Die vorliegende Erfindung betrifft neue substituierte
Phenylisothioharnstoffe, ein Verfahren zu ihrer Herstellung und ihre Verwendung als Arzneimittel, insbesondere als
Anthelmintika.
Es ist bereits bekannt geworden, daß das Präparat Thiabendazole der Formel
welches ein bekanntes, handelsübliches Anthelmintikum darstellt,
eine gute anthelmintische Wirkung aufweist. Die erfindungsgemäßen Verbindungen zeigen aber eine wesentlich
stärkere Wirkung gegenüber Helminthen als Thiabendazole.
Es wurde gefunden, daß die neuen substituierten Phenylisothioharnstoffe

Im Anmeldungstext wurden die jeweiligen Strukturformeln aus Gründen der Einheitlichkeit in allen Fällen gleichartig
formuliert.
Überraschenderweise zeigen die erfindungsgemäßen Isothioharnstoffe
eine erheblich höhere anthelmintische Wirkung als das handelsübliche Anthelmintikum Thiabendazole.Die erfindungsgemäßen
Stoffe stellen somit eine wertvolle Bereicherung der Pharmazie dar.
Zwei typische Beispiele erläutern die Herstellungsmethoden:
Verwendet man N-(2-Acetamido-4-phenoxy-phenyl)-N'-methoxycarbonyl-thioharnstoff,
Methyljodid und Natriumhydroxid als Ausgangsstoffe, so kann der Reaktionsverlauf durch das folgende

Äthylthio, n- und i-Propylthio, n-, i- und t--Butylthio genannt.
Als gegebenenfalls substituiertes Acyl R2 und R3 der Formel II
steht Acyl mit 1-6, vorzugsweise 2-4 Kohlenstoffatomen. Beispielhaft seien genannt: Formyl, Acetyl, Propionyl,
n-Butyryl, i-Butyryl.
Als gegebenenfalls substituiertes Carbalkoxy R2, R3 der Formel
II steht geradkettiges oder verzweigtes Carbalkoxy mit vorzugsweise 2-7, insbesondere 2-5 Kohlenstoffatomen.
Beispielhaft seien gegebenenfalls substituiertes Carbomethoxy, Carboäthoxy, Carbo-n- und -i-propyloxy und Carbo-n-, -i- und
-t-butyloxy genannt.
Halogenalkyl R2 und R3 der Formel II enthält vorzugsweise
1-4, insbesondere 1 oder 2 Kohlenstoffatome und vorzugsweise 1-5, insbesondere 1 - 3 gleiche oder verschiedene Halogen-

atome, wobei als ttalogenatome vorzugsweise Fluor, Chlor und
Brom, insbesondere Fluor und Chlor stehen. Beispielhaft seien Trifluormethyl, Chlor-di-fluormethyl, Brommethyl, 2,2,2-Trifluoräthyl
und Pentafluoräthyl genannt.
Als Halogen R , R^ in der Formel II steht vorzugsweise Fluor,
Chlor, Brom und Jod, insbesondere Fluor, Chlor und Brom.
Als gegebenenfalls substituiertes Aryl R und R in den Foimeln
II, IV und V steht Aryl mit vorzugsweise 6 oder vorzugsweise 6 - 1o Kohlenstoffatomen im Arylteil. Beispielhaft seien
gegebenenfalls substituiertes Phenyl oder Naphthyl genannt.

Als gegebenenfalls substituiertes Aralkyl R und R in den Formeln
II,IV und V steht gegebenenfalls im Arylteil und/oder Alkylteil substituiertes Aralkyl mit vorzugsweise 6 oder 1o,
insbesondere 6 Kohlenstoffatomen im Arylteil und vorzugsweise
I bis 4, insbesondere 1 oder 2 Kohlenstoffatomen im Alkylteil,
wobei der Alkylteil geradkettig oder verzweigt sein kann. Beispielhaft seien gegebenenfalls substituiertes Benzyl und Phenyläthyl
genannt.

