Beschreibung
Die Erfindung betrifft neue Acylderivate von Carnitin (ß-Hydroxy-γ-hutyrobetain),
ein Verfahren zu ihrer Herstellung sowie sie enthaltende pharmazeutische Mittel; die Erfindung betrifft insbesondere
neue Acylderivate von Carnitin der allgemeinen Formel

Die Erfindung umfaßt ferner auch die Verbindungen der oben angegebenen
allgemeinen Formel (i) in ihren optisch aktiven Formen (d.h. die entsprechenden D- und L-Isomeren) sowie in ihren racemischen
Formen (D, L-Formen) sowie auch ihre entsprechenden Salze, insbesondere ihre pharmazeutisch verträglichen Salze sowohl
in ihren optisch aktiven als auch in ihren racemischen Formen.
Die Erfindungen der oben angegebenen allgemeinen Formel (i)
können als solche oder in Form ihrer Salze mit Mineralsäuren oder aliphatischen und aromatischen Mono- oder Polycarbonsäuren
oder mit Sulfonsäuren oder Sulfamidsäuren hergestellt
werden.
Im allgemeinen besitzen die Verbindungen der allgemeinen Formel (i) und ihre pharmazeutisch verträglichen Salze interessante
cardiotrope, hyperlipoproteinämische und hyperlipidämische Eigenschaften.
Die Verbindungen der allgemeinen Formel (i) werden normalerweise
in Form ihrer Hydrochloride hergestellt. In der Tat ist es bevorzugt, das ß-Hydroxy-y-butyrobetain-hydrochlorid mit den
Acylchloriden, in denen der Acylrest R die oben angegebene

Bedeutung hat, umzusetzen. Die Umsetzung zur Herstellung dieser neuen Acylderivate wird normalerweise bei einer Temperatur zwischen
0 C und 80 C unter wasserfreien Bedingungen und in Gegenwart eines Überschusses von Trifluoressigsäure durchgeführt. Wenn das
Acylchlorid fest ist und in Trifluoressigsäure nicht leicht löslich ist, ist es möglich, seine Löslichkeit zu verbessern, um auf diese
Weise eine homogene Phase zu erhalten, indem man eine geringe
Menge eines Chlor enthaltenden Lösungsmittels, wie z.B. wasserfreies Methylenchlorid oder Chloroform, zugibt. Besondere Sorgfalt
muß darauf verwandt werden, die Reaktionszone unter wasserfreien Bedingungen zu halten durch Abschirmen der Reaktionszone mit CaClenthaltenden
Rohren. Nach Beendigung der Reaktion wird die dabei erhaltene Mischung abgekühlt und in der Regel mit Aceton behandelt;
ein eventueller Feststoff, der sich abscheiden kann, wird entfernt, während der Niederschlag, der sich bei der Zugabe von Athyläther
bildet, gesammelt wird. Das ausgefallene Produkt kann durch Kristallisation mit weiterem Äthyläther gereinigt werden. Im allgemeinen
reichen eine oder zwei Kristallisationen aus, um ein Produkt mit einer hohen Reinheit zu erhalten, die durch Dünnschichtchromatographie
unter Verwendung von Siliciumdioxidplatten und unter Verwendung verschiedener Eluierungsmittel, wie CHCl„/MeOH/-konzentriertesNH.OH
(Volumenverhältnis 50:30:8) oder n-BuOH/Essigsäure/H„0
(Volumenverhältnis 60:20:20) geprüft werden kann.
Im allgemeinen variieren die Reaktionsausbeuten von 60 bis 85 %,
ungeachtet einer möglichen Verringerung, die bei der Reinigung durch Kristallisation auftreten kann.
Zur Herstellung der Acylderivate von Carnitin, bei denen die Acylgruppe eine solche ist, die von einer a- oder ß-Ketosäure

