Es ist bereits bekannt, daß substituierte Diäther des Glycerins, insbesondere die Glycerinäther des Phenols, therapeutische Effekte
aufweisen. So sind in der DT-PS 730 789 1-0xyaryloxy-3-alkoxypropanole-(2)
als Sedativa und Hypnotica beschrieben, während in der US-PS 2 738 351 substituierte 1,3-Bisphenoxy-propanole-(2) offenbart
sind. Aus der DT-OS 2 120 396 ist außerdem bekannt, daß 1-Phenoxy-3-alkoxy-propanole-(2)
choleretische Eigenschaften besitzen, deren Aktivitäten durch Substitution am Phenyloxyrest variiert werden können,
wie dies in der DT-OS 2 207 254 gezeigt wird. Aus Journ. of Med. Chem.
1972, S. 286 ist bekannt, daß 1-Phenoxy-3-arylpiperazinyl-propanol-(2)-Derivate
auf das zentrale Nervensystem wirken und in der JA-PS 9 000 247 sind 1-0xyzimtsäureäther-Derivate des 3-Alkylamino-propanols-(2)
als ß-adrenergisch wirksame Verbindungen mit nervenblockierenden
Eigenschaften beschrieben.

Es wurde nun überraschenderweise festgestellt, daß bestimmte 1,J-disubstituierte
Propanol-(2)-Derivate und deren Mkotinsäureester starke hypolipämische Eigenschaften besitzen.
Es ist heute hinreichend bewiesen, daß erhöhte Serumlipide (Hypercholesterinämie
und/oder Bypertriglyceridämie) als sogenannte "Risikofaktoren erster Ordnung" die Entwicklung der Atherosklerose beschleunigen.
Atherosklerose, insbesondere atherosklerotisch bedingte koronare Herzkrankheiten sind heute in den hochindustrialisierten Ländern des
Westens zur häufigsten Todesursache aufgerückt.
Aus dieser Erkenntnis ergibt sich die Konsequenz für Prävention und Therapie. Der erkannte Risikofaktor Hyperlipämie muß beseitigt
werden, um eine Verbesserung der Prognose zu gewährleisten. Eingang in die Therapie der Hyperlipämien hat vor allem der 2-(4'-Chlorphenoxy)-2-methylpropionsäureäthylester
(CLOI1IBRAT) gefunden. Es hat sich jedoch gezeigt, daß in vielen Fällen Clofibrat einen nicht
befriedigenden therapeutischen Effekt zeitigt.

Aufgabe der vorliegenden Erfindung war es, neue therapeutisch wertvolle
Verbindungen zur Verfügung zu stellen, die bei sehr guter Verträglichkeit
insbesondere den Triglyceridspiegel und/oder den Gholesterinspiegel des Blutes wirkungsvoll verringern.
Gegenstand der vorliegenden Erfindung sind Isopropanolderivate der

Die Art und Weise, in der die Stellung des Restes -A-E in Formel
(j_) angegeben ist, soll zeigen, daß dieser Eest in Bezug
auf das Bindeglied Y sich in Position J oder 4 befindet.
Die Erfindung betrifft ferner ein Verfahren zur Herstellung neuer 1,5-disubstituierter Propanol-(2)-Derivate der allgemeinen Formel

und das Bindeglied X ein Sauerstoff- oder Schwefelatom bedeutet, wie oben beschrieben, umsetzt und das Reaktionsprodukt zur Herstellung
anderer Carbonylderivate wie folgt weiter verarbeitet.
Zur Herstellung der Säuren nach Formel Q) werden die Ester, vorzugsweise
die Methylester nach Formel QJ, in vorteilhafter Weise mittels wässrig-alkoholischer Alkalilauge verseift. Aus den in alkalischer
Lösung vorliegenden Alkalisalzen können die Säuren nach Formel Q) durch Zugabe von Mineralsäuren, wie z. B. Salzsäure oder Schwefelsäure
in Freiheit gesetzt werden.
Die Herstellung der Salze nach Formel QJ geschieht in der Weise, daß
man die entsprechenden Säuren bevorzugt in wässrig-alkoholischem Milieu mit Basen der Alkali- bzw. Erdalkalireihe, mit Aluminiumbasen, ferner
mit Ammoniak oder anderen therapeutisch . verwendbaren Aminen nach bekannten Verfahren reagieren läßt.
Ester nach Formel (jj werden, soweit deren Herstellung nicht nach dem
oben beschriebenen Prinzip erfolgt, nach Standardverfahren durch Veresterung der Carbonsäuren nach Formel QJ mit dem entsprechenden Alkohol
unter Verwendung eines sauren Katalysators erhalten oder durch Umesterung, wobei bevorzugt die Methylester nach Formel QJ mit einem anderen Alkohol
in Gegenwart eines sauren oder alkalischen Katalysators zur Reaktion gebracht werden.

