Die Erfindung betrifft Resorcine, insbesondere 2-Substitulert~5-alkylresorcine
der nachstehenden allgemeinen Pormel
in der R. ein Wasserstoffatom oder einen Nieder-alkanoylrest,
Rp einen gerad- oder verzweigtkettigen Alkylrest mit 1 bis
Kohlenstoffatomen und X ein Stickstoffatom oder ein methylsubstituiertes Kohlenstoffatom (C-Methyl) bedeuten.

Unter "Hleder-alkanoylresten" sind liier von Monocarbonsäuren
abgeleitete einwertige, gerad- oder verzweigtkettige, gesättigte
aliphatische Reste mit 2 Ms 6 Kohlenstoffatomen, wie
die Acetyl-, Propionyl-, a-Methylpropionyl-, Butyryl- oder
Hexanoylgruppe,zu verstehen.
Der Ausdruck "Alkylreste" "bezieht sich hier auf gerad- oder
verzweigtkettige, einwertige gesättigte aliphatische Reste mit 1 "bis 20 Kohlenstoffatomen, wie die Methyl-, n-Amyl-,
n-Hexyl-r 2-Heptyl-, n-Heptyl-, 3-Methyl-2-octyl-, n-Octyl-,
2-lTonyl-* 2-Tetradecyl-, n-Hexadecyl- oder 2-Eikonsanylgruppe.
Die erfindungsgemäßen Resorcine werden gemäß folgendem
Reaktionsschema hergestellt:

Die Benzopyranone (I) werden in Tetrahydrofuran mit Methylmagnesiumbromid
zu den Alkoholen (II) umgesetzt, welche
ihrerseits in Pyridin mit Essigsäureanhydrid zu den Diacetylderivaten umgesetzt werden. Die anschließende Dehydratisierung mit Thionylchlorid in Pyridin bei -10 bis O0C liefert
die Isopropenylderivate (III). Durch Umsetzung der Derivate (III) mit flüssigem Ammoniak bei Raumtemperatur in einer
Hochdruckbombe erhält man die Resorcine (IV). Durch Hydrierung der Isopropenylderivate (III) mit 5^iger Palladium-Aktivkohle in Äthylacetat werden die Isopropylderivate (V)
erhalten, welche in einer Hochdruckbombe bei Raumtemperatur mit flüssigem Ammoniak zu den deacetylierten Derivaten (71) umgesetzt werden.
ihrerseits in Pyridin mit Essigsäureanhydrid zu den Diacetylderivaten umgesetzt werden. Die anschließende Dehydratisierung mit Thionylchlorid in Pyridin bei -10 bis O0C liefert
die Isopropenylderivate (III). Durch Umsetzung der Derivate (III) mit flüssigem Ammoniak bei Raumtemperatur in einer
Hochdruckbombe erhält man die Resorcine (IV). Durch Hydrierung der Isopropenylderivate (III) mit 5^iger Palladium-Aktivkohle in Äthylacetat werden die Isopropylderivate (V)
erhalten, welche in einer Hochdruckbombe bei Raumtemperatur mit flüssigem Ammoniak zu den deacetylierten Derivaten (71) umgesetzt werden.

Die erfindungsgemäßen Verbindungen eignen sich als Analgetika,
Tranquilizer, sedativ-hypnotische Mittel und Antikonvulsiva.
Sie entfalten ihre Wirkung im allgemeinen bei Verabreichung in täglichen Dosen von 1 bis 10 mg/kg Körpergewicht. Ihre
analgetische Wirkung wurde mit Hilfe des Standard-Mäusekrampf- bzw.-krümmtests (standard mouse writhing test)[Whittle,
Brit. J. Pharmacol., 22 (1964), Seite 29öJ festgestellt und
durch den Heißplattenversuch [G-. ¥oolfe»und A.J. McDonald,
J. Pharmacol. Exper. Therap., 80 (1944), Seite 300] sowie
den Eatten-Schwanzschlagetest (rat tail flick test) bestätigt.