abgeleitet ist, ist es bevorzugt, zuerst die Ketogruppe zu schützen, indem man sie in ein Ketal überführt. Die Ketosäure
wird dabei zuerst in einen Ketoester und danach in ein Ketal überführt durch Umsetzung des Ketoesters mit Athylenglykol.
Das Ketal des Esters wird zu dem Säureketal hydrolysiert und dann mit Thionylchlorid in das Säurechlorid-Ketal umgewandelt.
Dieses Säurechloridketal wird bei der Umsetzung mit ß-Hydroxy-γ-butyrobetain
(Carnitin) unter Anwendung der vorstehend beschriebenen Verfahren eingesetzt. Die Carbonylschutzgruppe wird
während der Umsetzung hydrolysiert und das isolierte Rohmaterial enthält oder besteht aus dem gewünschten Acylcarnitin.
Die Erfindung, insbesondere das erfindungsgemäße Verfahren, wird durch die nachfolgenden Beispiele, in denen auch einige
physikalisch-chemische Daten der erfindungsgemäßen Hauptprodukte angegeben sind, näher erläutert, ohne jedoch darauf beschränkt
zu sein.
Zu einer Lösung von 3,94 g (0,02 Mol) Carnitinchlorid in 9 ml
Trifiuoressigsäure wurden 3,25 g (0,02 Mol) Dipropylessigsäurechlorid
zugegeben. Die Mischung wurde 24 Stunden lang unter Rühren bei Raumtemperatur gehalten. Dann wurden 70 ml Aceton
zugegeben und die Mischung wurde 2 Stunden lang unter Rühren bei 5 C gehalten. Das ausgefallene Carnitin wurde abfiltriert.
Zu der Lösung wurden außerdem 70 ml Äthyläther zugegeben und die Mischung wurde 30 Minuten lang unter Rühren bei 5 C gehalten.
Der gebildete Feststoff wurde abfiltriert. Das Rohprodukt wurde

in Isoproponol/Äthylathsr kristallisiert und ram erhielt ®s in

1,98 g (0,01 Mol) Camitinchlorid wurden in 3 ml CF3GOOH gelöst
und zu der Lösung wurde Überschüssiges Säurechlorid (7 ml) zugegeben.
Die Lösung wurde etwa 48 Stunden lang unter Rühren bei Raumtemperatur
gehalten. Danach wurde die Mischung mit 20 ml Aceton verdünnt und es wurde etwas Äther langsam zugegeben, bis die
Ausfällung vollständig war. Die Mischung wurde filtriert und der Niederschlag, der die Neigung hatte, hygroskopisch zu werden,
wurde schnell mit Äther gewaschen und unter Vakuum bei einer Temperatur von 50 C getrocknet. Dabei erhielt man 1,70 g (Ausbeute
60 %) eines Produkts mit den nachfolgend angegebenen Eigenschaften:

4,55 g (0,023 Mol) Carnitinchlorid wurden in 6,9 ml CF3COOH
gelöst und zu der Lösung wurde Überschüssiges Cinnamoylchlorid
(16 ml) zugegeben. Die Mischung wurde 4 bis 5 Stunden lang unter Ruhren bei 40 bis 45 C gehalten. Danach wurde die Mischung
mit 60 ml Aceton verdünnt und es wurde etwas Äther langsam

zugegeben bis die Ausfällung vollständig v/or. Die Mischung wurde
filtriert und der Niederschlag, der die Neigung hatte, hygroskopisch zu werden, wurde schnell mit Äther gewaschen und unter
Vakuum bei einer Temperatur von nicht über 500C getrocknet; dabei
erhielt man 5,3 g eines Produkts (Ausbeute 70 %) mit den folgenden

7,12 g (0,036 Mol) Carnitinchlorid wurden in 12 ml CF,CQOH gelöst
und zu der Lösung wurde überschüssiges p-Methoxycinnaraoylchlorid
(25 nil) zugegeben und die Mischung wurde 4 bis 5 Stunden lang
unter Rühren bei 40 bis 50 C gehalten» Danach wurde die Mischung mit 90 ml Aceton verdünnt und es wurde etwas Äther langsam
zugegeben bis die Ausfüllung vollständig wor. Die Mischung wurde filtriert und der Niederschlag, der die Neigung hatte, hygroskopisch

zu werden, wurde schnell mit Äther gewaschen und unter Vakuum bei einer Temperatur von nicht über 50 C getrocknet; dabei
erhielt man 9 g eines Produkts (Ausbeute 70 %) mit den folgenden

