Description
- Die vorliegende Erfindung betrifft Zwischenverbindungen, die zur Herstellung von 5-Amino-2,5-disubstituierte-4-hydroxypentansäurereste enthaltenden Peptidderivaten verwendbar sind, welche bei der Hemmung der Angiotensinogen-Spaltreaktion des Enzyms Renin nützlich sind.
- Das proteolytische Enzym Renin, das ein Molekulargewicht von etwa 40 000 besitzt, wird in der Niere produziert und von ihr in das Blut ausgeschieden. Es ist bekannt, daß es in vivo aktiv ist, indem es das natürlich vorkommende Plasma-Glycoprotein Angiotensinogen spaltet. Im Falle von menschlichem Angiotensinogen tritt die Spaltung an der Bindung zwischen den Leucin (10.)- und Valin (11.)-Aminosäureresten am N-terminalen Ende des Angiotensinogens ein:

- Das zirkulierende durch die obige Spaltungswirkung von Renin gebildete N-terminale Decapeptid (Angiotensin I) wird dann durch den Körper zu einem als Angiotensin II bekannten Octapeptid abgebaut. Angiotensin II ist als potente Drucksubstanz bekannt, d. h. eine Substanz, die in der Lage ist, eine beträchtliche Blutdrucksteigerung herbeizuführen, wobei angenommen wird, daß sie dadurch wirkt, daß sie eine Kontraktion der Blutgefäße und eine Ausschüttung des Natrium-zurückhaltenden Hormons Aldosteron aus der Nebenniere bewirkt.
- Man hat deshalb das Renin-Angiotensinogen-System als ursächlichen Faktor mit gewissen Formen der Hypertonie in Zusammenhang gebracht.
- Ein Mittel, die ungünstigen Folgen der Wirkungsweise des Renin-Angiotensinogen-Systems zu mildern, besteht in der Verabreichung einer Substanz, die die Angiotensinogen-Spaltwirkung von Renin hemmen kann.
- Eine Reihe derartiger Substanzen sind bekannt, einschließlich Antirenin-antikörper, Pepstatin und natürlich vorkommende Phospholipid-Verbindungen. Die Europäische Patentanmeldung Nr. 77 028 offenbart eine Reihe von Renin-hemmenden Polypeptidverbindungen, die ein nicht-terminales Statin (Sta; 4-Amino-3-hydroxy-6-methylheptansäure) oder Statin-Derivat aufweisen. Unter Einschluß von Sta enthält die große Mehrheit von exemplifizierten Verbindungen 6 oder mehr Aminosäurereste. Beispiele der wenigen dort offenbarten kürzesten Ketten sind:
- Acetyl-Phe-His-Sta-Leu-Phe-NH&sub2;, und
- t-Butyloxycarbonyl-Phe-His-Sta-Leu-Phe-NH&sub2;.
- Es gibt konstant mindestens zwei Aminosäurereste auf jeder Seite von Statin. Die Di- oder Polypeptidyl-statyl-Gruppe ist konstant mit einer lipophilen Aminosäure verbunden, am häufigstigen Leucin. (Siehe auch US-Patente 4 470 971 und 4 478 826).
- Die Europäische Patentanmeldung Nr. 45 665 und das US-Patent 4 424 207 offenbaren eine Reihe von Renin-hemmenden Polypeptid-Derivaten der Formel
- worin A zum Beispiel t-Butoxycarbonyl, B His oder eine andere basische Aminoacylgruppe, D Val, Ile oder ein anderer lipophiler Aminoacylrest, E Tyr, Phe, His oder ein anderer aromatischer Aminoacylrest, Ra und Rb je Isopropyl, Isobutyl, Benzyl oder eine andere Seitenkette vom Typus einer lipophilen Aminosäure und Ya eine terminale Gruppe vom Säure-, Ester- oder Amidtypus bedeuten. Unter Einschluß der zentralen 5-Aminopentansäurereste sind diese Verbindungen in jedem Fall Heptapeptide, d. h. N-Tetrapeptidyl-5-aminopentanoyl-lipophile- aminoacyl-aromatische-aminosäure-Derivate.
- Der Artikel "Pept. Struct. Funct.", Proc. 9th Am. Pept. Symp. 4615-4625 (1985) beschreibt eine stereogesteuerte 6stufige Synthese von Hydroxyethylendipeptid-Isosteren aus chiralen Aminoalkylepoxiden. J. Org. Chem. 45 28-29 (1980) und J. Med. Chem. 26 1457-1462 (1983) beschreiben die Zugabe organischer Lithiumverbindungen zu Aldehyden zur Bildung racemischer Mischungen von Alkoholen.
