Description
Die Erfindung betrifft ungesättigte Aldehyde der allgemeinen Formel

worin die gestrichelte linie eine von C-Atom4
ausgehende Doppelbindung darstellt, ein Verfahren zu ihrer Herstellung und deren Überführung in Verbindungen vom Typ des Sinensals.
Das erfindungsgemäße Verfahren zur Herstellung von Verbindungen der Formel I ist dadurch gekennzeichnet, daß man eine Verbindung der Formel

auf die im Patentanspruch angegebene Weise ozonisiert und das erhaltene Ozonisierungsprodukt in an sich
bekannter Weise mit einem milden Reduktionsmittel behandelt
Aus der Literatur ist eine O?onisier*mg von Myreen
bekannt, doch wird dort das Ozonisierungsprodukt oxydiert, und zwar zu Bernsteinsäure, s^ehe Pin der.

Als Ausgangsstoffe der Formel II kann man eis- oder
trans-Ocimen oder vorzugsweise Myreen verwenden.
Liegt in den Verbindungen der Formeln I und II die mit der terminalen Doppelbindung konjugierte Doppelbindung in der Kette (wie dies im Ocimen der Fall ist), so
sollen die Formeln I und It nicht nur die eis-, sondern
auch die trans-Isomeren darstellen.
Überraschenderweise verläuft die Ozonisierung der Triene II mit hoher Selektivität, indem die Ozonidbildung praktisch ausschließlich an der isolierten Doppel-
bindung erfolgt, die konjugierten Doppelbindungen also
mit dem Ozon praktisch nicht in Reaktion treten.
Die Ozonisierung kann nach an sich bekannten
Methoden dadurch vorgenommen werden, daß man ozonhaltiges Gas mit dem zu ozonisierenden Trien in so
Kontakt bringt, zweckmäßig durch Einleitung des Gases in eine vorzugsweise verdünnte Lösung des Triens. Als
Lösungsmittel eignen sich vor allem solche, die gegen Ozon inert sind oder wenigstens größere Stabilität
aufweisen als die zu ozonisierende Substanz; z.B. Alkane, wie Hexan, Petroläther, Cyclohexan; Benzol
und dessen Derivate; halogenierte Kohlenwasserstoffe,
wie Tetrachlorkohlenstoff, Chloroform, Methylenchlorid, Methylchlorid, Äthylchlorid, Äthylbromid; Ester,
wie Ameisensäure* oder Essigsäureester (Äthylacetat); μ Ketone, wie Aceton oder Me thy la thy Ike ton; Äther, wie
Dimethyläther, Diäthyläther, Tetrahydrofuran; Säureanhydride, wie Acetanhydrid; Säureamide, wie Formamid, Dimethylformamid; Nitromethan etc. Es kommen
auch solche Lösungsmittel in Frage, die mit dem primär gebildeten Ozonid in Reaktion treten, wie z. B.
Carbonsäuren, beispielsweise Ameisensäure, Essigsäure, Propionsäure; Alkohole, wie Methanol, Äthanol, Propa
nol; Wasser im Gemisch mit Aceton, Am besten eignen
sich Lösungsmittel, die die Ozonisierwngsprodukte fa
Lösung zu halten vermögen. Ferner sind niedrigsiedende Lösungsmittel vorzuziehen, da diese von den
Reaktionsprodukten in der Regel leichter abtrennbar sind. Für die Ozonisierung von Myreen und Ocimen
besonders geeignete Lösungsmittel sind z. B.: Methylchlorid, Chloroform, Tetrachlorkohlenstoff, Benzol,
Aceton, Äthylacetat, Methanol.
Die Konzentration der zu ozonisierenden Lösung kann innerhalb weiter Grenzen variieren. Verdünnte
Lösungen geben im allgemeinen bessere Ausbeuten. Aus praktischen Gründen wird man in der Regel
5—20%ige Lösungen verwenden.
Zweckmäßig läßt man auf das Trien II nicht mehr als etwa 1 Moläquivalent Ozon einwirken, um eine
Oxydation der Reaktionsprodukte zu vermeiden. Normalerweise wird ein Sauerstoffstrom mit einem
Ozongehalt von etwa 2—10% verwendet Es kommen jedoch auch verdünntere und konzentriertere Ozongemische in Frage. Es ist auch die Verwendung von
sauerstofffreiem Ozon (als Gas oder als Lösung) möglich.
Es empfiehlt sich, die Ozonisierung bei Temperaturen unterhalb Raumtemperatur, zweckmäßig bei Temperaturen unterhalb 00C, durchzuführen. Besonders gute
Ausbeuten werden bei Temperaturen im Bereiche von etwa -50 bis -900C erhalten.
Die Spaltung der primär erhaltenen Ozonisierungsprodukte zu den Verbindungen der allgemeinen
Formel I kann nach an sich bekannten Methoden vorgenommen werden.
Die — ebenfalls Gegenstand der vorliegenden Erfindung bildenden — Aldehyde der Formel
können aus den Ozonisierungsprodukten durch Behandlung mit einem milden Reduktionsmittel erhalten
werden, z. B. durch Behandlung mit einem Jodid (z. B,
Natrium- oder Kaliumiodid), Sulfit, Bisulfit (z.B.
Natriumbisulfit), mit Formaldehyd, Schwefeldioxyd, Pyridin, Hydrazinhydrat, mit einem Sulfid (z. B. Dirne*
thylsulfid), mit Hydrochinon, Zink oder Magnesium in
saurer Lösung, mit einem Silbersalz, mit Raney-Nickel
usw.
Die durch Ozonisierung der Triene Il und durch nachfolgende reduktive Spaltung der Ozonisierungsprodukte erhaltenen Aldehyde der Formel sind neue
Verbindungen, die sich als Zwischenprodukte zur Herstellung von Verbindungen mit Orangenaroma
verwenden lassen, insbesondere zur Herstellung des im Orangenöl (Citrus sinensis) vorkommenden /J-Sinensals
(trans*^*SinensaI·, 2,6-Dimgthyl* 1ö-methylen^t^y 1 -dodecatrienal) und von Isomeren desselben (cis-0-Sinensal, eis- und trans-&-Sinensal) (siehe z. B. C. A. 63
[1965], 4338 e, wo die Struktur von Sinensal angegeben
wird), weiche Verbindungen zur Aromatisierung, z. B. von Getränken, Verwendung finden können.
So läßt sich z. B. der aus Myreen erhältliche Aldehyd

