Description
- Die vorliegende Erfindung bezieht sich auf 4-Aminopiperidin-Derivate, auf Verfahren zu deren Herstellung, auf ihre Verwendung in der Therapie und auf pharmazeutische Zusammensetzungen, die diese enthalten. Spezieller sind diese Verbindungen zur Behandlung von Erkrankungen des zentralen und/oder pheripheren Nervensystems nützlich. Von besonderem Interesse ist die starke depressionshemmende Wirkung, die diese Verbindungen zeigen.
- Deshalb sind diese 4-Aminopiperidin-Derivate zur Vorbeugung und/oder Behandlung von Depression, schwerer Depression mit Angst, Angsterkrankungen und Affektstörungen besonders nützlich.
- Es wird berichtet, daß Depression bis zu 10 % der Bevölkerung mit einer Lebenszeitprävalenz von 19 % betrifft und mit einer signifikanten Sterberate verbunden ist. Der traditionelle Behandlungsansatz von Depression mit tricyclischen Antidepressiva erfuhr eine verringerte Popularität seit der Einführung von Medikamenten, die speziell auf das Gehirn-Serotonin-System gerichtet sind, nämlich spezifische Serotoninwiederaufnahmeinhibitoren (SSRIs), die von dem weit verbreiteten Fluoxetin repräsentiert werden. Wenn auch SSRIs im Vergleich zu tricyclischen Verbindungen tatsächlich verbesserte Nebenwirkungen aufweisen, zeigen die gesammelten Daten, daß diese Kategorie von Medikamenten die Symptome von Angst und Insomnie, die ein inhärenter Teil von Depression sind, nicht ausreichend behandeln. Außerdem können diese Substanzen selbst Nervosität, Insomnie und Angst induzieren. Daher benötigt eine wesentliche Anzahl von Patienten die Coverabreichung von Anxiolytika/Hypnotika, wie Benzodiazepine oder Antihistamine. Die letzteren Verbindungen, insbesondere Hydroxyzin, können geeigneter sein als Benzodiazepine bei der Coverabreichung mit SSRIs. Eine andere wichtige Nebenwirkung von SSRIs ist die Sexualdysfunktion, die scheinbar durch Serotonin-5-HT2-Rezeptoren vermittelt wird.
- Deshalb könnte, um die Mehrfachtherapie zu vermeiden, eine chemische Einheit, die die Wirksamkeit von SSRIs besitzt und zusätzliche Eigenschaften hat, die Nervosität, Angst und Sexualdysfunktion lindern könnten, für einen Patienten von großem Nutzen sein. Insbesonde re sollte ein solches Molekül wichtige Affinitäten zu Serotoninwiederaufnahmestellen (Hauptweise der Wirkung von SSRIs); Histamin-H1-Rezeptoren, um Nervosität, Angst zu verbessern und Schlaf zu erleichtern; und Serotonin-5-HT2-Rezeptoren haben, von deren Blockade erwartet wird, daß sie die Sexualnebenwirkungen verhindert.
- Unser Forschungsansatz in diesem Gebiet führte uns zur Entdeckung von Molekülen, die diese 3 Eigenschaften besitzen, d. h. Inhibierung der Serotoninwiederaufnahmestellen, Blockade von histaminergen H1-Rezeptoren und Blockade von Serotonin-5-HT2-Rezeptoren.
- Jüngste pharmakologische Studien, die durch die Anmelder durchgeführt wurden, offenbarten nicht erkannte und wirksame pharmakologische Eigenschaften der hierin erwähnten, neuen 4-Aminopiperidin-Derivate der Formel (I), die darauf schließen lassen, daß sie bei der Behandlung von Erkrankungen, wie die oben erwähnten, nützlich sind, aber nicht darauf beschränkt sind.
- US-Patent 5,461,066 beschreibt 4-Aminopiperidin-Derivate als Synthesezwischenprodukte. Die Zusammenfassung der Japanischen Patentanmeldung
JP 05148234 - Gemäß einem Aspekt stellt die vorliegende Erfindung folglich 4-Aminopiperidin-Derivate der Formel I, einschließlich pharmazeutisch akzeptabler Salze hiervon, bereit worin R1, R2, R3, R4 unabhängig voneinander aus Wasserstoff, Fluor, Chlor, Methyl und Trifluormethyl ausgewählt sind, mit der Maßgabe, daß, wenn R1, R2 und R4 Wasserstoff sind, dann R3 nicht Wasserstoff oder Halogen ist.
