oder 2-Hydroxy-3,5,6-trichlorphenylrest steht, sowie deren pharmazeutisch brauchbaren Salze.
Besonders wertvoll sind:
3.3',5,5',6'-Pentachlor-2,2'-dihydroxybenzanilid, 3,3'.5,5',6-Pentachlor-2,2'-dihydroxybenzariilid,
3,3',4',5,5'-6,6'-Heptachlor-2,2'-dihydroxybenz-
anilid und
3,3',5.5',6,6'-Hexachlor-2,2'-dihydroxybenzanilid.
3,3',5.5',6,6'-Hexachlor-2,2'-dihydroxybenzanilid.
Als pharmazeutisch brauchbare Salze der neuen Benzanilidderivate sollen z. B. Alkali- und Erdalkalisalze.
wie Natrium- und Calciumsalze, und die Salze von ungiftigen organischen Basen, wie Piperazin, genunnt
werden.
Die neuen Benzanilidderivate können durch eine an sich bekannte Umsetzung hergestellt werden, bei
welcher eine entsprechend substituierte Benzoesäure mit einem entsprechend substituierten Anilin umgesetzt
wird.
An Stelle der Benzoesäure können auch reaktionsfähige
Derivate, wie z. B. das Säurechlorid, verwendet werden.
Die Reaktion kann zweckmäßig in Gegenwart eines Verdünnungs- oder Lösungsmittels, z. B. Benzol,
Toluol, Xylol, Diäthyläther, Cyclohexan, Diisobutylen. Tetrahydrofuran, Dimethylformamid und
Di-n-butyläther durchgeführt werden, und sie kann durch Erwärmung beschleunigt oder zum Abschluß
gebracht werden.
Die Reaktion kann gegebenenfalls in Gegenwart einer Base, z. B. Pyridin, Triäthylamin, N,N-Dimethylanilin
oder eines Überschusses des genannten Aminophenolderivats durchgeführt werden.
Die Reaktion kann gegebenenfalls auch in Gegenwart eines anorganischen Katalysators, z. B. Aluminiumchlorid
oder Titanchlorid, durchgeführt werden.
Die Reaktion zwischen der genannten Carbonsäure und dem genannten Anilinderivat kann gegebenenfalls
in Gegenwart eines Entwässerungsmittels, z. B. Ν,Ν'-Dicyclohexylcyanamid, vorgenommen werden.
Die reaktionsfähigen Derivate der genannten Carbonsäure können zweckmäßig in Form einer Lösung
in einem inerten Lösungsmittel dargestellt werden, und die so erhaltene Lösung kann im oben angegebenen
Verfahren ohne Reinigung oder Abtrennung des reaktionsfähigen Derivats der genannten Carbonsäure
verwendet werden. Die Carbonsäure kann also in ein reaktionsfähiges Derivat dadurch umgewandelt
werden, daß sie mit- beispielsweise Thionylchlorid, Phosphor(III)-bromid oder Phosphor(III)-chIorid in
Gegenwart eines inerten Lösungsmittels, wie Benzol, und einer Base, wie Pyridin, in Reaktion gebracht
wird; die entstehende Lösung des reaktionsfähigen Derivats kann zur Herstellung der erfindungsgemäßen
Derivate im oben angegebenen Verfahren Verwendung finden.
Die Benzanilidderivate nach der Erfindung wirken zur Beseitigung von Helminthen, z. B. Lebergel, bei
Haustieren und sind deshalb wertvoll bei der Behänd- M
lung von Menschen und Tieren, die von Helminthen befallen sind. Sie sind dafür gebräuchlichen Mitteln
(Tetrachlorkohlenstoff, Hexachloräthan, »Hexachlorophen«) hinsichtlich des therapeutischen Verhältnisses
(LDs0ZED95) überlegen.
Die Erfindung wird an Hand der Beispiele näher erläutert, wobei alle angegebenen Teile Gewichtsteile
darstellen.
