Shin und Kim et al .
Biotechnology and Bioengineering 65 (1999), Seite 206-211, also mit einer Transaminase und einer Aminosäure (Alanin) aus dem Substrat Acetophenon nur Endausbeuten von 4.3% d. Th. 1-Phenylethylamin erhalten werden.
Durch Anwendung eines erfindungsgemäßen Reduktionssystems aus Aminosäure (Alanin) , Transaminase,
Aminosäuredehydrogenase, NADH, Formiatdehydrogenase und Ammoniumformiat wurde dagegen eine 3,5fach höhere Endausbeute an 1-Phenylethylamin aus Acetophenon erhalten (15,1 % d. Th. gemäß Beispiel 9) . Dies war überraschend und nicht vorhersehbar und bedeutet einen großen Vorteil gegenüber dem Stand der Technik.
Das erfindungsgemäße Verfahren ermöglicht somit die Herstellung von enantiomerenangereicherten Aminen aus Ketonen, ohne dass dazu stöchiometrische Mengen eines chiralen Hilfsstoffs erforderlich wären und hat gegenüber den bekannten Verfahren den Vorteil, dass durch eine geeignete Kombination von alpha-Aminosäure bzw. alpha- Ketosäure und Aminosäuredehydrogenase praktisch jede Aminosäuretransaminase für die Umsetzung des Ketons verwendet werden kann, so dass für jedes Keton jeweils die Aminosäuretransaminase eingesetzt werden kann, mit der sich die beste Enantioselektivität erzielen lässt.
Die nachfolgenden Beispiele sollen die Erfindung verdeutlichen, stellen jedoch keine Beschränkung in den Ausführungsformen dar.
Methoden:
Beispiel 1
Konstruktion der Expressionsstamme
Die Plasmide pGR15, ein E. coli Expressionsplasmid zur Expression der Transaminase aus Vibrio fluvialis unter dem Rhamnosepromotor (SEQ ID NO: 1) und pCR4, ein E. coli Expressionsplasmid zur Expression der Alanindehydrogenase aus Bacillus subtilis unter dem Rhamnosepromotor (SEQ ID NO: 2) wurden von Geneart (Regensburg, Deutschland) als synthetische Konstrukte bezogen.
Die Plasmide wurde nach den gangigen molekularbiologischen Methoden (z.B. Sambrook, J., E. F. Fritsch and T. Maniatis (1989) . Molecular cloning : a laboratory manual . CoId Spring Harbor, N. Y., CoId Spring Harbor Laboratory) in E. coli transformiert und auf LB . Agarplatten mit 100 μg/mL Ampicillin zur Selektion kuliviert.
Beispiel 2
Rekombinante Expression der Transaminase aus Vibrio fluvialis in E. coli DSM14459:
Zur Expression der Transaminase aus Vibrio fluvialis wurde in 50 ml LB Medium enthaltend 100 μg mL-1 Ampicillin zur Plasmidselektion E. coli DSM14459 (pGR15) kultiviert. Die Expression wurde sofort initiiert durch Zugabe von 2 g/L L- Rhamnose. Nach 24 h Inkubation bei 370C unter Schuttein wurden die Zellen abzentrifugiert, in 10 ml
Natriumphosphatpuffer (50 mM, pH 7,5) resuspendiert und mit Ultraschall aufgeschlossen. Der Überstand nach Abzentrifugieren der Zellbruchstucke wurde für weitere
Experimente eingesetzt (Zellrohextrakt) . Der Proteingehalt
wurde nach Bradford und die Enzymaktivitat wurde mit dem Transaminase Aktivitatstest bestimmt.
