本发明背景
在造成农作物损失和使它们得以控制的花费方面,昆虫和其它害虫每年都要耗费农民几十亿美元。昆虫在农业生产环境上造成的损失包括农作物产量的减少和农作物质量的降低。收获成本的增加也是昆虫大批出现的一个结果。
昆虫问题已部分地由栽培方法如庄稼轮作,以及由施肥刺激植物的状根生长的高水平的磷酸盐而解决。但是,庄稼轮作可被例如长角叶甲的出现两年滞育(或越冬)性状而破坏。化学杀虫剂严重地依赖于保证所需的控制水平。
化学杀虫剂提供了控制害虫的有效方法。但是,公众已开始关注在食物、地下水和环境中其它地方发现的残余化学药剂的数量。因而,越来越多的仔细审查合成的化学杀虫剂的潜在的对环境不利的后果。某些合成化学杀虫剂能够毒化土壤和下面的蓄水层,作为流走物的结果污染地表水,以及破坏非靶生物形式。某些合成化学控制药剂进一步的缺点是,在用于宠物、家畜或儿童可能接触到的区域时,成为公众安全公害。它们还会对施药的人产生健康损害,特别是他们没有遵守正确的施药方法时。世界上的管制机构正在限制和/或禁止多种合成杀虫剂的使用,特别是那些在环境中持续存在的和进入食物链的杀虫剂。对杀虫剂使用的新的严格限制和从市场消除某些有效的杀虫剂会限制控制花费很多的害虫的经济和有效的选择。
因为这些与使用多种合成化学杀虫剂相关的问题,所以存在对限制这些药剂使用和确定替代控制药剂的明确需要。取代合成的化学杀虫剂,或将这些杀虫剂和生物杀虫剂组合使用,能够降低环境中的毒性化学剂。
日益普及使用的生物杀虫剂是土壤微生物苏云金芽孢杆菌(Bacillus thuringiensis)(B.t.)。土壤微生物B.t.是革兰氏阳性、形成芽孢的细菌。许多B.t.菌株不显示杀虫活性。有些B.t.菌株产生杀虫的伴孢蛋白质包涵体。这种蛋白质包涵体在显微镜下常显现为特殊形状的晶体。晶体包涵体的形状和类型可用于表征B.t.菌株。在晶体包涵体中存在的典型的具有专一杀虫活性的“δ-内毒素”与不具有专一宿主范围的外毒素是不同的。
B.t.杀虫剂的商业使用起初限于小范围的鳞翅目(毛虫)害虫。例如,苏云金芽孢杆菌库斯塔克亚种(B.thuringiensis subsp.kustaki)的芽孢和晶体的制备物已经作为鳞翅目害虫的商品杀虫剂使用多年。但是最近,研究人员已发现对更广范围的害虫具有专一性的B.t.杀虫剂。例如,苏云金芽孢杆菌以色列亚种(B.t.israelensis)和莫里逊(morrisoni)亚种已经作为商品分别用于控制双翅目和鞘翅目昆虫。(Gaertner,F.H.[1989]“Cellular Delivery of CropProtection:Living and Non-LivingMicroorganisms”在ConlrolledDelivery of Crop Protection Agents,R.M.Wilkins,ed.,Taylor和Francis,New York和London,1990,PP.245-255”)。
已经鉴定了B.t.的新亚种,并分离和测序了活性δ-内毒素蛋白质的基因(Hfte,H.,H.R.Whiteley[1989]Microbiological Reviews52(2):242-255)。Hfte和Whiteley将B.t.晶体蛋白质基因分为四个主要种类。各种类为cryⅠ(鳞翅目专一的),cryⅡ(鳞翅目和双翅目专一的),cryⅢ(鞘翅目专一的),和cryⅣ(双翅目专一的)。已经报道了对其它害虫有专一毒性的菌株(Feitelson,J.S.,J.Payne,L.Kim[1992]Bio/Technology 10:271-275)。例如,已建议用CryⅤ和CryⅥ命名两种新的线虫-活性毒素。
多种苏云金杆菌δ-内毒素蛋白质由两个功能片段组成。这些“全长”蛋白质含有一个抗蛋白酶的核心毒素,对应于蛋白质分子的大约前半部分(N-末端部分)。已经知道了一种CryⅢA B.t.δ-内毒素核心片段的三维结构,并认为所有相关的毒素具有相同的整体结构(Li,J.,J.Carroll,D.J.Ellar[1991]Nature 353:815-821)。分子的第二半部分(C-末端部分)通常称为“前毒素片段”。相信前毒素片段参与毒素晶体的形成(Arvidson,H.,P.E.Dunn,S.Strand,A.I.Aronson[1989]Molecular Microbiology3:1533-1534;Choma,C.T.,W.K.Surewicz,P.R.Carey,M.Pozsgay,T.Raynor,H.Kaplan[1990]Eur.J.Biochem.189:523-527)。在昆虫肠内,典型的由蛋白酶将全长的130kDa毒素分子加工成抗性核心片段。因而前毒素片段可能通过降低蛋白酶对毒素分子的加工(Haider,M.Z.,B.H.Knowles,D.J.Ellar[1986]Eur.J.Biochem.156:531-540)或降低毒素溶解性(Aronson,A.I.,E.S.Han,W.McGaughey,D.Johnson[1991]Appl.Environ.Microbiol.57:981-986)限制核心对昆虫的可及性从而赋予毒素部分昆虫专一性。

