Description
Die Erfindung betrifft ein Verfahren zur Herstellung
des Decapeptids Ceruletid
und seiner Salze gemäß dem
voranstehenden Patentanspruch.
Nach der Nomenklatur, die gewöhnlich in der Peptid-Chemie
gebraucht wird, kann das therapeutisch wertvolle Decapeptid gemäß der Erfindung
durch die Formel
d. h. Pyroglutamyl-Glutaminyl-Asparaginyl-Tyrosyl (O-Sulfat)-
Threonyl-Glycyl-Tryptophyl-Methionyl-Asparaginyl-
Phenylalaninamid, worin die phenolische Hydroxylgruppe
des Tyrosin-Restes mit einer Sulfatgruppe verestert ist
und worin alle Aminosäuren (mit Ausnahme von Glycin)
die L-Konfiguration haben, dargestellt werden.
Bei dem Syntheseverfahren gemäß US-PS 34 72 832 erfolgt die Herstellung
durch Sulfatierung des geschützten Decapeptids
worin die Hydroxylgruppe des Threoninrestes acetyliert
ist, und anschließende Verseifung der Acetylgruppe. Bei
diesem Verfahren entstehen als Folge sowohl der
Sulfatierungsbedingungen im wasserfreien Medium als auch der
Verseifungsstufe mehrere Nebenprodukte, hauptsächlich
polysulfatierte, desamidierte und oxidierte Nebenprodukte
neben anderen nicht identifizierten Verunreinigungen.
Alle diese Verunreinigungen konnten nur durch sorgfältige
Reinigung durch Gegenstromverteilung mit dem hiermit
verbundenen Verlust an Zeit und Ausbeute entfernt werden.
Beim erfindungsgemäßen Verfahren wird das ungeschützte
Decapeptid
in einem wäßrigen Medium sulfatiert, wobei keine
abschließende Verseifung erforderlich ist. Unter diesen
milderen Bedingungen wird das Ceruletid in hohen
Ausbeuten (Umsatz 95% und mehr) und in hoher Reinheit
(etwa 90% gemäß HPLC) erhalten. Eine weitere Reinigung
kann durch Gegenstromverteilung vorgenommen werden.
Das ungeschützte Decapeptid III wird nach einem
Syntheseschema erhalten, bei dem nicht nur die Gefahr
der Racemisierung vermieden, sondern auch die Gefahr des
Abbaus des Tryptophanrestes durch wiederholte Behandlung
mit starken Säuren vermindert wird. Die Synthese dieses
Decapeptids besteht im wesentlichen in geeigneten
aufeinanderfolgenden Kondensationen von geschützten
Aminosäuren oder Peptidfragmenten. Die Kupplung wird so
durchgeführt, daß das erhaltene Decapeptid die gewünschte
Sequenz von zehn Aminosäuren hat. In den Aminosäuren und
Peptidfragmenten, die nach Verfahren, die in der Peptid-
Chemie an sich bekannt sind, kondensiert werden, sind die
Aminogruppen und Carboxylgruppen, die an der Bildung der
Peptidbindung unbeteiligt, jedoch reaktionsfähig sind,
durch eine Schutzgruppe, die durch Acidolyse, Verseifung
oder Hydrogenolyse entfernbar ist, blockiert.
Für den Schutz der Aminofunktion können beispielsweise
die folgenden Schutzgruppen verwendet werden: Carbobenzoxy
(Benzyloxycarbonyl), t-Butoxy (t-Butyloxycarbonyl),
Trityl (Triphenylmethyl), Formyl oder Trifluoracetyl.
Für den Schutz der Carboxylfunktion können beispielsweise
die folgenden Schutzgruppen verwendet werden: Methyl,
Äthyl, t-Butyl und Benzyl.