- Bevorzugte Verbindungen gemäß der Erfindung sind Verbindungen der Formel I, worin:
R2 Wasserstoff, Fluor oder Methyl ist,
R4 Wasserstoff, Fluor, Chlor oder Trifluormethyl ist,
R1 und R3 dieselben Definitionen, wie oben beschrieben, aufweisen, mit der Maßgabe, daß, wenn R1, R2 und R4 Wasserstoff sind, dann R3 nicht Wasserstoff oder Halogen ist,
oder ein pharmazeutisch akzeptables Salz hiervon. - Stärker bevorzugte Verbindungen gemäß der Erfindung sind Verbindungen der Formel I, worin:

R3 Wasserstoff, Fluor, Chlor oder Methyl ist,

oder ein pharmazeutisch akzeptables Salz hiervon. - Am stärksten bevorzugte Verbindungen gemäß der Erfindung sind Verbindungen der Formel I, worin:

N-(4-Methylbenzyl)-N-phenyl-4-piperidinamin; N-(4-Fluorbenzyl)-N-(3-fluorphenyl)-4-piperidinamin; N-(3-Fluorphenyl)-N-(4-methylbenzyl)-4-piperidinamin; N-(4-Fluorbenzyl)-N-(4-fluorphenyl)-4-piperidinamin; N-(4-Fluorphenyl)-N-(4-methylbenzyl)-4-piperidinamin oder pharmazeutisch akzeptable Salze hiervon. - Die besten Ergebnisse sind mit N-(4-Fluorbenzyl)-N-(3-fluorphenyl)-4-piperidinamin und N-(3-Fluorphenyl)-N-(4-methylbenzyl)-4-piperidinamin oder pharmazeutisch akzeptablen Salzen hiervon erhalten worden.
- Die erfindungsgemäßen „pharmazeutisch akzeptablen Salze" umfassen therapeutisch wirksame, nicht toxische Säuresalzformen, die die Verbindungen der Formel I bilden können. Die Säureadditionssalzform einer Verbindung der Formel I, die als ihre freie Form als eine Base auftritt, kann durch Behandeln der freien Base mit einer geeigneten Säure, wie eine anorganische Säure, beispielsweise eine Halogenwasserstoff-, wie Salz- oder Bromwasserstoff-, Schwefel-, Salpeter-, Phosphorsäure und dergleichen; oder eine organische Säure, wie beispielsweise Essig-, Hydroxyessig-, Propan-, Milch-, Pyruvin-, Malon-, Bernstein-, Malein-, Fumar-, Äpfel-, Wein-, Zitronen-, Methansulfon-, Ethansulfon-, Benzolsulfon-, p-Toluol- sulfon-, Cyclohexansulfamin-, Salicyl-, p-Aminosalicyl-, Pamoinsäure und dergleichen, erhalten werden.
- Umgekehrt können diese Salzformen in die freien Basenformen durch Behandlung mit einer geeigneten Base umgewandelt werden.
- Die Verbindungen der Formel I und ihre Salze können in Form eines Solvats vorliegen, das innerhalb des Umfangs der vorliegenden Erfindung einbezogen ist. Diese Solvate umfassen beispielsweise Hydrate, Alkoholate und dergleichen.
- Die vorliegende Erfindung betrifft ebenso Verfahren zur Herstellung von Verbindungen der Formel I.
- Die Verbindungen der Formel I gemäß der Erfindung können analog zu konventionellen Verfahren hergestellt werden, wie es einem Fachmann für synthetische organische Chemie verständlich ist.
- Die folgende Verfahrensbeschreibung stellt bestimmte Synthesewege in einer illustrativen Weise dar. Andere alternative und/oder analoge Verfahren werden dem Fachmann ohne weiteres offensichtlich sein.