Als gegebenenfalls substituiertes Cycloalkyl R , R in den Formeln II,IV und V steht mono- bi- und +"icyclisches Cycloalkyl
mit vorzugsweise 3 - 1o, insbesondere 3, 5. oder 6 Kohlenstoffatomen. Beispielhaft seien gegebenenfalls substituiertes
Cyclopropyl, C/clobutyl, Cyclopentyl, Cyclohexyl,
Cycloheptyl, Bicyclo-/~2,2,1_7-heptyl, Bicyclo-/~2,2,2_7-octyl
und Adamantyl genannt.
Als gegebenenfalls substituiertes Alkenyloxy R in den Formeln
II und V steht geradkettiges oder verzweigtes Alkenyloxy mit vorzugsweise
2-6, insbesondere 2-4 Kohlenstoffatomen. Beispielhaft seien gegebenenfalls substituiertes Äthenyloxy,
Propenyl-(1)-oxy, Propenyl-(2)-oxy und Butenyl-(3)-oxy genannt.

II, IV und V steht geradkettiges oder verzweigtes Alkinyl mit vorzugsweise 2 bis 6, insbesondere 2 bis 4 Kohlenstoffatomen.
Beispielhaft seien gegebenenfalls substituiertes Aethinyloxy, Propinyl-(1)-oxy, Propinyl-(2)-oxy und Butinyl-(3)-oxy genannt.
Die als Ausgangsstoffe zu verwendenden Alkylierungsmittel sind durch die Formel (III) definiert.
Als Beispiele für die erf indungsgerräß zu verwendenden Alkylierungsmittel
seien genannt:
Methyljodid, Aethyljodid, iso-Propyljodid, Cyclohexylbromid,
Dimethylsulfat, Toluolsulfonsäure-methylester, Allylbromid,
Benzylchlorid. Diese Verbindungen sind allgenein bekannt.
Als Substituenten für die Reste R , R , R , R und R kommen
infrage: Acyl mit vorzugsweise 2 bis 4 Kohlenstoffatomen, Carbalkoxy mit 2 bis 4 Kohlenstoffatomen, Alkoxyalkyl mit
jeweils 1 bis 4 Kohlenstoffatomen im Alkoxy und im Alkylteil, Aralkyl mit vorzugsweise 1 bis 4 Kohlenstoffatomen im Alkylteil
und vorzugsweise 6 Kohlenstoffatomen im Arylteil.
Besonders bevorzugt sind Verbindungen der Formel I, in denen R2 und R3 für Wasserstoff, Methyl, Äthyl, η-Butyl, Methoxy,
Athoxy, Fluor, Chlor, Brom, Cyan, Methylmercapto, Trifluormethyl, Acetyl, Propionyl, Acetylamino,
Carbomethoxyamino stehen,

Die als Ausgangsstoffe zu verwendenden substituierten Phenyithioharnstoffe
der Formel II und Anilinderivate der Formel IV sind zum Teil noch nicht bekannt. Sie können jedoch in Analogie
zu literaturbekannten Verfahren leicht hergestellt werden. So kann man die Anilinderivate der Formel IV aus den entsprechenden
Nitroanilinen durch Acylierung vorzugsweise mit Säurechloriden und nachträgliche Reduktion vorzugsweise mit
H2/Raney-Nickel gewinnen:

Die Verbindungen der Formel (II), in welcher X für SO steht, werden im allgemeinen durch Oxidation der entsprechenden Phenyl thioverbindungen
mit Wasserstoffperoxid in Acetanhydrid oder mit Benzoersäure in Dioxan oder Chloroform erhalten.
Durch Oxidation mit H2O2 in Eisessig bei erhöhter Temperatur
erhält man die entsprechenden Phenylsulfonylverbindungen der Formel (II):

Die Anilinderivate der Formel (IV),in welcher X für 30 bzw.
SO2 steht, können durch Umsetzung der entsprechenden 1-Nitroverbindungen mit H202/Acetanhydrid "bzw. H202/Eisessig, höhere Temperatur und anschließender Reduktion der Nitrogruppe zur
Aminogruppe zu den Verbindungen der Formel (IV), in welcher X für SO bzw. für SO2 steht, übergeführt werden. Die Reaktionsfolge soll durch das folgende Schema verdeutlicht werden:
SO2 steht, können durch Umsetzung der entsprechenden 1-Nitroverbindungen mit H202/Acetanhydrid "bzw. H202/Eisessig, höhere Temperatur und anschließender Reduktion der Nitrogruppe zur