Schema: Myrcen (H-I) wird durch Ozonisierung und nachfolgende milde reduktive Spaltung des Ozonisierungsprodukts in den Aldehyd I-l (4-Methylen-5-hexen-1 -al) Obergeführt. Der so erhaltene Aldehyd wird in
einer Wittig-Reaktion mit einem Phosphoran der allgemeinen Formel (II, worin Ph eine Phenylgruppe
darstellt und die beiden R1 niedere Alkylgruppen
bedeuten, die auch unter sich zu einer niederen
Alkylengruppe (z. B. Äthylen) verbunden sein können, zu einem Trienacetal der allgemeinen Formel IV-I
umgesetzt. Die ^ormel IV-I umfaßt das trans-Isomere
IVa-1 und das cis-Isomerc IVb-1.
Die Herstellung des Phosphorans III (wie auch der im folgenden noch genannten Phosphorane) und die
Umsetzung mit der Carbonylkomponente Ib-I können nach den an sich bekannten Methoden der Wittig-Reaktion
erfolgen (vgl. zum Beispiel Angewandte Chemie 71 [1959], 260). Man geht dabei zweckmäßig so vor, daß

man die Carbonylkomponente zu einer frisch bereiteten Lösung oder Suspension des Phosphorans zufügt.

In den nachfolgenden Beispielen sind die Temperaturen
in Celsiusgraden angegeben.
Beispie I 1
In eine auf -80° abgekühlte Lösung von 10g (73,5 mMol) Myrcen in 50 ml Methanol werden
73,5 mMol Ozon in ungefähr 3 Stunden eingeleitet. Dann wird die noch kalte Lösung kurz mit Stickstoff
durchgespült und mit 6,8 g (HOmMoI) Dimethylsulfid versetzt Hierauf wird das Kältebad entfernt und das
Reaktionsgemisch aufwärmen gelassen. Nach ungefähr 1,5 Stunden wird das Gemisch am Rotationsverdampfer
(Badtemperatur maximal 30°) auf ungefähr 1A des
Volumens eingeengt Der Rückstand wird in Äther aufgenommen und die Lösung zweimal mit Wasser

