Die Erfindung betrifft ein neues Verfahren aur Herstellung von Benzodiazepinen der allgemeinen Formel I

In der R,R5R und R die vorstehende Bedeutung haben, oder
dessen Salz, mit einem Oxydationsmittel behandelt.
Eh lot Überraschend, dass man nach dem erfindungogemäesen Verfahr
en, au ag eh end von einer Indol-Verbindungfden 5-gliedrigen
Ring zu einem 7-gliedrigen Ring erweitern kann-. Eine derartige
Ringerveiterung ist bisher in der Literatur nicht beschrieben.
Die im Verfahren der Erfindung eingesetzten 2-Aminomethylindole
der allgemeinen Formel II sind neue Verbindungen, die sich in einfacher Weise, z.Bo durch Reduktion von Indol-2-iminoalkyl-Derivaten
herstellen lassen. Diese Verbindungen werden ihrerseits duroh Umsetzung von Indol-2-carbonitrilen mit einem Alkylmagnesiumhalogenid
hergestellt.
Sämtliche vorgenannten Verbindungen können glatt und in hoher Ausbeute erhalten werden« Sc it ist das Verfahren der Erfindung
besonders wertvoll zur technischen Herstellung der Benzodiazepine.

Beispiele für Oxydationsmittel, die im Verfahren der Erfindung verwendet werden können 9 sind Oson, Wasserstoffperoxyd. Per«
säuren, wie Perameisensäure, Peressigsäure und Perbenzoesäure,
Chromsäure und Kaliumpermanganat. Chromsäureanhydrid und Oeon
sind die bevorzugten Oxydationsmitteln
Im allgemeinen wird die Umsetzung bei Raumtemperatur durchgeführt,
Jedoch ergeben manchmal höhere oder niedrigere Arbeits- tempeTatyLTen bessere Ergebnisse, Die Umsetzung wird vorzugsweise in Gegenwart eines Lösungsmittels, wie Wasser, Methanol,
Äthanol, Aceton, Tetrachlorkohlenstoff, Essigsäure, Schwefel-

Bekanntlich sind die Benzodiazepine der vorstehenden allgemeinen
Formel wertvolle Arzneimittel, die ale tPranquilizer, iiuakelrelarantien, Spaemolytika und Hypnotika verwendet werden; rgl.
L.H. Sternbach et al·, J. Org. Chern. Band 27, Seite 3788 (1962).
Eq sind verschiedene Verfahren zur Herstellung dieser Benzodiazepine bekannt. So erhält man sie z.B. in echlechter Auebeute
durch Umsetzung eines 2-Amiaobenzophenon-Deriveta mit o/-subetltuiertem ÜIycin-hydrοChlorid oder dessen Xthylester.
Hierbei fallen die in der 1-Stellung uneubatituierten Benzodiazepine an; vgl· PBP. 1 145 626. Gegebenenfalls kann stan diest
Verbindungen in der 1-Stellung alkylieren; vgl. L.H.Sterabach
et al., J. Org. Chem. Band 27, Seitt 3788 (1962). Benzodiasapine
werden auch durch ümeetaung «Ines Bronao«taBidobtnzoph«noiu Bit
Ammoniak hergestellt; vgl. J. Org. Chem. Band 2-7, Stlte 3788
(1962) und DBP. 1 136 709.
Aufgab· der Erfindung ist ea, ein neu4β und virtichaftlichere·
Verfahren zur Herstellung der Bonzodiasepin· dtr vorstehenden
allgemeinen Forael X zur Verfügung «u »teilen, das glatt und in
hoher Ausbaute verläuft.
Gegenstand der Erfindung iet eonit ein Verfahren zur Herstellung der Benzodiazepine der allgemeinen Torael I, das dadurch
gekennzeichnet ist, dass man ein 2-Aminomethylindol der allgemeinen Forael IZ

säure oder einem anderen inerten Lösungsmittel durchgeführt. Daβ Oxydaticnamlttel wird in mindestens stöchiometrischer Menge
τerwendet. Bei Verwendung von Chromtrioxyd in Essigsäure als
Oxydationsmittel verwendet man vorzugsweise die 2- bis 3-faohe
stöchiometrische Menge an Chromtrioxyd und führt die Umsetzung
bei Raumtemperatur durch. Das 2-Aminomethylindol oder dessen
Sal«, BoB. das Hydrochloric!, Hydrobromld, Sulfat, Nitrat oder
Acetat, wird im Lösungsmittel gelöst oder suspendiert und mit dem Oxydationsmittel unter Rühren versetzt, Die Umsetzung ist im
w allgemeinen innerhalb etwa 24 Stunden beendet. Das rohe Benzodiazepin kann nach der Isolierung ZcB. duroh Umkriatallisation
aus einem Lösungsmittel, wie Äthanol oder Isopropanol, gereinigt werden.
Die erhaltenen Benzodiazepine der allgemeinen Formel I können durch Behandlung mit einer anorganischen Säure, wie Salzsäure,
Schwefelsäure, Salpetersäure oder Phosphorsäure, oder einer organischen Säure, wie Maleinsäure, Fumarsäure, Bernsteinsäure,
Ameisensäure oder Essigsäure, in Ihre Salze überführt werden.
Beispiele für Halogenatome sind Fluor-, Chlor-, Brom- und Jodatome. Beispiele für Alkylreste mit 1 bis 3 Kohlenstoffatomen
sind die Methyl-, Äthyl-, n-Propyl- und Isopropylgruppe. Beispiele für Alkenylreete sind die Allyl-, Butenyl- und Pentenylgruppe. Beispiele für Halogenalkylreste mit 1 bis 3 Kohlenstoff atomen sind die 2,2,2-Trifluoräthyl-, 2-Chloräthyl- und
3-Chlorpropylgruppe. Beispiele für Alkoxygruppen mit 1 bis 3
im Cycloallrylrest