Die nachstehenden Beispiele sollen die Erfindung näher erläutern,
ohne sie jedoch zu "beschränken.
Beispiel 1

Ein Gemisch von 3,42 g 2-Benzyl-10-hydroxy-8-(3-methyl-2-octyl)-5-O3:o-1,2,3»4-tetrahydro-5H-[i]benzopyrano[4,3-cjpyridin,
1 g 10biger Palladium-Aktivkohle und 120 ml Xylol
wird unter Rühren 23 Std. unter Rückfluß gekocht. Anschliessend filtriert man den Katalysator vom Gemisch ab, dampft
das Eiltrat im Vakuum zur Trockene ein und kristallisiert den Rückstand aus Acetonitril um. Dabei erhält man die reine
Verbindung vom Ep. 122 bis 1230C

Eine Lösung von 16,97 g (0,05 Mol) 1O-Hydroxy-8-(3-methyl-2-octyl)-5-oxo-5H-[i]benzopyrano[4»3-e]pyridin
in 125 ml Tetrahydrofuran wird allmählich unter Rühren in eine lösung
von 125 ml 3m Methylmagnesiumbromid in Äther und 125 ml
Tetrahydrofuran eingetragen, so daß die erhaltene Lösung schwach unter Rückfluß kocht. Nach der Zugabe wird das
Reaktionsgemisch 90 Min. unter Rückfluß gekocht, abgekühlt und mit 200 ml gesättigter Jjamoniumchloridlösung zersetzt.
Anschließend extrahiert man zweimal mit Tetrahydrofuran und trocknet die Extrakte über wasserfreiem Magnesiumsulfat.
Beim Eindampfen im Vakuum erhält man die gewünschte Verbindung vom Pp. 82 bis 870C.

5-(3-methyl-2-octyl)-resorein

7»65 g Essigsäureanhydrid werden unter Rühren in eine eisbadgekühlte
Lösung von 9>73 g 2-{4-[(1-Hydroxy-1-methyl)-äthylJ-3-pyridyl}-5-(3-methyl-2-octyl)-resorcin
in 40 ml wasserfreiem Pyridin eingetropft. Das Gemisch wird 1 Std. "bei der Eisbadtemper&tur und dann über Nacht "bei Raumtemperatur
gerührt. Anschließend dampft man das Gemisch im Vakuum ein, gießt den Rückstand in 300 ml Eiswasser
und nimmt das gummiartige Material in Äther auf. Die ütherlö'sung wird zweimal mit Wasser gewaschen, über wasserfreiem
Natriumsulfat getrocknet und- im Yakuum eingedampft. Dabei erhält man die gewünschte Verbindung» die
ohne weitere Reinigung in der nächsten Stufe eingesetzt wird.

5 ml ThionylChlorid werden unter Rühren in eine Lösung von
1 , 27 g 1,3-Diacetyl-2-{4-[1-hydroxy-1-methyl)-äthyl]-3-pyridylj-5-(3-methyl-2-octyl)-resorcin
in 40 ml Pyridin, die im Eisbad auf -100C gekühlt wird» eingetropft. Das
Gemisch wird gerührt und innerhalb von 90 Min. auf O0C
erwärmen gelassen. Anschließend gießt man das Gemisch in Eiswasser ein» extrahiert mit Äther» wäscht die Extrakte mit
Wasser und trocknet sie über wasserfreiem natriumsulfat.
Der "beim Eindampfen in Vakuum erhaltene Rückstand wird an
einer ELorisiffisäule (149 "bis 250 μ; 60 bis 100 mesh) unter
Verwendung von abgestuften Methanol/Benzol-Iiösungsmittelmischungen
Chromatograph!ert; dabei erhält man die
reine Verbindung (Florisir— = Magnesiumsilikatgel).