分离新的B.t.分离物和它们的毒素基因,以及测定毒素的精确杀虫范围,是一个缓慢的经验的过程。随着广泛的研究和物力投资,发布了新B.t.分离物、毒素、基因以及已知B.t.毒素和分离物的新用途的专利。作为综述参看前面Feitelson等的文章。但是发现新的B.t.分离物以及已知B.t.分离物和毒素的新用途仍然是经验性的,不可预知的技术。
Smulevitch等([1991]FEBS Lett.293:25-26),Gleave等([1991]JGM 138:55-62),和Schevelev等([1993]FEBS Lett.336:79-82)描述了对鳞翅目昆虫有针对活性的Cry9毒素的特征。Lambert等(Lambert,B.,A.Van Vliet,M.Peferoen[1996]Appl.Environ.Microbiol 62(1):80-86)以及公布的PCT申请WO94/05771和WO94/24264中也描述了对鳞翅目害虫有针对活性的B.t.分离物和Cry9C毒素。
美国专利号5,126,133;5,188,960;和5,691,308公开了一个来自B.t.分离物PS81Ⅰ的Cry1Fa毒素和基因(81ⅠA)。美国专利号5,527,883;5,508,264;5,827,514;和5,840,554涉及CryⅠF嵌合毒素。WO99/24581公布了多种植物最优化的Cry1F基因。美国专利号5,686,069公布了来自B.t.分离物PS91C2的Cry1Fb毒素和基因。
使用基因工程技术,正在开发多种向农业环境施用B.t.毒素的方法。早在15年之前,在公开文献中已描述了在大肠杆菌(E.coli)中克隆和表达B.t.晶体蛋白质基因(Schnepf,H.E.,H.R.Whiteley[1981]Proc.Natl.Acad.Sci.USA 78:2893-2897)。美国专利号4,448,885和美国专利号4,467,036均公开了B.t.晶体蛋白质在E.coli中的表达。已经产生了基于-重组DNA的最B.t.产品并已获准使用,包括利用B.t.基因遗传工程操作植物以实现抗昆虫,以及使用稳定化的微生物细胞作为B.t.蛋白质传递载体。通过修饰B.t.毒素和/或其基因获得了多种改进。例如,美国专利号5,380,831和5,567,862涉及在植物中有增进表达的合成的杀昆虫晶体蛋白质基因的产生。因而,分离的B.t.内毒素基因成为有商业价值的。
B.t.毒素在农业中的成功应用的困难包括昆虫对B.t.毒素产生的抗性。此外,某些昆虫是B.t.的作用难以控制的。后者包括诸如棉铃象
和地老虎,迄今证实它们对多数B.t.δ-内毒素无明显敏感性。
地老虎发育进度中有多个作为幼虫的龄期。虽然较后龄期幼虫咬噬幼苗产生最明显的损害和经济损失,但食叶通常造成农作物诸如玉米的减产。在接近晚期龄期时(例如第三至四龄),幼虫开始咬噬植株或植株部分,特别是幼苗。由于进食方式的改变,在极少早期警告信号的情况下能够不可预见地形成造成经济损失的种群。大的地老虎每天可破坏数株幼苗,并且严重的害虫侵袭可除去整个群丛的农作物。地老虎的夜间习惯和向地下挖洞的行为也使观测和治理很成问题。
小地老虎(The black cutworm)(Agrotis ipsiion(Hufnagel);鳞翅目(Lepidoptera):夜蛾科(Noctuidae))是多种农作物包括玉米、棉花、十字花科芸苔属蔬菜(白菜(Bcassica),青花椰菜,甘蓝,大白菜)以及草坪的严重害虫。次级宿主植物包括甜菜根,辣椒,鹰嘴豆,faba beans,莴苣,紫花苜蓿(lucerne),洋葱,马铃薯,萝卜,油菜,水稻,大豆,草莓,甜菜,烟草,番茄,以及森林树木。在北美洲,地老虎属(Agrotis)的害虫以苜蓿,玉米(corn),烟草,大麻,洋葱,草莓,黑莓,覆盆子,紫花苜蓿(alfalfa),大麦,豆类,甘蓝,燕麦,豌豆,马铃薯,红薯(sweetpotatoes),番茄,花园花卉,草,紫花苜蓿(lucern),玉米(maize),芦笋,葡萄,几乎所有类型的叶子、野草和许多其它农作物和花园植物为食。在地老虎种群(the Tribe Agrotini)中的其它地老虎是害虫,特别是那些Feltia属(例如F.jaculifera(Guenée;相当于ducens subgothica)和Euxoa属(诸如E.messoria(Harris),E.scandens(Riley),E.auxiliarisSmith,E.detersa(Walker),E.tessellata(Harris)),E.ochrogaster(Guenée))。诸如Peridroma saucia的地老虎也可以是重要害虫。柑橘类的植物也可能是地老虎攻击的目标(“柑橘地老虎”Xylomyges curialis是一个例子)。
