tionssalze dieser Verbindungen und Arzneimittel, welche diese Verbindungen enthalten.
Die erfindungsgemäßen Verbindungen sind Fluorenone, wenn Z für Sauerstoff steht, und Fluorene, wenn Z
für H2 steht
Beispiele für Alkylengruppen, die durch A dargestellt
werden, sind: 1,2-Äthylen, Iß-Propylen, 1,4-ButyJen,
1,5-Pentylen, 1,6-Hexylen, 2-Methyl-l,4-butylen 2-Äthyl-1,4-butyIen und 3-MethyI-l,5-pentylen. Vorzugsweise ist
Beispiele für R und R1 sind geradketttge oder
verzweigtkettige Alkylgruppen, wie Methyl, Äthyl, n-Propyl, Isopropyl, η-Butyl, sea-Butyl, tert-Butyl,
Isoamyl, n-Pentyl und n-Hexyl.
Die Gruppen R und R1 können an juder basischen
Äthergruppe des Acyclischen Ringsystems gleich oder verschieden sein. Vorzugsweise sind jedoch beide
Gruppen R und beide Gruppen R1 in einer Vorbindung
gleich.
Die erfindungsgemäßen Verbindungen können mit pharmazeutischen Trägern zur Vorbeugung oder
Behandlung von Viruserkrankungen verabreicht werden. Beispielsweise kann die Verabreichung parenteral,
z. B. subcutan, intravenös, intramuskulär oder intrapere-
>ί toneal, oder topisch, beispielsweise intranasal oder
intravaginal, erfolgen. Alternativ oder gleichzeitig kann eine orale Verabreichung vorgenommen werden.
Die verabreichten Dosen hängen vom Virus, dessen Behandlung oder Prophylaxe erwünscht ist, von der Art
«) des Patienten, seinem Alter, seinem Gesundheitszustand, seinem Gewicht, dem Ausmaß der Infektion, der
Art einer gleichzeitigen Behandlung (sofern verwendet), der Häufigkeit der Behandlung und der Natur des
gewünschten Effektes ab. Beispiele für Dosen der
r> verabreichten aktiven Bestandteile sind: intravenös 0,1
bis ungefähr 10 mg/kg; intraperitoneal 0,1 bis ungefähr 50 mg/kg; subcutaii ),\ bis ungefähr 250 mg/kg; oral 0,1
bis ungefähr 500 mg/kg und insbesondere ungefähr 1 bis 250 mg/kg; intranasale Instillation 0,1 bis ungefähr
in 10 mg/kg; und aerosol 0,1 bis ungefähr 10 mg/kg
Körpergewicht des Patienten.
Die aktiven Bestandteile können zusammen mit pharmazeutischen Trägern in Einheitsdosierungsformen verwendet werden, z. B. als Feststoffe wie
4~< Tabletten, Kapseln, Pulver, als flüssige Lösungen, Suspensionen oder Elixiere für orale Verabreichung
oder als flüssige Lösungen für parenterale Verwendung. Die Menge des aktiven Bestandteils in der Dosis ist
natürlich von der Art der Einheitsdosis, der Art des Patienten und seinem Gewicht abhängig. So kann jede
Einheitsdosis ungefähr 1 mg bis ungefähr 30 mg aktiven Bestandteil und vorzugsweise ungefähr 25 bis 5000 mg
aktiven Bestandteil in einem pharmazeutischen Träger enthalten.
In den folgenden Schemata sind Verfahren dargestellt, die zur Herstellung der erfindungsgemäßen
Verbindungen verwendet werden können.

