Die Erfindung betrifft ferner ein Verfahren zur Herstellung der Verbindungen der allgemeinen Formel (I), welches dadurch gekenn-

angegebene Bedeutung haben, mit Ozon in einem inerten Lösungsmittel oxidiert.
Die niederen Alkylreste haben bis zu 6 Kohlenstoffatome; spezielle
Beispiele sind die Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-,
Isobutyl-, sek.-Butyl-, n-Pentyl- und n-Hexylgruppe. Halogensubstituierte
niedere Alkylreste enthalten bis zu 6 Kohlenstoffatome;
spezielle Beispiele sind die 2-Chloräthyl-, 3-Brompropyl- und

3-Fluorbuty!gruppe. Der drei- bis sechsgliedrige Ring kann ein
Aziridin-, Azetidin-, Pyrrolidin-, Piperidin- oder Morpholin-Ring
sein. Der Arylrest hat 6 bis 10 Kohlenstoffatome; spezielle Beispiele
sind die Phenyl-, ToIy1-, XyIyI-, Methoxyphenyl-, Chlorphenyl-,
Bromphenyl- und Naphthylgruppe. Der Aralkylrest hat 7 bis 10 Kohlenstoffatome; spezielle Beispiele sind die Benzyl-,
Phenyläthyl- und Phenylpropylgruppe.
Die im erfindungsgemäßen Verfahren als Ausgangsmaterial verwendeten
Phosphorsäurediamidester der Formel (II) werden beispielsweise dadurch hergestellt, daß man ein substituiertes oder unsubstituiertes
2-Propen-l-ol (n = 0) oder 3-Buten-l-ol (n = 1) der allgemeinen
Formel (III) mit Phosphoroxychlorid umsetzt, den erhaltenen Dichlorphosphorsäureester der allgemeinen Formel (IV) mit einem
sekundären Amin der allgemeinen Formel (V) oder mit Ammoniak kondensiert
und den erhaltenen Chlorphosphorsäureamidester der allgemeinen Formel (VI) mit einem primären Amin der allgemeinen Formel
(VII) oder mit Ammoniak in den Phosphorsäurediamidester der allgemeinen Formel (II) nach folgendem Reaktionsschema überführt:

Das erfindungsgemäße Verfahren wird auf bekannte Weise als Ozonoxidation
durchgeführt. Beispielsweise wird Ozon in einem Ozon-, generator hergestellt und zusammen mit Sauerstoff oder Luft in
die Lösung einer Ausgangsverbindung der allgemeinen Formel (II)
in einem inerten Lösungsmittel bei Raumtemperatur unter Eiskühlung eingeleitet. Geeignete Lösungsmittel sind solche, die
üblicherweise bei einer Ozonoxidation verwendet werden, z.B.

Methylchlorid, Äthylchlorid, Chloroform _Jlethylenchlorid,Dichloräthan,
Tetrachloräthan, Petroläther ,η-Hexan, Cyclohexan, Leichtbenzin,
Ligroin, Benzol, Toluol, Tetrahydrofuran, Dioxan, Aceton, Methyläthylketon oder ein Gemisch dieser Lösungsmittel mit oder
ohne Wasser. Das Ozon enthaltende Reaktionsgemisch wird im allgemeinen einige Stunden oder einige Tage lang bei Raumtemperatur
oder unter Kühlung (von 35 bis -200C) gehalten. Es empfiehlt sich,
das Reaktionsgemisch nach dem Einleiten des Ozons mit einem Peroxid, wie Wasserstoffperoxid, zu versetzen, jedoch ist diese
Maßnahme im erfindungsgemäßen Verfahren nicht unbedingt notwendig.
Die gewünschte Verbindung der allgemeinen Formel (I) kann aus dem Reaktionsgemisch isoliert und in herkömmlicher Weise durch Extraktion, Destillation, Chromatographie oder Umkristallisation gereinigt
werden.