地老虎也是北美洲以外地区的害虫,较具经济重要性的害虫攻击鹰嘴豆,小麦,蔬菜,甜菜(sugarbeet),紫花苜蓿,玉米,马铃薯,芜箐,油菜,莴苣,草莓,罗甘莓植物,亚麻,棉花,大豆,甜菜根(beetroot),大白菜,番茄,茄子,甘蔗,牧草,甘蓝,落花生,葫芦(Cucurbita),芜菁,向日葵,白菜(Brassica),洋葱,韭菜,芹菜,芝麻,芦笋,大黄,菊苣,温室农作物,以及菠菜。小地老虎(A.IPsilon)是北美洲以外,包括中美洲,欧洲,亚洲,澳大利亚,非洲,印度,台湾,墨西哥,埃及和新西兰出现的害虫。
阿根廷的主要地老虎品种是灰缘地夜蛾(Agrotis malefida),Porosagrotis gypaetiana,和小地老虎(Agrotis ipsilon(亦拼写为ypsilon))。这些地老虎攻击例如玉米,大豆和向日葵等培育植物,这些昆虫可严重减少幼苗种群,而且在有些情况下,可完全破坏整块土地。
对小地老虎(A.ipsilon)的培养控制诸如周边野草控制可以帮助防止严重的侵袭;但是,这种方法不是总能适用或有效。侵袭是散发性的,并且在种植前或种植时施用杀虫剂在过去是无效的。存在控制农作物中地老虎的一些诱饵。为了保护草皮草诸如creepingbentgrass,使用了化学杀虫剂。因为公众与这些区域的紧密接触,在草皮-覆盖区(例如高尔夫球草地,运动场,公园和其它娱乐区域,专业景观美化以及家庭草坪)使用化学杀虫剂受到特别的关注。天然产物(例如线虫和印苦楝子素(azadirachtin))通常效果不佳。
今天,由于迄今为止缺乏充分的效果,苏云金芽孢杆菌产品控制小地老虎没有得到广泛应用。地老虎的迅速取食和挖洞的行为使它们特别难以使用目前生物基础的害虫溶液来控制。例如Cry1A(b)毒素对地老虎是基本无效的。
本发明的简要总结
本发明涉及令人惊讶的发现,Cry1F蛋白质具有抗地老虎例如小地老虎(Agrotis ipsilon)的活性。这样,本发明提供了控制这些害虫的方法,其中上述方法包括将上述害虫与杀虫剂量的至少含有Cry1F毒素的杀虫部分的苏云金杆菌毒素接触。因此,本发明包括使用全长、截短和嵌合的Cry1F蛋白质和基因。在优选实施例中,Cry1F毒素是Cry1Fa毒素。可以按照本发明使用野生型和合成的Cry1F蛋白质。这样,本发明包括使用与已知Cry1F基因,优选的是核心毒素(core-toxin)编码部分杂交的多核苷酸(和/或它们的互补物,优选的是其全长互补物)。在优选的实施方案中使用了植物-最优化的多核苷酸。
本发明包括使用转基因宿主,包括植物宿主诸如玉米、棉花和向日葵。在一个优选实施方案中,本发明涉及用本发明的至少一种多核苷酸转化的植物细胞,从而使转化的植物细胞在目标昆虫消耗的组织中表达杀虫蛋白质。这种植物的转化可以通过使用本领域熟练技术人员熟知的技术来完成,并且典型的可包括修饰该基因来优化毒素在植物中的表达。

本发明的详细公开
本发明涉及令人惊讶的发现,Cry1F蛋白质具有抗地老虎例如小地老虎的活性。更令人惊讶的是发现本发明的毒素和基因可以控制第三龄或以上的幼虫。控制第一龄和第二龄的地老虎幼虫远非控制更晚龄期那么重要。尽管已知CryⅠ毒素一般是有抗鳞翅目活性的,但通常认为地老虎对B.t.毒素有抗性,如前面背景部分所祥述。
本发明包括使用重组宿主和提供植物最优化的(plant-optimized)多核苷酸序列。这些多核苷酸序列包括命名为1F1AB-PO、1F-T-PO、1F-7G-PO和1F-7Z-PO的植物最优化的基因。优选的植物宿主包括谷物(corn)、大豆、棉属、小麦、canola和向日葵。
本发明的一些实施方案中,基因编码与全长Cry1F毒素相比截短的Cry1F毒素。本发明截短的毒素典型地缺失全部或部分的前毒素节段。并且,本发明截短的基因可用于产生嵌合的基因和蛋白质。一个实施例是含有一个cryⅠF部分和一个cryⅠA(b)部分的植物最优化的基因,其中杂合基因编码一种嵌合毒素。优选的嵌合基因和毒素公布于美国专利号5,527,883和5,840,554中。根据本发明可以使用的其它嵌合基因和毒素公布于美国专利号5,508,264和5,827,514中。在一个优选实施方案中,嵌合基因的cryⅠF部分本身就是杀害虫的。也可依据本发明使用融合毒素。
在一个优选实施方案中,本发明涉及用至少一种多核苷酸序列转化的植物细胞,从而使转化的植物细胞在靶昆虫消耗的组织中表达杀虫毒素,进而使害虫接触杀虫蛋白质。这种植物的转化可以通过本领域熟练技术人员熟知的技术来完成,并且典型地可包括修饰该基因来优化毒素在植物中的表达。

基因和毒素可以使用本发明的多核苷酸形成完整“基因”在所需宿主细胞中编码蛋白质或多肽。例如,本技术领域熟练人员很容易认识到,所附的序列列表中有些多核苷酸没有显示终止密码子。并且,如本领域可容易的认识到,可以将本发明的多核苷酸置于目的宿主中的启动子控制下。
如熟练人员所容易的认识到,DNA典型的以双链形式存在。在这种排列中,一条链与另一条链互补,反之亦然。由于DNA在植物中复制(例如)产生了DNA的其它的互补链。这样,本发明包括对附录的序列表中例示的多核苷酸及其互补链的使用。本领域常使用“编码链”指与反义链结合的链。为了在体内表达蛋白质,典型将DNA的一条链转录为一条mRNA的互补链,它作为模板翻译出蛋白质。mRNA实际上是从DNA的“反义”链转录的。“有义”或“编码”链有一系列密码子(密码子是三个核苷酸,一次读三个可以产生特定氨基酸),其可作为开放阅读框(ORF)阅读来形成目的蛋白质或肽。本发明还包括与例示的DNA有相当功能的RNA和PNA(肽核酸)。

