Description
Bei den Cannabinoid-Verbindungen handelt es sich um aus Cannabis-
Arten isolierbare Inhaltsstoffe. Aufgrund der physiologischen
Wirksamkeit kommt dem Trans-delta-9-tetrahydrocannabinol (Δ⁹-THC)
eine wesentliche Bedeutung zu. Diese Verbindung wird auch als
6a,7,8,10a-Tetrahydro-6,6,9-trimethyl-3-pentyl-6H-
dibenzo[b,d]pyran-1-ol bezeichnet.
Die psychotropen aber nicht Sucht-erzeugenden Wirkungen machen
diese Verbindung als pharmazeutischen Wirkstoff interessant.
Aus dem Stand der Technik sind verschiedene Verfahren zur
Herstellung von Δ⁹-THC bekannt. Nachteilig bei den bekannten
Verfahren ist jedoch, daß bei den bekannten Synthesewegen eine
Anzahl von Nebenprodukten gebildet werden, die nur sehr schwer von
dem gewünschten Endprodukt abgetrennt werden können. Darüber
hinaus liegt das Endprodukt häufig in einer harzartigen Form vor,
was die Reinigung nicht gerade erleichtert. Aufgrund dieser
Nachteile stößt die Herstellung im größeren Maßstab auf erhebliche
Schwierigkeiten.
Das in der vorliegenden Anmeldung beschriebene 6,12-Dihydro-
6-hydroxy-cannabidiol wurde bisher weder synthetisch hergestellt,
noch als Zwischenprodukt zur Herstellung von Delta-9-tetrahydrocannabinol
eingesetzt. Eine Erwähnung
in der Literatur (Garrett et al., J. Pharm. Sci., 67 [1978],
S. 27-32] betrifft lediglich den analytisch-chromatografischen Spurennachweis
als eines unter vielen anderen Zersetzungsprodukten
von Delta-9-THC in saurer Lösung. Die Verbindung wurde also bisher weder
in präparativen Mengen hergestellt noch für irgendwelche Umsetzungen
verwendet. Auch die physikalischen Daten dieser Verbindung sind
erstmals durch die vorliegende Anmeldung bekanntgeworden.
Aufgabe der vorliegenden Erfindung ist es daher ein Verfahren zur
Verfügung zu stellen, mit dem Trans-delta-9-tetrahydrocannabinol
sowohl in ausreichender Reinheit wie auch in größerem Maßstab
hergestellt werden kann.
Erfindungsgemäß wird diese Aufgabe dadurch gelöst, daß zunächst
als Zwischenprodukt 6,12-Dihydro-6-hydroxy-cannabidiol
hergestellt wird, das durch Kristallisation leicht gereinigt
werden kann. Dieses Zwischenprodukt wird anschließend durch eine
Ringkondensation zu dem gewünschten Delta-9-THC umgesetzt.
Das erfindungsgemäße Herstellungsverfahren wird durch das
nachfolgend dargestellte Schema verdeutlicht:
Erfindungsgemäß wird zunächst das Zwischenprodukt 6,12-Diyhdro-
6-hydroxy-cannabidiol hergestellt, das auch als 1,3-Dihydroxy-2-
[6-(1-hydroxy-1-methyl-ethyl)-3-methyl-2-cyclohexen-1-yl]-5-
pentyl-benzol bezeichnet werden kann. Diese Verbindung ist im
obigen Formelschema mit der Bezeichnung (I) dargestellt.
Zur Synthese des erfindungsgemäßen Zwischenproduktes 6,12-
Dihydro-6-hydroxy-cannabidiol wird von den leicht zugänglichen
Ausgangsverbindungen Olivetol (Formel (II)) und cis-p-Menth-2-
en-1,8-diol (Formel (III)) ausgegangen. Die Umsetzung erfolgt in
einem geeigneten Lösungsmittel, wobei sich aromatische
Kohlenwasserstoffe, wie z. B. Benzol und Toluol, halogenierte
Kohlenwasserstoffe, wie z. B. Methylenchlorid, Chloroform,
Dichlorethan und Trichlorethylen, Ether, wie z. B. Diethylether,
Diisopropylether und Tetrahydrofuran als geeignet herausgestellt
haben. Auch Mischungen der genannten Lösungsmittel können
eingesetzt werden.
Bevorzugt werden bei der erfindungsgemäßen Umsetzung Toluol,
Benzol, Methylenchlorid und Chloroform verwendet.
Die erfindungsgemäße Umsetzung erfolgt in Gegenwart eines
geeigneten Katalysators, wobei Protonsäuren, wie beispielsweise
Halogenwasserstoffsäuren, Schwefelsäure, Phosphorsäure,
Perchlorsäure, organische Sulfonsäuren, wie beispielsweise
Methansulfonsäure und p-Toluolsulfonsäure, organische
Carbonsäuren, wie beispielsweise Oxalsäure, Trifluoressigsäure
oder andere Lewissäuren, wie beispielsweise Bortrifluorid,
Eisen-III-chlorid, Zinkchlorid, Zinkbromid, Zinn-IV-chlorid,
Titanchlorid oder Jod geeignet sind. Auch Mischungen der
einzelnen Katalysatoren können gegebenenfalls verwendet werden.
