Abstract

Preferably, the acne-removing composition is prepared from 1-10:1-10:1-10:1-10 by mass of radix scutellariae, cortex phellodendri bark, rhizoma coptidis and centella asiatica; further preferred are scutellaria root, phellodendron bark, coptis root and centella asiatica in a mass ratio of 5:3:3: 6.
"centella asiatica" refers to the whole plant of centella asiatica.
The baicalein, baicalin and other components contained in the scutellaria baicalensis roots influence the metabolism of arachidonic acid through various links, and inhibit the generation of prostaglandin E (PGE) and Leukotriene (LT) to different degrees, thereby relieving the expansion of blood vessels by inflammatory mediators, increasing the permeability of blood vessel walls and chemotactic effect of white blood cells; baicalin also has extremely high inhibitory activity on melanocytes, can remarkably inhibit the generation of lipid peroxide, and can relieve melanin pigmentation;
the phellodendron amurense bark has an anti-inflammatory effect, can directly support the muscle bottom to relieve pockmarks, can promote the generation of collagen to repair scars, combines the oxidation resistance of the collagen, and keeps the skin stable; cortex Phellodendri bark containing berberine, phellodendrine, obacunone, etc. has inhibitory effect on Staphylococcus aureus, Staphylococcus albus, Staphylococcus citricola, Diplococcus pneumoniae, Diplococcus meningitidis, Streptococcus viridis, Vibrio cholerae, Bacillus diphtheriae, Bacillus dysenteriae, Pseudomonas aeruginosa, etc.
The rhizoma coptidis root related by the invention contains various active ingredients such as coptisine and jateorhizine. Has remarkable antibacterial effect on gram-positive bacteria such as staphylococcus aureus, streptococcus pneumoniae, beta lactamase-producing streptococcus pneumoniae and the like, gram-negative bacteria such as pseudomonas aeruginosa, escherichia coli, drug-resistant acinetobacter baumannii, neisseria gonorrhoeae and the like, and fungi such as candida albicans, candida albicans and the like, and can balance grease.
The centella asiatica extract is rich in terpenoids, can obviously improve the content of collagen and cell layer fibronectin (fibronectin is widely involved in the processes of cell migration, adhesion, proliferation, hemostasis, skin tissue repair and the like), and can stimulate and promote the generation of type I and type III collagen, activate fibroblast genes and improve and repair the barrier function of the skin; can promote wound healing and scar repair, and reduce inflammatory reaction. In addition, the centella asiatica extract can reduce the production of proinflammatory mediators (IL-1, MMP-1), and improve and repair the barrier function of the skin.
On the other hand, the invention provides a preparation method of the acne-removing composition, which comprises the following steps:
(1) drying radix Scutellariae, cortex Phellodendri, rhizoma Coptidis, and herba Centellae, and pulverizing to obtain plant powder;
(2) adding water, butanediol and 1, 2-pentanediol into the plant powder obtained in the step (1), extracting and filtering.
Preferably, in the step (1), the pulverization is to 10-50 mesh.
Preferably, in the step (2), the mass ratio of the plant powder to the water, the butanediol and the 1, 2-pentanediol is 10-30:30-70:30-70:1-10, and is further preferably 17:50:50: 5.
Preferably, in the step (2), the extraction is performed by a low-temperature high-pressure difference method, and specifically comprises the following steps: regulating temperature to 30-50 deg.C, pressurizing to 50-100MPa, and soaking and extracting under stirring for 30-60 min.
The low-temperature high-pressure difference method is characterized in that loss of thermosensitive components can be avoided under low-temperature conditions, the solvent can more easily permeate into the plant powder under high-pressure conditions, effective components can be rapidly dissolved in the solvent, and tests prove that the extract obtained by the low-temperature high-pressure difference method has higher active components and better effect.
In another aspect, the invention also provides application of the acne-removing composition in cosmetics.
Preferably, in the application, the content of the acne-removing composition in the cosmetics is 2-10 wt%.
Finally, the invention provides oil-control acne-removing gel which comprises the composition.
Preferably, the oil-control acne-removing gel comprises an A phase and a B phase, wherein the A phase comprises water, allantoin, EDTA disodium, polyacrylate cross-linked polymer-6, 1, 2-pentanediol, menthol and the composition; and the phase B comprises hydroxyethyl acrylate/sodium acryloyldimethyl taurate copolymer, cyclopentadimethylsiloxane, cyclohexasiloxane and polydimethylsiloxane.