Beispiel 3
Rekombinante Expression der Alanindehydrogenase aus Bacillus subtilis in E. coli DSM14459:
Zur Expression der Alanindehydrogenase aus Bacillus subtilis wurde in 50 ml LB Medium enthaltend 100 μg/mL Ampicillin zur Plasmidselektion E. coli DSM14459 (pCR4) kultiviert. Die Expression wurde sofort initiiert durch Zugabe von 2 g/L L-Rhamnose. Nach 24 h Inkubation bei 300C unter Schuttein wurden die Zellen abzentrifugiert, in 10 ml Natriumphosphatpuffer (50 mM, pH 7,5) resuspendiert und mit Ultraschall aufgeschlossen. Der Überstand nach Abzentrifugieren der Zellbruchstucke wurde für weitere Experimente eingesetzt (Zellrohextrakt) . Der Proteingehalt wurde nach Bradford und die Enzymaktivitat wurde mit dem Aminosauredehydrogenase Aktivitatstest bestimmt.
Beispiel 4
Gaschromatographische Bestimmung der 1-Phenylethylamin- und Acetophenonkonzentration
Die Trennung von 1-Phenylethylamin und Acetophenon wurde mit einer WCOT Chrompack 7422-Saule (Lange: 25 m, CP-WAX Phase durchgeführt. Als Tragergas wurde Helium mit einem Fluß von 1 mL/min verwendet. Die Trennung erfolgte mit einem 2-stufigen Temperaturgradienten, beginnend bei 500C mit einer Aufheizrate von 0,5°C/min bis 600C, dann mit einer Aufheizrate von 100°C/min bis 2200C welche dann für 6 min gehalten wurden. Die Detektion erfolgte mit einem Flammenionisationsdetektor. Die Retentionszeit von 1-
Phenylethylamin betrug 24,2 min, die für Acetophenon betrug 24, 9 min.
Beispiel 5
Aminosauredehydrogenase Aktivität
Die Aminosauredehydrogenase Aktivität wurde spektrophotometrisch bestimmt. Dazu wurde die Abnahme der NADH Konzentration anhand der Abnahme der Absorption bei einer Wellenlange von 340 nm verfolgt. In einer 0,1 cm Quarzglaskuvette wurden 1 mL Substratlosung enthaltend
1 mmol/L NADH, 2,5 mmol/L Natriumpyruvat bzw. Natriumketoleucin, 400 mmol/L Ammoniumchlorid in einem Kaliumphosphatpuffer (50 mmol/L, pH 8,2) mit 10 μl der Enzymprobe vermischt und die Absorption bei einer Wellenlange von 340 nm über 10 min bei 300C vermessen. Mit dem Extinktionskoeffizienten ε=6300 1/ (mol cm) , der Schichtdichte und der Änderung der Absorption über die Zeit wurde die Aktivität berechnet.
Beispiel 6
Formiatdehydrogenase Aktivität
Die Formiatdehydrogenase Aktivität wurde spektrophotometrisch bestimmt. Dazu wurde die Zunahme der NADH Konzentration anhand der Zunahme der Absorption bei einer Wellenlange von 340 nm verfolgt. In einer 0,1 cm Quarzglaskuvette wurden 1 mL Substratlosung enthaltend
2 mmol/L NADH, 400 mmol/L Natriumformiat in einem Kaliumphosphatpuffer (50 mmol/L, pH 8,2) mit 10 μl der Enzymprobe vermischt und die Absorption bei einer Wellenlange von 340 nm über 10 min bei 300C vermessen. Mit dem Extinktionskoeffizienten ε=6300 1/ (mol cm), der Schichtdichte und der Änderung der Absorption über die Zeit wurde die Aktivität berechnet.
Beispiel 7
Glucosedehydrogenase Aktivität
Die Glucosedehydrogenase Aktivität wurde spektrophotometrisch bestimmt. Dazu wurde die Zunahme der NADH Konzentration anhand der Zunahme der Absorption bei einer Wellenlange von 340 nm verfolgt. In einer 1 cm Quarzglaskuvette wurden 1 mL Substratlosung enthaltend 0,25 mmol/L NADH, 100 mmol/L D-Glucose in einem Kaliumphosphatpuffer (100 mmol/L, pH 7,0) mit 10 μl der Enzymprobe vermischt und die Absorption bei einer Wellenlange von 340 nm über 10 min bei 300C vermessen. Mit dem Extinktionskoeffizienten ε=6300 1/ (mol cm), der Schichtdichte und der Änderung der Absorption über die Zeit wurde die Aktivität berechnet.