Die erfindungsgemäßen Verbindungen der allgemeinen Formel (I)
sind wirksame antibakterielle, immunosuppressive und/oder tumorhemmende Mittel.
1. Die antibakterielle Aktivität der erfindungsgemäßen Verbindungen
der allgemeinen Formel (I), z.B. 2-[Bis-(2-chloräthyl)-amino]-4-hydroperoxy-l-oxa-3-aza-2-phosphacyclohexan-2-oxid
(im folgenden als Verbindung A bezeichnet) und 2-(2-Chloräthyl)-amino-3-(2-chlor-
äthyl)-^-hydroperoxy-l-oxa-3-aza-2-phosphacyclohexan-2-oxid (Verbindung
B), wird in einem Agar-Verdünnungstest mit synthetischem
Medium (Davis & Mingioli, J.Bac, Band 60 (1950) Seite 17) bestimmt.
Hierbei ergibt sich eine minimale Hemmkonzentration gegen Bacillus subtilis PCI 219, Escherichia coli NIHJ JC-2, Salmonella
typhimurium und Pseudomonas aeruginosa von 6,3 χ 10~^ g/ml für
Verbindung A und 6,3 bis 12,5 χ 10~5 g/ml für Verbindung B.
2. Die immunosuppressive Wirkung der erfindungsgemäßen Verbindungen
der allgemeinen Formel (I) wird mit der von im Handel erhältlichen,
immünosüppressiven Mitteln, wie Azathlöprine und C'yclophosphamid
verglichen. Tabelle 1 faßt diese Ergebnisse zusammen.

Einer Gruppe von weiblichen DS-Miiusen werden rote Blutkörperchen

von Schafen (10 /Maus) intraperitoneal injiziert, um eine Immunreaktion
hervorzurufen. Gleichzeitig werden ebenfalls intraperitoneal die Testverbindungen verabreicht und die Antisera abgetrennt.
Der Häraagglutinationstiter der Antisera wird nach der Mikrotiter-Methode
gemessen und in log„-Reziprokwerten der Endpunktverdünnung ausgedrückt. Die gegen 2-Mercaptoäthanol resistenten Antikörper
werden auf ähnliche Weise bestimmt, nachdem die abgetrennten Antisera mit 0,1 m 2-Mercaptoäthanol behandelt wurden.
Aus der Tabelle I geht hervor, daß die erfindungsgemäßen Verbindungen
der Formel (I) als Immunosuppressor wirksamer als Azathioprine sind.

3. Die erfindungsgemäßen Verbindungen der Formel (I) zeigen ausgezeichnete
Antitumor-Wirkung gegen das Yoshida sarcoma, Leukämie L-1210-Zellen und andere Tumoren. In Tabelle II wird die tumorhemmende
Wirkung der erfindungsgemäßen Verbindungen gegen Leukämie L-1210-Zellen mit der des Handelspräparats Cyclophosphamid
verglichen.
Versuchsmethodik:
Weibliche (C57BL χ DBA/2) F.-Mäuse mit einem Körpergewicht von
16 bis 18 g werden jeweils intraperitoneal mit 15.000 Leukämie L-1210-Zellen geimpft. 2k Stunden nach der Impfung werden die
Testverbindungen ebenfalls intraperitoneal injiziert. Die beobachtete Überlebenszeit wird in Tagen gemessen, der Überlebensindex
errechnet sich aus dem Verhältnis der mittleren Überlebenszeiten von unbehandelten Mäusen (Kontrollversuch) und behandelten Mäusen.
Im Kontrollversuch wird den Mäusen jeweils 0,1 ml einer lOprozentigen Äthanollösung int-raperitoneal injiziert.
Die Tabelle II'zeigt, daß die erfindungsgemäßen Verbindungen der
Formel (I) gegen Leukämie L-1210-Zellen stärker tumorhemmend· wirksam sind als Cyclophosphamid.