- Die Schutzgruppe P kann irgendeine geeignete Aminschutzgruppe sein, wie beispielsweise Carbamat, Sulfenylderivate, Sulfonylderivate, Alkyl und Aryl. Nicht-einschränkende Beispiele sind Methoxycarbonyl, Ethoxycarbonyl, tert-Butoxycarbonyl (Boc), 9-Fluorenylmethoxycarbonyl (Fmoc), 9-(2-Sulfo)fluorenylmethoxycarbonyl, 9-(2,7-Dibrom)fluorenylmeth oxycarbonyl, 2,2,2-Trichlorethoxycarbonyl (Troc), 2-Phenylethoxycarbonyl, 2-Chlorethoxycarbonyl, Benzyloxycarbonyl, p-Methoxybenzyloxycarbonyl, Benzolsulfenyl, 2-Nitrobenzolsulfenyl, Tosyl, Benzolsulfonyl, Methyl, tert-Butyl, Allyl, Benzyl, Bis(4-methoxyphenyl)methyl oder 2,4-Dinitrophenyl. - Diese Umwandlung kann gemäß jeder Verfahrensweise, die dem Fachmann bekannt ist, durchgeführt werden.

- Diese Umwandlung kann in einem inerten Lösungsmittel, beispielsweise Acetonitril oder Dimethylformamid, in Gegenwart einer organischen oder anorganischen Base, beispielsweise Triethylamin oder Kaliumcarbonat, zwischen 50 und 100 °C durchgeführt werden.
- Die Verbindungen der Formel IV sind kommerziell erhältlich.

- Diese Umwandlung kann in einem alkoholischen Lösungsmittel, beispielsweise Methanol, in Gegenwart von 1 bis 2 Äquivalenten einer Säure, beispielsweise Essigsäure, und in Gegenwart von 1 bis 2 Äquivalenten eines Reduktionsmittels, wie Natriumcyanoborhydrid, durchgeführt werden.
- Die Verbindungen der Formel V und die Verbindungen der Formel VI sind kommerziell erhältlich.
- In einem anderen Aspekt stellt die Erfindung 4-Aminopiperidin-Derivate der Formel (I) bereit, einschließlich der pharmazeutisch akzeptablen Salze hiervon zu ihrer Verwendung als Medikament.
- Es ist schließlich herausgefunden worden, daß die Verbindungen der Formel (I) und die pharmazeutisch akzeptablen Salze hiervon besonders wirksame Antidepressiva sind.
- Deshalb sind diese Verbindungen zur Vorbeugung und/oder Behandlung von Depression und schwerer Depression mit Angst besonders nützlich.
- Diese Verbindungen können ebenso zur Vorbeugung und/oder Behandlung von anderen neurologischen Erkrankungen nützlich sein, einschließlich Angststörungen, insbesondere generalisierte Angststörung (GAD), Panikstörung (PD), posttraumatische Belastungsstörung (PTSD), soziale Angststörung (SAD), obsessiv-kompulsive Störung und Agoraphobie, bipolare Störungen, Manie, chronischer Schmerz, neuropathischer Schmerz, Migräne, Hirnischämie, Herzarrhythmie, Myotonie, Kokain- und Alkoholabhängigkeit, Schlaganfall, Myoklonus, Tremor, Neugeborenenhirnblutung, amyotrophe Lateralsklerose (ALS), Spastizität, Parkinson-Krankheit und andere neurodegenerative und Bewegungsstörungen.
- Daher betrifft die vorliegende Erfindung in einem weiteren Aspekt die Verwendung von Verbindungen der Formel (I) oder der pharmazeutisch akzeptablen Salze hiervon, wie oben definiert, zur Herstellung eines Medikaments für die Behandlung und/oder Prophylaxe von neurologischen Erkrankungen, wie oben erwähnt.
- Insbesondere betrifft die vorliegende Erfindung die Verwendung von Verbindungen der Formel (I) und der pharmazeutisch akzeptablen Salze hiervon, wie oben definiert, zur Herstellung eines Medikaments für die Behandlung und/oder Prophylaxe von Depression.