In den obigen Reaktionsschemata besitzen X, Z,
RR1N- und A die gleiche Bedeutung wie vorher angegeben, und jedes Hai bedeutet entweder Cl, Br oder
J.
Typische Beis^cie für Ausgangsmaterialien, die bei
den oben erläuterten Reaktionsschemata Verwendung finden können, sind Diphenole, wie

Typische Halogenalkylamine der Formel III, die im
Schema 1 brauchbar sind, sind z. B. N.N-Diäthyl-2-chlorolthylamin
und N-(2-Chloroälhyl)-piperidin. Typische Dihalogenalkane (V), die im Schema 2 brauchbar sind,
sind z.B. l-Bromo-2-chIoroätb..n und 1,6-Dibromohexan.
Amine (VII), die im Schema 2 brai, .hbar sind, sind sekundäre Amine, wie z. B. Dimethylamin.
In den obigen Reaktionsschemata kann die verwendete »Base« irgendeine Base sein, die mit einem Phenol
ein Salz bildet, wie z. B. Natriummethoxid, Natriumhydrid, Natriumamid, Natriumhydroxid und Kaliumhydroxid.
Die als Reaktionsmedium verwendeten Lösungsmittel können sehr verschieden sein und umfassen
aromatische Kohlenwasserstoffe, wie Benzol, Toluol und Xylol; halogenierte aromatische Kohlenwasserstoffe,
wie Chlorbenzol; aprotische Lösungsmittel, wie Ν,Ν-Dimethylformamid, N.N-Dimethylacelamid und
Dimethylsulfoxid; Alkohole, wie Äthanol und Isopropanol;
Ketone, wie Aceton und Butanon; Äther, wie Tetrahydrofuran und Dioxan; Wasser oder Gemische
daraus.
Bei den Syntheseverfahren, bei denen beispielsweise entweder Natriummcthuxid, Natriumamid oder Nntriumhydrid
als Base verwendet wird, wird die Reaktion in einem wasserfreien Medium, wie z. B. wasserfreies
Toluol oder Chlorbenzol, ausgeführt. Die Base (ungefähr 2,5 Äquivalente) wird zu einer Suspension von
beispielsweise einem Diphenol (1 Äquivalent) im wasserfreien Lösungsmittel zugegeben, und das Gemisch
wird zur Bildung des Diphenoxids erhitzt. In dem Fall, in dem Natriummethoxid verwendet wird, kann das
gebildete Methanol vörteilhafferweise durch azeolmpe
Destillation entfernt werden. Das Halogenid (III oder V) (ungefähr 2,5 Äquivalente) wird dann zugegeben, und
das Gemisch wird während eines Zeitraums von ungefähr 4 bis 24 Stunden unter Rückfluß erhitzt. Die
Produkte (I oder Vl) werden dann durch herkömmliche Verfahren isoliert, wobei I gewöhnlich als Bis-snureadditionssalz
isoliert wird.

