Description
Technical Field
The application relates to the field of repair skin care products, in particular to a repair cream containing carboxymethyl chitosan and a preparation method thereof.
Background
In recent years, the number of sensitive muscle people is increasing, and the sensitive muscle people comprise pathological skin diseases and neurogenic inflammation; external causes include environmental factors, lifestyle and messy skin care products, use of hormone containing products; the natural defense system of the skin is problematic, the immunity of the skin is low, the barrier function of the skin is too weak, and the skin cannot resist external stimulation.
More and more repair products appear in the market, and the requirements of consumers cannot be met. The main reasons are that the general repair cream on the market has limited repair capability or the safety of repair products is not high. Therefore, improvement is yet to be achieved.
Disclosure of Invention
In order to provide a repair cream with strong repair capability and high safety, the application provides a repair cream containing carboxymethyl chitosan and a preparation method thereof.
In a first aspect, the present application provides a repair cream containing carboxymethyl chitosan, which adopts the following technical scheme:
the repairing cream containing carboxymethyl chitosan comprises the following components in percentage by mass: phase A: 4-7% of humectant A, 1-3% of thickening agent, 0-1% of antioxidant and 1-3% of skin conditioner;
phase B: 0.1-1% of humectant B, 1-4% of carboxymethyl chitosan, 0.1-1.5% of tetrahydro-methyl pyrimidine carboxylic acid, 0.1-0.8% of kava root extract and 0.5-2% of dextrin;
the water is supplemented to 100 percent.
According to the technical method, an additive-free preservative system is adopted, so that irritation and allergy caused by a traditional preservative system are avoided, and the risk of skin damage caused by stress reaction of the skin is reduced; moreover, the technical scheme of the application preferentially selects the skin-friendly raw materials which are high in safety and free of risks of causing acne and allergy, so that the safety is ensured.
In the aspect of repairing effect, the technical scheme of the application is mainly used for repairing cream by specific matching of carboxymethyl chitosan, tetrahydro-methyl pyrimidine carboxylic acid and kava root extract, and shows excellent repairing effect. Clinical tests show that the red-fading effect on sensitive skin is particularly obvious, and the anti-inflammatory repair capability of the skin can be improved.
When the skin barrier is injured, the activated interleukin 1a is released along with the damage of the epidermis, and then new active factors such as interleukin 1a, TNF-a, GM-CSF, interleukin-6 and the like are generated and released, so that the migration of inflammatory cells is promoted, and macrophage-inducing factors are promoted to form inflammation.
The inventor finds that under the combined action of carboxymethyl chitosan, tetrahydro-methyl pyrimidine carboxylic acid and kava root extract, the compound has an inhibitory effect on active factors, and particularly can remarkably reduce the levels of interleukin-6 and TNF-a in cells, thereby relieving the severity of skin inflammation. And the cooperation of the three can promote the ability of the tetrahydro-methyl pyrimidine carboxylic acid to bind water molecules, form a stronger and tougher protective shell, play a certain role in blocking factors and further relieve inflammation.
In addition to relieving inflammation, the product of the present application is used for a long timeCan also effectively improve skin tolerance. The common matching of the kava root extract and other raw materials further improves the TRPV1 and Ca content of the kava root extract 2+ The inhibitory effect of the influx, thereby alleviating the stinging and burning sensation of the skin.
Preferably, the carboxymethyl chitosan, the tetrahydro-methyl pyrimidine carboxylic acid and the kava root extract are mixed in a mass ratio of 1: (0.2-0.4): (0.1-0.3).
By adopting the technical scheme, the use ratio of the carboxymethyl chitosan, the tetrahydro-methyl pyrimidine carboxylic acid and the kava root extract is further limited. Under the limited dosage and proportion, the three have more sufficient matching effect, and the repairing and anti-inflammatory effects can be further improved.
The inventor guesses that the reaction can be rapidly carried out after the charge on the surface of the carboxymethyl chitosan and the charge on the surface of the tetrahydro-methyl pyrimidine carboxylic acid generate the attraction effect of positive and negative charges; meanwhile, due to the attraction effect, the stability of the whole structure is improved. An optimal balance point exists between the positive charges and the negative charges, and the matching effect of the positive charges and the negative charges is better under the limited proportion.