butyl)-triphenyl-phosphoniumjodid werden in 10 ml e-methylen^t^-decadienaläthylenacetal) zum cis-Iso-
Tetrahydrofuran aufgeschlämmt und tropfenweise mit meren IVb-I (4-Methyl-8-methylen-4c3-decadienal-
einer Lösung von 4,5 mMol Butyllithium in Hexan äthylenaceta!) beträgt ca. 1:1. Das Isomerengemisch
versetzt. Unter Rotfärbung der Lösung geht dabei das Phosphoniumjodid als Phosphoran [(4-Äthylendioxybutylidenj-tnphenyl-phosphoranj
in Losung. Nacn ungefähr
10 Minuten gibt man zu der Lösung des Phosphorans 0,25 ml (4,5 mMol) Methyljodid. Dabei
wird die Lösung heller, und das gebildete Phosphoniumsalz
[(4-Äthylendioxy-1 -methylbutyl)-triphenyl-phosphoniumjodid] fällt zum Teil als Öl aus. Nach
10 Minuten tropft man 3,0 mMol Butyllithium in Hexan zu. Die Lösung wird dabei wieder dunkelrot.
Zu dieser, das Phosphoran IH (R1+ R1 = Ä'",ylen)
enthaltenden Lösung gibt man nach 10 Minuten 330 mg (3,0 mMol) 4-MethyIen-5-hexenaI (Ib-I), gelöst in 1 ml
Tetrahydrofuran, zu. Die Lösung entfärbt sich anschließend wieder teilweise. Nach 30 Minuten werden 100 mg
sublimiertes Kalium-tertButylat zugegeben. Das Gemisch
wird 13 Stunden weiter gerührt, dann in Pentan
aufgenommen und der ausgefallene unlösliche Teil abdekantiert Der Pentanextrakt wird anschließend mit
Wasser neutral gewaschen und über Natriumsulfat getrocknet Nach dem Entfernen des Lösungsmittels
wird der Rückstand destilliert Man erhält so 421 mg (63%) eines cis-trans-Isomerengemisches des Trienacetals
IV-1 (R1+ R' = Äthylen) in Form eines Öls mit
Siedepunkt 160°/0,1 mm; nf = 1,4930. IR-Banden bei
läßt sich gaschromalographisch auftrennen.
Das eingangs erwähnte Phosphoniumjodid, (4-Äthylendioxy-butyl)-triphenylphosphoniumjodid, Schmelzpunkt 172-177°, kann wie folgt erhalten werden:
4-ChIorbuttersäurechlorid wird nach Rosenmund zu 4-Chlorbutanal reduziert, dieser Aldehyd mit
Äthylenglykol acetalisiert, das erhaltene Produkt mit Natriumiodid in das entsprechende Jodidacetal (Äthylenacetal des 4-Jodbutarials) übergeführt und letzteres
mitTriphenylphosphin umgesetzt.
146 mg (0,65 mMol) des erhaltenen Trien-äthylenacetals IV-I (als cis-trans-Isomerengemisch) werden in
3,7 ml Dioxan und 1,2 ml 0,1 η-Schwefelsäure gelöst. Die Lösung wird 2 Stunden am Rückfluß gekocht, dann in
Äther aufgenommen, mit Natriumbicarbonatlösung neutral gewaschen und über Natriumsulfat getrocknet.
Rückstand destilliert. Man erhält so 97 mg (83%) eines cis-trans-lsomerengemisches des Trienaldehyds V-I,
4-Methyl-8-methylen-4,9decadienal, vom Siedepunkt 100°/0,1 mm.
Auf analoge Art werden die reinen eis- und trans-lsomeren IVb-I b/w. IVa-I (gewonnen aus dem
Isomerengemisch IV-I mittels präparativer Gaschromatographie) zum cis-lsomeren (4-Methyl-8-methylen-4c,9-decadienal) bzw. zum trans-lsomeren
Va-I (4-Methyl-8-methylen-4t,9-decadienal) verseift,
π -Wert und IR-Spektrum der beiden erhaltenen Isomeren sind identisch: η = 1,4909; IR-Banden bei
2700 m, 1725 s, 1600 m, 900 s cm -'.
175 mg (0,98 mMol) des erhaltenen trans-Trienaldehyds Va-I (4-Methyl-8-methylen-4t,9-decadienal) und
318 mg (I1OmMoI) des Phosphorans Vl, (oc-Formyläthyliden)-triphenylphosphoran, werden in 5 ml Benzol
gelöst. Die Lösung wird 40 Stunden am Rückfluß gekocht, das Benzol hierauf abgesaugt, der Rückstand
mit Pentan versetzt, das ausgefallene Phosphinoxyd abfiltriert und das Pentan wieder verdampft. Das
verbleibende öl wird destilliert. Man erhält so 161 mg (75%) gaschromatographisch reines trans-j3-Sinensal
VIIa-I (2,6-Dimethyl-10-methylen-2t,6t,l 1-dodecatrienal) vom ungefähren Siedepunkt 100°/0,1 mm; n'„"
= 1,0577; IR-Banden bei 1700 s, 160Ow^OOsCm-'.
Auf entsprechende Weise erhält man durch Umsetzung von 158 mg (0,89 mMol) des cis-Trienaldehyds
Vb-I mit 290 mg (0,91 mMol) des Phosphorans Vl 157 mg (81%) cis-0-Sinensal VlIb-I vom ungefähren
Siedepunkt 100°/0,1 mm; η T = 1,5078; IR-Banden bei 1700 s, 1600 w,900 s cm-'.

