Experiment 1
51 female SPF-grade Balb/c mice (purchased from institute of laboratory animals, university of cooperative and medical sciences, China pharmaceutical sciences) with the weight of 20 +/-2 g, which are aged for 8 weeks, are taken and raised for one week, and then the mice are divided into 17 groups of 3 mice each.
Then, local hair on the back of the mouse is shaved by using an electric hair cutter and an electric shaver, two skins with the area of 0.5cm multiplied by 0.5cm are selected as test parts on the back of the mouse, and the two test parts are respectively positioned on two sides of the spine. Then, the soothing essence prepared according to the above embodiment and the comparative preparation is correspondingly smeared on the skin of the test part on one side, the test part on the other side is not treated, and the UV dose is 1.5J/cm2The skin of the test parts on both sides was irradiated with ultraviolet rays for 20 min.
After completion of the irradiation treatment, the mice were anesthetized with ether, and skin tissue specimens were cut on the test sites, after which the wounds of the mice were sutured. Then, the skin tissue surface is scraped with a scalpel to remove sebum and dandruff, and subcutaneous fat is removed. Washing with phosphate buffer solution with pH of 7.4, adding EDTA separating solution, placing in water bath at 37 deg.C for 2 hr, taking out the skin, and separating with sharp forceps under dissecting microscope. The EDTA separation solution consists of the following components in parts by mass: 7.6g of tetrasodium ethylene diamine tetraacetate; 6.83g of sodium chloride; 0.2g of potassium chloride; 1.15g of disodium hydrogen phosphate; potassium dihydrogen phosphate 0.2 g; 1.2mL of 1% phenol red solution; distilled water was added to 1000mL and the pH was adjusted to 7.2.
After separation of true epidermis, Langerhans cells in epidermis are stained, and the specific steps are as follows:
step a, placing the epidermis in acetone for fixing for 10min at room temperature;
step b, taking out the epidermis, and washing the epidermis for 3 times by using phosphate buffer solution with pH of 7.4, wherein each time lasts for 10 min;
step c, putting the cleaned epidermis into an Eppendorf tube containing a 1:50 mouse anti-mouse H-2I-a monoclonal antibody, fully developing the epidermis in the antibody by using forceps, and standing for one night at the temperature of 4 ℃;
d, taking out the epidermis the next day, washing the epidermis for 3 times with phosphate buffer solution with the pH value of 7.4, 10min each time, putting 1:100Vectastain ABC reagent after washing, and incubating for 1.5h at 37 ℃;
step e, taking out the epidermis and washing the epidermis for 3 times by using phosphate buffer solution with the pH value of 7.4, wherein each time lasts for 10 min;
and f, adding the washed epidermis into the DAB kit, observing under a microscope, controlling the color development time to be 10min, washing with distilled water, and stopping the color development reaction.
And step g, paving the slices under a dissecting microscope, sealing the slices with glycerol gelatin, and observing the slices under the microscope.
The reagents used in the above experiments were as follows: biotin labeling of mouse anti-murine H-2I-Ad antibody (accession number 550554, BD PharMigen USA); 1:100 Vector ABC reagent (Vector Laboratories, Inc., U.S. A); DAB kit (purchased from gold bridge of China fir, Beijing, No. ZLI-9032).
Observing the stained slide under a microscope, wherein dendritic cells with brownish brown cell membranes and/or cytoplasms are Langerhans cells, randomly selecting 5 discontinuous fields for each slide under a 400-fold optical microscope, counting the number of positive cells in squares, and calculating the cell density (cells/mm) in unit area2) Firstly, the average value of the cell density obtained by calculation in 5 discontinuous visual fields of the same mouse is taken, and then the average value of the cell density of the skin tissue at the same side of a plurality of mice in the same group is calculated and recorded.
Experiment 2
The soothing essence prepared in the above examples and comparative examples was applied to the wound suture sites of mice in experiment 1,2 times a day, 1 time in the morning and evening, and 1mL each time, and the wound healing conditions of the mice in each group were observed and recorded.
Experiment 3
170 female volunteers with similar skin conditions are randomly selected, the age of the volunteers is 25-45 years, the female volunteers are randomly divided into 17 groups, the soothing essence prepared by the embodiment and the proportion is respectively and correspondingly used in each group, the soothing essence is uniformly applied to the cheek, and the female volunteers are used for 2 times every day, once in the morning and evening, 2mL each time and 30 days after insisting on. The following tests were also performed on the volunteers, and the average of the test results was recorded for each group of volunteers:

According to the comparison of the data of the example 3 and the comparative examples 1-2 in the table 2, the comparative example 2 is obtained by adding the carboxymethyl beta-dextran sodium on the basis of the comparative example 1, so that the effect of protecting the Langerhans cells of the soothing essence is improved, and the Langerhans cells are not easily damaged by ultraviolet rays; meanwhile, the preparation method is beneficial to activating macrophages to generate cytokines, epidermal growth factors and angiogenesis factors to a certain extent, so that the wound healing is promoted better, and the generation of wrinkles is delayed better. However, the effects of protecting langerhans cells, promoting wound healing and delaying wrinkle generation of comparative example 2 are all inferior to those of example 3 to a certain extent, which indicates that the problem of compatibility exists when carboxymethyl beta-dextran sodium is added alone, so that the effect of carboxymethyl beta-dextran sodium is easily influenced to a certain extent.
As can be seen from comparison of the data of example 3 with comparative example 1 and comparative examples 3 to 7 in table 2, the effect of protecting langerhans cells, the effect of promoting wound healing and the effect of delaying the generation of wrinkles of the soothing serum were affected regardless of the absence of any of the substances.
Comparison of the data from examples 3-5 in Table 2 shows that the addition of panthenol facilitates better activation of macrophages to produce epidermal growth factor and angiogenic factors, and thus better wound healing.
According to the comparison of the data of example 3 and examples 6-9 in table 2, the synergistic combination of panthenol and arginine is beneficial to activating macrophages to produce cytokines, epidermal growth factors and angiogenesis factors, so that the wound healing and wrinkle removal are promoted.
The above embodiments are preferred embodiments of the present application, and the protection scope of the present application is not limited by the above embodiments, so: all equivalent changes made according to the structure, shape and principle of the present application shall be covered by the protection scope of the present application.

Claims (10)

0.03-0.08% of carboxymethyl beta-glucan sodium;

8-10% of a humectant;