Die Kupplung zwischen einer Aminogruppe eines Moleküls
und einer Carboxylgruppe eines anderen Moleküls zur Bildung
einer Peptidbindung kann mit einem aktivierten
Acylderivat, z. B. einem gemischten Anhydrid, einem Azid,
einem p-Nitrophenyl- oder 2,4,5-Trichlorphenyl- oder
N-Hydroxysuccinimidylester oder durch direkte
Kondensation zwischen einer freien Aminogruppe und einer
freien Carboxylgruppe mit Hilfe eines Kondensationsmittels,
z. B. Dicyclohexylcarbodiimid oder 1-Cyclohexyl-3-
morpholinylcarbodiimid, allein oder in Gegenwart von 1-Hydroxybenzotriazol,
N-Hydroxysuccinimid und 8-Hydroxychinolin
erfolgen. Die Kondensation kann in einem Lösungsmittel,
beispielsweise einem N,N-Di-niederalkylformamid, einem
niederen aliphatischen Nitril, einem Pyridin oder Tetrahydrofuran,
beispielsweise in Dimethylformamid, Acetonitril,
Pyridin oder Tetrahydrofuran, durchgeführt werden.
Die Reaktionstemperatur kann zwischen -20°C und Raumtemperatur
liegen. Die Reaktionszeit beträgt im allgemeinen
6 bis 120 Stunden.
Die Sulfatierung
wird in einem
wäßrigen Medium mit dem Komplex B : SO3, worin B: eine
tertiäre Base ist, durchgeführt. Beispiele geeigneter
tertiärer Basen sind Pyridin oder substituierte Pyridine,
Trimethyl-, Triäthyl- oder niedere Trialkylamine, N-Niederalkyl-
morpholine, Dimethylanilin, Imidazol oder substituierte
Imidazole und Chinolin. Das Decapeptid III wird
in Wasser bei pH 9,5-10,5 durch Zusatz einer organischen
oder anorganischen Base oder beider Basen gelöst. Als
organische Basen kommen die vorstehend genannten Basen
infrage. Als anorganische Basen können NaOH, KOH oder Na2CO3
verwendet werden. Der Komplex wird als solcher oder als
Lösung in einem mit Wasser mischbaren organischen Lösungsmittel,
z. B. Dimethylformamid, Aceton und Acetonitril,
zugesetzt. Der pH-Wert des Reaktionsgemisches wird durch
Zusatz einer der vorstehend genannten Basen im Bereich
von 8,5-12,5 gehalten. Die Reaktionszeit beträgt im
allgemeinen 2-120 Stunden. Die erfindungsgemäße
Synthese von Ceruletid umfaßt die im Beispiel beschriebene
Sulfatierung mit anschließender Aufarbeitung des
Decapeptids. Das Decapeptid III wird durch
Kupplung des Hexapeptids IV mit dem Tetrapeptid V nach
der in Beispiel 3 beschriebenen Azidmethode hergestellt.
Die beiden Zwischenprodukte IV und V werden durch stufenweise
Verlängerung gemäß den Beispielen 1 bzw. 2
synthetisiert. Das vorstehend genannte Syntheseschema kann
wie folgt dargestellt werden:
Die Erfindung wird durch das folgende Beispiel weiter
erläutert.
Zu einer Lösung von 17,5 g Boc-Gly-OH in 300 ml wasserfreiem
Tetrahydrofuran werden 10,99 ml N-Methylmorpholin
und 9,60 ml Äthylchlorformiat nacheinander bei einer
Temperatur von -15°C gegeben. Das Gemisch wird 2 Minuten
bei dieser Temperatur gerührt, worauf 20,3 g
NH2-NH-Z·HCl (Z=Benzyloxycarbonyl) in 200 ml wasserfreiem
Dimethylformamid, das 10,99 ml N-Methylmorpholin
enthält, zugesetzt werden. Das Reaktionsgemisch wird
2 Stunden bei -10°C gerührt. Man läßt dann die Temperatur
in 2 Stunden von -10°C auf 0°C steigen. Das Lösungsmittel
wird unter vermindertem Druck entfernt und der
Rückstand in Äthylacetat aufgenommen und nacheinander
mit 1N-HCl bei 0°C, mit einer 5%igen wäßrigen Lösung
von NaHCO3 und mit einer gesättigten NaCl-Lösung neutral
gewaschen. Nach Entfernung des Lösungsmittels wird der
Rückstand aus Petroläther kristallisiert, wobei
29,1 g (Ausbeute 90%) Boc-Gly-NH-NH-Z (VI)
vom Schmelzpunkt 84-87°C erhalten werden.
