Description
- Die vorliegende Erfindung hat neue Indan-1,2-dion- und Indan-1,2,3-trionderivate, Verfahren zu ihrer Herstellung und ihre therapeutische Anwendung zum Gegenstand.
- Diese Derivate entsprechen genauer der Formel:
- in welcher R&sub2; und R&sub3; unabhängig voneinander H, C&sub1;-C&sub4;-Alkoxy oder OH darstellen und das Paar (A, B) den Wert annimmt:
- - (Sauerstoff, Sauerstoff), wobei in diesem Fall eines von R und R&sub1; OH, Halogen, (C&sub1;-C&sub4;-Alkyl)NH, N-Morpholino(C&sub1;-C&sub4;-alkyl)NH, 1-(Pyridyl)-4-piperazino oder 1- (Phenyl)-4-piperazino, dessen Phenylkern gegebenenfalls mit einem Halogen substituiert ist, darstellt und das andere eine Gruppe darstellt, die unter den folgenden ausgewählt wird:
- . NHCOR&sub4;, wobei R&sub4; = Phenyl, C&sub1;-C&sub4;-Alkyl oder C&sub1;-C&sub4;- Alkyl, das mit einem Halogenatom substituiert ist,

- mit Ausnahme der Derivate, für welche eines von R und R&sub1; OH oder Halogen darstellt und das andere NHCOR&sub4; darstellt, wobei R&sub4; = Phenyl oder C&sub1;-C&sub4;-Alkyl; der Derivate, für welche eines von R und R&sub1; (C&sub1;-C&sub4;-Alkyl)NH darstellt und das andere NHCOR&sub4; darstellt, wobei R&sub4; = C&sub1;-C&sub4;-Alkyl; und der Derivate, für welche eines von R und R&sub1; OH darstellt und das andere NHNHCOR&sub5; darstellt, wobei R&sub5; = Pyridyl oder Phenyl, das mit einer C&sub1;-C&sub4;-Alkoxygruppe substituiert ist,
- R und R&sub1; können außerdem zusammen eine Gruppe bilden:
- . =NNHCOR'&sub5;, wobei R'&sub5; = R&sub5;, Trifluoracetylaminophenyl, Acetylaminophenyl, Pyrazinyl oder Pyrrolyl,
- . NNHCOCONH&sub2;,
- . =NNHR&sub6;, wobei R&sub6; = Phenyl, Phenyl das mit einer oder zwei Gruppen substituiert ist, die ausgewählt werden unter C&sub1;-C&sub4;-Alkyl, Halogen und C&sub1;-C&sub4;-Alkoxy, Methylsulfonylphenyl und N-Methylmethansulfonamidophenyl,
- . =N-R&sub7;, wobei R&sub7; = Phenyl oder ein mit einer OH-Gruppe substituiertes Phenyl,
- . N-O-CO-R&sub8;, wobei R&sub8; = Phenyl oder ein mit einem Halogenatom substituiertes Phenyl, oder
- . C(CH&sub3;)-NH-NH-CO-R&sub9;, wobei R&sub9; = Phenyl, Phenylamino, Phenyl, das mit einem Halogenatom oder einer OH- Gruppe substituiert ist, oder Phenylamino, das mit einem Halogenatom oder einer OH-Gruppe substituiert ist,
- mit Ausnahme der Derivate, für welche R und R&sub1; zusammen eine Gruppe =NNHCOR'&sub5; bilden, wobei R'&sub5; = Pyridyl oder Phenyl, das mit C&sub1;-C&sub4;-Alkoxy substituiert ist, und von Derivaten, für welche R und R&sub1; zusammen eine Gruppe =N-NHR&sub6; bilden, wobei R&sub6; = Phenyl oder Phenyl, das mit einem Halogenatom oder einer C&sub1;-C&sub4;-Alkoxy- oder Alkylgruppe substituiert ist, oder
- - (NNHCXNHR&sub1;&sub0;, Sauerstoff), wobei X Sauerstoff oder Schwefel darstellt und R&sub1;&sub0; = H, Phenyl oder Phenyl, das mit einer oder zwei Gruppen substituiert ist, ausgewählt unter OH, CF&sub3;, C&sub1;-C&sub4;-Alkyl, C&sub1;-C&sub4;-Alkoxy, Halogen, Methylendioxy, Acetoxy und Hydroxyethyl, wobei in diesem Fall R und R&sub1; zusammen eine Gruppe =N-OH bilden.
- Unter den Umfang der vorstehenden Formel (I) fallen (i) die Verbindungen der Formel
- wobei R, R&sub1;, R&sub2; und R&sub3; dieselben Bedeutungen haben wie in Formel (I), wenn das Paar (A, B) dort den Wert (Sauerstoff, Sauerstoff) annimmt, und
- (ii) die Verbindungen der Formel
- wobei R, R&sub1;, R&sub2;, R&sub3;, X und R&sub1;&sub0; dieselben Bedeutungen haben wie in Formel (I), wenn das Paar (A, B) dort den Wert (=NNHCXNHR&sub1;&sub0;, Sauerstoff) annimmt.