4. Die cytostatische Wirkung der erfindungsgemäßen Verbindungen
der Formel (I).gegenüber HeLa-Zellen wird mit der von Cyclophosphamid
verglichen. Die ED50 der erfindungsgemäßen Verbindung A beträgt
O,9*1O~5 g/ml, die der Verbindung B 3,1I x 10"5 g/ml, während
die ED50 von Cyclophosphamid über 100 χ 10~5 g/ml beträgt. Die
Verbindungen der Formel (I) sind gegenüber HeLa-Zellen in vitro cytostatisch wirksam, während Cyclophosphamid keinerlei Wirkung
zeigt, -
Versuchsmethodik:·
i. einem Glas werden mit 0,03 prozentiger Trypsinlösung und
O3OiI prozentiger Ithylendiamintetraessigsäure-Lösung Zellen abgelöst.
Die dispergierten Zellen werden zentrifugiert und in möglichst wenig Eagle's-Medium wieder aufgeschlämmt, das mit 10 Proaent

Rinderserum ergänzt wurde. Etwa 100 000 Zellen, die in 1 ml Medium enthalten sind, werden in eine Reihe von jeweils 3 Kulturröhrchen
übertragen und bei 37°C bebrütet. Nach 3 Tagen wird das Medium entfernt und frisches Mediumj das die Testverbindungen enthält,
zugesetzt. Nach weiteren 2 Tagen wird die Anzahl der Zellen mit einem elektronischen Zellenzähler bestimmt. Die effektive
Dosis für eine 50prozentige Zellwachstumshemmung (ED^0) wird
graphisch ermittelt (J.Leiter und Mitarbeiter, Cancer Research, Band 22, Anhang (196 1) S. 837-841).
5.>Die akute Toxizität der erfindungsgemäßen Verbindungen der Formel
(I) wird bestimmt. Die LD1-Q der Verbindung A beträgt bei intraperitonealer
Verabreichung an Mäuse 181 mg/kg, die der Verbindung B 250 mg/kg.
Versuchsmethodik:
Die Verbindung wird einer Gruppe von weiblichen DS-Mäusen mit einem Körpergewicht von 16 bis 18 g intraperitoneal injiziert. Die
Mäuse werden die folgenden 50 Tage beobachtet und die LD1-Q nach
der Methode von van der Warden berechnet.
Die Erfindung betrifft daher ferner Arzneipräparate, die ein S-Amino^-hydroperoxy-l-oxa^-aza^-phosphacyclopentan- oder
-cyelohexan-2-oxid der allgemeinen Formel (I) sowie übliche Trägerstoffe, Hilfsstoffe und/oder Verdünnungsmittel enthalten.
Im allgemeinen enthalten die erfindungsgemäßen Arzneipräparate
die Verbindungen der allgemeinen Formel (I) in etwa derselben

Dosierung, in der Cyclophosphamid verabreicht wird, d.h.sie können
in einfach Dosen oder in geteilten Dosen mit 50 bis 500 mg der aktiven Verbindung verabreicht werden. Die Arzneipräparate werden zur
Bekämpfung verschiedener bakterieller Infektionen bei Säugetieren, Autoaggressionskrankheiten, Immunrejektionen und Tumoren verwendet.
Sie können als Tabletten, Kapseln, Pillen, Suspensionen, Emulsionen, Lösungen, Zäpfchen, Salben,Granulat oder Pulver verabreicht
werden. Zur Herstellung von Tabletten wird beispielsweise die Verbindung der Formel (I) mit einem Bindemittel, wie Traganth,
Gummi arabic um, -Getreidestärke oder Gelatine vermischt.. Gewöhnlich
empfiehlt sich die Zugabe von Verdünnungsmitteln,wie Getreidestärke,
Kartoffelstärke oder Alginsäure. Außerdem werden vorzugsweise Gleitmittel, wie Stearinsäure, Magnesiumstearat oder Talkum,
sowie Süßstoffe, wie Saccharin, zugemischt. Suspensionen und Lösungen werden beispielsweise mit Sirup oder Sorbit als Trägerstoffen,
Magnesiumalumin-iumsilikat, Methylcellulose oder Carboxymethylcellulose
als Viskositätsregler und mit geeigneten Konservie^ngsmitteln, wie Natriumbenzoat, p-Hydroxybenzoesäuremethyl-
oder -propylester, hergestellt.
Die erfindungsgemäßen Arzneipräparate enthalten die Verbindungen der allgemeinen Formel (I) in therapeutischen Tagesdosen, in geringerer
Dosis zur mehrfachen Applikation oder in höherer Dosis zur Aufteilung in kleinere Einheiten. Arzneipräparate zur parenteralen
Verabreichung können aus einzelnen Dosen bestehen oder in größeren Einheiten hergestellt werden, aus denen die Einzeldosis jeweils
entnommen wird.