Beispiel 8
Transaminase Aktivität
Die Transaminase Aktivität wurde gaschromatographisch bestimmt. Dazu wurde zu 1 ml Substratlosung enthaltend 10 mmol/L Natriumpyruvat, 50 mmol/L 1-Phenylethylamin in einem Kaliumphosphatpuffer (100 mmol/L, pH 7) mit der Enzymprobe vermischt und die Reaktionslosung bei 300C unter Schuttein inkubiert. In regelmäßigen Abstanden wurden 100 μL Reaktionslosung als Probe entnommen mit 50 μL 1 M Natronlauge versetzt und mit 500 μL Toluol extrahiert. Die organische Phase wurde abgenommen und der Gehalt an 1- Phenylethylamin und Acetophenon gaschromatographische bestimmt .
Beispiel 9
(mit Beeinflussung der Gleichgewichtslage, nach dem Schema auf S. 6 des Anmeldetextes) :
Es wurden in 1 mL Kaliumphosphatpuffer (100 mmol/L, pH 7) 20 mmol/L Acteophenon, 200 mmol/L Alanin, 90 mmol/L Ammoniumformiat und 0,3 mmol/L NADH gemischt und mit Vibrio fluvialis Transaminase bis zu einer Endkonzentraion von 20 U/mL, mit Alanindehydrognase aus Bacillus subtillis bis zu einer Endkonzentration von 400 U/mL und mit Formiatdehydrogenase aus Candida boidinii (Jülich, jetzt Codexis) bis zu einer Endkonzentration 24 U/mL versetzt. Diese Lösung wurde unter Schütteln bei 300C inkubiert und nach 1, 3, 5, 24 und 48 h der Acetophenon- und 1-
Phenylethylamingehalt bestimmt. Der maximale Umsatz von Acetophenon wurde nach 48 h erreicht.
Die Ausbeute an 1-Phenylethylamin betrug dabei 8,5% d. Th nach 24 h und 15,1% d. Th. nach 48 h (jeweils 100% Selektivität) .
Vergleichsbeispiel 1 (ohne Beeinflussung der Gleichgewichtslage) :
Es wurden in 1 mL Kaliumphosphatpuffer (100 mmol/L, pH 7) 30 mmol/L Acteophenon und 300 mmol/L Alanin gemischt und mit Vibrio fluvialis Transaminase bis zu einer Endkonzentraion von 20 U/mL versetzt. Diese Lösung wurde unter Schütteln bei 300C inkubiert und nach 1, 3, 6 und
24 h der Acetophenon und 1-Phenylethylamingehalt bestimmt. Der maximale Umsatz von Acetophenon wurde nach 24 h erreicht .
Dabei wurde 1-Phenylethylamin in einer Ausbeute vom 4,3% d. Th Lösung (100% Selektivität) erhalten.
Claims
1. Verfahren zur Herstellung eines enantiomerenangereicherten Amins aus einem Keton, dadurch gekennzeichnet, dass ein Keton mit Ammoniak oder einem Ammoniumsalz und einem Reduktionsmittel umgesetzt wird in Gegenwart eines katalytischen Systems, umfassend
a) eine Aminosäuretransaminase,
b) eine alpha-Aminosäure, die ein Substrat der
Aminosäuretransaminase ist,
c) eine zur Herstellung der alpha-Aminosäure geeignete Aminosäuredehydrogenase,
d) NAD(P)+ und
e) ein NAD(P)+ reduzierendes Enzym, das NAD(P)+ mit dem Reduktionsmittel zu NAD(P)H umsetzt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass die Aminosäuredehydrogenase die der alpha-Aminosäure entsprechende alpha-Ketosäure selektiv zum S-Enantiomer der alpha-Aminosäure umsetzt und die Aminosäuretransaminase selektiv das S- Enantiomer der alpha-Aminosäure mit dem Keton umsetzt.
3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass die Aminosäuredehydrogenase die der alpha-Aminosäure entsprechende alpha-Ketosäure selektiv zum R-Enantiomer der alpha-Aminosäure umsetzt und die Aminosäuretransaminase selektiv das R- Enantiomer der alpha-Aminosäure mit dem Keton umsetzt.