- Bestimmte Verbindungen besitzen außergewöhnlichen Wert als renininhibieruende Mittel. Diese Verbindungen sind Peptidderivate der Formel
- oder ein pharmazeutisch verwendbares Salz davon, worin bedeuten:

- Die vorliegende Erfindung betrifft die folgenden Verbindungen (III) und (IV), die zur Herstellung der obengenannten Zwischenverbindungen verwendbar sind:

- Die Erfindung betrifft auch ein stereoselektives Verfahren zur Herstellung der Verbindung der Formel (III), das die Umsetzung eines Aldehyds der Formel
- mit LiC CCOOR¹&sup5;, wobei R¹&sup5; (C&sub1;-C&sub3;)Alkyl ist, in einem reaktionsinerten Lösungsmittel bei -50ºC bis -80ºC zur Bildung einer Verbindung der Formel
- umfaßt.
- Diese Verbindung kann zur Bildung der Verbindung der Formel (III) hydriert werden, die dann zur Bildung einer Verbindung der Formel (IV) cyclisiert werden kann.
- Ferner betrifft die Erfindung ein stereoselektives Verfahren zur Herstellung einer Verbindung der Formel:
- worin R¹ wie oben definiert ist und R² (C&sub1;-C&sub6;)(2-Alkenyl), (C&sub4;-C&sub7;)(Cyclo-2-alkenyl), Benzyl oder Naphthyl oder eine dieser Gruppen bedeutet, die an einem aromatischen Ring mit den gleichen oder verschiedenen Substituenten, die (C&sub1;-C&sub3;)Alkyl, (C&sub1;-C&sub3;)Alkoxy, Fluor oder Chlor sind, mono- oder disubstituiert sein kann, das die Umsetzung eines Lactons der obigen Formel (IV) mit im wesentlichen einem Mol-Äquivalent eines organischen Halogenids der Formel R²X, worin X Cl, Br oder I ist, in Gegenwart einer starken Base geringer Nukleophilität umfaßt. R² ist vorzugsweise eine aktivierende Gruppe vom Allyl- oder Benzyl-Typus. X ist vorzugsweise Br.
- Die obengenannten Reaktionen werden wie folgt veranschaulicht:
- Die geschützte L-Aminosäure, von der ausgegangen wird, wird kommerziell oder durch auf dem Fachgebiet gut bekannte Standardmethoden erhalten, erwünschtenfalls unter Einschluß der Reduktion des aromatischen Rings, z. B. diejenige von N-(t-Butoxycarbonyl)phenylalanin-methylester, wie unten exemplifiziert. Der niedere Alkylester, vorzugsweise der Methylester wie gezeigt, wird leicht zum Aldehyd reduziert, zum Beispiel mit Diisobutylaluminiumhydrid in Toluol bei -50 bis -80ºC. Der Aldehyd seinerseits wird mit LiC CCOOR¹&sup5; (üblicherweise ist R¹&sup5; Äthyl, gebildet in situ aus Äthylpropiolat) umgesetzt, abermals bei -50 bis -80ºC in einem reaktionsinerten Lösungsmittel, wie Tetrahydrofuran. Im allgemeinen wird in dieser Stufe ein Stereoisomerenpaar gebildet, wobei das gewünschte Diastereoisomere (C) stark überwiegt. Das geringer vorhandene, unerwünschte Isomere wird vorzugsweise entfernt, worauf die Dreifachbindung hydriert wird (durchgeführt unter Standard-Hydrierbedingungen, z. B. über einem Palladiumkatalysator, vorzugsweise Pd in BaSO&sub4;, unter relativ milden Bedingungen) und Bildung des Lactons (z. B. durch Umsetzen in Toluol in Gegenwart von Essigsäure).
- Das gewünschte Lacton-Epimere, das die in Formel (E) und (F) gezeigte 4S-Stereochemie aufweist, wird dann mit einem Halogenid R²X (X=Cl, Br oder J; vorzugsweise X=Br) in Gegenwart eines wesentlichen Überschusses, z. B. 2 bis 2,5 Moläquivalente, einer starken Base mit niedriger Nucleophilität, wie LiN[CH(CH&sub3;)]&sub2; oder vorzugsweise Lithium-hexamethyldisilazid, kondensiert. Vorzugsweise ist das Halogenid eine solches vom Allyl- oder Benzyltypus (z. B. 2-Methyl-2-propenyl-bromid, Benzylbromid), wobei die Doppelbindung oder der aromatische Ring anschließend hydriert wird, falls die gesättigte Gruppe R² gewünscht wird, z. B.
- Abermals überwiegt das gewünschte Diastereoisomere, das die gezeigte trans (2R)-Stereochemie aufweist. Es wird durch Chromatographie abgetrennt (entweder vor oder nach irgendeiner zur Herstellung der gewünschten Gruppe R² erforderlichen Hydrierung). Alternativ kann die Hydrierung, insbesondere dann, wenn R² in der Stufe der Kondensation eine Benzylgruppe ist, auf später in der Gesamtsynthese der gewünschten Verbindung hinausgeschoben werden. Die bevorzugte Katalysatoren für die Hydrierung eines einfachen Olefins umfassen Pd oder Rh, während für die Reduktion von Phenyl zu Cyclohexyl Rh bevorzugt ist.