Die Beispiele erläutern die Erfindung.
Beispiel 1
Durch eine Lösung von 4,1 g N,N-Bis(2-chloräthyl)-phosphorsäurediamid-(3-butenyl)-ester
in 15 ml wäßrigem Aceton (Aceton/Wasser=
2:1) wird 50 Minuten lang unter Eiskühlung Ozon mit einer Geschwindigkeit von 20 bis 30 mg/min geperlt. Das Reaktionsgemisch wird
dann mit 2,0 ir.l- 30prozentigem Wasserstoffperoxid versetzt und 72-Stunden
lang bei 20C stehengelassen. Anschließend werden 100 ml
Äther zugesetzt, und das Gemisch wird mit Wasser gewaschen. Die Ätherphase wird an einer Silicagel-Säule chromatographiert und mit
Äther und Äthylacetat eluiert. Beim Eindampfen des Eluats erhält man Kristalle, die aus Aceton/Äther umkristallisiert werden ι Man
erhält 85Ο mg 2-|J3is-(2-chloräthyl)-amino3 -4-hydroperoxy-l-oxa-

Beispiel 2
Durch eine Lösung von 550 mg N,N-Bis-(2-chloräthyl)-phosphorsäurediamid-(3-butenyl)-ester
in wäßrigem Aceton (Aceton/Wasser = 2:1) wird 15 Minuten lang unter Eiskühlung Ozon mit einer Geschwindigkeit
von 15 mg/min geperlt. Das Reaktionsgemisch wird bei 20C
48 Stunden lang stehengelassen, dann mit 50 ml Äther versetzt und mit Wasser gewaschen. Die Ätherphase wird an einer Silicagel-Säule
chromatographiert, mit Äther und Äthylacetat eluiert und zur Trockene eingedampft. Die entstehenden Kristalle werden aus Aceton/Äther
umkristallisiert. Man erhält 45 mg des gleichen Produktes wie
Beispiel 1.
Durch eine Lösung von 498 mg N,N-Bis-(2-chloräthyl)-phosphorsäurediamid-(3-pentenyl)-ester
in 6 ml wäßrigem Aceton (Aceton/Wasser = 2:1) wird unter Eiskühlung 124 mg Ozon geperlt. Das Gemisch wird
mit 0,3 ml 30prozentigem Wasserstoffperoxid versetzt und über Nacht in einem Eisschrank stehengelassen. Das Gemisch wird mit
20ml Chloroform und 10 ml Wasser extrahiert, die wäßrige Phase wird nochmals mit '10 ml Chloroform extrahiert. Die Chloroformphasen
werden vereinigt, mit Wasser gewaschen, getrocknet und zur Trockene eingedampft. Das entstandene ölige Produkt wird aus Äther
umkristallisiert. Man erhält 220 mg des gleichen Produkts wie in Beispiel 1.

