Description
- Beschreibung
- Es besteht ein ständiges Interesse der Riechstoff-Industrie an neuartigen Riech- und Aromastoffen, deren olfaktorische oder gustatorische Eigenschaften die bisher bekannten und verwendeten Substanzen übertreffen bzw. in -Kompositionen wirkungsvoll ergänzen können. Der erste Schritt zu derartigen neuen Wirksubstanzen ist häufig die Analyse der natürlich vorkommenden Geruchskomplexe (ätherische Öle, Extrakte), die neben den bisher bekannten Hauptkomponenten, die oft den geruchlichen Grundakkord liefern, auch Substanzen geringer Konzentration enthalten.
- Derartige Neben- oder Spurenkomponenten sind häufig ohne Bedeutung für den geruchlichen Gesamteindruck-des Naturstoff-Gemisches. In bestimmten Fällen sind sie ab-er geeignet, den geruchlichen Gesarnteindruck erheblich zu beeinflussen. Solche Neben- oder Spurenkomponenten können in isoliertem Zustand ganz andere½Geruchseindrücke vermitteln, als sie dem ätherischen Öl oder Pflanzenextrakt zu eigen sind, dem sie entstammen. In besonderen Fällen können diese neuartigen Substanzen, die aus Naturstoffgemischen isoliert bzw. durch Synthese zugänglich gemacht worden sind, aufgrund ihrer individuellen Geruchseigenschaften äußerst wirkungsvoll in Parfümölen (Riechstoffmischungen) eingesetzt werden, deren Geruchseindruck grundlegend verschieden ist von dem des etherischen ÖLes oder Pflanzenextrakts: in dem die Substanz als Spurenkomponente vorkommt. Durch Zugabe der isolierten (oder synthetisierten) Substanz zu. einem Parfünöl kann ein ganz anderer Geruchseffekt erzielt werden, als dies durch Zugabe des ätherischen Öls oder Extrakts möglich ist, dem die Substanz entstammt. Diese Sachverhalte sind bei der Patenterteilung für das Menthon-thiol gewürdigt worden[GRUR, 702 (1978)].
- Das ostindische Sandelholzöl wird in der Parfümerie aufgrund seiner charakteristischen, geruchlichen und fixiercnden Eigenschaften geschätzt. Die begrenzte Verfügbarkeit und der hohe Preis geben Anlaß zur Synthese von gut zugänglichen Substanzen mit gleichen oder ähnlichen Geruchseigensciiaften.
- Die seit langem bekannten Hauptinhaltsstoffe des ostindischen Sandelholzöls sind @@ Sesquiterpenalkohole α-Santalol (2) und ß-santalol (3) (Über @cht: E.-J. Brunke und W. Rojahn, DRAGOCO-Report, 127 (1980)@ ine erneute gaschromatographische Analyse dieses Öls zeigte eine Reihe VOn Nebenkomponenten, die isoliert, spektroskopisch charakterisiert und olfaktorisch beurteilt worden sind [Übersicht: E.-J. Brunke, Parfümerie und Kosmetik 62, 73 (1981)].
- (+)-α-Santalol (-)-ß-Santalol (Z) (+)-epi-ß-Santalol (-)-(E)-ß-Santalol In dem hier abgebildeten Teil des Gaschromatogramms von ostindischem Sandelholzöl sind die anschließend diskutierten Verbindungen zugeordnet. Eine Geruchsbestimmung anhand der isolierten Sesquiterpenalkohole hat aufgezeigt, daß die typischen holziganimalischen Geruchsaspekte des Sandelholzöls vom (Z)-ß-Santalol (3a) bewirkt werden, das eine erhebliche Geruclisintensität aufweist. (E) -ß-Santalol (3b) riecht wenig angenehm holzig-medizinisch. Das α-Santalol (2) besitzt einen lediglich mild-holzigen Duft, der an reines Cedren erinnert; animalische oder ausstrahlende Effekte fehlen. Das epi-ß-Santalol (?f) riecht lediglich schwach holzig [E.-J. Brunke, H. Struwe und F.-J. Hamtnerschmidt, Tetrahedron Letters, 2405 (1980). G. Ohloff, R.L.Snowden und P. Sonnay, Helv. Chim. Acta 64, 25 (1981)].