Beispiel 5: p-Isobutylphenylacetylcarnitinchlorid
5,5 g (0,028 Mol) Carnitinchlorid wurden in 9 ml CF3COOH gelöst
und zu der Lösung wurde Überschüssiges Säurechlorid (20 ml)
zugegeben und die Mischung wurde 4 bis 5 Stunden lang unter Rühren bei 40 bis 45 C gehalten. Danach wurde die Mischung
in S-LO/CHCl, verteilt, die organische Phase wurde verworfen
und die wäßrige Phase wurde unter vermindertem Druck bei einer Badtemperatur von etwa 50 C eingeengt« Dabei erhielt man
ein gelatineartiges Rohmaterial, das in Isopropanol kristallisiert
wurde, der dabei erhaltene Niederschlag %#yrde abfiltriert, wegen
seiner Hygroskopizität schnell mit Äther gewaschen und unter
Vakuum bei einer Temperatur von nicht über 500C getrocknet; dabei
erhielt man 6,8 g eines Produkts (Ausbeute 65 %) mit den folgenden
Eigenschaften: F. 130 bis 132°C,

4,95 g (0,025 Mol) D^-Carnitinchlorid wurden in 8 ml CF..COOH
gelöst und zu der Lösung wurde Überschüssiges Säurechlorid
(16 ml) zugegeben und die Mischung wurde 4 bis 5 Stunden lang unter Ruhren bei einer Temperatur von 40 bis 45 C gehalten.
Danach wurde die Mischung zwischen H„0 und CHCl,, verteilt,
die organische Phase wurde verworfen, während die wäßrige Phase unter vermindertem Druck bei einer Badtemperatur von etwa 50 C
eingeengt wurde. Dabei erhielt man ein gelatineartiges Rohmaterial, das in Isopropanol kristallisiert wurde. Der erhaltene

Niederschlag wurde abfiltriert, wegen seiner Hygroskopizität schnell mit Äther gewaschen und unter Vakuum bei einer Temperatur

Beispiel 7: Glutarvlcarnitinchlorid
3,9 g (0,02 Mol) Carnitinchlorid wurden in 6,5 ml (0,06 Mol) Trifluoressigsäure gelöst und danach mit 3,0 g (0,02 Mol)
Glutarylchlorid umgesetzt in einem Kolben, der mit einem Rückflußkühler mit einem Cad« enthaltenden Rohr ausgestattet war
und auch einen MagnetrUhrer enthielt und in einem Bad gehalten wurde, dessen Temperatur während der gesamten Reaktionsdauer
(12 Stunden) bei einer Temperatur von 40 bis 45 C gehalten wurde.

Die Reaktionsmischung wurde unter Rühren mit 60 ml Aceton behandelt
und die geringe Menge an gebildetem Feststoff wurde abfiltriert« Unter Rühren wurden langsam 130 ml Äthyläther zugegeben, bis beim
Abkühlen in einem Eisbacl eine Ausfällung begann. Das etwas zerfließende
Rohprodukt, das durch Filtrieren gesammelt wurde (4,7 g)
wurde einmal kristallisiert und dabei erhielt man 4„02 g (71 %)
eines festen Produkts mit den folgenden Eigenschaften?