29,1 g Boc-Gly-NH-NH-Z (VI)
werden in 300 ml 99%iger Ameisensäure gelöst und 6 Stunden
bei Raumtemperatur stehen gelassen. Das Lösungsmittel
wird unter vermindertem Druck bei 30°C entfernt und
der ölige Rückstand aus Diäthyläther kristallisiert, wobei
23,0 g (Ausbeute 95%) H-Gly-NH-NH-Z·HCOOH (VII)
vom Schmelzpunkt 135-136°C erhalten werden.
18,75 g Boc-Thr-OH werden mit
23,0 g H-Gly-NH-NH-Z·HCOOH (VIII)
über das Äthoxyameisensäureanhydrid auf die für
die Herstellung der Verbindung VI beschriebene Weise
kondensiert, wobei
32,3 g (Ausbeute 89%) Boc-Thr-Gly-NH-NH-Z (VIII)
vom Schmelzpunkt 70-73°C erhalten werden.
32,3 g Boc-Thr-Gly-NH-NH-Z (VIII)
werden mit 350 ml Trifluoressigsäure bei 0°C 30 Minuten
behandelt. Das Lösungsmittel wird dann unter vermindertem
Druck bei 0°C abgedampft und der ölige Rückstand
aus Diäthyläther kristallisiert, wobei
33,0 g (Ausbeute 99%) H-Thr-Gly-NH-NH-Z·CF3COOH (IX)
vom Schmelzpunkt 140-145°C erhalten werden.
21,2 g Boc-Tyr-OH werden mit
33 g H-Thr-Gly-NH-NH-Z·CF3COCH (IX)
durch das Äthoxyameisensäureanhydrid
auf die für die Herstellung der Verbindung VI beschriebene
Weise kondensiert. Nach Kristallisation aus Äthylacetat-
Petroläther werden
39,0 g (Ausbeute 88%) Boc-Tyr-Thr-Gly-NH-NH-Z (X)
vom Schmelzpunkt 138-142°C erhalten.
Durch Acidolyse von
39,0 g Boc-Tyr-Thr-Gly-NH-NH-Z (X)
mit Trifluoressigsäure auf die für die Herstellung der
Verbindung IX beschriebene Weise werden
39,9 g (Ausbeute 100%) H-Tyr-Thr-Gly-NH-NH-Z·CF3COOH (XI)
vom Schmelzpunkt 145-150°C erhalten.
15,5 g Boc-Asp(OBzl)-OH werden mit
39,9 g H-Tyr-Thr-Gly-NH-NH-Z·CF3COOH (XI)
durch das Äthoxyameisensäureanhydrid in üblicher Weise kondensiert. Durch Kristallisation aus Äthylacetat werden

48,9 g (Ausbeute 93%) Boc-Asp(OBzl)-Tyr-Thr-Gly-NH-NH-Z (XII)
vom Schmelzpunkt 134-138°C erhalten.
Durch Acidolyse von
48,9 g Boc-Asp(OBzl)-Tyr-Thr-Gly-NH-NH-Z (XII)
mit Trifluoressigsäure in üblicher Weise werden
49,2 g (Ausbeute 99%) H-Asp(OBzl)-Tyr-Thr-Gly-NH-NH-Z·CF3COOH (XIII)
vom Schmelzpunkt 135-138°C erhalten.
In analoger Weise werden durch
Kondensation von 15,0 g Boc-Gln-OH mit
49,2 g H-Asp(OBzl)-Tyr-Thr-Gly-NH-NH-Z·CF3COOH (XIII)
durch das Äthyoxyameisensäureanhydrid nach Kristallisation aus Methanol
49,5 g (Ausbeute 88%) Boc-Gln-Asp(OBzl)-Tyr-Thr-Gly-NH-NH-Z (XIV)
vom Schmelzpunkt 200-203°C erhalten.