Ein Gemisch aus 6,2 Teilen 3,5-Dichlorsalicylsäure,
3,6 Teilen Thionylchlorid. 35Teilen Benzol und 0,1 Teil Pyridin wird ' J2 Stunde unter Rückfluß erwärmt. Die
entstehende Lösung wird abgekühlt und dazu anschließend 6,4 Teile 2-Amino-3,4,6-trichlorphenoI zugegeben.
Nach löstündiger Erwärmung unter Rückfluß wird das Gemisch abgekühlt und filtriert. Das
feste Filtrat wird mit 20 Teilen einer 9%igen wäßrigen Lösung von Natriumbicarbonat gerührt und das erhaltene
Gemisch filtriert. Das feste Filtrat wird dann aus o-Dichlorbenzol herauskristallisiert, so daß 3,3', A
5,5',6'-Pentachlor-2,2/-dihydroxybenzanilid erhalten ™
wird, das einen Schmelzpunkt von 224° C hat.
Das Verfahren nach Beispiel 1 wird wiederholt, mit dem Unterschied, daß die 6.2 Teile 3,5-Dichlorsalicylsäure
und die 6,4 Teile 2-Amino-3,4,6-trichlorphenol durch molekular äquivalente Mengen an entsprechend
substituierter Salicylsäure bzw. an entsprechend substituiertem 2-Aminophenol ersetzt werden.
Die folgenden Verbindungen werden dementsprechend erhalten:
3,3',5,5',6-Pentachlor-2,2'-dihydroxybenzanilid
(Schmelzpunkt 209 bis 21 IC),
(Schmelzpunkt 209 bis 21 IC),
(Schmelzpunkt 226" C),
3,3',4',5.5',6,6'-Heptachlor-2,2'-dihydroxybenzanilid
(Schmelzpunkt 243CC),
4-Brom-3,3',5,5',6,6'-hexachlor-2,2'-dihydroxybenzanilid
(Schmelzpunkt 2640C),
3.3',4'.5.5',6'-Hexachlor-2,2'-dihydroxybenzaniIid (Schmelzpunkt 215 bis 216 C),
anilid-(Schmelzpunkt 239CC).
21 Teile S.S-Dichlorsalicyloylchlorid werden allmählich
einer gerührten Suspension von 21 Teilen 2-Amino-3,4,6-trichlorphenol in 100 Teilen Toluol zugegeben,
worauf das Gemisch 16 Stunden bei 100° C gerührt wird. Das Reaktionsgemisch wird abgekühlt
und filtriert; das feste Filtrat wird mit Wasser gerührt und dann mit einer 5%igen wäßrigen Natriumbicarbonatlösung
unter Rühren vermengt. Das so erhaltene feste Filtrat wird aus o-Dichlorbenzol umkristallisiert,
wobei 3,3',5.5',6'-PentachIor-2,2'-dihydroxybenzanilid (Schmelzpunkt 224° C) entsteht.
3 a) 4,6 Teile Natrium werden in kleinen Mengen 500 Teilen Methanol zugegeben, und der entstehenden
Natriummethoxydlösung in Methanol 43,6 Teile 3,3',5,5',6,6'-Hexachlor-2,2'-dihydroxybenzanilid zugesetzt.
Das Gemisch wird so lange gerührt, bis sich der schwebende Feststoff gelöst hat, worauf die Lösung
unter vermindertem Druck verdunstet wird. Als Verdampfungsrückstand wird das Dinatriumsalz des
3,3',5,5',6,6'-HexachIor-2.2'-dihydroxybenzanilids erhalten.
3 b) Eine Lösung von 5 Teilen Calciumchlorid in 20 Teilen Wasser wird einer Lösung von 10 Teilen des
Dinatriumsalzes von 3,3'.5,5',6.6'-Hexachlor-2,2'-dihydroxybenzanilid in 100 Teilen Wasser zugegeben
und das Gemisch so lange gerührt, bis sich die Ausscheidung von Feststoff aus der Lösung vollendet hat.
Nach dem Filtrieren des Gemisches wird dieser Feststoff mit Wasser gespült und bei 50 C getrocknet.