Bei der Methode, bei der ein Alkalihydroxid, wie z. B.
Kaliumhydroxid, als Base verwendet wird, können zwei verschiedene Verfahren verwendet werden. Bei einem
Verfahren wird eine konzentrierte wäßrige Lösung (25 bis 50%) des Alkalihydroxids (ungefähr 2,5 Äquivalente)
zu einer Suspension von beispielsweise einem Diphenol (1 Äquivalent) in einem geeigneten aromatischen
Lösungsmittel, beispielsweise Xylol, zugegeben. Dieses Gemisch wird dann zum Sieden erhitzt, wobei
vorzugsweise gerührt wird, und das Wasser wird durch azeotrope Destillation entfernt; ein zweckmäßiges
Verfahren besteht darfc>. daß Wasser in einer Vorrichtung, wie z. B. in einer sogenannten Dean-Stark-Falle, zu
sammeln. Das Reaktionsgemisch, welches nunmehr weitgehend wasserfrei ist, wird dann mit dem Halogenid
(III oder V) (ungefähr 2,5 Äquivalente) wie oben beschrieben behandelt. Beim anderen Verfahren wird
die Reaktion in einem heterogenen Medium aus Wasser und einem aromatischen Kohlenwasserstoff, wie z. B.
Toluol oder Xylol, ausgeführt. Beispielsweise wird ein Diphenol (I Äquivalent) in dem aromatischen Kohlenwasserstoff
suspendiert. Dann wird gemäß Schema 1 eine Lösung von ungefähr 2,5 Äquivalenten eines
Hydrohalogenidsalzes des Aminohalo.renids (d. h. das
Hydrohalogenidsalz von III) im minimal möglichen Wasservolumen zugesetzt, und unter heftigem Rühren
wird eine konzentrierte wäßrige Lösung (25 bis 50%) des Alkalihydroxids (ungefähr 5 Äquivalente) zugesetzt.
Das Gemisch wird ungefähr 6 bis 24 Stunden auf Rückfluß erhitzt, und das Produkt wird von der
Kohlenwasserstoffschicht isoliert. Wenn in Schema 2 das wäßrige/aromatische Kohlenwasserstoff-Medium
zur Herstellung der Verbindungen des Typs Vl verwendet wird, welche keine Aminfunktionen enthalten,
dann wird das Alkalihydroxid nur in einem leichten Überschuß zu 2,0 Äquivalenten je Äquivalent Diphenol
(II) verwendet.
In Schema 2 kann die Reaktion zwischen dem Bis-(o)-halogenalkyl)-äther (Vl) und dem Amin (VII)
unter den verschiedensten Bedingungen ausgeführt werden. Beispielsweise kann die Verbindung (Vl)
gemeinsam mit einem großen Überschuß des Amins (VII) erhitzt werden, wobei das überschüssige Amin als
Reaktionsmedium und als Halogenwasserstoffakzeptor dient. Das Verfahren eignet sich besonders für die leicht
verfügbaren Amine, deren Überschuß leicht vom Rcaktionsgeinisch beispielsweise durch Destillation bei
vermindertem Druck oder durch Dampfdestillation entfernt werden kann. Man kann aber auch den
Bis-((i>-hiilogenalkyl)-älhcr (Vl) (I Äquivalent) und das
Amin (VII) (4 Äquivalente oder mehr) zusammen in einem Lösungsmittel erhitzen, wie z.B. in einem
aromatischen lösungsmittel, beispielsweise Benzol.
Toluol, Xylol oder Chlorbenzol, oder in einem niedrigmolekularen Alkohol, beispielsweise Methanol,
Äthanol oder Isopropanylalkohol, oder in einem niedrigmolekularen Keton, beispielsweise Aceton oder
Methylethylketon, Die Reaktion zwischen der Halogenverbindung und dem Amin wird gewöhnlich durch
Zusatz von Natrium- oder Kaliumiodid gefördert, wobei das Jodid entweder in -katalytischen oder stöchiometrischen
Mengen verwendet wird. In einigen Fällen, wie z. B. wenn das Amin teuer oder nur in ungenügender
Menge vorhanden ist, kann es vorteilhaft sein, nur 2
Äquivalente des Amins (VII) je Äquivalent Bis-(o)-halogenalkyl)-äther
(VI) zu verwenden, wobei ein Überschuß von Natrium- oder Kaliumcarbonat als Akzeptor
für den entstandenen Halogenwasserstoff verwendet wird. Im Falle von flüchtigen Aminen wird diese
Reaktion am besten unter Druck in einer Bombe oder in einem Atuoklaven ausgeführt.
Die obigen Verfahren, die durch die Schemata 1 und 2 dargestellt werden, werden im allgemeinen für die
Herstellung der zweifach basischen Fluorenon- und Fluorenäther bevorzugt, Schema I ist das Verfahren,
welchem im allgemeinen für diejenigen Verbindungen bevorzugt wird, in denen X für

in einer geraden Kette bezeichnet, während Schema 2 im allgemeinen für solche Verbindungen

Kette enthält.
Eine Abwandlung der obigen Verfahren, durch welche die zweifach basischen Äther hergestellt werden

Eine Lösung von 2-Diäthylaminomeihylchlorid [erhalten
aus 15,5 g (0.09 Mol) 2-Diäthylaminoäthylchloridhydrochlorid]
in 100 ml Toluol (getrocknet über Molekularsieben) wurde zu einer Mischung von 6,4 g
(0.03 Mol) 2,7-Dihydroxyfluoren-9on und 3,3g (0,06 :.
Mol) Natriummethoxid in 200 ml Toluol (getrocknet über Molekularsieben) zugegeben. Das Gemisch wurde
3 Stunden unter Rühren und Rückfluß gehalten. Nach dem Abkühlen wurde das Gemisch filtriert, um das
ausgefallene Natriumchlorid abzutrennen. Die Toluollö- in
sung wurde dreimal mit Wasser und dann einmal mit gesättigter Natriumchloridlösung gewaschen und über
wasserfreiem Magnesiumsulfat getrocknet. Dieses Gemisch wurde filtriert und das Filtrat mit ätherischer
Salzsäure gegenüber Kongorot angesäuert. Der ausgefallene Feststoff wurde abfiltriert und aus Butanon
umkristallisiert. wobei genug Methanol zur Bewirkiing
einer Auflösung zugesetzt wurde, worauf das Produkt 24 Stunden »n Vakuum bei 100°C getrocknet wurde. Fp