Ester nach Formel ('i), in der die Eindoglieder X und Y -0-bedeuten
und X auch -S- sein kann, die sich weder durch Veresterung der Carbonsäuren noch durch Umesterung der Hethylester nach Formel (_1_)
gewinnen lassen, werden bevorzugt dadurch hergestellt, daß man die am sekundären Hydroxyl mit einer Schutzgruppe, vorteilhaft mit einer
Acetyl- oder Benzylgruppe versehenen Carbonsäuren nach Formel (j[_)
mit üblichen Halogenierungsmitteln, bevorzugt Thiony!halogeniden,
in die reaktiven Säurehalogenide nach Formel QJ, in der E die
Bedeutung von -CO-Halogen besitzt, überführt, diese mit den entsprechenden
Alkoholen in an sich bekannter Weise umsetzt und anschließend die Schutzgruppe je nach Art hydrolytisch oder hydrogenolytisch selektiv
abspaltet, ohne daß gleichzeitig andere irreversible Veränderungen im Molekül erfolgen.
Verbindungen nach Formel (_1_), in der die Bindeglieder X und Y
-0- bedeuten und A in Position 3 oder 4 eine Äthylengruppe darstellt
und R eine Carboxylgruppe ist oder eine Alkoxycarbonylgruppe der allgemeinen Formel -COOR', wobei R' ein gerader oder verzweigter,
gesättigter Alkylrest mit 1-3 Kohlenstoffatomen bedeutet, lassen sich vor allem dadurch leicht herstellen, daß man die bequem zugänglichen
Verbindungen nach Formel (jj, in denen A in Position 3 oder 4
eine Vinylengruppe ist und R die oben genannte Bedeutung hat, einer
katalytischen Hydrierung unterzieht, wobei vorzugsweise Raney-Mckel
als Katalysator dient.
Verbindungen nach Formel (j_), in der die Bindeglieder X und Y ein Sauerstoffatom
darstellen und X auch -S- sein kann, A in Position 3 oder einen Bindestrich, eine Vinylen- oder Äthylengruppe bedeutet und der
Rest R die Funktion von -CONHOH besitzt, lassen sich aus den Estern
nach Formel (j_) bevorzugt aus den Methylestern, durch Reaktion mit
Hydroxylamin in protischem Milieu herstellen.

Verbindungen nach Formel (jL)» iQ eier A einen Bindestrich darstellt
und R die Bedeutung eines Acetalrestes hat von Verbindungen der allgemeinen Formel (Jd)
und R die Bedeutung eines Acetalrestes hat, können durch Umsetzung
in der A in Position 3 oder 4 einen Bindestrich bedeutet und R ein
Dimethoxymethyl—, Diäthoxymethyl— oder ein 1,3—Dioxolan-(2)—yl—Rest
ist, mit den Glycidäthern nach Formel (^) hergestellt werden, wobei man
vorteilhafterweise die Phenole nach Formel (jSj erst während der Reaktion
aus den entsprechenden Acetalen erzeugt.
Verbindungen nach Formel (_1_), in denen der Rest R eine Nicotinoylgruppe
darstellt, werden dadurch gewonnen, daß man Propanol-(2)-3)erivate
nach Formel (j_), in denen die Bindeglieder X und T —0- bedeuten und
X auch -S- sein kann, mit Nicotinsäurehalogeniden in an sich bekannter Weise umgesetzt. Propanol—(2)-Derivate nach Formel (J.), in denen R die
Bedeutung von -COOH besitzt, werden z. B. in Form ihrer Benzylester
mit Nicotinsäurehalogeniden zur Reaktion gebracht, wobei die Schutzgruppe anschließend durch Hydrogenolyse in Gegenwart von Palladiumkohle
selektiv abgespalten werden kann.
Glycidäther der allgemeinen Formel (j5) lassen sich durch Umsetzung

gesättigter oder ungesättigter Alkylrest mit 1-3 Kohlenstoffatomen ist, mit Epihalogenhydrinen, "bevorzugt Epichlorhydrin, in Gegenwart
von Basen, "bevorzugt Alkalihydroxiden gewinnen, wobei auf eine
Isolierung der primär gebildeten Halogenhydrine der allgemeinen Formel (8)