- Unter den Umfang der vorstehenden Formel (I) fallen
- (a) die Verbindungen der Formel
- wobei R&sub4; = durch ein Halogen substituiertes C&sub1;-C&sub4;-Alkyl,
- (b) die Verbindungen der Formel
- wobei R&sub5; ≠ Pyridyl und durch eine C&sub1;-C&sub4;-Alkoxygruppe substituiertes Phenyl,
- (c) die Verbindungen der Formel

- worin R&sub4; = durch ein Halogen substituiertes C&sub1;-C&sub4;-Alkyl,

- wobei eines von R und R&sub1; NHCOR&sub4; darstellt und das andere (C&sub1;-C&sub4;-Alkyl)NH, N-Morpholino(C&sub1;-C&sub4;-alkyl)NH, 1-(Pyridyl)- 4-piperazino oder 1-(Phenyl)-4-piperazino darstellt, dessen Phenylkern gegebenenfalls durch ein Halogen substituiert ist, mit Ausnahme der Verbindungen, bei denen eines von R und R&sub1; NHCOR&sub4; darstellt, wobei R&sub4; = C&sub1;-C&sub4;-Alkyl, und das andere (C&sub1;-C&sub4;-Alkyl)NH darstellt,
- (h) die Verbindungen der Formel
- wobei eines von R und R&sub1; NHNHCOR&sub5; darstellt und das andere (C&sub1;-C&sub4;-Alkyl)NH, N-Morpholino(C&sub1;-C&sub4;-alkyl)NH, 1-(Pyridyl)-4-piperazino oder 1-(Phenyl)-4-piperazino darstellt, dessen Phenylkern gegebenenfalls durch ein Halogen substituiert ist,
- (i) die Verbindungen der Formel
- wobei eines von R und R&sub1; NHNHCOCONH&sub2; darstellt und das andere (C&sub1;-C&sub4;-Alkyl)NH, Morpholino(C&sub1;-C&sub4;-alkyl)NH, 1-(Pyridyl)-4-piperazino oder 1-(Phenyl)-4-piperazino darstellt, dessen Phenylkern gegebenenfalls durch ein Halogen substituiert ist,

- In den vorstehenden Formeln (I') und (I") und (Ia) bis (In) ist R&sub2; insbesondere in Stellung 5 und R&sub3; in Stellung 6.
- Es ist ferner hinzuzufügen, daß in den Formeln (I') und (I") und (Ia) bis (In) das Paar (R&sub2;, R&sub3;) insbesondere (H, H), (5-OCH&sub3;, H), (5-OH, H) oder (5-OCH&sub3;, 6-OCH&sub3;) sein kann.
- Die vorliegende Erfindung umfaßt ferner die Salze der unter den vorstehend beschriebenen salzbildenden Verbindungen. Diese Salze umfassen die Additionssalze von Mineralsäuren, wie etwa Salz-, Bromwasserstoff-, Schwefel- oder Phosphorsäure, und die Additionssalze organischer Säuren, wie etwa Essig-, Propion-, Oxal- oder Zitronensäure.
- Die Erfindung umfaßt weiterhin alle möglichen Stereoisomeren der Derivate der Formel (I) und die Gemische derartiger Stereoisomeren ebenso wie die Metaboliten dieser Derivate.
- Ferner fallen die Verfahren zur Herstellung von Derivaten der Formel (I) unter den Umfang der vorliegenden Erfindung. Diese Verfahren bilden den Gegenstand der nachstehenden Schemata 1 bis 5, in welchen die Symbole R bis R&sub1;&sub0; und X ohne andere Angabe dieselben Bedeutungen wie in Formel (I) haben. - In den vorstehenden Schemata bezeichnen bis die verwendeten Verfahren und haben die folgenden Bedeutungen:
- Kondensation in einem Lösungsmittel wie etwa Ethanol in der Wärme, bevorzugt unter Rückfluß.
- Kondensation in einem Lösungsmittel wie etwa Benzol in der Wärme, bevorzugt unter Rückfluß.
- Reaktion mit einem Halogenierungsmittel, insbesondere einem Chlorierungsmittel und vorzugsweise Thionylchlorid in einem Lösungsmittel wie etwa THF in der Wärme, bevorzugt unter Rückfluß.
- Kondensation mit einem primären Amin (C&sub1;-C&sub4;- Alkyl)NH&sub2; oder N-Morpholino(C&sub1;-C&sub4;-alkyl)NH&sub2;] oder einem sekundären Amin [1-(Pyridyl)-4-piperazin oder 1-(Phenyl)-4-piperazin, dessen Phenylkern gegebenenfalls durch ein Halogenatom substituiert ist] in einem Lösungsmittel wie etwa Diethylether in Gegenwart einer Base wie etwa Triethylamin.
- Behandlung in der Wärme, bevorzugt unter Rückfluß, in Ethanol und in Anwesenheit von konzentrierter HCl.
- Kondensation in der Wärme, bevorzugt unter Rückfluß, in einem Lösungsmittel wie etwa Ethanol und in Anwesenheit von konzentrierter HCl.
- Erhitzen, vorzugsweise unter Rückfluß, einer Suspension von (Ia) oder (Ib) in Acetonitril.
- Kondensation in der Wärme, bevorzugt unter Rückfluß, in einem Lösungsmittel wie etwa Ethanol.