Es wird so das Calciumsalz des 3,3',5,5',6,6'-Hexachlor-2,2'-dihydroxybenzanilids
erhalten.
3 c) 20 Teile Piperazinhexahydrat werden einer Lösung von 40 Teilen 3.3 .5,5'.6.6'-Hexachlor-2,2'-dihydroxybenzanilid
in 300 Teilen Methanol zugegeben und das entstehende Gemisch so lange auf Raumtemperatur
gehalten, bis sich die Ausscheidung von Feststoff aus der Lösung im wesentlichen vollendet hat.
Nach Filtrieren des Gemisches wird dieser Feststoff mit Methanol abgespült und dann getrocknet und das
Monopiperazinsalz des 3,3'.5,5'.6,6'-Hexachlor-2.2'-dihydroxybenzanilids
erhalten.
Ein aus 10.75 Teilen 3.5.6-Trichlorsalicylsäure,
3,1 Teilen Thionylchlorid und 50 Teilen trockenem Benzol bestehendes Gemisch wird 30 Minuten unter
Rückfluß erwärmt. Der entstehenden klaren Lösung wird eine aus 4 Teilen Triäthylamin und 7,2 Teilen
2-Amino-4,6-dichlorphenol in 15 Teilen trockenem Benzol bestehende Lösung zugegeben. Nach 12stündiger
Erwärmung unter Rückfluß wird das Gemisch abgekühlt und anschließend mit 100 Teilen einer 5%igen
wäßrigen Natriumhydroxydlösung extrahiert. Der alkalische Extrakt wird mit 5%iger Salzsäure angesäuert,
der ausgeschiedene Feststoff abgefiltert und mit Wasser, gesättigter Natriumbicarbonatlösung und
Essigsäure nacheinander gewaschen. Der entstehende Rückstand wird aus Eisessig umkristallisiert. Es wird
so 3,3',5,5',6 - Pentachlor - 2,2' - dihydroxybenzanilid (Schmelzpunkt 209C) erhalten.
Eine aus 48,6 Teilen 3,5,6-Trichlorsalicylsäure und
35,6 Teilen 2-Amino-4,6-dichIorphenol in 400 Teilen trockenem Tetrahydrofuran bestehende Lösung wird
in einem Eisbad abgekühlt und gerührt, während eine aus 40,8 Teilen Dicyclohexylcarbodiimid in 100 Teilen
Tetrahydrofuran bestehende Lösung innerhalb von 2 Stunden zugegeben wird. Das entstehende Gemisch
wird weitere 12 Stunden bei 18°C gerührt und anschließend filtriert. Das Filtrat wird zur Trockne eingedampft,
worauf der feste Rückstand aus Essigsäure umkristallisiert wird. Es wird so 3,3',5,5',6-Pentachlor-2,2'-dihydroxybenzanilid
(Schmelzpunkt 2090C) erhalten.
Ein aus 48.3 Teilen 3,5,6-Trichlorsalicylsäure, 22 Teilen
Thionylchlorid und 400 Teilen trockenem Toluol bestehendes Gemisch wird gerührt und 45 Minuten
unter Rückfluß erwärmt. Das Toluol wird dann unter vermindertem Druck so weit abdestilliert, bis das
Volumen um etwa zwei Drittel vermindert ist. Dem Gemisch werden nun 25 Teile Pyridin und eine aus
35 Teilen 2-Amino-4,6-dichlorphenol in 250 Teilen warmem Toluol bestehende Lösung zugegeben. Das
entstehende Gemisch wird gerührt und 16 Stunden unter Rückfluß erwärmt. Dann wird unter vermindertem
Druck eingedampft, bis das Volumen um etwa zwei Drittel verringert ist, worauf das Gemisch abgekühlt
und filtriert wird. Das Filtrat wird mit einer gesättigten Natriumbicarbonatlösung und mit Wasser
gewaschen und dann aus Essigsäure umkristallisiert.
Es wird so 3.3',5,5',6-Pentachlor-2,2'-dihydroxybenzanilid (Schmelzpunkt 2090C) erhalten.