- Die vorliegende Beschreibung beschreibt ebenso ein Verfahren zur Behandlung von Depression bei einem Säuger, der eine solche Behandlung benötigt, umfassend die Verabreichung einer therapeutischen Dosis von mindestens einer Verbindung der Formel (I) und der pharmazeutisch akzeptablen Salze hiervon an einen Patienten.
- Die beschriebenen Verfahren umfassen die Verabreichung einer pharmazeutischen Zusammensetzung gemäß der Erfindung in einer Menge, die ausreichend ist, den Zustand zu lindern, an einen Säuger (vorzugsweise Menschen), der unter den oben erwähnten Krankheiten leidet. Die Verbindung wird günstigerweise in irgendeiner geeigneten Dosiereinheitsform verabreicht, einschließlich, aber nicht darauf beschränkt, einer, die 0,5 bis 500 mg, vorzugsweise 1 bis 100 mg Wirkstoff pro Dosiereinheitsform enthält.
- Unter dem Ausdruck „Behandlung", wie von dem Anmelder verwendet, ist die Heilbehandlung und prophylaktische Behandlung zu verstehen.
- Unter „heilend" verstehen wir die Wirksamkeit der Formel (I) bei der Behandlung der aktuellen Episode der depressiven Phase.
- Unter „prophylaktisch" oder „Erhaltung" verstehen wir die Vorbeugung des Wiederauftretens der depressiven Episoden.
- Zur Behandlung der Krankheiten können die Verbindungen der Formel (I) oder ihre pharmazeutisch akzeptablen Salze bei einer wirksamen täglichen Dosis eingesetzt und über eine pharmazeutische Zusammensetzung verabreicht werden.
- Deshalb ist eine andere Ausführungsform der vorliegenden Erfindung eine pharmazeutische Zusammensetzung, die eine wirksame Menge einer Verbindung der Formel I oder ihrer pharmazeutisch akzeptablen Salze oder ein Derivat in Kombination mit einem pharmazeutisch akzeptablen Träger für irgendeine der hierin beschriebenen Erkrankungen umfaßt.
- Um die erfindungsgemäße pharmazeutische Zusammensetzung herzustellen, werden eine oder mehrere der Verbindungen der Formel (I) oder ihre pharmazeutisch akzeptablen Salze gründlich mit einem pharmazeutischen Träger gemäß den konventionellen pharmazeutischen Mischtechniken vermischt, wobei der Träger eine breite Vielzahl von Formen in Abhängigkeit der Herstellungsform, die zur Verabreichung, beispielsweise oral, rektal oder parenteral, gewünscht ist, annehmen kann.
- Die vorliegende Erfindung erfordert die Verabreichung einer wirksamen Dosis der Verbindungen zur Behandlung und/oder Prophylaxe von Krankheiten. Die Dosis, die gemäß der Erfindung erforderlich ist, sollte ausreichend hoch sein, um die Linderung der Krankheiten zu ermöglichen. Pharmazeutische Zusammensetzungen, die die Verbindungen umfassen, können beispielsweise oral oder parenteral, d. h. intravenös, intramuskulär oder subkutan, intrathekal verabreicht werden.
- Pharmazeutische Zusammensetzungen, die zur oralen Verabreichung verwendet werden können, können Feststoffe oder Flüssigkeiten sein, und können beispielsweise in Form von Tabletten, Pillen, Dragees, Gelatinekapseln, Lösungen, Sirups und dergleichen vorliegen.
- Zu diesem Zweck können die Verbindungen gemischt mit einem inerten Verdünnungsmittel oder einem nicht-toxischen pharmazeutisch akzeptablen Vehikel, wie beispielsweise Stärke oder Laktose, verwendet werden. Gegebenenfalls können diese pharmazeutischen Zusammensetzungen ebenso ein Bindemittel, wie mikrokristalline Cellulose, Tragantgummi oder Gelatine, einen Lösungsvermittler, wie Alginsäure, ein Schmiermittel, wie Magnesiumstearat, ein Gleitmittel, wie kolloides Siliciumdioxid, ein Süßungsmittel, wie Saccharose oder Saccharin, oder Farbmittel oder ein Aromastoff wie Pfefferminz oder Methylsalicylat, enthalten.