- Alle hier gezeigten Strukturangaben der Stereochemie stellen die absolute Stereochemie dar. Die vorliegende Erfindung wird durch die folgenden Beispiele veranschaulicht. Selbstverständlich ist die Erfindung jedoch nicht auf die Einzelheiten dieser Beispiele beschränkt. Alle Temperaturen sind in ºC angegeben und sind, falls nicht anders ausgesagt, Umgebungstemperaturen. Jedes Strippen von Lösungsmitteln erfolgte im Vakuum. Alle Standardlösungen waren, falls nicht anders angegeben, in Wasser. THF bedeutet Tetrahydrofuran.
- Methyl-S-3-Cyclohexyl-2-(t-butoxycarbonylamino)-propionat (51,2 g, 0,179 Mol) wurde in 728 ml trockenem Toluol gelöst und auf -78ºC gekühlt. Diisobutylaluminiumhydrid (449 ml von 1M in Toluol, 0,449 Mol) wurde tropfenweise im Verlaufe einer Stunde zugesetzt, wobei -70º bis -78ºC aufrechterhalten wurde. Methanol (13 ml) wurde bei -70ºC zugesetzt, gefolgt von 608 ml halbgesättigter Natrium-Kaliumtartrat-Lösung, und die Mischung wurde auf Raumtemperatur erwärmt. Äther (300 ml) wurde zugesetzt und die organische Schicht abgetrennt und mit 1 l gesättigter Natrium-Kalium-tartrat-Lösung gewaschen. Die ursprüngliche wäßrige Schicht wurde mit 600 ml frischem Äther extrahiert und mit 600 ml frischer gesättigter Natrium-Kalium-tartrat-Lösung rückgewaschen. Die organischen Schichten wurden vereinigt, über MgSO&sub4; getrocknet und vom Lösungsmittel befreit, wodurch das Titelprodukt als ein Gummi erhalten wurde, verunreinigt mit Toluol auf Basis des ¹H-NMR, 45,6 g: DC Rf 0,45 (1 : 3 Äthylacetat : Hexan): ¹H-NMR (CDCl&sub3;) delta: 0,9 bis 2,3 (m), welches das t-Butyl-Singulett bei 1,4 einschließt, 3,0-4,8 (m), 4,9-5,2 (d), 9,6 (s).
- Trockenes, frisch destilliertes THF (117 ml) und Diisopropylamin (22,0 ml, 15,8 g, 0,156 Mol) wurden unter Stickstoff in einen mit der Flamme getrockneten Reaktionskolben gebracht und die erhaltene Lösung wurde auf -30º abgekühlt und im Verlaufe von 5 Minuten mit Butyllithium (76,9 ml von 1,6M in Hexan, 0,123 Mol) versetzt. Die Lösung wurde dann auf -78ºC gekühlt und im Verlaufe von 20 Minuten tropfenweise mit Äthylpropiolat (12,5 ml, 12,1 g, 0,123 Mol) versetzt, wobei eine Temperatur von -65º bis -78ºC aufrechterhalten wurde. Nach 30 Minuten bei -78º wurde im Verlaufe von 20 Minuten die Titelverbindung des vorhergehenden Beispiels (19,52 g, 0,0866 Mol) in 35 ml THF zugesetzt, wobei abermals -56º bis -78º aufrechterhalten wurde. Nach 2 Stunden wurden 200 ml 5 : 1 THF : Essigsäure zur Reaktionsmischung zugesetzt. Diese konnte sich auf Raumtemperatur erwärmen und wurde mit einem halben Volumen Äther und einem gleichen Volumen 10%iger Zitronensäure verdünnt.
- Die organische Schicht wurde abgetrennt, der Reihe nach mit 2·200 ml frischer 10%iger Zitronensäure, 200 ml gesättigter Kochsalzlösung und 2·200 ml gesättigter NaHCO&sub3; gewaschen, über MgSO&sub4; getrocknet und vom Lösungsmittel befreit, wodurch ein dunkelrotes Öl erhalten wurde, 38,2 g. Letzteres wurde an einer 10 cm · 42 cm Säule von Kieselgel mit DC-Überwachung und Elution mit 5 l 1 : 9 Äthylacetat : Hexan chromatographiert. Nach 1500 ml zur Entwicklung der Säule wurden 500 ml Fraktionen aufgefnagen. Die Fraktionen 29-37 wurden vereinigt und vom Lösungsmittel befreit, wodurch das Titelprodukt in Form eines Öls erhalten wurde, 15,3 g; DC Rf 0,44 (3 : 7 Äthylacetat : Hexan); ¹H-NMR (CDCl&sub3;) delta: 1,0-2,0 (m, 25H) umfassend das Singulett der t-Butylgruppe bei 1,5, 3,8-5 (m, 6H).
