gewünschlcnfalls in ein pharmazeutisch annehmbares Salz überführt.
Die erfindungsgemäßen Cephalosporine sind wertvolle Antibiotica hoher Aktivität, aber dennoch ungewohnlich
beständig. Sie sind nicht nur verhältnismäßig beständig gegen Leberenzyme, sondern auch beständig
gegen die Cephalosporinasen, die von gramnegativen Bakterien erzeugt werden. Deshalb bleibt die antimikrobielle
Aktivität der Verbindungen länger erhalten.
Kationen, die mit Säuren pharmazeutisch annehmbare Salze bilden, sind dem Fachmann bekannt. Zu
den für diesen Zweck häufig verwendeten Kationen gehören das Natrium-, Kalium- und Ammoniumion.
Auch die Aminogruppe in der Seitenkette kann durch Behandlung mit Säuren wie Chlorwasserstoffsäure,
Bromwasserstoffsäure und p-Toluolsulfonsäure in ein
Salz übergeführt werden.
Der Austausch der Acetoxygruppe in Stellung 3 kann in wäßriger Lösung bei Temperaturen zwischen
etwa 50 bis HX)C in Gegenwart einer milden Base wie Natriumbicarbonat durchgeführt werden. Vorzugsweise
arbeitet man mit einem Überschuß an Thiol. Die Alkylgruppe in Stellung 1 des verwendeten Tetrazols
oder in Stellung2 des Thiadiazols kann eine Methyl-, Äthyl- oder Propylgruppe sein.
Die eventuell erforderliche Acylierung der 7-Aminogruppc kann mit Hilfe der entsprechenden Säurenalogenide
oder gemischten Anhydride der Phenyl-u-amino- oder Phenyl-./-hydroxy-essigsäure durchgeführt
werden, wobei die <i-Amino- und «-Hydroxygruppen
der Säuren während der Acylierung geschützt sind. Hierzu kann man die Hydroxygruppe in den Ameisensäureester
überführen oder die Aminogruppe durch die tert.-Butyloxycarbonylgruppe oder die 2,2.2-Trichloräthoxycarbonylgruppc
oder durch das mil Methylaceloacetat erzeugte Enamin schützen. Diese
Schutzgruppen werden nach beendeter Acylierung wieder entfernt.
Die Erfindung wird an Hand der folgenden Beispiele
riher erläutert.
B e i s ρ i e 1 1
a) Hinc Mischung von 27,2 g (0,1 Mol) 7-Aminoeephalosporansäure
in 200 ml Wasser und KK) ml Aceton wird unter Rühren mit einer gesäugten Natriumbicarbonatlösung
bis zu einem pH-Wert von 7,9 versetzt. Diese Lösung wird in ein Bad von 80 C
gestellt und. wenn die Innentemperatur 45 C erreicht hat. mit einer Lösung von 19.6g (1.15 Mol) 2-Methyl-1.3.4-thiadiazol-5-thioi
in 2(K) ml Aceton versetzt. Die Mischung wird in dem Bad von 80 C 3 Stunden
lang erwärmt, dann auf 10 C abgekühlt und durch Zugabc von 6n-Salzsäurc auf pH 3,9 eingestellt. Die
kalte Mischung wird 15 Minuten lang gcrihrl, und
der feststoff wird nbfillriert. mit Aceton gewaschen und getrocknet. Is werden 24g (70%) 7-Amino-3-(2-methyl-1,3,4-thiadiazol-5-thiomethyl)-l*-ccphcm-4-carbonsäure
erhalten. Die erwartete Struktur wird durch Kernresonanz- und Ultraviolcltspcklioskopie
bestätigt.