- Kondensation in der Wärme, bevorzugt unter Rückfluß, in einem Lösungsmittel wie etwa wäßrigem Ethanol.
- Kondensation in der Wärme, bevorzugt unter Rückfluß, in einem Lösungsmittel wie etwa wäßrigem Ethanol.
- Kondensation von Chlorameisensäureisobutylester mit R&sub8;-COOH, insbesondere in einem Lösungsmittel wie etwa Essigsäureethylester in Anwesenheit einer Base wie etwa N-Methylmorpholin; das erhaltene gemischte Anhydrid wird anschließend mit dem Hydroxyimin kondensiert.
- Kondensation, insbesondere in einem Lösungsmittel, vorzugsweise Methanol, bevorzugt unter Rückfluß.
- Kondensation eines Semicarbazids oder Thiosemicarbazids in Form des Hydrochlorids in einem Lösungsmittel wie etwa einem Ethanol-Wasser-Gemisch in der Wärme, vorzugsweise unter Rückfluß
- Es ist noch hinzuzufügen, daß die Salze der salzbildenden Verbindungen unter denjenigen von Formel (I) zum Beispiel durch Einwirkung einer Säure auf die in einem geeigneten Lösungsmittel gelöste salzbildende Verbindung erhalten werden können.
- Die folgenden Herstellungen werden als Beispiele zum Veranschaulichen der Erfindung angegeben.
- Einer Suspension von 0,01 Mol 5-Acetoxy-2-carbethoxy-3- hydroxy-1-indanon in 80 ml Wasser fügt man 4 ml 10N Schwefelsäure zu. Das Reaktionsgemisch wird während 15 Minuten am Rückfluß gehalten. Das Unlösliche wird in der Wärme filtriert und der gebildete gelbe Niederschlag wird nach dem Abkühlen abgesaugt, mit Eiswasser gewaschen und dann getrocknet.
- Ausb.: 85%; Schmp.: 208ºC; IR: νOH = 3250 cm&supmin;¹, νCO = 1730 und 1680 cm&supmin;¹
- Einer Lösung von 0,02 Mol 5-Hydroxyindan-1,3-dion in 60 ml Chloroform fügt man tropfenweise 0,02 Mol Brom zu. Das Reaktionsgemisch wird 30 Minuten bei 50ºC gerührt und der nach Eindampfen des Lösungsmittels erhaltene Niederschlag wird umkristallisiert.
- Ausb.: 60%; Schmp.: 194ºC; IR: νOH = 3380 cm&supmin;¹, νCO = 1740 und 1710 cm&supmin;¹
- Umkristallisationslösungsmittel: Gemisch (Vol./Vol.) aus Ethylether/Petrolether
- Eine Lösung von 0,01 Mol 2-Brom-5-hydroxyindan-1,3-dion in 10 ml Dimethylsulfoxid wird 30 Minuten auf 80ºC erhitzt. Man fügt 50 ml 1N Salzsäure hinzu, dann verlängert man das Erhitzen um 30 Minuten. Nach dem Abkühlen wird die erhaltene ölige Suspension mit Ethylether extrahiert. Die organische Phase wird mit Wasser gewaschen und mit Magnesiumsulfat getrocknet.
- Der nach dem Abdampfen des Lösungsmittels erhaltene gelbe Niederschlag wird in Wasser umkristallisiert.
- Ausb.: 45%; Schmp.: > 265ºC; IR: νOH = 3340 cm&supmin;¹, νCO = 1750 und 1710 cm&supmin;¹
- Einer Lösung von 0,03 Mol 5,6-Dimethoxy-1-indanon in 100 ml Dioxan fügt man 0,06 Mol Selenoxid gelöst in 2,5 ml Wasser hinzu. Das Reaktionsgemisch wird 7 Stunden unter Rückfluß gehalten und der nach Abdampfen des Lösungsmittels erhaltene Rückstand wird in Essigsäureethylester gelöst. Die organische Phase wird mehrmals mit Wasser gewaschen, über Magnesiumsulfat getrocknet und anschließend entfärbt.
- Der nach Abdampfen des Lösungsmittels erhaltene Niederschlag wird in Ethylether umkristallisiert.
- Ausb.: 50%; Schmp.: 170ºC; IR: νCO = 1730 und 1700 cm&supmin;¹
- Einer Lösung von 0,03 Mol 6-Hydroxy-5-methoxy-1-indanon in 100 ml Dioxan fügt man 0,06 Mol Selenoxid gelöst in 2,5 ml Wasser hinzu. Das Reaktionsgemisch wird 7 Stunden unter Rückfluß gehalten und der nach Abdampfen des Lösungsmittels erhaltene Rückstand wird in Essigsäureethylester gelöst. Die organische Phase wird mehrmals mit Wasser gewaschen, über Magnesiumsulfat getrocknet und dann entfärbt.
- Der nach Abdampfen des Lösungsmittels erhaltene Niederschlag wird in Wasser umkristallisiert.
