Description
Verfahren zur Herstellung von Terpenderivaten Die Erfindung betrifft ein Verfahren zur Herstellung von Verbindungen,der Formel EMI1.1 worin die gestrichelten Linien eine vom C-Atom 3 ausgehende Doppelbindung darstellen. Das erfindungsgemässe Verfahren zur Herstellung von Verbindungen der Formel I ist dadurch gekennzeichnet, dass man eine Verbindung der Formel EMI1.2 ozonisiert und das erhaltene Ozonisierungsprodukt durch Behandlung mit einem milden Reduktionsmittel in einen Aldehyd der Formel EMI1.3 überführt und diesen Aldehyd mit einem Phosphoran der Formel EMI1.4 worin Ph eine Phenylgruppe bedeutet, zu einer Verbindung der Formel EMI1.5 umsetzt. Als Ausgangsstoff der Formel II kann man a-Farnesen oder ss-Famesen oder ein Gemisch der beiden verwenden. Die Formeln I, II und III sollen auch die zu den gezeichneten Strukturen cis-trans-isomeren Strukturen umfassen. So soll z.B. die Formel I sowohl die cisals auch die trans-Formen von a- bzw. ss-Sinensal bezeichnen. Überraschenderweise verläuft in der ersten Stufe des erfindungsgemässen Verfahrens die Ozonisierung des Tetraens II selektiv, indem die konjugierten Doppelbindungen mit dem Ozon praktisch nicht in Reaktion treten. Die Ozonisierung kann nach an sich bekannten Methoden dadurch vorgenommen werden, dass man ozonhaltiges Gas mit dem zu ozonisierenden Tetraen in Kontakt bringt, zweckmässig durch Einleitung des Gases in eine vorzugsweise verdünnte Lösung des Tetraens. Als Lösungsmittel eignen sich vor allem solche, die gegen Ozon inert sind oder wenigstens grössere Stabilität aufweisen als die zu ozonisierende Substanz; z. B. Alkane, wie Hexan, Petroläther, Cyclohexan; Benzol und dessen Derivate; halogenierte Kohlenwasserstoffe, wie Tetrachlorkohlenstoff, Chloroform, Methylenchlorid, Methylchlorid, Äthylchiorid, Athylbromid; Ester, wie Ameisensäure- oder Essigsäureester (Äthylacetat); Ketone, wie Aceton oder Methyläthylketon; Äther, wie Dimethyläther, Diäthyläther, Tetrahydrofuran; Säureanhydride, wie Acetanhydrid; Säureamide, wie Formamid, Dimethylformamid; Nitromethan usw. Es kommen auch solche Lösungsmittel in Frage, die mit dem primär gebildeten Ozonid in Reaktion treten können, wie z. B. Carbonsäuren, beispielsweise Ameisensäure, Essigsäure, Propionsäure; Alkohole, wie Methanol, Äthanol, Propanol; Wasser im Gemisch mit Aceton. Am besten eignen sich Lösungsmittel, die die Ozonisierungsprodukte in Lösung zu halten vermögen. Ferner sind niedrig siedende Lösungsmittel vorzuziehen, da diese von den Reaktionsprodukten in der Regel leichter abtrennbar sind. Besonders geeignete Lösungsmittel sind z. B.: Methylchlorid, Chloroform, Tetrachlorkohlenstoff, Benzol, Aceton, Äthylacetat, Methanol. Die Konzentration der zu ozonisierenden Lösung kann innerhalb weiter Grenzen variieren. Verdünnte Lösungen geben im allgemeinen bessere Ausbeuten. Aus praktischen Gründen wird man in der Regel 5-20 % ige Lösungen verwenden. Zweckmässig lässt man auf das Tetraen II nicht mehr als etwa 1 Moläquivalent Ozon einwirken, um eine Oxydation der Reaktionsprodukte zu vermeiden. Normalerweise wird ein Sauerstoffstrom mit einem Ozongehalt von etwa 2-10% verwendet. Es kommen jedoch auch verdünntere und konzentriertere Ozongemische in Frage. Es ist auch die Verwendung von sauerstofffreiem Ozon (als Gas oder als Lösung) möglich. Es empfiehlt sich, die Ozonisierung bei Temperaturen unterhalb Raumtemperatur, zweckmässig bei Temperaturen unterhalb 0 C, durchzuführen. Besonders gute Ausbeuten werden bei Temperaturen im Bereiche von etwa -50 bis -900 C erhalten. Die Spaltung der primär erhaltenen Ozonisierungsprodukte zu den Verbindungen der Formel III durch Behandlung mit einem milden Reduktionsmittel kann z. B. mit einem Jodid (z. B. Natrium- oder Kaliumjodid), Sulfit, Bisulfit (z. B. Natriumbisulfit), mit Formaldehyd, Schwefeldioxyd, Pyridin, Hydrazinhydrat, mit einem Sulfid (z.B. Dimethylsulfid), mit