- Sie umfassen ebenso Zusammensetzungen, die den Wirkstoff in einer kontrollierten Weise freisetzen können. Pharmazeutische Zusammensetzungen, die zur parenteralen Verabreichung verwendet werden, liegen in pharmazeutischen Formen vor, die für diese Verabreichungsweise bekannt sind, und liegen in Form von wässerigen oder öligen Lösungen oder Suspensionen vor, die im allgemeinen in Ampullen, Wegwerfspritzen, Glas- oder Kunststoffphiolen oder Infusionsbehältern enthalten sind.
- Zusätzlich zu der aktiven Verbindung können diese Lösungen oder Suspensionen gegebenenfalls ebenso ein steriles Verdünnungsmittel, wie Wasser zur Injektion, eine physiologische Kochsalzlösung, Öle, Polyethylenglykole, Glycerin, Propylenglykol oder andere synthetische Lösungsmittel, antibakterielle Mittel, wie Benzylalkohol, Antioxidationsmittel, wie Ascorbinsäure oder Natriumbisulfit, Chelatbildner, wie Ethylendiamin-tetra-essigsäure, Puffer, wie Acetate, Zitrate oder Phosphate, und Mittel zum Einstellen der Osmolarität, wie Natriumchlorid oder Dextrose sein.
- Diese pharmazeutischen Formen werden unter Verwendung der Verfahren, die routinemäßig von Pharmazeuten verwendet werden, hergestellt.
- Der Prozentsatz der Verbindung der Formel (I) in den pharmazeutischen Zusammensetzungen kann innerhalb eines weiten Bereiches an Konzentrationen fallen und hängt von einer Vielzahl von Faktoren, wie Geschlecht, Alter, Gewicht und medizinischem Zustand des Patienten sowie dem Verabreichungsverfahren ab. Daher beträgt die Menge an Verbindung der Formel (I) in den Zusammensetzungen zur oralen Verabreichung mindestens 0,5 Gew.-% und kann bis zu 80 Gew.-% in bezug auf das Gewicht der Zusammensetzung betragen.
- In einer anderen Ausführungsform betrifft die vorliegende Erfindung ebenso die Synthesezwischenprodukte der Formel II oder III, worin die Substituenten R und die Schutzgruppe P wie oben definiert sind.

- Die folgenden Beispiele werden nur zu illustrativen Zwecken bereitgestellt und sollen die Erfindung in keiner Weise einschränken. Der Fachmann wird einschätzen, daß Routineveränderungen und -modifikationen der folgenden Beispiele ohne Überschreitung des Geistes oder Umfangs der Erfindung gemacht werden können.
- Wenn nicht anders in den Beispielen angegeben, wurde die Charakterisierung der Verbindungen gemäß den folgenden Verfahren durchgeführt:
NMR-Spektren werden auf einem BRUKER AC 250 Fourier-Transformations-NMR-Spektrometer, ausgestattet mit einem Aspect-3000-Computer und einem 5 mm 1H/13C-Doppelprobenkopf, oder BRUKER DRX 400 FT NMR, ausgestattet mit einem SG Indigo2-Computer und einem 5 mm Umkehrgeometrie-1H/13C/15N-Dreifachprobenkopf, aufgezeichnet. Die Verbindung wird in DMSO-d6-(oder CDCl3-)-Lösung bei einer Probentemperatur von 313 K und bei Konzentrationen zwischen 2 und 20 mg/ml untersucht. Die Vorrichtung wird auf das Deuteriumsignal von DMSO-d6 (oder CDCl3) gesperrt. Die chemischen Verschiebungen werden in ppm nach tieferen Feldern von TMS verschoben, das als interner Standard verwendet wird, angegeben. DMSO-d6 (deuteriertes Dimethylsulfoxid). - Massenspektrometrische Messungen in LC/MS-Weise werden folgendermaßen durchgeführt:
HPLC-Bedingungen - Die Analysen werden unter Verwendung eines WATERS Alliance HPLC-Systems, ausgestattet mit einer INERTSIL ODS 3-, DP 5 μm, 250 × 4,6 mm Säule, durchgeführt.