- Als weitere Komponente (4-8 %) des Sandelholzöls wurde von uns das (-)-(Z)-$α-trans-Bergamotol (1a) isoliert und seine Struktur nach einer ersten Fehlbestimmung [E.-J. ßrunke und F.-J. Hammerschmidt, Vertrag beim Congress on Essential Oils, Cannes Okt.
- 1980] nunmehr eindeutig bestimmt. la ist eine neue Substanz.
- Zum Zwecke des Strukturbeweises wurde aus dem Alkohol la durch
Braunstein-Oxidation der ungesättigte Aldehyd 1b dargestellt, der sich von dem bisher in der Natur gefundenen 12-(E)-Isomeren 5 [3. Maurer und A. Grieder, Helv. Chim. Acta 60, 2177 (1977)] spektroskopisch und geruchlich unterscheidet. Durch Huang-Minlon Reduktion wurde aus dem Aldehyd ib das bekannte (-)-cx-brans-Bergamoten (6) erhalten, dessen Identität durch gleiche IR-, 1H-NMR- und MS-Spektren bewiesen wurde [Daten von (-)-a-trans-Bergamoten: E. Kovats, Helv. Chim. Acta 46, 2705 (1963). B. Maurer und A. Grieder, Helv. Chim. Acta 60, 2177 (1977)]. Die Geruchseigenschaften des (-)-(Z)-$α-trans-Bergamotols (la) können mit "hell, etwas holzig und Argumen-artig, von erheblicher Ausstrahlung" bezeichnet werden. Eine derartige Geruchsnote weicht erheblich von dem Geruchsprofil des ostindischen Sandelholzöls ab.< iM nO2*+ OH CHO la12-(2)] (Z )] / Huang-Min lon- Red. rY 6 t-)-Z-trans-Bergamoten Huang-Minton-Red. EHO+ CHO 7 5[12-(E)] - Es war daher neuartig und überraschend, daß das Bergamotol la, wenn es reinem ß-Santalol (3) zugegeben wird, das ja die typische Sandelholznote repräsentiert, diese Sandelholznote erheblich verstärkt und eine starke Ausstrahlung bewirkt, ohne selbst geruchlich hervorzutreten. Es hat sich gezeigt, daß das Bergamotol la auch ohne gleichzeitigen Einsatz von ß-Santalol vorteil haft in Parfüm-Kompositionen einzuarbeiten ist. Es verleiht Kompositionen mit verschiedenen Geruchsprofilen bereits bei Konzentrationen von 0,1 % deutlich eine höhere Frische und Ausstrahlung. Das Bergamotol la kann aber auch in höheren Konzentrationen in Parfümöle eingearbeitet werden.
- Der von Maurer und Grieder [Helv. Chim. Acta 60, 2177 (1977)] aus 5 durch Reduktion erhaltene Alkohol 7 mit 12-(E)-Geometrie besitzt nicht diese bemerkenswerten Geruchseigenschaften des 12-(Z)-Isomeren la. Der hier zu beobachtende Geruchsunterschied geometrischer Isomerer ist vergleichbar mit dem für (Z)- und (E) ß-Santalol (3a) und (3b) beschriebenen.
- Der durch Oxidation von la erhaltene Aldehyd lb weist einen ausstrahlenden weichen Holzduft auf, der an Sandelholzöl erinnert.
- Die durch Oxidation von 1b erhaltene neue Carbonsäure 1c besitzt holzig-säuerliche Geruchsaspekte, während die daraus durch übliche Veresterung erhaltenen Derivate if und 1g weich-holzige, etwas süße Geruchsnoten mit großer Ausstrahlung besitzen. Die durch übliche Veresterung von pa mit Essigsäure oder Propionsäureanhydrid erhaltenen Verbindungen id und le riechen ebenf'alls süß-holzig. Aufgrund ihrer Geruchseigenschaften können die neuen Verbindungen la-g vorteilhaft als Riechstoffe oder Bestandteil von Riechstof'f-Mischungen verwendet werden.
