According to the technical scheme provided by the invention, the plant composition with the acne preventing and removing effect and the nanogold with strong oxidation resistance are utilized to realize the synergistic effect, and the allergy relieving composition is combined to provide the mild and efficient preparation method of the acne preventing and removing essence, so that the defects of the acne preventing and removing effect and the mildness of the plant composition and the nanogold which are used independently are overcome. The present invention provides a composition comprising: the anti-acne and anti-acne tremella fuciformis tea comprises a tremella fuciformis fruiting body extract, an anti-acne and anti-acne component, a allergy-relieving component and nanogold. The composition has good antioxidant effect and certain antibacterial effect under proper proportion, so that the composition is mainly developed into a formula suitable for people who easily grow the acne from the antioxidant aspect, and solves the problems of primary acne and mature acne from the aspect of the protection of inhibiting further oxidation of fatty acid. Moreover, the components in the essence are beneficial to the exertion of the effect of the composition, the whole formula is very mild and has no irritation, and the application range of the essence is expanded.
Detailed Description
The invention provides a composition and application thereof in preparing an acne-removing product, and a person skilled in the art can use the contents to reference the contents and appropriately improve process parameters. It is expressly intended that all such similar substitutes and modifications which would be obvious to one skilled in the art are deemed to be included in the invention. While the methods and applications of this invention have been described in terms of preferred embodiments, it will be apparent to those of ordinary skill in the art that variations and modifications in the methods and applications described herein, as well as other suitable variations and combinations, may be made to implement and use the techniques of this invention without departing from the spirit and scope of the invention.
The test materials adopted by the invention are all common commercial products and can be purchased in the market. Wherein:
the cinnamon bark extract of Ceylon blue mainly contains flavone and tannin, and is prepared through crushing and sieving Ceylon blue bark, defatting, ultrasonic assisted alcohol extraction, filtering concentration, decolorizing, alcohol removal, purification and other steps.
The schisandra fruit extract has schisandra fruit extract with schisandra essence as main component and is prepared through crushing schisandra fruit, sieving, reflux extraction with water and alcohol, filtering, concentration, purification and other steps.
The extract of the root of Sophora flavescens ait contains matrine, matrine and the like as main ingredients, and is extracted by crushing and sieving the root of Sophora flavescens ait, extracting by water-alcohol reflux, filtering, concentrating, purifying and the like.
The ginseng root extract contains ginsenoside as main component, and is extracted through crushing and sieving ginseng root, reflux extraction with water and alcohol, filtering, concentration, purification, etc.
The echinacea purpurea extract mainly contains isobutyramide, echinacoside and the like, and is extracted by crushing and sieving echinacea purpurea, performing hydroalcoholic ultrasonic extraction, filtering, concentrating, purifying and the like.
The centella asiatica extract mainly contains asiaticoside, and is extracted by crushing and sieving centella asiatica, performing ultrasonic-assisted alcohol extraction, filtering and concentrating, decoloring, dealcoholizing, purifying and the like.
The main components of the giant knotweed rhizome extract comprise resveratrol, emodin and the like, and the giant knotweed rhizome extract is extracted by crushing and sieving the giant knotweed rhizome, extracting by petroleum ether, removing the petroleum ether, extracting by ultrasonic-assisted alcohol extraction, filtering and concentrating, decoloring, removing the alcohol, purifying and the like.
The extract of radix Scutellariae contains flavonoids such as baicalin and baicalein as main ingredients, and is prepared by pulverizing radix Scutellariae, sieving, ultrasonic extracting with water, precipitating at certain temperature for a certain time, filtering, washing with water, and drying.
The purslane extract mainly contains plant polysaccharide and vitamins, and is prepared by carrying out water and hydrolase enzymolysis extraction, alcohol extraction, filtration, concentration and the like on purslane.
The tea extract is rich in tea polyphenol, and is extracted by crushing tea, water extraction, alcohol extraction, enzymolysis, ultrafiltration, concentration, drying and the like.
The glycyrrhiza glabra root extract mainly comprises licoflavone, glycyrrhizin, glycyrrhiza polysaccharide and the like, and is extracted by crushing and sieving glycyrrhiza glabra root, performing ultrasonic-assisted alcohol extraction, filtering, concentrating, purifying and the like.
The flos Matricariae Chamomillae extract contains flavonoids, amino acids, chlorogenic acid, etc. as main ingredients, and is prepared by pulverizing flos Matricariae Chamomillae, sieving, extracting with ultrasound-assisted alcohol, filtering, concentrating, and purifying.