Bevorzugt verwendet werden im Rahmen der vorliegenden Erfindung
Salzsäure und p-Toluolsulfonsäure.
Das Verfahren kann bei Temperaturen zwischen etwa -30°C und
+50°C durchgeführt werden, wobei Temperaturen zwischen 0°C und
20°C bevorzugt sind.
Die Reaktionszeiten sind abhängig von dem verwendeten
Lösungsmittel, dem Katalysator und der Reaktionstemperatur. Es
können Reaktionszeiten zwischen wenigen Minuten und mehreren
Tagen erforderlich sein.
Das hergestellte Zwischenprodukt kann anschließend durch
Umkristallisation in bequemer Weise weiter gereinigt werden.
Hierbei hat sich die Verwendung von Petrolether zum
Umkristallisieren als geeignet herausgestellt.
Durch Ringkondensation kann aus 6,12-Dihydro-6-hydroxy-
cannabidiol das Trans-delta-9-tetrahydrocannabinol (Δ⁹-THC)
hergestellt werden. Diese Reaktion wird in einem geeigneten
Lösungsmittel unter Verwendung geeigneter Katalysatoren und
wasserbindender Mittel durchgeführt. Als Lösungsmittel werden
Kohlenwasserstoffe, wie beispielsweise Hexan, Heptan,
Cyclohexan, Petrolether, aromatische Kohlenwasserstoffe, wie
beispielsweise Benzol, Toluol, chlorierte Kohlenwasserstoffe,
wie beispielsweise Methylenchlorid, Chloroform, Dichlorethan
verwendet. Bevorzugt verwendet werden Methylenchlorid und
Chloroform. Auch Mischungen der Lösungsmittel können Verwendung
finden.
Als Katalysator für die Ringkondensation werden Lewissäuren, wie
beispielsweise Zinkchlorid, Zinkbromid, Bortrifluorid, Eisen-
III-chlorid, Zinn-IV-chlorid, Titanchlorid oder Jod verwendet,
wobei sich als besonders geeignet Zinkchlorid und Zinkbromid
herausgestellt hat.
Als wasserbindende Mittel werden neutrale Mittel wie
beispielsweise Magnesiumsulfat, Natriumsulfat, Kalziumsulfat
oder Molekularsiebe verwendet, wobei sich die letztgenannten als
besonders geeignet herausgestellt haben.
Durchgeführt wird die Reaktion bei einer Temperatur zwischen
etwa -20°C und dem Siedepunkt des entsprechenden Lösungsmittels.
Die benötigten Reaktionszeiten sind abhängig vom Katalysator,
Lösungsmittel, wasserbindenden Mittel und der
Reaktionstemperatur. Je nach den gewählten Bedingungen sind
Zeiten von wenigen Minuten bis zu mehreren Tagen erforderlich.
Für den Fachmann ist klar, daß die erfindungsgemäßen Umsetzungen
auch unter Verwendung von solchen funktionalisierten Derivaten
durchgeführt werden können, die ebenfalls zur Synthese von Δ-9-
THC herangezogen werden können.
Vorteilhaft bei den erfindungsgemäßen Verfahren ist, daß
ausgehend von leicht verfügbaren Ausgangssubstanzen das
Zwischenprodukt 6,12-Dihydro-6-hydroxy-cannabidiol auf einfache
Weise und in guter Ausbeute zugänglich ist und, daß dieses
Produkt ohne große Schwierigkeiten gereinigt werden kann.
Ausgehend von dem erfindungsgemäßen Zwischenprodukt kann sehr
reines, d. h. isomerenarmes Δ⁹-THC gewonnen werden, wobei die
Reinigung des so gewonnenen THC mit einer einfachen Eluierung
von einer Kieselgelsäule möglich ist. Mit Hilfe des
beschriebenen Verfahrens kann eine hohe reproduzierbare Ausbeute
an Δ⁹-THC erreicht werden, und die Herstellung des Endprodukts
ist auch im technischen Maßstab möglich.
90 g Olivetol wurden mit 85 g cis-p-Menth-2-en-1,8-diol und 4 g p-
Toluolsulfonsäure, H₂O in 4 l Methylenchlorid für 24 Stunden bei
20°C gerührt. Dann wurde einmal mit 200 ml 4%iger
Kaliumcarbonatlösung extrahiert und die organische Phase im
Vakuum zu einem Öl eingeengt. Dabei wurden etwa 170 g Rückstand
erhalten, der in 1 Liter Petrolether (50/70) gelöst wurde und
dreimal mit je 300 ml 0,5 N Natronlauge extrahiert wurde.
