Description
- Diese Erfindung betrifft ein Verfahren zur Herstellung von 9-Amino-1,2,3,4-tetrahydroacridin.
- Die Herstellung von Tacrin und dessen Derivaten ist gut bekannt.
- Im Journal of Heterocyclic Chemistry, Vol 5, 1968, S. 737-39 wird die Synthese von Tetrahydroacridonen unter Einsatz von p-Toluolsulfonsäure in der nachstehenden Reaktion offenbart.
- J. Bielavsky offenbart in Collection Czechoslov Chem. Commun., Vol. 42, 1977, S. 2862-2808 die Herstellung von Tacrin-Analogen mittels Hoffmannschem Abbau von Amiden. Insbesondere wird dieses Verfahren als vorteilhafte Methode zur Herstellung von Tacrin beansprucht. Es bietet jedoch nur Ausbeuten von etwa 65%.
- Tetrahedron Letters Nr.20, 1963, S. 1277-81 offenbart eine direkte Synthese von Aminochinolonen einschließlich Tacrin. Bei diesem Verfahren sind jedoch der Einsatz von Zinkchlorid und extrem hohe Temperaturen erforderlich.
- Tetrahedron Letters, Nr.27, 1986, S. 5323-26 beschreibt die Synthese von Tetrahydroacridin aus einem Imin unter Verwendung von 2,0 Äquiv. Lithiumdiisopropylamid (LDA) in Tetrahydrofuran. In dieser Quelle wird jedoch die Synthese von Tacrin nicht beschrieben.
- Synthesis, 1985, S. 547-8 beschreibt die Synthese von Tacrin unter Einsatz von Phosphorpentoxid mit einer Ausbeute von nur 30%.
- Diese Erfindung bietet ein Verfahren zur Herstellung von 9-Amino-1,2,3,4-tetrahydroacridin der Formel
- durch Umsetzung von 2-Aminobenzonitril und Cyclohexanon, wobei das resultierende Produkt mit wäßriger Natriumhydroxidlösung basisch eingestellt wird und das verfahren dadurch gekennzeichnet ist, daß die Reaktion in einem Schritt in Gegenwart eines aus der Gruppe p- Toluolsulfonsäure und deren Hydrate, Methansulfonsäure und Schwefelsäure ausgewählten Säurekatalysators in einem geeigneten Lösemittel erfolgt. Die freie Base kann unter Einsatz von Salzsäure leicht in das als Tacrin bekannte Hydrochloridsalz umgesetzt werden.
- Tacrin hat sich potentiell nützlich zur Behandlung verschiedener Gedächtnisstörungen erwiesen, die durch eine verringerte cholinergische Funktion gekennzeichnet sind, wie etwa die Alzheimer Krankheit.
- Vorzugsweise umfaßt diese Erfmdung die einfache Synthese von Tacrin durch Umsetzen von 2-Aminobenzonitril der Formel
- mit Cyclohexanon in Gegenwart von mehr als 1 Äquiv. p-Toluolsulfonsäuremonohydrat in Xylolen.
- Keines der vorgenannten bekannten Herstellungsverfahren weist die Vorteile der vorliegenden Erfindung auf, bei der das Produkt in hoher Ausbeute und Reinheit erzeugt wird.
- Zu den Vorteilen dieser neuen Methode zählen:
- 1. Die Vorteile des Verfahrens in einem Schritt, das für die kommerzielle Herstellung dieses wichtigen Produkts eingesetzt werden kann;
- 2. die Möglichkeit, Tacrin in hohen Ausbeuten zu erhalten;
- 3. das Verfahren bietet Sicherheit flir die Umwelt, da keine toxischen Metalle wie Zink eingesetzt werden;
- 4. das Verfahren läuft bei Temperaturen unter 150ºC und läßt sich problemlos diskontinuierlich betreiben;
- 5. die als Lösemittel eingesetzten Xylole können leicht zurückgewonnen und wiederaufbereitet werden, was zur Kosten- und Umweitwirksarukeit des Verfahrens beiträgt; und
- 6. das Verfahren ist leicht aufzuarbeiten, ohne daß flüchtige gasförmige Reagenzien oder Nebenprodukte entstehen, wie beispielsweise bei der Methode, die Phosphoroxychlorid verwendet, oder mit Ammoniak und Brom im Falle des Hoffmannschen Abbaus von Amiden.
