Description
Die Erfindung "betrifft ein Antitumor-Mittel, welches aus
p-Di(2-chloräthyl)amino-L-phenylalanin oder Ή, IT-Bis
(2-chloräthyl)p-phenylendiamin "besteht, die jeweils über
Peptidbindungen an einen tumorspezifischen Antikörper ge bunden sind.
Erfindungsgemäß werden also cytotoxisch wirkende Mittel,
sogenannte Cytotoxlka der !Formel
oder
in Gegenwart von einem in Wasser löslichen Carbodiimid
mit tumorspezifischen Antikörpern umgesetzt, so daß sich Peptidbindungen zwischen dem cytotoxischen Mittel und dem
tumorspezifischen Antikörper ausbilden.
Spezifische Antiseren werden mit üblichen Techniken der

Immunisation und Sammlung von Seren hergestellt. Diese
Antiseren v/erden dann, beispielsweise, von normalen Zellen absorbiert, um die unerwünschten Antikörper gegen andere
als die Zielzellen zu entfernen. Die verbleibenden Antikörper sind dann vorwiegend gegen die Zielzellen der
Wahl, beispielsweise Tumorzellen gerichtet. Ihre anschließende Bindung an ein toxisches Mittel ergibt ein
wirksames spezifisches cytotoxisches Mittel, welches gegen eine bestimmte ausgewählte Zielzelle gerichtet werden kann.
Tumorspezifische Antikörper werden beispielsweise gegen neoplastische Gewebe gezüchtet, und zwar mit Hilfe der
Techniken, die von Ghose et al»jBritish Medical Journal
(1972) 3, 495-499 beschrieben worden sind. So werden in einer Ausführungsform Antikörper gegen Mäuse-E14-lymphoma
(BL4 Immunoglobulin) mit p-Di(2~chloräthyl)amino-Ir-phenylalanin
in Gegenwart von l-Äthyl-3(3-dimethylaminopropyl)-carbodiimid-Hydrochlorid
umgesetzt, wodurch man einen cytotoxischen, tumorspezifischen Antikörperkomplex gewinnt,
in dem etwa 13 Moleküle des cytotoxischen Mittels pro Molekül Antikörper vorliegen.
Antikörper gegen Neoplasmen von lymphatischen oder hämatopoetischen
Geweben bei Lymphosarcom, chronischer lymphatischer leukämie, Hodgkin's Krankheit, Karzinomen der
Eierstöcke, der Brüste und der Hoden sowie weitere Epithelialgewebe und Melanome sind tumorspezifische Antikörper,
die erfindungsgemäß in Betracht gezogen sind. Die Globulanfraktion, die diesen Geweben entgegengerichtet und
von normalem Gewebe absorbiert ist, wird an ein cytotoxisehes Mittel wie p-Di(2-chloräthyl)amino-L-phenylalanin
gebunden, und zwar mit Hilfe von l-Äthyl-3(3-dimethylaminopropyl)carbodiimide
Es ist im Rahmen der vorliegenden Erfindung wesentlich, daß* nachdem der Komplex gebildet worden ist, der tumor-

spezifische Antikörper seine Spezifizität und das cytotoxisehe
Mittel seine Fähigkeit zur Alkylierung behalten. Beide wesentlichen Kriterien sind bei der Carbodiimid-induzierten
Peptidbindungsbildung erfüllt, bei welcher etwa
5-15 Moleküle eines der oben erwähnten cytotoxischen Mittel an einen geeigneten tumorspezifischen Antikörper
gebunden werden. Durch die Erfüllung dieser wesentlichen Kriterien ist es möglich, daß der tumorspezifische Antikörper
die gebundenen eytotoxischen Mittel an die Stelle bringt, an der die gewünschte Wirkung eintreten soll.
Die Dosierung ändert sich je nach dem tumorspezifischen
Antikörper, der Menge und der Art des an diesen gebundenen cytotoxischen Mittels, dem Ausmaß des Wachstums der
ITeoplasmen sowie der individuellen Toleranz gegenüber dem
tumorspezifischen Antikörper-Cytotoxicum-Komplex. Da toxische Nebenwirkungen des cytotoxischen Mittels durch die
Bindung desselben an den tumorspezifischen Antikörper weitgehend vermindert sind,können hohe Dosen des gebundenen
cytotoxischen Mittels ohne Schaden verabreicht werden. Die Dosierungen des p-Di(2-chloräthyl)amino-L-phenylalanin, die
in "Cuttings Handbook of Pharmacology", 4. Ausgabe, 1969, Appleton Century Crafts, auf Seite 139 beschrieben sind,
können um das zwei- bis fünffache erhöht werden.
Die folgenden Beispiele sollen zur weiteren Erläuterung der vorliegenden Erfindung dienen.
Es wurde ein Antiserum in Kaninchen gegen EL4-Tumorzellen
(transplantierbare lymphomzellen von C57Bl/6-Mäusen) gezüchtet.
Die EL4-Zellen, die den Mäusen entnommen worden sind, wurden intravenös Kaninchen injiziert f und zwar in
drei Dosen mit 10 Zellen pro Dosis in Abständen von 10
Tagen.