Eine Mischung aus 63,6 g (030 Mol) 2.7-Dihydroxyfluoren-9-on,
163,0 g (0,95 MoI) 2-DiäthyIaminoäthyl- t,n
chlorid-hydrochlorid und 132 g (2,0 MoI) 85%igem
Kaliumhydoxid in 900 ml Toluol und 300 ml Wasser wurde unter heftigem Rühren 20 Stunden auf Rückfluß
gehalten. Nach dem Abkühlen wurden die Schichten getrennt Die Toiuolschicht wurde dreimal mit Wasser t>5
und dann einmal mit gesättigter Natriumchloridlösung gewaschen und über wasserfreiem Magnesiumsulfat
getrocknet Das Gemisch wurde filtriert, und das Losungsmittel im Vakuum abgetrennt. Der Rückstand

und 1 Teil Methanol umkristallisiert, worauf das Produkt 24 Stunden unter Vakuum bei 100" C getrocknet wurde.

A. Zu einer gerührten, auf Rückfluß gehaltenen Mischung aus 21,2 g (0,10 Mol) 2.7-Dihydroxyfluoren-9-on
und 43,0 g (0,30 MuI) 1 -Brom-2-chloroäthan in
400 ml Wasser wurden tropfenweise während eines Zeitraums von 30 Minuten 80 ml (0,20 Mol) IO°/oiges
wäßriges Natriumhydroxid zugegeben. Nach beendeter Zugabe des Alkalis wurde das Gemisch unter Rühren 18
Stunden auf Rückfluß gehalten. Nach dem Abkühlen wurde die überstehende Schicht abdekantiert und der
Rückstand in Äthanol aufgenommen. Der ausgeschiedene Feststoff wurde abfiltriert, mit Äthanol gewaschen
und in Luft getrocknet; Fp 161 bis 174°C. Das Material
wurde in Chloroform aufgelöst, mit 10%igem wäßrigen Natriumhydroxid und mit Wasser gewaschen und über
wasserfreiem Magnesiumsulfat getrocknet Das Gemisch wurde filtriet, das Lösungsmittel eingedampft und
der Rückstand aus einer Mischung aus 5 Teilen Äthanol
und t Teil Chloroform umkristallisiert; das Produkt wurde an der Luft getrocknet Das Zwischenprodukt

