Die Verbindungen der Erfindung können durch Reduktion der ent sprechenden Verbindungen der allgemeinen Formel II
(II)
in der R , R und R·^ die in Anspruch 1 angegebene Bedeutung haben,
oder ihrer Salze mit Säuren, hergestellt werden.
Als Reduktionsmittel werden vorzugsweise Lösungen eines Alkalimetalls
in einem Alkohol oder Metallhydride eingesetzt. Die Reduktion kann jedoch auch elektrolytisch erfolgen.
Besonders "bevorzugte Beispiele für verwendbare Reduktionsmittel
sind Metallhydride, wie Lithiuraaluminiumhydrid, Natriumaluminium-

diäthyldihydrid und Natrium-bis-(2-methoxyäthoxy)-aluminiumhydrid,
die in einem inerten organischen Lösungsmittel, wie Diäthyläther, Diisopropyläther, Tetrahydrofuran, Dioxan, Äthylenglykoldimethyläther,
Heptan, Hexan, Cyclohexan, Benzol und Toluol, verwendet werden. Die Reduktion kann in einem verhältnismäßig
breiten Temperaturbereich durchgeführt v/erden, beispielsweise bei O0C bis zur Rückflußtemperatur des Reaktionsgemisches.
Ein weiteres Beispiel für ein bevorzugt verwendbares Reduktionsmittel
ist Natriumborhydrid, insbesondere wenn es in Verbindung mit einer Lewis-Säure, wie Aluminiumchlorid, oder nach Aktivierung
der Carboxamidgruppe in den Verbindungen der allgemeinen Formel II mit beispielsweise Triathyloxoniumfluoborat, verwendet
wird. Diboran kann ebenfalls als Reduktionsmittel verwendet werden.
Die Verbindungen der allgemeinen Formel I können auch durch Umsetzung
einer Verbindung der allgemeinen Formel III

in der R und R die vorstehende Bedeutung haben und X eine übliche "leaving-Gruppe", beispielsweise ein Halogenatom, wie
ein Chlor- oder Bromatom, oder eine Sulfonyloxygruppe, wie die
Methansulfonyloxy- oder p-Toluolsulfonyloxygruppe, mit Methyl-

amin oder Dimethylamin in einem inerten organischen Lösungsmittel,
wie Diäthyläther, Diisopropyläther, Tetrahydrofuran, Dioxan, Äthylenglykoldimethyläther, Äthanol, Methanol, Benzol, Toluol
oder Pyridin, gegebenenfalls in Gegenwart eines Säureacceptors hergestellt werden. Spezielle Beispiele für verwendbare Säureacceptoren
sind Pyridin, Picolin, Triäthylamin und Dimethylanilin. Die Umsetzung kann unter Eiskühlung bis zur Rückflußtemperatur
des Reaktionssystems durchgeführt werden.
Die nach den vorstehend beschriebenen Methoden erhaltenen Verbindungen
der allgemeinen Formel I werden nach dem Abtrennen auf übliche Weise gereinigt.
Die Salze der erfindungsgemäßen Verbindungen leiten sich von anorganischen oder organischen Säuren ab. Beispiele für die zur
Salzbildung verwendbaren Säuren sind Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Essigsäure, Oxalsäure, Citronensäure,
Weinsäure, Bernsteinsäure, Fumarsäure und Milchsäure. Mit beispielsweise Methylchlorid oder Methyljodid können auch quartäre
Ammoniumsalze hergestellt werden. Die Salze können auf übliche Weise wieder in die freien Basen überführt werden.
Die Verbindungen der Erfindung sind wertvolle Arzneistoffe. Sie sind Antagonisten von Tetrabenazin und Reserpin und heben die
ZNS-dämpfende Wirkung dieser Verbindungen auf. Ferner besitzen
sie eine stimraungsaufhellende und/oder appetitzügelnde Wirkung. Die Verbindungen der Erfindung können, mit üblichen Trägerstoffen
und/oder Verdünnungsmitteln und/oder Hilfsstoffen vermischt,
auf übliche Weise verabreicht werden. ' _j