Durch Behandlung von
49,5 g Boc-Gln-Asp(OBzl)-Tyr-Thr-Gly-NH-NH-Z (XIV)
mit Trifluoressigsäure in üblicher Weise werden
50,2 g (Ausbeute 100%) H-Gln-Asp-(OBzl)-Tyr-Thr-Gly-NH-NH-Z·CF3COOH (XV)
vom Schmelzpunkt 160-162°C erhalten.
Durch übliche Kondensation von
durch Äthoxyameisensäureanhydrid werden nach Kristallisation
aus Methanol,
vom Schmelzpunkt 220-225°C (Zers.) erhalten.
werden in 500 ml Dimethylformamid, das 2 Äquivalente
HCl enthält, gelöst und in einem offenen Gefäß in
strömendem Wasserstoff in Gegenwart von 14 g 10%iger
Palladiumkohle, die innerhalb von 12 Stunden portionsweise
zugesetzt wird, hydriert. Der Katalysator wird abfiltriert
und mit Dimethylformamid gut gewaschen. Der ölige Rückstand
wird aus absolutem Äthanol kristallisiert, wobei
vom Schmelzpunkt 145-150°C (Zers.) erhalten werden.
23,2 g Boc-Asp(OBzl)-OH werden mit 20,0 g H-Phe-NH2 durch
das Äthoxyameisensäureanhydrid in der für die Herstellung
der Verbindung VI beschriebenen Weise kondensiert. Nach
Kristallisation aus Äthylacetat werden
39,9 g (Ausbeute 85%) Boc-Asp(OBzl)-Phe-NH2 (XVII)
vom Schmelzpunkt 141 bis 142°C erhalten.
39,9 g Boc-ASp(OBzl)-Phe-NH2 (XVII)
das in 180 ml Dimethylformamid gelöst ist, werden bei Normaldruck in
Gegenwart von 29 g 10%iger Palladiumkohle hydrogenolysiert.
Nach Kristallisation aus Isopropanol werden
29,0 g (Ausbeute 90%) Boc-Asp-Phe-NH2 (XVIII)
vom Schmelzpunkt 196-198°C erhalten. -50,0° (c=1, DMF).
29,0 g Boc-Asp-Phe-NH2 (XVIII)
werden in 300 ml trockener
1,33N-HCl in Eisessig (HCl/AcOH) gelöst. Nach 25 Minuten
bei Raumtemperatur wird die Lösung unter vermindertem
Druck eingedampft und der Rückstand aus Diäthyläther
kristallisiert, wobei
24,2 g (Ausbeute 100%) H-Asp-Phe-NH2·HCl (XIX)
vom Schmelzpunkt 100-150°C (Zers.)
erhalten werden.
Durch Kondensation von
19,1 g Boc-Met-OH mit 24,2 g H-Asp-PHe-NH2·HCl (XIX)
durch das Äthoxyameisensäureanhydrid
in üblicher Weise und Kristallisation aus Äthylacetat werden
36,0 g (Ausbeute 92%) Boc-Met-Asp-Phe-NH2 (XX)
vom Schmelzpunkt 194-198°C erhalten. Durch Acidolyse von
36,0 g Boc-Met-Asp-Phe-NH2 (XX)
mit HCl/AcOH auf die für die Herstellung
der Verbindung XIX beschriebene Weise werden
31,5 g (Ausbeute 100%) H-Met-Asp-Phe-NH2·HCl (XXI)vom Schmelzpunkt 191-192°C erhalten.
21,5 g Boc-Trp-OH werden mit 31,5 g H-Met-Asp-Phe-NH2·HCl (XXI)
durch das Äthoxyameisensäureanhydrid in
üblicher Weise kondensiert, wobei nach Kristallisation
aus Äthylacetat
44,2 g (Ausbeute 90%) Boc-Trp-Met-Asp-Phe-NH2 (XXII)
vom Schmelzpunkt 199-200°C erhalten
werden. -34,5° (c=1, 95%iges AcOH).