此处使用的杂交的“严紧”条件是指达到与当前申请使用条件相同或近似相同程度的杂交专一性的条件。特别地,使用标准方法进行Southern印迹中固定化DNA与32P-标记的基因-专一探针的杂交(Maniatis,T.,E.F.Fritsch,J.Sambrook[1982]MolecularCloning:A Laboratory Manual,Cold Spring Harbor Laboratory,Cold Spring Harbor,NY)。通常,在能够检测靶序列的严紧条件下进行杂交和随后的洗涤。对于双链DNA基因探针,在DNA杂合体熔解温度(Tm)下20-25℃在6×SSPE,5×Denhardt溶液,0.1%SDS,0.1mg/ml变性DNA中过夜进行杂交。熔解温度由下列公式确定(Beltz,G.A.,K.A.Jacobs,T.H.Eickbush,P.T.Cherbas,和F.C.Kafatos[1983]Methods of Enzymology,R.Wu,L.Grossman和K.Moldave[eds.]Academic Press,New York 100:266-285)。
Tm=81.5℃+16.6 Log[Na+]+0.41(%G+C)-0.61(%甲酰胺)-600/双链碱基对长度。
典型的如下进行洗涤:(1)在室温下1×SSPE,0.1%SDS中洗涤15分钟共两次(低严紧洗涤)。(2)在Tm-20℃下0.2×SSPE,0.1%SDS中洗涤15分钟一次(中等严紧洗涤)。
对于寡核苷酸探针,在杂合体熔解温度(Tm)下10-20℃在6×SSPE,5×Denhardt溶液,0.1%SDS,0.1mg/ml变性DNA中过夜进行杂交。寡核苷酸探针的Tm由下列公式测定:

基因和毒素的修饰本发明中使用的基因和毒素不但包括特定的例示序列,还包括保存了此处特别例示的蛋白质的杀虫活性特征的部分和/或片段(包括与全长蛋白质相比在内和/或末端缺失)、变体、突变体、取代物(有替代氨基酸的蛋白质)、嵌合体和融合蛋白。此处使用的术语基因的“变体”或“变异”是指编码同一毒素或编码有杀虫活性的等价毒素的核苷酸序列。此处使用的术语“等价毒素”是指与权利要求的毒素具有相同或基本相同的抗靶害虫的生物活性的毒素。
使用标准技术可以修饰基因和容易的构建基因变异体。例如,本领域熟知制造点突变的技术。又例如美国专利号5,605,793描述了在随机断裂后使用DNA重装配产生其它分子多样性的方法。可以使用商品化核酸内切酶制造全长基因的片段,并且可以按照标准程序使用核酸外切酶。例如,可以使用酶诸如Bal31或定点诱变从这些基因的末端系统地切除核苷酸。还可以使用多种限制性酶获取编码活性片段的基因。可以使用蛋白酶直接获得这些毒素的活性片段。
使用此处提供的教导可以从B.t.分离物和/或DNA文库衍生出等价毒素和/或编码这些等价毒素的基因。有多种方法获取本发明的杀虫毒素。例如,可以使用此处公开和要求保护的杀虫毒素的抗体从蛋白质混合物鉴定和分离其它毒素。特别地,抗体可能是由毒素最恒定和与其它B.t.毒素最不同的毒素部分引起的。然后可以通过免疫沉淀、酶联免疫吸附测定(ELISA)或western印迹方法使用这些抗体专一的鉴定有特征活性的等价毒素。可使用本领域标准程序容易的制备此处公开的毒素或等价毒素或这类毒素的片段的抗体。然后可以从微生物中获得编码这些毒素的基因。
由于遗传密码子的丰余性,多种不同的DNA序列可以编码相同的氨基酸序列。产生这些编码相同或基本相同的毒素的可替代DNA序列正在本领域训练的人员技术水平内。这些不同的DNA序列包括在本发明的范围内。此处使用的“基本上相同的”序列是指有氨基酸取代、缺失、添加或插入但实质上不影响杀虫活性的序列。本定义中亦包括保留杀虫活性的片段。
等价毒素与例示的毒素有氨基酸相似性(和/或同源性)。氨基酸类似性/相同性典型的大于60%,优选的大于75%,更优选的大于80%,甚至更优选的大于90%,并且可以大于95%。也可以根据更特定的相同性和/或类似性范围定义本发明的优选的多核苷酸和蛋白质。例如与此处例示的序列有百分之49、50、51、52、53、54、55、56、57、58、59、60、61、62、63、64、65、66、67、68、69、70、71、72、73、74、75、76、77、78、79、80、81、82、83、84、85、86、87、88、89、90、91、92、93、94、95、96、97、98或99的相同性和/或类似性。除非特别说明,如此处使用的两个核酸之间的序列相同性和/或类似性百分数是使用Karlin和Altschul的算法确定的(1990),Proc.Natl.Acad.Sci.USA 87:2264-2268,改进的方法见Karlin和Altschul(1993),Proc.Natl.Acad.Sci.USA 90:5873-5877。