现有技术
蛋白酶用于洗涤、清洁和漂洗组合物,并且借助生物催化降解而有助于污垢的溶解,从而有助于清洁性能。在大多数的阴离子型表面活性剂体系中的蛋白酶的稳定性对配剂师而言仍然是挑战,特别是如果要制备具有长时间内储存稳定性且保留其酶活性的液体配制物。原则上,表面活性剂有助于蛋白质变性,从而导致酶结构的变性,因而导致酶活性的失活。
酶生产者使用蛋白质工程方法来开发相对于表面活性剂具有改进稳定性的酶。然而,这很复杂,并不是对于所有的酶是成功的,并且可能由于蛋白质结构的改变而导致比活性的降低。
或者,可以通过使用较温和的表面活性剂而使酶稳定。例如,将月桂基醚硫酸钠和/或脂肪醇乙氧基化物加入到用作主要表面活性剂的线性烷基苯磺酸盐中,参见Kravetz等,1985,Lund等,2012。
根据US5156773,甜菜碱也可用于使与阴离子表面活性剂相关的蛋白酶得以稳定。
DE102007005419公开了在宽范围内的含氮、非离子型氧化胺上作为酶稳定化剂。
EP0499434、EP2787065和EP2410039公开了单独或与其它阴离子表面活性剂组合的鼠李糖脂和槐糖脂,以及其对衣物的良好清洁效果。
WO2012010406的实施例中描述了与脂肪酶组合的鼠李糖脂的良好清洁效果。此时,不使用还含有其它阴离子表面活性剂的配制物。
在具有还含有蛋白酶活性的酶的液体表面活性剂体系的配制物中,还必须确保配制物中的蛋白酶活性通过合适的添加剂而得以抑制,这是因为否则的话可能在配制物中能够发生蛋白酶自发消化或者甚至其它酶的消化。为此,使用多元醇(例如1,2-丙二醇)、硼酸盐和其它抑制剂。尤其是,近年来,由于毒理学问题,硼酸盐已经声明狼藉,仍然需要廉价的、毒理学上可接受的替代物。
发明详述
已经令人惊奇地发现,通过添加生物表面活性剂,特别是鼠李糖脂和槐糖脂,可以提高尤其是阴离子表面活性剂体系中肽酶的稳定性。此外,可以减少额外需要的蛋白酶抑制剂的量,或者甚至可以完全排除蛋白酶抑制剂。
因此,本发明涉及包含如下的组合物:
A)至少一种肽酶,
B)至少一种生物表面活性剂,以及任选存在的
C)至少一种阴离子表面活性剂。
本发明组合物的一个优点是,基于组合物中存在的表面活性剂的总量,基于可再生原料中存在的的表面活性剂的比例优选大于50重量%。
本发明组合物的另一个优点是可以将糖或糖和甘油酯和/或脂肪酸用作生物表面活性剂的原料。
本发明组合物的另一个优点是它非常温和。
本发明的另一个优点在于,在本发明的组合物中,可以减少所需的蛋白酶抑制剂的量,或者甚至可以完全排除蛋白酶抑制剂。
与现有技术相比的其他优点是,在洗涤过程中酶稳定性的增加,以及与之相关的改进的清洁性能,例如在洗衣中。
本发明的另一个优点是,在本发明的组合物中,其中洗涤剂配制物中的稳定性不足时,也可以使用蛋白酶。
本发明的另一个优点是不必使用或使用更少量的配合剂(助洗剂)以确保在硬水中具有充分的洗涤性能。

在本发明中,术语“阴离子表面活性剂”应理解为其中在pH为7和20℃下至少90mol%的分子具有至少一个带负电荷基团的表面活性剂。优选,在作为背景电解质的10毫摩尔的氯化钾水溶液中进行测量时,它们在25℃下不具有pHIEP=2-12的等电点。
在本发明的上下文中,术语“使酶活性稳定”特别理解为当在30℃下储存一段长时间以致发生活性丧失时,在稳定剂的存在下(此时为生物表面活性剂),与没有稳定剂且在其他条件相同下而对比,所测试的酶损失了更少的活性。
本发明的组合物包含至少一种作为组分A)的肽酶。肽酶为是酶类(“EC”)3.4的酶。
