When the leontopodium alpinum extract is extracted by using ethanol with the volume fraction of 70-90%, the prepared leontopodium alpinum extract has a stronger acne removing effect by matching with other three components.
The specific value of 70-90% can be selected from 70%, 73%, 76%, 79%, 80%, 83%, 86%, 89%, 90%, etc.

In a third aspect, the invention provides a preparation with acne removing efficacy, and the preparation raw materials of the preparation comprise the acne removing composition according to the first aspect.
The preparation contains the composition of the first aspect, and has excellent effects of inhibiting bacteria, resisting oxidation, cleaning pores, promoting skin oil balance, relieving swelling and pain, and promoting acne mark repair.

Compared with the prior art, the invention has the following beneficial effects:
1. the narcissus bulb extract, the purslane fermented extract, the tea tree essential oil and the alpine edelweiss extract are creatively applied to the treatment of skin acne and comedo, and play a role mainly from 3 aspects: (1) softening stratum corneum and dredging pores; (2) controlling oil and cleaning oil secreted by the skin; and (3) inhibiting the growth of skin bacteria. The four components of the narcissus bulb extract, the purslane fermented extract, the tea tree essential oil and the alpine edelweiss extract supplement each other and have synergistic interaction, the composition prepared from the four components has stronger antibacterial and antioxidant effects, and further, a preparation product containing the four components has better acne-removing and swelling-reducing effects.
2. According to the preparation method of the acne-removing composition, the acne-removing composition can be prepared only by mixing the purslane fermented extract, the alpine edelweiss extract, the narcissus bulb extract and the tea tree essential oil, and the method is simple to operate and suitable for industrial amplification.
3. The preparation with acne removing effect contains the composition in the first aspect, and has excellent effects of inhibiting bacteria, resisting oxidation, cleaning pores, promoting skin oil balance, relieving swelling and pain and promoting acne mark repair.

The narcissus bulb extracts referred to in the following examples, comparative examples and test examples are products available from siennakang biotech ltd; example 7 the purslane extract was obtained from Canuo Biotech limited, guangzhou, model number purslane fermentation filtrate Hoshea TM Products of Portulaca; the tea tree essential oil is a product with industrial grade, which is purchased from Pan-Hei (Shanghai) International trade company; cinnamomum zeylanicum extract is a product available from Walt Laisi Biotech, lanzhou; the Salicis Babylonicae extract is obtained from Beijing Lingbao scientific and technological Co; rosa multiflora water is a product from Hubeixin Red chemical industry, chlorella vulgaris extract is a product from Shanyang Lianfeng Biotechnology, inc., piper methysticum extract is a product from Gansu Probiotics, inc., encapsulated salicylic acid is a product from Guangzhou Tinglan Biotechnology, inc., and dipeptide-15 is a product from Mecanin.
The examples do not specify particular techniques or conditions, and are to be construed in accordance with the description of the art in the literature or with the specification of the product. The reagents or apparatus used are conventional products commercially available from normal sources, not indicated by the manufacturer.
Preparation example 1
The preparation example provides a purslane fermentation extract, and the preparation method comprises the following steps:
(1) Cleaning herba Portulacae, removing root, oven drying, pulverizing, and sieving with 100 mesh sieve. Adding 1% of Thermus thermophilus culture medium (0.4% of yeast powder, 0.8% of polypeptone, 0.2% of sodium chloride, 2% of agar, and the balance of water, and sterilizing for later use).
(2) Inoculating Thermus thermophilus (ATCC 27634) into 2L Thermus thermophilus culture medium at 0.1%, culturing at 75 deg.C and 150rpm for 12 hr to obtain Thermus thermophilus activation solution; inoculating the Thermus thermophilus activation solution into the Thermus thermophilus culture medium obtained in the step (1) according to the inoculation amount of 1%, culturing at 75 ℃,150rpm for 20h, collecting fermentation liquor after fermentation is stopped, performing ultrasonic wall breaking treatment (working voltage 1800W) by using an ultrasonic crusher of Kunmansan Xiaomei model CB-26A, centrifuging at 3000rpm for 15min, taking lower layer cytolytic solution, and performing ultraviolet sterilization for 30min to obtain the purslane fermented extract.
Preparation example 2
The preparation example provides a leontopodium japonicum extract, and the preparation method comprises the following steps: chopping the edelweiss, soaking in 80% ethanol water solution for 30h, reflux-extracting for 3 times, 2h each time, concentrating the ethanol extract under reduced pressure, drying at 80 deg.C, and sieving with 80 mesh sieve to obtain the extract.
Preparation example 3
The preparation example provides a leontopodium alpinum extract, and the preparation method comprises the following steps: chopping the edelweiss, soaking in 65% ethanol water solution for 30h, reflux-extracting for 3 times, each for 2h, concentrating the ethanol extract under reduced pressure, drying at 80 deg.C, and sieving with 80 mesh sieve to obtain the extract.
Preparation example 4
The preparation example provides a leontopodium alpinum extract, and the preparation method comprises the following steps: chopping the leontopodium alpinum, soaking in 95% ethanol water solution for 30h, reflux-extracting for 3 times, each time for 2h, concentrating the ethanol extract under reduced pressure, drying at 80 deg.C, and sieving with 80 mesh sieve to obtain the extract.