Beispiel 4
N,N-Bis-(2-chloräthyl)-phosphorsäurediamid-(5-phenyl-3-pentenyl)-ester
wird gemäß Beispiel 1 mit Ozon oxidiert. Man erhält das gleiche Produkt wie in Beispiel 1.
Durch eine Lösung von 400 mg N,N-Bis-(2-chloräthyl)-phosphorsäurediamid-(4-phenyl-3-butenyl)-ester
in einem Gemisch aus 4 ml Aceton und 2 ml Wasser wird unter Eiskühlung 82 mg Ozon geperlt. Das
Reaktionsgemisch wird dann mit 0,3 ml 30prozentigem Wasserstoffperoxid
versetzt und 2 Tage lang unter Kühlung stehengelassen. Das Reaktionsgemisch wird dann mit 5 ml Wasser und 20 ml Chloroform
extrahiert, die wäßrige Phase wird nochmals mit 5 ml Chloroform extrahiert, und die Chloroformextrakte werden vereinigt, mit
Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockene eingedampft. Man erhält einen öligen Rückstand, der aus
Äther unter Kühlung umkristallisiert wird. Man erhält 80 mg des ' gleichen Produktes wie in Beispiel 1.
Durch eine Lösung von 12 g N,N-Bis-(2-chloräthyl)-phosphorsäurediamid-(2-butenyl)-ester
in 40 ml Aceton und 20 ml Wasser wird unter Eiskühlung 3,2 g Ozon geperlt. Das Reaktionsgemisch wird
dann mit 8 ml 30prozentigem Wasserstoffperoxid versetzt und bei

20C 4 Tage lang stehengelassen. Die entstandenen Kristalle werden
filtriert, mit Äther gewaschen, getrocknet und aus Methanol umkristallisiert. Man erhält 8,0 g 2-[Bis-(2-chloräthyl)-amino]-4-hydroperoxy-l~oxa-3-aza-2-phosphacyclcpentan-2-oxid,

Gemäß Beispiel 6 wird N,N-Bis-(2~chloräthyl)-phosphorsäurediamid-(3-phenyl-2-propenyl)-ester
mit Ozon oxidiert. Man erhält 2-[ßis-(2 -chloräthyl) -amino] -^-hydroperoxy-l-oxa-^-aza-^-phosphacy clopentn-2-oxid
in 54prozentiger Ausbeute.
Beispiel 8
Durch eine Lösung von 1,0 g N,N-Bis-(2-chloräthyl)-phosphorsäurediamid-(l-methyl-2-propenyl)-ester
in 20 ml wäßrigem Aceton (Aceton/Wasser -=2:1) wird im Verlauf von 12 Minuten unter Eiskühlung
264 mg Ozon geperlt. Das Gemisch wird mit 1,5 ml 30prozentigem
Wasserstoffperoxid versetzt und bei O0C 2 Tage lang stehengelassen.
Anschließend wird das Aceton im Vakuum abdestilliert und die zurückbleibende wäßrige Lösung mit Chloroform extrahiert. Der

Extrakt wird mit V/asser gewaschen, über wasserfreiem Natriumsulfat
getrocknet und zur Trockene eingedampft. Der kristalline Rückstand wird mit einem Gemisch aus Aceton und Ä'ther gewaschen und aus
Aceton/Chloroform umkristallisiert. Man erhält 48o mg 2-[ßis-(2-chloräthyl)-amino

Durch eine Lösung von 1,05 g N,N-Bis-(2-chloräthyl)-N'-phenylphosphorsäurediamid-(3-butenyl)-ester
in 10 ml wäßrigem Aceton (Aceton/Wasser - 2:1) wird unter Eiskühlung 216 mg Ozon geperlt.
Das Reaktionsgemisch wird mit 0,6 ml 30prozentigem Wasserstoffperoxid
versetzt und 20 Stunden lang bei 20C stehengelassen. Nach dem Einengen im Vakuum wird der Rückstand mit Chloroform
extrahiert, mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockene eingedampft. Das entstandene ölige
Material wird an einer Silicagel-Säule chromatographiert. Man erhält

Gemäß Beispiel 12 wird N,N'-Bis-(2-chloräthyl)-phosphorsäurediamid-(5-phenyl-3-pentenyl)-ester
mit Ozon oxidiert. Man erhält 2-(2-Chloräthyl)-amino-3-(2-chloräthyl)-4-hydroperoxy-l-oxa-3-aza-2-phosphacyclohexan-2-oxid
in M2,6prozentiger Ausbeute.
Durch eine Lösung von 1,4 g Piperidino-N-(2-chloräthyl)-amidophosphonsäure-(3~butenyl)-ester
in 12 ml wäßrigem Aceton (Aceton/ Wasser =2:1) wird unter Eiskühlung 36Ο ml Ozon geperlt. Das
Reaktionsgemisch wird mit 1,0 ml 30prozentigem Wasserstoffperoxid versetzt und 17 Stunden lang bei 20C stehengelassen. Nach dem
Eindampfen im Vakuum wird die zurückbleibende wäßrige Lösung mit