- Der Gradient läuft von 100 % Lösungsmittel A (Acetonitril, Wasser, TFA (10/90/0,1, V/V/V)) bis 100 % Lösungsmittel B (Acetonitril, Wasser, TFA (90/10/0,1 (V/V/V)) in 7 min mit einer Verweilzeit bei 100 % B von 4 min. Die Fließgeschwindigkeit wird auf 2,5 ml/min eingestellt und ein Abzweig von 1/10 wird noch vor der API-Quelle verwendet. Die Chromatographie wird bei 30 °C durchgeführt.
- MS-Bedingungen
- Die Proben werden in Acetonitril/Wasser, 70/30, V/V, bei der Konzentration von etwa 250 μg/ml gelöst. API-Spektren (+ oder –) werden unter Verwendung eines FINNIGAN (San Jose, Ca, USA) LCQ-Ion-Trap-Massenspektrometer durchgeführt. Die APCI-Quelle arbeitet bei 450 °C und der Kapillarheizer bei 160 °C. Die ESI-Quelle arbeitet bei 3,5 kV und der Kapillarheizer bei 210 °C.
- Massenspektrometrische Messungen in EI/DIP-Weise werden folgendermaßen durchgeführt:
Die Proben werden durch Erhitzen der Probe von 50 °C auf 250 °C in 5 min verdampft. EI-Spektren (Elektronenstoßspektren) werden unter Verwendung eines FINNIGAN (San Jose, CA, USA) TSQ 700 Tandem-Quadrupolmassenspektrometers aufgezeichnet. Die Quellentemperatur wird auf 150 °C eingestellt. - Massenspektrometrische Messungen auf einem TSQ 700 Tandem-Quadrupolmassenspektrometer (Finnigan MAT, San Jose, CA, USA) in GC/MS-Weise werden mit einem Gaschromatographmodell 3400 (Varian, Walnut Creek, CA, USA), ausgestattet mit einem Split-/splitlosen-Injektor und einer DB-5MS Quarzglassäule (15 m × 0,25 mm ID, 1 μm) von J & W Scientific (Folsom, CA, USA), durchgeführt. Helium (Reinheit 99,999 %) wird als Trägergas verwendet. Der Injektor (CTC A200S Autosampler) und die Übertragungsleitung arbeiten bei 290 bzw. 250 °C. Die Probe (1 μl) wird in splitloser Weise injiziert und die Ofentemperatur folgendermaßen programmiert: 50 °C für 5 min, erhöhen auf 280 °C (23 °C/min) und halten für 10 min. Der TSQ-700-Spektrometer arbeitet in Elektronenstoßweise (EI) oder chemischer Ionisierungsweise (CH/CH4) (Massebereich 33 bis 800, Scannzeit 1,00 s). Die Quellentemperatur wird auf 150 °C eingestellt.
- Der Wassergehalt wird unter Verwendung eines mikrocoulometrischen Karl-Fischer-Titrators von Metrohm bestimmt.
- Die präparativen, chromatographischen Trennungen werden auf Kieselgel 60 Merck, Teilchengröße 15 bis 40 μm, Referenz 1.15111.9025, unter Verwendung von betriebsintern modifizierten Säulen mit mittigem Druck vom Jobin Y von-Typ (80 mm ID), Fließgeschwindigkei ten zwischen 70 und 150 ml/min durchgeführt. Die Menge an Kieselgel und Lösungsmittelgemischen sind wie in den einzelnen Verfahrensweisen beschrieben.
- Die Schmelzpunkte werden auf einem Büchi-535-Totoli-Typ-Fusionometer, und werden nicht korrigiert, oder durch die Starttemperatur auf einem Perkin Elmer DSC 7 bestimmt.
- Wenn in den Beispielen nicht anders angegeben, werden die Verbindungen in der neutralen Form erhalten.
- 1.1 Synthese von tert-Butyl-4-(3-fluoranilino)-1-piperidincarboxylat 1.
