However, the actual efficacy of the hand cream is not mentioned in this document, and the effect of its use is not known. Chinese patent document CN105920325A discloses a traditional Chinese medicine moisturizing hand cream and a preparation method thereof, and the traditional Chinese medicine moisturizing hand cream comprises 3-9 parts of Chinese violet, 5-15 parts of mint, 5-10 parts of Chinese angelica, 8-15 parts of honeysuckle, 8-15 parts of wild chrysanthemum flower, 8-15 parts of dandelion, 5-10 parts of trogopterus dung, 2-8 parts of angelica dahurica, 2-6 parts of cortex moutan, 1-5 parts of liquorice, 10-18 parts of scutellaria baicalensis, 5-10 parts of kudzu root, 2-8 parts of pseudo-ginseng, 1-5 parts of hairyvein agrimony, 2-6 parts of schisandra chinensis and 1-3 parts of walnut kernel. The hand cream can prevent skin roughness, but the whitening and moisturizing effects are not disclosed. Therefore, in order to overcome the defects in the prior art, a traditional Chinese medicine hand cream with good moistening, whitening and rough skin improving functions needs to be developed, but the traditional Chinese medicine hand cream for moisturizing is not reported at present.

Example 4 hand cream irritation and allergy test
Taking 10 mice, and taking 16cm at back2The area was cut to remove hair, divided into two areas, one of which was applied with the hand cream prepared in examples 1-3 once a day, and the skin was wiped off the next day before application, and the adjacent area was observed as a control for 2 months. The results show that the skin in the test area and the control skin have no obvious difference in the experimental period, no irritation and anaphylactic reaction exist, and no abnormal reaction exists in mice, so that the hand cream is safe and non-toxic and has no irritation to the skin.
Example 5 determination of hand cream Properties
1. Measurement of wetting ability of hand cream
The determination was carried out by the pour point method, taking a 100mL beaker, weighing 20.0g of talc, gradually adding the hand cream of example 1, comparative example 2, mixing until the mixture formed a pourable homogeneous fluid, at which point the volume added dropwise was the pour point, and converting the base of the volume multiplied by 5 to the pour point value, the results are shown in table 1. From the analysis of the measurement results, compared with comparative example 1, comparative example 2 and the hand cream of example 1, the hand cream of example 1 has a lower pour point, which shows that the traditional Chinese medicine moisturizing hand cream of the invention has better wettability.
Table 1 comparison of wettability for different formulations
Item | Comparative example 1 | Comparative example 2 | Example 1 |
Flow point/mL | 62 | 42 | 37 |
2. Measurement of moisturizing ability of hand cream
The moisturizing effect of the backs of the hands smeared with the hand cream is measured by using a skin moisture in-vitro measuring instrument, and the specific method comprises the following steps: 0.5mL of the hand cream in comparative example 1, comparative example 2 and example 1 was applied to the pre-showered hands, a moisture measurement tank was placed in the middle of the back of the hand for 15min to measure the change in the relative rate of water vapor evaporation from the blank control within 10min, and the results showed that the relative rate of evaporation of the hand cream in the experimental group was 30% and 42% lower than that in the control group 1 and 2, respectively, indicating that the hand cream of the present invention has higher moisturizing ability than the hand cream in the control group.

2g of the hand cream prepared in comparative example 1, comparative example 2 and example 1 was taken, and 40ml of distilled water was added thereto and diluted for use. The B16 melanocytes were inoculated into a 96-well plate at 50. mu.l per well, and after culturing in an incubator for 24 hours, a hand cream diluent was added. After 5 days of drug action, the supernatant was discarded and washed twice with poly (butylene succinate) (PBS) pH 6.8, 40. mu.l of 1% (volume fraction) Triton X-100 in each well. Carrying out ultrasonication in ice bath, adding 10 μ L of 10 mmol/L-dopa (L-dopa) into each well, incubating at 37 deg.C for 90min, carrying out color comparison at 475nm, adjusting blank well to zero, and measuring absorbance value of each well.
Inhibition% [ (% 1-mean absorbance for drug group ÷ mean absorbance for control group) × 100%.

Taking comparative example 1, comparative example 2 and the example respectively1g of the hand cream prepared in example 1 was diluted with 40ml of distilled water. B16 melanoma cells from mice at 1x105The cells were inoculated in a culture dish at a density of one/ml and at 37 ℃ in 5% CO2Culturing for 1.5 days. Discarding the supernatant, adding hand cream diluent, discarding the supernatant after 3d of drug action, washing with PBS, digesting cells, and preventing over-digestion. After the mixture was blown and mixed, 0.5ml of each concentration was taken out and counted. Centrifuging the rest cell suspension at 2500r/min for 5min, discarding supernatant, adding NaOH solution into the precipitate, heating to dissolve melanin, and measuring absorbance value on enzyme-linked immunosorbent assay detector with 490nm wavelength.
Melanin synthesis inhibition [% 1- (drug well absorbance value ÷ drug well cell density) ÷ (control well absorbance value ÷ control well cell density) ] × 100%.