b) 15,2 g (0.1 Mol) D-Mandclsäurc werden in 250 ml
Ameisensäure eingerührt, und die Mischung wird 2 läge lang bei Raumtemperatur stehengelassen. Man
engt die Lösung im Vakuum ein, löst den Rückstand in Ikn/ol. wuscht die Benzollösung mit Wasser, trocknet
über Magnesiumsulfat, filtriert uiidengl mr Trockne ein, wodurch 13,2 g u-ManJelsäurcformialester
erhalten werden. Line Lösung von 3,6 g (0,02 Mol) des n-Mandelsäureformiatesters in 25 ml trockenem
Buizol mit Raumtempcratu! wird mit 5 ml Oxalylchlorid
und 1 Tropfen Dimethylformamid versetzt. Nach 2 Stunden langem Rühren bei Raumtemperatur
wird die Lösung im Vakuum eingeengt, wodurch der Formiatester von n-Mandclylchlorid als öl erhalten
wird, das in 40 ml Aceton gelöst wird. Die Säurechloridlösung wird zu einer eiskalten Lösung von
3,44g (0,01 Mol) des Produkts aus a) und 5 g Natriumbicarbonat
in 100 ml Wasser und 100 ml Aceton gegeben. Diese Mischung wird 1 Stunde in der Kälte
und dann 2 Stunden bei Raumtemperatur gerührt. Das Aceton wird unter vermindertem Druck entfernt,
und die erhaltene wäßrige Mischung wird unter Rühren zu einer kalten Mischung aus 100 ml Wasser
und 200 ml Äthylacetat gegeben. Der pH-Wert wird durch Zugabe von 6n-SaIzsäurc auf 2,0 eingestellt.
Nach Filtrieren durch eine Filterhilfe wird die wäßrige Schicht verworfen und die Äthylacetatschicht mit
Wasser gewaschen, über Magnesiumsulfat getrocknet und im Vakuum zu einem öl eingeengt. Das öl wird
mit Äthrr verrieben, wodurch es fest wird, und der Feststoff wird abfiltriert und getrocknet. Hs werden
4g 7-(D-2-Formyloxy-2-phenylacetamido)-3-(2-methyl
- 1,3.4 - thiadiozol - 5 - thiomethyl) - I3 - eephem-4-carbonsäure
erhalten.
2.64 g (0.54MoI) der formiatgeschützten Thiadiazolcephalosporansäure
in 30 ml Wasser werden mit 2,5 g Natriumbicarbonat versetzt, und die Mischung
wird 3 Stunden bei Raumtemperatur gerührt. Die Mischung wird mit Wasser auf 50 ml verdünnt, mit
50 ml Äthylacetat überschichtet, in einem fiisbad gekühlt
und durch Zugabe von Salzsäure auf pH 2 eingestellt. Die Schichten werden getrennt, und die Äthylacetatschicht
wird mit Wasser gewaschen, über Magnesiumsulfat getrocknet und mit Äther zur Abscheidung
des Produkts aufgeschlämmt. Nach Abfiltrieren werden 1,4g rohe 7-i)-Mandelamido-3-(2-methyl-l,3,
4 - thiadiazol - 5 - thiomethyl) - I3 - cephem - 4 - carbonsäure
erhallen. 1,3 g (0,0026 Mol) der rohen Säure 111
5 ml absolutem Äthanol werden mit 2,6 ml 1 n-Lösung von Natriumacetat in Methanol versetzt. Die Mischung
wird gekühlt und gerührt, und der abgeschiedene Feststoff wird abfiltriert, wodurch 1,1g des
Natriumsalzcs der Ccphalosporinsäurc erhalten werden. Dieses Natriumsalz wird durch Auflösen in Methanol
und Abscheiden durch Zugabe von Isopropanol umkristallisiert. Diese Behandlung liefert Natrium-7
- u-mandclamido- 3-(2- methyl- 1.3,4- thiadiazol-5-thiomethyl)-l'-cephem-4-carboxylat.
Die Struktur wird durch Kernresonanz- und Ultraviolett-Spektroskopic
bestätigt. Die Verbindung hat einen pKa-Wert von 4.9. Die freie Säure wird ferner in das Dicyclohexalaminsal/
übergeführt, dessen Klementaranalyse folgende Werte liefert:

a) Die Arbeitsweise von Beispiel 1 a) wird unter Verwendung von I-Methyl-1.2,3.4-tetrazol-5-thiol an
Stelle des Thiadiazols wiederholt. lis werden 25 g (76%) 7-Amino-3-(l -methyl-1,2.3,4-telrazol-5-thiomclhyl)-ΙΛ
- cephem-4-carbonsäure erhalten. Die
Struktur wird wiederum durch Kernresonanz- und Ultraviolettspektroskopie bestätigt.
b) Die Arbeitsweise vom Beispiel 1 b) wird unter Verwendung des Produkts vom Beispiel 2a)
wiederholt. Es werden 1,1g Natrium-7-i>mandelamido-3-(l
-methyl-1,2,3,4-tetrazol-5-thiomethyl)-l3-cephem-4-carboxylal
erhallen. Die Struktur wird durch Kernresonanz- und Ultraviolettspektroskopie
bestätigt. Das Produkt hat einen pKa-Wert von 5.2.