Die Verbindungen der allgemeinen Formel (_1_) und ihre Salze werden bevorzugt
oral verabreicht. Gewöhnlich beträgt die orale Tagesdosis bei Erwachsenen 0,1 bis 10 g, vorzugsweise 0,5 *>is 3 g·
Die Wirkstoffe können in üblicher l'orm zur oralen Verabreichung konfektioniert
werden, z. B. in Kapseln, in flüssiger Form, als Tabletten oder als Pulver. Durch Vermischen mit festen, pulverförmigen Trägerstoffen, wie Lactose,
Saccharose, Sorbit, Mannit, Kartoffel- oder Maisstärke, Cellulosederivaten oder Gelatine, gegebenenfalls unter Zusatz von Gleitmitteln, wie z. B.
Magnesium- oder Calciumstearat, Carb.owax^ ' oder Polyäthylenglycolen können

sie zu Tabletten oder zu Drageekernen verarbeitet werden.
Als weitere Verabreichungsformen eignen sich Steckkapseln, z. B. aus
Hartgelatine sowie geschlossene Weichgelatinekapseln mit einem Weichmacher, wie z. B. Glycerin. Die Steckkapseln enthalten den Wirkstoff
vorzugsweise als Granulat, z. B. in Mischung mit !Füllstoffen, wie Lactose, Saccharose, Mannit, Stärken, wie z. B. Kartoffelstärke,
Maisstärke oder Amylopectin, Cellulosederivaten, Gelatine oder hochdispersen Kieselsäuren. In weichen Kapseln ist der Wirkstoff
vorzugsweise in geeigneten Flüssigkeiten, z. B. in Pflanzenöl oder flüssigen Polyäthylenglycolen gelöst oder suspendiert.
Im folgenden wird die Erfindung anhand von Ausführungsbeispielen
näher erläutert.
Beispiel 1
5-(4>-Metho3tycarbonylpheno:xy)-1-(4>-chlorphenoxy)-propanol-(2)
Zur Lösung von 1,12 g (0,02 Mol) Kaiiumhydroxid in I50 ml absol.
Methanol werden 18,50 g (0,12 Mol) 4'-Hydroxybenzoesäuremethylester
gegeben und die Mischung langsam zum Sieden erwärmt. Im Verlaufe von 1 Stde. gibt man 18,46 g (0,1 Mol) 3-(4'-Chlorphenoxy)-1,2-epoxypropan
hinzu und erhitzt 21 Stdn. am Rückfluß. Anschließend wird das Lösungsmittel i. Vak. entfernt, der Rückstand in Ither aufgenommen und
mit Wasser bis zur neutralen Reaktion gewaschen. Nach dem Einengen
der über Natriumsulfat getrockneten organischen Phase i. Vak. verbleiben 25»05 g (74»4 f°) Rohprodukt, welches aus Tetrachlorkohlenstoff
umkristallisiert wird. Farblose Kristalle vom Schmp. 88-9O0Cj Ausbeute:

3-(4'-Chlorphenoxy)-1.2-epoxypropan werden mit der Lösung von 0.8 g
(0.02 Mol) Natriumhydroxid in 15Ο ml absolutem Methanol 18 Stdn. unter
Rückfluß erhitzt. Nach dem Entfernen des Lösungsmittels i. Vak. wird der Rückstand in Chloroform gelöst und mehrmals mit verd. Natriumhydroxid-Lösung
und anschließend mit Wasser gewaschen. Die über Natriumsulfat getrocknete und i. Vak. eingeengte organische Phase liefert ein öl, das
langsam erstarrt. Nach dem Umkristallisieren verbleiben 28.5 g (78.5 $)
farbloser Kristalle vom Schmp. 83-84 C (Ithanol/Wasser).