Ein aus 3.6 Teilen 2-Amino-4,6-dichlorphenol,
5,8 Teilen 3,5,6-Trichlorsalicylsäure und 40Teilen trokkenem
Toluol bestehendes Gemisch wird gerührt und auf 90 bis 95 C erwärmt, wobei 2,2 Teile Thionylchlorid
innerhalb von 45 Minuten langsam zugegeben werden. Das Gemisch wird dann 16 Stunden gerührt
und unter Rückfluß erwärmt. Das Gemisch wird anschließend unter vermindertem Druck eingedampft
und der dabei entstehende Rückstand mit 40 Teilen warmem Äthanol extrahiert. 10 Teile Wasser und
1 Teil Aktivkohle werden dem äthanolischen Extrakt zugegeben, und das erhaltene Gemisch dann filtriert.
Nun werden 20 Teile Wasser zugesetzt und der dabei ausgeschiedene Feststoff abgefiltert, mit gesättigter
Natriumbicarbonatlösung und dann mit Wasser gewaschen und anschließend aus Essigsäure umkristallisiert.
Es wird so 3,3',5,5',6-Pentachlor-2,2'-dihydroxybenzanilid (Schmelzpunkt 2090C) erhalten.
7a) Das Verfahren nach Beispiel 3a) wird wiederholt, mit dem Unterschied, daß die 43,6 Teile 3,3',5,5'.
6,6' - Hexachlor - 2,2' - dihydroxybenzanilid durch 40,1 Teile 3.3',5.5',6-Pentachlor-2,2'-dihydroxybenzanilid
ersetzt werden. Es wird so das Dinatriumsalz des 3,3'.5.5',6-Pentachlor-2,2'-dihydroxybenzanilids
erhalten.
7b) Das Verfahren nach Beispiel 3b) wird wiederholt, mit dem Unterschied, daß die 10 Teile Dinatriumsalz
von 3.3',5,5',6,6'-HexachIor-2,2'-dihydroxybenzanilid durch 9,2 Teile Dinatriumsalz von 3,3',5,5',
6-PentachIor-2,2'-dihydroxybenzanilid ersetzt werden.
Es wird also das Calciumsalz des 3,3',5,5',6-Pentachlor-2.2'-dihydroxybenzaniIids
erhalten.
7 c) Das Verfahren nach Beispiel 3 c) wird wiederholt, mit dem Unterschied, daß die 40 Teile 3,3',5,5',
6,6-Hexachlor-l^'-dihydroxybenzaniliddurch38TeiIe
3.3',5.5',6-Pentachlor-2,2'-dihydroxybenzaniIid ersetzt werden. Es wird so als Iederfarbener Feststoff das
Monopiperazinsalz des 3,3',5,5',6-PentachIor-2,2'-dihydroxybenzanilids
erhalten.
Ein Gemisch aus 10 Teilen 4'-NitrophenyI-3,5,6-trichlorsalicylat,
9,8 Teilen 2-Amino-4,6-dichIorphenoI und 120 Teilen Dimethylformamid wird 20 Stunden
auf 100: C erwärmt. Die entstehende Lösung wird im
wannen Zustand auf 100 Teile Eis gegossen. Der dadurch erzeugte feste Niederschlag wird aus Essigsäure
in Gegenwart von Knochenkohle umkristallisiert. Es wird so 3,3\5,5',6-Pentachlor-2,2'-dihydroxybenzanilid
(Schmelzpunkt 2090C) erhalten.
Das in diesem Verfahren als Ausgangsstoff benutzte 4'-Nitrophenyl-3,5,6-trichlorsalicylat war wie
folgt erzeugt worden:
Ein Gemisch aus 12,1 Teilen 3,5,6-Trichlorsalicylsäure,
7 Teilen 4-NitrophenoI, 3,5 Teilen Phosphorpentachlorid und 75 Teilen Xylol wird 3,5 Stunden
unter Rückfluß erwärmt. Die erhaltene Lösung wird 16 Stunden auf 18° C gehalten und dann einer Wasserdampfdestillation
unterworfen. Der feste Rückstand wird mit wäßriger Natriumbicarbonatlösung zerrieben
und aus Benzol umkristallisiert. Es wird so 4'-Nitrophenyl-3,5,6-trichlorsalicylat (Schmelzpunkt
178C) erhalten.