这种算法并入到Altschul等(1990),J.Mol.Biol.215:402-410的NBLAST和XBLAST程序中。使用分数(score)=100、字长度(wordlength)=12的参数通过NBLAST程序进行BLAST核苷酸检索,以获得有所需序列相同百分数的核苷酸序列。为了比较的目的获得有缺口的比对,使用了Altschul等(1997)Nucl.Acids Res.25:3389-3402中所述的Gapped BLAST程序。使用BLAST和Gapped BLAST程序时,使用了各程序(NBLAST和XBLAST)的缺省参数。参看网址http://www.ncbi.nih.gov。也可以使用上面背景部分中所述的Crickmore等的方法和算法计算相同性分数。
在决定生物活性的或参与确定最终决定生物活性的三维构型的毒素的关键区域,氨基酸同源性最高。在这点上,如果取代发生在对活性非关键的区域或是不会影响分子的三维构型的保守氨基酸取代,则这些氨基酸取代是可接受的和能够期望的。例如,氨基酸可以分为以下类别:非极性的,不带电极性的,碱性的和酸性的。只要取代本质上不影响化合物的生物活性,一个类别的某氨基酸被同一类别的另一个氨基酸替代的保守取代包括在本发明的范围内。表1提供了属于各种类的氨基酸的实例的列表。

在有些情况下,可以进行非-保守取代。关键因素是这种取代必须不会显著降低植物表达目的DNA序列的能力或显著降低毒素的生物活性。
此处使用的“分离的”多核苷酸和/或“纯化的”毒素是指这些分子不与它们天然相关的其它分子结合存在;这也包括它们在植物中的应用。这样,如此处所述“分离的”和/或“纯化的”意味着“人工处理”的参与。
虽然本发明提供了合成基因的特定实施方案,但是也可以使用与此处例示的基因功能上等价的其它基因来转化宿主,优选的是植物宿主。产生合成基因的其它指导可以在例如美国专利号5,380,831中找到。
重组宿主可以将本发明的编码毒素的基因导入到多种微生物或植物宿主。在优选实施方案中,毒素基因的表达直接或间接地导致在细胞内产生和保持杀虫剂。当害虫摄取转基因/重组/转化的宿主细胞时,害虫会摄取毒素。这是使害虫与毒素接触的优选方式。结果是控制了害虫(杀死或使其患病)。
本发明的有些实施方案中,可以在前期步骤中使用转化的微生物宿主来制备前体,例如,在优选实施方案中它最终会用于转化植物细胞和植物,使它们表达本发明的基因编码的毒素。这种方式转化和使用的微生物在本发明范围内。重组微生物可以是例如B.t.、E.coli或假单胞菌属(Pseudomonas)。
可由本技术领域熟练人员通过标准技术制造不同的重组生物体。这些转化需要的材料已在此处公开或熟练人员能够容易的得到。有多种方法将编码毒素的B.t.基因导入靶宿主,使该基因稳定保持和表达。这些方法是本领域熟练人员熟知的并举例描述于美国专利号5,135,867中,并于此处。
可以通过合适的载体将本发明的编码毒素的基因导入多种微生物和植物宿主。有多种目的农作物,诸如玉米、小麦、稻、棉花、大豆和向日葵。本发明的特定基因特别适于在转化的植物中稳定保持和表达能够表达多肽杀虫剂的基因,并能期望提供改进的保护以防止杀虫剂被环境降解和灭活。毒素基因的表达直接或间接导致在细胞内产生和保持杀虫蛋白质。这样,靶害虫可以通过摄取含有杀虫蛋白质的植物组织与对害虫有毒的杀虫蛋白质接触。结果是控制了害虫。另一种方法是,可以在害虫区域施用合适的微生物宿主例如荧光假单胞菌,它们的一些在那里能够增殖,并被靶害虫摄取。可以在延长毒素的活性和稳定细胞的条件下处理含有毒素基因的微生物。然后可将处理的保留毒素活性的细胞施用到有靶害虫的环境。
当通过适当的载体将B.t.毒素基因导入微生物宿主,并且该宿主能够以活体状态施用到环境,应当使用某些宿主微生物。选择已知占据一种或几种目的农作物的“植物圈”(叶面,叶际,根际,根表)的微生物宿主。选择这些微生物,以使它们有能力在特定环境中(农作物和其它昆虫栖息地)成功地与野生型微生物竞争,提供稳定保持和表达用以表达多肽杀虫剂的基因,并且理想地提供改进的保护以防止杀虫剂被环境降解和灭活。
已知多种微生物栖息于多种重要农作物的叶表(植物叶的表面)和/或根际(植物根的周围土壤)。这些微生物包括细菌、藻类和真菌。特别感兴趣的是微生物,诸如细菌,例如假单胞菌属、欧文氏菌属、沙雷氏菌属、克雷伯氏菌属、黄单胞菌属、链霉菌属、根瘤菌属、红假单胞菌属、嗜甲基菌属、土壤杆菌属、醋杆菌属、乳酸杆菌属、节杆菌属、固氮菌属、明串珠菌属和产碱菌属;真菌特别是酵母,例如酵母属、隐球酵母属、克鲁维氏酵母属、掷孢酵母属、红酵母属(Rhodotorula)和Aureobasidium。特别感兴趣的是植物圈细菌菌种诸如丁香假单胞菌、荧光假单胞菌、粘质沙雷氏菌、木醋杆菌、根癌土壤杆菌、球形红细菌、野油菜黄单胞菌、苜蓿中华根瘤菌、真养产碱菌、维涅兰德固氮菌;以及植物圈酵母菌种诸如深红酵母、粘红酵母、海滨红酵母、橙黄红酵母、浅白色隐球酵母、流散酵母、罗伦隐球酵母、罗茜酵母、善地酵母、酿酒酵母、掷孢酵母、香气掷孢酵母、佛地克鲁维氏酵母和出芽短柄霉。特别感兴趣的是产色素微生物。
