本发明属于酵素技术领域,特别涉及一种降尿酸的复合寡糖酵素酶及其制备方法。该降尿酸的复合寡糖酵素酶,主要由以下质量份数的原料制成:车前子提取物5‑8份、枸杞提取物3‑5份、锌酵母0.1‑0.2份、红糖5‑6份、茶树花提取物7‑8份、葡萄籽粉5‑7份、蒸馏水60‑62份、黄花菜粉10‑15份、壳寡糖1‑2份、维生素A0.01‑0.02份、迷迭香提取液0.05‑0.07份。上述原料经发酵、过滤除渣、混合陈化后,制得降尿酸的复合寡糖酵素酶。本发明原料易得、制备工艺稳定,制得的降尿酸的复合寡糖酵素酶可促进人体尿酸代谢,有效降低血尿酸值,且无明显副作用,适合进行推广。

技术领域
本发明属于酵素技术领域,特别涉及一种降尿酸的复合寡糖酵素酶及其制备方法。
背景技术
高尿酸血症是人体因嘌呤代谢发生紊乱,导致血液中尿酸增多而引起的一种代谢性疾病。高尿酸血症是痛风的发病基础,同时高尿酸血症还会导致高血压、糖尿病、肥胖、冠心病、肾病的发病率升高。
目前治疗高尿酸血症的药物分为两大类:一类是抑制尿酸生成的药物,另一类是促进尿酸排泄的药物;但这两类药物的副作用均较大。而基于中药等植物成分发酵制成的酵素,因其调理机体的作用相对温和、无明显副作用,成为控制或治疗高尿酸血症产品的研发方向。
发明内容
本发明为了弥补现有技术中治疗高尿酸血症药物副作用大的问题,提供了一种降尿酸的复合寡糖酵素酶及其制备方法。
本发明是通过如下技术方案实现的:
一种降尿酸的复合寡糖酵素酶,主要由以下质量份数的原料制成:车前子提取物5-8份、枸杞提取物3-5份、锌酵母0.1-0.2份、红糖5-6份、茶树花提取物7-8份、葡萄籽粉5-7份、蒸馏水60-62份、黄花菜粉10-15份、壳寡糖1-2份、维生素A0.01-0.02份、迷迭香提取液0.05-0.07份。
上述原料的优选质量配比为:
车前子提取物6.5份、枸杞提取物4份、锌酵母0.15份、红糖5.5份、茶树花提取物7.5份、葡萄籽粉6份、蒸馏水61份、黄花菜粉12.5份、壳寡糖1.5份、维生素A0.015份、迷迭香提取液0.06份。
本发明以利尿益肾的中药为主料,同时添加多种调节代谢的辅剂制得所述降尿酸的复合寡糖酵素酶;该降尿酸的复合寡糖酵素酶富含多种促进尿酸代谢的有效成分,服用后可显著降低血尿酸值。
本发明中,车前子提取物由车前子提取制成;车前子是车前科植物车前或平车前的干燥成熟种子,其味甘,性寒,具有清热利尿通淋、渗湿止泻、明目、祛痰的作用;用于治疗热淋涩痛,水肿胀满,暑湿泄泻,目赤肿痛,痰热咳嗽。
枸杞提取物由枸杞提取制成;枸杞主要含甜菜碱、阿托品、天仙子胺,对免疫功能有影响作用,还具有降血糖、调节血压、抗肿瘤等作用,入药具有养肝、滋肾、润肺的功效。
锌酵母的锌含量可达50000mg/kg,酵母锌的吸收利用率高达70%以上,补锌效果是传统补锌制剂的5-10倍,且消除了无机锌的副作用。
红糖是以甘蔗为原料制炼而成的粗糖,具有益气养血,健脾暖胃,驱风散寒,活血化淤之效;红糖还具有利尿作用。
茶树花提取物由茶树花提取制成;茶树花与茶叶一样,也有很多对人体有益的功效。茶树花中蛋白质、茶多糖、茶多酚、活性抗氧化物质均超出茶叶中同类物质含量,因而茶树花具有解毒、降脂、降糖、抗衰老、抗癌、滋补、壮体、养颜美容等功效。
葡萄籽粉由葡萄籽制成;葡萄籽含有多酚类物质,还含有粗蛋白、氨基酸、多种维生素及微量元素。葡萄籽具有抗氧化、降血脂、抗过敏、保护血管的功效,还能降低糖尿病、心脏病、关节炎等病症的发病风险。
