Description

Antibacterial itching-relieving red-fading acne-removing repairing composition as well as preparation method and application thereof
Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to a bacteriostatic itching-relieving red-fading acne-removing repairing composition as well as a preparation method and application thereof.
Background
Acne is also called acne and comedo, is a chronic inflammatory skin disease of pilosebaceous units, is frequently generated in teenagers, has great influence on the psychology and the social interaction of the teenagers, and can be naturally relieved or healed after the adolescence. Clinically, it is manifested by facial skin lesions of various shapes, such as acne, papule, pustule, and nodule. Acne occurs in close relation to factors such as hyperseborrhea, blockage of pilosebaceous ducts, bacterial infection and inflammatory reactions. After adolescence, the level of androgen, particularly testosterone, in a human body is rapidly increased, the development of sebaceous glands is promoted, and a large amount of sebum is produced. Meanwhile, abnormal keratinization of the pilosebaceous canal causes the canal to be blocked, sebum is obstructed to be discharged, and a keratoplug, namely micro acne, is formed. A number of microorganisms in the hair follicle, particularly propionibacterium acnes, multiply in number, and lipases produced by propionibacterium acnes break down sebum to produce free fatty acids, while chemotactic inflammatory cells and mediators, ultimately inducing and exacerbating the inflammatory response.
Skin lesions are well developed on the face and upper chest and back. Non-inflammatory lesions of acne manifest as open and closed comedones. A typical skin lesion of closed comedones (also known as whiteheads) is a skin-colored papule of about 1 mm in size, with no obvious hair follicle opening. Open comedones (also known as blackheads) exhibit a dome-shaped papule with a markedly dilated follicular opening. The further progression of the acne can evolve into various inflammatory lesions, manifested as inflammatory papules, pustules, nodules, and cysts. The inflammatory papules are red and have different diameters of 1-5 mm; the pustules are consistent in size and are filled with white pus; the diameter of the nodule is more than 5 mm, and the nodule can feel hard and painful when touched; the cyst is located deeper and filled with a mixture of pus and blood. These lesions may also fuse to form large inflammatory plaques and sinus tracts, etc. After the inflammatory lesions subside, pigmentation, persistent erythema, depressed or hypertrophic scars often remain.
Problems with acne removal products on the market: 1. easily cause skin allergy and has great skin irritation; 2. adding chemical bacteriostatic agent to destroy normal flora of skin; 3. adding some forbidden substances such as hormones like chloramphenicol; 4. adding heavy metal substances. Therefore, the current research and development direction is how to realize bacteriostasis, itching relieving, red fading and acne removing by combining the active ingredients of the traditional Chinese medicines.
For example, chinese patent application CN111603417A discloses a traditional Chinese medicine based facial mask for removing spots, acnes and whitening skin and a processing method thereof, comprising the following raw materials by weight: 9% of moistening raw materials, 8% of damp clearing raw materials, 7% of acne removing raw materials, 25% of freckle removing raw materials, 21% of whitening raw materials and 30% of distilled water; wherein the moistening raw materials comprise 10% of ginseng, 45% of angelica tail, 15% of centella asiatica, 23% of snow lotus and 7% of gardenia; the dampness eliminating raw materials comprise 40% of white wormwood, 29% of rhizoma atractylodis and 31% of radix sophorae flavescentis; the acne-removing raw materials comprise 10% of fleece-flower root, 45% of phellodendron, 30% of calamine and 20% of oldenlandia diffusa; the raw materials for removing the freckles comprise 10 percent of cortex moutan, 55 percent of angelica dahurica, 10 percent of safflower and 25 percent of mint; the whitening raw materials comprise 20% of white morning glory, 10% of cynanchum atratum, 50% of white peony root, 8% of bearberry leaf and 12% of liquorice. The beneficial effects are as follows: the use of the moistening raw materials has no any adverse stimulation to the skin, can expand the capillary vessels of the skin, promote the blood circulation of the skin, enhance the nutrition of the skin, and prevent the skin from dehydration, hardening and wrinkling, thereby enhancing the elasticity of the skin, regenerating cells, keeping the skin smooth and moist, preventing premature aging, and the use function of the freckle removing raw materials can promote blood circulation and remove freckles, treat chloasma on the face, dispel wind and heat, promote eruption and remove sores, soothe liver and remove freckles, has the efficacy of promoting blood circulation and nourishing blood, treats the facial yellowing and the dull complexion of women, has better acne removing effect, and the use of the whitening raw materials has the functions of whitening, fading freckles, moistening and cleaning the skin.