- Das Titelprodukt des vorhergehenden Beispiels (18,28 g) und 5% Pd/BaSO&sub4; (10,97 g) wurden mit 150 ml Äthylacetat vereinigt und 2 Stunden unter 4 Atmosphären Wasserstoffdruck hydriert. Der Katalysator wurde nach Filtration abgetrennt und das Filtrat vom Lösungsmittel befreit, wodurch das Zwischenprodukt Äthyl-4RS,5S-6-cyclohexyl-4-hydroxy-5-(t-butyloxyamino)hexanoat erhalten wurde, 19 g. Letzteres wurde in 250 ml 2,5%iger Essigsäure in Toluol aufgenommen, 2,5 bis 3 Stunden zum Rückfluß erhitzt, vom Lösungsmittel befreit und der Rückstand an einer 10 cm · 30 cm Säule von Kieselgel unter Überwachung durch DC und Elution mit 4 l 9 : 11 Äther : Hexan, 8 l 1 : 1 Äther : Hexan, 2 l 11 : 9 Äther : Hexan und schließlich 3 l 3 : 2 Äther : Hexan chromatographiert, wobei 28·400 ml Fraktionen, 6·150 ml Fraktionen und schließlich 11·400 ml Fraktionen gesammelt wurden. Die Fraktionen 17-24 wurden vereinigt und zusammen mit Äther vom Lösungsmittel befreit, wodurch das vorherrschende und gewünschte, weniger polare 4S-5S-Titelprodukt in Form eines Öls erhalten wurde, 9,13 g; DC Rf 0,5 (7 : 3 Äther : Hexan). Das mehr polare 4R,5S-Epimere des Titelprodukts wurde durch Entfernung des Lösungsmittels von den kombinierten Fraktionen 28-45 isoliert und kristallisierte durch Verreiben mit Hexan. 1,77 g; Schmp. 101,5-103,5º.
- Trockenes frisch destilliertes THF (30 ml) und Diisopropylamin (3,51 ml, 2,52 g, 0,0249 Mol) wurden unter Stickstoff in einen mit der Flamme getrockneten Reaktionskolben gebracht. Die erhaltene Lösung wurde auf -50º gekühlt und mit Butyllithium (13,9 ml von 1,6M in hexan, 0,0222 Mol) versetzt, und die Mischung wurde weiter auf -78º gekühlt. Das Titelprodukt des vorhergehenden Beispiels (2,77 g, 0,0089 Mol) in 15 ml THF wurde im Verlaufe von 10 Minuten tropfenweise zugesetzt. Während weiteren 20 Minuten bei -78º konnte sich das Enolat bilden, zu welchem Zeitpunkt 3-Brom-2-methyl-1-propen in 5 ml THF im Verlaufe von 10 Minuten zugesetzt wurde. Die Mischung wurde eine weitere Stunde bei -78º gerührt, mit 5 ml gesättigter NH&sub4;Cl abgeschreckt, auf Raumtemperatur erwärmt, mit einem halben Volumen Äther verdünnt, mit 2·50 ml 10%iger Zitronensäure, 2·50 ml gesättigter NaHCO&sub3; und 1·25 ml gesättigter Kochsalzlösung gewaschen, über MgSO&sub4; getrocknet und unter Bildung eines Öls vom Lösungsmittel befreit, 3,06 g, eine Mischung der Titelepimeren. Letztere wurden durch Chromatographie an 7 cm · 20 cm Kieselgel getrennt: Überwachung durch DC; Elution der Reihe nach mit 2 l 1 : 9 Äther : Hexan, 4 l 3 : 17 Äther : Hexan, 2 l 1 : 4 Äther : Hexan, 2 l 1 : 3 Äther : Hexan; Auffangen von 125 ml Fraktionen. Das weniger polare Titelprodukt mit der trans (2R)-Stereochemie wurde in den Fraktionen 30-48 gesammelt, vereinigt und vom Lösungsmittel befreit, wodurch dieses als Öl erhalten wurde, 1,17 g; DC Rf 0,45, (2 : 3 Äther : Hexan): ¹H-NMR (CDCl&sub3;) delta: 1,4 (s, 9H), 1,8 (s, 3H), 0,3-3,0 (m, 18H), 3,6-4,0 (m, 1H), 4,69 (s, 1H), 4,1-4,8 (m, 2H).
- Die Fraktionen 55-76 lieferten das mehr polare Titelprodukt auch in Form eines Öls, 0,358 g, mit cis (2S)-Stereochemie; DC Rf 0,36 (2 : 3 Äther : Hexan): ¹H-NMR identisch mit demjenigen des weniger polaren Epimeren.
- Das weniger polare Titelprodukt des vorhergehenden Beispiels (1,17 g) und 10% Pd/C (0,351 g) wurden in 20 ml Äthylacetat vereinigt und während 2,5 Stunden bei 4 Atmosphären Druck hydriert. Der Katalysator wurde durch Filtration abgetrennt und das Filtrat vom Lösungsmittel befreit, wodurch das weniger polare Titelprodukt (ebenfalls mit trans, d. h. 2B-Stereochemie) in Form eines Öls erhalten wurde, das beim Stehen kristallisierte, 1,20 g; Schmp. 88-93º; DC Rf 0,65 (1 : 1 Äther : Hexan), Rf 0,73 (2 : 1 Äthylacetat : Hexan). Das andere Isomere mit cis (2S)-Stereochemie wurde in gleicher Weise erhalten; DC Rf 0,59 (1 : 1 Äther : Hexan). In folgenden Beispielen, die das vorliegende weniger polare Epimere mit Rf 0,65 (1 : 1 Äther : Hexan) verwenden, ist die 2R-Stereochemie angegeben.