mit Triphenylphosphin zum Äthyl-triphenylphosphoniumjodid umgesetzt, und dieses mit Butyllithium und
Ameisensäuremethylester zur Reaktion gebracht.
2,0 g (8,0 mMol) von cis/trans-Trienacetal IV-I (R1
= C2H5) werden in 25 ml Dioxan und 8 ml
0,1 η-Schwefelsäure gelöst. Die Lösung wird 3 Stunden bei Raumtemperatur stehengelassen. Dann wird durch
Zugabe von festem Natriumbicarbonat neutralisiert und das Produkt mit Äther extrahiert. Nach Trocknen des
Ätherauszugs über Natriumsulfat, Abdampfen des Lösungsmittels und Destillation des Rückstandes erhält
man 1,1 g (77%) cis/trans-Trienaldehyd V-I vom r>
Siedepunkt 100°/0,1 mm; /?■;:'= 1,4831; IR-Banden bei
173Os, 1600 m, 900 scm-'.
Claims (1)
- Patentansprüche;CHOworin die gestrichelte Linie eine von C-Atorn 4 ausgehende Doppelbindung darstellt.Z Verfahren zur Herstellung der Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß man eine Verbindung der Formeldurch Einwirkung von nicht mehr als etwa zur Oberführung in den Aldehyd behandelt1 Moläquivalent Ozon auf 1 Mol der Verbindung (II) 3. Verwendung der Verbindungen nach Anspruchin Lösung unterhalb Raumtemperatur ozonisiert und 25 I zur Herstellung von einem Tetraenaldehyd derdas erhaltene Ozonisierungsprodukt in an sich folgenden Formelbekannter Weise mit einem milden ReduktionsmittelOHC-C=CH-CH2-CH2-C=CH-CH2-CH2-C-CH=Ch2 evil-!)I I IlCH3 CH3 CH2<qach an sich bekannten Methoden durch Umsetzung der Verbindung mit einem Phosphoran der allgemeiner FormelR1OCH-CH2-CH2-C=P(Ph), R1O CH3worin Ph eine Phenylgruppe und die Symbole R1 niedere Alkylgruppen, die auch unter sich zu einer niederen Alkylengruppe verbunden sein können,bedeuten, zu einem Trienacetal der allgemeinen FormelR1OR1OCH-CH2-CH2-C=CH-CH2-Ch2-C-CH=CH2(IV-I)CH2woriit R1 dasselbe wie oben bedeutet, und Hydrolyse des erhaltenen Trienacetals, gegebenenfalls durch Isolierung des eis- oder des trans-lsomeren, zum Trienaldehyd der FormelOHC-CH2-CH2-C=CH-CH2-Ch2-C-CH=CH2CH3 CH2und Umsetzung hiervon mit einem Phosphoran der allgemeinen FormelOHC-C=P(Ph)jCHjworin Ph die obige Bedeutung besitzt, zum Tetraenaldehyd (VlI-1).(V-I)(Vl)