Eine Lösung von 3 g Chrorasäureanhydrid in 3 ml WaBser wird zu
einem Gemisch aus 3 g 2-(ll-Amincäthyl)-5-chlor~3-phenylindolhydrochlorid
und 40 ml Essigsäure unter Kühlung und Rühren gegeben« Das Geraisch wird 15 Stunden bei Raumtemperatur gerührt,
danach mit wässrigem Ammoniak alkalisch gemacht und mit Benzol extrahiert. Die Benzollösung wird über Natriumsulfat getrocknet
und unter vermindertem Druck eingedampft. Der Rückstand wird mit Petroläther versetzt. Hierbei fällt das 7-Chlor-3-methyl»5-ph9nyl-l,3-dihydro»2H~l>4~benzodiazepin-2-on
aus. Nach Umk:. LStallisation aus einem Gemisch von Benzol und Petroläther
schmilzt die Verbindung bei 221 bis 2230C.
Gemäss Beispiel 1, jedoch unter Verwendung von 2~(1'-Aminoisopropyl)-5-chlor-3"phenylindol-hydrochlorid
erhält man das 7-Chlor~3-isopropyl-5~phenyl-l,3-dihydro~2H-lf4-benzodiazepin~
2-on, das nach ümkristallisation aus einer Mischung v.n Äther
und Petroläther bei 226 bis 227°C schmilzt.
Gemäes Beispiel 1, Jedoch unter Verwendung von 2-(ot-Aminobenzyl)-5=-chlor-3-phenylindol
erhält man das 7-Chlor~3,5™diphenyl-l,3~dihydro-2H-l,4-benzodiazepin-2~onf
das nach Umkristaliisatlon aus 2-Äthoxyäthanol bei 273 C schmilzt«
GemäsB' Beispiel 1, jedoch unter Verwendung von 2-(<* -Aminobenzyl)-5-chlor-l-methyl-3"Phenylindol^hydrcohlorid
erhält man daa

7-Chlor-315-diphenyl-l-oethyl-lf 3-dihydro-2H-l ( 4-benzodiazepin-2-on, das nach Umkristallisation aus einer Mischung von Methanol
und Benzol bei 223 his 2240C schallst.
In ähnlicher Welse erhält man gemäse Beispiel 1, jedoch unter
Verwendung von 2-(ll-Aminoäthyl)-5-chlor-3-(o-fluorphenyl)-lmethylindol-hydrochlorid, 2-(l'-Aminoäthyl)-5-chlor-l-äthyl-3-phenylindol-hydrochlorid, 2-(l'-Amlnoäthyi)-5-chlor-3-phenyl-l"
(2',2·,2«-trifluoräthyl)-indol-hydrochlorid, 2-(1·-Amlnoäthyl)-S-chlor-l-cyclopropylmethyl^-phenylindol-hydrochlorid,
2-( i'-Aininoäthyl)-5-inethoxy-l-methyl-3-phenylindol-hydrochlorld
und 2-(r-Aminoäthyl)-l,5~dimethyl-3-phenylindol, das 7-Chlor-5-(o-fluorphenyl)-l,3-dimethy1-, 7-Chior-l-äthyl-3-methy1-5-phenyl-, 7-Chlor-3-methy 1-5-pheny 1-1-(2 ·,2 ·, 2 ·-trlf luormethyl)..t
7-ChloΓ-l-cyclopΓopylmethyl-3-methyl-5·-phβnyl-, 7-Methoxy-l,3-dimethyl-5-phenyl- und lt3t7-Tri^nβthyl-5-phenyl-ll3-dihydro-2H··

Osonhaltiger Sauerstoff wird 6 Stunden bei 100C In ein Gemisch
aus 3 g 2-( C*-Aainobeniyl)-5-chlor-3-ph6nylindol-hydroohlorid
und 40 ml Basigeiure eingeleitet. Danach wird das Reaktionsgemlsch mit Elswaseer verdünnt, mit wässrigem Ammoniak neutralisiert und mit Methylenchlorid extrahiert. Die Methylenchlorldltisung wird getrocknet und das Lösungemittel unter verminderten
Druck abdestilliert. Der Rückstand wird aus einer Mischung von Methanol und Benzol umkristallisiert c Man erhält die gleiche
Verbindung, die in Beispiel 4 beschrieben ist, vom Schmelzpunk; 222 bis 2240C,