黄花菜粉由黄花菜制成;黄花菜,又名金针菜、柠檬萱草、忘忧草,属百合目百合科多年生草本植物。黄花菜性味甘凉,有止血、消炎、清热、利湿、消食、明目、安神、利尿消肿等功效,对吐血、大便带血、小便不通、失眠、乳汁不下等症有疗效。
壳寡糖能调节肠道内微生物的代谢活动,改善肠道微生物区系分布,从而提高机体免疫力,使肠道内pH下降,抑制肠道有害菌生长,产生B族维生素,分解致癌物质,促进肠蠕动,增进蛋白质吸收。壳寡糖还具有抗肿瘤、降血脂、调节血糖、改善肝脏和心肺功能等多种作用。
维生素A具有促进细胞增殖分化,维持骨骼、上皮组织、视觉正常功能等作用,维生素A还可调节机体免疫应答。
迷迭香提取液由迷迭香提取制成;迷迭香具有抗氧化性,已广泛用于医药、油炸食品、富油食品及各类油脂的保鲜保质,是一种天然食品防腐剂。此外,迷迭香具有镇静安神、醒脑的作用,对消化不良和胃痛均有一定疗效,还可改善语言、视觉、听力方面的障碍,提高注意力,治疗风湿痛,强化肝脏功能,降低血糖,有助于动脉硬化的治疗,帮助麻痹的四肢恢复活动能力。
上述降尿酸的复合寡糖酵素酶的制备方法,包括如下步骤:(1)将车前子提取物、枸杞提取物、锌酵母、红糖、茶树花提取物、葡萄籽粉、蒸馏水、黄花菜粉按配比加入发酵釜并搅拌均匀后,于43-45℃发酵60-62天,得到发酵产物;(2)发酵产物经过滤、除渣后,收集得到滤液;(3)将滤液加入调配釜,同时按配比向调配釜中加入壳寡糖、维生素A、迷迭香提取液并搅拌均匀后,陈化36小时,得到降尿酸的复合寡糖酵素酶;(4)对降尿酸的复合寡糖酵素酶进行无菌包装。
本发明利用车前子提取物、枸杞提取物、黄花菜粉等主料发酵制得发酵产物,再滤除发酵产物中不易被吸收的残渣,仅将发酵产物的滤液与壳寡糖、维生素A等辅剂混合陈化后,制得所述降尿酸的复合寡糖酵素酶;该降尿酸的复合寡糖酵素酶中的有效成分易于被人体吸收,从而促进人体尿酸代谢,有效降低血尿酸值。
本发明相对于现有技术,具有以下有益效果:
本发明以车前子提取物、枸杞提取物、黄花菜粉这些利尿促排益肾的中药为主料,同时配合茶树花提取物、葡萄籽这些促进代谢的植物成分发酵得到发酵产物,即酵素,所述发酵产物含有多种调节尿酸代谢的生物活性成分。为进一步提高本发明所述降尿酸的复合寡糖酵素酶在调节代谢方面的作用,向所述发酵产物经过滤、除渣后得到的滤液中加入壳寡糖、维生素A、迷迭香提取液,制得所述降尿酸的复合寡糖酵素酶;壳寡糖和维生素A均具有调节多种脏器功能的作用;迷迭香提取液可提高本发明所述降尿酸的复合寡糖酵素酶的保质期。本发明所述降尿酸的复合寡糖酵素酶经试验,可有效降低血尿酸值,取得了良好的应用效果。
本发明原料易得、制备工艺稳定,制得的降尿酸的复合寡糖酵素酶可促进人体尿酸代谢,有效降低血尿酸值,且无明显副作用,适合进行推广。
具体实施方式
下面结合具体实施例对本发明的技术方案进行详细说明,以便于对本发明的理解,但并不是对本发明的限制。
实施例1:一种降尿酸的复合寡糖酵素酶,主要由以下质量配比的原料制成:车前子提取物5kg、枸杞提取物3kg、锌酵母0.1kg、红糖5kg、茶树花提取物7kg、葡萄籽粉5kg、蒸馏水60kg、黄花菜粉10kg、壳寡糖1kg、维生素A0.01kg、迷迭香提取液0.05kg。
上述原料的具体制备方法如下:
(1)将车前子提取物、枸杞提取物、锌酵母、红糖、茶树花提取物、葡萄籽粉、蒸馏水、黄花菜粉按配比加入发酵釜并搅拌均匀后,于43℃发酵60天,得到呈深褐色并有甜酸味的发酵产物;(2)发酵产物经过滤、除渣后,收集得到滤液;(3)将滤液加入调配釜,同时按配比向调配釜中加入壳寡糖、维生素A、迷迭香提取液并搅拌均匀后,陈化36小时,得到降尿酸的复合寡糖酵素酶;(4)对降尿酸的复合寡糖酵素酶进行无菌包装。
应用试验:(1)试验分组:筛选得到年龄40-45周岁的男、女高尿酸血症患者各4名;将上述筛选得到的高尿酸血症患者随机分为试验组和对照组,每组高尿酸血症患者均为4名,且每组高尿酸血症患者男、女各2名;每名试验组高尿酸血症患者每日按1克每千克体重服用本实施例制得的降尿酸的复合寡糖酵素酶,并保持常规饮食,连续服用60天;每名对照组高尿酸血症患者每日按1克每千克体重服用与本实施例制得的降尿酸的复合寡糖酵素酶具有相同外观和口感的安慰剂,连续服用60天,该安慰剂由食用淀粉制成。(2)指标检测:分别检测两组高尿酸血症患者服用本实施例制得的降尿酸的复合寡糖酵素酶或安慰剂前1天、连续服用第15天、连续服用第30天和连续服用第60天的血尿酸值、尿液pH值、24h尿尿酸定量值。
试验结果:两组高尿酸血症患者服用本实施例制得的降尿酸的复合寡糖酵素酶或安慰剂前1天、连续服用第15天、连续服用第30天和连续服用第60天的血尿酸值、尿液pH值、24h尿尿酸定量值如下所示:
结果分析:本实施例中,连续服用本实施例制得的降尿酸的复合寡糖酵素酶的试验组高尿酸血症患者的血尿酸值持续降低,试验组高尿酸血症患者的尿液pH值逐渐升高,同时试验组高尿酸血症患者尿液中尿酸排泄量显著升高;而对照组高尿酸血症患者的各项指标未出现明显变化;服用本实施例制得的降尿酸的复合寡糖酵素酶或安慰剂后,两组高尿酸血症患者均未出现明显副作用。以上结果说明本实施例制得的降尿酸的复合寡糖酵素酶可促进人体尿酸代谢,长期服用可有效降低血尿酸值。
实施例2:一种降尿酸的复合寡糖酵素酶,主要由以下质量配比的原料制成:车前子提取物8kg、枸杞提取物5kg、锌酵母0.2kg、红糖6kg、茶树花提取物8kg、葡萄籽粉7kg、蒸馏水62kg、黄花菜粉15kg、壳寡糖2kg、维生素A0.02kg、迷迭香提取液0.07kg。
上述原料的具体制备方法如下:
(1)将车前子提取物、枸杞提取物、锌酵母、红糖、茶树花提取物、葡萄籽粉、蒸馏水、黄花菜粉按配比加入发酵釜并搅拌均匀后,于45℃发酵62天,得到呈深褐色并有甜酸味的发酵产物;(2)发酵产物经过滤、除渣后,收集得到滤液;(3)将滤液加入调配釜,同时按配比向调配釜中加入壳寡糖、维生素A、迷迭香提取液并搅拌均匀后,陈化36小时,得到降尿酸的复合寡糖酵素酶;(4)对降尿酸的复合寡糖酵素酶进行无菌包装。
应用试验:(1)试验分组:筛选得到年龄40-45周岁的男、女高尿酸血症患者各4名;将上述筛选得到的高尿酸血症患者随机分为试验组和对照组,每组高尿酸血症患者均为4名,且每组高尿酸血症患者男、女各2名;每名试验组高尿酸血症患者每日按1克每千克体重服用本实施例制得的降尿酸的复合寡糖酵素酶,并保持常规饮食,连续服用60天;每名对照组高尿酸血症患者每日按1克每千克体重服用与本实施例制得的降尿酸的复合寡糖酵素酶具有相同外观和口感的安慰剂,连续服用60天,该安慰剂由食用淀粉制成。(2)指标检测:分别检测两组高尿酸血症患者服用本实施例制得的降尿酸的复合寡糖酵素酶或安慰剂前1天、连续服用第15天、连续服用第30天和连续服用第60天的血尿酸值、尿液pH值、24h尿尿酸定量值。