所存在的肽酶优选选自蛋白酶。所有现有技术已知的蛋白酶均适用作蛋白酶,包括化学或基因改性的蛋白酶。这些特别包括EC 3.4.21的丝氨酸蛋白酶和EC 3.4.24的金属蛋白酶。所存在的肽酶优选EC 3.4.21.1的胰蛋白酶和糜蛋白酶样蛋白酶,EC 3.4.21.2和EC 3.4.21.4,特别优选EC 3.4.21.62的枯草杆菌蛋白酶。特别优选存在的金属蛋白酶选自EC 3.4.24.27的嗜热菌蛋白酶和EC 3.4.24.28的bacillolysine。
市售蛋白酶的实例包括KannaseTM、EverlaseTM、EsperaseTM、AlcalaseTM、NeutraseTM、DurazymTM、SavinaseTM、OvozymeTM、LiquanaseTM、Co-ronaseTM、PolarzymeTM、PyraseTM、胰腺胰蛋白酶NOVO(PTN)、Bio-FeedTM、Pro和Clear-LensTM(均来自Novozymes A/S,Bagsvaerd,丹麦)。其它市售蛋白酶包括RonozymeTM、Pro、MaxataseTM、MaxacalTM、MaxapemTM、Optic-leanTM、ProperaseTM、PurafectTM、Purafect OxTM、Purafact PrimeTM、ExcellaseTM、FN2TM、FN 3TM和FN4TM(Genencor International Inc.,Gist-Brocades,BASF,DSM)。Henkel/Kemira蛋白酶也合适,例如BLAP(US 5,352,604的图29中的序列,具有以下点突变:S99D+S101 R+S103A+V104I+G159S,下文称为BLAP),BLAP R(BLAP S3T+V4I+V199M+V205I+L217D),BLAP X(具有以下点突变的BLAP:S3T+V4I+V205I)和BLAP F49(具有以下点突变的BLAP:S3T+V4I+A194P+V199M+V205I+L217D)和KAP(来自Kao的嗜碱芽孢杆菌枯草杆菌蛋白酶,具有以下突变:A230V+S256G+S259N)。

可以使用的用于生产生物表面活性剂的原料为碳水化合物,特别是糖,例如葡萄糖,和/或亲脂性碳源,例如脂肪、油、偏甘油酯、脂肪酸、脂肪醇、长链饱和或不饱和烃。优选,在本发明的组合物中,不存在不是通过糖脂的发酵而生产的生物表面活性剂,其中所述糖脂例如为脂蛋白。
本发明的组合物优选包含作为组分B)的至少一种生物表面活性剂鼠李糖脂、槐糖脂、葡萄糖脂、纤维素脂、甘露糖醇酯脂(mannosylerythritol lipids)和/或海藻糖脂,优选鼠李糖脂和/或槐糖脂。生物表面活性剂,特别是糖脂表面活性剂,可以例如如EP 0 499434、US 7,985,722、WO 03/006146、JP 60 183032、DE 19648439、DE 19600743、JP 01304034、CN 1337439、JP 2006 274233、KR 2004033376、JP 2006 083238、JP 2006070231、WO 03/002700、FR 2740779、DE 2939519、US 7,556,654、FR 2855752、EP 1445302、JP 2008062179和JP 2007 181789或其中引用的文献而生产。可以例如由Soliance,France获得合适的生物表面活性剂。