B. Eine Mischung aus 4,4 g (0,013 Mol) 2,7-Bis-(2-chloroäthoxy)-fluoren-9-on,
2 g Kaliumiodid und 100 mi 40%igem wäßrigen Dimethylamin in 50 ml Tetrahydrofuran
wurde unter Rühren 16 Stunden in einem Parr-Druckreaktor auf 122° C erhitzt Das Lösungsmitte!
wurde im Vakuum abgetrennt und der Rückstand in Wasser aufgenommen. Die wäßrige Lösung wurde mit
10%igem Natriumhydroxid basisch gemacht und mit
W) Äther extrahiert. Die Älherlösung wurde zweimal mit
Wasser und dann mit gesättigter Natriumchloridlösung gewaschen und über wasserfreiem Magnesiumsulfat
getrocknet. Das Gemisch wurde filtriert und das Filtrat mit ätherischer Salzsäure gegenüber Kongorot sauer
gemacht. Der Äther wurde vom ausgefallenen (Λ abdekantiert und das öl umkristallisiert, und /war
einmal aus Butanon, wobei ausreichend Methanol zur Bewirkung einer Auflösung zugegeben wurde, und
zweimal aus Methanol allein. Das erhaltene Produkt wurde bei 100°C unter Vakuum getrocknet. Fp 278 bis
280°C(Zers.),A*;;'r' 268 mu, E^. 1800.
Herstellung von 2,7-Bis-[2-(dimethylamino)-äthoxy]-fluoren-9-on-dihydrochlorid
Eine Mischung von 21,2 g (0,10 Mol) 2,7-Dihydroxyliuoren-9-on,
16,0 g (0,296 Mol) Natriummethoxid, 250 ml Chlorbenzol und 60 ml Methanol wurde gerührt
und in einer solchen Weise erhitzt, daß das Methanol abdestillierte, bis das Reaktionsgemisch eine Temperatur
von 130° C erreichte. Das Gemisch wurde dann auf unter 1000C abgekühlt und 32,6 g (030 Mol) 2-Dimethylaminoäthylchlorid
in 71,4 g Chlorbenzol wurden zugegeben. Das Gemisch wurde 18 Stunden bei 1300C
gerührt und auf Rückfluß gehalten und dann auf ungefähr Raumtemperatur abgekühlt Zum Gemisch
wurden 300 ml Wasser und 40 ml 10%iges wäßriges Natriumhydroxid zugegeben, und das Gemisch wurde
30 Minuten gerührt Die obere wäßrige Schicht wurde abgetrennt und mit Chloroform extrahiert Die organischen
Fraktionen wurden vereinigt mit Wasser und

dann mit gesättigter Nalriiimchloridlösung gewaschen
und über wasserfreiem Magnesiumsulfat getrocknet.
Dhs Gemisch wurde abfiltriert und das Lösungsmittel im
Vakuum beseitigt. Der Rückstand wurde in JOO ml Methanol aufgenommen und mit alkoholischer Sal/säure
gegenüber Kongoroi sauer gemacht. Der ausgefallene Fesistoff wurde abfiltriert und aus Methanol
umkristallisiert, uH das Produkt wurde bei IOOnC 24

Herstellung von 2,7-Bis[2-(diäthylamino)-älhoxy]-fluorendihydrochlorid
Eine Lösung von 2-Diüthylaminoäthylchlorid [erhalten
aus 12.9g (0,075) 2-Diäthylaminoäthylchlorid-hydrnchlorid]
in 100 ml Toluol (getrocknet über Molcku-
lcirsieben) wurden zu einer Mischung von 4.9 g (0.025
Mol) 2,7-Dihydroxyi'!noren und 2,7 g (0,05 Mol) Natriummethoxid
in 200 ml Toluol (über Molekularsieben getrocknet) zugegeben. Das Gemisch wurde J Stunden
unter Rühren und Rückfluß erhitzt. Nach dem Abkühlen wurde das Gemisch filtriert, um das ausgefallene
Natriumchlorid zu beseitigen. Die Toluollösung wurde dreimal mit Wasser und dann einmal mit gesättigter
Natriumchloridlösung gewaschen und dann über wasserfreiem Magnesiumsulfat getrocknet. Dieses Gemisch
wurde filtriert, und das Filtrat wurde mit ätherischer Salzsäure gegenüber Kongorot sauer gemacht. Der
ausgefallene Feststoff wurde abfiltriert, in Butanon, welches eine zur Auflösung ausreichende Menge
Methanol enthielt, aufgelöst und mit Holzkohle entfärbt. Das erhaltene Produkt wurde bei 80"C unter Vakuum

Die Verbindung 2,7-Bis[2-(diiithylamino)-äihoxy]-fluorendihydrochlorid,
die im Beispiel 5 angegeben ist, kann auch durch katalytische Reduktion von 2,7-Bis-[2-
(diälhylaminoj-äthoxyj-fluoren^-on-dihydrochlorid
(Beispiel I) hergestellt werden.
Das Verfahren wird in einem Parr-Niedcrdruckhy· drierapparat mit ungefähr 4 g IO°/oigem Palladium auf
Kohle für jeweils 0,02 Mol des Fluorenons bei einer Reaktionstemperatur von 50°C und einer Reaktionszeit
bis zur Beendigung der Reduktion zum Fluoren von 3 bis 6 Stunden durchgeführt Der Anfangswasserstoffdruck
betrug 2,8 bis 3,5 at. Wasser wurde als Reaktionslösungsmittel eingesetzt. Das Produkt wurde
durch Filtrieren und Abdampfen isoliert und wie in Beispiel 5 aus Butanon und Methanol umkristallisiert. Fp