Hydrochinon, Zink oder Magnesium in saurer Lösung, mit einem Silbersalz, mit Raney-Nickel usw. vorgenommen werden. Die Herstellung des Phosphor ans IV sowie die Umsetzungen desselben mit dem Aldehyd III können nach den an sich bekannten Methoden der Wittig-Reaktion erfolgen (vgl. z. B. Angewandte Chemie 71 (1959), 260). Man geht dabei zweckmässig so vor, dass man den Aldehyd zu einer frisch bereiteten Lösung oder Suspension des Phosphorans zufügt. Wie im nachfolgenden Schema aufgezeigt, kann demgemäss z.B. trans-ss-Sinensal Ia aus trans-ss-Farnesen IIa über den Aldehyd IIIa erhalten werden. Schema EMI2.1 <tb> X <SEP> c4 <tb> <SEP> Ha <SEP> IIIa <tb> <SEP> P(Ph)a <tb> <SEP> w <tb> <SEP> Ph <SEP> = <SEP> Phenyl <tb> <SEP> I <tb> <SEP> ¸ <tb> <SEP> Ia <tb> Die erfindungsgemäss über die neuen Aldehyde III erhältlichen Verbindungen der Formel I stellen Substanzen mit Orangenaroma dar, insbesondere das im Orangenöl (Citrus sinensis) vorkommende ss-Sinensal (trans-ss-Sinensal: 2, 6-Dimethyl- 1 0-methylen- 2t,6t, 1 1-dodecatrienal), als auch Isomere desselben (cis-p-Sinensal, cis- und trans-a-Sinensal), aufgrund welcher Eigenschaften (Geschmack nach Citrusfrüchten) die Verbindungen I zur Aromatisierung, z.B. von Getränken, Verwendung finden können. In den nachfolgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben. Beispiel I 12,5 g (61,3 mMol) trans-ss-Farnesen werden in 120 ml absolutem Methanol gelöst. Bei -900 leitet man dann während 2 Stunden 49 mMol Ozon ein. Es wird dann kurz mit N2 gespült und das Reaktionsgemisch bei 0 mit 6,75 ml (92 mMol) Dimethylsulfid versetzt. Nach 1 :/2stündigem Rühren bei 0 wird das Methanol abdestilliert und der Rückstand an der 10flachen Menge Silicagel (Merck 0,05-0,2) chromatographiert. Durch Eluierung mit Hexan erhält man 6,4 g Ausgangsmaterial (trans-ss-Farnesen). Eluierung mit Benzol ergibt den Trienaldehyd (4-Methyl-8-methylen-4t,9- decadienal), der zwecks Reinigung im Kugelrohr de stilliert wird. (Sdp. 900/0,1 mm; nD =1,4887); IR Banden bei 1730 s; 1600 m; 900 s cm-l. Beispiel 2 175 mg (0,98 mMol) des erhaltenen trans-Trienaldehyds IIIa (4-Methyl-8-methylen-4t,9-decadienal) und 318 mg (1,0 mMol) des Phosphorans W, - (a-Formyl- äthyliden)-triphenylphosphoran, werden in 5 ml Benzol gelöst. Die Lösung wird 40 Stunden am Rückfluss gekocht, das Benzol hierauf abgesaugt, der Rückstand mit Pentan versetzt, das ausgefallene Phosphinoxyd abfiltriert und das Pentan wieder verdampft. Das verbleibende öl wird destilliert. Man erhält so 161 mg (75 SO) gaschromatographisch reines trans-ss-Sinensal Ia (2, 6-Dimethyl-1 0-methylen-2t, 6t, 11 -dodecatrienal) vom ungefähren Siedepunkt 1000/0,1 mm; nD = 1,0577; IR-Banden bei 1700 s, 1600 w, 900 s cm-1. Das Phosphoran W (Schmelzpunkt 220-222o) kann wie folgt erhalten werden: Ätkyljodid wird in Benzol mit Triphenylphosphin zum Sithyl-triphenylphosphoni- umjodid umgesetzt und dieses mit Butyllithium und Ameisensäuremethylester zur Reaktion gebracht.

- PATENTANSPRUCHVerfahren zur Herstellung von Verbindungen der Formel EMI2.2 worin die gestrichelten Linien eine vom C-Atom 3 ausgehende Doppelbindung darstellen, dadurch gekennzeichnet, dass man eine Verbindung der Formel EMI2.3 ozonisiert und das erhaltene Ozonisierungsprodukt durch Behandlung mit einem milden Reduktionsmittel in einen Aldehyd der Formel EMI3.1 überführt und diesen Aldehyd mit einem Phosphoran der Formel EMI3.2 worin Ph eine Phenylgruppe bedeutet, zu einer Verbindung der Formel I umsetzt.UNTERANSPRÜCHE 1. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man trans-ss-Farnesen als Ausgangsstoff der Formel II verwendet.2. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man trans-a-Farnesen als Ausgangsstoff der Formel II verwendet.3. Verfahren nach Patentanspruch, dadurch gekennzeichnet, dass man als Ausgangsstoff der Formel II ein Gemisch von a- und ss-Farnesen verwendet.
