Herstellung von Carotinoidverbindungen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Isozeaxanthin oder 4,4f-Dihydroxy-ß-carotin und Estern hiervon
aus Retrodehydro-ß-carotin. Diese Produkte sind Carotinoid-Pigmente, die zur Färbung von Nahrungsmitteln dienen oder zur
Herstellung anderer Nahrungsmittelpigmente, wie beispielsweise Canthaxanthin, dienen können.
Es ist bekannt, Isozeaxanthin aus von Vitamin A abstammenden Derivaten herzustellen, das seinerseits aus ß-Jonon synthetisiert
wird ,/HeIv. Chira. Acta, 3£, 449 0956^7» doch handelt es
sich um eine Synthese, die zahlreiche Stufen umfasst. Ein Ver- ™
fahren, das den Vorteil hat, dass ein natürliches Produkt als Ausgangsmaterial verwendet wird, besteht darin, ß-Carotin mit
einem Bromierungsmittel, wie N-Bromsuccinimid, Essigsäure und einem organischen alkalischen Mittel zu behandeln und dann den
erhaltenen Diester des 4l4l-Dihydr®xy-ß-carotins zu verseifen.
/HeIv. Chim. Acta, 4^, 402 (1958)7. Diese Synthese erfordert
jedoch niedrige Temperaturen und eine sorgfältige Durchführung,
die bei einer Fabrikation im Grossmassstab schwierig vorzunehmen
ist.
Es wurde nun ein Verfahren zur Herstellung von 4»4'-Dihydroxy-
ß-carotin und dessen Estern mit gesättigten Monocarbonsäuren

gefunden, das darin besteht, Retrodehydro-ß-carotin mit Jod in Anwesenheit einer gesättigten aliphatischen 1-lonocarbonnäure
und einem organischen alkalischen Mittel umzusetzen und gegebenenfalls
den so erhaltenen Ester des 4,-Ί'-Dihydrozy-ß-carotins
zu verseifen.
Die Reaktion der Herstellung des Diesters führt in dein Fall,
in welchem die gewählte aliphatische Säure Essigsäure ist, zum
Isozeaxanthin-diacetat der Formel ηηηητ
Das als Ausgangsprodukt verwendete Retrodehydro-ß-carotin kann nach verschiedenen Methoden aus Vitamin: A und dessen Derivaten
erhalten werden. Man kann es auch ganz einfach aus ß-Carotin
nach dem in der deutschen Patentanmeldung P 20 01 957.2 beschriebenen Verfahren herstellen. Dieses Verfahren
"besteht darin, ß-Carotin mit N-Bromsuccinimid umzusetzen
und dann das erhaltene Bromderivat in Anwesenheit eines Alkalijodids
einer Bromwasserstoffabspaltung zu unterziehen.
Die eingesetzte Jodmenge liegt in einem Molverhältnis von 1 :1 zu derjenigen des behandelten Retrodehydro-ß-carotins vor, und
die molare Menge an organischer Base ist zumindest derjenigen des Jods gleich, doch ist ein Überschuss nicht störend. Diese
organische Base ist vorzugsweise ein aliphatisches, aromatisches, oder heterocyclisches tertiäres Amin, wie beispielsweise
Trialkylamine, N-NlDialkylaniline, Pyridin und dessen Homologe,
N-Alkylpiperidine, W-Alkylmorpholine und dergleichen.
Allgemein können alle gesättigten Carbonsäuren in zumindest stöchiometrischer Menge verwendet werden. Falls man das Ibozeaxanthin
selbst herstellen will, eignet sich Essigsäure sehr gut.

Die Reaktion wird in einem Lösungsmittel für das Isοzeaxanthin,
beispielsweise einem chlorierten Lösungsmittel, wie Chloroform, einem benzolischen Kohlenwasserstoff, Dimethylformamid, Aceton,
Acetonitril, Nitrobenzol oder jedem anderen Lösungsmittel, das unter den Reaktionsbedingungen inert ist, durchgeführt.
Die Temperatur, bei der die Reaktion durchgeführt wird, liegt im allgemeinen zwischen -5 und +35 0C.
Wenn die Bildungsreaktion des Diesters beendet ist, kann das erhaltene Produkt aus dex" Reaktionsmasse isoliert und nach üblichen
Methoden gereinigt oder zur Freisetzung des Isozeaxanthins verseift werden. Man kann zu diesem Zweck ein anorganisches al- gm
kaiisches Mittel verwenden.
Das folgende Beispiel dient zur weiteren Erläuterung der Erfindung.
In einen 500 cm -Kolben, der mit einem Rührsystem, einem aufsteigenden
Kühler und einer Stickstoffzuleitung ausgestattet ist,
bringt man 0,200 g Retrodehydro-ß-carotln und 200 cm Chloroform
ein. Man kühlt den Kolben auf 0 0C ab und setzt 4 cm Essigsäure,
1,60 g N-Äthylmorpholin und dann I9,2 'cnr einer 0,5 % -igen
(Gewicht je Volumen) Lösung von Jod in Chloroform zu. ύ
Nach 1 3/4 stündigem Rühren bei 0 0C wird die Reaktionsmasse
nacheinander mit 100 cm"' V/asser, 100 cm einer wässrigen 2 $-igen
Schwefelsäurelösung und 100 cm einer wässrigen 5 $-igen Natriumcarbonatlösung
gewaschen. Man trocknet die Chloroformlösung über Natriumcarbonat und engt sie dann unter vermindertem Druck
(20 mm Hg) zur Trockne ein, wobei man die Temperatur bei 40 0C
hält. Der so erhaltene Rückstand wird in 4,8 cnr Benzol aufgenommen.
Zu der erhaltenen Lösung setzt man 40 cm einer methanolischen 15 #-igen Kaliumhydroxydlösung zu und rührt eine Stunde,
wobei die Temperatur bei 50 0C gehalten wird. Nach Abkühlen und
anschliessender Zugabe von 100 cm5 Wasser und 20 cnr5 Chloroform
trennt man die Chloroformschicht ab und trocknet sie über

Natriumcarbonat. Dann verdampft man das Chloroform und nimmt

den Rückstand mit 3 cnr Propionitril auf. Durch Erhitzen auf einem Wasserbad erhält man eine Lösung, die man 48 Stunden im Eisschrank stehen lässt. Man saugt über einer Glasfritte ab, wäscht das kristallisierte Produkt 5mal mit je T cm Methanol und trocknet es bei 25 0C unter vermindertem Druck (20 mm Hg). Man erhält so 0,1 g eines kristallisierten roten Produkts vom F = 170 °0 (Kofier-Bank), das bei der U.V.-Spektrographie in Lösung in Äthanol ein Absorptionsmaximum bei 450 nm (E 1 '00 = 215) aufweist. Das Isozeaxanthin wird so in reiner Form in einer Ausbeute von 47 <$, der Theorie erhalten.

Claims (1)
- PatentanspruchVerfahren zur Herstellung von 4,4'-Dihydroxy-ß-carotin und dessen Estern mit gesättigten Monocarbonsäuren, dadurch gekenn zeichnet , dass Retrodehydro-ß-carotin mit Jod in Anwesenheit einer gesättigten Monooarbonsäure und eines alkalischen organischen Mittels umgesetzt wird und gegebenenfalls der so erhaltene Ester des 4J4I-Bihydroxy-ßcarotins zur Freisetzung des 4,4t-Dihydroxy-ß-carotins ver-Beift wird.109830/2023