Methanol werden bei Raumtemperatur 20.07 S (0.1 Mol) 3-(4'-Chlorphenylmercapto)-1,2-epoxypropan
und 19.6 g (0.11 Mol) 4'-Hydroxyzimtsäuremethylester
eingetragen. Nach 19 Stdn. Erhitzen unter Rückfluß wird das
Lösungsmittel i. Vak. entfernt, der Rückstand in Chloroform aufgenommen und mit verd. Kaliumhydroxid-Lösung mehrmals extrahiert. Nach dem
Waschen und Trocknen über Natriumsulfat wird die organische Phase i. Vak. eingeengt, wobei 30.7 g (81.2 0Ja) Rohprodukt in Form eines gelben Öls

22.01 g (O.I Mol) nach bekanntem Verfahren (J. Amer. Chem. Soc. 72,
(195Ο) 37IO) hergestelltes 3-Chlor-1-(4'-chlorphenylamino)-propanol-(2)
in 100 ml absolutem Methanol werden mit 6.17 g (O.II Mol) Kaliumhydroxid
in 100 ml absolutem Methanol versetzt. Die Bildung des Epoxids läßt sich an
der Entstehung eines Niederschlags verfolgen. Nach Zugabe von 15·21 g
(O.I Mol) 4'-Hydroxybenzoesäuremethylester wird das Reaktionsgemisch
12 Stdn. unter Rückfluß erhitzt, das ausgeschiedene Kaliumchlorid abgetrennt und das Filtrat i. Vak. eingeengt. Der Rückstand wird mehrmals
mit Wasser digeriert und nach dem Trocknen über Phosphorpentoxid (26.4 g;
78.6 fo) zur weiteren Reinigung umkristallisiert. Farblose Kristalle vom
Schmp. 126-127° (Essigester).

Rückstand kristallisiert beim Digerieren mit Petroläther; farblose Kristalle vom Schmp. 114-115 (Essigester/Petroläther), Ausbeute:

22.44 S (0.4 Mol) Kaliumhydroxid, 6ö ml Äthanol und 30 ml Wasser wird 4 Stdn. unter Rückfluß
erwärmt, wobei eine klare Lösung entsteht. Bas Reaktionsgemisch wird
anschließend i. Vak. eingeengt, der Rückstand in Wasser gelöst und mit 5 η Schwefelsäure auf pH = 1 gestellt. Der niederschlag wird mit
Wasser neutral gewaschen, über Phosphorpentoxid i. Yak. getrocknet und
zur Reinigung umkristallisiert. Farblose Kristalle vom Schmp. 161-162°C (Methanol); Ausbeute 31.33 S (89.7%).

Die Suspension von 36.48 g (O.I Mol) 3-(4'-Carboxyphenoxy)-1-(4'-chlorphenoxy)-propanol-(2)-acetat
in 200 ml wasserfreiem Chloroform wird unter Rühren zum Sieden erwärmt. Anschließend entfernt
man die Heizung und gibt 17-85 S (0.I5 Mol) Thionylchlorid in einer
Weise zu, daß der Rückfluß erhalten bleibt. Nach dem Abklingen der
exothermen Reaktion wird weiter zum Sieden erwärmt, bis die Gasentwicklung beendet ist (ca. 4 Stdn.)j Chloroform und überschüssiges
Thionylchlorid i. Vak. entfernt und der Rückstand aus Tetrachlorkohlenstoff umkristallisiert.

in 50 ml absolutem Benzol werden 13.21 g (0.12 Mol) Pyridyl-(3)-methanol zugetropft und die
Mischung 5 Stdn. unter Rückfluß gerührt. Nach Zugabe von 50 ml
Benzol wird mehrfach mit Wasser extrahiert, die organische Phase über Natriumsulfat getrocknet und i. Vak. eingeengt.
Zur Entfernung der Schutzgruppe wird der Rückstand in 70 ml Methanol
gelöst, mit 20 ml konz. Ammoniak versetzt und die Mischung mehrere Stunden bei Raumtemperatur digeriert. Bann wird i. Vak. eingeengt,
der Rückstand in Chloroform aufgenommen und mit verdünnter Natriumhydrogencarbonatlösung
sowie mit Wasser gewaschen. Nach dem Trocknen über Natriumsulfat wird das Chloroform i. Vak. entfernt und das Rohprodukt
umkristallisiert. Farblose Kristalle vom Schmp. 1O7-9°C (Methanol). Ausbeute: 11.7 g (28.3 $).