Preferably, the humectant B is 1,2-hexanediol, 1,2-pentanediol or a mixture of two of the two.
Preferably, the humectant B is 1,2-hexanediol, and the mass ratio of 1,2-hexanediol to carboxymethyl chitosan is (0.1-0.3): 1.
as no preservative is adopted, the specific 1,2-hexanediol is selected to be matched with the carboxymethyl chitosan together, so that a certain antibacterial effect is achieved, and the integral storage performance of the product is improved.
Preferably, the humectant A is one or a mixture of glycerol, propylene glycol, butanediol, sodium hyaluronate and squalene.
Preferably, the humectant A is glycerol and sodium hyaluronate, and the mass ratio of the glycerol to the sodium hyaluronate is (90-120): 1, and mixing.
By adopting the technical scheme, the inventor finds that the more the glycerol and the sodium hyaluronate are used, the better the glycerol and the sodium hyaluronate are, and the two may generate antagonistic effect. Glycerin and sodium hyaluronate need to be limited in a certain proportion to exert good moisturizing effect, namely the range defined in the application.
Preferably, the carboxymethyl chitosan has a carboxylation degree of 80 to 90% and a molecular weight of 8000 to 9000.
By adopting the technical scheme, the hydroxylation degree and the molecular weight of the carboxymethyl chitosan are further limited. Under the limitation, the carboxymethyl chitosan can be mixed in the system more quickly and uniformly, so that the carboxymethyl chitosan is beneficial to being matched with the tetrahydro-methyl pyrimidine carboxylic acid and the kava root extract to improve the repairing and anti-inflammatory effects.
In a second aspect, the present application provides a method for preparing a repair cream containing carboxymethyl chitosan, which adopts the following technical scheme:
a preparation method of a repair cream containing carboxymethyl chitosan comprises the following steps:
mixing the water and the phase A together, heating to 80-88 ℃, and stirring until the water and the phase A are uniformly dissolved;
cooling to 35-45 deg.C, adding phase B, and stirring to obtain final product.
By adopting the technical scheme, various raw materials are mixed step by step, so that better mixing and dispersing effects are achieved, and the influence on the matching effect due to uneven mixing is reduced.
Preferably, the water and the phase A are stirred until the water and the phase A are uniformly dissolved, the temperature is kept for 18-30min, and then the phase B is cooled and added;
when mixing the phase B, adjusting the rotating speed to 550-650r/min.
The speed of agitation is further limited when mixing phase B. The inventor finds that the carboxymethyl chitosan, the tetrahydro-methyl pyrimidine carboxylic acid and the kava root extract have more sufficient mixing and dispersing effects at the rotating speed, and are favorable for promoting the attraction between the carboxymethyl chitosan and the tetrahydro-methyl pyrimidine carboxylic acid, so that the reaction speed is accelerated and a stable structure is formed.
In summary, the present application has the following beneficial effects:
1. according to the technical method, an additive-free preservative system is adopted, so that irritation and allergy caused by a traditional preservative system are avoided, and the risk of skin damage caused by stress reaction of the skin is reduced; moreover, the technical scheme of the application preferentially selects the skin-friendly raw materials which are high in safety and free of risks of causing acne and allergy, so that the safety is ensured.
2. The carboxymethyl chitosan is used for rapidly repairing damaged cuticle, improving skin barrier, achieving the effect of repairing efficacy, matching the effects of improving skin tolerance and relieving irritation of kava root extract, and multiplying the cell protection mechanism of tetrahydro-methyl pyrimidine carboxylic acid to form a protective shell, so that the skin cuticle barrier is improved and repaired.
3. Under the combined action of carboxymethyl chitosan, tetrahydro-methyl pyrimidine carboxylic acid and kava root extract, it has the effect of inhibiting active factor, and can notably reduce the level of interleukin-6 and TNF-a in cell, so that it can relieve the severity of scytitis. In addition, the cooperation of the three components can promote the ability of binding water molecules by the tetrahydro-methyl pyrimidine carboxylic acid, so that a stronger and tougher protective shell is formed, a certain blocking effect is achieved on factors, and inflammation is further relieved.