44,2 g Boc-Trp-Met-Asp-Phe-NH2 (XXII)
werden in 450 ml
99%iger Ameisensäure gelöst. Nach 5 Stunden bei Raumtemperatur
wird das Lösungsmittel unter vermindertem Druck
bei 30°C abgedampft und der Rückstand aus Isopropanol
kristallisiert, wobei
33,7 g (Ausbeute 89%) H-Trp-Met-Asp-Phe-NH2 (V)
vom Schmelzpunkt 205-207°C erhalten
werden.
Zu einer Lösung von
in 160 ml wasserfreiem Dimethylformamid
werden nacheinander 24 ml trockene 2,44 N-HCl in
wasserfreiem Tetrahydrofuran und 3,5 ml n-Butylnitrit
bei einer Temperatur von -25°C gegeben. Bei dieser Temperatur
wird 25 Minuten gerührt, worauf 13,1 ml N-Methylmorpholin
und anschließend eine Lösung von
14,8 g H-Trp-Met-Asp-Phe-NH2 (V)
in einem Gemisch von 120 ml
wasserfreiem Dimethylformamid und 40 ml Hexamethylphosphoramid,
das 2,8 ml N-Methylmorpholin enthält, zugesetzt
werden. Das Reaktionsgemisch wird 4 Stunden bei
-10°C stehen gelassen. Nach dem Abdampfen der flüchtigen
Lösungsmittel unter vermindertem Druck wird das Produkt
durch Verdünnen mit kalter wäßriger Lösung von Zitronensäure
in Wasser ausgefällt. Das rohe Material ergibt
nach Kristallisation aus Methanol
vom Schmelzpunkt 201-206°C (Zers.).
werden bei Raumtemperatur in 400 ml Wasser gelöst,
indem mit 1N-NaOH auf pH 10,5 eingestellt wird. 72 g
des Komplexes von Pyridin und Schwefeltrioxid werden
portionsweise zugesetzt, während 5 Stunden gerührt und
der pH-Wert mit 1N-NaOH bei 9-11 gehalten wird. Nach
Neutralisation der alkalischen Lösung mit festem CO2 wird
die Lösung durch Entfernen des Lösungsmittel unter
vermindertem Druck zur Trockne eingedampft und der Rückstand
in Dimethylformamid aufgenommen und zur Entfernung
der unlöslichen anorganischen Salze filtriert. Das Filtrat
wird erneut unter vermindertem Druck zu einem öligen
Rückstand eingedampft. Das Produkt wird dann in 400 ml
eines 1 : 1-Gemisches von Dimethylformamid und Wasser
gelöst und bei 0°C mit dem Ionenaustauscherharz "Dowex 50W"®
(H⁺) 5 Minuten unter Rühren behandelt. Das Gemisch wird
filtriert und mit Diäthylamin alkalisch gemacht. Das
Lösungsmittel wird durch Eindampfen auf ein kleines
Volumen unter vermindertem Druck entfernt und das Produkt
(13,5 g) durch Zusatz von Diäthyläther ausgefällt. Durch
Entsalzen mit Hilfe einer Säule des Adsorptionsmittels
"Sephadex G 10"® werden
als Diäthylaminsalz erhalten. -22° (c=1, DMF).
Claims (1)
- Verfahren zur Herstellung des Decapeptids Ceruletid der Formel und seiner Salze mit pharmazeutisch unbedenklichen Basen, worin alle Aminosäuren die L-Konfiguration haben, dadurch gekennzeichnet, daß man das Decapeptid der Formel in Wasser bei einem pH-Wert von 9,5-10,5 löst und im wäßrigen Medium mit einem Sulfatierungsmittel der Formel B : SO3, worin B: eine tertiäre Base bedeutet, bei einem pH-Wert zwischen 8,5 und 12,5 umsetzt, das hierbei gebildete Produkt in bekannter Weise isoliert und gegebenenfalls in ein Salz mit einer pharmazeutisch unbedenklichen Base überführt.