The rosemary leaf extract mainly contains terpene phenols and the like, and is extracted by crushing and sieving the rosemary leaves, performing ultrasonic-assisted alcohol extraction, filtering, concentrating, purifying and the like.
The magnolia flower extract contains flavonoid compounds as main components, and is extracted by crushing and sieving magnolia flower, ultrasonic water extraction, heat preservation for a certain time at a certain temperature, precipitation, filtration, water washing, drying and the like.
The snow lotus herb extract mainly contains flavonoid, polysaccharide and the like, and is extracted by carrying out ultrasonic-assisted alcohol extraction, filtration, concentration, purification and the like on the snow lotus herb.
The nanogold is composed of gold, tremella polysaccharide, water and 1, 2-hexanediol and is purchased from the Welcho chemical company Limited, Guangzhou.
In some embodiments, each 100g of the mild acne-preventing and acne-removing essence disclosed by the invention is prepared from the following components in parts by mass: 3g of butanediol, 8g of propylene glycol, 1g of glycerol polymethacrylate/propylene glycol/PVM/MA copolymer, 0.01g of tremella fruiting body extract, 1.0g of acne-preventing and acne-removing plant composition, 0.5g of allergy-relieving plant composition, 0.01g of nanogold, 0.4g of thickening agent, 0.03g of xanthan gum, 0.7g of p-hydroxyacetophenone, 0.2g of 1, 2-hexanediol, 0.4g of PEG-40 hydrogenated castor oil, 0.05g of essence, 0.4g of triethanolamine and the balance of water.
In some embodiments, each 100g of the mild acne-preventing and acne-removing essence disclosed by the invention is prepared from the following components in parts by mass: 7g of butanediol, 4g of propylene glycol, 4g of glycerol polymethacrylate/propylene glycol/PVM/MA copolymer, 0.05g of tremella fruiting body extract, 0.5g of acne-preventing and acne-removing plant composition, 0.1g of allergy-relieving plant composition, 0.06g of nanogold, 0.1g of thickening agent, 0.1g of xanthan gum, 0.2g of p-hydroxyacetophenone, 0.7g of 1, 2-hexanediol, 0.1g of PEG-40 hydrogenated castor oil, 0.01g of essence, 0.1g of triethanolamine and the balance of water.

The 4 compositions are sequentially marked as groups 1-4, and in order to prove the effect of the mild anti-acne and anti-acne essence provided by the invention, the mild anti-acne and anti-acne essence prepared by the invention is subjected to efficacy verification, and meanwhile, groups 5-10 are set as positive control samples. Taking samples of the groups 1-10 to perform propionibacterium acnes bacteriostasis tests (coating method), DPPH free radical clearance and ABTS free radical clearance in-vitro oxidation resistance tests and consumer use effect scoring tests, wherein:
group 5: the other components are the same as the component 3 except that 0.05g of the anti-acne and anti-acne plant composition is used for replacing 0.05g of the nanogold.
Group 6: the other components were the same as those of the component 3 except that the amount of the plant composition for preventing and removing acne was increased to 2 g.
Group 7: the other components are the same as the component 3, except that the anti-acne and anti-acne plant composition only contains schisandra chinensis fruit extract, sophora flavescens root extract, ginseng root extract and echinacea purpurea extract.
Group 8: the other components are the same as the component 3, except that the anti-acne and anti-acne plant composition only contains the extracts of the bark of the Cinnamomum zeylanicum, the root of the Sophora flavescens ait, the root of the ginseng and the Echinacea purpurea.
Group 9: the other components are the same as the component 3, except that the anti-acne and anti-acne plant composition only contains the extract of the bark of the Cinnamomum zeylanicum, the extract of the fruit of Chinese magnoliavine, the extract of the root of ginseng and the extract of the echinacea purpurea.
Group 10: the other components were the same as in group 3 except that the soothing botanical composition was not included.
The invention is further illustrated by the following examples:
example 1
1 part by mass of cinnamon bark extract of Ceylon, 2 parts by mass of fruit extract of Schisandra chinensis, 2 parts by mass of root extract of Sophora flavescens, 1 part by mass of root extract of Panax ginseng, and 1 part by mass of Echinacea purpurea extract are mixed to prepare the anti-acne and anti-acne plant component.
Example 2
Mixing herba Centellae extract, rhizoma Polygoni Cuspidati extract, Scutellariae radix extract, herba Portulacae extract, folium Camelliae sinensis extract, radix Glycytthizae extract, flos Matricariae Chamomillae extract, herba Rosmarini officinalis extract, flos Magnoliae extract, and herba Saussureae Involueratae extract at a certain mass ratio to obtain the desired component.