Eine Lösung von 4,37 g 1»3-Diacetyl-2-[4-isopropenyl-3-pyridyl]-5-(3-methyl-2-octyl)
-resorcin in 50 ml !Toluol wird in eine Hochdruckbombe mit 40 ml flüssigem Ammoniak
gegeben. Das Gemisch wird über Nacht bei Raumtemperatur geschüttelt. Anschließend läßt man das überschüssige
Ammoniak entweichen, reibt den Rückstand mit Wasser an und extrahiert mit Chloroform. Die Extrakte werden zweimal
mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und im Vakuum eingedampft. Der Rückstand wird aus
Äther/Petroläther (30 bis 600C) umkristallisiert; dabei
erhält man die reine Verbindung vom Ip. 128 bis 1290C.

Eine Lösung von 6 g 1, 3-Dia.cetyl-2-[4-isopropenyl-3-pyridyl]-5-(3-methyl-2-octyl)-resorcin
in 200 ml Ithylacetat wird in einem Parr-Apparat während 90 Min. in G-egenwart von 1,2g
5/£iger Palladium-Aktivkohle hydriert. Anschließend filtriert
man die Lösung, fügt nochmals 1,2 g 5fo±ge Palladium-Aktivkohle
hinzu und hydriert die Mischung 4 Std. Danach filtriert man den Katalysator ab und dampft das Piltrat im Vakuum zur
gewünschten Verbindung ein.

des Katalysators durch eine Florisir-^-Säule laufen gelassen.
Durch Eindampfen des Elutionsmittels im Vakuum erhält
man die gewünschte Verbindung.

mit Photozellen ausgestatteten Bewegungsaktivitätskammern bestimmt. Man verabreicht Gruppen von jeweils neun Hatten die
betreffende Verbindung oral 1 Std. vor dem lest und danach Methamphetamin (Desoxyn) in einer Dosis von 3 mg/kg (subkutan).
Dann werden die Hatten in die Kammer gegeben. Die Veränderungen der Bewegungsaktivität (motorischen Aktivität) werden
als Zählwerte der Photozellen registriert und den an (nur) mit Desoxyn behandelten Vergleichstieren erzielten Resultaten gegenübergestellt.

Die Prüfung der analgetischen Wirkung der erfindungsgemäßen Verbindungen erfolgt nach den Essigsäure-Krampf- bzw. -Krümmtest,
wie er von B.A. Whittle in Brit. J. Pharmacol 22 (1964),
Seite ZA-Sr beschrieben wird. Gruppen von weiblichen Swiss-Webster-Mäusen
werden orale Dosen des Wirkstoffs oder einer Vergleichsprobe verabreicht. Auf die Verwendung des intravenösen
Farbstoffs wird verzichtet. Eine Stunde später wird intraperitoneal 0,4 ml 0,5$ige Essigsäure verabreicht. Man
bestimmt die Anzahl der innerhalb von 20 Min. erfolgenden Krümmungen bzw. Krämpfe (Beginn der Zählung 5 Min. nach der
Essigsäureinjektion). Die analgetische Wirkung wird als prozentuale Hemmung des Krümmens gegenüber den Vergleichsproben oder als EDp-,w wiedergegeben.
Beim Test der analgetischen Wirkung nach der Ratten-Schwanzschlagemethode
[vgl. F.E. D'Amour und D.L. Smith, J. Pharmacol

Exp. Ther. 72 (1941), Seite 74] wird folgende Abwandlung
vorgenommen. Gruppen von männlichen Sprague-Dawley-Ratten mit einem Gewicht von jeweils 140 bis 170 g werden durch
Schwarz färbung der Schwänze mit Tusche präpariert. Dann wird die Wärme eines Strahlers so auf die Schwänze fokusiert
und eingestellt, daß die normalen Schmerzschwellen (Zeit in Sekunden bis zum Erfolgen einer Ausweichreaktion
oder eines Schwanzschlages) im Bereich von 8 bis 10 Sekun den liegen. Anschließend verabreicht man die zu testende
Verbindung oral und bestimmt die Schmerzschwellen der Ver
suchstiere stündlich. Die analgetischen Wirkungen sind in Tabelle I als prozentuale Erhöhung über die anfängliche
Schmerz schwelle 2 Std. nach der Verabreichung oder als Werte angegeben.