Chloroform extrahiert, der Extrakt wird mit Wasser gewaschen, über wasserfreiem Natriumsulfat getrocknet und zur Trockene eingedampft.
Der Rückstand wird an einer Silicagel-Säule chromatographiert
und mit Aceton eluiert. Das Eluat wird aus Aceton/Chloroform/ Äther umkristallisiert. Man erhält 565 mg 3-(2-Chloräthyl)-4-hydroperoxy-2-piper
idino-l-oxa^-aza^-phosphacyc lohe xan-2-oxid
als farblose Prismen, F. 119°C (Zersetzung).

Durch eine Lösung vom 2 g N,N-Bis-(2-chloräthyl)-N-methy1PhosPhor
säurediamid-(3-butenyl)-ester in 6 ml Aceton und 4 ml Wasser wird unter Eiskühlung 498 mg Ozon geperlt. Das Reaktionsgemisch wird
dann mit 2 ml 30prozentigem Wasserstoffperoxid versetzt und über Nacht bei Raumtemperatur stehengelassen. Nach dem Einengen im
Vakuum wird der Rückstand mit Chloroform extrahiert, der Extrakt wird mit Wasser gewaschen, getrocknet und zur Trockene eingedampft.
Die entstandenen Kristalle werden aus Aceton/Äther umkristallisiert. Man erhält 1,52 g 2- [Bis-(2-chloräthyl)-amino] -4-hydroperoxy-3-methyl-l-oxa-3-aza-2-phosphacyclohexan-2-oxid,

Gemäß Beispiel 15 wird N,N-Diäthyl-N!-(2-chloräthyl)-phosphorsäurediamid-(3-butenyl)-ester
mit Ozon oxidiert. Man erhält 2-Diäthylamino-3-(2-chloräthyl)-4-hydroperoxid-l-oxa-3-a.za-2-phosphacyclohexan-2-oxid,
F, 106 bis 1080C, in 47,5prozentiger Ausbeute.

Gemäß Beispiel 15 wird N,N,N'-Tris-(2-chloräthyl)-phosphorsäurediamid-(3-butenyl)-ester
mit Ozon oxidiert. Man erhält 2-[ßis-(2-chloräthyl)-amino]-3-(2-chloräthyl)-4-hydroperoxy-l-oxa-3-aza-2-phosphorcyclohexan-2*-oxid,
P. 115 bis 117°C in 57,Oprozentiger Ausbeute.
Gemäß Beispiel 15 wird H-(2-Chloi'äbhyD-N1 -phenyIphosphorsäurediamid-(3-butenyl)-ester
mit Ozon oxidiert.Man erhält 2-Phenylamino-

3-(2-chloräthyl)-4-hydroperoxy-l-oxa-3-aza-2-phosphacyelohexan-2-oxid
als öliges Produkt in 32,8prozentiger Ausbeute.

Verfahren nach Anspruch 14, dadurch gekennzeichnet, daß man als inerte Lösungsmittel Methylchlorid, Äthylchlorid, Chloroform, Methylenchlorid, Dichloräthan, Tetrachloräthan, Petroläther, η-Hexan, Cyclohexan, Leichtbenzin, Ligroin, Benzol,Toluol, Tetra-Methylathy^ketonhydrofuran, Dioxan, Acetonyoder em Gemisch dieser Lösungsmitteloder deren Gemisch mit Wasser verwendet.16. Verfahren nach Anspruch 14 und 15, dadurch gekennzeichnet, daß man die Oxidation bei einer Temperatur von -20 bis 35 C durchführt.17· Verfahren nach Anspruch Ik bis 16, dadurch gekennzeichnet,209882/1253daß man dem Reaktionsgemisch nach dem Einleiten des Ozons ein Peroxid, insbesondere Wasserstoffperoxid, zusetzt.18.