Performance test
1. And (3) detecting the anti-inflammatory effect:
1) Test samples: the products of examples 1-10, comparative examples 1-4.
2) Detection of the object: selecting 28 volunteers with good health, no infectious diseases, no allergic diseases, no cosmetics or other external preparations and no allergy history, wherein the age of the volunteers is 18-30 years old.
2 volunteers were assigned to 1 group, corresponding to one test sample.
3) The detection method comprises the following steps:
A. histamine stimulation, taken using an andrera 3D multifunctional 3D skin imager.
B. The corresponding test sample (about 1 g) is smeared on the cheek of the subject, after the test sample is absorbed by the skin, the skin of the subject is stimulated by histamine, and after the red and swollen stimulation reaction of the skin, the skin is photographed and observed. According to the following table 2, the anti-inflammatory effect is better when the face is recorded according to the face shooting conditions of 0h, 1h and 3h of the detection object, and the higher the score is.
TABLE 2
2. And (3) detecting the moisturizing effect:
in the detection method B of test 1, the skin moisture detector is used to detect the detection area of the detection object, and the detection nodes are before the test sample is applied to the subject and during 3 hours of shooting.
3. Storage performance: the products of examples 1-10 were placed in a room at 45 ℃ for 60 days, and after taking out, the paste was observed with the naked eye without change.
The results of the above tests 1-2 are shown in Table 3.
TABLE 3-1
TABLE 3-2
As can be seen from the comparison of the test data of examples 1-3 and comparative examples 1-3 in Table 3, at 0h, the skin reddens due to the stimulation of histamine, and the area of the facial redness of the user who uses the repair cream of examples 1-3 in advance is slightly smaller than that of comparative examples 1-3, and the depth of the redness is slightly lighter than that of comparative examples 1-3; and at 1 hour and 3 hours, the redness and swelling of the face of the user coated with the products of the examples 1 to 3 are quickly reduced and lightened, and the effect is obviously better than that of the products of the comparative examples 1 to 3. It is demonstrated that the use of the product of the present application in advance is beneficial for relieving inflammation of the skin. The inventors hypothesized that the co-formulation of carboxymethyl chitosan, tetrahydro-methyl pyrimidine carboxylic acid and kava root extract reduced the levels of interleukin-6 and TNF-a in the cells and formed a protective shell that served as a barrier effect, thus achieving anti-inflammatory repair.
According to the comparison of the detection results of examples 1 and 4-5 with those of examples 6 and 4, the redness and swelling of the face are better than those of comparative examples 1-3 after the product of comparative example 4 is used, but the inflammation relieving effect is far lower than that of examples 1 and 4-6. Example 6 is somewhat different from examples 1, 4-5. It is explained that the compounding effect of the raw materials can be sufficiently exhibited by not only limiting the use of carboxymethyl chitosan, tetrahydro-methyl pyrimidine carboxylic acid and kava root extract but also further limiting the use ratio and the use amount of the three.
According to the comparison of the test data of example 1 and examples 8-9, the anti-inflammatory repair effect of example 1 is similar to that of examples 8 and 9, and the differences between examples 8 and 9 and example 1 are only slight at 1h and 3h. The examples 1 and 8-9 slightly diverge in moisturizing effect, and the moisturizing effect of example 1 is better than that of example 8, and the moisturizing effect of example 8 is better than that of example 9. It is shown that glycerin and sodium hyaluronate need to be limited in a certain proportion to exert good moisturizing effect.
As can be seen from comparison of the test data of example 1 and example 10, the product of example 1 is superior to that of example 10 in the moisturizing effect and anti-inflammatory effect. It is necessary to further define the technological parameters for preparing the product of the application, so that the product has better mixing and dispersing effects.
The present embodiment is only for explaining the present application, and it is not limited to the present application, and those skilled in the art can make modifications of the present embodiment without inventive contribution as needed after reading the present specification, but all of them are protected by patent law within the scope of the claims of the present application.