Als "Verdünnungsmittel kommen bei der Durchführung der erfindungsgemäßen
Verfahrensvarianten a) und b) Wasser oder organische Lösungsmittel infrage, wie z. B. Alkohol, Aceton,
Dimethylsulfoxid, Dimethylformamid oder Acetonitril, entweder
allein oder in ihren Mischungen mit Wasser.
Die als Hilfsstoffe bei den erfindungsgsmäßen Verfahrensvarianten a) und b) zu verwendenden anorganischen und organischen

umfassen. Vorzugsweise verwendet man Jedoch Kaliumhydroxid, Natriumhydroxid, Natriumcarbonat, Nafcriumbicarbonat, Triäthylamin,
Pyridin, Collidin u. a.
Die Reaktionstemperaturen können bei besäen Varianten a) und
b) in einem größeren Bereich variiert werden. Im allgemeinen
arbeitet man zwischen -1o und +4o C, vorzugsweise zwischen
0 und +3o°C.
Bei der Durchführung der erfindungsgemäßen Verfahrensvarianten a) und b) setzt man im allgemeinen äquimolekulare
Mengen ein. Die Aufarbeitung kann so vorgenommen werden, daß man das Reaktionsgemisch - gegebenenfalls nach Abziehen des
Lösungsmittels - in Wasser gibt, das ausgefallene Produkt absaugt, trocknet und durch Umkristallisation reinigt.

Die Wirkung wurde im Tierversuch nach oraler und parenteraler
Applikation bei stark mit Parasiten befallenen Versuchstieren geprüft. Die angewendeten Dosierungen wurden sehr gut von den
Versuchstieren vertragen.
Die neuen Wirkstoffe können als Anthelmintika sowohl in der
Human- ale auch in der Veterinärmedizin verwendet werden.
Die neuen Wirkstoffe können in bekannter Weiss ui die
üblichen Formulierungen übergeführt werden.
Die neuen Verbindungen können entweder als solche oder aber
in Kombination mit pharmazeutisch annehmbaren Trägern zur Anwendung gelangen. Als Darreichungsformen in Kombination
mit verschiedenen inerten Trägern kommen Tabletten, Kapseln, Granulate, wäßrige Suspensionen, injizierbare Lösungen,
Emulsionen und Suspensionen, Elixiere, Sirup, Pasten und dergleichen in Betracht. Derartige Träger umfassen feste
Verdünnungsmittel oder Füllstoffe, ein steriles, wäßriges Medium sowie verschiedene nicht toxische organische Lösungemittel
und dergleichen. Selbstverständlich können die für eine orale Verabreichung in Betracht kommenden Tabletten
und dergleichen mit Süßstoffzusatz und ähnlichem versehen werden. Die therapeutisch wirksame Verbindung soll im vorgenannten
Fall in einer Konzentration von etwa 0,5 bis 90 Gew.-^ der Gesamtmischung vorhanden sein, d. h. in Mengen,
die ausreichend sind, um den obengenannten Dosierungespielraum zu erreichen.
Die Formulierungen werden in bekannter Weise hergestellt, z. B. durch Verstrecken der Wirkstoffe mit Lösungemitteln
und/oder Trägerstoffen, gegebenenfalls unter Verwendung von Emulgiermitteln und/oder Dispergiermitteln, wobei z. B.
im Fall der Benutzung von Wasser als Verdünnungsmittel
gegebenen!alls organische Lösungsmittel als Hilfelösungemittel
verwendet werden können.