优选,本发明的组合物具有鼠李糖脂作为生物表面活性剂,特别是单-,二-或多鼠李糖脂和/或槐糖脂。特别优选,本发明的组合物具有一种或多种具有式(I)、(II)或(III)的如EP 1 445 302A中所述的鼠李糖脂和/或槐糖脂。
在本发明的上下文中,术语“鼠李糖脂”应尤其理解为通式(I)的化合物或其盐,

存在于本发明组合物中的鼠李糖脂在给定的pH下,会至少部分地作为盐而存在。
在本发明的优选组合物中,存在的鼠李糖脂盐的阳离子优选选自Li+、Na+、K+、Mg2+、Ca2+、Al3+、NH4 +、伯铵离子、仲铵离子、叔铵离子和季铵离子。

特别优选的阳离子优选选自Na+、K+、NH4 +和三乙醇铵阳离子。
本发明的优选组合物的特征在于其包含鼠李糖脂的混合物,其中混合物中二-鼠李糖脂与单-鼠李糖脂的重量比大于51:49,优选大于75:25,特别优选97:3,特别是大于98:2。
本发明的优选组合物的特征在于,鼠李糖脂混合物包含51重量%至95重量%,优选70重量%至90重量%,特别优选75重量%至85重量%的diRL-C10C10和
0.5重量%至9重量%,优选0.5重量%至3重量%,特别优选0.5重量%至2重量%monoRL-C10C10,其中所述重量百分比是参考存在的鼠李糖脂的总量。
本发明的优选组合物的特征在于,除了上述diRL-C10C10和monoRL-C10C10含量之外,所述鼠李糖脂混合物包含:
0.5重量%至15重量%,优选3重量%至12重量%,特别优选5重量%至10重量%的diRL-C10C12:1,其中所述重量百分比是参考存在的鼠李糖脂的总量。
本发明的优选组合物的特征在于,除了上述diRL-C10C10和monoRL-C10C10含量之外,所述鼠李糖脂混合物包含:

此外,还优选本发明的组合物中存在的鼠李糖脂混合物仅包含少量式monoRL-CX或diRL-CX的鼠李糖脂。具体而言,本发明的组合物混合物优选包含:
0重量%至5重量%,优选0.001重量%至3重量%,特别优选0.01重量%至1重量%的diRLC10,其中所述重量百分比是参考存在的鼠李糖脂的总量,术语“0重量%”应理解为没有可检测到的量。
制备相关鼠李糖混合物的方法例如公开于EP2786743和EP2787065中。
槐糖脂可以在本发明中以酸的形式或内酯的形式使用。术语槐糖脂的“酸形式”是指EP2501813的通式(Ia),术语“内酯形式”是指EP2501813的通式(Ib)。
为了确定配制物中酸或内酯形式的槐糖脂的含量,参见EP 1 411 111B1,第8页,第[0053]段。
本发明的优选配制物包含作为组分B)的槐糖脂,其中内酯形式与酸形式的重量比为20:80至80:20,特别优选30:70至40:60。

本发明组合物的优选特征在于,在本发明的组合物中所有表面活性剂的总和的比例为0.1重量%至50重量%,优选1重量%至25重量%,优选5重量%至20重量%,特别优选10重量%至20重量%,其中所述重量百分比基于总的组合物。
生物表面活性剂在总表面活性剂中的比例基于本发明组合物中表面活性剂的用量优选5重量%至99重量%,优选20重量%至95重量%,特别优选25重量%至80重量%。
本发明的优选组合物包含水作为组分D)。
在优选的实施方案中,本发明组合物的含水量为0.001重量%至5重量%,优选0.01重量%至3重量%,特别优选0.1重量%至2重量%。该实施方案例如包括诸如呈粉末、颗粒或片剂形式的储存稳定的干洗剂。