Prüfung der Antivirusaktivität von 2,7-Bis-[3-(diäthylamino)-propoxy]-fluoren-9-on-dihydrochIorid.
Zwei Gruppen Mäuse wurden mit einer tödlichen Dosis (10 LDso) Encephalomyocarditis geimpft. Jede
Maus wog ungefähr 15 g, und jede der zwei Gruppen von Mäusen enthielt 10 bis 20 Tiere. Die Mäuse in einer
Gruppe wurden sowohl prophylaktisch als auch therapeutisch durch subcutane injektion von 2,7-Bis-[3-(diäthylamino)-propoxy]-nuoren-9-on-dihydrochk
>rid behandelt. Die Injektionen wurden 28,22 und 4 Stunden
vor dem Impfen mit dem Virus und 2, 20 und 26 .. jnden
nach dem Impfen durchgeführt. Das Volumen jeder Injektion betrug 0,25 ml und enthielt den aktiven
Bestandteil in einer Menge von 50 mg/kg, gelöst in sterilem Wasser, welches auch 0,15% Hydroxyäthylcellulose
enthielt. Die Vergleichstiere erhielten eine Leerdosis des gleichen Volumenträgers, welches keinen
aktiven Bestandteil enthält. Beobachtungen über 10 Tage zeigten, daß die behandelten Mäuse langer
überlebten als die Vergleichsmäuse.
Eine beispielhafte Zusammensetzung für eine parenterale Injektion ist die folgende, wobei die Mengen auf
Gewicht/Volumen-Basis bezogen sind.
a) 2,7-Bis-(3-piperidinopropoxy)-fluoren-9-on-dihydrochlorid
200 mg
b) Natriumchlorid q.s.
c) Wasser für Injektionen ad 10 ml
Die Zusammensetzung wird dadurch hergestellt, daß der aktive Bestandteil und ausreichend Natriumchlorid
in für Injektionen geeignetes Wasser aufgelöst werden, um eine isotonische Lösung herzustellen. Die Zusammensetzung
kann in einer einzigen Ampulle verkauft werden, die 200 mg aktiven Bestandteil enthält und für
mehrere Dosierungen reicht, oder sie kann in 10 Ampullen für eine einzige Dosierung vertrieben werden.
Fiine beispielhafte Zusammensetzung für harte
GelaliiKknpseln ist die folgende:

einem Nettofüllgewicht von 235 mg/Kapsel eingefüllt.

2.7 g (0,050 Mol) Natriiimmethoxid. 25 ml Methanol und 200 ml Chlorbenzol gerührt und
erhitzt, um das Methanol aus dem Gemisch zu
destillieren. Nachdem der Siedepunkt des Destillats I3(.'"C erreicht hatte, wurde das Gemisch auf unter
100"C abgekühlt, worauf eine Lösung von Din-propyl
aminoäthylchlorid [erhalten aus 21.0 g (0,105) Di-n-propylaminoäthylclilorid-hydrochlond]
in 100 ml Chlorben-/ol zugegeben wurde. Das resultierende Gemisch wun'.'
unter Rühren 6 Stunden auf Rückfluß erhitzt. Das Gemisch wurde wie in Beispiel 4 aufgearbeitet. Die
restliche Base wurde in Bulanon und nicht in Methanol aufgelöst, und das Dihydroehlorid wurde durch Zusatz
von alkoholischer Salzsäure ausgefällt. Der rosafarbene Feststoff wurde abfiltriert und dann aus Butanon
umkristallisiert, welchem eine zur Auflösung arsreichende
Menge Methanol zugesetzt worden war. Das umkristallisiertc Produkt wurde filtriert und unter
Vakuum bei lOOT getrocknet. Fn 194 his I97°C.