- Aufgrund all dieser Vorteile stellt das Verfahren eine Verbesserung gegenüber dem Stand der Technik dar.
- Tacrin wird gemäß der folgenden Reaktion hergestellt:
- Eine Lösung von 2-Aminobenzonitril in Xylolen mit 0,02 bis 0,05 Äquiv. p-Toluolsulfonsäuremonohydrat wird unter Rühren auf die Rückflußtemperatur erhitzt. Bei der Rückflußtemperatur wird Cyclohexanon in Xylolen zugegeben. Das Gemisch wird 8 bis 12 Stunden lang unter Rückfluß gekocht, abgekühlt, und weitere 1,0 bis 1,5 Äquiv. p-Toluolsulfonsäuremonohydrat werden zugegeben. Das Gemisch wird 3 bis 7 Stunden lang unter Rückfluß gekocht. Das Produkt als p-Toluolsulfonsäuresalz wird mittels Filtration isoliert. Durch Basischmachen dieses Produkts mit wäßriger Natriumhydroxidlösung und Extraktion mit Dichlormethan erhält man das gewünschte Produkt, Tacrin, als freie Base.
- Dieser Prozeß kann in einer Reihe von verschiedenen Arten von Lösemitteln wie aromatischen Kohlenwasserstoffen, halogenierten Aromaten oder aromatischen Ethern ablaufen. Beispiele aromatischer Kohlenwasserstoffe sind Xylole, Toluole und Mesitylen. Beispiele geeigneter halogenierter Lösemittel sind Chlorbenzol, p-, m- oder o-Dichlorbenzole und 2-, 3- oder 4- Chlortoluole. Aromatische Ether, die in diesem Verfahren eingesetzt werden können, umfassen Anisol, p-, m- oder o-Dimethoxybenzole und 2-, 3- oder 4-Methylanisol. Als Lösemittel bevorzugt werden Xylole als Gemisch von p-, m- und o-Isomeren.
- Das Verhältnis von Lösemittel zu den Reaktionspartnern bewegt sich Bereich von 5:1 bis 15:1 Volumen/Gewicht (Vol./Gew.). Das bevorzugte Verhältnis ist 10:1.
- Der Säurekatalysator wird zur Beschleunigung der Reaktion in einem Schritt eingesetzt, und die im allgemeinen verwendete Art ist eine Bronsted-Säure. Als Säurekatalysatoren können p-Toluolsulfonsäure und ihre Hydrate, Methansulfonsäure und Schwefelsäure eingesetzt werden. Bevorzugt wird p-Toluolsulfonsäuremonohydrat.
- Das Verhältnis des Katalysators zu dem Reaktionspartner, 2-Aminobenzonitril, bewegt sich im Bereich von 1,02 bis 1,6 Mol/Mol. Das bevorzugte Verhältnis ist 1,02.
- Die erfindungsgemäße Synthese erfolgt im allgemeinen bei einer Temperatur von etwa 110ºC bis etwa 180ºC. Der bevorzugte Temperaturbereich für die Durchführung der Synthese ist etwa 130ºC bis etwa 150ºC, insbesondere etwa 145ºC.
- Die Umsetzung benötigt bis zum Abschluß etwa 10 bis 15 Stunden. Die bevorzugte Reaktionszeit beträgt jedoch 11 bis 12 Stunden.
- Die Erfindung wird in den nachstehenden Beispielen ausführlicher beschrieben, in denen sich alle Teile, Anteile, Verhältnisse und Prozentsätze auf das Gewicht beziehen, sofern nichts anderes angegeben ist.
