b) Exposition der Testzelle mit einem bekannten GPCR-Agonisten unter Versuchsbedingungen und dann

worin die Translokation des detektierbaren Moleküls in der Testzelle die Aktivierung des GPCR-Stoffwechseis
anzeigt.
18. Verfahren gemäß Anspruch 17, worin die Versuchsbedingungen die Anwesenheit einer Test-Kinase
in der Testzelle bedeuten.
19. Verfahren gemäß Anspruch 17, worin die
Versuchsbedingungen die Anwesenheit eines Test-G-Proteins in der Testzelle bedeuten.

dessen Fragments auf die Fähigkeit, einen phosphorylierten GPCR zu binden, umfassend:
a) Bereitstellung einer Zelle, die: i) einen GPCR exprimiert und
ii) ein Konjugat enthält, das ein ß-Arrestin-Test-Protein
und ein visuell detektierbares Molekül umfasst;
b) Exposition der Zelle mit einem bekannten GPCR-Agonist und dann

c) Detektion der Translokation des detektierbaren
Moleküls vom Zytosol der Zelle zum Membranrand der Zelle;
worin die Translokation des detektierbaren Moleküls in der Testzelle die Bindung des ß-Arrestin-Proteins

23. Verfahren zum Screenen einer Testverbindung auf die Agonist-Aktivität für G-Protein-gekoppelten Rezeptor
(GPCR), umfassend:
a) Bereitstellung einer Zelle, die einen GPCR

Membranrand der Zelle zum Zytosol der Zelle nach der Exposition der Zelle mit der Testverbindung anzeigt,
dass die Testverbindung GPCR-Antagonist-Aktivität

59. Verfahren gemäß Anspruch 54, wobei die Testverbindung in wässriger Lösung ist.
60. Verfahren gemäß Anspruch 54, wobei die Zellen auf
einem Substrat aufgebracht sind.
61. Verfahren zum Screenen einer Zelle auf die Anwesenheit eines G-Protein-gekoppelten Rezeptors
(GPCR), umfassend:
a) Bereitstellung einer Testzelle, wobei die
Testzelle ein Konjugat enthält, das ein ß-Arrestin Protein und ein visuell detektierbares
Molekül umfasst;
b) Exposition der Testzelle mit einer Lösung, die einen GPCR-Agonist enthält und
c) Detektion der Translokation des detektierbaren Moleküls vom Zytosol der Zelle zum Membranrand
der Zelle;
worin die Bewegung des detektierbaren Moleküls vom Zytosol zum Membranrand der Testzelle nach der
Exposition der Testzelle mit dem GPCR-Agonist anzeigt, dass die Zelle einen GPCR enthält.
62. Verfahren gemäß Anspruch 61, worin das detektierbare
Molekül optisch detektierbar ist.
63. Verfahren gemäß Anspruch 61, worin das detektierbare Molekül grün fluoreszierendes Protein ist.
64. Verfahren zum Screenen einer Vielzahl von Zellen auf
solche Zelle, die einen G-Protein-gekoppelten Rezeptor (GPCR) enthalten, umfassend:

a) Bereitstellung einer Vielzahl von Testzellen, die ein Konjugat enthalten, das ein ß-Arrestin-Protein
und ein visuell detektierbares Molekül umfassen;
b) Exposition der Testzellen mit einem bekannten GPCR-Agonist und
c) Detektion solcher Zellen, in welchen das detektierbare Molekül vom Zytosol der Zelle zum
Membranrand der Zelle translokalisiert ist worin die Bewegung des detektierbaren Moleküls vom
Zytosol zum Membranrand der Zelle nach der Exposition mit dem GPCR-Agonist anzeigt, dass die Zelle einen
GPCR für diesen bekannten GPCR-Agonist enthält.
65. Verfahren gemäß Anspruch 64, worin das detektierbare Molekül optisch detektierbar ist.
66. Verfahren gemäß Anspruch 64 worin das detektierbare Molekül grün fluoreszierendes Protein ist.
67. Verfahren gemäß Anspruch 64, worin die Vielzahl der Testzellen in einem Gewebe enthalten sind.
68. Verfahren gemäß Anspruch 64, worin die Vielzahl der Testzellen in einem Organ enthalten sind.
69. Verfahren gemäß Anspruch 64, worin Schritt (b) die Exposition der Testzellen mit einer Vielzahl
bekannter GPCR-Agonisten umfasst.
70. Substrat, auf dem eine Vielzahl von Zellen aufgebracht ist, die einen GPCR exprimieren und ein
Konjugat enthalten, das ein ß-Arrestin-Protein und ein detektierbares Molekül umfasst.
71. Substrat gemäß Anspruch 70, worin das detektierbare Molekül ein optisch detektierbares Molekül ist.
72. Substrat gemäß Anspruch 70, worin das detektierbare
Molekül grün fluoreszierendes Protein ist.
73. Substrat gemäß Anspruch 70, worin die Zelle eine Säugetierzelle ist.
74. Substrat gemäß Anspruch 70, worin die Zelle ausgewählt ist aus der Gruppe bestehend aus
Bakterienzellen, Hefezellen, Pilzzellen, Pflanzenzellen und Tierzellen.
75. Substrat gemäß Anspruch 70, worin die Zelle einen olfaktorischen GPCR exprimiert.
76. Substrat gemäß Anspruch 70, worin die Zelle einen ßadrenergischen
GPCR exprimiert.
77. Substrat gemäß Anspruch 70, worin das Substrat
hergestellt wird aus einem Material, ausgewählt aus der Gruppe, bestehend aus Glas, Kunststoff, Keramik,
Halbleiter, Kieselgel, Glasfaser, Diamant, biokompatiblen Monomer- und biokompatiblen Polymer-Materialien.
78. Gerät zur Bestimmung der GPCR-Aktivität in einer
Testzelle, umfassend:
Vorrichtung zur Messung von Hinweisen auf die intrazelluläre Verteilung eines detektierbaren
Moleküls und
eine Computerprogramm-Vorrichtung, welche ein computerlesbares Speichermedium umfasst, das in
diesem Medium eine Vorrichtung für einen computerlesbaren Programmcode eingeschlossen enthält,
wobei die Vorrichtung für einen computerlesbaren Programmcode umfasst:
eine Vorrichtung für einen computerlesbaren Programmcode zur Bestimmung, ob die Hinweise auf
die Verteilung des detektierbaren Moleküls in der Testzelle die Konzentration des detektierbaren
Moleküls an der Zellmembran anzeigen, basierend
auf dem Vergleich der gemessenen Hinweise der intrazellulären Verteilung des detektierbaren
Moleküls in derKontrollzelle.
79. Gerät zur Bestimmung der GPCR-Aktivität in einer Testzelle, umfassend:
Vorrichtung zur Messung von Hinweisen auf die intrazelluläre Verteilung eines detektierbaren
Moleküls in wenigstens einer Testzelle zu vielen verschiedenen Zeitpunkten und eine Computerprogramm-Vorrichtung,
die ein computerlesbares Speichermedium umfasst, welches eine Vorrichtung für einen
computerlesbaren Programmcode in diesem Medium eingeschlossen hat, wobei die Vorrichtung für den
computerlesbaren Programmcode umfasst:
eine Vorrichtung für einen computerlesbaren Programmcode zu Bestimmung, ob die Hinweise auf
die Verteilung des detektierbaren Moleküls in der Testzelle zu vielen verschiedenen Zeitpunkten die
Translokation des detektierbaren Moleküls zur
Zellmembran anzeigen.
80. Gerät zur Bestimmung der GPCR-Aktivität in einer Testzelle, umfassend:
Vorrichtung zur Messung von Hinweisen auf die intrazelluläre Verteilung eines detektierbaren
Moleküls in mindestens einer Testzelle und eine Computerprogramm-Vorrichtung, die ein
computerlesbares Speichermedium umfasst, welches einen Vorrichtung für einen computerlesbaren
Programmcode in diesem Medium eingeschlossen enthält,

detektierbaren Moleküls an der Zellmembran anzeigt.

81. Gerät nach Anspruch 78, worin die genannten Hinweise auf die intrazelluläre Verteilung eines
detektierbaren Moleküls optische Hinweise sind.
82. Gerät nach Anspruch 78, worin die Messvorrichtung eine Vorrichtung zur Messung der
Fluoreszenzintensität umfasst.
83. Gerät nach Anspruch 78, worin das zu detektierende Molekül ein über die Fluoreszenz zu detektierendes
Molekül ist und worin der Schritt der Messung der Hinweise auf die intrazelluläre Verteilung des
detektierbaren Moleküls die Messung des Fluoreszenzsignale der Testzelle und von
Kontrollzellen umfasst.