Zehn Tage nach der letzten Dosis wurde das Kaninchen ausgeblutet
und das Serum aufgefangen. Das Serum wurde 30 Minuten auf 56° C erhitzt, um Nebenwirkungen zu zerstören.
Das aufgefangene Serum wurde von normalen Mäusezellen absorbiert, wodurch die tumorspezifischen Antikörper entstanden.
Das Serum wurde von einer Menge Mäusemilzzellen, die wenigstens einige von Mäusen der Art C57B1/6 oder ei-
.absorbiert.
ner ähnlichen Art enthielt , / Es wurden wiederholte Absorptionen mit dem Äquivalent von zehn Milzen auf einen Milliliter
Serum in 16 Stunden bei 4° C unter Rühren durchgeführt, bis das Serum gegen normale C57Bl/6-Stamm-Zellen in
üblichen cytotoxischen Prüftests keine Reaktivität mehr zeigte. In demselben Versuch und auch bei indirekten Immunofluorescenz-
oder Agglutinationstests wurde dasselbe Serum mit EL4-Tumorzellen umgesetzt, um die Tumorspec if izität des
hergestellten Serums zu zeigen.
Die Globulinfraktion wurde aus dem Serum durch Zugabe von
gesättigtem Ammoniumsulfat bis zu einer Endkonzentration
von 40 f> ausgefällt. Das ausgefällte EL4-Globulin (Ig)
wurde in Phosphat-gepufferter Salzlösung wieder gelöst, so daß sich eine schließliche Proteinkonzentration, gemessen
durch die Biuret-Reaktion, von 30- 50 mg pro Milliliter
ergab.
Die Peptidbindungsnzwischen einer Amino- oder Carboxylgruppe,
die in bestimmten cytotoxischen Mitteln bzw. Drogen vorhanden sind, und den entsprechenden Gruppen an den Antikörpermolekülen
lassen sich in Gegenwart von Carbodiimid in der nachfolgend beschriebenen Weise erzeugen.
Es wurden folgende Reagenzien hergestellt: (A) 10 ml E14-Immunoglobulinlösung

auf pH 6,5 mit HCl; (B) entweder (I) 57 mg Ν,ΪΤ-Bis
(2-chloräthyl)p-ph.enylendiamin-Hyärochloriä in 2 ml
Dioxan oder ( II) 57 mg p-Di(2-Chloräthyl)amino-Iiphenylalanin, gelöst in 0,8 ml einer Säure: Äthanolmischung
( 92 Jfi Äthanol, enthaltend 2 # Gew./Vol. HCl),
verdünnt mit einer 60 $igen Propylenglycol-PufferlÖsung, welche 1,2 fo Gew./Vol. Dikaliumhydrogenphosphat enthielt,
deren End-pH bei 6-6,5 lag; (C) 114 mg l-Äthyl-3
(3-dimethylaminopropyl)-carhodiimid-HCl, gelöst in 0,5 ml
Salzlösung. .
Die Reagenzien A,B und C wurden rasch fünf Sekunden lang
vermischt, worauf die Reaktion durch Zugabe von. 6 ml 1m Natriumacetat von pH 6,5 gestoppt wurde. Die entstandene
Lösung wurden gegen dreimal wechselnde 0,3 $ige Hatriumzitratlösung
24 Stunden bei 40C dialysiert, um nicht gebundene
Droge zu entfernen.
Der Drogen-Antikörper-Komplex wurde auf seinen Proteingehalt und seine Alkylierungswirkung analysiert. Der Proteingehalt
wurde mit Hilfe der üblichen Biuret-Versuchsanordnung
unter Verwendung von Schweineserum-Albumin als Standard gemessen. Die Alkylierungswirkung wurde nach der Epstein-Methode
festgestellt ( vergleiche J. Epstein, R.W. Rosenthal und J.R. Ess, Anal. Chem 27, 1434 (1955).
Das fertige Präparat des N,N-Bis(2-chloräthyl)p-phenylendiamin-Antikörper-Eomplexes
enthält 12,2 mg EL4-Immunoglobulin pro Milliliter und 122 ^g ir,lT~Bis(2-chloräthyl)
p-phenylendiamin. Dies bedeutet ein Verhältnis von .etwa
5,0 Drogenmoleküle pro Molekül Ig. In entsprechender Weise enthielt die fertige lösung des p-Di(2-chloräthyl)amino-L-phenylalanin-Antikörper-Komplexes
13,9 mg EIi4-Immunoglobulin pro Milliliter und 380 ^.g p-Di(2-chloräthyl)amino-1-phenylalanin,
was einem Verhältnis von etwa 13s 6 Drogenmolekülen
pro Molekül Antikörper entspricht.