Comparative example 1
2 parts by mass of shizandra berry extract, 2 parts by mass of kushen root extract, 1 part by mass of ginseng root extract and 1 part by mass of echinacea purpurea extract are mixed to prepare the acne-preventing and acne-removing plant component.
Comparative example 2
1 part by mass of cinnamomum zeylanicum bark extract, 2 parts by mass of sophora flavescens root extract, 1 part by mass of ginseng root extract and 1 part by mass of echinacea purpurea extract are mixed to prepare the acne-preventing and acne-removing plant component.
Comparative example 3
1 part by mass of cinnamon bark extract of Ceylon, 2 parts by mass of fruit extract of Schisandra chinensis, 1 part by mass of ginseng root extract and 1 part by mass of echinacea purpurea extract are mixed to prepare the anti-acne and anti-acne plant component.

The preparation method comprises the following steps:
1) weighing each component in proportion;
2) uniformly stirring and dispersing xanthan gum and tremella sporophore extract by using a certain amount of propylene glycol, and marking as prefabrication 1; adding the essence into PEG-40 hydrogenated castor oil, stirring and dissolving until the essence is transparent, and marking as a prefabricated product 2; adding p-hydroxyacetophenone into a proper amount of propylene glycol, heating to 60-70 ℃, stirring and dissolving until the mixture is transparent, and marking as a prefabricated product 3; triethanolamine was mixed with the appropriate amount of water and labeled preform 4.
3) Adding the rest water into an aqueous pot, slowly scattering the thickening agent on the water, soaking for 5-8 min, starting a stirrer, stirring for 2-4 min at 30-35 Hz, adding butanediol, the rest propylene glycol, the glycerol polymethacrylate/propylene glycol/PVM/MA copolymer, prefabricating for 1, heating to 85-90 ℃, preserving heat and stirring for 15-20 min;
4) cooling to 45-50 ℃, adding 1, 2-hexanediol, the acne-preventing and acne-removing plant composition, the allergy-relieving plant composition, the nanogold, the prefabricated product 2, the prefabricated product 3 and the prefabricated product 4, keeping the vacuum of-0.04 to-0.08 MPa, stirring for 10-15 min, cooling, sampling, detecting, and discharging after the product is qualified.
Effect verification
In order to prove the effect of the mild acne-preventing and acne-removing essence provided by the invention, the essences of groups 1-10 in example 3 are used for carrying out propionibacterium acnes bacteriostasis test (coating method), DPPH free radical clearance rate and ABTS free radical clearance rate in-vitro oxidation resistance test and consumer use effect scoring test:
1. verification method
1.1, Propionibacterium acnes bacteriostasis test (coating method):
firstly, preparing a double-layer plate (the lower layer is 10-15 mL of culture medium, the upper layer is 5mL of culture medium containing propionibacterium acnes) containing propionibacterium acnes (ATCC11827), wherein the bacterial content of the plate is 104~105CFU/plate, after the plate is cooled, accurately weighing 0.5g of sample, uniformly coating the sample on the plate containing bacteria by using a sterile cotton swab, culturing for 24 hours under the anaerobic condition of 36 +/-1 ℃, and then observing the growth condition of the bacterial colony in the plate. The judgment standard of the acne removing efficacy result of the sample propionibacterium acnes bacteriostasis test is shown in table 2:

1.2 DPPH free radical clearance rate: a certain amount of the material was mixed with a proper amount of DPPH solution, left in the dark for 30min, the absorbance at 517nm was measured, the radical scavenging activity was calculated as radical scavenging rate (%) against a blank control, and the test results are shown in Table 3.
1.3 ABTS free radical clearance: mixing and shaking 7mM ABTS aqueous solution and 2.45mM potassium persulfate solution according to the volume ratio of 1:1, standing at room temperature in a dark place for 16h, and diluting with absolute ethyl alcohol until the light absorption value at the wavelength of 734nm is less than 0.7 to obtain ABTS determination solution; a certain amount of the test material was taken, added to the ABTS measurement solution, left to stand for 5min, and the absorbance was measured at 734nm, and the radical scavenging activity was calculated as radical scavenging rate (%) with the test results shown in Table 3.