- 3-Fluoranilin (40 g) wurde in MeOH gelöst und die Lösung wurde auf 0 °C abgekühlt. tert-Butyl-4-oxo-1-piperidincarboxylat (79 g, 1,1 val), Essigsäure (26,8 ml, 1,3 val) und Natriumcyanoborhydrid (29 g, 1,3 val, portionsweise) wurden nacheinander zu der Lösung zugegeben. Das Reaktionsgemisch wurde bei Raumtemperatur bis zum Ende gerührt. Das Reaktionsgemisch wurde dann auf 0 °C abgekühlt und eine wässerige Lösung aus NaOH (20 %) wurde in das Gemisch gegossen (pH ~ 10). Der Niederschlag wurde abfiltriert, mit Wasser gewaschen, getrocknet und aus iPrOH umkristallisiert, wodurch tert-Butyl-4-(3-fluoranilino)-1-piperidincarboxylat 1 (68,5 g) erhalten wurde.
Ausbeute: 63 %.
MS (MH+): 295. - Die Verbindungen der Formel III, die in Tabelle 1 aufgelistet sind, können in einer analogen Weise synthetisiert werden.
- 1.2 Synthese von tert-Butyl-4-[3-fluor(4-fluorbenzyl)anilino]-1-piperidincarboxyl
- tert-Butyl-4-(3-fluoranilino)-1-piperidincarboxylat 1 (20 g) wurde in Acetonitril (250 ml) gelöst. Kaliumcarbonat (33,2 g) und 1-(Brommethyl)-4-fluorbenzol (11 ml) wurden zu der Lösung zugegeben. Das Reaktionsgemisch wurde unter Rückfluß über Nacht erhitzt, dann abgekühlt, konzentriert und der Rest wurde in Dichlormethan und Wasser aufgenommen. Die wässerige Schicht wurde zweimal mit Dichlormethan gewaschen. Die vereinigten organischen Schichten wurden über MgSO4 getrocknet und konzentriert. Der resultierende weiße Feststoff wurde in Hexan zerrieben und abfiltriert, wodurch tert-Butyl-4-[3-fluor(4-fluorbenzyl)anilino]-1-piperidincarboxylat 8 (25,5 g) erhalten wurde.
Ausbeute: 89 %.
MS (MH+): 403. - Die Verbindungen der Formel II, die in Tabelle 2 aufgelistet sind, können in einer analogen Weise synthetisiert werden.
- 1.3 Synthese von N-(4-Fluorbenzyl)-N-(3-fluorphenyl)-4-piperidinamin 43.
- tert-Butyl-4-[3-fluor(4-fluorbenzyl)anilino]-1-piperidincarboxylat 8 (25,5 g) wurde in Dichlormethan gelöst und die Lösung wurde auf 0 °C abgekühlt. TFA (30 ml) wurde zu der Lösung zugegeben und das Reaktionsgemisch wurde bei Raumtemperatur gerührt. Nach 1 h 30 min wurde TFA (20 ml) zugegeben und das Reaktionsgemisch wurde für weiter 30 Minuten gerührt. Das Lösungsmittel wurde unter Vakuum entfernt und der Rest wurde in Dichlormethan und Wasser aufgenommen. Die wässerige Phase wurde durch Zugabe von NaOH alkalisiert und mit Dichlormethan gewaschen. Die vereinigten organischen Schichten wurden über MgSO4 getrocknet und konzentriert. Das Rohprodukt wurde durch Chromatographie gereinigt (Elutionsmittel: CH2Cl2/MeOH/NH4OH 95/5/0,5), wodurch N-(4-Fluorbenzyl)-N-(3-fluorphenyl)-4-piperidinamin (18,4 g) als freie Base erhalten wurde.
- Eine Lösung aus Ether, gesättigt mit HCl, wurde zu N-(4-Fluorbenzyl)-N-(3-fluorphenyl)-4-piperidinamin, gelöst in Ether, zugegeben. Der Niederschlag wurde abfiltriert, mit Ether gewaschen und in einem Ofen (50 °C) unter Vakuum getrocknet, wodurch 17,51 g N-(4-Fluorbenzyl)-N-(3-fluorphenyl)-4-piperidinamin 43 (2HCl, 0,5 H2O) als weißer Feststoff erhalten wurden.
Ausbeute: 72 %.
MS (MH+): 303. - Die Verbindungen der Formel I, die in Tabelle 3 aufgelistet sind, können in einer analogen Weise synthetisiert werden.