有多种方法将编码毒素的B.t.基因导入目的宿主,其是在使该基因稳定保持和表达的条件下。这些方法为本领域熟练人员熟知,并例如描述于美国专利号5,135,867中,其以参考文献形式并于本文。
处理细胞如上所述,可以处理表达B.t.毒素的B.t.或重组细胞以延长毒素活性和稳定细胞。形成的杀虫剂微囊将B.t.毒素包含在稳定化的细胞结构中并在将微囊施用到靶害虫的环境中时保护毒素。合适的宿主细胞可以包括原核或真核细胞,通常限于那些不产生对高等生物诸如哺乳动物有毒物质的细胞。但是,也可以使用产生对高等生物有毒物质的生物体,其中的有毒物质是不稳定的或使用时的水平足够低,避免了对哺乳动物宿主的可能毒性。特别感兴趣的宿主是原核或低等真核生物,诸如真菌。
在处理时,虽然有时可以使用芽孢,但是细胞通常是完整的并且实际上处于增殖形式,而不是芽孢形式。
只要该技术不有害地影响毒素的性质或减少细胞对毒素的保护能,就可以使用化学或物理手段或将化学和/或物理手段组合来处理微生物细胞例如含有B.t.毒素基因的微生物。化学试剂的实例是卤化试剂,特别是原子序号17-80的卤素。更特别的,可以在温和的条件下使用碘并持续足够时间以获得期望的结果。其它的技术包括用醛处理,诸如戊二醛;抗-感染剂,诸如氯化苄基二甲基烷基铵和十六烷基氯化吡啶鎓;醇,诸如异丙基和乙醇;多种组织固定剂,诸如路戈尔碘液(Lugol iodine),布英固定剂(Bouin’s fixative),多种酸和海利固定剂(Helly’s fixative)(见:Humason,Gretchen L.,AnimalTissue Techmiqnes,W.H.Freeman and Company,1967);或将物理(加热)方法和化学试剂组合,在将细胞施用于宿主环境时,保持和延长细胞内产生的毒素的活性。物理方法的例子是短波辐射诸如γ-射线和X-射线、冷冻、紫外光照射、冷冻干燥等等。处理微生物细胞的方法公开于美国专利号4,695,455和4,695,462中,以参考文献形式并于此处。
细胞通常会有增强的结构稳定性,从而加强对环境条件的抗性。当杀虫剂是前体形式时,应当选择细胞处理的方法使其不会抑制靶害虫病原体将前体处理成杀虫剂成熟形式的过程。例如,甲醛会交联蛋白质从而抑制多肽杀虫剂的前体形式的处理。处理方法应当至少保留毒素的实质部分的生物利用度或生物活性。
为了生产目的的选择宿主细胞时特别感兴趣的特征包括,将B.t.基因导入宿主的容易度,存在表达系统,表达效率,杀虫剂在宿主中的稳定性,以及存在辅助性遗传能力。为了用作杀虫微囊的感兴趣特征包括杀虫剂的保护质量,诸如厚的细胞壁,色素形成,和细胞内包装或形成包涵体;在液体环境中存活;缺少哺乳动物毒性;吸引害虫摄食;易于杀死和固定而不破坏毒素;诸如此类。其它的考虑包括易于形成制剂和处理,经济,储藏稳定性,诸如此类。
细胞的生长可以在任何方便的营养培养基培养含有B.t.杀虫剂基因的细胞宿主,优选地DNA构建体有选择性优势,提供的选择性培养基使得基本所有或所有细胞保留B.t.基因。然后可以施用传统方法收获这些细胞。或者,可以在收获前处理细胞。
可以使用标准培养基和发酵技术培养本发明的B.t.细胞。完成发酵循环后可首先使用本领域熟知方法从发酵肉汤中分离B.t.芽孢和晶体来收获细菌。通过加入表面活性剂、分散剂、惰性载体和其它组分将回收的B.t.芽孢和晶体制成可湿粉剂、液体浓缩剂、颗粒或其它制剂,以促进处理和应用于特定的靶害虫。本技术领域熟知这些制剂和应用的程序。
制剂含有引诱剂和B.t分离物的芽孢和晶体或含有从此处公开的B.t分离物可获得的基因的重组微生物的制剂化的诱饵颗粒可以施用于土壤。制剂化产品也可以用作种子涂层或根处理或在农作物循环的晚期阶段进行全植物处理。通过与多种惰性材料诸如无机矿物质(页硅酸盐,碳酸盐,硫酸盐,磷酸盐,等等)或植物材料(粉状玉米芯,稻壳,胡桃壳,等等)混合,能够以可湿性粉剂、颗粒或粉尘形式对植物和土壤施用B.t细胞。该制剂可能包括撒布器-粘着剂辅剂、稳定剂、其它杀虫剂添加剂或表面活性剂。液体制剂可以是基于水的或非水的,并以泡沫、凝胶、悬浮液、可乳化浓缩物或类似形式施用。成分可以包括流变剂、表面活性剂、乳化剂、分散剂或多聚体。
按照特定制剂的性质,特别是它是浓缩剂或是直接使用,本技术领域熟练人员将会意识到杀虫剂的浓度可以变化很大。杀虫剂将会至少占重量的1%并可达到100%。干燥制剂将会含有大约1-95%重量的杀虫剂,而液体制剂中液相中通常将会含有大约1-60%重量的固体物。制剂中通常将会含有大约102至104细胞/毫克。将会在每公顷使用大约50毫克(液体或干剂)至1公斤这类制剂。
通过喷洒、撒粉、喷淋或类似方法将制剂施用到害虫环境例如土壤和叶片。
突变体可以使用本技术领域熟知方法制造本发明的分离物的突变体。例如通过甲磺酸乙酯(EMS)诱变分离物可以获得不产孢子的突变体。可以使用本领域熟知方法通过紫外线和亚硝基胍制造突变体。