the invention has the beneficial effects that:
(1) the acne-removing composition provided by the invention has the effects of removing acne marks, removing acne scars, controlling oil and removing acnes. The function purposes, the function routes and the function mechanisms of each component in the composition are different, and the components show the optimal synergistic effect when in proper dosage proportion.
(2) The effective components are plant sources, and have no irritation to skin, mild components and safety.
(3) The low-temperature high-pressure difference method is characterized in that loss of thermosensitive components can be avoided under low-temperature conditions, the solvent can more easily permeate into the plant powder under high-pressure conditions, effective components can be rapidly dissolved in the solvent, and tests prove that the extract obtained by the low-temperature high-pressure difference method has higher active components and better effect.

The present invention will be further explained with reference to specific examples in order to make the technical means, the technical features, the technical objectives and the effects of the present invention easier to understand, but the following examples are only preferred embodiments of the present invention, and not all embodiments of the present invention. Based on the embodiments in the implementation, other embodiments obtained by those skilled in the art without any creative efforts belong to the protection scope of the present invention. In the following examples, unless otherwise specified, all the operations were performed by conventional methods, all the equipments were performed by conventional methods, and the materials of the equipments used in the respective examples were the same. For process parameters or conditions not specifically mentioned, it can be carried out with reference to conventional techniques.
In the following examples, the term "acne removing agent" refers to the composition prepared by the present invention, including scutellaria root extract, phellodendron bark extract, coptis root extract, centella asiatica extract, and is abbreviated as "yellow" because it contains scutellaria root extract, phellodendron bark extract, and coptis root extract.
Example 1
A preparation method of a three-yellow acne removing agent comprises the following steps:
(1) drying radix scutellariae, cortex phellodendri bark, rhizoma coptidis root and centella asiatica, wherein the mass ratio of the radix scutellariae, the cortex phellodendri bark, the rhizoma coptidis root and the centella asiatica is 1:10:1: 1; pulverizing to 50 mesh to obtain plant powder;
(2) adding water, butanediol and 1, 2-pentanediol into plant powder, wherein the mass ratio of the plant powder to the water, the butanediol and the 1, 2-pentanediol is 17:50:50: 5; mixing, putting into extraction kettle, adjusting temperature to 30 deg.C, pressurizing to 50MPa, soaking and extracting under stirring for 60 min, centrifuging, collecting supernatant, and filtering with 1000 mesh filter cloth.

Test example 1 anti-Propionibacterium acnes Effect
Propionibacterium acnes is the primary pathogen that causes acne. In the antibacterial experiment, propionibacterium acnes is used as a bacterial model, and in order to verify the antibacterial effect of the composition, the compositions in the embodiments 1 to 3 and the comparative examples 1 to 7 of the invention are subjected to antibacterial performance measurement by referring to GB15979-2002 (hygienic Standard for Disposable sanitary articles) appendix C4 'test method for antibacterial performance of dissolution antibacterial (inhibiting) products':
1) test samples: the compositions described in examples 1-3 and comparative examples 1-7 were diluted to 2% dilutions, respectively.
2) Preparation of bacterial suspension: a24-hour culture of the propionibacterium acnes is washed by 0.03mol/L phosphate buffer solution to prepare 104-106CFU/mL bacterial suspension.
3) Respectively taking 1g of test sample, adding 0.1mL of the bacterial suspension, uniformly coating and mixing, starting timing, adding 9mL of 0.03mol/L phosphate buffer solution after 20min, fully and uniformly mixing, taking 3 dilutions, respectively taking 1mL of the dilutions, placing the dilutions in two parallel plates, adding corresponding culture medium, and culturing for 24 hours at 36 +/-1 ℃ under anaerobic condition to count bacterial colonies. The above experiment was repeated 3 times, and the average was taken and a blank control group was set.
The calculation formula of the bacteriostatic rate is as follows: and X is (A-B)/A X100%, wherein X is the bacteriostasis rate, A is the average recovered colony of a blank control sample, and B is the average recovered colony of a test sample.
The evaluation criteria for efficacy were: (1) the effect is obvious when the bacteriostasis rate is more than or equal to 90 percent; (2) the bacteriostatic rate is 40-90% and the effect is normal; (3) the bacteriostasis rate is less than 40 percent, which indicates no effect. The results are shown in the following table.