- Ausb.: 50%; Schmp.: > 265ºC; IR: νCO = 1740 und 1700 cm&supmin;¹
- Einer Suspension von 0,01 Mol 5-Hydroxyindan-1,3-dion in 6 ml 2N Schwefelsäurelösung fügt man tropfenweise 0,02 Mol Natriumnitrit gelöst in 8 ml Wasser zu, wobei die Temperatur bei 5ºC gehalten wird. Das Reaktionsgemisch wird 4 Stunden bei 5ºC gerührt. Das Unlösliche wird abgesaugt, mit Wasser gewaschen, getrocknet und in Aceton umkristallisiert.
- Ausb.: 65%; Schmp.: 220ºC; IR: νOH = 3400 und 3260 cm&supmin;¹; νCO = 1730 und 1690 cm&supmin;¹
- Im folgenden verwendet man die Namen Ninhydrin, Indan- 1,2,3-trion-monohydrat und 2,2-Dihydroxy-1,3-dioxo-2H- inden unterschiedslos.
- Einer Lösung von 0,01 Mol Ninhydrin in 20 ml Ethanol fügt man 0,01 Mol H&sub2;N-NH-CO-R&sub5; oder H&sub2;N-NH-CO-CO-NH&sub2; gelöst in 20 ml Ethanol zu.
- Das Reaktionsgemisch wird 5 bis 15 Minuten auf 60ºC erhitzt und der gebildete weiße Niederschlag wird abgesaugt, mit Ethylether gewaschen und dann getrocknet.
- Einer Lösung von 0,04 Mol Ninhydrin in 40 ml Benzol fügt man 0,04 Mol NH&sub2;-CO-R&sub4; zu. Das Reaktionsgemisch wird eine Stunde am Rückfluß gehalten. Der nach Abkühlen in einem Eisbad gebildete weiße Niederschlag wird abgesaugt und in 150 ml Diethylether gelöst. Die organische Phase wird drei Mal mit 100 ml Wasser gewaschen und über Magnesiumsulfat getrocknet. Die nach Eindampfen des Lösungsmittels gebildeten weißen Kristalle werden umkristallisiert.
- Einer Lösung von 6 g (0,02 Mol) 2-Benzamido-2-hydroxyindan-1,3-dion in 20 ml Tetrahydrofuran fügt man tropfenweise 3,5 ml (0,04 Mol) Thionylchlorid zu. Das Reaktionsgemisch wird eine Stunde am Rückfluß gehalten. Der nach Abdampfen von überschüssigem Thionylchlorid und Tetrahydrofuran erhaltene ölige Rückstand kristallisiert nach dem Waschen mit Petrolether.

- Einer Lösung von 0,007 Mol 2-Benzamido-2-chlorindan-1,3- dion in 60 ml Ethylether fügt man 1 ml Triethylamin zu. Man läßt 5 Minuten rühren und fügt dann 0,007 Mol Amin zu. Das Reaktionsgemisch wird 2 Stunden bei Raumtemperatur gerührt. Der gebildete Niederschlag wird abgesaugt, mehrmals mit Wasser gewaschen, getrocknet und umkristallisiert.

- Einer Suspension von 0,008 Mol Verbindung (Ia) oder (Ib) in 100 ml Ethanol setzt man 5 Tropfen konzentrierte Salzsäure (10N) zu. Das Reaktionsgemisch wird 30 Minuten am Rückfluß gehalten. Der gebildete gelbe Niederschlag wird in der Kälte abgesaugt, getrocknet und Umkristallisiert.
- Einer Lösung von 0,01 Mol Ninhydrin in 50 ml Ethanol setzt man nacheinander 0,01 Mol des in 50 ml Ethanol gelösten entsprechenden Hydrazids und 5 Tropfen konzentrierte Salzsäure (10N) zu.
- Das Reaktionsgemisch wird 30 Minuten am Rückfluß gehalten, man läßt auf etwa 50ºC abkühlen und der erhaltene gelbe Niederschlag wird abgesaugt, getrocknet und umkristallisiert.
- Eine Suspension von 1,5 g Verbindung (Ia) oder (Ib) in 100 ml Acetonitril wird 30 Minuten am Rückfluß gehalten. Die erhaltene Lösung wird bis zum Auftreten eines gelben Niederschlags, den man nach dem Abkühlen absaugt, 30 Minuten am Rückfluß gehalten.
- Einer Lösung von 0,01 Mol Ninhydrin in 60 ml Ethanol fügt man 0,01 Mol Hydrazid zu. Das Reaktionsgemisch wird 2 Stunden am Rückfluß gehalten. Der erhaltene orangefarbene Niederschlag wird abgesaugt, getrocknet und umkristailisiert.

- Einer Lösung von 0,01 Mol Ninhydrin in 200 ml Ethanol fügt man 0,01 Mol 2-Hydroxyanilin zu. Das Reaktionsgemisch wird eine Stunde am Rückfluß gehalten. Der erhaltene Niederschlag wird abgesaugt, mit Ethylether gewaschen und aus Acetonitril umkristallisiert.