较少百分数的不产孢子突变体在延长的发酵时期内将会保持完整并且不裂解;这些菌株定义为裂解负的(-)。通过在摇瓶培养物中筛选不产孢子突变体并且选择在发酵末期仍然完整和含有毒素晶体的突变体鉴定了裂解负菌株。裂解负菌株适于能够产生保护的囊化毒素蛋白质的细胞处理程序。
为了制备上述不产孢子突变体的抗噬菌体变种,将等份的噬菌体裂解物涂布到营养琼脂并干燥。然后将一份噬菌体敏感菌株直接铺平板到干燥的裂解物上面并干燥。将平板在30℃培养。将平板培养2天,在这时,会看到许多菌落在琼脂上生长。挑选部分菌落并亚克隆到营养琼脂平板。通过与噬菌体裂解物交叉划线培养检测这些有表观抗性培养物的抗性。在平板上划一条噬菌体裂解物线并干燥。然后将假定的抗性培养物与噬菌体线交叉划线。在30℃培养过夜后,抗性细菌在与噬菌体线交叉划线的任何地方都不出现裂解。然后通过将抗性培养物在营养琼脂平板铺成菌苔再次证实对噬菌体的抗性。也以同样方式把敏感菌株涂布平板作为阳性对照。干燥后,将一滴噬菌体裂解物滴在平板中央并干燥。在30℃培养24小时后抗性培养物在放置噬菌体裂解物的区域不出现裂解。
只要不与本说明的明确教导矛盾,将把参考和引用的所有专利、专利申请、临时专利和公开出版物以参考文献形式完整地并入本发明。
下面是说明实施本发明的流程的实施例。这些实施例不应解释为限制。实施例1-Cry1F抗小地老虎(Agrotis ipsilon;鳞翅目:夜蛾科)幼虫的生物检测
使用MR872(美国专利号5,840,554中公开的表达cry1Fa/cry1Ab嵌合基因的荧光假单胞菌克隆)冻干粉剂制备物利用标准生物检测方法测定Cry1F抗小地老虎(BCW)的生物学活性。通过将检测样品掺入人工饲料并用处理和未处理的饲料喂食幼虫进行BCW的生物检测。所有检测均使用BCW人工饲料(BioServ Corporation,Frenchtown,NJ)进行。通过将样品以6毫升悬浮液加54毫升饲料的比率与人工饲料(首先降温至55℃)混合进行饲料掺入。通过系列稀释产生多种处理浓度。涡流混合后,将混合物注入3毫升分区的塑料盘(Nutrend ContainerCorporation,Jacksonville,FL),比例大约是每个24孔盘30毫升饲料(每孔填充一半)。有较大孔的Nutrend盘用于三龄或更老昆虫的生物检测;它们亦使用人工饲料填充大约一半。不含实验物质的水空白作为对照。随后至少30分钟使饲料冷却和固定,将幼虫(French AgResources,Lamberton,MN)放置在饲料混合物上面,每孔一个幼虫。使用定位焊接烙铁(tacking iron)用Mylar片(Mylar sheeting)密封孔,并在每孔扎几个针孔进行空气交换。在25℃存储室(holdingroom)以14∶10(光∶暗)持续6天。在大约6天后记录死亡率和发育迟缓。对于LC50生物检测,至少用3个平行,各自有5-7种剂量并且24只昆虫/每剂量。结果显示于表2(测定MR872 Cry1F冻干粉剂制备物抗第一和第三龄小地老虎幼虫的生物检测)。

使用诱饵制剂测定了Cry1F抗小地老虎的活性。将MR872克隆与商品化诱饵(SoilServ,Salinas,CA)混合并将产生的混合物覆盖到盆栽麦苗(Freekes growth stage 1.5)的土壤表面,比率是50磅诱饵/英亩。通过向诱饵中添加不同量的毒素测定了两种浓度的Cry1F,产生的比率相当于6.25克和12.5克Cry1F毒素/英亩。然后以大约每5株植物一只昆虫的比例用3或4龄幼虫感染植物,在24-48小时后测量植物损伤。损伤的程度级别(rating scale)是0-4,0是指植株完全咬切至地面,4是指没有可见损伤。对照包括未添加诱饵的植株(“无诱饵”),添加不含毒素的诱饵(“空白诱饵”)以及无诱饵或昆虫的植株(“对照”)。结果显示于图1和2。实施例3-通过昆虫生长和死亡率以及植物损伤测定的转基因Cry1F向日葵植株抗小地老虎的效率
植株处理:
·V2期对照幼苗

实验:B.t.幼苗:5个重复(repetition),每个重复5株幼苗,每株幼苗一只幼虫。对照:5个重复,每个重复5株幼苗,每株幼苗一只幼虫。
流程:
·每一重复含有播种到透明矩形盘(15厘米×25厘米)的五颗种子(seeds),并加盖以限制昆虫幼虫。
·每一重复使用5只幼虫感染。将盘置于25℃和75%湿度的室中。感染后24,48,72和96小时评估感染。
·评估
1.植株-分类为未损伤的,损伤的(咬了茎,子叶或叶的)或在子叶下切割的。对各重复亦进行分级:1(未损伤或茎上有一些咬痕的)至9(整个植株被切掉并且80%以上的组织被食)。
2.昆虫——统计感染后96小时的死幼虫并将存活的幼虫称重。
3.通过可能性表分析(contingency table analysis)来分析数据。
结果:有图显示4个重复(感染48和96小时后)。在下列表中报告数据,显示用小地老虎感染后48,72和96小时各重复的评估。

概要将十株Cry1F玉米(实质上表达序列3的多核苷酸)在田间条件下控制小地老虎(Agrotis ypsilon(Hufnagel))的能力与两株非-Bt杂合体(对照1和对照2)进行了比较,为了比较的目的,一个杂合体表达Cry1Ab,另一个杂合体表达Cry9C蛋白质。与非-Bt杂合体相比,所有的Cry1F事件提供抗植丛损失(stand loss)的良好保护。