5- C4' -(1.5-Dioxolan-2-yl )-T?henoxy ]-1 -(-4' -chlorphenoxy )-propanol-( 2 )
Die Lösung von 20.82 g (0.1 Mol) 2-(4'~Acetoxyphenyl)-1.3-dioxolan
(hergestellt in Analogie zu dem im Bull. Soc. Chim. Prance 1972 (6),
2287 aufgeführten Verfahren) und 6.74 g (0.12 Mol) Kaliumhydroxid in
100 ml Isopropanol wird im Verlauf von 1 Stde. mit 1.8 ml (O.I Mol)
Wasser versetzt und 5 Stdn. bei Baumtemperatur gerührt. Anschließend
werden I6.56 g (O.O9 Mol) 3-(4*-Chlorphenoxy)-1,2-epoxypropan zugefügt
und das Reaktionsgemisch weitere 6 Stdn. "bei 60 C gehalten. Nach dem
Einengen des Lösungsmittels i. Vafc. wird der Rückstand in Äther aufgenommen, die Ätherphase mit 2 J&Lger Kaliumcarbonatlösung mehrfach extrahiert,
mit Wasser neutral gewaschen und üper Natriumsulfat getrocknet.
Der Äther wird i. Yak. entfernt und der Rückstand (20.9 g>
66.4 %) bei 1900C / 1 χ 10"1 mm destilliert.

enthaltendes Arzneimittel
100 g 3-(4'-Methoxycarbonylphenoxy)-1-(4'-chlorphenoxy)-propanol-(2)
werden mit 16 g Haisstärke und 6 g hochdispersem Siliciumdioxyd gut
vermischt, anschließend mit einer Lösung von 2 g Stearinsäure, 6 g Acetylcellulose und 6 g Stearin in 70 ml Isopropanol befeuchtet und
granuliert. Das getrocknete Granulat wird durch ein Sieb geschlagen und nach Vermischen mit 16 g Maisstärke, 16 g Talk und 2 g Magnesiumstearat
zu 1000 Drageekernen verpreßt. Nach Überziehen mit einem Sirup von 2 g Lacca, 7>5 g Gummi arabicum, 0,15 g i'arbstoff, 2 g kolloidale Siliciumdioxyd,
25 g Talk und 53» 35 S Saccharose v/erden nach dem Trocknen
tausend 260 mg schwere Dragees mit je 100 mg Wirkstoff erhalten.

Pharruakologische Testung
1. Orale Verträglichkeit
Nach oraler Applikation der Testverbindungen an Mäusen vom Stamm IMR-I, mit einem Gewicht von 15-20g wurde die alcute Toxizität
bestimmt. Die LDr -Werte wurden nach Litchfield-V/ilcoxon (J. Pharmak.
exp. Therap. ji6_, 99 (1949)) bereclmet und beziehen sich auf den
achten Tag nach der Behandlung.
Die LDcq für Clofibrat betrug I9OO mg/kg. Die erfindungsgemäßen
Substanzen waren durchweg verträglicher und dem Clofibrat überlegen.
2. Lipidsenkende Wirkung-
Die lipidsenkende Wirkung wurde an Gruppen von jeweils 10 normal
gefütterten ("ssniff"-Alleinfutter), normolipämischen, männlichen,
200 - 220 g schweren Wistar-Eatten (Ivaiiovas-Kis;.jIegg) geprüft.
Die Testverbindungen wurden in einer wässrigen Lösung von 0,25 c/°
Agar und 0,85 i° NaCl aufgenommen und oral verabreicht. ITach Applikation
von 4 x 100 mg/kg über einen Zeitraum von 3 Tagen wurden
die Tiere im Anschluß an einen vierstündigen Putterentzug durch

Die Bestimmung der Seruialipide wurde mit einem 'l'echnicon Auto-,
analyzer durchgeführt. Das Gesamtcliolesterin (TC) wurde mit der
Lieberinann-Burchard-Iteaktion erfaßt. Mo quantitative Analyse der
i'riglyceride (ΐ&) erfolgte enzymatisch nach Eggstein und Kreutz
(lilin. "Wschr. AjI, 2G2 (19£>«))>
modifiziert für den Autoanalyzer.
Die lipidsenkende Wirkung iat ausgedrückt als die prozentuale
Senkung des Gesamtcholesterins und der Triglyeeride gegenüber
der Kontrolle. Unter oben angeführten Yersuohsbediiigungen konnte
mit Clofibrat eine durchschnittliche Senkung des Gesaiatcholesterins
um 37·4 i 13·ύ '/β und der Trigiyceride uin 50.4 ± 20.0 ^o

(jeweils X +_ S ) erzielt werden . Sämtliche getesteten Verbindungen
waren in mindestens einem ¥ert dem Clofibrat überlegen.