- 34 mmol in 68 ml Essigsäureethylester gelöstem Arylcarbonsäurechlorid fügt man bei Raumtemperatur 34 mmol Chlorameisensäureisobutylester und 34 mmol N-Methylmorpholin, dann bei -10ºC tropfenweise 34 mmol in 25 ml DMF gelöstes 2-Hydroxyiminoindan-1,3-dion hinzu. Nach einer Stunde bei -10ºC, 15 Stunden bei Raumtemperatur und 5 Stunden bei 40ºC wird der gebildete Niederschlag durch Filtration über eine Glasfritte entfernt und mit Essigsäureethylester gewaschen. Das Filtrat wird anschließend mit einer gesättigten Natriumhydrogencarbonatlösung gewaschen, über Magnesiumsulfat getrocknet und unter vermindertem Druck eingedampft. Die erhaltenen gelben Kristalle werden umkristallisiert, dann eine Nacht im Vakuum bei 50ºC getrocknet.

Das Reaktionsgemisch wird 1 Stunde am Rückfluß gehalten. Der gebildete Niederschlag wird in der Wärme abgesaugt, getrocknet und in Acetonitril umkristallisiert.

- Es sollte deutlich gemacht werden, daß die vorstehenden Infrarotspektren mit KBr-Tabletten aufgenommen wurden.

- mit Ausnahme der Verbindungen, bei denen R und R&sub1; zusammen eine Gruppe =N-NH-CO-R'&sub5; bilden, wobei R'&sub5; = Pyridyl, und
- andererseits der pharmazeutisch annehmbaren Additionssalze von salzbildenden Verbindungen unter denen vorstehenden Formel (A)
- Die nicht von der Formel (A) umfaßten Verbindungen und ihre etwaigen Salze sind bekannt und können nach den vorstehenden Schemata bis hergestellt werden, wobei die entsprechenden Ausgangsmaterialien eingesetzt werden.
- Die Untersuchung der Verbindungen der Formel (A) und ihrer etwaigen Salze hat gezeigt, daß sie verschiedene pharmakologische Eigenschaften besitzen. So sind sie venotonisch und beeinflussen in den meisten Fällen das arterielle System nicht: außerdem erhöhen sie die Kapillarresistenz, vermindern die durch gewisse entzündungsverursachende Mittel erzeugte Gefäßhyperpermeabilität und zeigen Antilipoperoxid-, Antiradikal- und entzündungshemmende Eigenschaften wie auch eine Aktivität bei septischem Schock.
- Diese Eigenschaften zeigen sich bei Säugern wie Ratten, Meerschweinchen und Kaninchen unter in-vitro- (isolierte Gefäße oder Gefäßgewebe) und in-vivo-Bedingungen.
- Für die in-vitro-Untersuchung werden die Verbindungen in reinem oder DMSO oder Alkohol enthaltendem Wasser gelöst.
- Für die in-vivo-Untersuchung werden sie gelöst in reinem Wasser auf intravenösem Weg, in wäßriger, DMSO oder keines enthaltender Lösung auf intraperitonealem Weg oder in Lösung oder in Suspension in Carboxymethylcellulose oder in einer zusammengesetzten, Tween und in gewissen Fällen DMSO enthaltenden wäßrigen Lösung auf oralem Weg verabreicht.
- Die Kontraktionswirkung wird in vitro gemessen:
- - durch die Kontraktionskraft, die durch Gefäßringe entwickelt wird, welche sich in Ruhe befinden oder stimuliert werden (entweder elektrisch oder durch physiologische Mittel) und unter isometrischen Bedingungen gehalten werden.
- - durch den Druck, der durch das mit konstanter Leistung perfundierte Gefäßgewebe entwickelt wird.
- In vivo werden der arterielle und venöse Druck unter Normalbedingungen und nach dem Herzstillstand gemessen. Bei dem Herzstillstand wird der venöse Tonus aus dem am Gleichgewicht gemessenen venösen und arteriellen Druck berechnet und in Abhängigkeit von den relativen Unterschieden der Volumendehnbarkeit zwischen den beiden Geweben korrigiert (Samar und Coleman, Am. J. Physiol., 1978, 234:H94-100; Yamamoto et al., Am. J. Physiol., 1980, 238:H823-828).
- Die Zunahme der Kapillarresistenz wird durch die Anderung des Patechienindex (das Austreten von Erythrozyten aus den Gefäßen auslösender negativer Druck) abgeschätzt, welcher durch ein vom der Parrotschen Angiosterrometer stammendes Verfahren gemessen wird.
- Die Gefäßpermeabilität wird in vivo und in vitro durch die Messung des Austretens von Albumin oder eines an Albumin gebundenen Farbstoffs (Evans-Blau) aus den Gefäßen untersucht. In vivo wird die Hyperpermeabilität durch die Injektion einer Lösung von Histamin, Bradykinin oder Zymosan ausgelöst. Die in-vitro-Modelle gestatten das Verwirklichen von Überdrücken (auf einem isolierten Gefäßgebiet) und/oder von entzündlichen Gefäßreaktionen.
- Die entzündungshemmende Aktivität wird durch die Messung der Hemmung des Ödems und der Leukozytenwanderung nach dem Auslösen einer Rippenfellentzündung bei der Ratte durch eine Carragenininjektion in den Brustkorb gezeigt (Almeida et al., J. Pharmacol. Exp. Therap., 1980, 214:74).