在另一优选的实施方案中,本发明组合物的含水量为10重量%至95重量%,优选20重量%至90重量%,优选30重量%至80重量%。该替代实施方案例如包括储存稳定的液体清洁剂。
优选本发明的组合物的pH为4至12.5,优选5至10,特别优选5.5至9.0。
若本发明组合物例如用于衣物洗涤剂中,则优选其pH为7至12.5,优选7.5至12,特别优选8至12。
若本发明组合物例如用于人工洗碗剂中,则优选其pH为4至8,优选4.5至7.5,特别优选5至6.5。
与本发明相关的“pH”定义为使用依据ISO 4319(1977)的校准pH电极在25℃下搅拌5分钟后对有关物质测量的值。
本发明的优选组合物包含至少一种作为组分E)的蛋白酶抑制剂。
所存在的优选蛋白酶抑制剂选自可逆的蛋白酶抑制剂。
常用作可逆蛋白酶抑制剂是盐酸苄甲脒、硼砂、硼酸、硼酸化物或其盐或酯,特别是包括具有芳族基团的衍生物,例如邻位、间位或对位取代的苯基硼酸,特别是4-甲酰基苯基硼酸,或其指定的化合物盐或酯。也用于此的还有肽醛类,即具有降低的C-末端的低聚肽,特别是由2至50个单体组成的那些。所述肽可逆蛋白酶抑制剂尤其包括卵类粘蛋白和亮肽酶。枯草杆菌蛋白酶的特异性可逆肽抑制剂以及蛋白酶的融合蛋白和特异性肽抑制剂也适用于此。
特别优选使用硼酸和/或其盐作为组分E)。
根据本发明,优选若硼酸和/或其盐作为组分E)而存在,则还包括多元醇。这些通过与硼酸和/或其盐以及生物表面活性剂的相互作用而使配制物中的肽酶活性得以进一步稳定。所用的优选多元醇为1,2-丙二醇、乙二醇、四氢呋喃二醇、甘油、山梨醇、甘露醇、葡萄糖、果糖和乳糖。根据本发明,硼酸和/或其盐与多元醇的重量比为1:0.1至1:10,优选1:0.3至1:5。
本发明的特别优选的组合物包含
A)至少一种肽酶,
B)至少一种生物表面活性剂,
C)至少一种阴离子表面活性剂,
D)水,和
E)至少一种蛋白酶抑制剂,
其中:
所述肽酶选自EC 3.4.24.28的杆菌溶素组,EC 3.4.24.27的嗜热菌蛋白酶组和EC3.4.21.62的枯草杆菌蛋白酶组,
所述生物表面活性剂选自鼠李糖脂和槐糖脂,
所述阴离子表面活性剂选自任选的烷氧基化的磺酸盐、烷基硫酸盐和烷基醚硫酸盐,
所述蛋白酶抑制剂选自硼酸及其盐,
并且所述组分基于总组成的存在量:
A)0.001重量%至10重量%,优选0.05重量%至5重量%,优选0.1重量%至3重量%,
B)0.5重量%至50重量%,优选2重量%至40重量%,优选5重量%至30重量%,
C)0.1重量%至40重量%,优选1重量%至35重量%,优选2重量%至30重量%,
D)0.001重量%至95重量%,优选1重量%至90重量%,优选10重量%至60重量%
E)0.001重量%至10重量%,优选0.01重量%至5重量%,优选0.1重量%至3重量%。
本发明还涉及一种使肽酶稳定的方法,包括以下方法步骤:
a)提供至少一种肽酶和
b)加入至少一种生物表面活性剂,
以获得肽酶稳定的组合物。
本发明的方法优选进行以使根据本发明优选的本发明组合物作为肽酶稳定的组合物而获得。
本发明还涉及至少一种用于使至少一种特别是在本发明的组合物中肽酶的酶活性得以稳定的生物表面活性剂。
在根据本发明的用途的情况下,本发明组合物及其优选组分的特别优选的实施方案是以优选量而使用。