Die Beispiele erläutern die Erfindung.
Eine Lösung von 0,30 g Lithiumaluminiumhydrid in 10 ml Diäthyläther
wird unter Eiskühlung mit einer Lösung von 0,813 g 2,2-Diphenylcyclopropancarbonsäure-N,N-dimethylamid in 15 ml
Diäthyläther versetzt. Das Gemisch wird zwei Stunden unter Rückfluß erhitzt und gerührt, sodann abgekühlt, mit 20 ml 1Cprozentiger
Natronlauge versetzt und mit Diäthyläther extrahiert. Der Ätherextrakt wird über Magnesiumsulfat getrocknet und eingedampft.
Ausbeute 0,701 g N,N-Dimethyl-2,2-diphenylcyclopropylmethylamin als Öl. Das Hydrochlorid schmilzt bei 231 bis
233°C
Auf ähnliche Weise werden die folgenden Verbindungen erhalten:
N-Methyl-2,2-diphenylcyclopropylmethylamin,
F. 240 bis 243°C (Hydrochlorid);
N, N-Dimethyl-2,2-di- (p-f luorphenyl) -cyclopropylmethylamin,
F. 186 bis 188°C (Hydrochlorid) und
N-Methyl-2,2-Di-(p-fluorphenyl)-cyclopropylmethylamin,
F. 124 bis 126°C (Hydrochlorid).
Eine Lösung von 10 ml einer 40prozentigen wäßrigen Lösung von Dimethylamin in 10 ml Äthanol wird bei Raumtemperatur mit einer
Lösung von 0,50 £ 2,2-Diphenylcyclopropylmethylchlorid in 10 ml
Äthanol und 1,0 g Triäthylamin versetzt. Das Gemisch wird 32 Stunden auf 35 bis 40°C erwärmt und gerührt. Sodann wird das

Reaktionsgemisch eingedampft, der Rückstand in lOprozentige
Natronlauge gegossen und mit Diäthyläther extrahiert. Der Ätherextrakt wird getrocknet und chromatographiert. Das Eluat
wird eingedampft. Es wird das N,N-Dimethyl-2,2-diphenylcyclo~ propylmethylamin als Öl erhalten. Das Hydrochlorid schmilzt
bei 231 bis 233°C.
Auf ähnliche Weise werden folgende Verbindungen hergestellt:
N-Methyl-2#2-diphenylcyclopropylmethylamin,
F. 240 bis 2430C (Hydrochlorid),
N, N-Dimethyl-2,2-di- (p-fluorphenyl)-cyclopropylmethylamin,
F. 186 bis 1880C (Hydrochlorid) und
N-Methyl-2,2-di-(p-fluorphenyl)-cyclopropylmethylamin,
F. 124 bis 1260C (Hydrochlorid).

Claims (7)
- Γ -7-Patentansprüche 1. Cyclopropylmethylamin-Derivate der allgemeinen Formel I1 2in der R und R Wasserstoff- oder Halogenatome bedeuten undR^ ein Wasserstoffatom oder eine Methylgruppe darstellt, ihre Salze mit Säuren und quartären Ammoniumsalze.
- 2. N,N-Dimethyl-2,2-diphenylcyclopropylmethylamin und seine Salze mit Säuren und quartären Ammoniumsalze.
- 3. N-Methyl-2,2-diphenylcyclopropylmethylamin und seine Salze mit Säuren und quartären Ammoniumsalze.
- 4. N, N-Dimethyl-2,2-di-(p-fluorphenyl)-cyclopropylmethylamin und seine Salze mit Säuren und quartären Ammoniumsalze.
- 5. N-Methyl-2,2-di-(p-fluorphenyl)-cyclopropylraethylamin und seine Salze mit Säuren und quartären Ammoniumsalze.
- 6. Verfahren zur Herstellung der Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß man entv/eder609809/η 995Γ . - β- 7536165(a) eine Verbindung der allgemeinen Formel II12 3
in der R , R und R die in Anspruch 1 angegebene Bedeutunghaben, oder ihr Salz mit einer Säure reduziert oder (b) eine Verbindung der allgemeinen Formel III(IIIin der R und R die vorstehende Bedeutung haben und X eine übliche "leaving-Gruppe" darstellt, mit Methylamin oder Dimethylamin zur Umsetzung bringt und gegebenenfalls die nach (a) oder (b) erhaltene Verbindung in an sich bekannter Weise mit einer Säure in ein Salz oder mit einem Quaternierungsmittel in das quartäre Ammoniumsalz überführt. - 7. Arzneimittel, bestehend aus einer Verbindung nach Anspruch bis 5 und üblichen Trägerstoffen und/oder Verdünnungsmitteln und/oder Hilfsstoffen.L J609809/0995