- Trockenes Diisopropylamin (2,48 ml, 0,0177 Mol; destilliert über CaH&sub2;) in trockenem THF (7,4 ml, destilliert aus K) wurde unter Stickstoff auf 0º abgekühlt. Während 5 Minuten wurde tropfenweise Butyllithium (11,0 ml 1,62M in Hexan, 0,0177 Mol) zugesetzt. Nach dem Rühren während 15 Minuten bei 0º wurde die Mischung auf -78º abgekühlt und während 5 Minuten das weniger polare 4S,5S-Titelprodukt von Beispiel 3 (2,30 g, 0,0074 Mol) in 3,7 ml trockenem THF zugesetzt. Nach Rühren während weiteren 30 Minuten bei -78º wurde während 5 Minuten tropfenweise Benzylbromid (0,923 ml, 0,0078 Mol) in 3,7 ml trockenem THF zugesetzt. Nach 1 Stunde bei -78º wurde die Reaktion durch Zusatz von 10 ml gesättigter NH&sub4;Cl abgebrochen, auf Raumtemperatur erwärmt, mit 25 ml Äther verdünnt und die Schichten getrennt. Die organische Schicht wurde mit 2·15 ml gesättigter NaHCO&sub3; gewaschen, getrocknet (MgSO&sub4;) und zu einem Öl eingeengt (3,29 g). Das Öl wurde an 150 g Kieselgel unter Verwendung von 2,5 Liter 1 : 9 Äthylacetat : Hexan als Elutionsmittel und Überwachung durch DC chromatographiert. Die reinen Produktfraktionen wurden vereinigt und vom Lösungsmittel befreit, wodurch das gereinigte Titelproukt in Form eines weißen öligen Feststoffs erhalten wurde, 1,46 g; DC Rf 0,60 (1 : 1 Äthylacetat : Hexan). Das mehr polare Ausgangsmaterial (0,91 g gelbes Öl, etwa 70% rein, DC Rf 0,4 im gleichen System) wurde ebenfalls aus der Säule gewonnen.
- N-(t-Butoxycarbonyl)leucin-methylester (28,0 g 0,114 Mol) wurde nach der Methode des Beispiels 1 in das vorliegende Titelprodukt übergeführt, 21,7 g (88%), in Form eines schwach gelben Öls; DC RF 0,36 (2 : 3 Äthylacetat : Hexan); ¹H-NMR (CDCl&sub3;) delta (90 MHz) 0,97 (d, J=6, 6H), 1,1-1,8 (m, 3H), 1,4 (s, 9H), 3,3-5,0 (m, 2H), 9,53 (s, 1H).
- Das Produkt des vorhergehenden Beispiels (0,4 g, 0,048 mMol) wurde nach der Methode des Beispiels 2 in das vorliegende Titelprodukt umgewandelt, 5,45 g; DC Rf 0,40 (3 : 7 Äthylacetat : Hexan); IR (CHCl&sub3;), 3438, 3340, 2233, 1711 cm&supmin;¹; ¹H-NMR (CDCl&sub3;) delta (300 MHz) 0,97 (t, J=7, 6H), 1,34 (t, J=6, 3H), 1,48 (s, 9H), 1,48 (m, 2H), 1,70 (m, 1H), 3,3-3,4 (m, 1H), 3,81-3,96 (m, 1H), 4,28 (q, J=7, 2H), 4,45-4,58 (m, 1H), 4,68-4,78 (m, 1H).

- Das Produkt des vorhergehenden Beispiels wurde nach der Methode des Beispiels 3, ausgenommen die Anwendung von Gradientelution mit 2 : 3 bis 1 : 0 Äthylacetat : Hexan bei der Chromatographie, in das gewünschte weniger polare (4S,5S)-Lacton-Produkt umgewandelt und durch Verreiben mit Hexan kristallisiert, 3,10 g (78%); Schmp. 76-77º; IR (CHCl&sub3;) 3439, 1775, 1711 cm&supmin;¹; ¹H-NMR (CDCl&sub3;) delta (300 MHz) 0,92 (d, J=6, 6H), 1,44 (s, 9H), 1,28-1,81 (m, 3H), 2,06-2,32 (m, 2H), 2,48-2,58 (m, 2H), 3,79-3,92 (breites s, 1H), 4,42-4,58 (m, 2H).

- Das mehr polare (4R,5S)-Lacton wurde auch bei der Chromatographie in wesentlich niedrigerer Ausbeute isoliert und durch Verreiben in Hexan kristallisiert, 0,68 g; Schmp. 113,5-116ºC.