材料和方法在第一天使用Almaco锥形播种器将每一记录项(entries)播种25粒种子。播种了四组重复--分别随机分开。行是成对的,每对行之间有未种植的人行道。各小块土地包围在8英寸高,2.5英尺宽和6英尺长的电镀钢栅栏内。在第13和14天将栅栏敲入地下大约2英寸深。用凡士林处理栅栏边缘阻碍幼虫逃跑。在栅栏内撒麦杆为地老虎提供隐蔽处。幼苗长出后不久,用ELISA条带测定含有Cry1F的记录项植株,除去不表达植株或过表达植株将植丛间苗至10株。有3小块土地,最终植丛是8或9株植物。还将非-Cry1F的植株间苗到10株。如果除去了植株,在各植株后面放置一块塑料桩使我们知道除去了一株植株。在第15天,当植株到了V1或V2早期,在每株植物放入3只三龄小地老虎幼虫。在第19天每株植物放入2只四龄BCW进行第二次感染。
从第16天至26天每天检查各小块土地。在第29天再进行分级,在第34天进行最后分级。每一损伤的植株前面均用小塑料桩标记,并在数据册中记录损伤类型。实验结束时进行最后的植丛计数。
结果在本实验中获得对小地老虎的中等水平压力,在有些非-Bt记录项中导致植丛减少。由于未知的原因,本实验中BCW有导致枯心(dead-heart)植株而不是咬植株的倾向。由于这种原因,还比较了损伤的植株的资料。
表8显示植物类型和每种类型植株的植丛减少(Stand Reduction)百分数(%SR)和损伤植株百分数(%Dam)。植丛减少意思是植株被杀死。损伤的植株有明显的摄食损伤但是未损失它们的生长点。

明显地,需要能够在植物种表达该基因地启动子区。因而,为了植物内表达,本发明的DNA在合适的启动子区域控制下并和它可操作地连接。本领域熟知使用这种构建体获得植物内表达的技术。
可以使用多种本技术领域熟知的技术将此处公开的编码杀虫剂毒素的基因插入到植物细胞。例如,存在多种克隆载体,包含在E.coli中的复制系统和筛选转化细胞的标记,可用于将外源基因插入到高等植物。载体包括,例如pBR322,pUC系列,M13mp系列,pACYC184等等。相应地,可将编码B.t.毒素的序列在适当的限制性位点插入载体。使用产生的质粒转化入E.coli。在适当的营养培养基中培养E.coli细胞,然后收获并裂解。回收质粒。通常作为检测方法进行序列测定,限制性分析,电泳和其它生物化学-分子生物学方法。每一次操作,可以切割使用的DNA序列并加入下一DNA序列。每一个质粒序列均可克隆到相同或其它质粒。
根据将目的基因插入植物的方法,可能需要其它DNA序列。如果,例如使用Ti或Ri质粒转化植物细胞,那么至少右侧边界,但常常是Ti或Ri质粒T-DNA的右侧和左侧边界,必须作为要插入基因的侧翼区域参加。使用T-DNA转化植物细胞已经得到集中研究并充分的描述于EP 120516;Hoekema(1985)在:The Binary Plant Vector System,Offset-durkkerij Kanters B.V.,Albasserdam,Chapter 5;Fraley等,Crit.Rev.Plant.Sci.4:1-46;and An等(1985)EMBO J.4:277-287。

现有多种技术可以将DNA插入植物宿主细胞。这些技术包括使用根瘤土壤杆菌或毛根土壤杆菌作为转化试剂用T-DNA,融合,注射,biolistics(微粒轰击),或电穿孔以及其它可能的方法进行转化。如果使用土壤杆菌进行转化,必须将要插入的DNA克隆到特定的质粒,即克隆到中间载体或二元载体。由于序列与T-DNA中的序列同源,通过同源重组可以将中间载体整合到Ti或Ri质粒。Ti或Ri质粒还包含对转移T-DNA必须的vir区域。中间载体本身在土壤杆菌中不能复制。通过辅助质粒方法(接合)可以将中间载体转移到根瘤土壤杆菌。二元载体在E.coli和土壤杆菌中可以自己复制。它们含有一个选择性标记基因和接头或多接头,其通过右侧和左侧T-DNA边界区域构架。它们可以直接转化到土壤杆菌(Holsters等[1978]Mol.Gen.Genet.163:181-187)。用作宿主细胞的土壤杆菌包含一个携带vir区域的质粒。vir区域对于将T-DNA转移到植物细胞是必需的。可以含有其它的T-DNA。这样转化的细菌用于转化植物细胞。为了将DNA转移到植物细胞,可以有利地将植物外植体与根瘤土壤杆菌或毛根土壤杆菌一起培养。然后在可以包含用于选择的抗生素或杀生物剂的合适的培养基中,可以从感染的植物材料(例如叶片,茎的一部分,根,但是也可以是原生质体或悬浮培养的细胞)产生整个植物。然后可以检测这样获得的植物中插入DNA的存在。在注射和电穿孔时对质粒没有特别要求。可以使用常规质粒,诸如pUC衍生物。
转化的细胞以常规方式在植物内生长。它们可以形成生殖细胞并将转化的特性转移到子代植株。这些植物能够以正常方法生长并且与具有相同转化的遗传因子或其它遗传因子的植株杂交。产生的杂交个体具有相应的表型特征。