- Die pauschale Wirkung als "Freiradikalfänger" wird in vitro durch ein Modell untersucht, das 1,1-Diphenyl-2- picrylhydrazyl (DPPH) als stabiles freies Radikal verwendet, ein Verfahren, das von dem von Lamaison et al., Plantes Médic. et phytothérapie, XXII, 1088, 231-234, beschriebenen abgeleitet ist.
- Die Antioxidationsaktivität wird in vitro durch ein Modell der Lipidperoxidation untersucht, welche auf der Peroxidation einer Linolsäureemulsion durch Eisen beruht, ein Verfahren, das hinsichtlich des von Sutherland et al., Arch. Biochem. Biophys., 1982, 214, 1-11, beschriebenen abgeändert wurde.
- Die Aktivität bei septischem Schock wurde an der Ratte nach dem Auslösen durch Lipopolysaccharid-Endotoxin (15 mg/kg) untersucht, ein dem von Terashita et al., Eur. J. Pharmacol., 109, 257-261, 1985, beschriebenen ähnliches Verfahren.
- Die Verbindungen der Formel (A) und ihrer etwaigen Salze erhöhen die Kontraktion tierischer Saphenavenen, welche durch Noradrenalin und Depolarisierung (Hyperkalilösung) erzeugt wurde, ohne in der Mehrzahl der Fälle die arteriellen Kontraktionsantworten zu beeinflussen. So erhöhen 2-Hydroxy-2-(2-methoxybenzoylhydrazino)-indan-1,3-dion und die Verbindung des Beispiels 22 (10 mN bis 30 µM) die durch KCl (40 mM) erzeugten Kontraktionen der Saphena-Venen von Kaninchen um mehr als 50% (ED&sub5;&sub0; ±0,3 µMol).
- Die Verbindungen der Beispiele 3 und 12 und 2-(3-Methylphenylhydrazono)-indan-1,3-dion erhöhen bei ihrer Höchstkonzentration von 30 bis 200% die Konzentration in den Saphenavenen des Kaninchen als Antwort auf 0,3 Mikromol Noradrenalin.
- Die Zielverbindungen der Beispiele 3, 6 und 11 erhöhen den venösen Grundtonus der Ratte um mehr als 20%, ohne den arteriellen Druck bei Dosen von 1 bis 3 Picomol und i.v. -Verabreichung zu beeinflussen.
- Als Beispiel erhöhen die Verbindungen der Beispiele 17, 22 und 54 und 2-Isonicotinoylhydrazonoindan-1,3-dion die Kapillargrundresistenz um 10 bis 100%, wenn diese eine Stunde bis zwei Stunden nach der Verabreichung von 5-20 mg/kg/i.p. und vier bis sechs Stunden nach der oralen Verabreichung von 5-20 mg/kg bei der Ratte gemessen wird.
- Die Verbindungen der Beispiele 1, 3 und 11 und 2-Hydroxy- 2-(3-methoxybenzoylhydrazino)-indan-1,3-dion vermindern die Gefäßpermeabilität um 10 bis 50%, wenn diese ein bis zwei Stunden nach der Verabreichung von 5-20 mg/kg/i.p. und zwei bis vier Stunden nach der oralen Verabreichung von 5-20 mg/kg bei der Ratte gemessen wird.
- Die zwei Mal i.p. in einer Dosis von 20 mg/kg verabreichte Verbindung von Beispiel 9 und 2-Isonicotinoylhydrazonoindan-1,3-dion hemmen im Rippenfellmodell bei der Ratte das Ödem und die Leukozytenwanderung in den Brustkorb 6 Stunden nach der Carragenininjektion.
- Die Verbindungen der Beispiele 9 und 56 und 2-(3-Fluorphenylhydrazono)-5-methoxyindan-1,3-dion zeigen im DPPH verwendenden Modell eine maximale Antiradikalwirkung von mehr als 95%.
- Darüber hinaus sind die Verbindungen der Formel (A) und ihre etwaigen Salze sehr wenig toxisch. Zum Beispiel wird nach der einmaligen Verabreichung per os an die Maus bei einer Dosis von 1 g/kg bei 2-Phenylhydrazonoindan-1,3- dion keine Sterblichkeit beobachtet. Die einzigen beobachteten Wirkungen sind in gewissen Fällen gefärbte Diarrhöen und gefärbter Urin, letzterer ist ein Beweis für die Resorption des Produkts.
- Das Vorstehende zeigt, daß die Verbindungen der Formel (A) und ihre etwaigen Salze in der menschlichen und tie rischen Therapie verwendet werden können. Sie sind durch ihre Gefäß- und entzündungshemmende Komponente besonders angezeigt bei funktioneller organischer venöser Insuffizienz und Hämorrhoidalerkrankungen wie auch bei typischen Entzündungsbeschwerden (der Knochengelenke, der Haut oder der Herzgefäße) und bei Schockzuständen, die aus einem bedeutsamen Abfall des arteriellen Drucks bestehen, insbesondere bei Zuständen eines septischen Schocks (endotoxisch).