Alkohole (z". B. Äthylalkohol, Glycerin), Glykole (ζ. Β. Propylenglykol, Polyäthylenglykol) und Wasaer; feete
Trägeratoffe, wie z. B. natürliche Geateinamehle (z. B.
Kaoline, Tonerden, Talkum, Kreide), synthetiache Gesteinemehle
(z. B. hochdisperse Kieselsäure, Silikate), Zucker (z. B. Roh-, Milch- und Traubenzucker); Smulgi "littel,
wie nicht ionogene und anionische Emulgatoren (ζ. Β. PoIyoxyäthylen-Pettsäure-Ester,
Polyoxyäthylen-Fettalkohol-Äther, Alkylsulfonate und Arylsulfonate), Dispergiermittel
(z. B. Lignin, Sulfitablaugen, Methylcellulose, Stärke und
Polyvinylpyrrolidon) und Gleitmittel (z.. B. Magnesiumstearat,
Talkum, Stearinsäure und Natriumlaurylsulfat).
Im Falle der oralen Anwendung können Tabletten selbstverständlich aufler den genannten Trägeretoffen auch Zusätze,
wie Natriumeitrat, Calciumcarbonat und Dicalciumphosphat, zusammen mit verschiedenen Zuschlagstoffen, wie Stärke,
vorzugsweise Kartoffelstärke, Gelatine und dergleichen, enthalten. Weiterhin können Gleitmittel, wie Magnesiumetearat,
Natriumlaurylsulfat und Talfcum, zum Tablettieren
mitverwendet werden.
Im Falle wäßriger Suspensionen und/oder Elixieren, die für
orale Anwendungen gedacht sind, können die Wirkstoffe außer mit den obengenannten Hilfsstoffen mit verschiedenen
Geschmacksaufbesserern oder Farbstoffen versetzt werden.
Für den Fall der parenteralen Anwendung können Lösungen der
Wirkstoffe unter Verwendung geeigneter flüssiger Trägermaterialen
eingesetzt werden.

Die Wirkstoffe können in Kapseln, Tabletten, PartÄa4n!," ^
Dragees, Ampullen usw. auch in Form von Dosierungaeinheiten
enthalten sein, wobei jede Dosierungseinheit so angepaßt
ist, daß sie eine einzelne Dosis des aktiven Bestandteile liefert.
Die neuen Verbindungen können in den Formulierungen auch in Mischungen mit anderen bekannten Wirkstoffen vorliegen.
Die neuen Wirkstoffe können in üblicher Weise angewendet
werden. Die Applikation erfolgt vorzugsweise oral, eine
parenterale, insbesondere subkutane, aber auch eine dermale
Applikation sind jedoch ebenfalls möglich.
Im allgemeinen hat es sich als vorteilhaft erwiesen, Mengen von etwa 1 bis etwa 100 mg der neuen Verbindungen je kg
Körpergewicht pro Tag zur Erzielung wirksamer Ergebnisse zu verabreichen.
Trotzdem kann es gegebenenfalls erforderlich sein, von den genannten Mengen a\ zuweichen, und zwar in Abhängigkeit vom
Körpergewicht des Versuchstieres bzw. der Art des Applikationsweges, aber auch aufgrund der Tierart und deren
individuellem Verhalten gegenüber dem Medikament bzw. der Art von dessen Formulierung und dem Zeitpunkt bzw. Intervall,
zu welchem die Verabreichung erfolgt. So kann es in einigen Fällen ausreichend sein, mit weniger als der vorgenannten
Mindestmenge auszukommen, während in anderen Fällen die genannte obere Grenze überschritten werden muß.
Im Fall der Applikation größerer Mengen kann es empfehlenswert sein, diese in mehrere Einzelgaben über den Tag zu
verteilen. Für die Applikation in der Human- und Veterinärmedizin ist der gleiche Dosierungsepielrauir vorgesehen.
Sinngemäß gelten auch die weiteren obigen Ausführungen.

Experimentell mit Uncinaria stenocephala infizierte Hunde wurden nach Ablauf der Praepatenz der Parasiten behandelt.
Die Wirkstoffmenge wurde als reiner Wirkstoff in Gelatinekapseln
oral appliziert.
Der Wirkungsgrad wird dadurch bestimmt, daß man die nach der Behandlung abgetriebenen und die im Versuchstier verbliebenen
Würmer nach Sektion zählt und den Prozentsatz der abgetriebenen Würmer errechnet.
In der nachfolgenden Tabelle sind die Wirkstoffe, die Parasitenart und die geringste Dosierung, die den Wurmbefall
der Versuchstiere um mehr als 9o % ("Red.^ 9o%") reduziert,
im Vergleich zum Thiabendazole aufgeführt.
Die Dosierung wird in mg Wixksubstanz pro kg Körpergewicht angegeben.
Der Ausdruck "3 x 2,5" bedeutet, daß dreimal 2,5 mg/kg appliziert worden sind.