Ein Gemisch aus 12 Teilen 3,5,6-Trichlorsalicylsäure,
2,5 Teilen Phosphortrichlorid und 24 Teilen Toluol wird 2 Stunden unter Rückfluß erwärmt.
Diesem Reaktionsgemisch werden dann 8 Teile 2 - Amino - 4,6 - dichlorphenol zugegeben und dann
12 Stunden gerührt und unter Rückfluß erwärmt. Das Gemisch wird nach erfolgter Abkühlung filtriert,
das Filtrat mit Wasser und dann mit gesättigter wäßriger Natriumbicarbonatlösung gewaschen. Der feste
Rückstand wird aus Essigsäure umkristallisiert. Es wird so 3,3',5,5',6-Pentachlor-2,2'-dihydroxybenzanilid
(Schmelzpunkt 209 bis 2110C) erhalten.
Ein Gemisch aus 6 Teilen 3,5,6-TrichIorsalicylsäure,
4 Teilen Phosphortrichlorid und 24 Teilen Toluol wird 30 Minuten unter Rückfluß erwärmt. Diesem
Reaktionsgemisch werden dann 8 Teile 2-Amino-4,6-dichlorphenoI
zugegeben, 16 Stunden gerührt und unter Rückfluß erwärmt. Anschließend wird abgekühlt
und filtriert. Das Filtrat wird aus Essigsäure umkristallisiert. Es wird so 3,3',5,5',6-Pentachlor-2,2'-dihydroxybenzanilid
(Schmelzpunkt 209 bis 21Γ C) erhalten.
Claims (1)
- Patentansprüche:
1. Benzanilid der allgemeinen FormelHO ClR-CO-NHin der R für den 2-Hydroxy-3,5,6-trichlorphenylrest steht, sowie deren pharmazeutisch brauchbaren Salze.
2. Benzanilide der allgemeinen FormelHO CIR-CO-NHCl Clin der R für den 2-Hydroxy-3,5-dibromphenyl-, 2-Hydroxy-3,5-dichlorphenyl-, 2-Hydroxy-3,5,6-trichlorphenyl- oder 2-Hydroxy-3,5,6-trichlor-4-bromphenylrest steht, sowie deren pharmazeutisch brauchbaren Salze.
3. Benzanilide der allgemeinen FormelHO ClR-CO-NHCl CIin der R für den 2-Hydroxy-3,5-dichlorphenyl- oder 2-Hydroxy-3,5,6-trichlorphenylrest steht, sowie deren pharmazeutisch brauchbaren Salze.4. Verfahren zur Herstellung des Benzanilids nach Anspruch 1, dadurch gekennzeichnet, daß man in an sich bekannter Weise 2-Hydroxy-3,5,6-trichlorbenzoesäure mit 2-Hydroxy-3,5-dichloranilin umsetzt und gegebenenfalls mit einer Base reagieren läßt.5. Verfahren zur Herstellung der Benzanilide nach Anspruch 2, dadurch gekennzeichnet, daß man in an sich bekannter Weise 2-Hydroxy-3,5-dibrom-, 2 - Hydroxy - 3,5 - dichlor -, 2 - Hydroxy-3,5,6 - trichlor - oder 2 - Hydroxy - 3,5,6 - trichlor-4-brombenzoesäure mit 2-Hydroxy-3,5,6-trichloranilin umsetzt und gegebenenfalls mit einer Base reagieren läßt.6. Verfahren zur Herstellung der Benzanilide nach Anspruch 3, dadurch gekennzeichnet, daß man in an sich bekannter Weise 2-Hydroxy-3,5-dichlor- oder 2-iIydroxy-3,5,6-trichlorbenzoesäure mit 2-Hydroxy-3,4,5,6-tetrachloranilin umsetzt und gegebenenfalls mit einer Base reagieren läßt.