- Die venöse funktionelle Insuffizienz ist durch eine Erweiterung und eine Überdehnbarkeit der oberflächlichen Venen der unteren Gliedmaßen gekennzeichnet und wird von funktionellen Symptomen begleitet: Schmerzen der unteren Gliedmaßen, Ödeme, unruheartige Parästhesie, angespannte Beine. Dieser Krankheitstyp kann sich zu organischer venöser Insuffizienz (Krampfadern, tiefe Klappeninsuffizienz...), ja sogar zur Phlebothrombose und Ulzerläsionen entwickeln.
- Bei dieser Venenerkrankung baut sich in den ersten Stadien eine Entzündungskomponente auf und zeigt sich deutlicher in den fortgeschrittenen Stadien.
- Die vorliegende Erfindung umfaßt folglich die Verwendung der vorstehend beschriebenen Verbindungen der Formel (A) und ihrer etwaigen Salze als aktive Substanzen zur Herstellung von Arzneimitteln und pharmazeutischen Zusammensetzungen zur humanen oder veterinärmedizinischen Verwendung, welche mindestens eine der besagten Verbindungen zusammen mit einem physiologisch annehmbaren Träger oder Verdünnungsmittel enthalten.
- Die Form dieser Arzneimittel und pharmazeutischen Zusammensetzungen hängt ganz offensichtlich vom gewünschten Verabreichungsweg, insbesondere oral, parenteral und rektal, ab und sie können nach klassischen Techniken unter Einsetzen üblicher Träger und Hilfsmittel formuliert werden.
- So können sie im Fall einer Verabreichung auf oralem Weg die Form von Pastillen, Tabletten, Kapseln, Lösungen, Sirupen und Suspensionen annehmen.
- Die Pastillen, Tabletten und Kapseln enthalten die aktive Substanz in Verbindung mit einem Verdünnungsmittel (zum Beispiel Lactose, Dextrose, Sucrose, Mannit, Sorbit oder Cellulose), einem Gleitmittel (zum Beispiel Siliziumoxid, Talk oder ein Stearat wie Magnesiumstearat), einem Bindemittel (zum Beispiel Amidon, Methylcellulose oder Gummiarabicum), einem Zerfallshilfsmittel (zum Beispiel ein Alginat) und sie werden durch bekannte Techniken, zum Beispiel Mischen, Granulieren, Tablettieren, Umhüllung usw. hergestellt.
- Die Sirupe können als Träger Glycerin, Mannit und/oder Sorbit enthalten. Die Lösungen und Suspensionen können Wasser und einen Träger wie etwa einen Naturgummi, Agar- Agar, Natriumalginat oder Polyvinylalkohol umfassen.
- Zur Verabreichung auf parenteralem Weg können die Arzneimittel und Zusammensetzungen die Form von Lösungen, Emulsionen oder Suspensionen annehmen, welche die aktive Substanz und einen geeigneten Träger, wie etwa sterilisiertes Wasser oder isotone, sterile, wäßrige Kochsalzlösungen, enthalten.
- Zur Verabreichung auf rektalem Weg können sie die Form von Suppositorien annehmen, welche die aktive Substanz und einen geeigneten Träger, wie etwa Kakaobutter oder Polyethylenglykol enthalten.
- Die therapeutische Dosis der aktiven Substanzen kann gemäß dem Verabreichungsweg, dem Alter, dem Gewicht und dem Zustand der zu behandelnden Person und der therapeutischen Wirksamkeit der eingesetzten Substanz 1000 mg/Tag erreichen.

1. Verbindungen, entsprechend der Formel:

mit Ausnahme von Derivaten, für welche R und R&sub1; zusammen
eine Gruppe =N-NH-CO-R'&sub5; bilden, wobei R'&sub5; = Pyridyl; und
andererseits der pharmazeutisch annehmbaren Salze von
salzbildenden Verbindungen unter denen der oben
angegebenen Formel (A) mit anorganischer oder organischer
Säure.
6. Pharmazeutische Zusammensetzung, dadurch gekennzeichnet,
daß sie einen Trägerstoff oder ein physiologisch
annehmbares Verdünnungsmittel und mindestens eine Verbindung oder
ein pharmazeutisch annehmbares Salz dieser Verbindung, wie
in Anspruch 5 definiert, umfaßt.
7. Verfahren zur Herstellung von Verbindungen der Formel (I)
gemäß Anspruch 1, für welche das Paar (A, B) =
(Sauerstoff, Sauerstoff), wobei eines von R und R&sub1; OH oder
Halogen darstellt, und das andere NHCOR&sub4;, NHNHCOR&sub5; oder
NHNHCOCONH&sub2; darstellt, dadurch gekennzeichnet, daß es die
Kondensation von Verbindungen der Formel:
H&sub2;N-CO-R&sub4;,
H&sub2;N-NH-CO-R&sub5;, oder
H&sub2;N-NH-CO-CO-NH&sub2;
mit Indan-1,2,3-trion-monohydrat der Formel:
umfaßt, wobei R&sub2;, R&sub3;, R&sub4; und R&sub5; dieselbe Bedeutung haben
wie in Anspruch 1, gegebenenfalls gefolgt von der Reaktion
der somit erhaltenen Verbindungen mit einem
Halogenierungsmittel.