试验结果:两组高尿酸血症患者服用本实施例制得的降尿酸的复合寡糖酵素酶或安慰剂前1天、连续服用第15天、连续服用第30天和连续服用第60天的血尿酸值、尿液pH值、24h尿尿酸定量值如下所示:
结果分析:本实施例中,连续服用本实施例制得的降尿酸的复合寡糖酵素酶的试验组高尿酸血症患者的血尿酸值持续降低,试验组高尿酸血症患者的尿液pH值逐渐升高,同时试验组高尿酸血症患者尿液中尿酸排泄量显著升高;而对照组高尿酸血症患者的各项指标未出现明显变化;服用本实施例制得的降尿酸的复合寡糖酵素酶或安慰剂后,两组高尿酸血症患者均未出现明显副作用。以上结果说明本实施例制得的降尿酸的复合寡糖酵素酶可促进人体尿酸代谢,长期服用可有效降低血尿酸值。
实施例3:一种降尿酸的复合寡糖酵素酶,主要由以下质量配比的原料制成:车前子提取物6.5kg、枸杞提取物4kg、锌酵母0.15kg、红糖5.5kg、茶树花提取物7.5kg、葡萄籽粉6kg、蒸馏水61kg、黄花菜粉12.5kg、壳寡糖1.5kg、维生素A0.015kg、迷迭香提取液0.06kg。
上述原料的具体制备方法如下:
(1)将车前子提取物、枸杞提取物、锌酵母、红糖、茶树花提取物、葡萄籽粉、蒸馏水、黄花菜粉按配比加入发酵釜并搅拌均匀后,于44℃发酵61天,得到呈深褐色并有甜酸味的发酵产物;(2)发酵产物经过滤、除渣后,收集得到滤液;(3)将滤液加入调配釜,同时按配比向调配釜中加入壳寡糖、维生素A、迷迭香提取液并搅拌均匀后,陈化36小时,得到降尿酸的复合寡糖酵素酶;(4)对降尿酸的复合寡糖酵素酶进行无菌包装。

结果分析:本实施例中,连续服用本实施例制得的降尿酸的复合寡糖酵素酶的试验组高尿酸血症患者的血尿酸值持续降低,试验组高尿酸血症患者的尿液pH值逐渐升高,同时试验组高尿酸血症患者尿液中尿酸排泄量显著升高;而对照组高尿酸血症患者的各项指标未出现明显变化;服用本实施例制得的降尿酸的复合寡糖酵素酶或安慰剂后,两组高尿酸血症患者均未出现明显副作用。以上结果说明本实施例制得的降尿酸的复合寡糖酵素酶可促进人体尿酸代谢,长期服用可有效降低血尿酸值。

1.一种降尿酸的复合寡糖酵素酶,其特征为,主要由以下质量份数的原料制成:车前子提取物5-8份、枸杞提取物3-5份、锌酵母0.1-0.2份、红糖5-6份、茶树花提取物7-8份、葡萄籽粉5-7份、蒸馏水60-62份、黄花菜粉10-15份、壳寡糖1-2份、维生素A0.01-0.02份、迷迭香提取液0.05-0.07份。
2.根据权利要求1所述的一种降尿酸的复合寡糖酵素酶,其特征为,主要由以下质量份数的原料制成:车前子提取物6.5份、枸杞提取物4份、锌酵母0.15份、红糖5.5份、茶树花提取物7.5份、葡萄籽粉6份、蒸馏水61份、黄花菜粉12.5份、壳寡糖1.5份、维生素A0.015份、迷迭香提取液0.06份。
3.根据权利要求1所述的一种降尿酸的复合寡糖酵素酶的制备方法,其特征为,包括如下步骤:(1)将车前子提取物、枸杞提取物、锌酵母、红糖、茶树花提取物、葡萄籽粉、蒸馏水、黄花菜粉按配比加入发酵釜并搅拌均匀后,于43-45℃发酵60-62天,得到发酵产物;(2)发酵产物经过滤、除渣后,收集得到滤液;(3)将滤液加入调配釜,同时按配比向调配釜中加入壳寡糖、维生素A、迷迭香提取液并搅拌均匀后,陈化36小时,得到降尿酸的复合寡糖酵素酶;(4)对降尿酸的复合寡糖酵素酶进行无菌包装。