- Das Titelprodukt des vorhergehenden Beispiels (das weniger polare 4S,5S-Epimer; 0,51 g, 0,0019 Mol) wurde nach der Methode des Beispiels 4 in das vorliegende Titelprodukt (zum größeren Teil), zusammen mit dessen mehr polaren 2S,4S,5S-Epimer, umgewandelt. Die Rohprodukte (0,60 g) wurden an einer 4,5·20 cm Säule von Kieselgel unter Elution mit je 1 l 1 : 9, 3 : 17, 1 : 5, 1 : 4, 3 : 7 und 1 : 1 Äther : Hexan und Auffangen von 23 ml Fraktionen chromatographiert. Die Fraktionen 51-85 lieferten das gereinigte Titelprodukt, 0,21 g; Schmp. 128-132º.
- [alpha]30/D -23,7º (c=0,529, CH&sub3;OH)
- Das mehr polare 2S,4S,5S-Epimere wurde aus den Fraktionen 89-120 isoliert, 105 mg; ¹H-NMR (CDCl&sub3;) enthält delta 4,8 (2s, 2H, Vinyl-Protonen) und 1,75 (s, 3H, Vinylmethyl): ein Teil dieses Epimers wurde durch langsames Eindampfen aus CH&sub2;Cl&sub2; : Hexan kristallisiert, was nadelförmige Kristalle vom Schmp. 99-101º lieferte.
- Zu einer Suspension von Lithium-hexamethyldisilazid bei -78ºC, hergestellt durch tropfenweisen Zusatz von 5,1 ml (8,11 mMol) einer 1,6M Lösung von n-Butyllithium in Hexan zu 1,79 ml (1,39 g, 8,49 mMol) Hexamethyldisilazan in 3,5 ml THF bei 0º, wurde tropfenweise eine Lösung von 1,00 g (3,69 mMol) 4S,5S-Lacton des vorhergehenden Beispiels in 3 ml THF zugesetzt. Am Ende des Zusatzes wurde die Lösung klar und wurde weitere 15 Minuten bei -78ºC rühren gelassen. Eine Lösung von 0,548 g (4,06 mMol) frisch destilliertem Methallylbromid in 2 ml THF wurde dann im Verlauf von 5 Minuten tropfenweise zugesetzt. Die Mischung konnte sich während 2 Stunden langsam auf -40ºC erwärmen, bevor sie mit 2 ml gesättigter NH&sub4;Cl abgeschreckt wurde. Nach dem Erwärmen auf Raumtemperatur wurde die Reaktionsmischung zwischen 30 ml Äther und 30 ml 10%iger Zitronensäure verteilt. Die organische Schicht wurde abgetrennt und mit 10%iger Zitronensäure (3·30 ml) und gesättigter NaHCO&sub3; gewaschen, getrocknet (MgSO&sub4;) und zu 1,11 g einer rohen Mischung von cis(2S) und trans(2R) eingedampft. Diese Lactone wurden an 88 g Kieselgel mit Äther-Hexan (1 : 9 bis 3 : 7) als Elutionsmittel getrennt. Die Fraktionen, die das weniger polare trans(2S)-Lacton [DC Rf 0,55 (1 : 1 Äther : Hexan)] enthielten, wurden vereinigt und zu 0,613 g (51%) eines weißen Feststoffs eingedampft, Schmp. 132-135ºC. Geringe Verunreinigungen (wie das DC zeigte) wurden durch Verreiben in Hexan entfernt, wodurch 0,562 g (47%) des analytisch reinen Titel- 2R,4S,5S-Lactons erhalten wurde, Schmp. 33-135ºC. Für die Röntgenanalyse geeignete Kristalle wurden durch langsames Eindampfen aus Hexan-Methylenchlorid erhalten. ¹H-NMR (CDCl&sub3;) delta (250 MHz) 0,90 (J, J=6, 3H), 0,92 (d, J=6, 3H), 1,42 (s, 9H), 1,70 (s, 3H), 1,92-2,15 (m, 2H), 2,26-2,39 (m, 1H), 2,57 (dd, J=15 und 3, 1H), 2,72-2,88 (m, 1H), 3,77-3,90 (m, 1H), 4,34 (d, J=9, 1H), 4,33-4,51 (m, 1H), 4,70 (s, 1H), 4,81 (s, 1H); ¹³C-NMR (75 MHz) delta 21,8, 23,0, 24,7, 28,3, 30,0, 37,9, 39,5, 41,8, 51,7, 79,8, 80,7, 112,8, 141,9, 156,0, 179,3; IR (CHCl&sub3;) 3439, 1768, 1712, 1654 cm&supmin;¹; [alpha] 30/D-25,0º (C=0,5, CH&sub3;OH). Anal. ber. für C&sub1;&sub8;H&sub3;&sub1;NO&sub4;: C, 66,43; H, 9,60; N, 4,30. Gef. C, 66,47; H, 9,59; N, 4,27. Einkristall-Röntgenanalysen bestätigten die Richtigkeit der Struktur und die stereochemische Zuordnung für diese Verbindungen.