As can be seen from Table 1, the compositions of examples 1 to 3 have an obvious bacteriostatic effect on Propionibacterium acnes, and the compositions of comparative examples 1 to 7 have a general bacteriostatic effect; experiments show that the scutellaria baicalensis roots, the phellodendron amurense barks, the coptis roots and the centella asiatica can obviously inhibit the proliferation of propionibacterium acnes when being used in a combined way according to a proper proportion, and the inhibition effect is better than that when the components are used independently; can effectively prevent or improve the acne symptom of the skin.

Test example 2 oil control acne removal test
The oil-controlling and acne-removing efficacy of the oil-controlling and acne-removing condensation prepared in examples 4-6, comparative examples 8-13 and blank control group was evaluated. The blank control group was different from example 6 in that it did not contain the acne removing agent.
The oil control effect tester is a skin oil tester. The working principle of the skin cuticle oil absorption rubber belt is that a special rubber belt is adopted, oil on the surface of the skin cuticle can be absorbed quickly, the transmittance of the rubber belt can be changed by the oil, and the oil content of the skin cuticle can be obtained by measuring the transmittance of the rubber belt before and after use through a photometer arranged in an instrument.
The test method comprises the following steps: 100 volunteers with oily skin and whelk are recruited and divided into 10 groups of 10 persons each, wherein one group uses the oil-controlling and acne-removing gel of a blank control group, three groups use the oil-controlling and acne-removing gels prepared in examples 4-6 respectively, and six groups use the oil-controlling and acne-removing gels prepared in comparative examples 8-13 respectively, and the oil-controlling and acne-removing gels are used once in the morning and evening every day with the use amount of 0.2 g/time, smeared on the face and lightly massaged until being absorbed. The oil content of the forehead of the volunteer was measured every other week with a skin oil tester, each part was tested 3 times, the average was taken, and the specific test record is shown in table 2. The acne removing condition is recorded by taking pictures every other week, the acne number is reduced by more than 1/3 and the judgment is effective by comparing the picture with the picture before using the product, and the specific test records are shown in the table.

As can be seen from tables 11 and 12, the oil-controlling acne-removing gel prepared by the embodiment of the invention has good oil-controlling acne-removing effect; and the combination of the radix scutellariae, the bark of amur corktree, the root of coptis and the centella asiatica according to a proper proportion has better acne removing effect than that of single use.

The oil-controlling and acne-removing gel prepared in examples 4-6 and comparative examples 8-13, the oil-controlling and acne-removing gel prepared in a blank control group and the commercial acne-removing products prepared in comparative examples 14-15 were evaluated for the efficacy of removing acne marks and scars, wherein the blank control group is different from the example 6 in that no acne removing agent is contained.
The test method comprises the following steps: 120 volunteers with newly generated acne marks and scars (generated within one month) are gathered and divided into 10 groups, 10 persons are gathered, one group of the groups uses the oil-control acne-removing gel of a blank control group, three groups of the groups use the oil-control acne-removing gel prepared in examples 4-6 respectively, six groups of the groups use the oil-control acne-removing gel prepared in comparative examples 8-13 respectively, two groups of the groups use the commercially available acne-removing products of comparative examples 14-15 respectively, and the groups are used once a day in the morning and evening with the use amount of 0.2 g/time, smeared on the face and gently massaged until the gel is absorbed. The conditions of the acne marks and the acne scars after the volunteers use the product for one month are compared, and specific test records are shown in the table.