B ei spi el 3
Eine Lösung von 2,46 g (0,01 Mol) n-Phenylglycin,
dessen Aminogruppe durch die tert.-Butyloxycarbonylgruppe geschützt ist, in 50 ml Tetrahydrofuran wird
in einem Eis/Aceton-Bad auf —10 C gekühlt Diese
kalte Lösung wird mit 1,4 ml Triäthylamin und 1,3 ml Isobutylchlorformiat versetzt. Die Lösung des erhaltenen
gemischten Anhydrids wird 20 Minuten lang in der Kälte gerührt und dann unter Rühren zu einer
kalten Lösung von 3,44 g (0,01 Mol) des Produkts vom Beispiel 1 und 1,4 ml Triäthylamin in 50 ml Wasser
und 50 ml Tetrahydrofuran gegeben. Diese Reaktionsmischung wird 1 Stunde in der Kälte und 2 Stunden
bei Raumtemperatur gerührt. Das Tetrahydrofuran wird im Vakuum entfernt, und die wäßrige Mischung
wird unter Rühren in eine kalte Mischung aus 50 ml Wasser und 100 ml Äthylacetat gegossen. Der pH-Wert
wird durch Zugabe von 6n-Salzsäure auf 2 eingestellt. Die Mischung wird durch eine Filterhilfe filtriert, und
die wäßrige Schicht und die Äthylacetatschicht werden getrennt.
Die Äthylacetatschicht wird mit Wasser gewaschen, über Magnesiumsulfat getrocknet, nitriert und im
Vakuum zur Trockne eingedampft. Der Rückstand wird mit Äther verrieben und nitriert, wodurch 4 g
festes Produkt erhalten werden. Zur Entfernung der tert.-Butyloxycarbonylgruppe wird dieser Feststoff
5 Minuten in 20 ml eiskalter Trifluoressigsäure gerührt, und dann wird die Mischung im Vakuum eingeengt.
Nach Zugabe von 10 ml Äthylacetat wird die Mischung erneut im Vakuum eingedampft. Der Rückstand wird
mit 30 ml Wasser und 30 ml Methylisobutylketon gerührt und bis zu pH 4,0 mit Tributylamin versetzt.
Mach Abfiltrieren des abgeschiedenen Feststoffs werden 0,75 g rohe 3-{2-Mcthyl-l,3,4-thiadiazol-5-thiomethyl)
- 7 - D - phenylglycylamido - I3 - cephem - 4 - carbonsäure
erhalten. Die Wasserschicht wird mil Methylisobutylketon gewaschen, zur Trockne eingedampft,
und der Rückstand wird mit Acetonitril aufgeschlämmt und filtriert, wodurch weitere 0,86 g festes
Produkt erhalten werden. Zur Gewinnung von reinem Produkt werden 0,59 g des bei pH 4 abgeschiedenen
Feststoffs mit 3 ml Formamid. 3 ml Wasser und 1 ml Methanol gerührt, die Lösung wird filtriert, und
es werden 20 ml Acetonitril zugesetzt. Diese Mischung wird mehrere Stunden lang gekühlt, und der abgeschiedene
Feststoff wird abfiltriert, wodurch 0.15 g reines Produkt erhalten werden. Das Kernresonanzspektrum
stimmt mit der angenommenen Struktur überein, und im Bioautogramm ergibt das Produkt
einen einzigen Fleck.

Nach der Arbeitsweise vom Beispiel 3 wird mit 1 - Methyl - 1,2,3.4 - tetrazol - 5 - thiol an Stelle des
Thiadiazols 3-(I -Methyl-1,2,3,4-tetrazol-5-thiomethyl)-7-n-phcnylglycylamidol3-cephem-4-carbon·
säure erhalten. Das Kernresonanzspektrum und da; Bioautogramm des rohen Produkts bestätigen, daß die
gewünschte Verbindung entstanden ist.

Toxizitätsuntersuchung
Zur Ermittlung der Toxizitäten der erfindungsgemäßen Verbindungen werden die entsprechenden Verbindungen
intravenös an Mäuse verabreicht. Die LD50 für die Verbindung gemäß Beispiel 2 b) beträgt
demzufolge 4047 bis 5301 mg/kg. Dieser Wert bewegt sich in dem für Cephalosporine allgemein anerkannten
niedrigen Toxizitätsbereich.