Audiogener Anfalltest . , (an Mäusen)
Man verwendet bei diesem Test männliche Mäuse vom O'Grady-St^TTiTn
mit einem Gewicht von jeweils 14 bis 16 g» welche speziell für die Empfänglichkeit gegenüber audiogenen Anfällen
gezüchtet wurden. Der audiogene Apparat besteht aus einem Holzkasten, der einen Metallbehälter mit zwei im
oberen Teil angebrachten Türklingeln einschließt. Haeh der Wirkstoffverabreichung gibt man die Versuchstiere in die
audiogene Kammer, betätigt die Klingeln 1 Min. lang und beobachtet die Krämpfe bzw. Zuckungen der Tiere [vgl. N.P.
Plotnikoff und D.M. Green, J. Pharmacol. Exp. Ther., 119 (1957), Seite 294].

Die Erfindung "betrifft ferner Arzneimittel, welche als Wirkstoff
komponente mindestens eine erfindungsgemäße Verbindung in Kombination mit einem pharmakologisch verträglichen Träger
oder Verdünnungsmittel enthalten. Die Verbindungen der Erfindung weisen sowohl orale als auch parenterale Wirksamkeit
auf und können zu Präparaten für orale, parenterale oder rektale Zwecke formuliert werden.
Beispiele für feste Verabreichungsformen für orale Zwecke sind Kapseln, Tabletten, Pillen, Pulver und Granulate. Bei
solchen Pestpräparaten wird der Wirkstoff bzw. die aktive Verbindung mit mindestens einem inerten Verdünnungsmittel,
wie Rohrzucker, Milchzucker oder Stärke, vermischt. Derartige Präparate können ferner gemäß der üblichen Praxis
zusätzliche Substanzen (außer inerten Verdünnungsmitteln), wie Gleitmittel (z.B. Magnesiumstearat), Süßstoffe und
Geschmacks- bzw. Geruchskorrigentien und Farbstoffe (flavoring agents), enthalten. Tabletten und Pillen können
außerdem mit magensaftresistenten, dünndarmlöslichen Überzügen
(enteric coatings) versehen werden.
Beispiele für flüssige Verabreichungsformen für orale Zwecke sind pharmakologisch verträgliche Emulsionen, lösungen,
Suspensionen, Sirups und Elixiere, welche die üblicherweise verwendeten inerten Verdünnungsmittel, wie Wasser, enthalten.
Außer inerten Verdünnungsmitteln können derartige Präparate auch Hilfsstoffe, wie Netzmittel» Emulgier- und Suspendiermittel,
Süßstoffe, Parfümierungsmittel, Geschmacksbzw. Geruchskorrigentien und Farbstoffe enthalten.
Beispiele für erfindungsgemäße Arzneimittel, welche sich zur parenteralen Verabreichung eignen, sind sterile, wäßrige
oder nicht-wäßrige lösungen, Suspensionen oder Emulsionen. Beispiele für nicht-wäßrige Lösungsmittel oder Medien sind
Propylenglykol, Polyäthylenglykol, pflanzliche Öle, wie Olivenöl, und injizierbare organische Ester, wie Esteroleat (Öl-