From 13 and 14, the acne mark removing and conditioning essence (containing salicylic acid) of the comparative example 14 has a good acne mark removing effect and a poor acne scar removing effect; the centella asiatica acne cream of the comparative example 15 has good scar removing effect and poor acne mark removing effect; the oil-control acne-removing gel prepared by the embodiment of the invention has good acne mark-removing and scar-removing effects, and has better acne-removing effect than that of each component used independently.
Test example 4 irritation test (Patch test)
Test samples: oil-controlling acne-removing gel prepared in examples 4-6 and commercially available acne-removing products of comparative examples 14-15.
Subject: selecting 30 volunteers (the same batch of subjects is used for each test sample);
the test method comprises the following steps: a closed test is adopted, 0.02g of test sample is placed in a patch applicator, a hypoallergenic adhesive tape is applied to the arm curve side of a tested subject, the test area is at least 5 x 5cm, the tested subject straightens the arm when applying, the test sample is applied from the lower part to the upper part, the adhesive tape is lightly pressed by the palm to discharge air, the test sample is tightly attached to the skin for 24 hours, and the blank control group is a test sample which is not used.
And (4) observing results: the patch was removed 30min and then interpreted according to the interpretation criteria described below, and the statistical test results are shown in the table below.
Interpretation criteria: (one) negative reaction; (+ -suspicious reaction: only mild erythema; (+) weak positive: erythema, infiltrates, and possibly small amounts of papules; strong positive (++): erythema, infiltrates, papules, blisters; (+++) very positive: erythema, infiltrates, papules, blisters, bulla.

As can be seen from the table above, the oil-controlling and acne-removing condensation prepared in the embodiments 4-6 of the invention has low skin sensitization, and no positive reaction is detected in 30 subjects, which indicates that the composition has no stimulation to skin, mild components and safety.
In conclusion, compared with the comparative examples, the three-yellow acne removing agent composition obtained in examples 1-3 has more remarkable effects of reducing skin grease secretion and better acne removing effect, and especially, the effect of example 3 is the most ideal; the experimental result shows that the composition can embody the best oil-controlling and acne-removing efficacy by matching and combining the components in a proper proportion, and has the best acne mark-removing and scar-removing efficacy. The analysis proves that the efficacy of the composition is due to the fact that the action purposes, the action routes and the mechanisms of all the components in the composition are different, and the components show the best synergistic effect when being used in the right dosage proportion; and tests prove that the composition has no stimulation to skin, mild components and safety.

Claims (10)
1. An acne-removing composition with the effects of removing acne marks and acne scars is characterized by comprising a scutellaria baicalensis root extract, a phellodendron amurense bark extract, a coptis chinensis root extract and a centella asiatica extract.
2. The acne-removing composition according to claim 1, wherein the raw materials of the composition are scutellaria baicalensis root, phellodendron amurense bark, coptis chinensis root and centella asiatica in a mass ratio of 1-10:1-10: 1-10.
3. The acne treatment composition according to claim 2, wherein the raw materials of the composition are scutellaria baicalensis root, phellodendron amurense bark, coptis chinensis root and centella asiatica in a mass ratio of 5:3:3: 6.
4. A method for preparing the acne-removing composition according to any one of claims 1 to 3, which comprises the following steps:
(1) drying radix Scutellariae, cortex Phellodendri, rhizoma Coptidis, and herba Centellae, and pulverizing to obtain plant powder;
(2) adding water, butanediol and 1, 2-pentanediol into the plant powder obtained in the step (1), extracting and filtering.
5. The preparation method according to claim 4, wherein in the step (2), the mass ratio of the plant powder to the water, the butanediol and the 1, 2-pentanediol is 10-30:30-70:30-70: 1-10.
6. The preparation method according to claim 5, wherein in the step (2), the mass ratio of the plant powder to the water, the butanediol and the 1, 2-pentanediol is 17:50:50: 5.
7. The preparation method according to claim 4, wherein in the step (2), the extraction is performed by a low-temperature high-pressure difference method, and specifically comprises the following steps: regulating temperature to 30-50 deg.C, pressurizing to 50-100MPa, and soaking and extracting under stirring for 30-60 min.

9. An oil-controlling acne-removing gel, which is characterized by comprising the acne-removing composition as described in any one of claims 1 to 3 or prepared by the preparation method as described in any one of claims 4 to 7.

The oil-controlling acne-removing gel according to claim 9, characterized by comprising phase a and phase B, wherein phase a comprises water, allantoin, disodium EDTA, polyacrylate crosspolymer-6, 1, 2-pentanediol, menthol and the acne-removing composition; the phase B comprises hydroxyethyl acrylate/sodium acryloyldimethyl taurate copolymer, cyclopentadimethylsiloxane, cyclohexasiloxane and polydimethylsiloxane;