Die Petroletherphase wurde nun auf eine Kieselgelsäule (ca. 500 g
Kieselgel) aufgetragen und mit einer Mischung von Petrolether
(50/70) und Diisopropylether (2 : 1) eluiert. Das Eluat wurde im
Vakuum eingeengt und der Rückstand (ca. 102 g) aus 600 ml
Petrolether (50/70) umkristallisiert. Erhalten wurden dabei 77 g
des gewünschten Produktes. Bei der Charakterisierung von 6,12-
Dihydro-6-hydroxy-cannabidiol wurden folgende Meßwerte erhalten:
Schmelzpunkt: 77-77,5°C.
Optische Rotation: [α]=-70,3°C (c=1,00; CHCl₃).
Ultraviolett-Spektrum: (UV)
λmax (Ethanol, 95%) 275 nm (ε 1024);
λmax (Ethanol, 95%) 281 nm (ε 987).
Infrarot-Spektrum:
(IR, KBr-Pellet) [cm]-1 3392, 2956, 2930, 2859, 1630, 1582, 1446, 1379, 1369, 1335, 1308, 1231, 1202, 1186, 1158, 1076, 1046, 1028.
Massenspektrum:
(MS, 70 eV, EI) m/z 332 (M⁺, 10), 314 (29) 299 (37), 297 (10), 286 (12), 271 (44), 258 (13), 246 (11), 243 (13), 231 (100), 193 (25), 174 (12), 137 (14), 93 (18), 59 (25), 43 (63); TMS-Derivat: m/z 548 (M⁺).

10 g 6,12-Dihydro-6-hydroxy-cannabinol wurden in 300 ml
Methylenchlorid gelöst und in Gegenwart von 20 g Zinkbromid und
15 g Molekularsieb 3 Å 24 Stunden unter Rückfluß gehalten.
Anschließend wurde filtriert und das Filtrat eingeengt. Der
Rückstand wurde in 100 ml Petrolether aufgenommen und auf eine
Kieselgelsäule (ca. 100 g Kieselgel) aufgetragen. Anschließend
wurde mit Petrolether eluiert und das Eluat eingeengt. Als
Rückstand konnte Δ⁹-THC mit einer Reinheit von 96% erhalten
werden.
Claims (14)
1. Verfahren zur Herstellung von 6,12-Dihydro-6-hydroxycannabidiol
der folgenden Formel
worin Olivetol mit folgender Formel
mit cis-p-Menth-2-en-1,8-diol der Formel
in einem Lösungsmittel unter Verwendung eines Katalysators
umgesetzt wird und das Reaktionsprodukt gegebenenfalls durch
Kristallisation weiter gereinigt wird.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß das
Lösungsmittel ausgewählt ist unter aromatischen
Kohlenwasserstoffen, halogenierten Kohlenwasserstoffen,
Ethern oder Mischungen davon.
3. Verfahren nach Anspruch 2, dadurch gekennzeichnet, daß das
Lösungsmittel ausgewählt ist aus der Gruppe bestehend aus
Toluol, Benzol, Methylenchlorid und Chloroform oder
Mischungen davon.
4. Verfahren nach einem der vorhergehenden Ansprüche, dadurch
gekennzeichnet, daß als Katalysator Protonsäuren, organische
Sulfonsäuren oder andere Lewissäuren verwendet werden.
5. Verfahren nach Anspruch 4, dadurch gekennzeichnet, daß als
Katalysator Salzsäure oder p-Toluolsulfonsäure verwendet
wird.
6. Verfahren nach einem der vorhergehenden Ansprüche, dadurch
gekennzeichnet, daß das 6,12-Dihydro-6-hydroxy-cannabidiol
durch Kristallisation aus Petrolether weiter gereinigt wird.
7. 6,12-Dihydro-6-hydroxy-cannabidiol.
8. Verfahren zur Herstellung von Trans-delta-9-
tetrahydrocannabinol der Formel
worin bei 6,12-Dihydro-6-hydroxy-cannabidiol, das nach einem
Verfahren gemäß einem der Ansprüche 1 bis 7 herstellbar ist,
der Formel
eine Ringkondensation durchgeführt wird.
9. Verfahren gemäß Anspruch 8, dadurch gekennzeichnet, daß die
Ringkondensation in einem Lösungsmittel ausgewählt aus einer
Gruppe bestehend aus Kohlenwasserstoffen, aromatischen
Kohlenwasserstoffen oder chlorierten Kohlenwasserstoffen
durchgeführt wird.
10. Verfahren nach Anspruch 9, dadurch gekennzeichnet, daß als
Lösungsmittel Methylenchlorid oder Chloroform verwendet
wird.
11. Verfahren nach einem der Ansprüche 8 bis 10, dadurch
gekennzeichnet, daß die Ringkondensation in Gegenwart einer
Lewissäure als Katalysator durchgeführt wird.
12. Verfahren nach Anspruch 11, dadurch gekennzeichnet, daß als
Katalysator Zinkchlorid oder Zinkbromid verwendet wird.
13. Verfahren nach einem der Ansprüche 8 bis 12, dadurch
gekennzeichnet, daß die Ringkondensation in Gegenwart eines
wasserbindenden Mittels durchgeführt wird.
14. Verfahren nach Anspruch 13, dadurch gekennzeichnet, daß das
wasserbindende Mittel ein Molekularsieb ist.