- Die für die Verwendung von Parfümölen in. verschiedenen technischen Medien (Waschpulver, Wäscheweich, Shampoo, etc.) zu fordernde Produktstabilität kann durch selektives Hydrieren der Seitenkette der Verbindungen 1a-1g verbessert werden. Hierbei treten allerdings geringfügige Verschiebungen der Geruchsprofile ein. Die katalytische Hydrierung tRaney-Nickel) von lb ergab den einfach gesättigten Aldehyd 1i, der zu dem entsprechenden Alkohol lh reduziert wurde (NaBH4). Übliche Veresterung von 1h ergab das Acetat 1l und das Propionat Im. Selektive katalytische Hydrierung der Ester 1f und 1g führte zu 1n bzw. 1o, deren Verseifung (NaOH/EtOII) die Carbonsäure lk ergab. Die neuen Verbindungen 1h-1o besitzen interessante holzige Geruchsnoten, die jeweils an die entsprechende ungesättigte Verbindungen erinnert, jedoch etwas härter und kräftiger wirken. Bei 11 wird gegenüber lb eine aldehydische Grünnote betont, während 1k etwas harziger wirkt als le. Aufgrund dieser Geruchsnoten können die neuen Verbindungen lh-lo vorteilhaft als Riechstoffe eingesetzt werden.
- In dem Schweizer Patent Nr. 622'946 (Anmeldetag 22.8.1977) ist die Photooxidation des «-Bergamotens (6) beschrieben, die zu dem tertiären Alkohol 8 und dem sekundären. Alkohol 9 fWirt. Der Verbindung 8 wird eine anhaftende und elegante Geruchsnote zu- geschrieben, die an Vetyver oder Ceder erinnert soll. Der Duftcharakter des ebenfalls in dieser Schrift beschriebenen (E)-konfigurierten Aldehyds 5 wird als "holzartig, mit einem Sandelholzaspekt, sowie mit Cumin-artigen und animalischen Noten angegeben. Es war daher überraschend, daß der primäre Alkohol la eine reine, helle und ausstrahlende Holznote ohne krautige oder harzige Aspekte besitzt, wie sie 8 zugeschrieben worden sind.
- Die Synthese der Verbindungen der Formeln la-o erfolgte ausgehend von a-trans-Bergamoten (5), das durch fraktionierte Destillation aus Opoponax-Öl, Bergamott-Öl oder Olibanum-Öl optisch aktiv gewonnen werden kann oder durch Totalsynthese [E.-J. Corey, D.E. Cane und L. Libit, J. Xmer. Cheni. Soc., 93, 7016 (1971)] in racemischer Form'zugänglich ist. Durch Selendioxid -Oxidation unter üblichen Bedingungen wurde aus 6 der (E)-konfigurierte Aldehyd 5 erhalten [Schweizer Patent Nr. 622 9146].
- Aus Q läßt sich durch Umsetzen mit Kaliumcyanid und Braunstein in Analogie zu einer von Corey et al. gegebenen Vorschrift [J.
- Amer. Chem. Soc., 90, 5617 (1968)] der E-konfigurierte Methylester 1o darst'ellen und in Analogie zu Kretschmar und Erman trahedron Letters, 41 (1970)] durch Bestrahlung mit UV-Licht zum Z-konfigurierten Ester 1f (1f:10=70:30) isomerisieren. das durch Säulenchromatographie zu isolierende Isomere lf läßt sich mit Lithium-aluminiumhydrid auf übliche Waise zu a-trans-Bergamo;tol (la) reduzieren. Aus la sind die übrigen Derivate (lb-lo) auf den bereits beschriebenen Wegen zuganglicll.
- Beispiel 1 Darstellung von (E)-a-trans-Bergamotol (5) Eine Lösung von 50 g a-trans-Bergamoten (6,[α]D= -30°) und 30 g Selendioxid in 500 ml Ethanol (96%ig) wurde 12 Stdn. bei-Stedetemp. gerührt. Das nach Einengen verbleibende Rohprodukt (gelbes Öl), bestehend aus 5 und 2 (2:1), wurde in 300 m£ n-Hexan gelöst, mit 30 e Mangandioxid versetzt und 3 Stdn. bei Raumtemp.
- gerührt. Die Reaktionslösung wurde filtriert und eingeengt. man erhielt 51 g 5 als gelbes Öl. Durch Schichtchromatographie (Cyclohexan/Essigester, 3:1) wurde eine analytische Probe gereinigt.