4. Verfahren nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass das Reduktionsmittel ein Salz der Ameisensäure und das NAD(P)+ reduzierende Enzym eine Formiatdehydrogenase ist.
5. Verfahren nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass das Reduktionsmittel Glucose und das NAD(P)+ reduzierende Enzym eine Glucosedehydrogenase ist.
6. Verfahren nach einem der vorangehenden Ansprüche dadurch gekennzeichnet, dass die Aminosäuredehydrogenase ausgewählt ist aus Leucindehydrogenasen, Alanindehydrogenasen,
Phenylalanindehydrogenasen und Glutamatdehydrogenasen.
7. Verfahren nach einem der vorangehenden Ansprüche dadurch gekennzeichnet, dass Aminosäuretransaminase, Aminosäuredehydrogenase und das NAD(P)+ reduzierende Enzym in Form eines rekombinanten Ganzzellkatalysators eingesetzt werden.
8. Verfahren nach Anspruch 7, dadurch gekennzeichnet, dass der rekombinante Ganzzellkatalysator ein Bakterium, vorzugsweise ein Escherichia coli Bakterium, ist, das Aminosäuretransaminase,
Aminosäuredehydrogenase und das NAD(P) reduzierende Enzym überexprimiert .
9. Verfahren nach einem der vorangehenden Ansprüche dadurch gekennzeichnet, dass das Keton ausgewählt ist aus Dialkylketonen, Alkylarylketonen,
Alkylheteroarylketonen und Alkylaralkylketonen, wobei die Alkylgruppen, Arylgruppen und Heteroarylgruppen mit nicht enzyminhibierenden Gruppen substituiert sein können .
10. Verfahren nach einem der vorangehenden Ansprüche dadurch gekennzeichnet, dass zu Beginn der Umsetzung an Stelle der alpha-Aminosäure die der alpha-Aminosäure entsprechende alpha-Ketosäure vorgelegt wird.
11. Verfahren nach einem der vorangehenden Ansprüche dadurch gekennzeichnet, dass die Umsetzung in einem wässrigen Reaktionsmedium bei einem pH-Wert im Bereich von 6 bis 9 erfolgt.
12. Verfahren nach einem der vorangehenden Ansprüche dadurch gekennzeichnet, dass die Umsetzung in einem Zweiphasensystem aus einer wässrigen und einer organischen Phase erfolgt.
13. Verfahren nach einem der vorangehenden Ansprüche dadurch gekennzeichnet, dass zu Beginn der Umsetzung nur ein Teil des umzusetzenden Ketons vorgelegt wird und der restliche Teil des umzusetzenden Ketons entsprechend dem Umsatz an Keton zudosiert wird.
14. Verfahren nach einem der vorangehenden Ansprüche dadurch gekennzeichnet, dass die Gesamtmenge an alpha-Aminosäure und der alpha-Aminosäure entsprechender alpha-Ketosäure, bezogen auf die Gesamtmenge an Keton, im Bereich von 1 bis 50 mol-%, vorzugsweise 2 bis 10 mol-%, liegt.
15. Verfahren nach einem der vorangehenden Ansprüche dadurch gekennzeichnet, dass die Gesamtmenge an NAD(P)+ und NAD(P)H, bezogen auf die Gesamtmenge an Keton, im Bereich von 0,001 bis 5 mol-%, vorzugsweise 0,01 bis 1 mol-% liegt.
16. Verwendung eines katalytischen Systems, umfassend
a) eine Aminosäuretransaminase,
b) eine alpha-Aminosäure, die ein Substrat der Aminosäuretransaminase ist, c) eine zur Herstellung der alpha-Aminosäure geeignete Aminosäuredehydrogenase, d) NAD(P)+ und
e) ein NAD(P)+ reduzierendes Enzym, das NAD(P)+ mit dem Reduktionsmittel zu NAD(P)H umsetzt, zur Herstellung eines enantiomerenangereicherten Amins aus einem Keton.