7,2 § (0,062 Mol) Lövulinsöore würden mit 8 ml konzentrierter
HLSO^, und 200 ml ebsolutero XtOH verestert. Der dabei erhaltene
Äthylester (7,0 fl, 0,048 Mol) wurde mit 8,2 ml Ä'thylenglykol
und 0,112 g p-Toluolsulfonsäore 96 Stunden lang bei 1700C in
wasserfreiem Toluol behandelt. Nach Beendigung der Reaktion wurde die organische Phase mit einer Lösung von gesättigtem
NaHCO^ und Ho0 gewaschen und anschließend über wasserfreiem
-λ/. Λ
Na„SO. getrocknet. Nach dem Trocknen der Lösung erhielt man
5 g (55 %) des Ketals des Lävulinsäureäthylesters. Das dabei
erhaltene Produkt wurde in 40 ml Methanol und 40 ml 1 η NaOH gelöst und 2 Stunden lang bei Raumtemperatur gehalten. Man erhielt
4 g des Ketals der Lävulinsäure, die mit 5 ml SOCl« 4 Stunden lang
bei 80 C behandelt wurden zur Herstellung des Säurechlorids des Ketals der Lävulinsäure. Dabei erhielt man etwa 5 g eines
schwach dunklen Säurechlorids, die zu 4,5 g (0,023 Mol) Carnitinchlorid, gelöst in 10 ml Trifluoressigsäure, zugegeben wurden.
Die Reaktionsmischung wurde Über Nacht unter Ruhren bei 50 C gehalten.
Nach der Zugabe von 40 ml Aceton entstand ein dUnner Niederschlag, der abfiltriert wurde. Zu der Mischung wurden 138 ml kalter
Äthyläther (von 0 C) zugegeben und die Mischung wurde über Nacht unter Rühren stehen gelassen. Aus der Lösung schieden sich 5,4 g
eines weißen Rohproduktes aus, das in Äthyläther weiter kristallisiert wurde. Dabei erhielt man 4,7 g eines Produkts mit den folgenden

6,4 g (0,043 Mol) ß-Ketoglutarsäure wurden 4 Stunden lang bei 80 C mit 6 ml konzentrierter HpSO, und 200 ml absolutem Äthylalkohol behandelt. Der entstehende Äthylester (6,5 g, 0,037 Mol)
wurden mit 9 Mol Äthylenglykol und 0,120 mg p-Toluolsulfonsäure
unter Rückflußbedingungen 72 Stunden lang in wasserfreiem Toluol umgesetzt. Die Reaktionsmischung wurde mit einer gesättigten
NaHCO_-Lösung, mit Wasser gewaschen und nach den Trocknen Über
wasserfreiem Na«SO. unter Vakuum zur Trockne eingedampft. Nach
dem Trocknen der Lösung erhielt man 4,8 g (65 %) des Ketals des
ß-Ketoglutarsäureäthylesters. Der dabei erhaltene Feststoff wurde
in 45 ml Methylalkohol und 40 ml 1 η NaOH gelöst und unter Ruhren
2 Stunden lang bei Raumtemperatur gehalten. Dabei erhielt man 3,8 g (0,02 Mol) (90 %) des Ketals der ß-Ketoglutarsäure, die mit
6 ml Thionylchlorid 6 Stunden lang bei 80 C behandelt wurden zur Herstellung des entsprechenden Säurechlorids. Das erhaltene
Säurechlorid wurde langsam zu 4,2 g (0,022 Mol) Carnitinchlorid,
gelöst in Trifluoressigsäuren zugegeben, die Mischung wurde 12
Stunden lang unter Ruhren bei 60 C gehalten und dann auf 0 C
ebgekUhlt. Danach wurden 40 ml Aceton zugegeben. Es bildete sich
eine schwache Wolke, die durch Zentrifugieren eliminiert wurde. Zu der zentrifugierten Lösung wurden 130 ml Äthyläther bei 0°C
zugegeben. Es entstand ein weißer Niederschlag, der noch einmal

Zu 4,5 g (0,023 Mol) Carnitinchlorid, gelöst in 10 ml Trifluoressigsäure und erhitzt auf 400C, wurden 3,09 g (0,023 Mol)
Fumarsäurechlorid unter starkem Rühren langsam zugegeben. Die
Reaktionsmischung wurde 12 Stunden lang gerUhrt, wobei derauf geachtet wurde, daß die Temperatur 40 C nicht Überstieg, und wobei
der Reaktionskolben durch ein CaCl0 enthaltendes Rohr abgeschirmt
wurde. Die Mischung wurde dann mit einem Eisbad auf 0 C abgekühlt
und es wurden langsam 60 ml Aceton zugegeben. Die Lösung wurde trUbe und sie wurde dann zentrifugiert. Zu der klaren Lösung
wurden langsam 120 ml einer Mischung aus Äthyläther und Hexan (i:i) zugegeben. Dabei erhielt man 5,4 g eines Rohprodukts in
Form eines Öls, das langsam fest wurde. Nach der Kristallisation aus