因此,根据本发明的特别优选的用途的特征在于所述肽酶选自EC 3.4.24.28的杆菌溶素组,EC 3.4.24.62的枯草杆菌蛋白酶组和EC 3.4.21.27的嗜热菌蛋白酶组,并且所用生物表面活性剂选自鼠李糖脂和槐糖脂。
以下引用的实施例以示例的方式对本发明进行说明,而不意图限制本发明,其应用范围来自整个说明书和权利要求书以及实施例中指定的实施方案。

图1:加入月桂基醚硫酸钠(SLES)对具有线性烷基苯磺酸盐(LAS)的配制物中的0.8L储存稳定性的影响(参见表1)。与存储在冰箱中酶的活性对比图。SLES几乎不能促进酶的稳定。

实施例:
实施例1:在线性烷基苯磺酸盐(LAS),LAS与月桂基醚硫酸钠(SLES)的混合物、LAS与鼠李糖脂的混合物以及LAS与槐糖脂的混合物中,蛋白酶的储存稳定性的对比
本研究应显示SLES、鼠李糖脂和槐糖脂在LAS存在下对蛋白酶Neutrase和Alcalase的稳定作用。为了研究所述储存稳定性,在0.1M三乙醇胺缓冲液(TEA)pH=8中制备了表面活性剂混合物。也加入蛋白酶抑制剂1,2-丙二醇和硼酸。按需要,通过加入酸或碱而将混合物调节至pH=8。将液体制剂的蛋白酶加入到相关混合物中,混合并将混合物储存在30℃(参见表1和2)。所述蛋白酶是产品0.8L和2.4LFG(枯草杆菌蛋白酶)。在各个时间点从这些组合物中提取样品,并用0.1M磷酸盐缓冲液(pH=7)稀释100倍。将这些各个稀释的溶液中的100μl用滴管加入到96孔的微孔板的孔中。然后向其中加入100μl 0.4mg/ml的底物N-琥珀酰-Ala-Ala-Pro-Phe对硝基酰苯胺(Sigma Aldrich)在0.1M磷酸盐缓冲液(pH=7)中的溶液中,并且经由底物在37℃下的水解,在410nm下在微孔板读数器(Tecan,Infitite M200 Pro)中测量酶的活性。活性是由初始斜率而计算,并与存储在冰箱中的酶的酶活性对比。
在所有实施例中如下混合物用作鼠李糖脂:
通过HPLC证实的鼠李糖脂:
在所有实施例中使用的槐糖脂来自Ecover的槐糖脂,其酸与内酯的比例为60:40。
表1:0.1M TEA缓冲液中以重量比%计的组成。将组合物的pH调节至pH=8。
表2:包含LAS且具有和不具有各种稳定表面活性剂以及2.4LFG的组合物。数据是以在0.1M TEA缓冲溶液中以重量比%而计。将组合物的pH调节至pH=8。
实施例2:在不存在蛋白酶抑制剂下酶稳定性的提高
储存稳定性实验和活性测量以类似于实施例1而进行。制备了其他的组合物,其中并没有加入蛋白酶抑制剂1,2-丙二醇和硼酸。
表3:包含LAS且具有和不具有各种稳定表面活性剂、多元醇、抑制剂和Alcalase2.4L FG的组合物。数据是以在0.1M TEA缓冲溶液中以重量比%而计。将组合物的pH调节至pH=8。

实施例4:通过将阴离子型表面活性剂加入生物表面活性剂而改进蛋白质溶解性
除了酶在液体洗涤剂配制物中具有足够的保存稳定性以外,在应用中肽酶或蛋白酶的活性也极其重要。在与表面活性剂的相互作用中,它们有助于水不溶性蛋白质和水不溶性蛋白质土壤的溶解。
各种表面活性剂混合物可单独使用并且与蛋白酶组合使用以研究水不溶性但水分散性模型玉米蛋白的溶解度。玉米蛋白为来自玉米的储存蛋白的混合物。