- Die Fraktionen, die das mehr polare cis(2S)-Lacton (DC Rf 0,44 1 : 11 Äther : Hexan) enthielten, wurden vereinigt und zu 39 mg (3%) eines weißen Feststoffs eingedampft, Schmp. 96-98ºC: ¹H-NMR (CDCl&sub3;) delta (250 MHz) 0,92 (d, J=6, 6H), 1,43 (s, 9H), 1,72 (s, 3H), 2,02-2,14 (m, 1H), 2,23-2,36 (m, 1H), 2,60-2,87 (m, 2H), 3,74-3,89 (m, 1H), 4,35-4,47 (m, 2H), 4,69 (s, 1H), 4,78 (s, 1H): ¹³C-NMR (75 MHz) delta 21,9, 22,0, 23,0, 24,8, 28,3, 30,7, 38,8, 38,9, 42,3, 50,1, 79,6, 80,4, 112,6, 142,0, 155,9, 178,7; IR (CHCl&sub3;) 3443, 1774, 1714, 1656 cm&supmin;¹; [alpha]30/D-0,6º (C=0,5, CH&sub3;OH). Anal. ber. für C&sub1;&sub8;H&sub3;&sub1;NO&sub4;: C, 66,43; H, 9,60; N, 4,30; Gef. C, 66,94; H, 9,45; N, 4,27.
- Eine Lösung von 438 mg (1,35 mMol) des Titellactons des vorhergehenden Beispiels in 10 ml Äthylacetat, enthaltend 44 mg 10%iges Pd/C, wurde 2 Stunden auf einer Parr Shaker-Apparatur bei 50 psi hydriert. Nach dem Abfiltrieren des Katalysators und Abdampfen des Lösungsmittels wurden 437 mg (99%) des vorliegenden Titelprodukts in Form eines weißen Feststoffs erhalten, Schmp. 130-131ºC. ¹H-NMR (CDCl&sub3;) delta (300 MHz) 0,84-0,97 (m, 12H), 1,41 (s, 9H), 1,86-1,96 (m, 1H), 2,30-2,42 (m, 1H), 2,56-2,68 (m, 1H), 3,76-3,89 (m, 1H), 4,35 (d, J=8, 1H), 4,45 (breites t, 1H); ¹³C-NMR (75 MHz) delta 21,3, 21,8, 22,9, 23,0, 24,8, 26,1, 28,3, 31,0, 37,7, 40,5, 41,9, 51,7, 79,8, 80,5, 156,0, 180,3; IR (CHCl&sub3;) 3439, 1769, 1713 cm&supmin;¹; [alpha]30/D-32,1º (C=1,0, CH&sub3;OH). Anal. ber. für C&sub1;&sub8;H&sub3;&sub3;NO&sub4;: C, 66,02; H, 10,16; N, 4,28. Gef. C, 66,07; H, 10,03; N, 4,05.
- Das weniger polare 4S-5S-Produkt des Beispiels 3 (2,5 g, 0,008 Mol) und 4-Chlorbenzylbromid (1,81 g, 0,0088 Mol) wurden nach der Methode des Beispiels 6 in das vorliegende chromatographierte Titelprodukt in Form eines farblosen gummiartigen Produkts umgewandelt, 1,91 g; DC Rf 0,6 (3 : 1) Hexan : Äthylacetat) 0,7 (2 : 1 Hexan : Äthylacetat mit 1% Essigsäure).
- Die Methode des Beispiels 6 unter Verwendung von 5 : 1 Hexan : Äthylacetat als Elutionsmittel bei der Chromatographie wurde verwendet, um das 4S,5S-Produkt des Beispiels 3 (1,5 g, 0,0048 Mol) und alpha-Brom-p-xylol (0,98 g, 0,0053 Mol) in das vorliegende Titelprodukt in Form eines weißen gummiartigen Produkts umzuwandeln, 0,985 g; DC Rf 0,5 (2 : 1 Hexan : Äthylacetat), 0,75 (1 : 1 Hexan : Äthylacetat).
- Das 4S,5S-Produkt des Beispiels 8 (3,79 g, 0,018 Mol) und p-Methoxybenzylbromid (2,61 g, 0,130 Mol) wurden nach der Methode des Beispiels 13 in das chromatographierte Titelprodukt in Form eines weißen gummiartigen Produkts umgewandelt, 1,06 g; DC Rf 0,4 (3 : 1 Hexan : Äthylacetat).
- Das 4S,5S-Produkt des Beispiels 3 (2,0 g, 0,0064 Mol) und 3,5-Dichlorbenzylbromid (1,68 g, 0,007 Mol) wurden nach der Methode des Beispiels 6 unter Verwendung von 3 : 1 Hexan : Äthylacetat als Elutionsmittel bei der Chromatographie in das Titelprodukt in Form eines klaren gummiartigen Produkts umgewandelt, 1,36 g; DC Rf 0,22 (3 : 1 Hexan : Äthylacetat), 0,9 (1 : 2 Hexan : Äthylacetat mit 1% Essigsäure).
- Das 4S,5S-Produkt des Beispiels 3 (2,0 g, 0,0064 Mol) und o-Chlorbenzylchlorid (1,44 g, 0,007 Mol) w urden nach der Methode des Beispiels 6 unter Verwendung von 6 : 1 Hexan : Äthylacetat als Elutionsmittel bei der Chromatographie in das Titelprodukt in Form eines klaren gummiartigen Produkts umgewandelt, 1,51 g; DC Rf 0,75 (3 : 1 Hexan : Äthylacetat), 0,65 (2 : 1 Hexan : Äthylacetat mit 1% Essigsäure).