For another example, chinese granted patent CN103202797B discloses an anti-acne lotion with anti-acne efficacy and a preparation method thereof, wherein the used traditional Chinese medicine extract is obtained by extracting the following raw material medicines in parts by weight by a conventional extraction method in the field of traditional Chinese medicine: 20-30 parts of artemisia capillaries, 10-30 parts of fructus kochiae, 10-20 parts of cortex moutan, 10-20 parts of gallnut, 10-20 parts of spina gleditsiae and 10-20 parts of selfheal.
For another example, chinese patent application CN110448472A discloses a water-oil two-phase instant mixing facial mask solution and a preparation method thereof, belonging to the technical field of skin care products, wherein the facial mask solution comprises a water phase and an oil phase; the water phase and the oil phase are separately packaged and are instantly mixed when in use; the mixing mass ratio of the water phase to the oil phase is (20-30) to (2-5); the water phase comprises a humectant, a thickening agent, a water-soluble functional additive, an auxiliary agent and water, and the mass ratio of the humectant to the water phase is (0.1-20): 0.1-15): 0.1-5): 45-99.5; the oil phase comprises an emollient, an oil-soluble functional additive and an emulsifier, and the mass ratio of the emollient to the oil-soluble functional additive to the emulsifier is (80-100): 0.1-15): 0.1-5); the water-soluble functional additive comprises one or more of carnosine, acetyl hexapeptide-8, oligopeptide-1, nonapeptide-1, collagen, yeast extract, vitamin C, tranexamic acid, licoflavone, ferulic acid, arginine, lysine, centella asiatica extract and lactobionic acid; the oil-soluble functional additive comprises one or more of oil-soluble astaxanthin, campesterol, camellia seed oil, jojoba seed oil, tocopherol, hydrogenated soybean lecithin, Daphnia giraldii Nitsche fruit oil, extract of flos seu radix camptosorus/leaf/vine, artichoke fruit oil, sunflower seed oil unsaponifiable matter, sweet almond oil, Chinese chestnut seed oil, peony seed oil and lecithin; the anti-sensitivity agent is one or more selected from dipotassium glycyrrhizinate, bisabolol, herba Portulacae extract, allantoin, tetrandrine, herba Polygoni Hydropiperis extract, radix Cynanchi Atrati extract, and folium Artemisiae Argyi extract. The facial mask liquid is fresh and mild in texture, is not sticky, does not irritate the skin, and does not aggravate the skin burden.
However, although the prior art reports that the cynanchum atratum extract, the centella asiatica extract and other components are combined to prepare the external composition for skin, the composition in the prior art has a complex formula, so that the production cost is overhigh, and the composition reported in the prior art is difficult to simultaneously exert the functions of inhibiting bacteria, relieving itching, fading red and removing acnes. Therefore, the invention aims to overcome the defects of the prior art and provide a composition which is simple in formula, does not contain additional active ingredients and preservatives, and can simultaneously exert the functions of inhibiting bacteria, relieving itching, fading red and removing acnes and application thereof.

Pharmacodynamic experiment
In vitro bacteriostasis experiment
A quantitative sample (prepared in examples 1-3 and comparative examples 1-2) of 0.1ml was applied to the medium inoculated with the test species, and the sample was allowed to diffuse and permeate within the agar medium over a certain period of time. Within the range of the inhibitory concentration, the growth of the indicator bacteria is inhibited, creating a sterile transparent area called zone of inhibition. Three replicates were run for each sample and the final experimental result was the average of the three replicates. Under a certain condition and in a certain range, the diameter of the inhibition zone and the logarithm of the concentration of the sample form a linear function relationship. By using the principle, the virulence of the bactericide can be compared, or the content of the effective components in the sample can be determined. The results of the zone of inhibition in vitro bacteriostatic experiments are shown in table 1.

And (3) judging standard of in vitro bacteriostasis qualitative result: the diameter of the inhibition zone is less than 8mm and is insensitive (-), 8-13 mm is low-degree sensitive (+), 13-19 mm is medium-degree sensitive (+ +), and more than 19mm is high-degree sensitive (+ +++). The experimental results show that the compositions of examples 1-3 and comparative example 1 are highly sensitive compositions, but the in vitro bacteriostatic effect of examples 1-3 is more excellent.