- C15H22O (218.34) 15 22 Beispiel 2 Darstellung von (Z)-$α-trans-Bergamotsäure-methylester (1f) Eine Lösung von 15 g Aldehyd 2r (nach Beispiel 1) und 23 g Kaliumcyanid in 2.5 e dest. Methanol wurde mit 500 g Mn02 und 65 m2 Essigsäure versetzt und 14 Stdn. bei Raumtemp. gerührt. Nach Filtration wurde dio Reaktionslösung eingeengt, mit 100 ml Wasser versetzt und aufgearbeitet (Vorsicht, HCN!). Nach Einengen des Lösungsmittels verblieben 14.5 g des Methylesters 10 als gelbes Öl. Das Rohprodukt wurde in 250 ml Methanol aufgenommen und in einer Bestrahlungsapparatur bei 200C 4 Stdn. mit W-Licht bestrahlt. Das nach Einengen vorliegende Produktgemisch bestand aus dem Ester mit (Z)-Konfiguration 1f und dem (E)-Ester 8 (7:3). Durch fraktionierte Destillation wurde 1f (94%ig nach GC) erhalten. Kp.(O.l Torr)=1050C. Eine analytische Probe wurde durch Säulenchromatographie isoliert. [Geruch: holzig, süß]. NMR (CCl4):#=0.83, s (6-CH3), 1.65, "s" (2-CH3), 1.87, "s" (13-CH3, transoid), 3.70, s (-COOCH3), 5.33, m (3-H), 5.93 ppm, "t", J=7 Hz (12-H), MS:m/z(%)=248(2, M+), Z33(1), 216(4), 201(3), 189(5), 173(3), 161(5), 145(5), 135(10), 121(20), 119 (99), 114(38), 107(29), 105(21), 101(15), 93(100).
- O16H2402(248.36) Beispiel 3 Darstellung von (Z)-α-trans-Bergamotol (la) Eine Lösung von 10 g des (Z)-Methylesters 1f (nach Beispiel 2) in 200 ml trockenem Ether wurde unter Rühren zu einer Aufschlämmung von 8 g Lithium-aluminiumhydrid in 200 me trockenem Ether hinzugetropft. Nach 1 Stde. Rühren bei Siedetemp. wurden 20 me Ethylacetat zugegeben. Nach 10 min Rühren bei Raumtemp. gab man 50 g Na2SO4.10 H20 hinzu, filtrierte und engte ein. Es verblieben 7.9 g la als farbloses Öl. [Geruch: angenehm, ausstrahlend holzig, etwas wachsig]. NMR (CCl4):#= 0.83, s (6-OH3), 1.64, "s" (2-CH3), 1.74, "s" (13-CH3, transoid), 4.05, s (CH2-15), 5.15, m (3-H), 5.23 ppm, "t" (12-H). IR: 3320 cm (OH). Die Mischungen c und e zeichen sich gegenüber Mischung a durch eine deutliche Abrundung aus. Der Dufteindruck der Mischung d ist ausstrahlender und etwas holziger als die Mischung a.

Claims (4)
- Verwendung von α-trans-Bergamotol-Derivaten als Riech- und Aromastoffe Pa tentansprüche 1. Verwendung als Riech- und Aromastoffe der a-trans-Bergamotol-Derivate der allgemeinen Formel 1, wobei R eine Hydroxymethyl-, Formel, Carboxyl-, Acetoxymethyl-, Propoxymethyl-, Carbmethoxy- oder Carbethoxygruppe
und die gestrichelte Linie eine fakultative Doppelbindung mit (Z)-Geometrie bedeutet.R CH2OH CHO COOH CH2OAc CH2OPr COOMe COOEt (#12) a b c d e f g (12-,13-H) h i k l m n o - 2. Verwendung der a-trans-Bergamotol-Derivate der Formel 1 als Bestandteil von Parfüm- oder Aroma-Komposionen aus einheitlichen Riechstoffen natürlichen oder synthetischen Ursprungs.
- 3, Verwendung der α-trans-Bergamotol-Derivate der Formel 1 als Bestandteil von Parfüm- oder Aroma-Komposionen aus ätherischen Ölen und Extrakten.
- 4.. Verwendung der α-trans-Bergamotol-Derivate der Formel 1 als Bestandteil von Parfüm- oder Aroma-Kompositionen aus einheitlichen Riechstoffen, ätherischen Ölen und Extrakten.