在0.1M TEA缓冲液(pH=8)中制备了所有溶液/分散体。通过用超声波在超声波浴中处理30分钟并在磁性搅拌器进一步搅拌以使玉米蛋白颗粒均匀分散以制样而制备0.5重量%玉米蛋白(Sigma-Aldrich)的储备分散体。同样制备了10重量%的Alcalase 2.4L FG储备溶液。制备了由线性烷基苯磺酸盐(LAS,Marlon ARL,Sassol)以及总表面活性剂含量为0.11重量%的生物表面活性剂组成的表面活性剂储备溶液。所述线性LAS和生物表面活性剂的重量比例为100:0、75:25、50:50、25:75和0:100。将表面活性剂、酶和玉米蛋白储备溶液在微孔板(总液体体积220μl)中进行混合,并在微孔板读数器(Tecan,Infitite M200Pro)中在600nm(OD 600)下测量浊度。建立以下浓度:0.25重量%玉米蛋白,0.05重量%表面活性剂(混合物),0.45%Alcalase 2.4L FG。初始加入表面活性剂混合物和酶,然后通过加入玉米蛋白分散体而使反应开始。微孔板读数器中的所有溶液和测量中将温度控制在25℃。在80分钟内每分钟测量一次浊度的变化。在各自测量之间将板在读数器中晃动10秒钟。浊度的降低可能是玉米蛋白溶解的结果。
仅添加表面活性剂会导致玉米蛋白分散体的部分溶解(图9和图10)。加入蛋白酶但没有表面活性剂会导致缓慢但几乎完全溶解(图10和11)。与没有表面活性剂的酶相比,加入生物表面活性剂和蛋白酶会导致轻微加快的溶解。随着LAS的增加(基于表面活性剂总量的比例),溶解性越来越加快。这是由于蛋白质与表面活性剂(混合物)在蛋白质溶解中的协同作用。因此,LAS和生物表面活性剂的混合物是最佳的,以使与蛋白酶组合下达到快速的蛋白质溶解,并且同时获得尽可能高的表面活性剂混合物中蛋白酶的储存稳定性。在表4中对该组合物进行额外的描述。

Claims (10)
1.一种组合物,其包含:
A)至少一种肽酶,
B)至少一种生物表面活性剂,以及任选
C)至少一种阴离子表面活性剂。
2.权利要求1的组合物,其特征在于所述肽酶选自蛋白酶,特别是选自EC 3.4.21的丝氨酸蛋白酶和EC 3.4.24的金属蛋白酶。
3.权利要求1或2的组合物,其特征在于所述生物表面活性剂选自鼠李糖脂和槐糖脂。
4.前述权利要求至少之一的组合物,其特征在于存在的阴离子表面活性剂选自包含以下组分的组中,或者选自优选由以下组分组成的组中:烷基硫酸盐、烷基醚硫酸盐、任选烷氧基化磺基琥珀酸盐、任选烷氧基化甲基磺基琥珀酸盐、任选烷氧基化磺酸盐、任选烷氧基化甘氨酸盐、任选烷氧基化谷氨酸盐、任选烷氧基化羟乙基磺酸盐、任选烷氧基化羧酸盐、任选烷氧基化茴香酸盐、任选烷氧基化乙酰丙酸盐、任选烷氧基化酒石酸盐、任选烷氧基化乳酸盐、任选烷氧基化牛磺酸盐、任选烷氧基化丙氨酸盐、任选烷氧基化磷酸盐、任选烷氧基化磺基乙酸盐、任选烷氧基化磺基琥珀酰胺盐、任选烷氧基化肌氨酸盐和任选烷氧基化膦酸盐,尤其是任选烷氧基化磺酸盐、烷基硫酸盐和烷基醚硫酸盐。
5.前述权利要求至少之一的组合物,其特征在于所有表面活性剂总和的比例为0.1重量%至50重量%,优选1重量%至25重量%,优选5重量%至20重量%,特别优选10重量%至20重量%,其中所述重量百分比基于总的组合物。
6.前述权利要求至少之一的组合物,其特征在于基于组合物中表面活性剂的总量,所有表面活性剂中生物表面活性剂的比例为5重量%至99重量%,优选20重量%至95重量%,特别优选25重量%至80重量%。
7.前述权利要求至少之一的组合物,其包含
D)水。
8.前述权利要求至少之一的组合物,其包含
E)至少一种蛋白酶抑制剂。
9.权利要求8的组合物,作为蛋白酶抑制剂其包含硼酸和/或其盐。
10.至少一种生物表面活性剂用于稳定至少一种肽酶的酶活性的用途。