Zu einer Lösung von 1,58 g (0,008 Mol) Carnitinchlorid in
5 ml Trifluoressigsäure wurden 1,78 g (0,008 Mol) des Säurechlorids von NjS-Diacetylcystein in 8 ml Trifluoressigsäure

Zu 2,8 g (0,014 Mol) Carnitinchlorid, gelöst in 8 ml Trifluoressigstiure, wurden unter konstantem Ruhren langsam 2,48 g (0,014
Mol) Acetylvalinchlorid zugegeben. Das Acetylvalinchlorid wurde vorher durch Umsetzung von Acetylvalin mit Überschüssigem
Thionylchlorid hergestellt. Die Mischung wurde Über Nacht unter
starken Rühren in einen thernostatischen Bad bei 400C unter
vorsichtigen Ausschluß von Feuchtigkeit in Gegenwart eines CaCl^ enthaltenden Rohres reagieren gelassen. Der nach der
Zugabe von 40 nl Aceton erhaltene dUnne Niederschlag wurde

Zu einer Mischung aus 10,6 g (0,1 Mol) wasserfreiem Natriumcarbonat, 0,1 ml wasserfreiem Dimethylformamid und 13,9 ml (0,2 Mol)

Brenztraubensäure in 125 ml wasserfreiem Äthyläther, der bei O C
gehalten wurde, wurde Oxalylchlorid, verdünnt in 25 ml wasserfreiem
Äther,in einer Menge äquimolar zu der Brenztraubensäure (17,1 ml,
0,2 Mol) zugetropft. Die dabei erhaltene Mischung wurde 24 Stunden lang unter Ruhren stehen gelassen. Die Reaktionsmischung wurde
dann filtriert und das Filtrat wurde destilliert, wobei die bei 53 C/126 Torr destillierende Fraktion gesammelt wurde, Ausbeute
20 %.
Alternativ wurde das Brenztraubensäurechlorid auch hergestellt
durch Zutropfen von 9 ml (etwa 0,1 Mol) Methyldichlormethyläther
zu 7 ml (etwa 0,1 Mol) Brenztraubensäure bei Raumtemperatur unter Rühren. Nach der Zugabe wurde die Reaktionsmischung auf 50 C erhitzt
und 30 Minuten lang bei dieser Temperatur gehalten. Die Mischung wurde dann destilliert, wobei die bei 53 C/126 Torr destillierende
Fraktion gesammelt wurde.
Das nach einem der vorstehend beschriebenen Verfahren erhaltene Pyruvylchlorid wurde zu einer Lösung von 10 g Carnitinhydrochlorid,,
gelöst in 25 ml Trifluoressigsöure, zugegeben. Die dabei erhaltene
Mischung wurde bei 40 C über Nacht stehen gelassen. Die Mischung wurde dann mit Eis gekühlt und es wurden 40 ml Aceton zugegeben«
Nach 2 Stunden wurde der Niederschlag abfiltriert. Zu dem Filtrat wurden 100 ml Äthylather zugetropft, wobei man ein öl erhielt.
Dieses öl wurde durch Auflösen in einem ÄtOH/Aceton (5/1)-Gemisch
gereinigt, dann wurde Äthylather zugegeben, was zur Bildung eines Niederschlages führte, dessen Analyse ergab, daß es sich dabei um
die Titelverbindung handelte.