Ein völliges Sistieren der Eiausscheidung nach der Behandlung
bedeutet, daß die Würmer abgetrieben wurden oder so geschädigt sind, daß sie keine Eier mehr produzieren können (Dosis
effectiva).

Herstellung neuer Ausgangsstoffe:
Beispiel A 1)

2-Amino-5-phenoxy-acetanilid
2-Amino-5-phenoxy-acetanilid
2-Amino-5-phenoxy-acetanilid kann in einem zweistufigen
Prozess aus 2-Nitro~5--phenoxy-anilin wie folgt gewonnen werden:
1. Stufe:
NH-CO-CH3
Zu einer Lösung von 23 g (o,1 Mol) 2-Nitro-5-phenoxy-anilin
vom Fp. 1420C und 7,9 g (o,1 Mol) Pyridin in 25o ml trockenem
Benzol wird unter Rühren bei Raumtemperatur 7,9 g (o,1 Mol) Acetylchlorid in 5o ml trockenem Benzol zugetropft.
Nach dem Abklingen der leicht exothermen Reaktion wird 1,5 Stunden bei Zimmertemperatur und weitere 2 Stunden bei Siedetemperatur
nachgerührt. Danach wird vom ausgeschiedenen, wasserlöslichen Pyridinhydrochlorid heiß abfiltriert, das
Lösemittel im Vakuum abdestilliert, der Rückstand mit verdünnter Salzsäure gut verrührt, abgesaugt, getrocknet und aus

Aethanol umgelöst. Man erhält das 2-Nitro-5-phenoxy-acetanilid
vom Fp. 9o°C in einer Ausbeute von 73 % der Theorie,
2. Stufe:
o-o-
NH-CO-CH2 NH-CO-CH7
54,5 g (o,2 Mol) 2-Nitro-5-phenoxy-acetanilid vom Fp. 9o C
werden in 5oo ml Tetrahydrofuran mit 5 g Raney-Nickel über
eine Reaktionsdauer von 1,5 Stunden bei 5o atu Wasserstoff
hydriert. Die Temperatur steigt dabei von 13 auf 21 C an; der Hp-Verbrauch entspricht der berechneten Menge. Nach dem Erkalten
wird vom Katalysator abgesaugt, das Lösemittel verdampft und der Rückstand aus Toluol umgelöst. Man erhält
labei das 2-Amino-5-phenoxy-ace-Ausbeute von 77 % der Theorie.
Beispiel A 2)
dabei das 2-Amino-5-phenoxy-acetanilid vom Fp. 12o C in einer

N-(2-Acetamino-4-phenoxy-phenyl)-N1-methoxycarbonyl-thioharnstoff
Zu einer Lösung von 121 g (o,5 Mol) 2-Amino-5-phenoxy-acetanilid
vom Fp. 12o°C in 1ooo ml Aceton wird unter Rühren bei Raumtemperatur eine Lösung zugetropft, die folgendermaßen
hergestellt wird:

Zu 97,2 g (1 Mol) Kaliumrhodanid (= Kaliumthiocyanat) in
365 ml trockenem Aceton werden 94,5 g (1 Mol) Chlorameisensäuremethylester
unter Rühren und Wasserkühlung zugetropft. Gegen Ende des Eintropfens läßt man die Temperatur auf 4o C
ansteigen, rührt hierbei noch 1 Stunde nach und filtriert anschließend (man geht dabei von einer 5o%igen Umsetzung aus).
Nach Beendigung des Eintragens dieser Lösung aus Kaliumrhodanid und Chlorameisensäuremethylester wird anschließend
für 1 Stunde bei Raumtemperatur und 4 Stunden bei 60 C
nachgerührt. Anschließend wird auf 1o C abgekühlt, der dabei auskristallisierende N-(2-Acetamido-4-phenoxy-phenyl)-N'-methoxycarbonyl-thioharnstoff
wird mit wenig Aceton nachgewaschen und getrocknet. Zur weiteren Reinigung kann aus Dioxan
umgelöst werden. Fp. 2040C (unter Zersetzung), Ausbeute: 64 g.