Es ist infolgedessen möglich, die beschriebenen Drogen mit Hilfe einer Peptidbindung Covalent an tumorspezi- .
fische Antikörper zu "binden. Die Wirkung derartiger
Drogen-Antikörper-Komplexe ist im folgenden Beispiel 2 gezeigt.
Verwendung der Drogen-Antikörper-Komplexe.
Die in der vorstehend beschriebenen Weise hergestellten Drogen-Antikörper-Komplexe wurden dazu verwendet, Mause
gegen die lethale Wirkung der Injektion von EL4-Tumorzellen
zu schützen.
Gruppen von Mäusen wurden intraperitoneal etwa 10,000
lethale Dosen ( 5 x 10^) Tumorzellen injiziiert, die dem
Unterleib zuvor infiziierter Mäuse entnommen worden waren.
Vierundzwanzig Stunden nach der Injektion der Zellen erhiel ten die Tiere die erste von vier täglichen Dosen eines
therapeutischen Mittels oder eines Kontrollmittels entsprechend den folgenden Angaben.

Die Drogen-Antikörper-Komplexe gemäß vorliegender Erfindung zeichnen sich also durch eine erheblich größere Schutzwirkung
auf Mäuse aus als entweder das rmmuno-EL4-globulin allein oder als die Droge in Bindung an ein nicht spezifisches
Globulin. Darüberhinaus sind die äquivalenten Dosen der toxischen Mittel allein für Mäuse lethal, während die
maximal verträgliche Dosis der Droge bei alleiniger Verwendung nicht ausreicht, den Ausgang unter den Bedingungen
der vorliegenden Versuchsanordnung nennenswert zu beeinflussen.

Claims (9)
1. Antitumor-Mittel, "bestehend aus p-Di(2-chloräthyl)amino-L-phenylalanin
oder N,N-Bis(2-chloräthyl)p-phenylendiamin-Hydrochlorid
als cytotoxischem Mittel, welches durch Peptidbindungen
an einen tumorspezifischen Antikörper gebunden ist.
2. Antitumor-Mittel "nach Anspruch 1, dadurch gekennzeichnet, daß 5-15 Moleküle des cytotoxischen Mittels pro Molekül
des tumorspezifischen Antikörpers gebunden sind.
3. Antitumor-Mittel nach Anspruch 1, dadurch gekennzeichnet, daß p-Di(2-chloräthyl)amino-L-phenylalanin über Peptidbindungen
an gegen E14-Iiymphom spezifische Antikörper gebunden
ist.
4. Antitumor-Mittel nach Anspruch 1, dadurch gekennzeichnet, daß N,üT-Bis(2-chloräthyl)p-phenylendiamin über Peptidbindungen
an gegen EL4-lymphom spezifische Antikörper gebunden
ist.
5. Verfahren zur Herstellung des Antitumor-Mittels gemäß
Anspruch 1, dadurch gekennzeichnet, daß man ein cytotoxisches Mittel mit einem tumorspezifischen Antikörper in
Gegenwart eines in Wasser löslichen Carbodiimids umsetzt, so daß es zur Ausbildung von Peptidbindungen zwischen dem cytotoxischen Mittel und dem tumorspezifischen Antikörper kommt.
Anspruch 1, dadurch gekennzeichnet, daß man ein cytotoxisches Mittel mit einem tumorspezifischen Antikörper in
Gegenwart eines in Wasser löslichen Carbodiimids umsetzt, so daß es zur Ausbildung von Peptidbindungen zwischen dem cytotoxischen Mittel und dem tumorspezifischen Antikörper kommt.
6. Verfahren nach Anspruch 5, dadurch gekennzeichnet, daß das cytotoxische Mittel aus p-Di(2-chloräthyl)amino-L-phenylalanin
besteht.

7. Verfahren nach Anspruch 5, dadurch gekennzeichnet, daß das cytotoxische Mittel aus ΪΓ,Ν-Bis(2-chloräthyl)p-phenylendiamin-Hydrochlorid
besteht.
8. Verfahren nach Anspruch 5, dadurch gekennzeichnet, daß in dem Antitumor-Mittel 5 "bis 15 Moleküle des cytotoxischen
Mittels an ein Molekül des tumorspezifischen Antikörpers
gebunden sind.
9. Verfahren nach Anspruch 5, dadurch gekennzeichnet, daß man als Carbodiimid 1-A"thyl-3-(3-dimethylaminopropyl)-carbodiimid-Hydrochlorid
verwendet.