Second, testing oil control effect of product
An experimental instrument: skin oil tester
Figure BDA0003404858180000061
SM815
Sample preparation: the bacteriostatic, itching-relieving, red-fading and acne-removing repairing compositions prepared in examples 1-3 and comparative examples 1-2.
Blank: the test area was free of any cosmetic.
Experimental population
The subjects were 30 persons in total, and were divided into 6 groups on average. The specific gender and age composition of each group was determined randomly. In the meantime, if 2 persons had adverse reactions, the test was terminated.
Experimental methods
Selecting the forehead (divided into a left part and a right part) of a subject, and sequentially marking in a circulating way: a test area sample group and a blank control area;
secondly, the technician uses the skin oil tester to test the tested part for 3 to 5 times, and the average value is taken and recorded as the initial value.
Thirdly, smearing a sample on the experimental part of the subject, and using the bacteriostatic, itching-relieving, red-fading and acne-removing repairing composition.
And fourthly, after continuously using the antibacterial itching-relieving red-fading acne-removing repairing composition for one hour, two hours and four hours, measuring for 3-5 times by a technician by using a skin oil tester, and taking an average value.
And fifthly, counting the measured values and analyzing the change rule of the skin oil content.
Analysis of results
Change in oil content
The change of the oil content is reflected in a test period, and the oil content in an experimental area changes along with time. The larger the value, the larger the oil content, and conversely, the smaller the oil content.
As shown in FIG. 1, the differences between examples 1 to 3 and comparative examples 1 to 2 after 1 hour of use and the blank group are significant; the oil and fat contents of examples 1 to 3 and comparative examples 1 to 2 were significantly lower than those of the blank group within 4 hours of use of examples 1 to 3 and comparative examples 1 to 2, and the oil and fat contents of examples 1 to 3 were significantly lower than those of comparative examples 1 to 2.
Change in oil increase rate
Oil increase rate (sample group oil content-initial oil content)/initial oil content
As shown in fig. 2, in the test period, after 1 hour of applying the bacteriostatic, antipruritic, red-fading, acne-removing repairing composition to the tested part, the increase rate of skin oil secretion is obviously lower than that of the blank group and is always lower than that of the blank group compared with the initial value.
Fat suppression rate
The relative increase rate of the grease, namely the grease inhibition rate (the grease content of the sample group-the grease content of the blank group)/the grease content of the blank group; the smaller this value, the more significant the effect of fat inhibition of the test sample relative to the blank sample.
As shown in fig. 3, after the bacteriostatic, antipruritic, red-fading and acne-removing repairing composition is applied to the tested part in the test period, the fat inhibition rate is always negative.
Conclusion of the experiment
After the bacteriostatic itching-relieving red-fading acne-removing repairing composition prepared in the embodiment 1-3 of the invention is used, the grease secretion of the tested part is obviously lower than that of the tested area without being smeared with a sample, and the effect is more obvious than that of the comparative example 1-2, which shows that the bacteriostatic itching-relieving red-fading acne-removing repairing composition has the capability of inhibiting the grease secretion of skin.
Third, acne removing test
Trial objects: 120 volunteers with acne on face were selected as test subjects, the 120 test subjects were equally divided into 6 groups of 20 subjects, and the bacteriostatic, antipruritic, reddish-fading and acne-removing repairing compositions prepared in examples 1-3 and comparative examples 1-2 were respectively tried on one group without any treatment. During the trial period, the subjects refrain from using other skin care products and should maintain good working and resting time and eating habits. The grading criteria for acne in patients are shown in table 2 below.
Table 2: grading Standard for acne

Figure BDA0003404858180000091
The results of the trial show that the bacteriostatic, itching-relieving, red-fading and acne-removing repairing and protecting composition prepared in the embodiments 1-3 (particularly in the embodiment 1) can effectively improve the problem of facial acne, the whole part of III-grade acne is improved to II-grade or I-grade, even a small part of acne patients can completely recover to 0-grade, and the effective rate is close to 100%. Patients with acne grade II, more than 90% converted to grade I and even partially returned to grade 0. While the control group and comparative example 2 had less significant relief of acne symptoms than before treatment. Although comparative example 1 had a significant relieving effect on acne symptoms, the relieving effect was significantly inferior to examples 1 to 3 of the present invention.
The foregoing describes preferred embodiments of the present invention, but is not intended to limit the invention thereto. Modifications and variations of the embodiments disclosed herein may be made by those skilled in the art without departing from the scope and spirit of the invention.