- Beispiel 2: Bindungsassays
- 2.1 H1-Bindung.
- Die Affinität der Testverbindungen für den menschlichen Histamin-H1-Rezeptor wurde durch einen [3H]-Mepyramin-Bindungsassay bewertet. Diese Bindung wurde, wie von Gillard et al. (Gillard M., Van der Perren C., Moguilevsky N., Massingham R., Chatelain P., Mol. Pharmacol. (2002), 61, 391–399) beschrieben, durchgeführt.
- 2.2 5-HT-Aufnahme.
- Die Affinität der Testverbindungen für den Serotonin-Transporter wurde durch einen [3H]-Paroxetin-Bindungsassay bewertet. Diese Bindung wurde, wie von Marcusson et al. (Marcusson J. O., Bergstrom M., Eriksson K., Ross S. B., J. Neurochem. (1988), 50, 1783) beschrieben, mit leichten Modifikationen durchgeführt. Die Membranproteine (100 bis 200 μg) aus Rattenhirnrinde wurden für 120 min bei 25 °C in 2 ml eines 50 mM Tris-HCl-(pH 7,4)-Puffers, enthaltend 2 mM MgCl2 und 0,05 nM Radioligand, inkubiert. Nicht-spezifische Bindung, die als die restliche Bindung definiert wird, wurde in Gegenwart von 5 μM Imipramin gemessen.
- 2.3 5-HT2-Bindung
- Die Affinität der Testverbindungen für die 5-HT2-Rezeptoren wurde durch einen [3H]-Ketanserin-Bindungsassay bewertet. Diese Bindung wurde gemäß Leysen et al. (Leysen J. E., Niemegeers C. J., Van Nueten J. M., Laduron P. M., Mol. Pharmacol. (1982), 21, 301 – 314) mit leichten Modifikationen durchgeführt. Kurz, es wurden 250 μg Membranproteine aus Rattenhirnrinde für 60 min bei 25 °C in 1 ml eines 50 mM Tris-HCl-(pH 7,4)-Puffers, enthaltend 2 mM MgCl2 und 0,2 nM Radioligand, inkubiert. Nicht-spezifische Bindung, die als die restliche Bindung definiert wird, wird in Gegenwart von 1 μM Chlorpromazin gemessen.
Claims (14)
- 4-Aminopiperidin-Verbindung der Formel I, einschließlich eines pharmazeutisch akzeptablen Salzes hiervon worin R1, R2, R3, R4 unabhängig voneinander aus Wasserstoff, Fluor, Chlor, Methyl und Trifluormethyl ausgewählt sind, mit der Maßgabe, daß, wenn R1, R2 und R4 Wasserstoff sind, dann R3 nicht Wasserstoff oder Halogen ist.
- 4-Aminopiperidin-Verbindung nach Anspruch 1, worin R2 Wasserstoff, Fluor oder Methyl ist; und R4 Wasserstoff, Fluor, Chlor oder Trifluormethyl ist.
- 4-Aminopiperidin-Verbindung nach Anspruch 1, worin R1 Wasserstoff, Fluor oder Chlor ist; R2 Wasserstoff, Fluor oder Methyl ist; R3 Wasserstoff, Fluor, Chlor oder Methyl ist; und R4 Wasserstoff, Fluor oder Chlor ist.
- 4-Aminopiperidin-Verbindung nach Anspruch 1, worin R1 Wasserstoff oder Fluor ist; R2 Wasserstoff oder Fluor ist; R3 Fluor oder Methyl ist; und R4 Wasserstoff ist.
- 4-Aminopiperidin-Verbindung nach Anspruch 1, ausgewählt aus der Gruppe, bestehend aus N-(4-Methylbenzyl)-N-phenyl-4-piperidinamin; N-(4-Fluorbenzyl)-N-(3-fluorphenyl)-4-piperidinamin; N-(3-Fluorphenyl)-N-(4-methylbenzyl)-4-piperidinamin; N-(4-Fluorbenzyl)-N-(4-fluorphenyl)-4-piperidinamin; N-(4-Fluorphenyl)-N-(4-methylbenzyl)-4-piperidinamin oder pharmazeutisch akzeptablen Salze hiervon.