Verfahren b); Bei diesem Verfahren erhielt man zuerst das gemischte
Anhydrid der Brenztraubensäure, das dann mit Carnitinperchlorat umgesetzt wurde, wobei man die Titelverbindung (isoliert als Pyruvylcarnitinperchlorat) erhielt.
8,8 g (0,1 Mol) Brenztraubensäure wurden in 100 ml Acetonitril
gelöst und bei einer Temperatur zwischen -10 und 0 C wurden äquimolare
Mengen (in bezug auf die Brenztraubensäure) Triäthylamin (10,1 g, 0,1 Mol) und Äthyl- oder Isobutylchlorformiat (0,1 Mol) zugegeben.
Die dabei erhaltene Reaktionsmischung wurde 1 Stunde lang bei 0 C gehalten; das Triäthylaminhydrochlorid wurde abfiltriert und das
das gemischte Anhydrid enthaltende Filtrat wurde zu einer Lösung von Carnitinperchlorat zugegeben, die wie folgt hergestellt worden
war: 10 g Carnitinhydrochlorid wurden in 150 ml CH-CN suspendiert
und es wurden 12 g Silberperchlorat zugegeben. Die Mischung wurde im Dunkeln unter Rühren etwa 30 Minuten lang stehen gelassen und
dann wurde das ausgefallene Silberchlorid abfiltriert.
Die Reaktionsmischung wurde über Nacht bei 40 C gehalten. Dann wurde die Mischung abgekühlt und filtriert. Zu dem Filtrat wurde
Äthyläther zugegeben, wobei man ein öl erhielt, das durch Auflösen
in ÄtOH/Aceton (5/1) und weitere Ausfällung mit Äther gereinigt

Therapeutische Anwendung der erfindungsgemäßen Acylcarnitinderivate
der allgemeinen Formel Cl) *_____»___«___________-____-__-___,-__-_
Unter Anwendung des Weil-Verfahrens (l) wurde die Verträglichkeit
der Acylcarnitine der allgemeinen Formel (I) sowie ihrer Salze,
die auf intraperitonealem Wege verabreicht wurden, bei Mäusen
untersucht. Wie aus den in der nachfolgenden Tabelle I angegebenen LD-n-Werten hervorgeht, weisen alle Acylderivate von Carnitin eine
gute Verträglichkeit auf.
Es wurde auch der cardiokinetische Effekt am isolierten Herzen wie
nachfolgend angegeben untersucht: Kaninchenherzen, die nach dem
Langendorff-Verfahren isoliert worden waren, wurden mit einer mit Sauerstoff angereicherten Ringer-Lösung bei 38,2°C perfundiert.
Die isometrischen Kontraktionen, das Elektrocardiogramm und der
Coronardurchfluß wurden unter Verwendung eines Battaglia-Rangoni-Polygraphen
aufgezeichnet. Nach der Entfernung des Sauerstoffs aus der Perfusionsfiüssigkeit wurde in dem Herzmuskel eine Stoffwechselschädigung
induziert bis zu einer Abnahme der Herzkontsaktionskraft

um 80 %. Unter diesen Bedingungen der verlängerten Anoxie wurde
die aerobe Glykolyse des Myocardiums verringert, begleitet von
der Speicherung der sauren Katabolite sowohl aufgrund der Anreicherung von Brenztraubensäure als auch aufgrund ihrer Umwandlung
in Milchsäure, die wegen der Depression der Pyridinenzyme, wie z.B. LDH (Lactodehydrogenase),nicht ausgenutzt werden kann. Dies
hat Auswirkungen auf die anaerobe Glykolyse, die eine stets zunehmende Anzahl von Enzymen betraf, begleitet von einer
progressiven und zunehmenden kritischen Erschöpfung des Myocardiums. Dabei trat eine ganze Serie von HerzmuskelermUdungserscheinungen auf, die an Hand des Verhaltens der untersuchten
Parameter, insbesondere der Kontraktionskraft, des Coronardurchflusses, der Herzgeschwindigkeit und des Herzrhythmus, beobachtet
werden konnten. Sobald die Kontraktionskraft um 80 % herabgesetzt worden war, wurde die PerfusionsflUssigkeit erneut mit Sauerstoff
angereichert entweder ohne Zugabe von anderen Verbindungen (Kontrollen) oder unter Zugabe der untersuchten Verbindungen.
In der nachfolgenden Tabelle II sind die Prozentwerte der Kontraktionskraft des Herzens angegeben, die einen positiven ionotropen
Effekt anzeigen,errechnet 10 Minuten nach der Unterbrechung der Anoxieperiode (Myocarderholung).
Die unter Anwendung des Studenten-t-Tests ausgewerteten Ergebnisse
zeigen, daß bei gleichen Konzentrationen in der PerfusionsflUssigkeit Trifluormethylcinnamoyl, Cinnamoyl, Pivaloyl, Brompropionyl,
Dipropylacetyl und Pyruvyl einen größeren positiven ionotropen Effekt induzieren als die anderen untersuchten Verbindungen, wobei
statistisch signifikante Unterschiede im Vergleich ζυ den Kontroll-