- In einen mit einem hängenden Rührer, Dean-Stark-Abscheider und Thermometer ausgestatteten dreihalsigen Rundkolben wurden 6,67 g 2-Aminobenzonitril, 66,7 ml Xylole und 0,213 g p-Toluolsulfonsäuremonohydrat gegeben. Die gerührte Lösung wurde auf die Rückflußtemperatur erhitzt. Bei der Rückflußtemperatur wurde eine Lösung von 8,78 ml Cyclohexanon in 8,8 ml Xylolen über einen Zeitraum von 40 Minuten zugetropft, während gleichzeitig mittels azeotroper Destillation Wasser entfernt wurde. Das Gemisch wurde weitere 10 Stunden unter Rückfluß gekocht, dann leicht abgekühlt, weitere 10,71 g p-Toluolsulfonsäuremonohydrat wurden zugegeben, und das Gemisch wurde weitere 5 Stunden unter Rückfluß gekocht. Nach Abkühlen auf Raumtemperatur wurde das Produkt mittels Filtration und Trocknen unter Vakuum isoliert. Man erhielt 20,4 g Feststoff. Davon wurden 19 g auf 95 ml Dichlormethan und 38 ml 5%ige wäßrige Natriumhydroxidlösung aufgeteilt. Die basische wäßrige Phase wurde mit weiteren
- 47,5 ml Dichlormethan extrahiert, und die vereinte organische Phase wurde mit 0,56 g Aktivkohle und 11 g Kaliumcarbonat gerührt, durch Kieselgur filtriert, mit Dichlormethan gespült, und das Filtrat wurde einrotiert. Man erhielt 9,7 g (Gesamtausbeute 93,4%) des gewünschten Produkts, 9-Amino-1,2,3,4-tetrahydroacridin, als Feststoff mit einer Reinheit von > 99% gemäß GC-Analyse.
- Tacrin (freie Base, 10 g) in 6N Salzsäure (50 ml) wird gerührt und erhitzt, bis sich der gesamte Feststoff gelöst hat. Die heiße Lösung wird in einen sauberen Kolben umgegossen, und Acetonitril (50 ml) wird zugegeben. Das HCl-Salz von Tacrin wird beim Abkühlen ausgefällt. Dieser Feststoff wird bei 0ºC isoliert und unter Vakuum getrocknet; man erhält 90% Feststoff
- Das HCl-Salz von Tacrin (7 g) wird aus 3:1 (Vol./Vol.) Acetonitril/wasser (42 ml) umkristallisiert. Nachdem der Feststoff ausgefällt wurde, wird mehr Acetonitril (28 ml) zugegeben, um die Aufschlämmung röhrfähiger zu machen. Der Feststoff wird bei 0ºC isoliert und unter Vakuum getrocknet; man erhält 90% Feststoff (Reinheit > 99,9% gemäß HPLC-Analyse).
Claims (5)
1. Verfahren zur Herstellung von 9-Amino-1,2,3,4-tetrahydroacridin, die Umsetzung von
2-Aminobenzonitril und Cyclohexanon und Basischmachen des Produkts mit wäßriger
Natriumhydroxidlösung umfassend und dadurch gekennzeichnet, daß die Reaktion in
einem Schritt in Gegenwart eines aus der Gruppe p-Toluolsulfonsäure und deren Hydrate,
Methansulfonsäure und Schwefelsäure ausgewählten Säurekatalysators in einem
geeigneten Lösemittel erfolgt.
2. Verfahren gemäß Anspruch 1, wobei das Lösemittel aus Xylolen, Toluol, aromatischen
Ethern wie Anisol und Dimethoxybenzol und halogenierten Aromaten wie beispielsweise
Chlorbenzol und Dichlorbenzol ausgewählt wird.
3. Verfahren gemäß Anspruch 2, wobei als Lösemittel Xylole eingesetzt werden.
4. Verfahren gemäß Anspruch 1, wobei der Säurekatalysator
p-Toluolsulfonsäuremonohydrat ist und als Lösemittel Xylole eingesetzt werden.
5. Verfahren gemäß Anspruch 1, wobei das Produkt als freie Base durch Umsetzen der
freien Base mit Salzsäure in das Hydrochloridsalz umgesetzt wird.