8. Verfahren zur Herstellung von Verbindungen der Formel (I)
gemäß Anspruch 1, für welche das Paar (A, B) =
(Sauerstoff, Sauerstoff) wobei eines von R oder R&sub1; (C&sub1;-C&sub4;-
Alkyl), N-Morpholino(C&sub1;-C&sub4;-alkyl)NH, 1-(Pyridyl)-4-
piperazino oder 1-(Phenyl)-4-piperazino darstellt, dessen
Phenylkern gegebenenfalls mit einem Halogen substituiert
ist, und das andere NHCOR&sub4;, NHNHCOR&sub5; oder NHNHCOCONH&sub2;
darstellt, dadurch gekennzeichnet, daß es die Kondensation
von (C&sub1;-C&sub4;-Alkyl) NH&sub2;, N-Morpholino(C&sub1;-C&sub4;-alkyl)NH&sub2;,
1-(Pyridyl)-4-piperazin oder 1-(Phenyl)-4-piperazin,
dessen Phenylkern gegebenenfalls mit einem Halogen
substituiert ist, jeweils mit den Verbindungen der Formel:
umfaßt, wobei R&sub2; und R&sub3; dieselbe Bedeutung haben wie in
Anspruch 1, Hal = Halogen und R&sub1; = NHCOR&sub4;, NHNHCOR&sub5; oder
NHNHCOCONH&sub2;
9. Verfahren zur Herstellung von Verbindungen der Formel (I)
gemäß Anspruch 1, für welche das Paar (A, B) =
(Sauerstoff, Sauerstoff) und R und R&sub1; zusammen eine Gruppe
=N-NH-CO-R'&sub5; oder =N-NH-CO-CO-NH&sub2; bilden, dadurch
gekennzeichnet, daß es das Erhitzen der Verbindungen der Formel
(I) gemäß Anspruch 1, für welche das Paar (A, B) =
(Sauerstoff, Sauerstoff) und eines von R und R&sub1; OH darstellt und
das andere NH-NH-CO-R&sub5; oder NH-NH-CO-CO-NH&sub2; darstellt, in
Ethanol in Anwesenheit von konzentriertem HCl oder in
Acetonitril, und gegebenenfalls das Einwirken einer
Mischung aus Trifluoressigsäure und
Trifluoressigsäureanhydrid oder einer Mischung aus Essigsäure und
Essigsäureanhydrid auf die somit erhaltenen Verbindungen
umfaßt.
10. Verfahren zur Herstellung von Verbindungen der Formel (I)
gemäß Anspruch 1, für welche das Paar (A, B) =
(Sauerstoff, Sauerstoff) und R und R&sub1; zusammen eine Gruppe
=N-NH-CO-R'&sub5;, =N-NH-R&sub6; oder =NH-R&sub7; bilden, dadurch
gekennzeichnet, daß es die Kondensation von
H&sub2;N-NH-CO-R'&sub5;, von H&sub2;N-NH-R&sub6;-Hydrochlorid oder H&sub2;N-R&sub7;-
Hydrochlorid mit Indan-1,2,3-trion-monohydrat der Formel:
umfaßt, wobei R&sub2;, R&sub3;, R'&sub5;, R&sub6; und R&sub7; dieselbe Bedeutung
haben wie in Anspruch 1
11. Verfahren zur Herstellung von Verbindungen der Formel (I),
gemäß Anspruch 1, für welche das Paar (A, B) =
(Sauerstoff, Sauerstoff) und R und R&sub1; zusammen eine Gruppe
=N-O-CO-R&sub8; bilden, dadurch gekennzeichnet, daß es die
Kondensation von Chlorameisensäureisobutylester jeweils mit
den Verbindungen der Formel:
R&sub8;-COOH,
gefolgt von der Kondensation des daraus resultierenden
gemischten Anhydrids mit der Verbindung der Formel umfaßt:
wobei R&sub2;, R&sub3; und R&sub8; dieselben Bedeutungen haben wie in
Formel (I).
12 Verfahren zur Herstellung von Verbindungen der Formel (I)
gemäß Anspruch 1, für welche das Paar (A, B) =
(Sauerstoff, Sauerstoff) und R und R&sub1; zusammen eine Gruppe
=C(CH&sub3;)-NH-NH-CO-R&sub9; bilden, dadurch gekennzeichnet, daß es
die Kondensation der Verbindungen der Formel:
R&sub9;-CO-NH-NH&sub2;
jeweils mit den Verbindungen der Formel:
umfaßt, wobei R&sub2;, R&sub3; und R&sub9; dieselben Bedeutungen haben wie
in Formel (I).
13. Verfahren zur Herstellung von Verbindungen der Formel (I)
gemäß Anspruch 1, für welche das Paar (A, B) =
(N-NH-CX-NH-R&sub1;&sub0;, Sauerstoff) und R und R&sub1; zusammen eine
Gruppe =N-OH bilden, dadurch gekennzeichnet, daß es die
Kondensation der Verbindung der Formel:
H&sub2;N-NH-CX-NH-R&sub1;&sub0;
in Form des Hydrochlorids jeweils mit den Verbindungen der
Formel:
umfaßt, wobei R&sub2;&sub1; R&sub3;, X und R&sub1;&sub0; dieselben Bedeutungen haben
wie in Formel (I).