versuchen auftraten.
Der coronarvasodilatorische Effekt wurde ebenfalls untersucht,
wobei die nachfolgend angegebenen Ergebnisse erhalten wurden. Alle untersuchten Verbindungen der Formel (i) riefen im Vergleich
zu den Kontrollen eine geringe statistisch nicht-signifikante Zunahme des Coronardurchflusses hervor.
Es wurde auch der/chronotrope Effekt untersucht und alle geprüften
Verbindungen der Formel (i) führten zu keiner signifikanten
Änderung der Herzgeschwindigkeit gegenüber den Kontrollen.
Außerdem wurde der Antiarrhythmieeffekt untersucht und es wurde
gefunden, daß bei Anwendung des isolierten Ratten-Herzvorhof-Verfahrens von M. Libonati und G. Segre (2) unter den geprüften
Verbindungen der Formel (I) Pyruvyl, Trifluormethylcinnamoyl, Pivaloyl, Chlorbutyryl, Dipropylacetyl besonders vorteilhaft waren,
da sie die ausgeprägtesten Antiarrhythmieeigenschaften aufwiesen. Diese Ergebnisse sind in der nachfolgenden Tabelle III angegeben.
Außerdem wurde der Antiarrhythmie-Effekt der Verbindungen bei
Mäusen nach dem Verfahren von P.W. Nwangwu und T. Holcslow (4) untersucht. Bei Verwendung von Aconitin (5 γ /ml) als
arrhythmogenes Mittel wurden die Änderungen des Herzrhythmus der Tiere aufgezeichnet und die Ausbrudisztitder anfänglichen
Arrhythmie und/oder der Ventrileular-Tachycardie wurden als Endpunkt verwendet. Die Antiarrhythmiemittel fuhren zu einer
Zunahme der Latenzzeit der anfänglichen ECG-Änderung.

Die in der folgenden Tabelle IV angegebenen Ergebnisse zeigen, daß die Verbindungen eine Antiarrhythmie-Aktivität aufweisen,
die ausgeprägt ist bei Dipropylacetyl, Cyclohexylpropionyl, Trifluormethylcinnamoyl, Methoxycinnamoyl und Pyruvyl.
Ferner wurde der Antagonismus zu der durch Adrenalin induzierten
Toxizität untersucht, wobei die folgenden Ergebnisse erhalten wurden: Gruppen von 10 männlichen Albino-Mäusen vom Stamme
Swiss wurden in zunehmenden logarithmischen Dosen Adrenalin (Tartrat) intraperitoneal injiziert. Anderen ähnlichen Tiergruppen wurden die untersuchten Verbindungen auf dem gleichen
Wege 30 Minuten vor der Adrenalinverabreichung in einer Dosis von 150 mg/kg injiziert. Nach dem Verfahren von Litchfiled und
Wilcoxon (3) wurde 36 Stunden nach der Adrenalinverabreichung die Mortalität bestimmt. Die dabei erhaltenen Ergebnisse sind in der
nachfolgenden Tabelle V angegeben.
Alle oben-genannten Verbindungen und ihre pharmazeutisch verträglichen Salze sind für die Therapie von Herzerkrankungen vom
Anoxie-, Ischämie-, Arrhythmie- und cardiotoxischen Typ sowie in solchen Fällen, in denen der Energiebedarf des Herzens steigt,
am meisten bevorzugt.
Der antilipämische Effekt einiger Acylderivate der Formel (i)
wurde unter zwei verschiedenen experimentellen Bedingungen untersucht:
Bei Ratten, die 17 Stunden lang gefastet hatten, wurden die F.F.A. (fettfreien Säuren)-Plasmagehalte durch eine einzige