1.4, a consumer use effect scoring experiment test method:
selecting a subject with the age of 16-32 years, wherein the number of the subjects is 300, the skin characteristics are oily skin and mild acne skin, randomly dividing the skin into 6 groups, dividing the skin into left and right side faces, using a control sample (application group 4-10) on the left side, using a test sample (group 1-3) on the right side, respectively 1 time in the morning and evening (the specific use condition is shown in table 4), and evaluating the use feelings of the subjects for preventing acne (mainly reflecting the capacity of preventing new acne and removing nascent acne), removing acne (removing mature acne), stimulating performance and the like after 14 days, wherein the test results are shown in table 5, wherein the score of the test results is from 0 to 10, and the higher the score is, and the more obvious effect is achieved.
2. Verification of effects
The judgment results of the acne removing efficacy of the sample propionibacterium acnes bacteriostasis test and the results of DPPH and ABTS free radical clearance rate test are shown in Table 3:

The results show that the bacteriostatic effects of groups 1-10 are all low, and the bacteriostatic effect of group 6 is moderate. However, the radical scavenging effect was strong in each group, and the groups 6, 1, 3, 2, 4, and 10 were in this order. The inhibition of propionibacterium acnes and the removal of free radicals are two important conditions for producing an anti-acne effect, and the two conditions complement each other. The composition disclosed by the invention has a low antibacterial effect, but can generate an acne removing effect through a good effect of removing free radicals.
Therefore, the essence has the anti-acne effect with different degrees.

As can be seen from Table 5, each group had certain acne removing and acne preventing effects.
Wherein the groups 1-4, 6 and 10 have higher scores in the aspects of the acne prevention effect and the acne removal effect. This indicates that these groups of samples can achieve good anti-acne and anti-acne effects by scavenging free radicals. The groups 5 and 7 to 9 had low scores in both the anti-acne effect and the anti-acne effect, and could not exert a good anti-acne effect, which was consistent with the effect of scavenging free radicals described in table 3.
The samples of groups 6 and 10 were inferior in irritation to the other samples, and the subjects considered that the samples were somewhat irritating and not mild enough.
In combination with the results, the anti-acne and anti-acne component provided by the invention can realize good anti-acne and anti-acne effects through removing free radicals and matching with the nano-gold and the allergy-relieving component,
comparing group 3 with groups 7 to 9, it can be seen that the anti-acne and anti-acne component provided in example 1 with the same amount has better effect of scavenging free radicals and better effect of preventing acne and removing acnes compared with comparative examples 1 to 3 which lack some components. The components for preventing and removing the acnes are reasonable in composition and all the components are absent. However, excessive addition of this component can cause irritation and is not conducive to use by people with sensitive skin. Therefore, the amount of the anti-acne and anti-acne component added to every 100g of essence is 0.5-1.0 g.
Comparing the samples of groups 1-6, it can be seen that the effect of the acne removing component of the invention can be further improved by adding nanogold, and the effect of the sample added with nanogold is better than that of the group 5 without adding nanogold in the aspects of oxidation resistance, acne removing and acne preventing. In addition, in a certain concentration range, the content of the nano-gold is increased, so that the effects of resisting oxidation, preventing acnes and removing acnes can be improved, but because the cost of the nano-gold raw material is high, the addition amount of the nano-gold in every 100g of essence is more appropriate from the comprehensive consideration of ensuring the using effect, the mildness and the cost.
Comparing group 10 with group 3, it can be seen that group 3 with the addition of the soothing component does not exhibit irritation at the same amount of the anti-acne component and nanogold, while group 10 receives feedback of higher irritation, and the soothing component provided by the present invention can reduce skin sensitivity.
The essential liquid of the group 1-3 is considered in all aspects, and good acne prevention and removal effects are obtained by removing free radicals and reducing sensitivity, so that the reasonable components and the appropriate concentration are shown. And further research shows that the matrix components of the essence in the groups 1-3 have good protection effects on the stability of the allergy-relieving component and the acne-preventing and acne-removing component.
The above is only a preferred embodiment of the present invention, and it should be noted that, for those skilled in the art, a plurality of modifications and embellishments can be made without departing from the principle of the present invention, and these modifications and embellishments should also be regarded as the protection scope of the present invention.

the acne-preventing and acne-removing component consists of a cinnamomum zeylanicum bark extract, a schisandra chinensis fruit extract, a sophora flavescens root extract, a ginseng root extract and an echinacea purpurea extract; wherein the mass ratio of the cinnamomum zeylanicum bark extract to the schisandra chinensis fruit extract to the sophora flavescens root extract to the ginseng root extract to the echinacea purpurea extract is 1:2:2:1: 1;