- Das 4S,5S-Produkt des Beispiels 3 (2,0 g, 0,0064 Mol) und m-Chlorbenzylchlorid (0,92 ml, 1,44 g, 0,007 Mol) wurden nach der Methode des vorhergehenden Beispiels in das chromatographierte Titelprodukt in Form eines farblosen gummiartigen Produkts übergeführt, 2,11 g; DC Rf 0,4 (3 : 1 Hexan : Äthylacetat), 0,75 (2 : 1 Hexan : Äthylacetat mit 1% Essigsäure).
Claims (4)
1. Stereoselektives Verfahren, das umfaßt die Umsetzung
eines Aldehyds der Formel
worin R¹ (C&sub1;-C&sub6;)Alkyl, (C&sub1;-C&sub6;)Alkenyl, Phenyl, Naphthyl, (C&sub4;-
C&sub7;)Cycloalkyl, (C&sub4;-C&sub7;)Cycloalkenyl, (C&sub7;-C&sub9;)Phenylalkyl, (C&sub1;&sub1;-
C&sub1;&sub3;)Naphthylalkyl, (C&sub5;-C&sub1;&sub0;)(Cycloalkyl)alkyl oder eine dieser
Gruppen bedeutet, die am aromatischen Ring mit den gleichen
oder unterschiedlichen Gruppen, ausgewählt aus (C&sub1;-C&sub3;)Alkyl,
(C&sub1;-C&sub3;)Alkoxy, Fluor oder Chlor, mono- oder disubstituiert
sein kann,
mit LiC CCOOR¹&sup5;,
worin R¹&sup5; (C&sub1;-C&sub3;)Alkyl ist,
in einem reaktionsinerten Lösungsmittel bei -50 bis -80ºC zur
vorherrschenden Bildung einer Verbindung der Formel
2. Verfahren nach Anspruch 1, das ferner die Schritte umfaßt:
(a) Hydrierung der Verbindung der Formel (C) zur Bildung
einer Verbindung der Formel
und (b) Cyclisation der Verbindung der Formel (D) zur Bildung
einer Verbindung der Formel
3. Verbindung der Formel
oder
worin R¹&sup5; (C&sub1;-C&sub3;)Alkyl ist und
R¹ (C&sub1;-C&sub6;)Alkyl, (C&sub1;-C&sub6;)Alkenyl, Phenyl, Naphthyl, (C&sub4;-
C&sub7;)Cycloalkyl, (C&sub4;-C&sub7;)Cycloalkenyl, (C&sub7;-C&sub9;)Phenylalkyl, (C&sub1;&sub1;-
C&sub1;&sub3;)Naphthylalkyl, (C&sub5;-C&sub1;&sub0;)(Cycloalkyl)alkyl oder eine dieser
Gruppen bedeutet, die am aromatischen Ring mit den gleichen
oder unterschiedlichen Gruppen, ausgewählt aus (C&sub1;-C&sub3;)Alkyl,
(C&sub1;-C&sub3;)Alkoxy, Fluor oder Chlor, mono- oder disubstituiert
sein kann.
4. Stereoselektives Verfahren zur Herstellung einer Verbindung
der Formel.
worin R¹ (C&sub1;-C&sub6;)Alkyl, (C&sub1;-C&sub6;)Alkenyl, Phenyl, Naphthyl, (C&sub4;-
C&sub7;)Cycloalkyl, (C&sub4;-C&sub7;)Cycloalkenyl, (C&sub7;-C&sub9;)Phenylalkyl, (C&sub1;&sub1;-
C&sub1;&sub3;)Naphthylalkyl, (C&sub5;-C&sub1;&sub0;)(Cycloalkyl)alkyl oder eine dieser
Gruppen bedeutet, die am aromatischen Ring mit den gleichen
oder unterschiedlichen Gruppen, ausgewählt aus (C&sub1;-C&sub3;)Alkyl,
(C&sub1;-C&sub3;)Alkoxy, Fluor oder Chlor, mono- oder disubstituiert
sein kann, und R² (C&sub1;-C&sub6;)(2-Alkenyl), (C&sub4;-C&sub7;)(Cyclo-2-alkenyl),Benzyl
oder Naphthylmethyl oder eine dieser Gruppen bedeutet,
die an einem aromatischen Ring mit den gleichen oder
unterschiedlichen Substituenten, die (C&sub1;-C&sub3;)Alkyl, (C&sub1;-C&sub3;)Alkoxy,
Fluor oder Chlor sind, mono- oder disubstituiert sein
kann,
das umfaßt die Umsetzung eines Lactons der Formel
mit im wesentlichen einem Mol-Äquivalent eines organischen
Halogenids der Formel
R²X,
worin X Chlor, Brom oder Jod ist, in Gegenwart eines erheblichen
Überschusses einer starken Base geringer Nukleophilität.