Bei den Ratten wurde das durch eine Ölverabreichung mittels des Futters geänderte Lipoproteinmuster nach einer einzigen Behandlung
mit d,l-Dipropyl-ACAR; d,l-Äthylhexanoyl-CAR, d,l-3-»Brompropionyl-CAR
und d,l-Pyruvyl-CAR wieder rückgebildet. Den ausgeprägtesten
Effekt, angezeigt durch die Zunahme der HDL (der Lipoproteinfraktion mit hoher Dichte) und die Abnahme der LDL und VLDL
(der Lipoproteinfraktion mit niedriger Dichte und der Lipoproteinfraktion
mit sehr niedriger Dichte) wies das d,1-Pyruvy!derivat
auf, das gegenüber den Triglyceriden und den Cholesterinplasma-Gehalten,
die nach der ölaufnahme anstiegen, aktiv war. Die Ergebnisse sind in der folgenden Tabelle VI angegeben.
1.) Lösungen und sterile wäßrige Lösungen, die Acylcarnitine in
Konzentrationen von 25 mg bis 500 mg pro ml enthalten
a) Der Arzneimittelträger für injizierbore Ampullen/Phiolen wurde
unter Anwendung der nachfolgenden, die Erfindung nicht beschränkenden Zusammensetzung hergestellt:
Natriumcarboxymethylcellulose (mit niedriger
Viskosität) ■' 10 mg/ml
Polysorbate 80 4 mg/ml
Polyparaben 0,4 mg/ml
Wasser für die Injektionen in einer ausreichenden
Menge für 1 ml-, 2 ml-, 5 ml- und 10 ml-Ampullen/dPhiolen

b) Der Arzneimittelträger für Phleboclysis-Flaschen, die 50 ml,
100 ml, 250 ml, 500 ml und 1000 ml enthielten, wurde unter Anwendung der nachfolgend angegebenen, die Erfindung nicht beschränkenden Zusammensetzung hergestellt:
NaCl 8,6 g/l
KCl 0,3 g/l
CaCl2 0,33 g/l
c) Der Arzneimittelträger für Flaschen für die orale Verwendung,
die 5 ml bis 100 ml enthielten, wurde unter Anwendung der folgenden, die Erfindung nicht begrenzenden Zusammensetzung hergestellt:
Vitamin B-« 3 μg/ml
gereinigtes Wasser in genügender Menge
2.) Tabletten, die 20 Mg bis 500 mg Acylcarnitin oder irgendeines
der geprüften Derivate enthalten
Der Arzneimittelträger wurde unter Anwendung der nachfolgend angegebenen, die Erfindung nicht beschränkenden Zusammensetzung
hergestellt:

2. Acylderivate von Carnitin nach Anspruch 1, dadurch gekennzeichnet, daß sie in ihren optisch aktiven Formen vorliegen.
3. Acylderivate von Carnitin nach Anspruch T, dadurch gekennzeichnet, daß sie in ihrer racemischen Form vorliegen.
4. Acylderivate von Carnitin nach einem der Ansprüche 1 bis
3, dadurch gekennzeichnet, daß sie in Form eines pharmazeutisch

verträglichen Salzes vorliegen.
5. Verfahren zur Herstellung der Acylderivate von Carnitin nach
einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, daß man
a) das Chlorid der entsprechenden Säure herstellt und
b) das Chlorid der Stufe (a) in Gegenwart von Trifluoressigsäure
bei einer Temperatur zwischen etwa Raumtemperatur und etwa
60 C mit Carnitinhydrochlorid umsetzt.
6. Pharmazeutisches Mittel, insbesondere fUr die Behandlung von
Herzstörungen, Hyperlipoproteinämien und Hyperlipämien, dadurch
gekennzeichnet, daß es besteht aus oder enthält eine wirksame Menge mindestens eines Acylderivats von Carnitin nach einem der
Ansprüche 1 bis 4.