säureester). Solche Präparate können ferner Hilfsstoffe, wie
Konservierungs-, Netz-, Emulgier- und Dispergiermittel, enthalten.
Die Präparate können sterilisiert werden, beispielsweise durch Filtration durch ein Bakterienfilter, durch Einverleibung
von Sterilisierungsmitteln, durch Bestrahlen oder durch Erhitzen. Man kann sie auch in Form steriler Festpräparate
erzeugen, welche unmittelbar vor dem. Gebrauch in sterilem Wasser oder einem anderen sterilen, injizierbaren
Medium gelöst werden können.
Präparate für die rektale Verabreichung sind Suppositoryen,
welche außer dem Wirkstoff Bindemittel, wie Kakaobutter oder ein Suppositorienwachs, enthalten können.
Die Wirkstoffdosis in den erfindungsgemäßen Arzneimitteln
kann variiert werden. Es ist jedoch notwendig, daß die Wirkstoffmenge so bemessen wird» daß man eine geeignete Yerabreichungsform
erhält. Die jeweils gewählte Dosis hängt vom angestrebten therapeutischen Effekt, vom Verabfolgungsweg
und von der Behandlungsdauer ab.

Claims (24)
- Patentansprüche1/ί 2-Substituiert-5-alkylresorcine der nachstehenden allgemeinen Formelin der R, ein Wasserstoff atom oder einen Meder-alkanoylrest, R2 einen gerad- oder verzweigtkettigen Alkylrest mit 1 bis 20 Kohlenstoffatomen und X ein Stickstoffatom oder ein methylsubstituiertes Kohlenstoffatom (C-Methyl) bedeuten«
- 2. Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß Ε., ein Wasserstoff atom, R2 einen Alkylrest und X ein Stickstoffatom bedeuten.
- 3. 2-{4-[(i-Hydro2:y-1-methyl)--äthyl]-3-pyridyl}-5-(3-methyl-2-octyl)-resorcin, eine Verbindung nach Anspruch 2.

- 6. Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß R1 -C(O)CH5, R2 einen Cg-Alkylrest und X ein Stickstoffatom bedeuten.
- 7. 1»3-Diacetyl-2-[4-isopropenyl-3-pyridylJ-5-(3-methyl-2-octyl)-resorcin, eine Verbindung nach Anspruch 6.
- 8. Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß die Reste R1 Jeweils ein Wasserstoffatom, R2 einen Cg-Alkylrest und X ein Stickstoffatom bedeuten.
- 9. 2-[4-Isopropenyl-3-pyridyl]-5-(3-methyl-2-octyl)-resorcin, eine Verbindung nach Anspruch 8.
- 10. Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß R1 -C(O)GH-, R2 einen Cg-Alkylrest und X ein Stickstoffatom bedeuten.
- 11. 1,3-3)iacetyl-2-[4-isopropyl-3-pyridyl ]-5-( 3-methyl-2-octyl)-resorcin, eine Verbindung nach Anspruch 10.
- 12. Verbindungen nach Anspruch 1, dadurch gekennzeichnet» daß die Reste R.. Jexveils ein Wasserstoff atom, Rp einen Cg-Alkylrest und X ein Stickstoffatom bedeuten.
- 13. 2-[4-Isopropyl-3-pyi*idylJ-5-(3-methyl-2-octyl)-resorcin, eine Verbindung nach Anspruch 12.

- .18. Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß die Reste R1 jeweils -C(O)CH3, R2 einen Cg-Alkyl~ rest und X C-CH^ bedeuten.
- 19. 1»3-Diacetyl-2-[(2-isopropenyl-5-methyl)-phenyl]-5-(3-methyl-2-octyl)-resorcin, eine Verbindung nach Anspruch 18. ,
- 20. Verbindungen nach Anspruch 1, dadurch gekennzeichnet» daß die Reste R1 jeweils -C(O)CH5, R2 einen Cg~Alkylrest und X C-CH, bedeuten.
- 21. 1,3-Macetyl-2-[(2-i sopropyl-5-methyl)-phenyl ]-5-( 3-methyl-2-octyl)-resorcin, eine Verbindung nach Anspruch 20.
- 22. Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß die Reste R. jeweils ein -Wasserstoffatom, R2
einen Cq-Alkylrest und X C-CH^ bedeuten. - 23. 2-[ (2-Isopropyl-5-methyl)-phenyl]~5-(3-methyl-2-octyl)· resorcin, eine Verbindung nach Anspruch 22.