The formulations prepared in examples 1 to 7 and comparative examples 1 to 9 were evaluated for acne removal effect by the following specific evaluation methods: (1) 160 volunteers with mild/moderate acne skin between the ages of 18 and 45, with hyperpigmentation, persistent erythema or depressed scarring, were selected to maintain regular life during use. Volunteers were randomly divided into 16 groups of 10 persons each. (2) The face was divided into two, the left side was the test area and the right side was the comparison area, the left side was the test sample and the right side was wiped with clear water for 14 consecutive days. examples 5 and 6 show that the leontopodium anisopliae extract prepared by extracting with 70-90% ethanol can play a better acne removing role when being matched with other raw materials, because the ethanol volume fraction used for extraction is not in a preferred range of 70-90%, and the acne removing effect score is poorer than that of example 1.

Examples 5 to 6 show that the antibacterial rate of the edelweiss extract prepared by using the 70 to 90 percent aqueous solution of ethanol as the raw material has better antibacterial effect when the volume fraction of the ethanol in the aqueous solution of ethanol is outside the preferable range when the edelweiss extract is prepared by using the ethanol aqueous solution in the examples 5 to 6.
The preparation raw materials of comparative examples 1-8 contain one or three of narcissus bulb extract, tea tree essential oil, leontopodium alpinum extract and purslane fermented extract, and the bacteriostasis rate is lower than that of example 1, which shows that the compounded application of the narcissus bulb extract, the tea tree essential oil, the leontopodium alpinum extract and the purslane fermented extract can realize synergistic synergism in the aspect of bacteriostasis, and further achieve stronger acne removing effect.

The antioxidant properties of the formulated products prepared in examples and comparative examples were evaluated by testing the removal efficiency of DPPH. The specific evaluation method comprises the following steps: 0.5g of the formulation product was immersed in 4mL of each DPPH (100. Mu.M in 99wt% ethanol) solution (using DPPH only solution as a control) and incubated in the dark for 60min. The preparation raw materials in comparative examples 1-8 contain one or three of a narcissus bulb extract, a tea tree essential oil, a leontopodium alpinum extract and a purslane fermented extract, and DPPH clearance rates of the preparation raw materials are lower than those of example 1, which shows that the narcissus bulb extract, the tea tree essential oil, the leontopodium alpinum extract and the purslane fermented extract have stronger effects of resisting oxidation and scavenging free radicals when being compounded and applied.
Test example 4
The ATP contents of the preparations prepared in example 1 and example 7 were determined in the following manner: taking 1g of the composition with the effects of inhibiting melanin protein and improving black eye and pouch prepared in each embodiment, and adding 10mL of physiological saline for dissolving; (2) ATP was detected using an ATP detection kit (Biyun day, model S0026), luminescence values at 5min and 10min were measured on a fluorescence chemiluminescence analyzer, and the ATP content in example 1 and example 7 was compared according to the magnitude of the fluorescence value.

As can be seen from table 5, the higher the fluorescence value, the higher the ATP content, and the purslane extract used in example 7 was a commercially available purslane extract, which was lower in ATP content at 5min and 10min than in example 1, and the ATP content at 10min of example 1 was higher than 5min, indicating that the purslane fermented extract prepared by fermentation and extraction using thermophilic bacteria had more molecular motors and ATP, and further, more molecular motors could be used as skin energy factors, promoting synthesis of skin ATP energy, and bidirectionally regulating skin function state.
Test example 5
Safety evaluations were made with respect to the formulations prepared in examples 1 to 9. The specific evaluation method comprises the following steps: the test was carried out according to the human skin patch test method in the seventh chapter human safety test method in the cosmetic safety technical Specification 2015 edition. preferably, the antibacterial agent comprises any one of or a combination of at least two of 0.05-1 part of quaternary ammonium salt, 0.5-3 parts of nonandiamide MEA (membrane electrode assembly) or 0.1-1 part of hexamidine diisethionate in parts by mass.
4. The acne removal composition of any one of claims 1-3, further comprising any one or a combination of at least two of Cinnamomum zeylanicum extract, epilobium frechelii extract, rosa water, chlorella extract, or Piper methysticum extract;
preferably, the acne-removing composition further comprises any one or a combination of at least two of 0.1-1 part of cinnamomum zeylanicum extract, 0.1-1 part of salix flexuoskii extract, 3-10 parts of rose water, 0.2-2 parts of chlorella extract or 0.5-5 parts of kava extract in parts by mass.
5. The acne treatment composition according to any one of claims 1-4, wherein the acne treatment composition further comprises a functional auxiliary;
preferably, the functional auxiliary agent comprises any one or combination of at least two of heparin sodium, nicotinamide, coated salicylic acid, 3-o-ethyl ascorbic acid, dipeptide-15 or lactic acid;
preferably, the functional auxiliary agent comprises any one or combination of at least two of 0.05-1 part of heparin sodium, 0.5-3 parts of nicotinamide, 1-10 parts of coated salicylic acid, 1-5 parts of 3-o-ethyl ascorbic acid, 0.005-0.01 part of dipeptide-15 or 1-5 parts of lactic acid in parts by mass.
6. The method for preparing the acne-removing composition according to any one of claims 1-5, wherein the method for preparing the acne-removing composition comprises the following steps:
mixing the narcissus bulb extract, the purslane fermented extract, the tea tree essential oil and the alpine edelweiss extract to prepare the acne-removing composition.
